This study analysed the water quality of 156 samples collected from 13 different points within the Chief Albert Luthuli Local Municipality between January and December 2021, focusing on compliance with the South African National Standard 241 (SANS 241). Microbiological assessments revealed widespread contamination, with heterotrophic plate counts (HPCs) exceeding the recommended range in nearly all samples. The median HPC levels were particularly concerning, with the untreated source (RE000) exhibiting a median of 2555 CFU/mL. Total coliforms and Escherichia coli were present at alarming levels, indicating significant faecal contamination; the highest total coliform count, 2420 CFU/100 mL, was recorded at RE000. Physically, while all samples complied with electrical conductivity standards (0–70 µS/cm), turbidity levels showed variability, with nine samples exceeding the acceptable threshold of 1 NTU. The highest turbidity, 7.55 NTU, was recorded at the untreated source (RE000). Chemically, pH levels fell within safe ranges (6.0–9.0), with all samples being compliant with standards. However, only one sample (E001) met the free chlorine standard of 0.3–0.6 mg/L, suggesting inadequate disinfection practices. The untreated source was classified as 'Unsuitable for drinking' (WQI = 207.27). Access to safe drinking water sources and appropriate sanitation facilities remains a dream in low and middle-income countries including South Africa. This study identified the origin of faecal pollution by using quantitative polymerase chain reaction (qPCR) targeting host-specific Bacteroidales genetic markers to track the distribution of human-specific (BacHum) and animal-specific (cattle—BacCow, chicken—Cytb, pig—Pig-2-Bac, dog—BacCan) markers in water sources used by rural communities of the Vhembe District Municipality (VDM). Results revealed the prevalence of BacHum, BacCow, and BacCan in all surface water sources in Thulamela Local Municipality (TLM) and Collins Chabane Local Municipality (CLM) during wet (100%) and dry seasons (50–75%). Cytb was not detected in untreated spring water in TLM and CLM, and Pig-2-Bac was not detected in untreated hand-dug well water in TLM during both seasons. Household-level analysis detected Cytb (28.8% wet, 17.5% dry), BacHum (34.4% wet, 25% dry for Pig-2-Bac) in stored untreated spring water in CLM, and Cytb (42.9% wet, 28.5% dry) in untreated hand-dug well water in TLM. Background: Escherichia coli, a ubiquitous inhabitant of the gut microbiota, has been recognized as an indicator of fecal contamination and a potential reservoir for antibiotic resistance genes. Its prevalence in drinking water sources raises concerns about the potential dissemination of antibiotic resistance within aquatic ecosystems and the subsequent impact on public health. The ability of E. coli to acquire and transfer resistance genes, coupled with the constant exposure to low levels of antibiotics in the environment, underscores the need for comprehensive surveillance and rigorous antimicrobial stewardship strategies to safeguard the quality and safety of drinking water supplies, ultimately mitigating the escalation of antibiotic resistance and its implications for human well-being. Methods: WG5D strain, isolated from a drinking water distribution source in North-West Province, South Africa, underwent genomic analysis following isolation on nutrient agar, anaerobic cultivation, and DNA extraction. Paired-end Illumina sequencing with a Nextera XT Library Preparation kit was performed. The assembly, annotation, and subsequent genomic analyses, including phylogenetic analysis using TYGS, pairwise comparisons, and determination of genes related to antimicrobial resistance and virulence, were carried out following standard protocols and tools, ensuring comprehensive insights into the strain's genomic features. Results: This study explores the notable characteristics of E. coli strain WG5D. This strain stands out because it possesses multiple antibiotic resistance genes, encompassing tetracycline, cephalosporin, vancomycin, and aminoglycoside resistances. Additionally, virulence-associated genes indicate potential heightened pathogenicity, complemented by the identification of mobile genetic elements that underscore its adaptability. The intriguing possibility of bacteriophage involvement and factors contributing to pathogenicity further enriches our understanding. We identified E. coli WG5D as a potential human pathogen associated with a drinking water source in South Africa. The analysis provided several antibiotic resistance-associated genes/mutations and mobile genetic elements. It further identified WG5D as a potential human pathogen. The occurrence of E. coli WG5D raised the awareness of the potential pathogens and the carrying of antibiotic resistance in drinking water. Conclusions: The findings of this study have highlighted the advantages of the genomic approach in identifying the bacterial species and antibiotic resistance genes of E. coli and its potential as a human pathogen. Rivers in Africa have experienced dire pollution as a result of the poor management of wastewater effluent emanating from water resource recovery facilities (WRRFs). An integrated wastewater resource recovery model was developed and applied to identify ideal wastewater resource recovery technologies that can be used to recover valuable resources from a mixture of wastewater effluents in a case study in the Burgersfort WRRF in the Limpopo province, South Africa. This novel model incorporates the process of biological nutrient removal (BNR) with an extension of conventional methods of resource recovery applicable to wastewater. The assessment of results of effluent quality from 2016 to 2022 revealed that ammonia, chemical oxygen demand, total coliform, fecal coliform, and Escherichia coli levels were critically non-compliant with the permissible effluent guidelines, indicating a stable upward trend in terms of concentrations, and scored a very bad wastewater quality index rating. All variables assessed showed a significant loading, except for orthophosphates, and significant correlations were observed among the variables. The results of the integrated wastewater resource recovery model revealed a high probability of reclaiming recoverable resources such as nutrients, sludge, bioplastics, biofuel, metals, and water from wastewater, which have economic, environmental, and social benefits, thereby improving the effluent quality of a WRRF. We assessed the prevalence, distribution, and antibiotic resistance patterns of Escherichia coli and Enterococcus spp. isolated from raw and treated wastewater of a major wastewater treatment plant (WWTP) in KwaZulu-Natal, South Africa and the receiving river water upstream and downstream from the WWTP discharge point. Escherichia coli and enterococci were isolated and counted using the Colilert®-18 Quanti-Tray® 2000 and Enterolert®-18 Quanti-Tray 2000 systems, respectively. A total of 580 quantitative PCR-confirmed E. coli and 579 enterococci were randomly chosen from positive samples and tested for in vitro antibiotic susceptibility using the disk diffusion assay against 20 and 16 antibiotics, respectively. The removal success of the bacterial species through the treatment procedure at the WWTP was expressed as log removal values (LRVs). Most E. coli were susceptible to meropenem (94.8%) and piperacillin-tazobactam (92.9%), with most Enterococcus susceptible to ampicillin (97.8%) and vancomycin (96.7%). In total, 376 (64.8%) E. coli and 468 (80.8%) Enterococcus isolates showed multidrug resistance (MDR). A total of 42.4% (246/580) E. coli and 65.1% (377/579) enterococci isolates had multiple antibiotic resistance indices >0.2. The LRV for E. coli ranged from 2.97 to 3.99, and for enterococci the range was observed from 1.83 to 3.98. A high proportion of MDR E. coli and enterococci were present at all sampled sites, indicating insufficient removal during wastewater treatment. There is a need to appraise the public health risks associated with bacterial contamination of environmental waters arising from such WWTPs to protect the health of users of the receiving water bodies. Incorporation of black soldier fly larvae (BSFL) in fecal sludge management shows promise as a resource recovery strategy. BSFL efficiently convert organic waste into valuable lipids and protein, which can be further processed into commercial products. Ensuring the microbial safety of waste-derived products is critical to the success of resource-oriented sanitation and requires the development of effective sludge treatment. This study evaluates the microbial treatment efficacy of the viscous heater (VH) for fecal sludge management and potential application of the VH in BSFL production. The VH is a heat-based fecal sludge treatment technology that harnesses the viscosity of fecal sludge to achieve pasteurization temperatures. Inactivation of in situ Escherichia coli, total coliform, heterotrophic bacteria, and somatic coliphage was evaluated in fecal sludge that was treated for 1–6 min at VH temperature set-points of 60°C and 80°C. The VH inactivated in situ E. coli, total coliform, and somatic coliphage in fecal sludge to below the limits of detection (1- to 5-log10 inactivation) when operated at the 80°C set-point with a 1-min residence time. Both temperature set-points achieved 1- to 3-log10 inactivation of in situ heterotrophic bacteria. The VH was also evaluated as a potential pretreatment step in BSFL production. BSFL grown in untreated and VH-treated fecal sludge demonstrated similar results, indicating little impact on the BSFL growth potential by VH-treatment. However, BSFL bioconversion rates were low for both substrates (1.6% ± 0.6% for untreated sludge and 2.1 ± 0.4 VH-treated fecal sludge). [ABSTRACT FROM AUTHOR]

National and regional water quality monitoring networks have been operated in South Africa since the early 1970s. These originally had text-based inventories that were convenient for specialists who were familiar with the national networks and knew the locations of their sites of interest. However, within two decades the networks had expanded in geographical extent and variables monitored to such an extent that users needed spatial context in order to locate sites that fitted their information requirements. Mapping applications running on the Internet, such as Google Earth and Leaflet, form the foundation of a system for providing online inventories and summaries of the data available on the water quality database. The interfaces were constructed using available software, mainly ArcInfo and R. Groundwater is a valuable source of drinking water worldwide, recognized as an improved drinking water source. However, on-site sanitation systems may put groundwater at risk of fecal contamination. In the present study, two approaches were used to ascertain the sources of fecal contamination in groundwater used by communities of the Vhembe District Municipality. Overall, 87.5% of boreholes (n = 70) in the wet and 72.5% in the dry season were contaminated with Escherichia coli, and septic tank (n = 18) wastewaters displayed up to 10 4  cfu/mL E. coli. Host-specific Bacteroidales quantitative polymerase chain reaction (qPCR) assays established the presence of human (BacHum and HF183) and animal (Cytb, BacCan, and Pig-2Bac) genetic markers in groundwater from 15.7% of boreholes (wet) and 10% of boreholes (dry). No strong associations were founded between culturable E. coli counts and the presence/absence of marker genes for all the markers except for Cytb marker, which showed a weak significant correlation (r = 0.217; p = <0.01) between E. coli and the Cytb marker under dry seasonal conditions. Human markers and Cytb were present in the household septic tank wastewater samples. Significant differences in marker genes distribution in wastewater were observed using the Chi-squared test: HF183 (p = <0.001) and BacHum (p = <0.001). Overall, no association was recorded between markers in groundwater and in wastewater for 18 households' septic tanks. A combined culturable E. coli and host-specific Bacteroidales qPCR assays remain an appropriate approach for the identification of fecal contamination of groundwater. PRACTITIONER POINTS: Households primarily used private boreholes for drinking water, as a primary source. Most households used on-site sanitation systems, including ventilated improved pit latrines and flush toilets connected to septic tanks. Escherichia coli was detected in groundwater, and the sources of fecal contamination were humans and animals (pigs, dogs, and chickens). The presence of human and animal markers in groundwater suggests that humans and animals are liable for fecal contamination. Numerous behavioral and ecological factors are associated with parasite transmission. One factor explored in human research, but absent from nonhuman primate research, is parasite transmission from soil ingestion. Human studies suggest geophagy, the regular and deliberate consumption of soil, increases risk of soil-transmitted helminth (STH) infection. Geophagy, which is prevalent in nonhuman primates, has several positive associations: gastrointestinal distress alleviation, possible mineral supplementation, and bacterial infection prevention. Our objective was to determine whether STH transmission was possible from deliberate or accidental soil ingestion, in a troop of chacma baboons ( Papio hamadryas ursinus) that engaged in geophagy, foraged on the terrestrial substrata, and had a Trichuris sp. sample prevalence of 100%. We collected and analyzed 80 soil samples from geophagy and ground foraging sites on and around Wildcliff Nature Reserve, South Africa. Forty soil samples were collected from sites where soil was consumed, and 40 were collected from sites where soil was not consumed. At geophagy sites, the number of Trichuris sp. eggs recovered varied significantly between areas used and unused by the baboons, suggesting behavior is an important factor. In contrast, at foraging sites, there was only a tendency to recover more eggs at used than unused areas, and we propose egg recovery was influenced by fecal contamination that occurred throughout foraging stands. The difference in egg recovery between used areas at geophagy sites and used areas at foraging sites was not significant. These preliminary findings suggest both geophagy and foraging sites are a potential source of STH infection for this troop. Whether geophagy benefits outweigh the potential cost of parasite reinfection for this baboon troop is unknown, so we encourage future research on the influence that host foraging behavior may have on parasite reinfection. Total coliforms are a group of bacteria found in high numbers in mammalian intestines; hence their presence in water indicates the possible contamination with faecal material. Total and faecal coliform counts were monitored over a period of 18 months using mFC, m-Endo and CM1046 media together with enzymatic assays on 215 environmental water samples obtained from the Eastern Cape Province of South Africa. A positive correlation, with an R2 value of 0.9393 was observed between faecal and total coliform colony units employing mFc and m-Endo media, and 0.8818 using CM1046 media. Also, a positive correlation was observed between Escherichia coli colony-forming units and β-D-galactosidase (B-GAL) activity (R2=0.8542). Overall, this study indicated that faecal contamination of environmental water samples could be monitored by measuring total coliform β-galactosidase activity and total coliform colony-forming units. This study assessed the groundwater quality around two municipal solid waste landfill sites, in the city of Bloemfontein, Free State Province, South Africa. The two landfill sites are located in two contrasting geological terrains, with both lacking some basic facilities found in a well-designed landfill. A total of eight groundwater samples were collected from pollution monitoring boreholes near the two landfill sites, with five samples representing the northern landfill site and three samples representing the southern landfill site. The samples were collected in the autumn and winter seasons to assess any possible seasonal variations. They were analysed for physicochemical (pH, electrical conductivity (EC), total dissolve solids (TDS), chemical oxygen demand (COD) and total organic carbon (TOC)) and microbiological parameters (Escherichia coli, total coliform). The results of the analysis showed that the waters from both landfills were generally dominated by Ca, Mg, SO4, and HCO3 ions. Some of the major anions and cations in the water samples were above the South African National Standard (SANS241:2015) and World Health Organisation (WHO) permissible limits for drinking water. Majority of the boreholes had total dissolved solids and electrical conductivity values exceeding the SANS 241:2015 and WHO permissible limits. Piper trilinear plots for the two landfill sites showed that Ca(Mg)HCO3 water type predominates, but Ca(Mg)SO4 and Ca(Mg)Cl were also found. These water types were further confirmed with expanded Durov diagrams, indicating that that the boreholes represented a water type that is seldom found which is undergoing ion exchange, typical of sulphate contamination. From the SAR diagrams, boreholes in the northern landfill site had a high salinity hazard with only one borehole in the southern landfill site having a high salinity hazard. The geology was found to play a significant role in the distribution of contaminants into the groundwater systems in the study area. The study concluded that the northern landfill site had a poorer water quality in comparison to the southern landfill site based on the analysed physicochemical parameters. However, the southern landfill site showed significant microbial contamination, due to the elevated amount of E. coli and total coliform concentrations. The high permeability of the weathered dolerites in the northern landfill site might have enabled the percolation of contaminants into the groundwater resulting in the poorer water quality. Background: Access to safe water for drinking and domestic activities remains a challenge in emerging economies like South Africa, forcing resource-limited communities to use microbiologically polluted river water for personal and household purposes, posing a public health risk. This study quantified bacterial contamination and the potential health hazards that wastewater treatment plant (WWTP) workers and communities may face after exposure to waterborne pathogenic bacteria in a WWTP and its associated surface water, respectively. Results: Escherichia coli (Colilert®-18/ Quanti-Tray® 2000) and enterococci (Enterolert®/ Quanti-Tray® 2000) were quantified and definitively identified by real-time polymerase chain reaction targeting the uidA and tuf genes, respectively. An approximate beta-Poisson dose-response model was used to estimate the probability of infection (Pi) with pathogenic E. coli. Mean E. coli concentration ranged from 2.60E+ 02/100 mL to 4.84E+ 06/100 mL; enterococci ranged from 2.60E+ 02/100 mL to 3.19E+ 06/100 mL across all sampled sites. Of the 580 E. coli isolates obtained from this study, 89.1% were intestinal, and 7.6% were extraintestinal pathogenic E. coli. The 579 enterococci obtained were 50.4% E. faecalis (50.4%), 31.4% E. faecium, 3.5%, E. casseliflavus and 0.7% E. gallinarum. The community health risk stemming from the use of the water for recreational and domestic purposes revealed a greater health risk (Pi) from the ingestion of 1 mL of river water from upstream (range, 55.1–92.9%) than downstream (range, 26.8–65.3%) sites. The occupational risk of infection with pathogenic E. coli for workers resulting from a once-off unintentional consumption of 1 mL of water was 0% (effluent) and 23.8% (raw influent). Multiple weekly exposures of 1 mL over a year could result in a Pi of 1.2 and 100% for the effluent and influent, respectively. Conclusion: Our findings reveal that there is a potentially high risk of infection for WWTP workers and communities that use river water upstream and downstream of the investigated WWTP. This work evaluates and analyses the Greenhouse Gas (GHG) emission reduction potential of Solar Water Heaters (SWH) in Daveyton, in Ekurhuleni. As an energy intensive country, with its energy mix dominated by fossil-fuel energy sources, South Africa is among the largest emitters of GHGs globally. Consequently, the country made commitments to substantially reduce its carbon footprint from the energy sector, through renewable energy initiatives. A solar water heater programme was subsequently introduced as an initiative to reduce GHG emissions in the residential sector. Objectives of the study were to determine the amount of energy saved and GHG emission reduction through the installation of SWHs; to determine the efficiency and sufficiency of SWHs for the average family; and to determine the contribution of SWHs towards reduction of GHG emissions. These objectives were achieved by conducting a survey in low-income households that were piloted for the SWH installation programme in Daveyton, Ekurhuleni. The findings of the study suggested that a total saving of 89.74 kWh of energy per day and reduction of 63.1 kg of carbon dioxide (CO 2) equivalent were achieved through the SWH programme for the entire sample area per day. Persistent organic pollutants are lipophilic contaminants which can accumulate in marine food chains, causing health issues for human consumers. Farmed C. meridionalis and M. galloprovincialis were collected and analysed for POPs, extracted using QuEChERS method and analysed via GC-MS/MS. Detected OCPs (trans -permethrin > cis -permethrin > dieldrin > chlordane > chloro-benzilate > endosulfan > nonachlor > DDD) were highest contributors to POP contamination (average: 0.7 – 48 ng/g dry weight), followed by PAHs (flourene > pyrene > fluoranthene > benzo(a)pyrene > benz(a)anthracene; average: 1 – 13.4 ng/g d.w.), and PCBs (180 > 149 > 28 > 110 > 153 > 52 > 18 > 44 > 118 > 138; average: 0.3 – 3.2 ng/g d.w). Temporal differences were detected in both species for cis - and trans -permethrin, and PCBs 18, 118 and 149, with seasonal changes linked gametogenic cycle (fat) of mussels.
