The aim of this study was to investigate the test-retest reliability of the motives for undertaking physical activity (PA) items from the Health Behavior in School-Aged Children (HBSC) study questionnaire among Slovak and Czech adolescents and to determine whether this reliability differs by gender, age group and country. We obtained data from 580 students aged 11 and 15 years old (51.2% boys) who participated in a test and retest study with a four-week interval in 2013 via the Health Behavior in School-Aged Children cross-sectional study in the Czech Republic and Slovakia. We estimated the test-retest reliability of all 13 dichotomized motives by using Intraclass Correlation Coefficients (ICC) and Cohen's Kappa statistics, for continuous and dichotomized motives, respectively. Test-retest reliability showed moderate agreement for nine motives (ICC from 0.41 to 0.60) and fair agreement for four motives (ICC from 0.33 to 0.40). Kappa statistics were similarly moderate to large (0.33 to 0.61), except for three motives with small or trivial correlations. The motives "To improve my health" and "To enjoy the feeling of using my body" had consistently low Kappas and correlations. Overall, the results of this study suggest that most questions on motives for PA on the HBSC questionnaire have acceptable test-retest characteristics for use among adolescents.

The development of the new technologies such as the next-generation sequencing (NGS) makes more accessible the diagnosis of genetically heterogeneous diseases such as Lynch syndrome (LS). LS is one of the most common hereditary form of colorectal cancer. This autosomal dominant inherited disorder is caused by deleterious germline mutations in one of the mismatch repair (MMR) genes - MLH1, MSH2, MSH6 or PMS2, or the deletion in the EPCAM gene. These mutations eventually result in microsatellite instability (MSI), which can be easily tested in tumor tissue. According to the actual recommendations, all patients with CRC that are suspect to have LS, should be offered the MSI testing. When the MSI is positive, these patients should be recommended to genetic counseling. Here we report a pilot study about the application of NGS in the LS diagnosis in patients considered to have sporadic colorectal cancer. The inclusion criteria for the NGS testing were MSI positivity, BRAF V600E and MHL1 methylation negativity. We have used 5 gene amplicon based massive parallel sequencing on MiSeq platform. In one patient, we have identified a new pathogenic mutation in the exon 4 of the MSH6 gene that was previously not described in ClinVar, Human Gene Mutation Database, Ensembl and InSight databases. This mutation was confirmed by the Sanger method. We have shown that the implementation of new criteria for colorectal patients screening are important in clinical praxis and the NGS gene panel testing is suitable for routine laboratory settings.

Objectives: Physical activity significantly contributes to overall health and the level of activity during adolescence has been associated with physical activity in adulthood. The aim of this paper was to analyse motives for young people undertaking leisure time physical activity in the Czech and Slovak Republics., Methods: Eleven to fifteen year old schoolchildren (N=9,014; mean age=13.59) participating in the Health Behaviour in School-aged Children 2009/2010 study in the Czech and Slovak Republics completed 12 items measuring physical activity motivations. To analyse the factor structure of motives for physical activity, Confirmatory Factor Analysis (CFA) was conducted using Principal Component Analysis (PCA) with varimax rotation., Results: Contrary to previous research that reported three main motives for physical activity (health, social, achievement), our study suggests four motives. The additional motive is a combination of internal and external motivations and was called the "Good Child Motive"., Conclusions: Better understanding of motives behind physical activity or inactivity of young people could significantly contribute to evidence based planning and development of national strategies for public health and active living., (Copyright© by the National Institute of Public Health, Prague 2015.)

Objectives: Better assessment of the reliability of the physical activity and sedentary behaviour items across countries in all WHO regions is highly needed. The aim of the study was to examine the test-retest reliability of selected physical activity and sedentary behaviour items of the HBSC questionnaire in Czech, Slovak and Polish adolescents., Methods: We obtained data from 693 Czech, Slovak and Polish (50.9% boys) primary school pupils, grades five (mean age = 11.08; SD = 0.45) and nine (mean age = 15.12; SD = 0.45), who participated in a test-retest study in 2013. We used the single measures of Intraclass Correlation Coefficients (ICC) and Cohen's Kappa statistic to estimate the test-retest reliability of all selected items within the sample and stratified by gender, age group and country., Results: Both physical activity items (VPA and MVPA) and most of the sedentary behaviour items showed moderate agreement (ICC 0.41-0.60) and a similarly moderate correlation (Cohen's Kappa 0.3-0.5) after dichotomization., Conclusions: The physical activity and sedentary behaviour items of the HBSC questionnaire seem to be at the borderline of reliability to be used in adolescents.
