Study Question: Do distinct subpopulations of decidual stromal cells (DSCs) exist and if so, are given subpopulations enriched in recurrent miscarriage (RM)?Summary Answer: Three subpopulations of DSCs were identified from which inflammatory DSCs (iDSCs) and glycolytic DSCs (glyDSCs) are significantly enriched in RM, with implicated roles in driving decidual inflammation and immune dysregulation.What Is Known Already: DSCs play crucial roles in establishing and maintaining a successful pregnancy; dysfunction of DSCs has been considered as one of the key reasons for the development of RM.Study Design, Size, Duration: We collected 15 early decidual samples from five healthy donors (HDs) and ten RM patients to perform single-cell RNA sequencing (scRNA-seq). A total of 43 RM patients and 37 HDs were enrolled in the validation cohort.Participants/materials, Setting, Methods: Non-immune cells and immune cells of decidual tissues were sorted by flow cytometry to perform scRNA-seq. We used tissue microarrays (TMA) to validate three distinct subpopulations of DSCs. The expression of inflammatory and glycolytic proteins by DSCs was validated by immunohistochemistry (IHC) and multiplex immunohistochemistry (mIHC). Different subsets of decidual NK (dNK) cells and macrophages were also validated by multicolor flow cytometry and mIHC. Cell ligand-receptor and spatial analyses between DSCs and immune cells were analyzed by mIHC.Main Results and the Role Of Chance: We classify the DSCs into three subtypes based on scRNA-seq data: myofibroblastic (myDSCs), inflammatory (iDSCs) and glycolytic (glyDSCs), with the latter two being significantly enriched in RM patients. The distribution patterns of DSC subtypes in the RM and HD groups were validated by mIHC. Single-cell analyses indicate that the differentiation of iDSCs and glyDSCs may be coupled with the degrees of hypoxia. Consequently, we propose a pathological model in which a vicious circle is formed and fueled by hypoxic stress, uncontrolled inflammation and aberrant glycolysis. Furthermore, our results show that the inflammatory SPP1+ macrophages and CD18+ dNK cells are preferentially increased in the decidua of RM patients. To explore the heterogeneity of CD24+ decidual stromal cells (DSCs) in patients with recurrent miscarriages (RMs). We have discerned that the expression of CD24 serves to differentiate two stable and functionally distinct lineages of DSCs. The heterogeneity of CD24+ DSCs has been scrutinized, encompassing variances in stromal markers, transcriptional profiles, metabolic activity, and immune regulation. A total of 129 early decidual samples were obtained, comprising 36 from healthy donors and 93 from patients with RMs. Blood samples were collected before the surgical procedure. Paraffin-embedded segments from 20 decidual samples of patients with RMs were obtained. None. The flow cytometry was used to quantify the expression of CD24+ DSCs in both healthy donors and patients with RMs, although it also evaluated the cellular heterogeneity. To ascertain the transcriptomic profiles of CD24+ DSCs by reanalyzing our single-cell transcriptomic data. Additionally, to measure the metabolomic activity of CD24+ DSCs from patients with RMs, ultraperformance liquid chromatography-mass spectrometry was employed. Through the implementation of a coculture system, we unraveled the role of CD24+ DSCs in immune regulation. Patients with RMs exhibit a notable enrichment of CD24+ DSCs, revealing a pronounced heterogeneity characterized by variations in stromal markers and transcriptional profiles. The heightened enrichment of CD24+ DSCs may play a pivotal role in triggering decidual inflammation and dysfunction in decidualization. Furthermore, CD24+ DSCs showed diverse metabolic activities and impeded the induction of naïve CD4+ T cells into regulatory T cells through the abundant secretion of 3-hydroxyisovaleric acid. Finally, our investigations have revealed that intraperitoneal administration of 3-hydroxyisovaleric acid in mouse models can elevate the risk of RM. We have successfully identified a disease-associated subset of CD24+ decidual stromal cells that could potentially contribute to the development of RM through the impairment of decidual immune tolerance. During rapid urbanization, microclimate environment deterioration through events such as haze pollution and heat waves has continuously occurred in cities, which greatly affects the living environment, production activities, and health of urban residents. Therefore, it is particularly necessary to explore methods for controlling and optimizing the urban microclimate environment. In this paper, based on the mechanism of the effect of urban spatial structure at street-level on the distribution of atmospheric particulate matter, an indicator system that can be employed to comprehensively describe and quantify urban morphological structure at street-level was constructed from eight aspects: the spatial morphology of street-valleys, intensity of land use and development, geometric structure of buildings, inhomogeneity of buildings, roughness of the underlying surface, distribution of ecological landscapes, 3D architectural landscape morphology, and ventilation potential. Furthermore, using satellite remote sensing images and vector thematic maps of Shanghai, indicator factors were quantified by applying GIS technique. The intrinsic mechanism of the influence of the urban morphology on the diffusion and transport of atmospheric particulate matter was comprehensively analyzed by combining statistical methods and data mining algorithm, and eight key dominant factors were identified that can be considered to improve the urban ventilation conditions and help control urban air pollution, namely, the land use intensity, urban canopy resistance, vegetation cover, spatial congestion rate, comprehensive porosity, height-to-gross floor area ratio, building density, and average building volume ratio. As such, according to the quantitative analysis results for various combinations of the dominant factors, a spatial optimization strategy at street-level that can help improve the urban air quality was proposed in terms of identifying the pathways through which urban spatial elements affect the distribution of particulate matter, i.e., controlling the source–flow diversion–flow convergence process. Cognitive impairment using education-based cutoff points for CMMSE scores in elderly Chinese people of agricultural and rural Shanghai China. Objectives - To evaluate cognitive impairment (CI) in rural China using the Chinese version of the Mini-Mental Status Examination (CMMSE) and compare the prevalence of CI using two different cutoff points. Materials & methods - A population-based survey was conducted of 2809 people aged 60 years and above in a community of two towns (Huaxin and Xujing) in the Qingpu district, located in the western suburb of Shanghai. Face-to-face interviews were carried out to collect relevant information with questionnaires. The Chinese version of the Mini-Mental State Examination with either a 23/24 cutoff point or a cutoff point varying according to education level (AEL) was used to screen subjects for CI. Results - Among these subjects, the mean age was 70.6 years (SD = 6.6) and ranged from 60 to 92 years and included 1010 (36.0%) men and 1799 (64.0%) women. The mean age was 70.7 years (SD = 6.4) for men and 70.5 years (SD = 6.7) for women. Of the 2809 subjects, 2010 (71.5%) had no formal education, 607 (21.6%) completed 1-6 years of education, and 173 (6.2%) completed more than 6 years of school education. The prevalence of CI was 35.6% (95% CI: 33.8-37.4) for both genders when the cutoff point of 23/24 was used. However, when the cutoff point was altered with respect to different education levels, the prevalence of CI was 7.0%. For each item of the CMMSE, increased years of education correlated with a higher item score, with the exception of the 'Naming' item score. Conclusions - This study demonstrates that screening of CI using the AEL cutoff scores is feasible in a low-education population. The COVID-19 lockdown has deepened inequity among vulnerable groups, such as the elderly. Reducing inequity in access to essential service facilities is an effective way to improve community resilience in dealing with pandemics. In this research, three indexes were created to measure community resilience in accessing essential services. Specifically, we have considered the different walking capacity and different needs of the elderly and the general population. We selected Shanghai as the case for our research and analyzed the spatial patterns of both space-based and population-based essential service facilities. The Lorenz curve and the Gini coefficient were used to measure the spatial equity. And, we attempted to reveal the relationships between the population density and three indexes through bivariate Local Indicators of Spatial Association. The results suggest that the Diversity Index enjoys the highest equity, followed by the Demand Accessibility Index, and the equity of the Per Capita Quantity Index is the lowest. Furthermore, the accessibility of essential services in urban areas is excellent, while in some suburban areas it is low. In March 2022, a new wave of COVID-19 outbreak occurred in Shanghai due to the widespread transmission of the Omicron variant. A two-month citywide lockdown was implemented from April 1st to May 31st, adopting measures such as zone-based classification and grid management. This unique social event provided an "ideal air quality experiment" for pollution research. The rapid reduction in economic activities during the lockdown had many positive impacts on the environment, leading to overall improvements in air quality. Particularly, the concentration of NOx, one of the precursors to O3, significantly decreased. However, O3, as a typical secondary pollutant, showed a noticeable increase. This study uses the WRF-CAMx-OSAT air quality model method to analyze the source of O3 pollution in Shanghai from April to May 2022. The impact of O3 precursor control, sector sources, and regional contributions on the formation of O3 pollution in Shanghai is analyzed in depth. During the pandemic lockdown period, it was found that, in Shanghai, the overall O3 levels were controlled by VOCs (Volatile Organic Compounds), and controlling VOCs proved to be an effective measure in reducing O3 concentrations in Shanghai. Compared with the same period in 2021, the proportion of road traffic sources contributing to ozone concentration has significantly decreased from 70.61% to 64.3%, but they are still the largest contributor. The contribution of industrial emissions to the ozone concentration has significantly risen from 20.71% to 26.36%, making them still the second largest contributor. Industrial and traffic sources are emission sources that require particular attention. The contribution ratio of local sources to external transport is about 7:3, which is higher than the ratio of local sources to external transport in the same period of 2021, which is about 6:4. The local ozone is the main source of ozone concentration in Shanghai, and controlling local source emissions is the key to controlling ozone concentration in the Shanghai area. When excluding the impact of long-range transport, the main areas contributing to O3 formation from local sources are Baoshan District, Jiading District, Qingpu District, and Chongming District, accounting for approximately 41.12% of the total absolute contribution. Different source regions exhibit significant spatial variations in their contributions to the ozone concentration. Anthropogenic sources of fine particulate matter (PM2.5) threaten ecosystem security, human health and sustainable development. The accuracy prediction of daily PM2.5 concentration can give important information for people to reduce their exposure. Artificial neural networks (ANNs) and wavelet-ANNs (WANNs) are used to predict daily PM2.5 concentration in Shanghai. The PM2.5 concentration in Shanghai from 2014 to 2020 decreased by 39.3%. The serious COVID-19 epidemic had an unprecedented effect on PM2.5 concentration in Shanghai. The PM2.5 concentration during the lockdown in 2020 of Shanghai is significantly reduced compared to the period before the lockdown. First, the correlation analysis is utilized to identify the associations between PM2.5 and meteorological elements in Shanghai. Second, by estimating twelve training algorithms and twenty-one network structures for these models, the results show that the optimal input elements for daily PM2.5 concentration predicting models were the PM2.5 from the 3 previous days and fourteen meteorological elements. Finally, the activation function (tansig-purelin) for ANNs and WANNs in Shanghai is better than others in the training, validation and forecasting stages. Considering the correlation coefficients (R) between the PM2.5 in the next day and the input influence factors, the PM2.5 showed the closest relation with the PM2.5 1 day lag and closer relationships with minimum atmospheric temperature, maximum atmospheric pressure, maximum atmospheric temperature, and PM2.5 2 days lag. When Bayesian regularization (trainbr) was used to train, the ANN and WANN models precisely simulated the daily PM2.5 concentration in Shanghai during the training, calibration and predicting stages. It is emphasized that the WANN1 model obtained optimal predicting results in terms of R (0.9316). These results prove that WANNs are adept in daily PM2.5 concentration prediction because they can identify relationships between the input and output factors. Background: Brain Magnetic Resonance Imaging (MRI) examination of cerebral small vessel disease (CSVD) may help screen vascular cognitive impairment. A recently estimated CSVD score system was suggested to capture the overall CSVD burden. The study aimed to detect the association between systemic evaluation score of cerebral vascular imaging parameters with cognitive functions. Methods: This was a cross-sectional study in community settings. From October 2017 to September 2018, elder (≧60) residents were recruited through on-site visit in 6 communities from Shanghai, China. The participants underwent brain MRI, carotid ultrasound, laboratory tests of blood and urine samples. Cognitive function was evaluated using Mini-Mental State Examination (MMSE). MRI score of CSVD was calculated according to the 2012 standard for the evaluation of statistical changes in imaging. Results: Total 171 subjects completed survey and examinations. There were 55 participants diagnosed with cognitive impairment, with a total percentage of 32.2%. Participants with and without cognitive impairment showed significant differences in age, BMI and education level. Cognitive impaired participant had more disease history/comorbidity of hypertension and chronic renal insufficiency, higher level of creatinine, as well as lower level of full blood count (FBC) and alanine aminotransferase (ALT). A significant difference was detected in CSVD score between participants with and without cognitive impairment. Results of linear regression analysis showed significant negative correlations between MMSE score and both left and right carotid artery peak systolic velocity (PSV), however the CSVD score was only borderline (P = 0.0566) positively correlated with MMSE. Multivariate linear correlation analysis including all collected risk factor data showed that left carotid artery PSV score was among the independent negative correlated factors of MMSE. Multivariate binary logistic analysis showed that age, education and history of hypertension were the only statistically associated factors of cognitive impairment. Conclusions: The current study identified high prevalence of cognitive impairment in a Chinese community. In addition, correlations between cerebral vascular disease imaging status and cognitive functions were confirmed although the sample size limited the possibility of screening cognitive impairment with imaging technique. Vulnerability research is the core issue and one of the research hotspots of sustainable development science. Vulnerability and its evaluation framework provide a new perspective for rural social-ecosystem studies. This paper introduced the 'input-output' efficiency theory and constructed the 'SEE-PSR' framework for the analysis of social-ecosystem vulnerability in the rural area in Qingpu District of Shanghai City. The DEA models, spatial autocorrelation model, multivariate logistic regression model, geographical detector and hierarchical cluster model were used to analyze the spatial differences of social-ecosystem vulnerability, and its geographical impact mechanisms and ecological restorations, in 184 administrative villages in this area. The results can be divided into three main points. (1) The results of the 'input-output' efficiency model of the EW-DEA based on entropy weight aggregation crossover was more reliable and accurate for the evaluation of rural social-ecosystem vulnerability. The vulnerability of the social-ecosystems in the administrative villages showed a trend of gradual decline from east to west, with an average value of vulnerability of 0.583, and the vulnerability of social systems had become an important factor in constraining the decrease of the vulnerability of the social-ecosystems in the region. (2) The distances from the center of Shanghai City, from Dianshan Lake, from the center of Qingpu District and from the water area were the four dominant geographical factors affecting the vulnerability of the social-ecosystem in this region. The geographical impacts exhibited the spatial differentiations of systemic structure, the substitution of typological attributes and the transformation level. (3) The geographical factors coupling the impact types of the social-ecosystem vulnerability were divided spatially into 10 types. The geographic multi-factor coupling impact types were dominant, which presented multi-cyclic spatial patterns and were dominated by the central multi-factor which was surrounded by the single factor types on both sides. According to the different types, some feasible ways of ecological restoration were proposed, which drew on the experiences of integrated territory consolidation to remediate the vulnerability of rural social-ecological systems. The results of this study can provide scientific reference for rural spatial reconstruction, regional ecological restoration and sustainable development for the regions characterized by conflict in the 'strict protection of the ecological environment and vigorous development of the economy'. Background: H type hypertension is defined as homocysteine (Hcy) ≥ 10 μmol/L in combination with primary hypertension. Studies demonstrated that the existence of hyperhomocysteine (HHcy) in hypertensive exacerbates the poor outcome of cardiocerebral incidents. This study was to investigate the current epidemic situation of H type hypertension and determine the risk factors in order to find intervention targets for H type hypertensives.Methods: We conducted a cross-sectional study using cluster sampling design in Shanghai, China from July 2019 and April 2020. 23,652 patients with primary hypertension were enrolled in this study. Their medical information was recorded, and the level of Hcy concentrations and methylenetetrahydrofolate reductase (MTHFR) C677T polymorphisms were detected.Results: In total, 22,731 of 23,652 patients were recorded. The mean age was 68.9 ± 8.6 y and 43% were men. 80.0% of the enrolled patients had H type hypertension. The frequency of allele T was 40.9%, and the proportions of the CC, CT, and TT genotypes were 36.1%, 46.0%, and 17.9%, respectively. Compared with the TT genotype, the plasma Hcy concentration levels were lower in patients with the CC/CT genotype (18.96 ± 13.48 μmol/L vs. 13.62 ± 5.20/14.28 ± 5.36, F = 75.04, p < 0.01). The risk for H type hypertension was higher in elderly people. Men had ~ 5.55-fold odds of H type hypertension compared with women. Patients with CT genotype and TT genotype had ~ 1.36- and ~ 2.76-fold odds of H type hypertension compared with those with CC genotype, respectively. Smoking and diabetes were not significantly associated with H type hypertension.Conclusions: The prevalence of H type hypertension in patients with primary hypertension was 80.0%, which was higher than the 75% found in prior report in China. The configuration of a multifunctional rural landscape is critical for its protection. Although studies on multifunctional rural landscapes have been conducted, there is a lack of information regarding the spatiotemporal characteristics and tradeoff/synergy relationships of rural landscape functions in the time series on the administrative unit scale. The purposes of this study were to (1) analyze the spatial-temporal distribution characteristics of the tradeoff and synergy from the perspective of multifunctionality for efficient use of rural landscape resources and (2) formulate regional sustainable development policies to minimize the conflict between people and nature. Aiming at the scientific representation of landscape function and the quantification of landscape multifunctional relationships, and by taking Qingpu District of Shanghai as an example, six kinds of rural landscape functions were constructed according to the functional framework of "productive function, living function and ecological function". Based on the data for 1980 to 2018, the characteristics of variations of multifunctional tradeoff and synergy relationships of the rural landscape in 184 administrative villages were studied by the methods of Spearman rank correlation coefficient analysis and bivariate spatial autocorrelation. The following results were obtained. 1) The fine division of rural landscape function types was realistic and necessary for analyzing the regional multi-function relationships in the regions with rapid development. In the process of rapid urbanization, the rural landscape functions of urban suburban areas changed under the combined action of natural resource endowment, social and economic conditions and other internal and external factors. As a result, the agricultural production function could not replace the economic development function and become the function of rural landscape production. The research results of Qingpu District showed that the agricultural production function was no longer the primary functional form, yet the economic development function became the dominant function in this area. 2) Temporal and spatial analysis methods of rural landscape functions can accurately and comprehensively reflect the evolution of the characteristics of multifunction tradeoffs and synergies. According to the Spearman rank correlation analysis of the multifunction value of the rural landscape in the time dimension, the results masked the differences of resource and environment carrying capacity caused by the differences of regional landscape resource endowment in the spatial dimension. 3) The spatial and temporal differences of the multi-functional tradeoffs and synergies of the rural landscape in Qingpu District from 1980 to 2018 were significant. There was significant heterogeneity of tradeoffs and synergies between functions in the spatial pattern, with clustering characteristics. Meanwhile, as for the temporal pattern, the tradeoffs and synergies of functions changed differently in terms of Moran's I and the correlation coefficient. Background: Screening for cognitive impairment (CI) is often hampered by lack of consensus as to which screening instrument to use. The aim is to assess the consistence and applicability of different CI screening tools.Method: In a cross-sectional study from October 2017 to September 2018 in 7 communities in Shanghai, China, elder (≧60) residential volunteers with no history of major cardiovascular diseases, cancers and other comorbidities known to affect cognitive functions were recruited. The participants underwent tests with 7 cognitive function screening instruments. Multivariate linear regressions were performed to test correlations between demographic characteristics, including gender, age, education, and marital status, with cognitive test scores. Mini-Mental State Examination (MMSE) score adjusted according to the correlation coefficients was used to detect CI with a cutoff of 24. Other cognitive function scores were compared between participants with and without CI. In addition, Pearson's correlation test was used to detect association between different test scores.Results: 172 participants with relatively low education levels were included. Age and education showed significant association with cognitive test scores. Using adjusted MMSE, 39.6% of participants were identified with CI, while the percentage was 87.2% when adjusted Montreal Cognitive Assessment (MoCA) with cutoff of 26 was used. Analysis of "abnormal" test scores showed that MMSE had the highest percentage of valid data (98.8%). MoCA and Isaacs test of Verbal Fluency (VF) score had correlation with most the other scores, while MMSE only significantly associated with VF and MoCA.Conclusions: MMSE may still present the most applicable tools for quick screen of cognitive functions, especially when environmental conditions may interfere with participants' attention. This paper contributes to an in-depth understanding of how the mega- event contributes glurbanization of entrepreneurial city through a case study of Expo 2010 in Shanghai. It argues that spatial-related transformation is central to mega-event approach to glurbanization yet the soft power building is uncertain. It implies that the domestic impacts of mega- events are likely to be more profound than their global influences. This corresponds to the capitalist transformation from Fordist-Keynesianism to neoliberalism, in which mega-events such as Olympic Games and World Exposition have increasingly been incorporated into urban development plan to boost urban agenda. Although the profile of world fairs is reduced and does not have the international impacts that they used to have, Shanghai Expo 2010, the first Expo ever held in a developing country, is pinned hope on as the "Turn to Save the World Expo" and is unusually ambitious to bring opportunities in urban transformation. With a well-developed framework of glurbanization entailed by entrepreneurial city, this research enriches glurbanization theory by a thorough examination of Shanghai Expo. It finds that Expo-led landscape reconfiguration, spatial restructuring, and new sources provision effectively transformed Shanghai, propelling glurbanization in diminutive spatial scale. Yet, it remains powerless to impress the world as the voice of domestic propaganda is limited in the Western mainstream media. In all, the Expo case well exemplifies the power of mega-event approach to advancing local agenda, especially in spatial transformation per se, as well as its constraints in (re)shaping a global discourse. China has experienced an unprecedented level of farmland conversion for urban development despite the fact that farmland remains a highly scarce and valuable resource in this densely populated country. The causes of this presumably unreasonable phenomenon are examined from the perspective of competing claims for farmland conversion in Shanghai. The reasons behind such excessive farmland conversion for urban development in transitional China are generalized as follows: (1) the present urban-rural dualistic structure influences farmers to pursue economic rather than agricultural claims, (2) local governments with economic claims exercise strong incentives and power in converting farmland for urban purposes, (3) local governments largely undermine the enforcement of the central government's policies (which upholds agricultural claims), (4) other groups with noneconomic claims are excluded from the farmland conversion process, and (5) the village cadres and role model campaigns effectively reduce the conflict between farmers and local governments regarding the amount of compensation. The aim of the current study is to analyze and extract the useful patterns from Location-Based Social Network (LBSN) data in Shanghai, China, using different temporal and spatial analysis techniques, along with specific check-in venue categories. This article explores the applications of LBSN data by examining the association between time, frequency of check-ins, and venue classes, based on users' check-in behavior and the city's characteristics. The information regarding venue classes is created and categorized by using the nature of physical locations. We acquired the geo-location information from one of the most famous Chinese microblogs called Sina-Weibo (Weibo). The extracted data are translated into the Geographical Information Systems (GIS) format, and after analysis the results are presented in the form of statistical graphs, tables, and spatial heatmaps. SPSS is used for temporal analysis, and Kernel Density Estimation (KDE) is applied based on users' check-ins with the help of ArcMap and OpenStreetMap for spatial analysis. The findings show various patterns, including more frequent use of LBSN while visiting entertainment and shopping locations, a substantial number of check-ins from educational institutions, and that the density extends to suburban areas mainly because of educational institutions and residential areas. Through analytical results, the usage patterns based on hours of the day, days of the week, and for an entire six months, including by gender, venue category, and frequency distribution of the classes, as well as check-in density all over Shanghai city, are thoroughly demonstrated.
