Aims Although elasmobranchs are consumed worldwide, bacteriological assessments for this group are still sorely lacking. In this context, this study assessed bacteria of sharks and rays from one of the most important landing ports along the Rio de Janeiro coast. Methods and results Bacteria were isolated from the cloacal swabs of the sampled elasmobranchs. They were cultured, and Vibrio, Aeromonas , and Enterobacterales were isolated and identified. The isolated bacteria were then biochemically identified and antimicrobial susceptibility assays were performed. Antigenic characterizations were performed for Salmonella spp. and Polymerase Chain Reaction (PCR) assays were performed to identify Escherichia coli pathotypes. Several bacteria of interest in the One Health context were detected. The most prevalent Enterobacterales were Morganella morganii and Citrobacter freundii , while Vibrio harveyi and Vibrio fluvialis were the most prevalent among Vibrio spp. and Aeromonas allosacharophila and Aeromonas veronii bv. veronii were the most frequent among Aeromonas spp. Several bacteria also displayed antimicrobial resistance, indicative of Public Health concerns. A total of 10% of Vibrio strains were resistant to trimethoprim-sulfamethoxazole and 40% displayed intermediate resistance to cefoxitin. Salmonella enterica strains displayed intermediate resistance to ciprofloxacin, nalidixic acid and streptomycin. All V. cholerae strains were identified as non-O1/non-O139. The detected E. coli strains did not exhibit pathogenicity genes. This is the first study to perform serology assessments for S. enterica subsp. enterica isolated from elasmobranchs, identifying the zoonotic Typhimurium serovar. Salmonella serology evaluations are, therefore, paramount to identify the importance of elasmobranchs in the epidemiological salmonellosis chain. Conclusions The detection of several pathogenic and antibiotic-resistant bacteria may pose significant Public Health risks in Brazil, due to high elasmobranch consumption rates, indicating the urgent need for further bacteriological assessments in this group. However, anthropogenic activities associated to land use changes and accelerated urban development has led to significant amounts of pollutant inputs, especially trace elements, into these environments. To assess land use changes due to accelerated urban development during the last decades the state of Rio de Janeiro, 15 surface sediment samples and one sediment core were analysed for the first time at Padre Lagoon, in southeastern Rio de Janeiro, with regard to radiocarbon dating, grain size, and pseudototal trace element concentrations. The age of the bottom core ranged from 470–314 cal yr BP determined by 14C radiocarbon dating. Pseudototal trace element concentrations were significantly different between the sediment core and the surface sediments and higher trace element levels were observed at the top of the sediment core and in all surface sediment samples, indicating high anthropogenic influence to the lagoon over time, both due to local urbanization and from atmospheric pollution from the industrial metropolitan region of the city of Rio de Janeiro. The current climate change scenario caused by anthropogenic activities has resulted in novel environmental pressures, increasing the occurrence and severity of fungal infections in the marine environment. Research on fungi in several taxonomic groups is widespread although not the case for elasmobranchs (sharks and rays). In this context, the aim of the present study was to screen the oral fungal microbiota present in artisanally captured Rioraja agassizii, a batoid that, although endangered, is highly fished and consumed worldwide. Oropharyngeal samples were obtained by swabbing and the samples were investigated using morphological and phenotypic methods by streaking on Sabouraud Dextrose Agar (SDA) and subculturing onto CHROMagar Candida (BD Difco) and CHROMagar Candida Plus (CHROMagarTM), as well as molecular techniques by amplification of the ITS1-5.8S-ITS2 ribosomal DNA region and a MALDI-TOF MS assessment. The findings indicated the presence of Candida parapsilosis (seven isolates), Candida duobushaemulonii (one isolate) and Rhodotorula mucilaginosa (three isolates), several of these reported for the first time in Rioraja agassizii. In addition, a 100% agreement between the MALDI-TOF results and partial ITS region sequencing was noted, demonstrating that the MALDI-TOF MS is a rapid and effective alternative for yeast identification in Rioraja agassizii isolates and potentially in other elasmobranch species. These findings highlight the need for further research to determine the potential impact on elasmobranch health, ecology, and commercial fisheries. Furthermore, this research is paramount in a One Health framework and may be employed to predict elasmobranch responses to an evolving ocean, keep healthy populations in check, monitor species, and assess the public health consequences of consuming these species. Simple Summary: There is a lack of detailed information on the capture pressures and records regarding the tiger shark Galeocerdo cuvier (Péron & Lesueur, 1822) (Carcharhinidae) for the state of Rio de Janeiro, Brazil. This study aimed to expand the tiger shark record database and to improve upon future conservation and management strategies in this area. A total of 23 new records were obtained, increasing the number of tiger shark records off the coast of the state of Rio de Janeiro approximately 5-fold. Possible seasonality patterns concerning tiger shark sizes were noted, indicating the need to consider the coast of Rio de Janeiro as an especially relevant area for at least part of the life history of tiger sharks. The tiger shark Galeocerdo cuvier (Péron & Lesueur, 1822) (Carcharhinidae) is classified as near-threatened along the Brazilian coast, in line with its global categorization. Although Rio de Janeiro, located in southeastern Brazil, is internationally identified as a priority shark conservation area, many shark species, including tiger sharks, are landed by both industrial and artisanal fisheries in this state. However, there is a lack of detailed information on the species capture pressures and records for the state of Rio de Janeiro. Subcellular metal distribution assessments are the most adequate biomonitoring approach to evaluate metal toxicity, instead of total metal assessments This study aimed to assess subcellular metal distributions and associations to the main metal exposure biomarker, metallothionein (MT), in two bromeliad species (Tillandsia usneoides and Tillandsia stricta) exposed established in industrial, urban, and port areas in the metropolitan region of Rio de Janeiro, southeastern Brazil, through an active biomonitoring approach conducted one year. Metals and metalloids in three subcellular fractions (insoluble, thermolabile and thermostable) obtained from the MT purification process were determined by inductively coupled plasma mass spectrometry (ICP-MS). Lower MT concentrations were observed both during the dry sampling periods, associated to the crassulacean acid metabolism (CAM) and during the COVID-19 pandemic, due to reduced urban mobility, decreasing pollutant emissions. The percentage of non-bioavailable metals detected in the insoluble fraction increased throughout the sampling period for both species. Several metals (Cr, Co, Cu, Cd, Mn, Ni, Se, and Zn), most associated with vehicle emissions, the main pollutant source in urban centers, were detected in the thermostable fraction and are, thus, associated with MT through the MT-metal detoxification route. Insoluble metal concentrations were higher in T. stricta , indicating that this species seems less susceptible to cellular metal exposure damage. Sponges are not routinely employed as metal bioindicators in Brazil. In this sense, this study reports baseline metal and metalloid concentrations, determined by inductively coupled plasma mass spectrometry, for two Demospongiae sponge species, Hymeniacidon heliophila and Desmapsamma anchorata , sampled from two Southeastern Brazil areas. Sponges from Ilha Grande Bay, an Environmental Protection Area, exhibited higher Al, As, Cd, Co, Cr, Fe, and Ni levels compared to Vermelha Beach, a metropolitan area in the Rio de Janeiro city. Several strong correlations were noted between elemental pairs, indicating common contamination sources and/or similar metabolic detoxification routes. Comparisons of the means determined herein for each study site to other reports indicate mostly lower Ag, As, Co, Cd, and Cu levels, while Al levels were higher than other studies, and Cr, Ni, and Fe were within reported ranges. These baseline data further knowledge on metal pollution in Desmspongiae members, which are still limited. Endocrine disrupting compounds (EDCs) can be found in domestic sewage, wastewater treatment plant effluents, natural water, rivers, lakes and in the marine environment. Jurujuba Sound, located in the state of Rio de Janeiro, Southeastern Brazil, receives untreated sewage into its waters, one the main sources of aquatic contamination in this area. In this context, the aim of the present study was to evaluate the estrogenic potential of water sampled from different depths and from areas with differential contamination levels throughout Jurujuba Sound. Water quality was evaluated and acute toxicity assays using Allviibrio fischeri were conducted, while estrogenic activity of the water samples was determined by a Yeast Estrogen Screening assay (YES). Water quality was mostly within the limits established for marine waters by the Brazilian legislation, with only DOC and ammoniacal nitrogen levels above the maximum permissible limits. No acute toxicity effects were observed in the Allivibrio fisheri assay. The YES assay detected moderate estrogenic activity in bottom water samples from 3 sampling stations, ranging from 0.5 to 3.2 ng L −1 , as well as in one surface water sample. Estrogenic activity was most frequently observed in samples from the bottom of the water column, indicating adsorption of estrogenic compounds to the sediment. [ABSTRACT FROM AUTHOR]

Numerous countries have been confronted with infectious diseases in mariculture activities, including fungi infections, although reports in scallops are scarce. Thus, this study aimed to investigate the occurrence of filamentous fungi in Nodipecten nodosus specimens from three marine farms in Southeastern Brazil. Eight fungi genera were observed in the branchial arches, intestine and muscle tissue of the scallop specimens. These include potentially toxin-producing species, such as Aspergillus , Penicillium and Fusarium . Their presence may lead to potential public health concerns, since all sampling sites showed the presence of fungi in all scallop organs, with special concern regarding edible muscle tissue. A significant number of species was observed at one of the study areas, which could indicate a previously unknown source of contamination, since increases in fungi species richness in polluted coastal waters have been reported. We evaluated concentrations of eight heavy metals Cr, Zn, Pb, Ni, Cu, Cd, Co and V, in tissues of representative macrofauna species from 68 sandy beaches from the coast of Rio de Janeiro state. The links between contamination levels and community descriptors such as diversity, evenness, density and biomass, were also investigated. Metal concentrations from macrofaunal tissues were compared to maximum permissible limits for human ingestion stipulated by the Brazilian regulatory agency (ANVISA). Generalized linear models (GLM's) were used to investigate the variability in macrofauna density, richness, eveness and biomass in the seven different regions. A non-metric multidimensional scaling analysis (n-MDS) was used to investigate the spatial pattern of heavy metal concentrations along the seven regions of Rio de Janeiro coast. Variation partitioning was applied to evaluate the variance in the community assemblage explained by the environmental variables and the heavy metal concentrations. Our data suggested high spatial variation in the concentration of heavy metals in macrofauna species from the beaches of Rio de Janeiro. This result highlighted a diffuse source of contamination along the coast. Most of the metals concentrations were under the limits established by ANVISA. The variability in community descriptors was related to morphodynamic variables, but not with metal contamination values, indicating the lack of direct relationships at the community level. This study presents a classification method combining logistic regression and fuzzy logic in the determination of sampling sites for feral fish, Nile Tilapia ( Tilapia rendalli). This method statistically analyzes the variable domains involved in the problem, by using a logistic regression model. This in turn generates the knowledge necessary to construct the rule base and fuzzy clusters of the fuzzy inference system (FIS) variables. The proposed hybrid method was validated using three fish stress indices; the Fulton Condition Factor (FCF) and the gonadossomatic and hepatossomatic indices (GSI and HSI, respectively), from fish sampled at 3 different locations in the Rio de Janeiro State. A multinomial logistic regression allowed for the FIS construction of the proposed method and both statistical approaches, when combined, complemented each other satisfactorily, allowing for the construction of an efficient classification method regarding feral fish sampling sites that, in turn, has great value regarding fish captures and fishery resource management. Total mercury (Hg) was determined in muscle tissue of 20 Guiana dolphins stranded along the coast of Rio de Janeiro state, Brazil, with a mean of 1.07 μg/g wet weight. Mercury concentrations were positively related to body length, possibly related to the capacity of the dolphins to bioaccumulate this element throughout life. The Hg concentrations were not significantly different between males and females, although females (1.08 μg/g) showed slightly higher levels than males (1.04 μg/g). Concentrations were low when compared to results of studies carried out with small cetaceans in the Northern Hemisphere, and with previous studies in the Southeastern Brazil. [ABSTRACT FROM AUTHOR]

Although shark meat is consumed worldwide, elevated arsenic (As) concentrations have been increasingly reported. The Caribbean sharpnose shark (Rhizoprionodon porosus) is a widely consumed fishing resource in Brazil, with scarce information on As burdens to date. Herein, commercial-sized juvenile Caribbean sharpnose sharks from Rio de Janeiro (Brazil) were assessed in this regard, presenting significantly higher hepatic As concentrations in males (8.24 ± 1.20 mg kg-1 wet weight; n = 12) compared to females (6.59 ± 1.87 mg kg-1 w.w.; n = 8), and a positive correlation (r = 0.74) was noted between female muscle and liver As concentrations, indicating systemic inter-tissue transport not evidenced in males. Arsenic concentrations were over the maximum permissible As limit established in Brazilian legislation for seafood and calculated Target Hazard Quotients suggest consumption risks, although cancer risks were not evidenced. Therefore, Public Health concerns with regard to Caribbean sharpnose shark As burdens deserve careful attention. • Arsenic burdens were determined in Rhizoprionodon porosus from Brazil. • Sex-dependent hepatic and systemic As transport was observed. • Muscle As concentrations were over the maximum permissible limit set in Brazil. • Calculated THQ indicates risks concerning shark consumption. • The calculated TCR was lower than the threshold for cancer development. This study comprises the first record of a juvenile Giant Devil Ray specimen for Rio de Janeiro, Southeastern Brazil, and its metal and metalloid contents. A scientometric assessment was also performed for the Manta and Mobula genera. Only five records were found, and only As, Cd, Pb, Hg, Pt, Pd and Rh have been assessed. All studies but one concerned human consumption. A significant knowledge gap on metal and metalloid ecotoxicology for mobulid rays is noted, indicating the emergence of a new field of research that th may be applied for wildlife conservation and management in response to anthropogenic contamination.
