This paper is the editorial for the virtual special issue (VSI) of Renewable and Sustainable Energy Reviews (RSER) dedicated to three Sustainable Development of Energy Water and Environment Systems (SDEWES) Conferences held in 2022, namely: (i) the 5th South East European Conference SEESDEWES 2022 held from May 22 to 26, 2022 in Vlore, Albania; (ii) the 3rd Latin American LASDEWES 2022 held from July 24 to 28, 2022 in Sao Paulo, Brazil and (iii) the 17th SDEWES 2022 held from November 6 to 10, 2022 in Paphos, Cyprus. The VSI collected both high quality review papers and original research articles presented at these conferences, aligning with the aims and scope of RSER. Following a rigorous peer review process, seventeen articles were accepted and published. These articles explore techniques and technologies essential for advancing feasible Renewable and Sustainable Energy Systems. Spanning multidisciplinary topics, many of them address almost all the Sustainable Development Goals, offering feasible solutions for much-needed decarbonization pathways. • The VSI SDEWES 2022 presents the ready solutions for Renewable and Sustainable Energy Systems. • Five clusters for presenting the Renewable and Sustainable energy cutting edge solutions. • The positioning in the scientific context of the Renewable and Sustainable Energy Systems. • Feasible solutions for much-needed decarbonization pathways. Brazil's Atlantic Forest (BAF) is a highly fragmented, strategic environmental and socio-economic region that represents the fourth biodiversity hotspot while also producing many commodities that are exported globally. Human disturbance plays a pivotal role as a driver of BAF's soil dynamics and behaviors. The soils under Late Primary and Secondary Semideciduous Seasonal Forests (LPSF and LSSF) were characterized by high to moderate resilience, with improved chemical properties as human disturbance decreased. The Transitional Forest to Cerrado (TFC) had the worst soil conditions. Disturbed Primary and Secondary Semideciduous Seasonal Forests (DPSF and DSSF) represent a transitional stage between LPSF/LSSF and TFC. Accordingly, SOCs stocks increased from TFC << DPSF, DSSF < LPSF, LSSF. In BAF soils, to avoid unreliable data, SOCs measurements should be (i) conducted to at least 1 m soil depth and (ii) quantified with a CHN analyzer. Human disturbance strongly affected the positive feedback between vegetation succession, SOCs, and soil nutrition. Soil development decreased as human disturbance increased, thus negatively affecting SOCs. Soils in the BAF require a long time to recover after the end of human disturbance, thus suggesting that preservation strategies should be prioritized in remnant BAF fragments. This paper investigates the mystery of baryogenesis for isotropic and anisotropic universe models in the context of Rastall's theory of gravity (RTG). In the first part, we utilize the Rastall field equations (FEs) for the flat Friedmann Robertson Walker (FRW) metric to find the solutions of density ρ and Hubble parameter H. Additionally, Rastall parameter plays a crucial role to discuss baryogenesis and fulfilling its criteria on the observational ground. It is found that very large positive/negative values of Rastall parameter like λ ≥ 1 0 5 4 (for ω = 0) and λ ≤ − 1 0 2 2 (for ω = 1) are required to satisfy the observational bounds on baryon asymmetry factor (BAF), see Fig. 1. In the second part, we develop the FEs for RTG using the Locally Rotationally Symmetric (LRS) Bianchi type-1 line-element. Then we find the solutions of the FEs and use them to find the BAF using Rastall parameter as λ ≤ − 1 0 3 7 and anisotropic parameter n ≥ 2 to meet the consistency with the experimental limit, see Fig. 3. For n = 1 , the anisotropic model reduces to the isotropic FRW model, where we have a zero BAF. Finally, we conclude that RTG with both universe models is a good candidate to study baryogenesis in the early universe and the variable λ has a considerable impact on the BAF. We present sub-50 fs soliton pulse generation from a diode-pumped Kerr-lens mode-locked laser based on an Yb3+-doped BaF2 crystal. Utilizing a spatially single-mode, fiber-coupled InGaAs laser diode at 976 nm as a pump source, the Yb:BaF2 laser generates pulses as short as 46 fs at 1060.1 nm with an average output power of 45 mW at a pulse repetition rate of ~65.6 MHz via soft-aperture Kerr-lens mode locking. To the best of our knowledge, this represents the first demonstration of Kerr-lens mode-locked operation of the Yb:BaF2 crystal, as well as the shortest pulse duration ever achieved from any diode-pumped mode-locked laser based on an Yb3+-doped alkaline-earth fluoride crystal. The SWI/SNF chromatin remodeling complex consists of more than ten component proteins that form a large protein complex of >1 MDa. The catalytic proteins Smarca4 or Smarca2 work in concert with the component proteins to form a chromatin platform suitable for transcriptional regulation. However, the mechanism by which each component protein works synergistically with the catalytic proteins remains largely unknown. Here, we report on the function of Smarce1, a component of the SWI/SNF complex, through the phenotypic analysis of homozygous mutant embryonic stem cells (ESCs). Disruption of Smarce1 induced the dissociation of other complex components from the SWI/SNF complex. Histone binding to DNA was loosened in homozygous mutant ESCs, indicating that disruption of Smarce1 decreased nucleosome stability. Sucrose gradient sedimentation analysis suggested that there was an ectopic genomic distribution of the SWI/SNF complex upon disruption of Smarce1, accounting for the misregulation of chromatin conformations. Unstable nucleosomes remained during ESC differentiation, impairing the heterochromatin formation that is characteristic of the differentiation process. These results suggest that Smarce1 guides the SWI/SNF complex to the appropriate genomic regions to generate chromatin structures adequate for transcriptional regulation. [ABSTRACT FROM AUTHOR]

Lead telluride and germanium telluride are well-known IV-VI semiconductors, which is now the focus of research due to the perspective of application as thermoelectrics for midrange temperatures. Solid solutions and heterostructures on this basis, obtained by molecular beam epitaxy, are a promising direction for the development of these materials. In this paper, we have focused on the Raman spectra excited by the 514.5 nm laser line (out of resonance) of PbTe, GeTe, (Pb, Ge)Te, and (Pb, Ge, Eu)Te layers grown on BaF2 (111) monocrystalline substrates. The obtained phonon properties are related to the properties of the corresponding bulk materials or can be explained by a model that takes into account the difference in the masses of the constituent elements only, as is the case with the local mode of Ge in PbTe (registered at about 181 cm−1). Multiphonon processes registered for this phonon are a consequence of the change in the electronic structure of PbTe and electron-phonon interaction. In this paper, Er3+ doped TeO 2 -ZnF 2 -BaF 2 -KF-Ta 2 O 5 tellurite glasses with low hydroxyl content (∼0.03 × 10−19 cm−3) were investigated employing both glass composition and glass melting process optimization. The Raman spectra and physical properties were characterized to analyze the structure of the glasses. Under the pumping of 980 nm LD laser, intense up-conversion fluorescence at 1.5 and 2.7 μm of samples and their lifetimes were detected and analyzed, and the related transition mechanisms with gradient-varying Er3+ doping concentrations were discussed. The maximum absorption and emission cross section at 2.7 μm was calculated to be 6.4 × 10−21 cm2 and 6.8 × 10−21 cm2, correspondingly, which were higher than those of traditional tellurite glasses. Using the calculated and measured spectroscopic parameters of bulk tellurite glass, a dual-wavelength pumping model was established to verify the feasibility of mid-infrared laser output in similar tellurite glass fiber. Experimental results support the assertion that the Er3+ doped tellurite glasses hold promise as a candidate laser gain medium for mid-infrared fiber laser systems. [ABSTRACT FROM AUTHOR]

Prior to the next generation sequencing and characterization of the tumor genome landscape, mutations in the SWI/SNF chromatin remodeling complex and the KEAP1-NRF2 signaling pathway were underappreciated. While these two classes of mutations appeared to independently contribute to tumor development, recent reports have demonstrated a mechanistic link between these two regulatory mechanisms in specific cancer types and cell models. In this work, we expand upon these data by exploring the relationship between mutations in BAF and PBAF subunits of the SWI/SNF complex and activation of NRF2 signal transduction across many cancer types. ARID1A/B mutations were strongly associated with NRF2 transcriptional activity in head and neck squamous carcinomas (HNSC). Many additional tumor types showed significant association between NRF2 signaling and mutation of specific components of the SWI/SNF complex. Different effects of BAF and PBAF mutations on the polarity of NRF2 signaling were observed. Overall, our results support a context-dependent functional link between SWI/SNF and NRF2 mutations across human cancers and implicate ARID1A inactivation in HPV-negative HNSC in promoting tumor progression and survival through activation of the KEAP1-NRF2 signaling pathway. The tumor-specific effects of these mutations open a new area of study for how mutations in the KEAP1-NRF2 pathway and the SWI/SNF complex contribute to cancer. [ABSTRACT FROM AUTHOR]

A bubble-based acoustofluidic filtration (BAF) microfluidic device, which employs cross-flow filtration (CFF) and acoustic streaming, separates cells with high efficiency for forensic analysis. Forensic samples are typically complex and contain a substantial number of squamous epithelial cells from the female vagina, which tend to have fouling problems during filtration due to their morphological and cell adhesion differences. To overcome this issue, the BAF device utilizes bubble oscillation by bulk acoustic wave (BAW) to generate acoustic streaming, which offers additional hydrodynamic forces for side flushing cleaning and achieves effective removal within a mere 0.5 seconds. Our device is tested with imbalanced cell mixtures of sperm and epithelial cells with large disparity ratios. By concurrently employing CFF and acoustic streaming, the samples with our sperm-enrichment can achieve 91.72–97.78% for the recovery rate and 74.58–89.26% for the purity in the sperm enrichment. They are further subjected to short tandem repeat (STR) profiling, enabling the identification of perpetrators. Notably, even samples with minimal sperm cells demonstrated a significant increase in the male donor DNA ratio, while the peak heights of female alleles became virtually undetectable. With the arrival of the space economy, the miniaturization of spacecraft and their multi-functional trends have put a greater demand on space thermal control systems. Smart radiation devices (SRDs) are gaining attention as a low-cost, light-weight material for the thermal control of spacecraft. Previous works have focused on how to enhance IR emittance tunability of SRDs. In fact, the SRDs performance and lifetime may be affected by the external space environment. Thus, researchers proposed to cover the SRDs with a protective layer to resist oxidation and other problems. However, the influence of protective layer on the performance of SRDs, including IR emittance tunability Δ ε and solar absorption α s , has not been studied systematically yet. In this paper, the impact of protective layer on the performance of SRDs is comprehensively evaluated in theory. Firstly, a three-layer SRD is composed of a vanadium dioxide (VO 2) film, BaF 2 , and a silver (Ag) substrate, which is optimized using the transfer matrix method. The IR emittance tunability and solar absorption can reach 0.64 and 0.29, respectively. Secondly, the smart thermal control device with three protection materials (BaF 2 , Al 2 O 3 and Si) is investigated. The results show that the performance of SRD can be enhanced or deteriorated, depending on the types of protection materials and the thickness. When the protection material is Si, the IR emittance tunability can be enhanced to 0.65, while the solar absorption can be decreased to 0.18. The performance of the proposed SRD meets the requirement of the spacecraft thermal control devices. This paper presents an overview for the Special Issue of Clean Technology and Environmental Policy journal (CTEP), and it includes accepted papers from: 5th South East European Conferences on Sustainable Development of Energy, Water and Environment Systems (SDEWES) held from May 22–26, 2022, in Vlore, Albania. 3rd Latin American Conferences on Sustainable Development of Energy, Water and Environment Systems (SDEWES) held from July 24–28, 2022, in Sao Paulo, Brazil. 17th Conferences on Sustainable Development of Energy, Water and Environment Systems (SDEWES) held from November 6–10, 2022, in Paphos, Cyprus. Considering CTEP's policy of high-quality research papers, guest editors have invited 35 research articles, presented at the SDEWES 2022 conference. After a vigorous review process, 13 papers have been accepted for publication in this special issue. All 13 accepted papers are briefly presented in this overview together with a wider view that presents research efforts within the SDEWES community published through previous SDEWES special issues. Purpose: Acute lung injury (ALI) is a severe pulmonary disease characterized by damage to the alveoli and pulmonary blood vessels, leading to severe impairment of lung function. Studies on the effect of capsaicin (8-methyl-N-geranyl-6-nonamide, CAP) on lipopolysaccharide (LPS)-induced ALI in bronchial epithelial cells transformed with Ad12-SV40 2B (BEAS-2B) are still limited. This study aimed to investigate the effect and specific mechanism by which CAP improves LPS-induced ALI.Methods: The present study investigated the effect of CAP and the potential underlying mechanisms in LPS-induced ALI in vitro and vivo via RNA sequencing, Western blotting (WB), quantitative real-time reverse transcription PCR (qRT‒PCR), enzyme-linked immunosorbent assay (ELISA), and transmission electron microscopy (TEM). The TRPV1 inhibitor AMG9810 and the AKT agonist SC79 were used to confirm the protective effect of the TRPV1/AKT axis against ALI. The autophagy agonist rapamycin (Rapa) and the autophagy inhibitors 3-methyladenine (3-MA) and bafilomycin A1 (Baf-A1) were used to clarify the characteristics of LPS-induced autophagy.Results: Our findings demonstrated that CAP effectively suppressed inflammation and autophagy in LPS-induced ALI, both in vivo and in vitro. This mechanism involves regulation by the TRPV1/AKT signaling pathway. By activating TRPV1, CAP reduces the expression of P-AKT, thereby exerting its anti-inflammatory and inhibitory effects on pro-death autophagy. Furthermore, prior administration of CAP provided substantial protection to mice against ALI induced by LPS, reduced the lung wet/dry ratio, decreased proinflammatory cytokine expression, and downregulated LC3 expression.Conclusion: Taken together, our results indicate that CAP protects against LPS-induced ALI by inhibiting inflammatory responses and autophagic death through the TRPV1/AKT signaling pathway, presenting a novel strategy for ALI therapy. ATP-dependent chromatin remodeling SWI/SNF complexes exist in three subcomplexes: canonical BAF (cBAF), polybromo BAF (PBAF), and a newly described non-canonical BAF (ncBAF). While cBAF and PBAF regulate fates of multiple cell types, roles for ncBAF in hematopoietic stem cells (HSCs) have not been investigated. Motivated by recent discovery of disrupted expression of BRD9, an essential component of ncBAF, in multiple cancers, including clonal hematopoietic disorders, we evaluate here the role of BRD9 in normal and malignant HSCs. BRD9 loss enhances chromatin accessibility, promoting myeloid lineage skewing while impairing B cell development. BRD9 significantly colocalizes with CTCF, whose chromatin recruitment is augmented by BRD9 loss, leading to altered chromatin state and expression of myeloid-related genes within intact topologically associating domains. These data uncover ncBAF as critical for cell fate specification in HSCs via three-dimensional regulation of gene expression and illuminate roles for ncBAF in normal and malignant hematopoiesis. BRD9 is a core non-canonical BAF component. Here the authors show that BRD9 plays a pivotal role in regulating the disease-related cell fate of hematopoietic stem cells. The current applications in the energy sector are based largely on fossil fuels which release greenhouse gas emissions to the atmosphere. To face the issue of global warming, the energy sector has to transfer to and develop sustainable energy solutions that do not release carbon emissions. This is one of the primary motivators for the SDEWES conference as well as for this review, and previous ones, examining the most recent works based on sustainable and green energy production in such fields. The 17th Conference on the Sustainable Development of Energy, Water, and Environment Systems (SDEWES) was held on 6–10 November 2022 in Paphos, Cyprus. The SDEWES conference aims at solving complex and ongoing concerns that approach a long-term perspective and supporting innovative solutions and continuous monitoring and evaluation. This review paper aims at collecting the main presented papers focused on the following hot topics: low-carbon technologies based on renewable and clean-energy systems, including mainly biomass, solar, and wind energy applications; energy storage systems; hydrogen-based systems; energy-saving strategies in buildings; and the adoption of smart management strategies using renewable energy systems. These topics are investigated in order to propose solutions to address the issues of climate change, water scarcity, and energy saving. From the analyzed works, we note that some key issues for sustainable development remain to be further addressed: such as novel and advanced energy storage systems, green hydrogen production, novel low-temperature district heating and cooling networks, novel solar technologies for the simultaneous production of power and high temperature heat, solar desalination for hydrogen production systems, and agrivoltaic systems for the production of power and food. Ever since 2002, when the first Conference on Sustainable Development of Energy, Water and Environment Systems (SDEWES) was held in Dubrovnik, the SDEWES conferences series has been providing a forum for world-wide scientists and those interested in learning about the sustainability of development, to present research progress and to discuss the state of the art, the future directions and priorities in the various areas of sustainable development. One of the main issues of the coming decades is to improve efficiencies by integrating various energy systems, using excess from one, as resource in another in the correct moment. Integrating electricity, heating, cooling, transport, water, buildings, waste, wastewater, industry, forestry and agriculture systems will be pivotal towards sustainable development. SDEWES has maintained high publishing standard as 2408 SDEWES papers were published in special issues of leading scientific journals including "Energy - The International Journal". In 2022, the 17th SDEWES Conference was held during November 6–10 in Paphos, Cyprus, the 5th SEE Conference took place in May 22–26, 2022 in Vlorë, Albania, the 3rd Latin American Conference was held through a virtual platform during July 24–28, 2022. The Energy journal has continued its cooperation with SDEWES launching a special issue dedicated to the 2022 Conferences. The special issue includes papers which traditionally cover a range of energy issues - higher renewables penetration and various technologies and fuels assessments at energy supply side, as well as, energy efficiency in various sectors, buildings, district heating, electric vehicles and demand modelling at energy demand side. This paper presents the results of ceramic synthesis in the field of a powerful flux of high-energy electrons on powder mixtures. The synthesis is carried out via the direct exposure of the radiation flux to a mixture with high speed (up to 10 g/s) and efficiency without the use of any methods or means for stimulation. These synthesis qualities provide the opportunity to optimize compositions and conditions in a short time while maintaining the purity of the ceramics. The possibility of synthesizing ceramics from powders of metal oxides and fluorides (MgF2, BaF2, WO3, Ga2O3, Al2O3, Y2O3, ZrO2, MgO) and complex compounds from their stoichiometric mixtures (Y3Al3O12, Y3AlxGa(5−x) O12, MgAl2O4, ZnAl2O4, MgWO4, ZnWO4, BaxMg(2−x) F4), including activators, is demonstrated. The ceramics synthesized in the field of high-energy electron flux have a structure and luminescence properties similar to those obtained by other methods, such as thermal methods. The results of studying the processes of energy transfer of the electron beam mixture, quantitative assessments of the distribution of absorbed energy, and the dissipation of this energy are presented. The optimal conditions for beam treatment of the mixture during synthesis are determined. It is shown that the efficiency of radiation synthesis of ceramics depends on the particle dispersion of the initial powders. Powders with particle sizes of 1–10 µm, uniform for the synthesis of ceramics of complex compositions, are optimal. A hypothesis is put forward that ionization processes, resulting in the radiolysis of particles and the exchange of elements in the ion–electron plasma, dominate in the formation of new structural phases during radiation synthesis. Malignant rhabdoid tumor (MRT) is a highly malignant and often lethal childhood cancer. MRTs are genetically defined by bi-allelic inactivating mutations in SMARCB1, a member of the BRG1/BRM-associated factors (BAF) chromatin remodeling complex. Mutations in BAF complex members are common in human cancer, yet their contribution to tumorigenesis remains in many cases poorly understood. Here, we study derailed regulatory landscapes as a consequence of SMARCB1 loss in the context of MRT. Our multi-omics approach on patient-derived MRT organoids reveals a dramatic reshaping of the regulatory landscape upon SMARCB1 reconstitution. Chromosome conformation capture experiments subsequently reveal patient-specific looping of distal enhancer regions with the promoter of the MYC oncogene. This intertumoral heterogeneity in MYC enhancer utilization is also present in patient MRT tissues as shown by combined single-cell RNA-seq and ATAC-seq. We show that loss of SMARCB1 activates patient-specific epigenetic reprogramming underlying MRT tumorigenesis. The regulatory landscape of malignant rhabdoid tumor (MRT) due to SMARCB1 loss remains to be explored. Group 3 medulloblastoma is one of the most aggressive types of childhood brain tumors. Roughly 30% of cases carry genetic alterations in MYC, SMARCA4, or both genes combined. While overexpression of MYC has previously been shown to drive medulloblastoma formation in mice, the functional significance of SMARCA4 mutations and their suitability as a therapeutic target remain largely unclear. To address this issue, we combined overexpression of MYC with a loss of SMARCA4 in granule cell precursors. Both alterations did not increase proliferation of granule cell precursors in vitro. However, combined MYC overexpression and SMARCA4 loss successfully induced tumor formation in vivo after orthotopic transplantation in recipient mice. Resulting tumors displayed anaplastic histology and exclusively consisted of SMARCA4-negative cells although a mixture of recombined and non-recombined cells was injected. These observations provide first evidence for a tumor-promoting role of a SMARCA4 deficiency in the development of medulloblastoma. In comparing the transcriptome of tumors to the cells of origin and an established Sonic Hedgehog medulloblastoma model, we gathered first hints on deregulated gene expression that could be specifically involved in SMARCA4/MYC driven tumorigenesis. Finally, an integration of RNA sequencing and DNA methylation data of murine tumors with human samples revealed a high resemblance to human Group 3 medulloblastoma on the molecular level. Altogether, the development of SMARCA4-deficient medulloblastomas in mice paves the way to deciphering the role of frequently occurring SMARCA4 alterations in Group 3 medulloblastoma with the perspective to explore targeted therapeutic options. This paper concerns the Hellenistic and Early Roman cooking pottery originating from the area of Nea Paphos, Cyprus. Macroscopic examinations conducted on a few thousand local vessels and their fragments provided the detailed characterization of this group of pottery in terms of the appearance of fabric, typology, and chronology. The integration of these data with the results of laboratory analysis of selected samples revealed that the local production of cooking pottery changed through time in terms of clay composition, shapes, surface treatment, and firing techniques. The transformations observed within the local cooking pottery manufacture seem to correlate with political and social changes as well as the development of Nea Paphos. [ABSTRACT FROM AUTHOR]

A series of CoCrFeNiMn high-entropy alloy matrix self-lubricating composites were prepared by spark plasma sintering. The composites are composed of an FCC phase, Cr7C3, Ag, and eutectic fluoride BaF2/CaF2 phases. The microstructure of the composites is uniform. The additional phases distribute along the boundary of equiaxed grains of the FCC phase. The compressive yield strength and fracture toughness decrease with the increase of eutectic fluoride BaF2/CaF2. The composites are susceptible to brittle cleavage fracture. [ABSTRACT FROM AUTHOR]

Biological aerated filters (BAF) are attached growth aerobic systems used for the treatment of wastewater. This system can establish the physical filtration of solids along with the biological decomposition of organic matter. The performance of the BAF depends on the type of support media and its ability to meet objective of the treatment process. Natural zeolites, with properties such as ion exchange, larger surface area, capability to form biofilm, adsorption, regenerative properties etc., are a promising filter material in BAF systems. This paper investigates and discusses the effect of various factors like hydraulic retention time (HRT), carbon to nitrogen (C/N) ratio, aeration rate, depth of filter and particle size of natural zeolite which influence the performance of BAF. Based on the experimental results and optimization study using response surface methodology, carried out on the BAF system with varying process variables like HRT, C/N ratio, aeration rate and depth of filter, the natural zeolite media with particle size 1-3 mm range showed improved performance as compared to the coarser particles, that is, 3-5 mm and 5-10 mm. On conducting a final run using natural zeolite particle size of 1-3 mm under optimized value of HRT = 3.25 h, C/N = 6.2, aeration rate = 69 L/h, depth of filter = 83 cm, the BAF system observed chemical oxygen demand, biological oxygen demand, NH3-N, turbidity and total dissolved solids removal percentage of 97%, 96%, 99%, 98% and 58%, respectively. TeO2-BaF2-Er2O3-Dy2O3 laser glasses were prepared using the melt-quenching method. The bound water that can capture the excited state energy was reduced by physical and chemical methods. We did not observe a significant Er3+ emission peak at 2.7 μm in fluorescence spectra, which may be due to the efficient energy transfer process (ET2). Meanwhile, we found a broadband gain span of approximately 400 nm in fluorescence spectra at the 2.85 μm band, attributed to the 'vector summation' of the energy level radiation transition and the change of the glass network. Subsequently, we explored the structural properties of the glass. The results indicated that the Gaussian peak located at 250 cm−1 drifts toward 370 cm−1, which may be caused by the fracture or recombination of Te-O-Te and a decrease in the bridge oxygen content with the increasing concentration of Er2O3. The topology cage structure around the luminescence center of rare earth ions is changed and the stability of the optically active center is enhanced, finally contributing to the enhancement of luminescence. Meanwhile, the maximum σemi and gain coefficient of Dy3+ reach up to 7.22 × 10−21 cm2 and 7.37 cm−1, respectively. The comprehensive results show that the fluorotellurite glass designed in this study is expected to be a gain medium for mid-infrared lasers in remote sensing monitoring, military, and other fields. Alternative techniques have been investigated for effectiveness in caries removal because conventional metallic dental burs can lead to an excessive loss of sound tissue. The aim of the present study is to realize a preliminary approach in obtaining effective polymer mixtures for polymeric bur development, capable of removing primary dental caries using combinations of polymers to ensure the requirements for such instruments, but also a greater compatibility with the teeth structure. This study assessed the main mechanical properties, water sorption, solubility and microscopic structure of four new polymer mixture recipes to provide essential features in obtaining experimental self-limited dental burs. Two mixtures have in their composition polymer mixtures of Bis-phenol A diglycidyl ether dimethacrylate/Triethylene glycol dimethacrylate/Urethane dimethacrylates (R1, R2), and two other mixtures have Bis-phenol A diglycidyl ether dimethacrylate/Polymethyl methacrylate/Methyl methacrylates (R3, R4). The incorporation of nanoparticles into the polymer matrix has become essential due to the need of polymer biocompatibility increasing along with teeth surface remineralization, so that the powder charge was added to four recipes, such as 5% glass with BaF2 and 0.5% graphene with silver particles. All data sets were analyzed using the One-Way ANOVA test. R3, R4 showed higher compressive strength and diametrical compression values; these values increased when glass and graphene were added. Moreover, the addition of glass particles lead to an increase in flexural strength. Regarding the sorption, sample R3 had the most significant differences between day 69 and the rest of the investigation days, while the solubility varied at different intervals. From the mechanical evaluation, we could conclude that the Bis-GMA/PMMA/MMA mixtures fit the mechanical characteristics supported by polymer burs, following future studies regarding their use on the affected dentin. Background: Disrupted intestinal epithelial barrier is one of the major causes of Crohn's disease (CD). Novel molecular targets for intestinal epithelial barrier are essential to treatment of CD. Transmembrane and immunoglobulin domain-containing protein 1 (TMIGD1) is an adhesion molecule that regulates cell adhesion, migration, and enterocyte differentiation. However, the function and mechanism of TMIGD1 in CD and intestinal epithelial barrier has rarely been studied. Furthermore, the association between TMIGD1 and the clinical features of CD remains unclear. Methods: Transcriptome analysis on colonic mucosa from CD patients and healthy individuals were performed to identify dysregulated genes. Multi-omics integration of the 1000IBD cohort including genomics, transcriptomics of intestinal biopsies, and serum proteomics identified the association between genes and characteristics of CD. Inflammation was assessed by cytokine production in cell lines, organoids and intestinal-specific Tmigd1 knockout (Tmigd1INT-KO) mice. Epithelial barrier integrity was evaluated by trans-epithelium electrical resistance (TEER), paracellular permeability, and apical junction complex (AJC) expression. Co-immunoprecipitation, GST pull-down assays, mass spectrometry, proteomics, and transcriptome analysis were used to explore downstream mechanisms. Results: Multi-omics integration suggested that TMIGD1 was negatively associated with inflammatory characteristics of CD. TMIGD1 was downregulated in inflamed intestinal mucosa of patients with CD and mice colitis models. Tmigd1INT-KO mice were more susceptible to chemically induced colitis. In epithelial cell lines and colonic organoids, TMIGD1 knockdown caused impaired intestinal barrier integrity evidenced by increased paracellular permeability and reduced TEER and AJC expression. TMIGD1 knockdown in intestinal epithelial cells also induced pro-inflammatory cytokine production. Mechanistically, TMIGD1 directly interacted with cytoplasmic BAF nuclear assembly factor 1 (BANF1) to inhibit NF-κB activation. Exogenous expression of TMIGD1 and BANF1 restored intestinal barrier function and inhibited inflammation in vitro and in vivo. TMIGD1 expression predicted response to anti-TNF treatment in patients with CD. Fluorine (F) plays an important role in biology and geology but is hard to analyse and quantify using element-specific techniques. This is related to its high first ionisation potential and analytical methods depending on high energy sources for excitation and/or ionisation of F. In particular ICP-MS was initially found incapable of detecting F; however, recent methodological advances, i.e., the application of Ba as a plasma modifier and the application of tandem mass spectrometry, enabled its analysis by targeting BaF+. In this study, we suggest this approach in conjunction with laser ablation to perform the mapping of F in both biological and geological samples. In a proof of concept, tooth samples as biological samples and a Rhynie chert as a geological sample were analysed. For method development and to evaluate the performance of the developed method, gelatine-based F-standards were prepared and characterised using combustion ion chromatography. Standards were further interrogated to estimate LODs and LOQs. Depending on the required spatial resolution, figures of merit in the upper ng g−1 range and lower μg g−1 range were achievable. This is the first example of F mapping using LA-ICP-MS instrumentation and the developed methods close an important analytical gap by enabling the spatially resolved F analysis at relevant biological and geological concentrations. Simple Summary: The generation of an artificial intestine could minimize the use of in vivo trials, which are presently necessary for testing aquafeeds. As well as being lengthy and expensive, they also require the use of many animals. Furthermore, although in vivo screening allows for a precise evaluation of the final effect of each feed, it does not improve our basic knowledge of the cellular and molecular mechanisms that determine the end results. This, in turn, severely limits our ability to understand and predict the biological value of ingredients, both individually and in differing combinations thereof. To circumvent these constraints, we previously derived and characterized two novel cell lines from the proximal (RTpi-MI) and distal intestine (RTdi-MI) of rainbow trout (Oncorhynchus mykiss), demonstrating that the two cell lines retain the main properties of the intestinal tracts from which they originate. In the present work, we used these cells to set up a functional intestinal barrier on a permeable membrane support to investigate the digestion and absorption of dietary amino acids in fish feed. The results demonstrate that the procedure we developed enables the quantification of individual amino acids that cross the intestinal wall. This new platform will now be used to screen and select new feed ingredients/formulations. The ever-increasing number and variation of raw materials utilized to provide alternative feed formulations continues to allow for a more sustainable and flexible approach. Testing all these options in vivo is still the most robust and reliable manner to pick the best raw material candidates, but it requires the use of large numbers of animals and is time-consuming and expensive. Therefore, we are developing an in vitro platform that can provide a reliable evaluation of new ingredients. The main aim of this work was to combine an in vitro digestion protocol of extruded, commercially relevant aquafeeds with the exposure of intestinal epithelial cells to the extracted bio-available fraction (BAF). The results show that 250,000 cells/cm2 represents the optimal seeding density and that up to 50% BAF concentration for up to 24 h had no negative effects on the epithelial barrier morphology and function. It is possible to determine amino acid digestibility and bioavailability in all the experimental conditions (with and without BSA, at 25% and 50% dilution) and at all time points (0, 6, and 24 h). However, BAF concentration, the medium used for its dilution, and the length of exposure to the different epithelial cell lines can all influence the results and, therefore, must be selected according to the final aim of the experiment. Heavy metals (HMs) have the characteristics of high toxicity, long residual time, and difficult degradation, which pose a potential threat to aquatic ecosystems. The distribution and migration of HMs in different media can reflect their potential impact on aquatic ecosystem health. In this study, we analyzed the distribution characteristics of seven HMs (Zn, Cu, Cr, Ni, Hg, As, and Pb) in the water and sediment and five groups of organisms (fish, Crustacean, cephalopod, bivalvia, and gastropoda) in the sea ranching area of Haizhou Bay and explored the regularity of HM accumulation from water and sediments to organisms. The results showed that in the water, Zn and Hg had the smallest concentrations in the nearshore area, Cr had the lowest concentrations in the offshore area, and As, Cu, Pb, and Ni had the lowest concentrations in the central area. In the sediment, Hg had the smallest concentrations in the nearshore area and As, Pb, Cr, Cu, Ni, and Zn had the lowest concentrations in the offshore area; the average contents of seven heavy metals all meet the Class I standard of sediments in China. The concentrations of Cu and Zn in crustaceans were significantly higher than those in fishes (p < 0.05), and As showed a higher enrichment effect in cephalopods. Under the influence of feeding habits and habitat environment, the BAF value of benthic crustaceans and bivalvia accumulated HMs from sediments is high, and the BAF value of fish accumulated HMs from water and sediments is low. Overall, the concentration of HMs in water is high, and HMs in sediments are in good condition, crustaceans and bivalviaa have a strong ability to accumulate HMs from water and sediments, while fish are weak. WRKY transcription factors (TFs) play an important role in regulating plant growth and responses to environmental stress. However, the molecular mechanism of WRKY to cadmium (Cd) stress is unclear, which prevents phytoremediation of Cd-contaminated soil from widely application. To determine the underlying mechanism, PyWRKY48 -overexpressing poplars were obtained (OE-32 and OE-67) to study the Cd tolerance and accumulation in poplars. Results showed that the Cd content in the aboveground part of the two transgenic poplar lines were 1.57 and 1.99 times higher than that of wild type (WT), and lateral roots, GSH, PCs content and GST activity increased significantly. RNA-seq. data about transgenic and WT poplars revealed that 2074 differentially expressed genes (DEGs) in roots, 4325 in leaves, and 499 in both tissues. And these DEGs were mainly concentrated in ABC transport protein (PaABC), heavy-metal binding protein (PaHIPP), and transportation and loading of xylem (PaNPF , PaBSP) proteins, and they enhanced Cd accumulation. Meanwhile, PyWRKY48 increased the Cd tolerance of transgenic poplars by up-regulating the expression of PaGRP , PaPER and PaPHOS , which encode cell wall proteins, antioxidant enzyme, and heavy metal-associated proteins, respectively. In addition, overexpression PyWRKY48 promoted poplar growth by increasing the chlorophyll a nd carotenoid content. This study generated PyWRKY48 -overexpressing poplars and functionally verified them in Cd-contaminated soil, to analyze the effects of the gene on poplar growth, Cd tolerance and Cd accumulation. RNA seq. data revealed that several genes are involved in Cd exposure. This may provide a strong molecular basis and new ideas for improving the phytoremediation efficiency of Cd-contaminated soils. Growing concerns over environmental issues and sustainable living have resulted in increased interest in renewable energy and energy efficiency. The real estate market is no exception, with homeowners increasingly considering the market value of green and sustainable buildings, which can offer both energy efficiency and potential health benefits. This study investigates the level of interest among homeowners in investing in renewable energy sources and energy efficiency measures for their homes and how it relates to their perception of the market value of green or sustainable buildings in the real estate market. A survey was conducted in the Paphos urban complex in Cyprus, with 180 participants over the age of 18. The participants were selected through a random sampling method and were representative of the general population in terms of gender, age, and income. Data were collected on their attitudes towards renewable energy sources and energy efficiency, as well as their perceptions of the market value of green buildings. The data collected were analyzed using various statistical methods, including Cronbach's α coefficient, the non-parametric Friedman test, descriptive statistics, and factor analysis, with the Statistical Package for the Social Sciences (SPSS) being used for coding and analysis. Results indicate that 64% of the homeowners surveyed were interested in investing in renewable energy sources, and 72% were interested in energy efficiency measures. Additionally, findings suggest a moderate level of interest (58%) among homeowners in investing in renewable energy sources and that this is positively associated with their perception of the market value of green buildings. Furthermore, homeowners with higher income and education levels tend to be more interested in investing in renewable energy sources and energy efficiency measures and perceive green buildings as having higher market value. This study provides insights into the factors that drive homeowners' investment in renewable energy sources and energy efficiency measures, shedding light on the relationship between homeowners' perceptions of the market value of green buildings and their interest in such investments. Perfluorooctanoic acid (PFOA) is a representative persistent organic pollutant and its disposal by mechanochemical (MC) technology emerges in recent years. However, degradation mechanism of PFOA especially rupture of C−F bonds during MC process is still unclear. Therefore, we innovatively employed barium titanate as co-milling reagent in MC system to disclose an electron-dominated reduction process. By stimulating piezoelectric effect of BaTiO 3 under MC impact, free electrons were generated. The results implied more than 95.00% degradation and 60.00% defluorination efficiency were obtained after 6 h′ ball milling. DPPH• was used as probe to confirm the existence of piezo-excited electrons, which were further verified to be major reactive species by atmosphere experiments. Thus, PFOA destruction was dominated by reduction process, characterizing by breakage of C−F bonds induced by electrons. Accordingly, the fate of organic fluorides was explored and BaF 2 was identified as final product. The cleavage of carboxyl group initiated PFOA decomposition, following by successive removal of CF 2 groups and elimination of F−. Moreover, the practical experiments and reusable trials implied promising application of this method. Diffuse midline gliomas (DMGs) including diffuse intrinsic pontine gliomas (DIPGs) bearing lysine-to-methionine mutations in histone H3 at lysine 27 (H3K27M) are lethal childhood brain cancers. These tumors harbor a global reduction in the transcriptional repressive mark H3K27me3 accompanied by an increase in the transcriptional activation mark H3K27ac. We postulated that H3K27M mutations, in addition to altering H3K27 modifications, reprogram the master chromatin remodeling switch/sucrose nonfermentable (SWI/SNF) complex. The SWI/SNF complex can exist in two main forms termed BAF and PBAF that play central roles in neurodevelopment and cancer. Moreover, BAF antagonizes PRC2, the main enzyme catalyzing H3K27me3. We demonstrate that H3K27M gliomas show increased protein levels of the SWI/SNF complex ATPase subunits SMARCA4 and SMARCA2, and the PBAF component PBRM1. Additionally, knockdown of mutant H3K27M lowered SMARCA4 protein levels. The proteolysis targeting chimera (PROTAC) AU-15330 that simultaneously targets SMARCA4, SMARCA2, and PBRM1 for degradation exhibits cytotoxicity in H3.3K27M but not H3 wild-type cells. AU-15330 lowered chromatin accessibility measured by ATAC-Seq at nonpromoter regions and reduced global H3K27ac levels. Integrated analysis of gene expression, proteomics, and chromatin accessibility in AU-15330-treated cells demonstrated reduction in the levels of FOXO1, a key member of the forkhead family of transcription factors. The altered lysosomal function can induce drug redistribution which leads to drug resistance and poor prognosis for cancer patients. V-ATPase, an ATP-driven proton pump positioned at lysosomal surfaces, is responsible for maintaining the stability of lysosome. Herein, we reported that the potassium voltage-gated channel subfamily J member 15 (KCNJ15) protein, which may bind to V-ATPase, can regulate the function of lysosome. The deficiency of KCNJ15 protein in breast cancer cells led to drug aggregation as well as reduction of drug efficacy. The application of the V-ATPase inhibitor could inhibit the binding between KCNJ15 and V-ATPase, contributing to the amelioration of drug resistance. Clinical data analysis revealed that KCNJ15 deficiency was associated with higher histological grading, advanced stages, more metastases of lymph nodes, and shorter disease free survival of patients with breast cancer. KCNJ15 expression level is positively correlated with a high response rate after receiving neoadjuvant chemotherapy. Moreover, we revealed that the small molecule drug CMA/BAF can reverse drug resistance by disrupting the interaction between KCNJ15 and lysosomes. We have optimized the growth conditions of the buffer layer for further deposition of Pb1–xSnxTe (x ≥ 0.4), which has the properties of a crystalline topological insulator. To this end, a three-component heterostructure consisting of CaF2, BaF2, and Pb0.7Sn0.3Te:In layers was formed and optimized on the Si(111) surface. The surface morphology of this structure was studied depending on the temperature growth regimes and the optimal combination of growth parameters was selected from the point of view of smoothness and crystalline quality. Cancer development is a complex process involving both genetic and epigenetic changes. The SWI/SNF (switch/sucrose non‐fermentable) chromatin remodelling complex, one of the most studied ATP‐dependent complexes, plays an important role in coordinating chromatin structural stability, gene expression and post‐translational modifications. The SWI/SNF complex can be classified into BAF, PBAF and GBAF according to their constituent subunits. Cancer genome sequencing studies have shown a high incidence of mutations in genes encoding subunits of the SWI/SNF chromatin remodelling complex, with abnormalities in one or more of these genes present in nearly 25% of all cancers, which indicating that stabilizing normal expression of genes encoding subunits in the SWI/SNF complex may prevent tumorigenesis. In this paper, we will review the relationship between the SWI/SNF complex and some clinical tumours and its mechanism of action. The aim is to provide a theoretical basis to guide the diagnosis and treatment of tumours caused by mutations or inactivation of one or more genes encoding subunits of the SWI/SNF complex in the clinical setting. This work aimed to detect changes in trace element chemical speciation in sewage sludge (SS) after the ozonation process. The modified Community Bureau of Reference (BCR) sequential extraction procedure was performed to determine the chemical speciation of trace elements in SS after the ozonation process. To assess potential soil contamination with trace elements from sewage sludge after the ozonation process, the risk assessment code (RAC) coefficient was used. The bioaccumulation factor (BAF) and translocation factor (TF) values were also calculated to characterize the efficiency of trace element accumulation in the studied plant species from soil fertilized with sludges after the ozonation process. Generally, the mean concentration of total trace elements in the SS after the ozonation process was higher, but the differences were statistically significant only in the case of Mn, Cu, Pb, and Cd. The dominant fraction of Fe, Cr, Pb, and Cd was the residual fraction F4, while the extractable/exchangeable fraction F1 was present in the smallest amount. Therefore, in the case of Mn, Zn, and Ni, the ozonation process had a significant impact on the increase in the content of these elements in the F1 fraction. The application of the SS stabilized by ozonation process for maize and wheat fertilization did not significantly affect the bioaccumulation of most of the analyzed metals in aboveground biomass. Higher values of BAF coefficients after the application of ozonated SS were found only in the case of Cu and Ni. In turn, the determined TF coefficients were lower than 1 in most cases. The obtained results showed that the slight change in the concentration of Zn, Mn, and Ni in fraction F1 causes a specific risk of their mobility in the soil environment. It should be noted that due to the variable composition of sewage sludge, an analysis of the content of individual trace elements in chemical fractions should be carried out to assess its actual impact on the environment. Elemental analysis of fluorochemicals has received renewed attention in recent years stemming from the increased use of fluorinated compounds. However, fundamental drawbacks of in-plasma ionization have hindered ICPMS applications in this area. Recently, we have introduced post-ICP chemical ionization for BaF+ formation using Ba-containing reagent ions supplied by nanospray, leading to major improvements in F detection sensitivity. Here, we present further insights into this post-plasma chemical ionization. First, we examine the effect of oxygen introduced into the plasma (a necessity for organic solvent introduction) on BaF+ ion formation. The results indicate that excess plasma oxygen leads to abundant HNO3 in the post-plasma flow, shifting ionization reactions toward BaNO3+ formation and suppressing BaF+ sensitivity. To amend this, we utilize reagent ions with other metal centers to impart selectivity toward F detection. Our investigations show that robustness of F detection in the presence of abundant HNO3 improves in the order Al3+ ≈ Sc3+ > La3+ > Mg2+ > Ba2+ as the metal center in the reagent ions, consistent with the stronger metal–F bond in the series. Sc-based ionization resulting in ScNO3F+ shows the best balance between sensitivity and robustness in the presence of nitric acid. Similarly, this ion shows an improved tolerance relative to BaF+ for a Cl-containing matrix where HCl interferes with ionization. Finally, we demonstrate a unique feature of post-plasma chemical ionization for real-time flagging of matrix effects via monitoring reagent ions. These findings provide significant improvements of post-plasma chemical ionization for elemental F analysis, particularly for online chromatographic detection where solvent gradients are utilized. The primary source of soil pollution is a complex mixture of numerous inorganic and organic compounds (including chlorinated compounds, nutrients, and heavy metals, etc.). The presence of all of these compounds makes remediation and cleanup difficult. In this study, the phytoremediation ability of Jatropha curcas and Pongamia pinnata was tested to remove nickel (Ni) and Zinc (Zn) from paper mill and municipal landfill contaminated soils, to understand the uptake potential and to estimate the accumulation pattern of Ni and Zn in the vegetative parts of the plant. The experiments were carried out in pots (3 kg capacity) and the different combinations of soil were made by mixing the contaminated soil with a reference soil (forest soil) as T 0 , T 25 , T 50 , T 75 and T 100. The plant biomass, chlorophyll content, proline, nitrate reductase activity and metal removal efficiency (%)were determined after 120 DAS (i.e., the days after sowing). The results of the study showed that with increasing metal stress, there is a reduction in the above-ground biomass content in both the plant species with a slightly less impact on the root biomass. Over a period of 4 months, J. curcas and P. pinnata removed 82–86% and 93-90% Ni, respectively. The removal of Zn was significantly less as compared to Ni as most of the Zn remained in the belowground part (roots) and in the soil. Besides, the phytostabilization capacities of the plants were calculated on the basis of their tolerance index (TI), bioaccumulation factor (BAF) and translocation factor (TF). Gammaherpesviruses, including Kaposi sarcoma-associated herpesvirus (KSHV) and Epstein-Barr virus (EBV), are DNA viruses that are globally associated with human cancers and establish lifelong latency in the human population. Detection of gammaherpesviral infection by the cGAS-STING innate immune DNA-sensing pathway is critical for suppressing viral reactivation from latency, a process that promotes viral pathogenesis and transmission. We report that barrier-to-autointegration factor 1 (BAF)-mediated suppression of the cGAS-STING signaling pathway is necessary for reactivation of KSHV and EBV. We demonstrate a role for BAF in destabilizing cGAS expression and show that inhibiting BAF expression in latently infected, reactivating, or uninfected cells leads to increased type I interferon-mediated antiviral responses and decreased viral replication. Furthermore, BAF overexpression resulted in decreased cGAS expression at the protein level. These results establish BAF as a key regulator of the lifecycle of gammaherpesviruses and a potential target for treating viral infections and malignancies. The short-wavelength sub-nanosecond luminescence in BaF 2 material, discovered in the early 80s, gave a new direction to research into fast scintillators and phosphors. In contrast to 'typical' luminescence occurring within the forbidden band gap, the new type of emission was due to transitions between the upper core and valence bands. Thus it was named core-valence luminescence (CVL); the terms cross-luminescence and Auger-free luminescence have also been used. Given its unusual nature, this new luminescence process has attracted considerable interest in the field of condensed matter physics. During the sequent decades (till the end of the previous century) CVL has been experimentally observed in several dozen compounds and the conditions for detecting CVL have been established. Interest in this kind of luminescence increased in recent years due to the emergence of new methods of crystal synthesis, the development of high-speed solid-state photodetectors, and the construction of unique modern set-ups using synchrotron radiation. This paper analyzes the numerous studies of CVL, obtained mainly over the past decade. Experimental and theoretical data on this topic are considered in detail. Special attention is given to new results on scintillation characteristics of BaF 2 in the form of single crystals, ceramics, nanoparticles, and composites. The features of CVL in binary and ternary compounds are discussed. Some Cs- and Ba-based ternary halides show relatively high light yields and represent promising fast scintillators. Although China has been the main manufacturer and consumer of phthalate esters (PAEs), human health ambient water quality criteria (AWQCs) have not been proposed for these chemicals. In this study, the distribution and bioaccumulation of six PAEs (dimethyl phthalate (DMP), diethyl phthalate (DEP), di-n-butyl phthalate (DBP), butyl benzyl phthalate (BBP), bis (2-ethylhexyl) phthalate (DEHP), and di-n-octyl phthalate (DnOP)) were investigated in 11 edible fish species collected from Poyang Lake, China. The results showed that the total concentrations of the six PAEs in the fish ranged between 118.63 and 819.84 μg/kg wet weight (mean of 327.50 ± 190.44 μg/kg). DMP, DEP, DBP, and DEHP were detected in all samples, of which DEHP and DBP were two of the most predominant phthalates, accounting for more than 90% of the total PAEs. The DEHP concentrations in fish with different habitat preferences were different, demersal species were significantly higher than pelagic species (p < 0.05). The mean natural logarithmic bioaccumulation factors (log BAFs) of PAEs increased with increasing lipophilicity of the substances, which yielded the following regression equation: log BAF (L/kg) = 0.103 log Kow + 2.158 (r2 = 0.940, p < 0.05, n = 4). Using this quantitative structure–activity relationship to calculate BAFs for the remaining undetected substances (BBP and DnOP) to derive AWQCs. According to the natural parameters, the human health AWQCs relating to PAE concentrations for water and fish consumption were derived as 9.4 × 103 (DMP), 5.0 × 102 (DEP), 4.2 × 101 (DBP), 1.1 (BBP), 8.6 × 10− 2 (DEHP), and 2.0 (DnOP) μg/L. Human health risk assessment indicated that the dietary intake of DEHP may exert a carcinogenic effect on residents of the Poyang Lake region. Mutations in DNAJC5/CSPα are associated with adult neuronal ceroid lipofuscinosis (ANCL), a dominant-inherited neurodegenerative disease featuring lysosome-derived autofluorescent storage materials (AFSMs) termed lipofuscin. Functionally, DNAJC5 has been implicated in chaperoning synaptic proteins and in misfolding-associated protein secretion (MAPS), but how DNAJC5 dysfunction causes lipofuscinosis and neurodegeneration is unclear. Here we report two functionally distinct but coupled chaperoning activities of DNAJC5, which jointly regulate lysosomal homeostasis: While endolysosome-associated DNAJC5 promotes ESCRT-dependent microautophagy, a fraction of perinuclear and non-lysosomal DNAJC5 mediates MAPS. Functional proteomics identifies a previously unknown DNAJC5 interactor SLC3A2/CD98hc that is essential for the perinuclear DNAJC5 localization and MAPS but dispensable for microautophagy. Importantly, uncoupling these two processes, as seen in cells lacking SLC3A2 or expressing ANCL-associated DNAJC5 mutants, generates DNAJC5-containing AFSMs resembling NCL patient-derived lipofuscin and induces neurodegeneration in a Drosophila ANCL model. These findings suggest that MAPS safeguards microautophagy to avoid DNAJC5-associated lipofuscinosis and neurodegeneration. BAF and PBAF are mammalian SWI/SNF family chromatin remodeling complexes that possess multiple histone/DNA-binding subunits and create nucleosome-depleted/free regions for transcription activation. Despite previous structural studies and recent advance of SWI/SNF family complexes, it remains incompletely understood how PBAF-nucleosome complex is organized. Here we determined structure of 13-subunit human PBAF in complex with acetylated nucleosome in ADP-BeF3-bound state. Four PBAF-specific subunits work together with nine BAF/PBAF-shared subunits to generate PBAF-specific modular organization, distinct from that of BAF at various regions. PBAF-nucleosome structure reveals six histone-binding domains and four DNA-binding domains/modules, the majority of which directly bind histone/DNA. This multivalent nucleosome-binding pattern, not observed in previous studies, suggests that PBAF may integrate comprehensive chromatin information to target genomic loci for function. Our study reveals molecular organization of subunits and histone/DNA-binding domains/modules in PBAF-nucleosome complex and provides structural insights into PBAF-mediated nucleosome association complimentary to the recently reported PBAF-nucleosome structure. Fast and heavy inorganic scintillators with suitable radiation tolerance are required to face the challenges presented at future hadron colliders of high energy and intensity. Up to 5 GGy and 5 × 1018 neq/cm2 of one-MeV-equivalent neutron fluence is expected by the forward calorimeter at the Future Hadron Circular Collider. This paper reports the results of an investigation of proton- and neutron-induced radiation damage in various fast and heavy inorganic scintillators, such as LYSO:Ce crystals, LuAG:Ce ceramics, and BaF2 crystals. The experiments were carried out at the Blue Room with 800 MeV proton fluence up to 3.0 × 1015 p/cm2 and at the East Port with one MeV equivalent neutron fluence up to 9.2 × 1015 neq/cm2, respectively, at the Los Alamos Neutron Science Center. Experiments were also carried out at the CERN PS-IRRAD proton facility with 24 GeV proton fluence up to 8.2 × 1015 p/cm2. Research and development will continue to develop LuAG:Ce ceramics and BaF2:Y crystals with improved optical quality, F/T ratio, and radiation hardness. [ABSTRACT FROM AUTHOR]

Double-PHD fingers 3 (DPF3) is a BAF-associated human epigenetic regulator, which is increasingly recognised as a major contributor to various pathological contexts, such as cardiac defects, cancer, and neurodegenerative diseases. Recently, we unveiled that its two isoforms (DPF3b and DPF3a) are amyloidogenic intrinsically disordered proteins. DPF3 isoforms differ from their C-terminal region (C-TERb and C-TERa), containing zinc fingers and disordered domains. Herein, we investigated the disorder aggregation properties of C-TER isoforms. In agreement with the predictions, spectroscopy highlighted a lack of a highly ordered structure, especially for C-TERa. Over a few days, both C-TERs were shown to spontaneously assemble into similar antiparallel and parallel β-sheet-rich fibrils. Altered metal homeostasis being a neurodegeneration hallmark, we also assessed the influence of divalent metal cations, namely Cu2+, Mg2+, Ni2+, and Zn2+, on the C-TER aggregation pathway. Circular dichroism revealed that metal binding does not impair the formation of β-sheets, though metal-specific tertiary structure modifications were observed. Through intrinsic and extrinsic fluorescence, we found that metal cations differently affect C-TERb and C-TERa. Cu2+ and Ni2+ have a strong inhibitory effect on the aggregation of both isoforms, whereas Mg2+ impedes C-TERb fibrillation and, on the contrary, enhances that of C-TERa. Upon Zn2+ binding, C-TERb aggregation is also hindered, and the amyloid autofluorescence of C-TERa is remarkably red-shifted. Using electron microscopy, we confirmed that the metal-induced spectral changes are related to the morphological diversity of the aggregates. While metal-treated C-TERb formed breakable and fragmented filaments, C-TERa fibrils retained their flexibility and packing properties in the presence of Mg2+ and Zn2+ cations. Haemoglobinopathies, including thalassaemias and sickle-cell syndromes, are demanding, lifelong conditions that pose a significant burden to patients, families, and healthcare systems. Despite the therapeutic advances and the resulting improvements in prognosis accomplished in past decades, these patients still face important challenges, including suboptimal access to quality care in areas with developing economies, changing epidemiology due to massive migration flows, an evolving clinical spectrum due to ageing in well-treated patients, and limited access to novel high-cost therapies. We herein describe the organization of healthcare services for haemoglobinopathies in Cyprus--with particular focus on beta-thalassaemia, the most prevalent condition in this region--along with selected patient outcomes. This report aims at underscoring the fact that nationally funded and well-coordinated prevention and care programmes for chronic and complex conditions, such as haemoglobinopathies, with active involvement from patient organizations lead to effective disease control and excellent outcomes in survival, quality of life, social adaptation, and public health savings, and allow timely and effective responses to emerging crises, such as the COVID-19 pandemic. The Cyprus paradigm could therefore serve as a blueprint for the organization or adaptation of haemoglobinopathy programs in other countries since these disorders are still widely occurring. Objective: To search and analyze the related genes of liver hepatocellular carcinoma (LIHC) by using bioinformatics technology. Methods: Gene expression omnibus (GEO) was used to retrieve the entry of "Liver hepatocellular carcinoma", and GSE109903 chip data was downloaded. The differentially expressed genes in the control group and liver hepatocellular carcinoma group were screened by bioinformatics analysis. GO enrichment analysis, KEGG pathway analysis, differential gene expression analysis and visualization processing were performed for the differentially expressed genes; Protein interaction network analysis and visualization processing were used to screen core genes EEF1A1 and HK2 with strong correlation with liver hepatocellular carcinoma. GEPIA, Kaplan-Meier plotter, GeneMANIA and Timer 2.0 databases were used to analyze the differential expression, prognostic value and immune cell infiltration of key genes in LIHC. Results: A total of 1 059 differentially expressed genes were screened, including 637 up-regulated genes and 872 downregulated genes. Functional analysis showed that differentially expressed genes were mainly involved in the process of positive transcription regulation, nucleosome function, chromatin function and RNA binding. Pathway analysis showed that differentially expressed genes were involved in systemic lupus erythematosus, alcoholism and RNA polymerase I promoter opening pathway. The analysis of differential gene expression showed that the drugs with similar gene characteristics were mainly CDC inhibitors, prostaglandins, serotonin receptor antagonists, BAF transcriptional repression inhibitors, tyrosine phosphatase inhibitors, etc; Protein interaction network analysis showed that the main genes associated with LIHC were EEF1A1, HK2, FAM38A and LAMB3; EEF1A1 and HK2 genes were further analyzed by GEPIA. The results showed that the expression of EEF1A1 and HK2 mRNA in LIHC tissues was higher than that in normal tissues, and was significantly correlated with the pathological stage, overall survival rate and disease-free survival rate of LIHC patients. EEF1A1 and HK2 may be potential prognostic biomarkers for LIHC patients. In addition, the functions of EEF1A1 and HK2 were mainly related to translation factor activity, molecular chaperone mediated autophagy and carbohydrate catabolism, and purine nucleoside diphosphate metabolism, respectively. Immunocyte infiltration showed that the expression of EEF1A1 and HK2 was significantly correlated with the infiltration of a variety of immune cells, including six types of immune cells: CD4+T cells, macrophages, neutrophils, B cells, CD8+T cells and dendritic cells. Conclusion: By screening differentially expressed genes, we can identify the key genes in the development of liver hepatocellular carcinoma, and screen potential prognostic biomarkers for the survival of patients with liver hepatocellular carcinoma. A series of high quality x%Pr3+:BaF2 (x = 0.6, 1, 2); (1%Pr3+, 2%Na+):BaF2; (1%Pr3+, y%Ca2+):BaF2 (y = 3, 5) and (0.6%Pr3+, 10%R3+):BaF2 (R = Y, Lu, Gd) crystals were grown by temperature gradient technique (TGT). The Raman spectra, absorption spectra, fluorescence spectra, and fluorescence decay lifetime were measured. XRD patterns indicate that the as-synthesized samples are pure BaF2 phase. The absorption cross-section of Pr3+ ions in the BaF2 matrix can be greatly improved by co-doping R3+ ions, while improvement by Na+ and Ca2+ ions is marginal. The fluorescence characteristic of Pr3+ ions can be effectively regulated by co-doping Na+, Ca2+, R3+ (R = Y, Lu, Gd) ions of different valence states. When R3+ ions are introduced, the fluorescence spectra show the strongest orange emission and the CIE chromaticity coordinates of Pr3+ ions are also in the orange region. When single-doped Pr3+ ions and Na+ or Ca2+ ions are introduced, the fluorescence lifetime of the 3P0 level is double exponential decay, and the short lifetime is the main proportion. However, the fluorescence centers tend to homogeneous and stable by R3+ ions co-doping and the fluorescence lifetime exhibits single-exponential decay, which is fitted to 49.7 μs, 47.8 μs and 49.6 μs of (0.6%Pr3+, 10%R3+):BaF2 (R = Y, Lu, Gd), respectively. All the results show that (Pr3+, Gd3+):BaF2 and (Pr3+, Na+):BaF2 are excellent laser gain media for high-efficiency orange and red laser operation. [ABSTRACT FROM AUTHOR]

Four-wave mixing processes are ubiquitous in ultrafast optics and the determination of the coefficients of the χ(3) tensor is thus essential. We introduce a novel time-resolved ultrafast spectroscopic method to characterize the third-order nonlinearity on the femtosecond time-scale. This approach, coined as "nonlinear chirped interferometry," makes use of the variation of the optical group delay of a transmitted probe under the effect of an intense pump pulse in the nonlinear medium of interest. The observable is the spectral interference between the probe and a reference pulse sampled upstream and the metric is the transient swing of the probe group delay. We show that the detected signal is enhanced when the pulses are weakly chirped, and that, although interferometric, the method is intrinsically less sensitive to environmental phase fluctuations and drifts. By chirping adequately the reference pulse, the transient frequency shift of the probe pulses is also detected in the time domain and the detected nonlinear signal is enhanced. Nonlinear phase shifts as low as 10 mrad, corresponding to a frequency shift of 30 GHz, i.e., 0.01% of the carrier frequency, are detected without heterodyne detection or active phase-stabilization. The diagonal and/or non-diagonal terms of reference glasses (SiO2) and crystals (Al2O3, BaF2, CaF2) are characterized.
