Buffalopox (BPX) is a highly contagious disease that causes high morbidity and production losses in buffaloes. During this study, seroprevalence, effect of various associated risk factors, and pathological studies of BPX were recorded in the Punjab province. A total of 97 blood samples and 63 scabs were collected from clinically pox suspected buffaloes. Serum was harvested to perform single radial hemolysis to assess the seroprevalence, and scabs were subjected to PCR for BPX virus confirmation. Results revealed that, animal demographics and environmental associated factors showed significant effect (p⟨0.05,1⟨R2⟩0) on BPX occurrence. The overall BPX seroprevalence was recorded 4.18% in the Punjab province. The BPX was recorded 5.48% in Nili Ravi breed during winter (7.42%), aged 5-7 years (7.46%) under loose housing (5.51%) in the Faisalabad region (8.03%). Further, BPX was 5.37% in pregnant, 6.86% pregnant milking buffaloes during the 3rd lactation period (7.28%) in dairy herds (5.20%). The BPX was 5.22% in non-vaccinated buffaloes where multiple animals were reared together (4.99%) in the herds having 21-30 total number of animals. A total of 49 scab samples were found positive for the BPX virus via PCR with C18L gene amplification. Grossly, inflammatory lesions with pits in the center and wart-like nodules were seen on teats and udder of buffaloes. Increased leukocytes, especially neutrophils and lymphocytes, were seen in the blood of the infected animals. These results provide a broader window to understand the effect of associated risk factors, strengthen the diagnostic aid, and to contain the current spread of BPX in Pakistan to safeguard large ruminant-based livelihood., (Copyright© by the Polish Academy of Sciences.)

Background: Neonatal sepsis is a leading cause of morbidity and early-life mortality worldwide, and previous data have reported the highest neonatal mortality rate in Pakistan., Purpose: The present study aimed to decipher the prevalence of group B Streptococcus (GBS)-associated sepsis, coinfections, and antibiotic susceptibility of isolated microbes in neonates., Methods: Blood samples of 100 cases of neonatal sepsis were subcultured on blood agar, GBS agar, and MacConkey agar for isolation of GBS and suspected microbes., Results: Of 100 neonatal blood samples, 85 blood samples were culture-positive, including mixed culture growth (n = 18), 25 samples as early-onset neonatal sepsis (EONS) and 60 as late-onset neonatal sepsis (LONS). Staphylococcus aureus showed high percent positivity (31.4%), followed by Candida sp (16.5%), GBS (14.1%), Klebsiella (14.1%), Staphylococcus epidermidis (11.8%), Pseudomonas (9.4%), Acinetobacter (9.4%), Esherichia coli (8.2%), and Enterococcus (5.9%). GBS was isolated more frequently from EONS than from LONS with 50% coinfections. Mode of delivery, gender, and respiratory distress in neonates were significantly associated with culture-positive sepsis. GBS isolates were highly (91.7%) susceptible to vancomycin, cefotaxime, and chloramphenicol, followed by penicillin (83.3%), ampicillin, and tetracycline (75%). GBS isolates were resistant to erythromycin, clindamycin, ciprofloxacin, and linezolid., Implications for Practice: Our findings evidenced GBS-associated risk factors and antibiotic susceptibility pattern of neonatal sepsis, which will help clinicians in management, control, and treatment of neonatal sepsis., Implications for Research: The epidemiological evidence of GBS-associated neonatal sepsis, demographic characteristics, risk factor data analysis, and drug resistance pattern has disease prevention implications in neonates in low-income countries including Pakistan.

Toxoplasmosis, an infection caused by Toxoplasma (T.) gondii Apicomplexa protozoan, is widespread in humans and other animal species, having already been reported in many countries and different climates. In Pakistan, no data is available on this aspect among food animals. This study was undertaken to determine the seroprevalence of T. gondii infection in sheep and goats. A total of 200 serum samples from sheep and goats, were collected from urban area of Rahim Yar Khan (Punjab), Pakistan and tested for Toxoplasmosis with a commercial latex agglutination kit (Eiken Chemical Co., Ltd. Japan). The overall seroprevalence of Toxoplasmosis was 19%. Goats had a significantly higher (p < 0.01) prevalence (25.4%) as compared to the sheep (11.2%); and higher (p < 0.01) in the female (24%) than in the males (19%) for both species. In the present study the male (both in sheep and goat) are found less seropositive T. gondii (OR = 0.23; 99% C.I. = 0.01, 1.81) as compared to female sheep and goat. The prevalence was significantly higher (p < 0.01) in adult sheep than younger animals. Among both the sheep and goats the group from 1-1.5 years are highly seropositive (OR = 1.75; 99% C.I. = 0.47, 6.51) as compared to the group less than one year of age followed by the 2-2.5 years age group (OR = 1.63; 99% C.I. = 0.50, 5.74) whereas group with more than 3 years of age least seropositive.

The present paper describes the field evaluation of local gametocyte vaccine and its comparative efficacy with commercial anticoccidial vaccine, LivaCox, used in breeder and broiler flocks in Pakistan. Humoral immune response in vaccinated and control chickens was monitored by enzyme-linked immunosorbent assay. Results demonstrated significantly elevated antibody titres in vaccinated groups as compared to control groups conducted both in Laboratory and field experiments. Significantly (P < 0.01) higher antibody titres in local gametocyte-vaccinated group as compared to LivaCox-vaccinated chickens were recorded. Splenic cell migration inhibition assay was used to detect the cell-mediated immune (CMI) response, and results were expressed in terms of per cent migration index. Lower per cent migration index in LivaCox-vaccinated chickens indicated the higher CMI response, as compared to local gametocyte-vaccinated chickens, although the difference was statistically non-significant (P > 0.05). Results of the challenge studies in laboratory experiments revealed significantly higher (P < 0.05) oocyst count in LivaCox-vaccinated group as compared to local gametocyte-vaccinated chickens.Maximum protection (75%) against mixed species of genus Eimeria was recorded in chickens vaccinated with gametocyte vaccines as compared to LivaCox-vaccinated group. The mean body weight gains in chickens vaccinated with local gametocyte vaccine were significantly better (P < 0.05) than in chickens vaccinated with LivaCox vaccine, both in laboratory and field experiments. Majority of the chickens (70-72%) in control group demonstrated severe lesions (3.0-4.0), while 20-26% chickens showed moderate lesions (2.0). On the other hand, local gametocyte- and LivaCox-immunized chickens developed 78% and 85% mild to moderated lesions (1.0-2.0), respectively. Results of the present study provide a probable explanation for cross-protection induced by Eimeria tenella gametocyte vaccines against other species of genus Eimeria.

Angiotensin-converting enzyme (ACE) plays an important role in the development of systemic lupus erythematosus (SLE) because its end-product, angiotensin II, plays an integral role in the regulatory system responsible for endothelial control and vascular tone, systems that are commonly affected in patients with SLE. Additionally, ACE inhibitors have been shown to retard the progression of SLE and lupus nephritis. Our goal was to investigate whether ACE gene polymorphisms are associated with increasing severity of SLE. We genotyped 39 SLE patients of varying disease severity from a homogenous Asian population and 79 control subjects for ACE I/D and 2350 G > A dimorphisms. All patients met the American College of Rheumatology (ACR) criteria for SLE and their disease severity was measured using Systemic Lupus Activity Measure (SLAM). The "A" allele was found to be associated with increase in severity of SLE with the AA genotype present only in severe disease. No association with SLE in general, compared to healthy subjects, was found with either dimorphism. We also examined the transmission of haplotypes as defined by these polymorphisms. The D and A alleles were found in strong linkage disequilibrium especially in severe SLE. The DA-haplotype was more frequent in severe SLE, than mild to moderate disease. Our findings suggest that DNA sequence variation in the ACE gene influences disease progression and severity of SLE.
