The genus Trichoderma holds economic significance due to its widespread distribution and diverse applications, including biological control, enzyme production, and various biotechnological uses. The accurate identification of Trichoderma species is crucial given their close association with human activities. Despite previous efforts in classification, a comprehensive analysis combining morphological and molecular approaches is necessary. This study focuses on the isolation of four Trichoderma species from industrial wastewater in Pakistan, expanding on the known diversity in the region; isolation involved collecting samples from industrial wastewater effluents at specific sites in Punjab, Pakistan. Trichoderma strains were cultured and purified on solid media, with subsequent biomass production for bisorptional activity. Morphological characterization included colony features and microscopic examinations. DNA extraction, polymerase chain reaction (PCR), and sequencing of the internal transcribed spacer (ITS) region were conducted for molecular analysis. Phylogenetic analysis was performed using the Maximum Likelihood Algorithm. The study identified three Trichoderma species, viz. T. citrinoviride, T. erinaceum, and T. longibrachiatum. Each species was characterized morphologically and supported by molecular–phylogenetic analysis. Illustrations of microscopic features and a phylogenetic tree based on the ITS-nrDNA region were recorded. T. citrinoviride and T. longibrachiatum, isolated from steel mill and tanneries wastewater, respectively, were differentiated based on morphological characteristics such as phialides and conidia. The combination of morphological and molecular techniques enhances the accuracy of species identification. The study highlights the significance of Trichoderma in industrial wastewater environments and underscores the need for continued research in this area. Future research should focus on exploring the ecological roles and potential applications of the newly identified Trichoderma species. Additionally, further investigations into the biotechnological potential of these species, including enzyme production and bioremediation capabilities, would contribute to their practical applications. The Peelu (Salvadora oleoides Decne.) fruit is well known for its nutritional and medicinal values. The current study analyzed the chemical composition of Salvadora oleoides fruit. Fresh Peelu fruits were harvested, and physicochemical properties, proximate composition, macro- and micronutrients, and phytochemical properties were determined. Moreover, ethanol and methanol fruit extract was analyzed for physicochemical properties. The Peelu fruit seemed to be a potential source of essential macro- ((nitrogen (N), phosphorus (P), potassium (K), calcium (Ca), and magnesium (Mg)) and micronutrients (zinc (Zn), manganese (Mn), iron (Fe), and copper (Cu)). The fruit had significant biochemical properties (total soluble solids (TSS), total acidity (TA), and TSS : TA ratio) with appreciable moisture, crude fiber, and ash contents. The fruit extracts demonstrated significantly higher antioxidants and phenolics, ascorbic acid contents, and carotenoids. Phytochemical screening of fruit revealed the presence of coumarins, flavonoids, phlobatannins, tannins, and terpenoids. Physicochemical and sensory evaluation of extracts indicated its potential for further in vivo study trials. The Peelu fruit was found to be a good source of mineral nutrients, proximate contents, vitamins (ascorbic acid and carotenoid), phytochemicals (total phenolic sand antioxidant contents), and pharmaceutically important metabolites that can be used as functional drink. The pollution is hot issue of current era in world and the current study was carried to explore impacts of brickkilns' emanations on physiochemical properties of agricultural lands from District Bhimber of Azad Jammu and Kashmir (AJK) Pakistan. In this research, various edaphic characteristics: pH, soil organic matter, organic carbon, water holding capacity, cation exchange capacity and heavy metal contamination in soils nearby of brickkilns were determined. The pH of soil ranged from 5.55 to 7.50, soil organic matter was 0.35–0.90% and organic carbon content was 0.65–1.40%. The water holding capacity ranged from 2.10 to 3.20 mgL-1 and carbon exchange capacity was 1250 to 4202 meq/100g. The contamination profile of heavy metal depicted that Pb showed highest conc. 0.065 mg/g followed by Co (0.053 mg/g) and Ni with 0.52 mg/g in the soil. Pb and Cr had high conc. in soil samples around brickkilns due to burning of coal and rubber tyres as fuel. The conc. of sulphate and nitrate ranged from 0.90±0.50 mol L-1 to 4.25±0.65 mol L-1 and 2.30±0.50 mol L-1 to 6.55±0.25 mol L-1, respectively. The fertility of agriculture lands was depicted that edaphic properties were directly related while nutritive features were inversely commensurate to distance from brickkilns. The research proved that emanations of brickkilns causes severe impact on quality of agriculture land, plant growth and its yield. Hence, reclamation measures should be taken to mitigate and/or eradicate nuisance of brickkilns emanations by using environmental friendly strategies. [ABSTRACT FROM AUTHOR]

Loquat [Eriobotrya japonica (Thunb.) Lindl.] is an important fruit crop in Pakistan; however, a constant decline in its production is noted due biotic and abiotic stresses, particularly disease infestation. Fungal pathogens are the major disease-causing agents; therefore, their identification is necessary for devising management options. This study explored Taxila, Wah-Cantt, Tret, Chatar, Murree, Kalar-Kahar, Choa-Saidan-Shah and Khan-Pur districts in the Punjab and Khyber Paktoon Khawa (KPK) provinces of Pakistan to explore the diversity of fungal pathogens associated with loquat. The samples were collected from these districts and their microscopic characterizations were accomplished for reliable identification. Alternaria alternata, Curvularia lunata, Lasiodiplodia theobromae, Aspergilus flavis, Botrytis cinerea, Chaetomium globosum, Pestalotiopsis mangiferae and Phomopsis sp. were the fungal pathogens infesting loquat in the study area. The isolates of A. alternata and C. lunata were isolated from leaf spots and fruit rot, while the isolates of L. theobromae were associated with twig dieback. The remaining pathogens were allied with fruit rot. The nucleotide evidence of internal transcribed spacer (ITS) regions (ITS1, 5.8S, and ITS2) were computed from all the pathogens and submitted in the database of National Center for Biotechnology Information (NCBI). For multigene analysis, beta-tubulin (BT) gene and glyceraldehyde 3-phosphate dehydrogenase (GAPDH) regions were explored for A. alternata and C. lunata isolates, respectively. The virulence scales of leaf spots, fruit rot, and twig dieback diseases of loquat were developed for the first time through this study. It is the first comprehensive study with morpho-molecular identification, and newly developed virulence scales of the fungal pathogens associated with loquat, which improves the understanding of these destructive diseases. Mungbean yellow mosaic virus (MYMV) is an important constraint in successful production of mungbean (Vigna radiata L.) in many countries, including Pakistan. The MYMV spreads by insect vector whitefly (Bemisia tabaci Gennadius). The use of resistant cultivars is the most effective management tactics for MYMV. Twenty mungbean varieties/lines were screened against insect vector of MYMV under field condition in the current study. Resistance levels for varieties/lines were assessed through visual scoring of typical disease symptoms. Furthermore, the impacts of two insecticides 'Imidacloprid' and 'Thiamethoxam' and two plant extracts, i.e., neem (Azadirachta indica), and Eucalyptus (Eucalyptus camaldulensis) were tested on the suppression of whitefly. Field screening indicated that none of the tested varieties/lines proved immune/highly resistant, while significant variations were recorded among varieties/lines for resistance level. All varieties/lines were systemically infected with MYMV. The varieties 'AARI-2006' and 'Mung-14043' were considered as resistant to MYMV based on visual symptoms and the lowest vector population. These varieties were followed by 'NM-2006' and 'NL-31', which proved as moderately resistant to MYMV. All remaining varieties/lines were grouped as moderately to highly susceptible to MYMV based on visual symptoms' scoring. These results revealed that existing mungbean germplasm do not possess high resistance level MYMV. However, the lines showing higher resistance in the current study must be exploited in breeding programs for the development of resistant mungbean varieties/lines against MYMV. Imidacloprid proved as the most effective insecticide at all concentrations to manage whitefly population. Peanut (Arachis hypogea L.) is an important nut crop extensively grown in rainfed regions of Pakistan. The crop requires low inputs; thus, could grow successfully under diverse environmental conditions. Due to pegging ability, peanut grows aggressively in sandy and sandy-loam soils. However, it has not introduced to Thal region of southern Punjab, Pakistan. A two-year field experiment was conducted to optimize sowing dates for two peanut genotypes ('BARI-2016' and 'NO-334') in Thal region (Layyah). Similarly, a yield trial was conducted at Chakwal where both genotypes are extensively grown. Five sowing dates (10th April, 1st May, 20th May, 10th June and 30th June) were included in the study. The highest seed yield was obtained with early sown crop (10th April) during both years. Pod formation reduced with increasing atmospheric temperature and no pods were formed on the plants sown on 30th June. Decreased pod formation seemed a major reason for low yield in late-sown crop. The highest yield was observed for the crop sown on 10th April, which was decreased by 40% for the crop sown on 1st May. Genotype 'BARI-2016' performed better for seed yield at both locations compared with 'NO-334'. The results suggested that genotype 'BARI-2016' is more adaptive to arid and semi-arid condition under rainfed or irrigated conditions. Sowing peanut at optimum time would increase seed yield in arid and semi-arid regions. Nonetheless, 'BARI-2016' can be grown under rainfed and irrigated conditions successfully. Cotton (Gossypium hirsutum L.), being a cash and fiber crop is of high significance in Pakistan. Numerous insect pests and viral diseases in Pakistan and around the world attack cotton crop. Genetically modified cotton (transgenic, resistant to lepidopteran insects), hereafter written as 'Bt-cotton' has been introduced in many regions of the world to combat bollworms. However, cultivars differ in their pest susceptibility, yield response and fiber quality traits. Nonetheless, recent studies have indicated that lepidopteran pests are evolving resistance against 'Bt-cotton'. Several 'Bt-cotton' cultivars have been developed in Pakistan in the past decade; however, limited is known about their pest susceptibility, seed-cotton yield and fiber quality traits. This two-year field study evaluated pest susceptibility, yield and fiber quality traits of thirteen newly developed 'Bt-cotton' cultivars in Pakistan. The cultivars differed in their susceptibility to sucking insects during both years of study. The cultivars 'FH-647', 'SLH-8', 'FH-Lalazar' and 'IUB-013' were more susceptible to jassid, whereas 'BS-52' exhibited higher susceptibility to whitefly during both years of study. Similarly, cultivars 'AGC-999' and 'MNH-992' proved highly susceptible to thrips during each study year. Although 'Bt-cotton' is resistant to bollworms, cultivars 'SLAH-8', 'VH-305' and 'BH-184' were susceptible to spotted bollworm, while 'SLAH-8', 'RH-647' and 'VH-305' were infested by American bollworm. The most susceptible cultivars to cotton leaf curl virus (CLCuV) attack were 'RH-647', 'IR-NIBGE-7' and 'VH-305'. The highest seed-cotton yield was recorded for 'FH-Lalazar' during both years of study. Similarly, the highest ginning out turn was recorded for cultivars 'BS-52', 'VH-305', 'RH-647', 'IUB' and 'AA-919'. The cultivar 'FH-Lalazar' exhibited low pest susceptibility and CLCuV infestation compared to the rest of cultivars. The highest and the lowest gross and net incomes and benefit:cost ratio were noted for 'FH-Lalazar' and 'RH-647, respectively. Keeping in view the low pest susceptibility and high seed-cotton yield, 'FH-Lalazar' could be recommended for higher yield and economic returns in Multan, Pakistan.
