Introduction: Herbicide application in tree nurseries is limited because of the potential for chemical injury to the large diversity of trees species grown, the lack of registered products, and increasing restrictions on herbicide use, necessitating the costly practice of hand weeding. Soil solarization can reduce the weed seedbank by trapping solar energy under clear plastic film, resulting in high soil temperatures lethal to imbibed weed seeds and seedlings. The objective of this study was to determine if soil solarization would be an effective weed management strategy in Pacific Northwest, USA, tree production systems. Methods: Field studies were conducted at three commercial tree nurseries in Oregon and Washington over two years to test soil solarization in reducing the naturally occurring weed seedbank and the time required to hand weed fields. Further field and laboratory tests were conducted with five weed species: Poa annua, Polygonum pensylvanicum, Amaranthus retroflexus, Portulaca oleracea, and Cyperus esculentus. Weed seeds and tubers were buried in packets at 5 and 10 cm to determine their viability after 6 weeks of solarization. A laboratory study was conducted with all but C. esculentus to quantify the exposure time at 45, 50, and 55°C required for 90% death (T90). Results: Soil solarization was particularly effective in reducing the emergence of naturally occurring weeds in the fall and winter, when weed emergence was reduced by 94-96%. Emergence was reduced 67-81% during the subsequent spring and early summer. Nine to ten months after solarization, solarized areas had a 52 - 69% reduction in hand weeding time compared to non-solarized areas. In field trials with buried seed and tuber packets, mortality differed by location and depth, with P. annua and P. pensylvanicum having the greatest percent seed mortality followed by A. retroflexus and variable results for P. oleracea and C. esculentus. In lab studies, seed mortality differed depending on species and temperature; however, at 55°C, there was a relatively rapid drop in seed viability for all species, and T90 values ranged from 1.2 to 41 h whereas at 45° C the range was 47 to > 3000 h. Similar to the field studies, P. annua and P. pensylvancium were more sensitive to heat, followed by A. retroflexus and P. oleracea. Conclusion: Soil solarization can be an effective weed management tool in reducing the weed seedbank in Pacific Northwest tree nurseries and other fallsown crops but may not work for certain, thermotolerant weed species such as C. esculentus. Abstract: BACKGROUND: Farmers in the low‐rainfall region of eastern Oregon rely on repeated applications of non‐selective herbicides, predominately glyphosate, to control Salsola tragus in no‐till fallow systems. Reports of poor glyphosate effectiveness have increased in recent years. Reduced efficacy is often attributed to dust, water stress, or generally poor growing conditions during application. Inadequate control also may be the result of the evolution of glyphosate resistance. Therefore, studies were undertaken to determine if glyphosate‐resistant S. tragus populations occur in Oregon. RESULTS: Results from dose–response studies confirmed glyphosate resistance in three of 10 Oregon Salsola tragus populations. The ratio I50R/I50S from dose–response curves was, on average, 3.1 for the relative dry biomass per plant and 3.2 for the % of surviving plants per pot in these three populations. Plant mortality at recommended glyphosate doses for the resistant populations was less than 30% 3 weeks after treatment. CONCLUSIONS: Glyphosate resistance in S. tragus highlights the imperative need to diversify weed control strategies to preserve the longevity and sustainability of herbicides in semi‐arid cropping systems of the Pacific Northwest. © 2017 Society of Chemical Industry [ABSTRACT FROM AUTHOR]

Evolved resistance to the herbicide glyphosate has been reported in eleven weed species, including Lolium multiflorum. Two glyphosate-resistant L. multiflorum populations were collected, one from Chile (SF) and one from Oregon, USA (OR), and the mechanisms conferring glyphosate resistance were studied. Based on a Petri dish dose–response bioassay, the OR and the SF populations were two and fivefold more resistant to glyphosate when compared to the susceptible (S) population, respectively; however, based on a whole-plant dose–response bioassay, both OR and SF populations were fivefold more resistant to glyphosate than the S population, implying that different resistance mechanisms might be involved. The S population accumulated two and three times more shikimic acid in leaf tissue 96 h after glyphosate application than the resistant OR and SF populations, respectively. There were no differences between the S and the glyphosate-resistant OR and SF populations in 14C-glyphosate leaf uptake; however, the patterns of 14C-glyphosate translocation were significantly different. In the OR population, a greater percentage of 14C-glyphosate absorbed by the plant moved distal to the treated section and accumulated in the tip of the treated leaf. In contrast, in the S and in the SF populations, a greater percentage of 14C-glyphosate moved to non-treated leaves and the stem. cDNA sequence analysis of the EPSP synthase gene indicated that the glyphosate-resistant SF population has a proline 106 to serine amino acid substitution. Here, we report that glyphosate resistance in L. multiflorum is conferred by two different mechanisms, limited translocation (nontarget site-based) and mutation of the EPSP synthase gene (target site-based). Concerns about genetically modified (GM) crops include transgene flow to compatible wild species and unintended ecological consequences of potential transgene introgression. However, there has been little empirical documentation of establishment and distribution of transgenic plants in wild populations. We present herein the first evidence for escape of transgenes into wild plant populations within the USA; glyphosate-resistant creeping bentgrass ( Agrostis stolonifera L.) plants expressing CP4 EPSPS transgenes were found outside of cultivation area in central Oregon. Resident populations of three compatible Agrostis species were sampled in nonagronomic habitats outside the Oregon Department of Agriculture control area designated for test production of glyphosate-resistant creeping bentgrass. CP4 EPSPS protein and the corresponding transgene were found in nine A. stolonifera plants screened from 20 400 samples (0.04 ± 0.01% SE). CP4 EPSPS-positive plants were located predominantly in mesic habitats downwind and up to 3.8 km beyond the control area perimeter; two plants were found within the USDA Crooked River National Grassland. Spatial distribution and parentage of transgenic plants (as confirmed by analyses of nuclear ITS and chloroplast matK gene trees) suggest that establishment resulted from both pollen-mediated intraspecific hybridizations and from crop seed dispersal. These results demonstrate that transgene flow from short-term production can result in establishment of transgenic plants at multi-kilometre distances from GM source fields or plants. Selective pressure from direct application or drift of glyphosate herbicide could enhance introgression of CP4 EPSPS transgenes and additional establishment. Obligatory outcrossing and vegetative spread could further contribute to persistence of CP4 EPSPS transgenes in wild Agrostis populations, both in the presence or absence of herbicide selection. DNA sequence analysis of the psbA gene encoding the D1 protein of photosystem II (PS II), the target site of PS II-inhibiting herbicides, identified a point mutation (Asn266 to Thr) in a bromoxynil-resistant Senecio vulgaris L. population collected from peppermint fields in Oregon. Although this mutation has been previously reported in Synechocystis, this is the first report of this particular point mutation in a higher plant exhibiting resistance to PS LI-inhibiting herbicides. The resistant population displayed high-level resistance to bromoxynil and terbadil (R1S ratio 10.1 and 9.3, respectively) and low-level resistance to metribuzin and hexazinone (RJS ratio 4.2 and 2.6, respectively) when compared with the susceptible population. However, the population was not resistant to the triazine herbicides atrazine and simazine or to the urea herbicide diuron. A chlorophyll fluorescence assay confirmed the resistance levels and patterns of cross-resistance of the whole-plant studies. The resistant S. vulgaris plants produced fewer seeds. Differences in cross-resistance patterns to PS II-inhibiting herbicides and the difference in fitness cost could be exploited in a weed management program. [ABSTRACT FROM AUTHOR]

Black leg (caused by Plenodomus lingam and P. biglobosus ) and chlorotic leaf spot (caused by Pyrenopeziza brassicae ) are economically important fungal diseases of Brassicaceae crops. Surveys of seed fields and weed hosts were conducted to understand the distribution and prevalence of these diseases in Oregon after black leg and chlorotic leaf spot outbreaks occurred in Brassicaceae crops in 2014. Postharvest black leg ratings were conducted in seed fields of canola, forage rape, and turnip in 2015 and 2016. The incidence of black leg was greater for turnip (51%) than for canola (29%) and forage rape (25%). The overall average disease incidence was greater for seed crops harvested in 2015 (46%) than for crops harvested in 2016 (28%). A disease survey of wild Brassicaceae plants was conducted along Interstate 5 in Oregon. Brassicaceae weed population sites were identified and 40 sites were sampled for these diseases. Black leg and chlorotic leaf spot were present in 60 and 45%, respectively, of the sampled sites. Both species of Plenodomus were detected in weed populations, with P. lingam being the predominant species recovered (95%). The northernmost sample site with black leg was <32 km from the Oregon-Washington border, and the southernmost site with black leg was within 32 km of the Oregon-California border. Chlorotic leaf spot was detected <32 km from the Oregon-Washington border, whereas the southernmost site where it was detected was approximately 164 km from the Oregon-California border. Based on this study, infected crop residues and weed hosts may facilitate the persistence and spread of these pathogens.

Glufosinate-resistant Lolium perenne L. spp. multiflorum biotypes from Oregon exhibited resistance levels up to 2.8-fold the field rate. One resistant biotype (MG) had an amino acid substitution in glutamine synthetase 2 (GS2), whereas the other (OR) exhibited the wild-type genotype. We hypothesized that the amino acid substitution in GS2 is involved in the resistance mechanism in MG and that non-target site resistance mechanisms are present in OR. OR metabolized glufosinate faster than the other two biotypes, with >75% of the herbicide metabolized in comparison to 50% in MG and the susceptible biotype. A mutation in GS2 co-segregating with resistance in MG did not reduce the enzyme activity, with results further supported by our enzyme homology models. This research supports the conclusion that a metabolism mechanism of glufosinate resistance is present in OR and that glufosinate resistance in MG is not due to an altered target site.

Background: Multiple-herbicide resistance in Lolium perenne spp. multiflorum has evolved in many areas in Oregon. To manage the resistant populations, the resistance patterns must be determined. In this study, a population (CT) suspected to be resistant to sulfometuron and hexazinone was collected from a Christmas tree plantation., Results: The CT population is resistant to at least six herbicides with four different mechanisms of action: atrazine (>16-fold), diuron (2.4-fold), glyphosate (7.4-fold), hexazinone (3.1-fold), imazapyr (1.8-fold) and sulfometuron (>16-fold). Two mutations, Trp-591-Leu and Ser-264-Gly, were identified in the acetolactate synthase (ALS) and psbA gene respectively. No previously reported mutation in the 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS) gene was found. Less shikimic acid accumulated in the CT plants than in the susceptible plants after treatment with glyphosate at 0.6 kg AE ha(-1) ., Conclusion: This study suggests that the multiple resistance patterns of Lolium perenne spp. multiflorum populations can be complex, but that chemical control options to manage these populations exist. These remaining chemical options should be integrated with non-chemical management strategies to slow the spread of multiple-resistant biotypes in agroecosystems., (© 2013 Society of Chemical Industry.)
