To examine the potential for the autogenic ecosystem engineers, crustose coralline algae (CCA), to serve as seed banks or refugia for life stages of other species, it is critical to develop sampling protocols that reflect the diversity of life present. In this pilot study on two shallow water species of CCA collected from Raoul Island (Kermadec Islands; Rangitāhua) New Zealand, we investigated two preservation methods (ethanol vs. silica gel), sampled inner and outer regions of the crusts, and used DNA metabarcoding and seven genes/gene regions (16S rRNA, 18S rRNA, 23S rRNA, cox1, rbcL, and tufA genes and the ITS rRNA region) to develop a protocol for taxa identification. The results revealed immense diversity, with typically more taxa identified within the inner layers than the outer layers. As highlighted in other metabarcoding studies and in earlier work on rhodoliths (nodose coralline algae), reference databases are incomplete, and to some extent, the use of multiple markers mitigates this issue. Specifically, the 23S rRNA and rbcL genes are currently more suitable for identifying algae, while the cox1 gene fares better at capturing the diversity present inclusive of algae. Under the ocean acidification (OA) conditions predicted for 2100, the larval stages of temperate sea urchins are smaller, with reduced and abnormal skeleton and changes in metabolic rate. Here, we measure the impact of near-future and long-term predictions of atmospheric pCO2 levels on the early development of the echinometrid sea urchin Evechinus chloroticus using single male:female crosses, effect sizes and a within-study meta-analysis. Using the developmental signpost of the 16-cell stage, we show a mean delay of 5.11 min at pH 7.7 and 11.85 min at pH 7.5, which may have flow-on effects to later embryo and larval stages. Echinopluteus larvae raised in OA conditions (pH 7.7, 7.5) were ~ 5–10% smaller in body length and 10–12% smaller in arm length than controls. Metabolic rate was highly variable between single male:female crosses—increasing in some crosses, decreasing in others—with a non-significant effect size in the meta-analysis. Preliminary experiments suggest that metabolic rate changes may be impacted by loss of mitochondrial function at low pH. Single male:female crosses showed variable OA responses in all measurements, suggesting that when assessing the population-level impacts of OA on early development of sea urchins there is a need to include high levels of biological replication. The threat posed by ocean acidification (OA) to the diversity and productivity of New Zealand marine ecosystems is assessed in a synthesis of published trends and impacts. A 20-year time series in Subantarctic water, and a national coastal monitoring programme, provide insight into pH variability, and context for experimental design, modelling and projections. A review of the potential impact of changes in the carbonate system on the major phyla in New Zealand waters confirms international observations that calcifying organisms, and particularly their early life-history stages, are vulnerable. The synthesis considers ecosystem and socio-economic impacts, and identifies current knowledge gaps and future research directions, including mechanistic studies of OA sensitivity. Advanced ecosystem models of OA, that incorporate the indirect effects of OA and interactions with other climate stressors, are required for robust projection of the future status of New Zealand marine ecosystems. The physiology of the New Zealand sea urchin Evechinus chloroticus was evaluated through feeding, respiration, growth and gonad growth in adult animals acclimated for 90 days at 18 °C (annual mean temperature) and 24 °C (ambient summer temperature (21 °C) + 3 °C). Measured parameters with representative rates of assimilation efficiency were used to calculate scope for growth (SfG) for each treatment. All physiological parameters were negatively affected at 24 °C, showing a decrease in feeding rate which coincided with negative growth and gonad development at the end of the acclimation period, and a decrease in respiration rate suggesting metabolic depression. Histology of gonad samples after the acclimation period also showed no gametic material in animals acclimated at 24 °C. All animals acclimated at 24 °C had negative growth, differing from the calculated SfG which indicated that the animals had sufficient energy for production. The results suggest that calculated SfG in echinoderms should be used together with actual measurements of growth in individuals as, by itself, SfG may underestimate the actual effect of ocean warming when animals are exposed to stressful conditions. Overall, considering the total loss of reproductive output observed in E. chloroticus at higher temperatures, an increase in seawater temperature could dramatically influence the persistence of northern populations of this species, leading to flow-on effects in the subtidal ecosystem. Temperature and salinity are important environmental factors affecting the normal functioning of marine animals, particularly animals such as sea urchins living in shallow waters and tide pools. Here, we evaluated the effect of different combinations of temperature and salinity on early embryos of the endemic New Zealand sea urchin Evechinus chloroticus. Animals were collected at Matheson's Bay (36º18′17′′S; 174º47′51′′E) in north-eastern New Zealand in February 2013. Embryos were exposed to five salinities (29, 31, 34, 35 and 37 ppt) and two temperatures (18 and 21 °C) during the first 24 h of development. Low salinity (29 ppt) affected all parameters (fertilization, development rate, gastrulation and normal development), with ca. 50 % of embryos surviving at 29 ppt, whereas seawater temperature only affected development rate and gastrulation. An increase in temperature from 18 to 21 °C minimized the negative effect of low salinity (≤31 ppt) on development rate and gastrulation of E. chloroticus. Overall, the results of this study suggest that early embryos of E. chloroticus have developmental plasticity to withstand reductions in salinity up to 29 ppt; however, it is still unknown whether the surviving embryos will be able to complete larval development at low salinities, particularly whether the embryos and larvae are carried into extreme environments such as estuaries where salinity is even lower. Seawater temperature is an important environmental factor for the early life stages of marine invertebrates. In this study we evaluated and described the effects of temperature during early development of E. chloroticus, identifying the optimum temperature range and upper thermal limit for successful development. The temperature range evaluated was between 15–24°C which included the normal seawater temperatures during the spawning season in northern New Zealand, as well as the highest temperature projected by the IPCC for this region due to global warming (1–3°C by the year 2100). Gametes from several females and males were used in the experiment. Fertilization was carried out at different temperatures and development was monitored at different time points after fertilization in each temperature. The development rate of E. chloroticus increased with an increase in seawater temperature. However, at temperatures higher than 21.5°C the amount of abnormal development reached ∼30%. The optimum temperature for early development was between 15–21°C, whereas the upper thermal limit was ∼24°C. Therefore, early development of E. chloroticus is negatively affected by an increase in seawater temperature of ∼3–4°C above current seawater temperature levels in northern New Zealand. The thermal sensitivity of early life stages of E. chloroticus could affect survival rates during early development of this species in a global warming scenario, which could impair recruitment in populations which are exposed to higher temperatures, leading to possible distributional shifts of this species. The ascidian Styela clava, native to the north-west Pacific, is an invasive species affecting New Zealand's marine ecosystems, biodiversity and aquaculture operations. To provide detailed information on the reproductive biology of S. clava in New Zealand for post-border biosecurity management, long-term seasonal patterns of gametogenesis were determined from May 2006 to May 2008 in Auckland's Waitemata Harbour (36°49′20″S, 174°45′85″E). Of particular interest was whether the critical 15°C threshold spawning temperature for reproduction observed in the Northern Hemisphere applied here to the first Southern Hemisphere study. S. clava gametogenesis followed a regular seasonal cycle with ripe gametes appearing as early as September and persisting to June; this time frame corresponds to the period when sea surface temperatures in the region first reach 15°C and with spawning occurring mainly during late summer to early autumn. From photoperiod manipulation, it was determined that spawning occurred at approximately 18:20. The extended reproductive period and a short generation time in the Waitemata Harbour provides a lengthy opportunity for S. clava to spread. Abstract: Plastics pollution in the ocean is an area of growing concern, with research efforts focusing on both the macroplastic (>5mm) and microplastic (<5mm) fractions. In the 1990s it was recognized that a minor source of microplastic pollution was derived from liquid hand-cleansers that would have been rarely used by the average consumer. In 2009, however, the average consumer is likely to be using microplastic-containing products on a daily basis, as the majority of facial cleansers now contain polyethylene microplastics which are not captured by wastewater plants and will enter the oceans. Four microplastic-containing facial cleansers available in New Zealand supermarkets were used to quantify the size of the polythelene fragments. Three-quarters of the brands had a modal size of <100 microns and could be immediately ingested by planktonic organisms at the base of the food chain. Over time the microplastics will be subject to UV-degradation and absorb hydrophobic materials such as PCBs, making them smaller and more toxic in the long-term. Marine scientists need to educate the public to the dangers of using products that pose an immediate and long-term threat to the health of the oceans and the food we eat. [Copyright &y& Elsevier]

In the family Asterinidae, development through a planktonic lecithotrophic brachiolaria larva is common and has evolved independently several times. Here, we describe the lecithotrophic development of the asterinid Stegnaster inflatus, a species endemic to New Zealand. Early development through the blastula and gastrula stages is short, with hatching at the brachiolaria stage occurring within 48 hr. After hatching, larvae are negatively buoyant, and without aeration remain near the bottom of the culture containers. The settled benthic juvenile stage was reached in ~2 weeks. The brachiolaria of S. inflatus shares common characteristics with the planktonic brachiolaria of other asterinids in that the brachiolar attachment apparatus comprises three brachia and a central adhesive disc, although the latter is thin and appears to be reduced. Mortensen (1925, Videns kabelige Meddelelser fra Dansk naturhistorisk Forening i København, 79(15), 261‐420) had hypothesized that individuals of S. inflatus might brood within the "cave" formed in the interambulacral space between the arms. We found no evidence for brooding, but hypothesize that S. inflatus may have demersal development, on or near the bottom, which has implications for larval dispersal and population structure. Seawater temperature is projected to increase globally due to climate change, affecting physiological responses, fitness and survival of marine organisms. Thermal tolerance studies are critical to determine the ability of animals to adapt to future environmental conditions. In this study, we aimed to determine if the thermal limits of the New Zealand Evechinus chloroticus would shift with animal's thermal history. We tested the effect of six thermal regimes on the righting ability, temperature of loss of righting (T LOR), median lethal temperature (LT 50), lethal temperature (LT) and the gene expression of the heat shock protein 70 (hsp70) of the New Zealand sea urchin E. chloroticus when exposed to a thermal shock of 1 °C day−1 (duration of 7–16 days depending on the treatment). Treatments consisted of laboratory acclimation for one and four weeks to 18 °C and 24 °C (mean winter (15 °C) and summer temperature (21 °C) + 3 °C of warming, respectively), compared to non-acclimated sea urchins collected during winter (14.6 °C) and summer seasons (20.4 °C). Thermal history did not have a significant effect on the righting ability of E. chloroticus (T LOR ranged between 28 and 29 °C for all treatments) and LT 50 (ranged between 29 and 30 °C for all treatments). However, LT of E. chloroticus collected during winter season was significantly lower than animals acclimated for one week at 18 ° C. Maximum expression of hsp70 mRNA (T max) was observed at around 27–28 °C regardless of treatment; however, relative hsp70 mRNA levels were significantly higher in animals acclimated for four weeks at 24 °C. Despite proving to be a thermotolerant species with LTs around 30 °C, E. chloroticus was unable to increase thermal tolerance and T max when acclimated to high temperatures, suggesting that E. chloroticus may have a limited adaptive capacity to modify its phenotype; however, evolutionary adaptations may allow E. chloroticus to adapt to future ocean temperatures. In sea urchins, spermatozoa are stored in the gonads in hypercapnic conditions (pH<7.0). During spawning, sperm are diluted in seawater of pH>8.0, and there is an alkalinization of the sperm's internal pH (pHi) through the release of CO2 and H+. Previous research has shown that when pHi is above 7.2-7.3, the dynein ATPase flagellar motors are activated, and the sperm become motile. It has been hypothesized that ocean acidification (OA), which decreases the pH of seawater, may have a narcotic effect on sea urchin sperm by impairing the ability to regulate pHi, resulting in decreased motility and swimming speed. Here, we used data collected from the same individuals to test the relationship between pHi and sperm motility/performance in the New Zealand sea urchin Evechinus chloroticus under near-future (2100) and far-future (2150) atmospheric PCO2 conditions (RCP 8.5: pH 7.77, 7.51). Decreasing seawater pH significantly negatively impacted the proportion of motile sperm, and four of the six computer-assisted sperm analysis (CASA) sperm performance measures. In control conditions, sperm had an activated pHi of 7.52. Evechinus chloroticus sperm could not defend pHi in future OA conditions; there was a stepped decrease in the pHi at pH 7.77, with no significant difference in mean pHi between pH 7.77 and 7.51. Paired measurements in the same males showed a positive relationship between pHi and sperm motility, but with a significant difference in the response between males. Asteroid wasting events and mass mortality have occurred for over a century. We currently lack a fundamental understanding of the microbial ecology of asteroid disease, with disease investigations hindered by sparse information about the microorganisms associated with grossly normal specimens. We surveilled viruses and protists associated with grossly normal specimens of three asteroid species (Patiriella regularis, Stichaster australis, Coscinasterias muricata) on the North Island / Te Ika-a-Māui, Aotearoa New Zealand, using metagenomes prepared from virus and ribosome-sized material. We discovered several densovirus-like genome fragments in our RNA and DNA metagenomic libraries. Subsequent survey of their prevalence within populations by quantitative PCR (qPCR) demonstrated their occurrence in only a few (13%) specimens (n = 36). Survey of large and small subunit rRNAs in metagenomes revealed the presence of a mesomycete (most closely matching Ichthyosporea sp.). Survey of large subunit prevalence and load by qPCR revealed that it is widely detectable (80%) and present predominately in body wall tissues across all 3 species of asteroid. A new malyngamide, S (1), was isolated from a New Zealand collection of the sea hare Bursatella leachii and structurally characterized by spectroscopic methods and chemical degradation. Malyngamide S exhibited cytotoxicity and antiinflammatory properties.
