Purpose: Disseminated nontuberculous mycobacterial (DNTM) infection can involve multiple organs, including the lungs, skin and soft tissues and lymph nodes. However, NTM infection leading to osteolysis has been rarely reported. Here, we analyzed the clinical features, osteolytic mechanisms, treatment and prognosis of patients with DNTM disease with osteolytic lesions. Patients and Methods: This retrospective study was conducted between January 1, 2011, and December 31, 2020, at the First Affiliated Hospital of Guangxi Medical University and the Fourth People's Hospital of Nanning City. Patients who had culture and/or histopathological proof of DNTM disease with osteolytic lesions were included. Results: Ten HIV-negative patients with DNTM disease with osteolytic lesions were enrolled. Five of these patients had underlying diseases. Seven and three of the patients were positive and negative for anti-interferon-γ autoantibodies (AIGAs), respectively. The AIGA positivity rate was 70% (7/10). Ostealgia and anemia were the most common symptoms, followed by fever, emaciation, cough, expectoration, anorexia, subcutaneous abscesses and lymphadenopathy. Leukocyte and neutrophil counts were increased. The most common sites were the vertebrae, sternum, clavicle and ribs, although the femur, ilium, humerus, and scapula were also involved. Radiography and computed tomography (CT) showed moth-eaten or irregular destruction of bone, bone defects, pathological fracture, periosteal proliferation and surrounding abscesses. Emission CT (ECT) bone scans showed significantly increased uptake in many skeletal regions. Positron emission tomography(PET)/CT showed metabolic activity in multiple bones. All patients received anti-nontuberculous therapy, and five underwent surgery. Two died during treatment. Conclusion: DNTM infection of bone and leading to osteolysis usually occurs in patients with AIGA-positive antibodies. DNTM disease with osteolysis is characterized by increased leukocytes and neutrophil counts, focal suppurative granulomas, and multiple areas with moth-eaten or irregular destruction of bone with increased radioactive concentrations. However, the role of DC in the context of Talaromyces marneffei (T. marneffei) infection has not been determined. In this study, we aimed to study the effect of the yeast form of T. marneffei yeasts on DCs, as well as the role of DCs in modulating T helper 17 (Th17) and regulatory T (Treg) cell responses to the pathogen. Methods: Mouse bone marrow-derived DCs were stimulated with T. marneffei yeasts for 24 h. Frequencies of CD80 and CD86 expression on DCs and the levels of IL-6, IL-10 and TGF-β in the culture supernatant of yeast-stimulated DCs were detected by flow cytometry and ELISA, respectively. In co-culture experiments, CD4+ T lymphocytes of mice were isolated from the spleen using magnetic beads and co-cultured with T. marneffei yeasts, with or without DCs for 24 h. The proportions of Th17 and Treg cells in co-culture were detected by flow cytometry. The mRNA levels of RORγt and Foxp3 were detected by RT-PCR. Levels of IL-10 and TGF-β in the co-culture supernatant were detected by ELISA. Results: The expressions of CD80 and CD86 on DCs were increased, as well as IL-6, IL-10 and TGF-β levels in the culture supernatant of T. marneffei-stimulated DCs were higher than those in DCs cultured without T. marneffei. In co-culture experiments, in the presence of DCs, T. marneffei promoted Treg expansion and Foxp3 up-regulation but limited Th17 and downregulated RORγt. Levels of IL-10 and TGF-β were higher in the co-culture containing DCs than without DCs. Conclusion: Our findings demonstrated that the interaction between DCs and T. marneffei could promote Treg expansion but not Th17 generation. [Display omitted] • A novel ECOFB-MES was applied to restore practical polluted urban water bodies. • The TN removal load of ECOFB-MES was 12.7–16.6 times higher than normal ECOFB. • ECOFB-MES increased TC removal efficiency of sediment by 2.4 times. • MES enhanced pollutants removal via electrons compensation under low-carbon water. An innovative ecological floating bed coupled with sedimental microbial electrochemical system (ECOFB-MES) was designed for urban water remediation. A demonstration-scale ECOFB-MES of approximately 500 m2 was firstly constructed to perform the remediation of 8040 m2 urban water located in Nanning, Guangxi Zhuang Autonomous Region, China. During one-year operation, the COD, NH 4 +–N, NO 3 ¯–N and TP concentrations stabilized around 10.3 ± 4.0 mg/L, 0.04 ± 0.02 mg/L, 0.61 ± 0.27 mg/L and 0.12 ± 0.05 mg/L from May 2021 to December 2021. The COD, TN and TP removal loads in April when MESs were connected were 72.47 g/d/m2, 4.26 g/d/m2 and 0.39 g/d/m2, which were 3.6, 15.3 and 2.8 times higher than those in January (open circuit). The average TC removal efficiency of sediment near anode was 36.7 %, which was 2.4 times higher than that of sediment far from anode (15.5 %). The average current of the MESs improved from 0.6 ± 0.1 mA to 32.0 ± 2.9 mA with reducing external resistance gradually, which was responsible for higher TC removal efficiency and lower dissolved oxygen (DO) concentration (7.78 ± 1.35 mg/L), which could enhance nitrogen removal. The MES enhanced the total number of species and the bacteria diversity of polyester fibers and roots in the same coupled system. The dominant functional bacteria such as Pseudomonas , Gemmobacter , Aeromonas and Arthrobacter beneficial for oxygen reduction and nitrogen removal were found on biocathode, and this indicated that biocathode of present system could reduce nitrate mainly through heterotrophic nitrification and aerobic denitrification.
