Purpose: Arbuscular mycorrhizal fungi (AMF) play a key role in medicinal plant species, besides their ecological role in shaping plant communities. Knowledge about the diversity and structure of AMF communities associated with the endangered Prunus africana is valuable in the conservation and domestication of the species for its medicinal products. Methods: We investigated the diversity and structure of AMF species communities in the rhizosphere soils of P. africana trees occurring in four fragmented Afromontane forests found in Cameroon (Mount Cameroon and Mount Manengouba) and Kenya (Chuka and Malava) using Illumina Miseq sequencing of 18S rRNA gene amplicons. Results: A total of 64 virtual taxa (VT) belonging to eight genera were detected, namely Glomus (43 VT), Claroideoglomus (6 VT), Paraglomus (5 VT), Acaulospora (4 VT), Diversispora (3 VT), and Archaeospora, Pacispora, and Scutellospora with 1 VT each. Scutellospora heterogama VTX00286 was the most abundant and common species in all four sites (49.62%). Glomeraceae and Gigasporaceae were the most abundant families found across the sites, while Acaulosporaceae, Pacisporaceae, and Archaeosporaceae were rare, represented by < 1% of all the detected taxa. Conclusion: Our data shows a high diversity of AMF species associated with P. africana and variable community structure partially shaped by local edaphic factors. [ABSTRACT FROM AUTHOR]

Prunus africana bark contains phytochemical compounds used in the treatment of benign prostatic hyperplasia and prostate cancer. It has been shown that this plant establishes association with arbuscular mycorrhizal fungi (AMF). AMF are involved in nutrient uptake, which may also affect plant growth and secondary metabolites composition. However, there is no information regarding the role of AMF in the growth and phytochemical content of P. africana. A pot experiment was carried out to assess the response of 8 months old vegetatively propagated P. africana seedlings inoculated with indigenous AMF collected from Mount Cameroon (MC) and Mount Manengumba (MM) in Cameroon, Malava near Kakamega (MK) and Chuka Tharaka-Nithi (CT) in Kenya. Mycorrhizal (frequency, abundance and intensity), growth (height, shoot weight, total weight, number of leaf, leaf surface) and phytochemical (total phenol, tannin and flavonoids) parameters were measured three months after growth of seedlings from two provenances (Muguga and Chuka) with the following inoculation treatments: MK, CT, MC, MM, non-sterilized soil (NS) and sterilized sand as non-inoculated control. Results showed that seedling heights were significantly increased by inoculation and associated with high root colonization (>80%) compared to non-inoculated seedlings. We also found that AMF promoted leaf formation, whereas inoculation did not have any effect on the seedling total weight. AMF inoculum from MM had a higher tannin content, while no significant difference was observed on the total phenol and flavonoid contents due to AMF inoculation. Pearson’s correlation was positive between mycorrhizal parameters and the growth parameters, and negative with phytochemical parameters. This study is the first report on the effect of AMF on the growth and phytochemical in P. africana. Further investigations are necessary to determine the effect of single AMF strains to provide better understanding of the role of AMF on the growth performance and physiology of this important medicinal plant species. Abstract: Legume biological nitrogen fixation is an environmentally friendly and economical means that can reduce low resource farmer dependence on expensive chemical nitrogen (N) fertilizers. We investigated the effect of two cowpea (Vigna unguiculata (L.) Walp) varieties (IT95K-52-34, an international variety from IITA and Kang’au, a local variety) under an integrated soil fertility management trial on indigenous symbiotic rhizobia in a semi-arid farmer''s field in eastern Kenya. The ox-ploughed field trial had the following treatments: an unamended control, manure applied at 2.5tha−1, triple superphosphate (TSP as (P2O5, 0:46:0) at 15kgha−1; and a combination of manure and TSP applied at the single rates. Soil samples were collected from each treatment during planting and harvesting of the cowpea crop and used in most probable number (MPN) plant infection assays with the two cowpea varieties as traphosts in Leonard jar growth systems and grown under glasshouse conditions. Generally, soil amendments enhanced cowpea rhizobial populations which varied from 4.89×102 rhizobiag−1 soil to 1.074×103 rhizobiag−1 soil. The highest shoot biomass accumulation occurred on cowpea variety IT95K-52-34 plants inoculated with soils from the manure applied plots. We isolated 150 fast- and slow-growing cowpea rhizobia. Contrary to most previous studies, the bulk (97%) of the isolates was fast growing which grouped into 9 types on the growth characteristics on yeast extract agar (YEMA). A field study was carried out on a six-year-old on-farm field trial during long-rains season (April–August) 2003 to investigate the effect of improved fallow systems and phosphorus application on arbuscular mycorrhiza fungi (AMF) symbiosis in maize. The trial comprised of maize rotated with a fast growing leguminous Crotalaria grahamiana fallow and a non-leguminous Tithonia diversifolia fallow for 3 years followed by continuous maize. The experiment was randomized complete block design with three cropping (continuous maize, Crotalaria fallow and Tithonia fallow) systems and two phosphorus levels (0 and 50 kg P/ha). AMF colonization in maize roots, maize yield and macro-nutrients uptake were recorded. Phosphorus applications improved ( P < 0.05) early (<8 weeks old maize) AMF colonization, nutrient uptake and maize yield in improved fallow systems. Greater differences due to phosphorus application were noted in maize in Tithonia fallow than in Crotalaria fallow. Following phosphorus application, a positive relationship existed between early AMF colonization and maize yield ( r = 0.38), and phosphorus and nitrogen uptake ( r = 0.40 and r = 0.43, respectively), demonstrating the importance of phosphorus fertilization in enhancing low-input technologies (improved fallows systems) in phosphorus deficient and acidic soils of western Kenya.
