Following the Fukushima nuclear accident in March 2011, radiocesium (rCs) contamination in deciduous trees remains over 10 years later even though the trees were leafless at the time of the accident. This phenomenon is considered to be the result of repeated retranslocation of rCs that initially penetrated the bark into the internal tissues. To implement effective measures after a possible accident in the future, it is necessary to clarify how rCs is translocated in the tree after penetration. In this study, rCs translocation was dynamically visualized using a positron-emitting tracer imaging system (PETIS) and autoradiography after the bark of apple branches was removed. The PETIS results showed the translocation of 127 Cs from the branch to young shoots and the main stem in apple trees under controlled spring growing conditions. The transport velocity of rCs in the branch was faster than that in the main stem. The transport of rCs, which was either acropetal or basipetal, in the main stem through the branch junction favored basipetal movement. Autoradiography of transverse sections of the main stem indicated that basipetal translocation was due to transport in the phloem. This study demonstrated the initial translocation responses of rCs similar to previous field research, which indicates that rCs transport to the young shoots tends to be higher under controlled conditions. We present how radioactive Cs was deposited on wheat, rice, peach tree and soil after nuclear accident in Fukushima. The deposition of radioactive Cs was found as spots at the surface of the leaves, branch or trunk of the trees, as well as in soil using one of the imaging method, autoradiography. The deposited radioactive Cs was not easily washed out, even with the treatment of acid solution. When the wheat was harvested 2 months after the accident, high radioactivity of Cs was found only on the leaves developed and expanded at the time of the accident. In the case of the rice grain, most of the radioactivity was found in bran and the radioactivity was drastically reduced in milled rice. Most of the radioactive Cs accumulation in rice plants was estimated from the absorption of the Cs ion dissolved in water, rather than Cs adsorbed in soil. [ABSTRACT FROM AUTHOR]

A nondestructive analytical method based on autoradiography and gamma spectrometry was developed to perform activity distribution analysis for particulate samples. This was applied to aerosols collected in Fukushima Japan, 40 km north of the Daiichi nuclear power plant for a 6 week period beginning shortly after the March 2011 tsunami. For an activity distribution of 990 'hot particles' from a small filter area, the hottest particle was nearly one Bq Cs but most of the activity in the filter was produced by particles having <50 mBq each. Cs/Cs activity ratios corrected to March 20, 2011 ranged from 0.68 ( u = 28 %) to 1.3 ( u = 15 %). The average ratio for a large quantity of particles was 0.92 ( u = 4 %). Virtually all activity collected was beta and not alpha, suggesting little if any direct fuel debris was present at this site and time. These findings are expected to assist with separate efforts to better understand the emission events, radionuclide transport and potential environmental or biological uptake. We analyzed fresh and dead leaves collected in forests in Fukushima after the Fukushima Daiichi Nuclear Power Plant (FDNPP) accident, using autoradiography. Both fresh and dead leaves of Cryptomeria japonica were contaminated by radionuclides (Cs and Cs). Contamination of the fresh leaves was possibly attributed to interception of radionuclides by tree canopies, whereas the dead leaves indicated the direct deposition of radionuclides by fallout and/or washout of radionuclides intercepted by tree canopies. Translocation of radiocesium from a contaminated branch to new leaves growing after the FDNPP accident was not clearly observed, although transfer of radiocesium from leaf parts to male flowers occurred. Fallen leaves of Quercus serrata, which started growing after the FDNPP accident, did not show radioactivity, indicating that significant amounts of translocation from other parts to new leaves did not occur. Fallen leaves of Q. serrata collected from a litter showed hot spots originating from direct fallout. Needles of Pinus densiflora were also contaminated by fallout. Leaching with pure water removed soluble fractions of radiocesium and hot particles from the surface of the contaminated leaves, but significant amounts of radioactivity remained. This means that foliar absorption occurred in both fresh and dead leaves. Further leaching experiments using surfactant and acetone could not remove the remaining radiocesium from the leaves. The leaching experiments indicate that radiocesium in the contaminated leaves is strongly fixed in leaf tissues and is not readily released unless leaf tissues are decomposed. [ABSTRACT FROM AUTHOR]

We measured the concentrations of cesium isotopes ( 133 Cs, 134 Cs, and 137 Cs) in zooplankton samples collected in waters off the east coast of Japan from May 2015 to June 2020. By combining these data with those obtained previously from May 2012 to February 2015, we evaluated the long-term impacts of the Fukushima Dai-ichi Nuclear Power Plant accident on marine zooplankton. Relatively high 137 Cs concentrations in zooplankton, exceeding 10 Bq/kg-dry weight, were sporadically observed until June 2016, regardless of year or station. After May-June 2017, 137 Cs concentrations decreased to below 1 Bq/kg-dry at most stations, and by May 2020, concentrations were below 0.5 Bq/kg-dry except those off Fukushima Prefecture. Since the accident, the 137 Cs/ 133 Cs atom ratios of zooplankton samples were higher than those of ambient seawater until 2019, but in May-June 2020 the ratios matched those of seawater except off Fukushima Prefecture. Highly radioactive particles were not detected in zooplankton samples by autoradiography using imaging plates after May-June 2017, although they were before. Therefore, the persistence of elevated 137 Cs/ 133 Cs ratios in zooplankton relative to seawater for nine years after the accident was probably due to the incorporation of highly radioactive particles (cesium-bearing particles or clay-mineral aggregates with highly adsorbed radiocesium) onto/into zooplankton for several years after the accident. However, since at least May-June 2017, these elevated ratios have likely been caused by small highly radioactive particles (or larger particles disaggregated into small pieces) entering the ocean from land via rivers or directly discharged from the Fukushima Nuclear Power Plant. AbstractThree cases of general paresis were successfully treated with high-dose penicillin. In all cases, cerebrospinal fluid cell counts decreased to normal and mental status improved rapidly. Cerebrospinal fluid protein concentrations decreased and IQ scores and overall levels of functioning improved gradually over 1 year. Regional cerebral blood flow (CBF) changes were analyzed longitudinally for 1 year using single photon emission computed tomography (SPECT). Autoradiography method and region of interest (ROI) analyses were used for quantitative CBF assessment and 3D stereotactic surface projections (3D-SSP) analyses were used for qualitative CBF pattern assessment. 3D-SSP could not reveal remarkable CBF pattern changes through the courses. The ROI analyses showed remarkable CBF decreases in all brain regions 1 month after the treatment, which recovered to normal levels 1 year after the treatment. These results suggest that remarkable decreases of quantitative CBF counts reflect the disappearance of encephalitis, while their gradual recovery reflects the gradual improvement of cerebral functional activity. As Treponema pallidum infection affects whole brain and CBF changes globally, quantitative CBF assessment may be more efficient than qualitative CBF pattern analyses for the purpose of understanding the pathophysiology of general paresis. Radioactive cesium concentrations in the suspended matter of the coastal waters around the Fukushima Daiichi Nuclear Power Plant (FDNPP) were investigated between January 2014 and August 2015. The concentrations of radioactive cesium in the suspended matter were two orders higher in magnitude than those determined in the sediment. In addition, we discovered highly radioactive Cs particles in the suspended matter using autoradiography. The geometrical average radioactivity of particles was estimated to be 0.6 Bq at maximum and 0.2 Bq on average. The contribution ratio of highly radioactive Cs particles to each sample ranged from 13 to 54%, and was 36% on average. A major part of the radioactive Cs concentration in the suspended matter around the FDNPP was strongly influenced by the highly radioactive particles. The subsequent resuspension of highly radioactive Cs particles has been suggested as a possible reason for the delay in radioactive Cs depuration from benthic biota., (Copyright © 2018 Elsevier Ltd. We conducted an elution experiment with contaminated soils using various aqueous reagent solutions and autoradiography measurements of contaminated bamboo shoots and shiitake mushrooms to determine the physical and chemical characteristics of radioactive Cs from the Fukushima Daiichi Nuclear Power Plant accident. Based on our study results and data in the literature, we conclude that the active Cs emitted by the accident fell to the ground as granular non-ionic materials. Therefore, they were not adsorbed or trapped by minerals in the soil, but instead physically adhere to the rough surfaces of the soil mineral particles. Granular Cs* can be transferred among media, such as soils and plants. Analysis of (137)Cs trapped in biomass in highly contaminated zones is crucial in predicting the long-term fate of (137)Cs following the explosion at the Fukushima Daiichi Nuclear Power Plant. We surveyed forest 20-50 km from the plant in July and September 2011 to evaluate (137)Cs trapped in biomass within 20 km of the plant. We determined the ambient dose rate and collected forest soils and twigs at 150 sampling points. Removability from the canopy was evaluated by washing leaves and branches with water and organic solvents. The biomass of the forest canopy was then calculated. (137)Cs fallout was simulated with an atmospheric transport model. The modeled dose rate agreed with observations (n = 24) (r = 0.62; p < 0.01). Washing experiments demonstrated that unremovable portions accounted for 53.9 ± 6.4% of (137)Cs trapped by deciduous canopy (n = 4) and 59.3 ± 13.8% of (137)Cs trapped by evergreen canopy (n = 10). In total, it was estimated that 74.5 × 10(12) Bq was trapped by canopy in the forest within the no-go zone, with 44.2 × 10(12) Bq allocated to unremovable portions, and that 0.86% of the total release was trapped in biomass as of September 2011.

Irradiation from internally deposited radionuclides induces malignant tumors. Ingested radionuclides accumulate in specific organs, which are irradiated over a lifelong period. Our aim is to elucidate why the development of malignant tumors requires long-term internal exposure, on the order of decades, despite the fact that irradiation is continuous over this period. Three major factors are considered to be responsible for the long incubation time in carcinogenesis caused by internally deposited alpha-emitters: uneven distribution of radionuclides, limited range of irradiation, and dynamic movement of tumor precursor cells. We hypothesized that target cells susceptible to malignant transformation may undergo one event by alpha particles and may then migrate outside of the range of alpha particles, thereby avoiding immediate induction of successive additional events that would lead to cell death or neoplastic changes. Based on this hypothesis, we further proposed a mathematical model to predict the relationship between dose rate and incubation period of tumors induced by internally deposited alpha-emitters. The function was non-linear and included terms of both direct and indirect radiation effects. It well fitted both human Th-ICC cases and rat Pu-induced lung cancer, suggesting that indirect radiation effects are independent from dose rate. The significance of parameters of the model is discussed.

Functional Bacteria and Archaea community structures of a full-scale anaerobic sludge digester were investigated by using a full-cycle 16S rRNA approach followed by microautoradiography (MAR)-fluorescent in situ hybridization (FISH) technique and micromanipulation. FISH analysis with a comprehensive set of 16S and 23S rRNA-targeted oligonucleotide probes based on 16S rRNA clone libraries revealed that the Gram-positive bacteria represented by probe HGC69A-hybridized Actinobacteria (8.5+/-1.4% of total 4', 6-diamidino-2-phenylindole (DAPI)-stained cells) and probe LGC354-hybridized Firmicutes (3.8+/-0.8%) were the major phylogenetic bacterial phyla, followed by Bacteroidetes (4.0+/-1.2%) and Chloroflexi (3.7+/-0.8%). The probe MX825-hybridized Methanosaeta (7.6+/-0.8%) was the most abundant archaeal group, followed by Methanomicrobiales (2.8+/-0.6%) and Methanobacteriaceae (2.7+/-0.4%). The functional community structures (diversity and relative abundance) of major trophic groups were quantitatively analyzed by MAR-FISH. The results revealed that glucose-degrading microbial community had higher abundance (ca. 10.6+/-4.9% of total DAPI-stained cells) and diversity (at least seven phylogenetic groups) as compared with fatty acid-utilizing microbial communities, which were more specialized to a few phylogenetic groups. Despite the dominance of Betaproteobacteria, members of Chloroflexi, Smithella, Syntrophomonas and Methanosaeta groups dominated the [(14)C]glucose-, [(14)C]propionate-, [(14)C]butyrate- and [(14)C]acetate-utilizing microorganism community, and accounted for 27.7+/-4.3%, 29.6+/-7.0%, 34.5+/-7.6% and 18.2+/-9.5%, respectively. In spite of low abundance (ca. 1%), the hitherto unknown metabolic functions of Spirochaeta and candidate phylum of TM7 as well as Synergistes were found to be glucose and acetate utilization, respectively.

Genetic diversities of the nuclear and mitochondrial genomes in Pinus parviflora were studied in 16 populations, which were distributed across most of the species' range in Japan. Six mitochondrial DNA haplotypes were identified among the 16 populations. The intrapopulation diversity of allozymes was similar to that of other endemic woody species (H(S)=0.259). Although P. parviflora is distributed in discrete populations, differentiation between these was very low (G(ST)=0.044). In addition, the extent of genetic differentiation between two varieties (var. pentaphylla and var. parviflora) was extremely low (G(VT)=0.001). Intrapopulation diversity of mitochondrial DNA was also very low (H(S)=0.098), but population differentiation was high (G(ST)=0.863). Moreover, the distribution of haplotypes reflected the taxonomic differences between P. parviflora var. pentaphylla and var. parviflora. The populations of var. pentaphylla and var. parviflora contained different haplotypes. Differing modes of inheritance may account for the differences in nuclear and mitochondrial genetic diversity.

We carried out screening for mutations in the GH-1 gene in 29 sporadic Japanese subjects with severe Isolated Growth Hormone Deficiency (IGHD) by dideoxy fingerprinting (ddF). Three of 29 (approximately 10%) were heterozygous for each of the following GH-1 gene mutations including: 1) an G-->A transition in the third codon of the GH-1 signal peptide of exon 1 resulting in a Threonine to Alanine substitution, 2) a G-->A transition in the first base of the donor splice site of IVS 3 (+1G-->A) and 3) a G-->A transition in the 183rd codon of the GH-1 mature peptide of exon 5 resulting in an Arginine to Histidine substitution. One of three was heterozygous for both mutations of 1) and 2). The IVS 3 (+1G-->A) mutation has been previously reported in affected individuals from three unrelated families with IGHD type II (autosomal dominant form). This mutation destroys the GH IVS 3 donor splice site, causing skipping of exon 3 and loss of the codons for amino acids 32-71 of the mature GH peptide. Our findings indicate that 1) ddF screening of genomic DNAs provides a practical tool to detect GH gene mutations and 2) some sporadic cases of IGHD may be caused by GH gene alternations.

Recent studies have shown both association and linkage between a Gly40-Ser mutation in the glucagon receptor gene and NIDDM in French patients with familial NIDDM. This mutation was present in heterozygous form in 4.6% of diabetic probands but only 1% of the French population, suggesting that it was an important risk factor in the development of NIDDM. A total of 348 unrelated Japanese subjects (220 with NIDDM, 53 with impaired glucose tolerance (IGT) and 75 normal subjects) were screened for the presence of the Gly40-Ser mutation. Seventy-two percent of the NIDDM patients and 52% of IGT subjects had a positive family history of NIDDM. The Gly40-Ser mutation, which could be readily detected in a positive control subject, was not found in any of the 348 Japanese subjects studied. Thus, the Gly40-Ser mutation does not play an important role in the pathogenesis of NIDDM in Japanese patients.

The first Japanese patient with mitochondrial trifunctional protein deficiency has been identified. The patient's alpha and beta-subunits were synthesized, transported into the mitochondria, and converted to the mature size, but rapidly disappeared. The newly synthesized mature alpha and beta-subunits in the control cells were incorporated into the enzyme complex, alpha4beta4, whereas those in the patient's cells were present as monomers. We propose that formation of the enzyme complex is required for stabilization of trifunctional protein.

Study Design: The present study analyzed the restriction fragment length polymorphism patterns of alpha 2(XI) collagen, bone morphogenetic protein-2, alkaline phosphatase, and tumor necrosis factor-alpha genes in patients with ossification of the posterior longitudinal ligament. This study investigates the genetic polymorphism of bone-induced factors in patients with ossification of the posterior longitudinal ligament and compares it with healthy control subjects., Objectives: To clarify the genetic markers linked to ossification of the posterior longitudinal ligament., Summary of Background Data: Ossification of the posterior longitudinal ligament is a genetic disease associated with abnormal calcium metabolism involving the posterior longitudinal ligament. Previous genetic studies have not identified the pathologic mechanism of ossification of the posterior longitudinal ligament. Histopathologic studies of ossification of the posterior longitudinal ligament and the animal model, the spinal hyperostotic mouse, have revealed an increase in Type XI collagen and bone morphogenetic protein-2 expression., Methods: Eighteen Japanese patients with ossification of the posterior longitudinal ligament and 51 healthy, unrelated control subjects were investigated for the restriction fragment length polymorphism patterns of COL11A2, bone morphogenetic protein-2, alkaline phosphatase, and tumor necrosis factor-alpha, genes with various restriction endonucleases., Results: The gene frequencies of COL11A2 obtained with BamHl (10.0 kb fragment) and HindIII (19.0 kb fragment) observed in patients with ossification of the posterior longitudinal ligament were higher compared with control subjects (0.43 and 0.14, respectively). These differences were statistically significant (BamHl P = 0.018; Hindlll P = 0.046). Two new restriction fragment length polymorphism patterns were detected of the bone morphogenetic protein-2 gene with Mspl and Taql and one already known restriction fragment length polymorphism pattern of the tumor necrosis factor-alpha gene with Ncol. However, they were not significantly different from the control subjects., Conclusions: Seven restriction fragment length polymorphisms of COL11A2 gene were identified. Two of them (BamHl, 10.0/10.0 kb genotype; HindIII, 19.0/19.0 kb genotype) were significantly different in patients with ossification of the posterior longitudinal ligament.

Since abnormalities of brain development play a role in the aetiology of schizophrenia, growth factors, known to play a role in neurodevelopment, such as neurotrophin-3 (NT-3), are therefore candidate genes for this disorder. The A3/147 bp allele of a dinucleotide repeat polymorphism in the promoter region of the NT-3 gene has been reported as occurring more frequently in a sample of Japanese schizophrenics compared to controls. We have determined the frequency of alleles of this polymorphism in 175 Caucasian schizophrenic patients and 147 control subjects. The patient and control samples showed no significant deviation from Hardy-Weinberg equilibrium and, in a test of allalleles, the patients and controls did not differ significantly in allele frequencies. However, the male schizophrenics were more likely than male controls to have the A3/147 bp allele (P = 0.029).

The relationship between the genotypes of Xba I and Bam H I restriction fragment length polymorphisms (RFLPs) at a gene for debrisoquine hydroxylase (CYP2D6) and phenotypes of the metabolic function of debrisoquine/sparteine, the EM and the PM in a healthy Japanese population was investigated. The genotypes of Xba I 11.5 kb and Xba I 44 kb-Bam H I 2.3 kb- were responsible for the Japanese PM. Genotype of Xba I RFLP at CYP2D6 locus was analyzed in 43 healthy individuals and 51 patients with IDP. The relative risk of IDP was 2.15 times more for individuals with the Xba I 44 kb allele compared to those without the allele (chi 2 = 4.149, d.f. = 1, p < 0.05) and it was 6.32 times greater for the Xba I 44 kb homozygotes than the Xba I 29 kb homozygotes (chi 2 = 4.935, d.f. = 1, p < 0.05). These data suggest that the PM for debrisoquine/sparteine hydroxylase might be one of the genetic factors making humans susceptible to IDP acquisition.

Twenty four cloned isolates of Aujeszky's disease virus collected from outbreaks of Aujeszky's disease from 1981 through 1989 in Japan were characterized by their restriction endonuclease (RE) cleavage patterns, virulence for mice and thymidine kinase (TK) activity. All of the isolates belonged to Type II of the four types classified by Herrmann et al. (1984). Based on the number and migration rate of the restriction fragments, the isolates were divided into 7 groups with Bam HI, 9 groups with Kpn I, 3 groups with BstE II and 2 groups with Sal I. The results indicate that the RE analysis, especially with Bam HI and Kpn I, provides useful epidemiological information about field isolates of Aujeszky's disease virus. All of the isolates showed virulence for mice ranging from 6.9 to 63.0 (PFU/LD50). It was interesting that the Nagano S87 strain, which had the highest virulence for the mouse, showed unique RE cleavage patterns with four enzymes. On the other hand, ara-T-resistant, TK-negative strain, was avirulent for mice (greater than 10(6.4) PFU/LD50). All of the isolates investigated in this study showed TK activity by the thymidine plaque autoradiography.

A radiograph of the incorporation of 3H-phenylalanine and 3H-tyrosine into cultured lymphocytes in a phenylketonuria patient indicates phenylalanine hydroxylase deficiency in the peripheral lymphocytes. Cultured lymphocytes from the patient showed much less uptake of 3H-phenylalanine and a slightly higher incorporation of 3H-tyrosine than the control. It is possible that a lack of phenylalanine hydroxylase in the peripheral lymphocytes of PKU patients prevents oxidation of phenylalanine to tyrosine.

A gene product (p42) of the long open reading frame, now termed tax, of the viral genome of human T-cell leukemia virus type I (HTLV-I) may be related to the transformation of T cells in adult T-cell leukemia-lymphoma (ATLL). To evaluate its association with the disease, we compared the prevalence of antibody to p42 in sera obtained from 105 HTLV-I carriers and 64 ATLL patients from southwest Japan. The prevalence of the anti-p42 antibody reactivity was 63% among carriers and 31% among cases. The cases were more than 3 times as likely to lack antibody to p42 than carriers, the relative odds (OR) = 3.4, p = 0.001. When the samples were tested for antibody against p24, the most immunogenic core protein, the prevalence was somewhat higher among carriers (65%) than in cases (52%), but not significantly so (p = 0.15). Among the healthy carriers, the correlation between the prevalence of both antibodies was high (p = 0.001), and only 25% of those who had antibody to p24 lacked antibody to p42. However, among the cases, reactivity to both antigens was independent (p = 0.52), and 65% of those with antibody to p24 lacked antibody to p42, OR = 6.3, p = 0.0004. Thus the strongest serologic marker of ATLL following diagnosis was lack of reactivity to p42, particularly among those subjects with anti-p24. Whether this altered response is present prior to disease remains to be determined.

A DNA sequence polymorphism, revealed by digestion of genomic DNA with the endonuclease Xba1 and hybridisation with a complementary DNA clone for a human glucose transporter, yields two alleles (sizes 6.2 kbp, the X1 allele; or 5.9 kbp, the X2 allele). The genotype frequencies were investigated in three non-insulin-dependent diabetic populations. The frequencies (%) of X1.X1, X1.X2, and X2.X2 were 13, 51, and 36 among 89 North European diabetic subjects, and 8, 38, 54 among their 104 controls (chi 2 test p less than 0.02; G-test p less than 0.02). For 53 South European diabetic patients the frequencies were 19, 50, 31, and for their 41 controls they were 2, 58, 40 (chi 2 test p less than 0.02; G-test p less than 0.01). The corresponding figures were 6, 55, 39 for 45 Japanese patients and 0, 28, 72 for a further 49 controls (chi 2 test p less than 0.01; G-test p less than 0.001). The occurrence of the association of the X1 allele with diabetes in three separate populations suggests that the polymorphic site may be close to a diabetogenic locus on chromosome 1.

Diagnosis of familial amyloid polyneuropathy (FAP) was investigated by recombinant DNA techniques using Southern blot hybridization in 49 constituents of FAP pedigrees originated from Nagano and Hiroshima Prefectures in Japan. The results were correlated with clinical features of the patients. Twenty-nine patients originated from Ogawa Village in Nagano Prefecture of the central part of Japan had typical clinical features of Andrade type FAP including polyneuropathy with autonomic disorders. The variant prealbumin gene with a substitution of methionine (Met) for valine (Val) at position 30 was detected in their DNA analysis (positive DNA diagnosis). In 5 cases from Nagano and Hiroshima Prefectures who had clinical features of Andrade type FAP but no family history, DNA diagnosis was positive. In another 5 members of FAP pedigrees originated from Ogawa Village without clinical features of FAP, DNA diagnosis was positive. Two cases originated from Iiyama City in Nagano, who had central nervous system disorders resembling spinocerebellar degeneration with spastic paraplegia besides clinical features of Andrade type FAP, had the Val-Met interchange variant prealbumin gene. In a case from the same Iiyama family who had only the central nervous system disorders without clinical features of FAP, DNA diagnosis was negative. These findings indicate that the presence of the variant prealbumin gene closely linked to the clinical manifestations of Andrade type FAP. The diagnostic DNA analysis was considered to be useful for the diagnosis of Andrade type FAP for sporadic cases as well as so far nonsymptomatic gene carriers. In conclusion, FAP patients originated from Iiyama City and Ogawa village were considered to have the same mutation despite the variances in clinical features.
