Background: Pigs are susceptible to several ruminant pathogens, including Coxiella burnetti, Schmallenberg virus (SBV) and bovine viral diarrhea virus (BVDV). These pathogens have already been described in the pig population, although the dynamics of the infection and the impact on pig farms are currently unclear. The aim of this work was to evaluate the presence of these infections in the pig population of the Campania region, southern Italy, and to evaluate the risk factors associated with a greater risk of exposure. Results: A total of 414 serum samples belonging to 32 herds were tested for the presence of antibodies against SBV, Coxiella, and BVD using commercial multispecies ELISA kits. SBV (5.3%) was the most prevalent pathogen, followed by Coxiella (4.1%) and BVD (3%). The risk factors included in the study (age, sex, province, farming system, ruminant density and major ruminant species) had no influence on the probability of being exposed to BVD and Coxiella, except for the location, in fact more pigs seropositive to Coxiella were found in the province of Caserta. However, the univariate analysis highlighted the influence of age, location, and sex on exposure to SBV. The subsequent multivariate analysis statistically confirmed the importance of these factors. The presence of neutralizing antibodies for SBV and BVDV, or antibodies directed towards a specific phase of infection for Coxiella was further confirmed with virus-neutralization assays and phase-specific ELISAs in a large proportion of positive samples. The presence of high neutralizing antibody titers (especially for SBV) could indicate recent exposures. Twelve of the 17 positive samples tested positive for antibodies against Coxiella phase I or II antigens, indicating the presence of both acute and chronic infections (one animal tested positive for both phases antibodies). Conclusions: Our study indicates a non-negligible exposure of pigs from southern Italy to the above pathogens. Further studies are necessary to fully understand the dynamics of these infections in pigs, the impact on productivity, and the public health consequences in the case of Coxiella. Simple Summary: African swine fever (ASF) is a highly lethal viral disease affecting suids and caused by the African Swine Fever Virus (ASFV). ASF was described for the first time in 1921 in Kenya. The ASF genotype I virus was introduced to Europe in 1957, marking the onset of the first European wave. In 2007, ASFV genotype II was detected in Georgia, affecting domestic pigs and wild boars, and it later spread to several European and extra-European countries, including Italy. This report focuses on the strategic challenges encountered in the attempt to eradicate ASFV amongst domestic pigs in the Lombardy region. The joint efforts that were implemented facilitated the eradication of ASF in domestic pigs in just over 1.5 months, representing an example of effective and timely cooperation to mitigate both the spread of the infection and the economic repercussions for the Italian and global pig industries. Two months later, it was diagnosed in a commercial pig farm as a consequence of the disease's spread amongst wild boars and an increase in the viral environmental load. This report aims to describe the features of ASFV domestic pig outbreaks that occurred in the Zinasco municipality (Lombardy) and the joint efforts to mitigate potential direct and indirect economic impacts on the Italian and global pig industry. The epidemiological investigation and the measures implemented, which were all performed according to national and European regulations, as well as exceptional ad hoc measures aimed at protecting the pig industry, are described in order to provide a practical and effective approach to combating ASF. Monitoring disease among wildlife is critical to preserving health in both domestic animals and wildlife, and it becomes much more critical when the diseases cause significant economic damage to the livestock industry or threaten public health. Given the continuous increase in populations and its role as a reservoir for several infections, wild boar (Sus scrofa) requires special attention regarding disease surveillance and monitoring. In this study, we investigated the molecular prevalence of selected pathogens in the wild boar population of Campania, southern Italy. The prevalence of pathogens causing reproductive problems in pigs (Sus domesticus), including porcine parvovirus (PPV), porcine circovirus types 2 and 3 (PCV-2 and PCV-3), pseudorabies virus (PRV), Coxiella burnetii, and Brucella suis, was evaluated by testing the reproductive organs collected from 63 wild boars with polymerase chain reaction. The most common pathogens were PPV (44.4%) and two porcine circoviruses (14.3%). PRV and C. burnetii, on the other hand, showed a significantly lower prevalence (1.6%). No reproductive organs tested were positive for B. suis. Risk factor analysis revealed a correlation between age and PCV-2 positivity, with animals less than 12 months old having significantly higher prevalence rates. Our findings suggest that wild boars hunted in the Campania region harbour several infections potentially transmissible to other mammals' reproductive tracts. Furthermore, our results emphasized the importance of strict adherence to biosecurity protocols on domestic swine farms, especially on free-range farms, to avoid interactions between domestic and wild animals. The presence of carbapenem–resistant bacteria and carbapenem resistance genes (CRGs) in livestock is increasing. To evaluate the presence of carbapenemase-producing Enterobacteriaceae (CPE) and the main CRGs along swine food chains of the Marche Region (Central Italy), samples of faeces, feed, and animal-food derived products were collected from seven small/medium, medium, and large-scale pig farms. A total of 191 samples were analysed using a culture-dependent method, with the aim of isolating CPE. Isolates were analysed for their resistance to carbapenems using a modified Hodge test and the microdilution method for the minimum inhibitory concentration (MIC) determination. Moreover, the extraction of microbial DNA from each sample was performed to directly detect selected CRGs via qPCR. Among the 164 presumptive resistant isolates, only one strain from a liver sample, identified as Aeromonas veronii, had an ertapenem MIC of 256 μg/mL and carried a carbapenemase- (cphA) and a β-lactamase- (blaOXA-12) encoding genes. A low incidence of CRGs was found; only nine and four faecal samples tested positive for blaNDM-1 and blaOXA-48, respectively. Overall, the importance of monitoring CPE and CRGs in livestock and their food chains should be stressed to control all potential non-human CPE and CRGs reservoirs and to determine safety levels for human health. Rotavirus H (RVH) has been detected in humans, pigs and bats. Recently, RVH infections were reported in different porcine farms worldwide, suggesting epidemiological relevance. However, to date, the genome information of RVH strains has been limited due to the scarcity of deposited sequences. This study aimed to characterize the VP7, VP4, VP6 and NSP4 genes of RVHs from 27 symptomatic pigs, in Italy, between 2017 and 2021. RVH genes were amplified via RT-PCR using specific primers, and the amplicons were sequenced. By coupling the data generated in this study with the sequences available in the databases, we elaborated a classification scheme useful to genotype the VP7, VP4, VP6 and NSP4 genes. The nucleotide identity and phylogenetic analyses unveiled an impressive genetic heterogeneity and allowed the classification of the Italian RVH strains into 12G (VP7), 6P (VP4), 8I (VP6) and 8E (NSP4) genotypes, of which 6I, 5E and the totality of the G and P genotypes were of novel identification. Our data highlight the high genetic variability of the RVH strains circulating in pigs and underline the importance of a robust classification system to track the epidemiology of RVHs. African swine fever (ASF) is a devastating infectious disease of domestic pigs and wild boar that is spreading quickly around the world and causing huge economic losses. Although the development of effective vaccines is currently being attempted by several labs, the absence of globally recognized licensed vaccines makes disease prevention and early detection even more crucial. ASF has spread across many countries in Europe and about two years ago affected the Italian susceptible population. In Italy, the first case of ASF genotype II in wild boar dates back to January 2022, while the first outbreak in a domestic pig farm was notified in August 2023. Currently, four clusters of infection are still ongoing in northern (Piedmont-Liguria and Lombardy), central (Lazio), and southern Italy (Calabria and Campania). In early September 2023, the first case of ASFV genotype II was detected in a domestic pig farm in Sardinia, historically affected by genotype I and in the final stage of eradication. Genomic characterization of p72, p54, and I73R/I329L genome regions revealed 100% similarity to those obtained from isolates that have been circulating in mainland Italy since January 2022 and also with international strains. The outbreak was detected and confirmed due to the passive surveillance plan on domestic pig farms put in place to provide evidence on genotype I's absence. Porcine Circovirus type 2 (PCV2) is the etiological agent of a disease syndrome named Porcine Circovirus disease (PCVD), representing an important threat for the pig industry. The increasing international trade of live animals and the development of intensive pig farming seem to have sustained the spreading of PCVD on a global scale. Recent classification criteria allowed the identification of nine different PCV2 genotypes (PCV2a–i). PCV2a was the first genotype detected with the highest frequency from the late 1990s to 2000, which was then superseded by PCV2b (first genotype shift). An ongoing genotype shift is now determining increasing prevalence rates of PCV2d, in replacement of PCV2b. In Italy, a complete genotype replacement was not evidenced yet. The present study was carried out on 369 samples originating from domestic pigs, free-ranging pigs, and wild boars collected in Sardinia between 2020 and 2022, with the aim to update the last survey performed on samples collected during 2009–2013. Fifty-seven complete ORF2 sequences were obtained, and the phylogenetic and network analyses evidenced that 56 out of 57 strains belong to the PCV2d genotype and only one strain to PCV2b, thus showing the occurrence of a genotype shift from PCV2b to PCV2d in Sardinia. Hepatitis E virus (HEV) is an important cause of acute viral hepatitis in humans worldwide. The food-borne transmission of HEV appears to be a major route in Europe through the consumption of pork and wild boar meat. HEV epidemiology in wild boars has been investigated mainly in Northern and Central Italian regions, whilst information from Southern Italy is limited. We investigated the occurrence of HEV in wild boar in the Apulia and Basilicata regions (Southern Italy). Thirteen (10.4%) out of one hundred and twenty-five wild boar samples tested positive for HEV using a quantitative reverse transcription PCR. HEV prevalence was 12% in Apulia and 9.3% in Basilicata. Seven samples were genotyped, and different subtypes (c, f, m) of genotype 3 were identified. The complete genome of a 3m strain was determined, and the virus showed the highest nucleotide identity to a human HEV strain identified in France in 2017. These findings demonstrate the substantial circulation of HEV in the wild boar population in Italian Southern regions. Gathering information on the HEV strains circulating in different geographical areas is useful for tracking the origin of HEV outbreaks and assessing the epidemiological role of wild boar as a potential virus reservoir for domestic pigs. Simple Summary: Porcine Reproductive and Respiratory Syndrome (PRRS) is an infectious disease of viral etiology characteristic of the swine species. Although almost three decades have passed since its emergence, control of the disease still presents significant issues, representing a source of concern for veterinarians and breeders. In addition to management difficulties, the PRRS virus (PRRSV) causes severe economic losses in terms of abortions, a decrease in animal growth, increased mortality, and the massive use of drugs. Understanding the genetic markers involved in the response to the infection is challenging and crucial and represents the main goal of this study. CD163, which encodes the membrane receptor used by the PRRSV to enter macrophages and initiate infection, has been identified as one of the most promising marker genes associated with genetic susceptibility to the disease. In this study, detection by sequencing of the more significant polymorphisms on the CD163 gene was conducted for the first time on 377 pigs reared in different farms distributed in some areas of Central Italy. The genotyping data obtained in this work, together with the assessment of the virological status of the animals and the comparison with the findings from other PRRSV conditioned and experimental infection trials, will allow a better understanding of whether some Italian pig populations can represent a good genetic resource and a reservoir of resistance/"resilience" markers to PRRS. Indeed, marker-assisted selection (MAS) could represent an alternative and a more valid tool than vaccination to control the spread of this impactful disease. Porcine Reproductive and Respiratory Syndrome (PRRS) caused by the PRRS virus affects farmed pigs worldwide, causing direct and indirect losses. The most severe manifestations of PRRS infection are observed in piglets and pregnant sows. The clinical outcome of the infection depends on the PRRSV strain's virulence, the pregnancy state of the female, environmental factors, the presence of protective antibodies due to previous infections, and the host's genetic susceptibility. The latter aspect was investigated in this study, in particular, evaluating the most significant polymorphisms (SNPs) of the CD163 gene in slaughtered pigs reared in Central Italy. Total RNAs were extracted from 377 swine samples and subjected to RT-PCR targeted to the CD163 gene, followed by sequencing analysis. Contextually, the viral RNA was detected by RT-qPCR in order to phenotypically categorize animals into infected and not infected. In particular, 36 haplotypes were found, and their frequencies ranged from 0.13% to 35.15%. There were 62 resulting genotypes, three of which were associated with a putative resistance to the disease. Both the haplotypes and genotypes were inferred by PHASE v.2.1 software. To the best of our knowledge, this type of investigation was conducted for the first time on pig livestock distributed in different regions of Central Italy. Thus, the obtained findings may be considered very important since they add useful information about swine genetic background in relation to PRRS infection, from the perspective of adopting Marker-Assisted Selection (MAS) as a possible and alternative strategy to control this still widespread disease. Simple Summary: Lung lesions, which are common findings in finisher pigs, are considered useful indicators of animal health and welfare at the slaughterhouse. Several methods for evaluating such lesions have been developed, and selecting the best system is critical for obtaining reliable and consistent data. In this view, the aim of this study was to compare two well-known scoring methods (Madec and Blaha methods) using data on lung lesions collected by two operators in an Italian high-throughput abattoir under routine slaughter conditions. Overall, there was a satisfactory level of agreement between the two methods, although a discrepancy in accurately recognizing healthy lungs and minor injuries between the two systems has been observed. According to our results, the Blaha method is a visual-only system applicable to very large abattoirs. It can be easily used to provide an overview of the respiratory health status of pigs, although it can yield a higher percentage of false negative results. On the other hand, the Madec method can provide more detailed results but would be more challenging to use for continuous monitoring in large abattoirs because it requires both visual and palpation assessments. Respiratory diseases continue to pose significant challenges in pig production, and the assessment of lung lesions at the abattoir can provide valuable data for epidemiological investigations and disease surveillance. The evaluation of lung lesions at slaughter is a relatively simple, fast, and straightforward process but variations arising from different abattoirs, observers, and scoring methods can introduce uncertainty; moreover, the presence of multiple scoring systems complicates the comparisons of different studies, and currently, there are limited studies that compare these systems among each other. The objective of this study was to compare validated, simplified, and standardized schemes for assessing surface-related lung lesions in slaughtered pigs and analyze their reliability under field conditions. This study was conducted in a high-throughput abattoir in Italy, where two different scoring methods (Madec and Blaha) were benchmarked using 637 plucks. Statistical analysis revealed a good agreement between the two methods when severe or medium lesions were observed; however, their ability to accurately identify healthy lungs and minor injuries diverged significantly. These findings demonstrate that the Blaha method is more suitable for routine surveillance of swine respiratory diseases, whereas the Madec method can give more detailed and reliable results for the respiratory and welfare status of the animals at the farm level. Simple Summary: Reducing the sources of stress on farms allows for enhanced animal welfare and productivity. Aerial contaminants and pollutants that can be found in indoor animal houses are among these stressors. In Italy, the guidelines to assess animal welfare in pig farming are displayed in a protocol named "ClassyFarm", based on European legislation. Specific indications are given on the microclimatic conditions of livestock indoor environments (temperature, relative humidity, dustiness) and air quality, especially regarding harmful gases such as ammonia (NH3) and carbon dioxide (CO2). Nevertheless, the recommended measurement techniques for dust and harmful gases are not satisfactory. In this review, the effects of these pollutants that have been studied in the scientific literature are reported, as well as some tips that can help in carrying out proper measurements. The presence of harmful bioaerosol on farms cannot be neglected since it affects not only breeding animals' well-being but also the health of people living in the surrounding area. On 24 September 2019, the Ministry of Health issued an explanatory circular containing clarifications on the implementation methods of the National Improvement Plan for the application of Legislative Decree 122/2011. The Plan states that "In all farms where weaning or fattening pigs are raised and in breeding farms which wean piglets (excluding those for self-consumption), a risk assessment is carried out by the veterinarian on the basis of three levels: insufficient, room for improvement and optimal". ClassyFarm, a risk assessment tool for livestock farming, is applied in Italy to evaluate the level of welfare and management of animals from a variety of points of view. Essentially, the categorization risk introduced by ClassyFarm in pig farming depended on the obligation stated by the EU in Decree 122/2011 to avoid tail docking in piglets and, at the same time, to reduce the stressor aspects able to induce aggressive behavior among pigs, improving the welfare and health status of animals. Since ClassyFarm evaluates many aspects of the management of animal farming, our aims in this review are to discuss the topic from an environmental point of view: (1) to frame the indications of ClassyFarm to make a farm risk assessment based on pigs' welfare; (2) to review environmental quality assessment in pig farms, and its repercussions on animal health and welfare; (3) to describe the most used sampling techniques of air pollutants measurements. Simple Summary: Maternal behaviour is a common trait in many mammalian species, and mothers can invest a lot of energy in parental care, ensuring a higher probability of survival for their infants. An interesting question is whether these behaviours are influenced by particular mechanisms and how the offspring can effectively attract the mother's attention. In order to try to answer this question, we studied a group of sows reared in northern Italy (ethical farm Parva Domus, Cavagnolo, Turin) in which lactating and non-lactating females were present. Isolation calls emitted by the piglets were able to generate a state of anxiety in the mothers and can be therefore a valid example of a useful mechanism to request maternal care effectively. Furthermore, we not only observed that lactating females responded and reacted more to the vocalisations of piglets from other broods, but also that less aggressive mothers responded more. Finally, we found that certain vocalisation characteristics may influence the type of response by the mothers. Therefore, several factors seem to play a key role in eliciting response behaviour in sows. Domestic pigs (Sus scrofa) possess complex socio-cognitive skills, and sows show high inter-individual variability in maternal behaviour. To evaluate how females—reared under natural conditions—react to the isolation calls of their own piglets or those of other females, we conducted observations and experimental trials. In January–February 2021, we conducted all-occurrences sampling on affiliation, aggression, and lactation (daily, 7:30–16:30 h) on six lactating and four non-lactating females at the ethical farm Parva Domus (Turin, Italy). The trials (30 s each, n = 37/sow) consisted of briefly catching and restraining a piglet. We recorded the sow response (none/reactive/proactive movement towards the piglet; self-directed anxiety behaviours such as body shaking) before and during the trial and under control conditions. Increased levels of anxiety behaviour in sows were accompanied by an increased frequency of responses. Less aggressive sows and lactating sows showed the highest frequencies of response. Finally, the isolation calls' maximum intensity had an influence on the type of response observed, with higher proactive response frequencies following lower intensity isolation calls. Our results suggest that being under lactation could play a key role in increasing sow response levels and that specific acoustic features may influence the response. [ABSTRACT FROM AUTHOR]

Objectives To investigate the optrA -carrying genetic elements and their transferability in two linezolid-resistant Streptococcus dysgalactiae subsp. equisimilis (SDSE) strains of swine origin. Methods SDSE strains (V220 and V1524) were phenotypically and genotypically characterized. Transferability of oxazolidinone resistance genes (filter mating), genetic elements and relatedness between isolates (WGS) were analysed. Excision of the genetic elements was assayed by inverse PCR. Results SDSE isolates were resistant to chloramphenicol, florfenicol and linezolid, but susceptible to tedizolid and both carried the optrA gene. In SDSE V220 optrA was located on a 72.9-kb ICE Sdy V220 inserted in the 3′ end of the chromosomal rum gene. It was 94%–96% identical (coverage, from 31% to 61%) to other optrA -carrying ICEs. In-depth ICE Sdy V220 sequence analysis revealed that optrA was carried by an IME Sdy V220 (17.9 kb), also containing the tet (O/W/32/O) gene. Inverse PCR assays excluded the ICE Sdy V220 mobility. In SDSE V1524, optrA was carried by the Φ Sdy V1524 prophage, integrated near the 5′ end of the chromosomal had gene, showing a genetic organization similar to that of other streptococcal phage. Conjugation and transduction assays failed to demonstrate the optrA transferability to streptococcal recipients. V220 and V1524 belonged to two novel sequence types (ST704 and ST634, respectively). Conclusions To the best of our knowledge, this is the first identification of the optrA gene on a prophage and an ICE in SDSE isolates from swine brain. These findings are consistent with the current belief in the key role of bacteriophages and ICEs in the streptococcal evolution and adaptation. [ABSTRACT FROM AUTHOR]

The recovery of Nero Lucano (NL) pigs in the Basilicata region (Southern Italy) started in 2001 with the collaboration of several public authorities in order to preserve native breeds that can play a significant economic role both due to their remarkable ability to adapt to difficult environments and the value of typical products from their area of origin. In this study, by using the Illumina Porcine SNP60 BeadChip, we compared the genetic structures of NL pigs reared in a single farm in two different periods separated by a time interval corresponding to at least three generations. The results showed an increase in the percentage of polymorphic loci, a decrease in the inbreeding coefficient calculated according to ROH genome coverage (FROH), a reduction in the number of ROH longer than 16 Mb and an increase in ROH with a length between 2 and 4 Mb, highlighting a picture of improved genetic variability. In addition, ROH island analysis in the two groups allowed us to identify five conserved regions, located on chromosomes 1, 4, 8, 14 and 15, containing genes involved in biological processes affecting immune response, reproduction and production traits. Only the conserved ROH island on chromosome 14 contains markers which, according to the literature, are associated with QTLs affecting thoracic vertebra number, teat number, gestation length, age at puberty and mean platelet volume. Simple Summary: Little is known about the interdependencies between livestock production and biodiversity. The swine sector is characterized by a diversity of extensive and intensive production practices, which rely on a wide range of natural and manmade activities (e.g., different use of the land) that can reduce its adverse effects on biodiversity. The Nero d'Abruzzo pig is a black domestic pig from the Abruzzo region, Italy, that is reared with extensive practice. This study using a proteomics approach aimed to investigate breed differences between the autochthon Nero d'Abruzzo and commercial-hybrid pigs commonly used by the industry. The comparison of the protein profile of muscle exudate obtained following centrifugation from the diaphragm of the two pig breeds highlight significant differences in the protein expression of a large pool of proteins. Among the proteins that were upregulated or downregulated differently in the two pigs, three—fatty acid synthase, catalase and glutathione peroxidase—may contribute in part to explaining, according to their functions, the phenotypic differences between the pigs. This work has identified a cohort of proteins that can be potentially useful to define new molecular phenotypes for novel applications in pig breeding programs and are of great interest for future work. Capital-driven animal husbandry systems undertaken in the last century led to the abandoning of many pig breeds that were not profitable. These local pig breeds and their respective production systems have great potential as they are able to respond to the high criteria and needs of modern society concerning some environmental aspects, animal-welfare, healthiness, etc. This is the case of the black pigs of Italy. The Apulo-Calabrese is a breed of black pig, known by many other names such as Nero d'Abruzzo. In order to further understand the biological differences between different types of porcine genetics (Nero d'Abruzzo and commercial-hybrid) we used a label-free LC-MS strategy and Western-blot to characterize the proteomes of muscle-exudate collected from these pigs. This proteomics approach identified 1669 proteins of which 100 changed significantly in abundance between breeds. Bioinformatics functional analysis indicated that differentially expressed proteins were involved in several biological processes related to energy-metabolism and response to oxidative stress, suggesting that these functions might distinguish between these pigs. Fatty-acid synthase, catalase and glutathione-peroxidase, involved in enzymatic activity were found to be more represented in samples obtained from the Nero d'Abruzzo. Simple Summary: In swine, the diagnosis of enteric diseases is challenging due to simultaneous presence of one or more microbic agents sharing similar clinical signs and pathological lesions. Therefore, the study of microscopic lesions is crucial in establishing the role of pathogens as causative agents in cases of co-infections; this can be confirmed by demonstrating agent–lesion co-localization. Although PCR is more sensitive than IHC, the latter provides a positive result only when the amount of antigen is significant, and therefore is likely the cause of the enteric pathology. Lawsonia intracellularis is the aetiologic agent responsible for Porcine proliferative enteropathy. However, it is complex to determine whether this intracellular bacterium is the cause of clinical disease due to its high prevalence in the field. In this study, we investigate the agreement between PCR and IHC results for L. intracellularis evaluation, and the infection and co-infection by porcine circovirus type 2 and L. intracellularis in the ilea of pigs presented with suspected proliferative enteropathy in Italy. The objective of this study was to employ a diagnostic algorithm, which involves detecting positive farms by stool PCR followed by PCR and histology/immunohistochemistry on ileum samples, for diagnosing Lawsonia intracellularis proliferative enteritis in Northern Italy. The primary aim was to examine the relationship between the gold standard of L. intracellularis diagnostics, namely histology and immunohistochemistry, and PCR in acute and chronic cases of L. intracellularis enteritides. An additional goal was to investigate the coinfection of L. intracellularis with porcine circovirus type 2 (PCV2). Twenty-eight ileum samples, including four from acute cases and 24 from chronic cases, were collected. PCR yielded positive results in 19 cases (four acute and 15 chronic cases). In comparison, immunohistochemistry was positive in 16 cases (four acute and 12 chronic cases), with an observed agreement of 89%. The findings suggest that performing the two tests in series can increase the specificity of the causal diagnosis. PCR may be used as a screening tool to identify the presence of the microorganism, and only positive cases will be examined by histology and immunohistochemistry to confirm the causative role of L. intracellularis. Co-infection with PCV2 was demonstrate in two out of four acute cases and in two out of 24 chronic cases, providing further evidence to support the hypothesis that when the infection starts with ubiquitous pathogens such as L. intracellularis, it may boost the possibility of PCV2 replication, especially in acute cases. Simple Summary: The use of boars for breeding purposes is common on small farms in rural regions of Italy. These animals are butchered at the end of their reproductive life for the production of charcuterie. In this regard, orchiectomy is required to remove boar taint caused by testosterone metabolites. The purpose of this study is to validate the efficacy of a safe, rapid and inexpensive castration procedure. To accomplish this objective, 91 boars were orchiectomised using the suture ligation technique. Following castration, characteristics of the severed ductus deference were evaluated in order to understand how much force would be required to tie knots without breaking the deferens and achieving proper haemostasis. After identifying a correlation between tensile strength and the age and weight of boars, a sutureless technique was successfully applied to 20 animals. The heterogeneity of Italian manufacturing processes results in the production of a large variety of pork products. In Sardinia, boars are raised and butchered to produce charcuterie. These animals are castrated before slaughter as androstenone would otherwise taint the meat, rendering it unfit for human consumption. However, to date, the literature concerning surgical orchiectomy in adult boars is limited. The goal of this study is to assess whether a sutureless swine orchiectomy procedure is feasible. Additionally, this study aims to determine the appropriate traction force needed to tie knots in the deferens duct of pigs of different weights and ages. Two groups were created: the first (n = 91) underwent orchiectomy by suture ligation; the second (n = 20) was castrated using the sutureless technique. Deferens ducts of animals in the first group (n = 182) were collected following castration, and their tensile strength was measured. Pearson's linear correlation was used to determine the relationship between the maximum traction force and weight and age groups. A correlation of 0.99 and 0.96 was shown between traction force and age and traction force and weight, respectively. In accordance with these results, sutureless castration was performed on 20 animals, calibrating the pulling force needed according to the age and weight of the boars. Background: Hepatitis E Virus (HEV) is recently considered an emerging public health concern. HEV genotypes 1 and 2 are widely distributed and pathogenic only for humans. In contrast, HEV, genotypes 3 and 4 are observed in swine, deer, wild boars and rabbits and can also be transmitted to humans. The presence of HEV in the liver, muscle, faeces, blood, and bile was detected by real-time RT-PCR in 156 pigs belonging to twenty different farms, ranging from 1 to 8 months of age. The phylogenetic analysis was performed on the viral strain present in the positive biological matrix, with the lowest Ct. HEV-IgG and HEV-IgM in the sera were analysed by two different ELISA kits. Results: Twenty-one pigs, i.e., 13.46% of them (21/156, 95% CI: 8.53%-19.84%), tested positive for HEV in at least one biological matrix by real-time RT-PCR, while phylogenetic analysis revealed the presence of HEV subtypes 3f and 3c. Pig serums analysed by ELISA showed an overall prevalence of 26.92% (42/156, 95% CI: 20.14%-34.60%) for HEV-IgG, whereas the 28.95% (33/114, 95% CI: 20.84%-38.19%) of them tested negative resulted positive for the HEV-IgM. Conclusions: The faeces are the biological matrix with the highest probability of detecting HEV. The best concordance value (Kappa Kohen index) and the highest positive correlation (Phi index) were observed for the correlation between bile and liver, even when the number of positive liver samples was lower than the positive bile samples. This finding may suggest that a higher probability of HEV occurs in the bile, when the virus is present in the liver, during the stages of infection. Finally, the presence of HEV in muscle was observed in 11 pigs, usually used for the preparation of some dishes, typical of the Italian tradition, based on raw or undercooked meat. Porcine epidemic diarrhea virus (PEDV), belonging to the genus Alphacoronavirus, can cause serious disease in pigs of all ages, especially in suckling pigs. Differences in virulence have been observed between various strains of this virus. In this study, four pigs were inoculated with PEDV from Germany (intestine/intestinal content collected from pigs in 2016) and four pigs with PEDV from Italy (intestine/intestinal material collected from pigs in 2016). The pigs were re-inoculated with the same virus on multiple occasions to create a more robust infection and enhance the antibody responses. The clinical signs and pathological changes observed were generally mild. Two distinct peaks of virus excretion were seen in the group of pigs inoculated with the PEDV from Germany, while only one strong peak was seen for the group of pigs that received the virus from Italy. Seroconversion was seen by days 18 and 10 post-inoculation with PEDV in all surviving pigs from the groups that received the inoculums from Germany and Italy, respectively. After fifty years of spread in the European continent, the African swine fever (ASF) virus was detected for the first time in the north of Italy (Piedmont) in a wild boar carcass in December, 2021. During the first six months of the epidemic, the central role of wild boars in disease transmission was confirmed by more than 200 outbreaks, which occurred in two different areas declared as infected. The virus entered a domestic pig farm in the second temporal cluster identified in the center of the country (Lazio). Understanding ASF dynamics in wild boars is a prerequisite for preventing the spread, and for designing and applying effective surveillance and control plans. The aim of this work was to describe and evaluate the data collected during the first six months of the ASF epidemic in Italy, and to estimate the basic reproduction number (R0) in order to quantify the extent of disease spread. The R0 estimates were significantly different for the two spatio-temporal clusters of ASF in Italy, and they identified the two infected areas based on the time necessary for the number of cases to double (td) and on an exponential decay model. These results (R0 = 1.41 in Piedmont and 1.66 in Lazio) provide quantitative knowledge on the epidemiology of ASF in Italy. These parameters could represent a fundamental tool for modeling country-specific ASF transmission and for monitoring both the spread and sampling effort needed to detect the disease early. Simple Summary: An interesting anthropological question is whether, over the last 10,000 years, humans have domesticated animals by actually selecting the tamest and least aggressive individuals. These characteristics—known as 'domestication syndrome'—should be present from the very early stages of life of domesticated animals. Because social play is the most important 'friendly' contact between very young individuals, we verified whether piglets (with both domestic pig parents) were more playful and less aggressive than hybrid pups (with wild boar father and domestic pig mother). To this purpose, we investigated three litters of piglets and three litters of wild boar hybrids, all raised in northern Italy (Parva Domus farm, Cavagnolo, Torino) in the same grassland/woodland environment and under similar farming conditions. We found that piglets played more and in a 'less chaotic' way than wild boar hybrids, especially after the first three weeks of life but also that piglets were more aggressive than hybrids, with piglet aggression being more frequently unbalanced in favour of one individual. Thus, the domestication syndrome does not fully apply to either social play or aggression, possibly because artificial selection may have produced greater tameness of pigs towards humans than towards other pigs. The 'domestication syndrome' defines a suite of features that domesticated animals possess as the result of the artificial selection operated by Homo sapiens since the Neolithic. An interesting anthropological question is whether such features, including increased tameness and reduced aggression, apply to all domesticated forms. We investigated this issue in the domestic pig (Sus scrofa). We video-recorded and analysed aggression and social play (mostly play-fighting) sessions from piglets (three litters; n = 24) and wild boar hybrids (domestic pig mother x wild boar father; three litters; n = 27) from 6–50 days of age, raised in the same woodland/grassland habitat and extensive farming management (ethical farm 'Parva Domus', Cavagnolo, Torino). Play and aggression session structure was assessed via Asymmetry (AI; offensive/defensive pattern balance), Shannon (H′; pattern variability), and Pielou (J; pattern evenness) indices. We found that piglets played more (especially after the 20th day of life) and engaged in less variable and uniform sessions than wild boar hybrids. Compared to hybrids, piglets showed less variable but more frequent (especially when approaching weaning) and asymmetrical aggressive events. Thus, the domestication syndrome does not seem to fully apply to either social play or aggression, possibly because artificial selection has produced greater tameness of pigs towards humans than towards conspecifics. Aims: Bacillus cereus is often responsible for foodborne diseases and both local and systemic infections in humans. Cases of infection in other mammals are rather rare. In this study, we report a B. cereus feed‐related outbreak that caused the death of 6234 pigs in Italy. Methods and Results: Massive doses of a Gram‐positive, spore‐forming bacterium were recovered from the animal feed, faeces of survived pigs and intestinal content of dead ones. The B. cereus MM1 strain was identified by MALDI‐TOF MS and typified by RAPD‐PCR. The isolate was tested for the production of PC‐PLC, proteases, hemolysins and biofilm, for motility, as well as for the presence of genes encoding tissue‐degrading enzymes and toxins. Antimicrobial resistance and pathogenicity in Galleria mellonella larvae were also investigated. Our results show that the isolated B. cereus strain is swimming‐proficient, produces PC‐PLC, proteases, hemolysins, biofilm and carries many virulence genes. The strain shows high pathogenicity in G. mellonella larvae. Conclusions: The isolated B. cereus strain demonstrates an aggressive profile of pathogenicity and virulence, being able to produce a wide range of determinants potentially hazardous to pigs' health. Significance and Impact of Study: This study highlights the proficiency of B. cereus to behave as a devastating pathogen in swine if ingested at high doses and underlines that more stringent quality controls are needed for livestock feeds and supplements. Background: African Swine Fever (ASF) is a challenge for pig health worldwide. The disease has spread to multiple countries on five continents. ASF‐free countries need to apply effective strategies to prevent the introduction of infection. Methods: Italy implemented a surveillance and prevention plan for ASF in 2020, supported by a dedicated information system. Several pillars for action have been identified: passive surveillance in both domestic pigs and wild boar populations, verification of the implementation of biosecurity measures on farms and an awareness campaign for all involved stakeholders. Results: There were some regional differences in the management of passive surveillance. In order to identify all critical points and apply corrective measures, regional authorities were called to carry out a gap analysis exercise in July 2020. There were an adequate number of samples collected from wild boar but the number of samples collected from domestic pigs was below the target in most regions. Furthermore, sample distribution within the country was not homogeneous. Eucoleus garfiai (syn. Capillaria garfiai) is a nematode infecting lingual tissue of domestic and wild swine. Prevalence data for this parasite are scant and often related to accidental findings, occurring only in Japan and a few European countries. In this study, an epidemiological survey was performed in order to identify E. garfiai in wild boar from the Campania region, southern Italy. A total of 153 wild boar carcasses were inspected over the course of two hunting seasons (2019–2020). Histological examinations were performed on tongue samples fixed and stained with haematoxylin and eosin. The scraping of dorsal tongue tissue was carried out to collect adult worms for parasitological examination. Out of 153 wild boars, 40 (26.1%, 95% CI: 19.8–33.6%) tested positive for helminths and/or eggs in tongue tissues. Parasites were identified morphologically and identification was confirmed by molecular analysis of the 18S rRNA gene, showing a 99% nucleotide match with E. garfiai sequences available in literature. No statistically significant differences were found according to age, sex nor hunting province. Our findings agree with previous histopathological data confirming the low pathogenic impact of this nematode. The present study represents the first report of E. garfiai in wild boar from Italy. Hepatitis E virus (HEV) is a zoonotic pathogen, responsible for numerous cases of infection in humans. Transmission occurs through the orofecal route, and ingestion of contaminated foods represents an important risk factor for final consumer's health. Wild animal species, in particular wild boar (Sus scrofa), are the main virus reservoirs; liver is the target organ, from which, through the hematic diffusion, HEV reaches different tissues and organs, as muscular one. The hygienic-sanitary critical issues connected with game meat food chain in general, and particularly wild boar, with special regards to any geographical area where this animal species can be directly in contact with humans, domestic ones (i.e., domestic pig), and other wild reservoirs (i.e., wild ruminants), finds favorable environmental conditions, have induced us to conduce the present scientific investigation. During the hunting season 2019/2020, a total of 156 wild boar livers were collected from provided plucks at slaughterhouse in Ascoli Piceno. Nested RT-PCR was used for the viral RNA detection. Results demonstrated a positivity of 5.12% (8/156), and the circulation in the screened area of genotype 3 subtype c, which is frequently identified in Central Italy. HEV sanitary relevance and the emerging role of any food chains in its transmission impose further detailed studies. Simple Summary: Hepatitis E virus (HEV) is an emerging pathogen, causing an increasing number of autochthonous cases in industrialized countries. In Europe, infections are associated with the zoonotic HEV-3 and HEV-4 genotypes and pigs and wild boars are the main reservoirs. A major concern of infections is linked to its foodborne transmission, due to consumption of raw or undercooked pork products infected by HEV-3 or HEV-4. HEV-3 is widespread in farmed pigs, mainly aged 3–4 months. Besides a decline with age, infected pigs have been observed at slaughterhouses, representing a risk for both the consumers and the workers of the pig industry. HEV is transmitted by the fecal–oral route and shed in feces in large amounts. The risk of viral spreading in farm and presence of infected pigs at slaughtering was evaluated by assessing the quantity and the duration of HEV-3 shedding in feces of infected pigs. Feces of 23 HEV-3 positive pigs were assayed during their fattening, shortly before their slaughtering. Results confirmed a long period of viral shedding in feces with a large amount of the virus released in the environment (mean 105 GC/g). Prevalence and quantity of the virus declines with the age of animals. The study provides information on the dynamic of the infection in pigs, important to prevent HEV occurrence and circulation in farms. Genotype 3 of hepatitis E virus (HEV-3) is the most common in Europe in both humans and pigs. HEV-3 strains are zoonotic, and foodborne cases associated with consumption of raw and undercooked pork products, mainly liver sausages, have been described. HEV-3 circulates largely in European pig farms, maybe due to its long persistence in the environment. Animals get infected around 3–4 months of age; shortly after, the infection starts to decline up to the age of slaughtering (8–9 months of age in Italy). With the purpose to understand the duration in farmed pigs of the shedding of the virus and its quantity, HEV-RNA detection was performed by Real-time RT-PCR from feces collected individually from two groups of 23 pigs. Sampling was conducted for 4 months shortly before slaughtering age. At 4-months-old, all animals were shedding HEV-3 to high load around 105 genome copies per gram (GC/g). Prevalence was higher in growers than in fatteners, with most of the pigs still positive around 166 days of age. Beyond some difference among individual pigs, the amount of HEV in feces decreased with the age of animals. Neospora caninum (Apicomplexa, Sarcocystidae) is a major cause of reproductive failure in cattle. In pigs, only a few studies investigated the effects of this parasite on reproductive efficiency. Considering the relevance of swine farms in northern Italian regions, an epidemiological survey was designed to investigate the spread of N. caninum infection. Three hundred seventy fattening pigs and sows from 23 intensive farms in Lombardy were sampled. Sera were analyzed by a commercial immunofluorescence antibody test. Statistical analysis through univariate and multivariate generalized linear models was conducted to detect farm management practices enhancing the risk of infection. At the farm level, 52.1% (12/23) of the selected farms, 72.7% housing sows and 40% fattening pigs, scored positive. At the individual level, 25 animals (25/370, P = 6.7%) were positive to N. caninum antibodies: one fattening pig and two sows showed an antibody titer of 1:100, and in two sows, an antibody titer of 1:400 and 1:6400 was evidenced. A higher seroprevalence was detected in sows (17/151, P = 11.2%) if compared to fattening pigs (8/219, P = 3.6%) (OR = 1.19, P value = 0.000 in sows). Moreover, a higher seroprevalence was recorded in farms with low and moderate sanitary score (P = 100% and P = 64.2%, respectively) if compared to farms with high sanitary score (P = 22.2%) (OR = 1.24, P value = 0.007 in score = 1 and OR = 1.10, P value = 0.050 in score = 2). This study provides the first data on the circulation of N. caninum in intensive swine farms in Italy, demonstrating the spread of the parasite in fattening pigs and sows in Lombardy region. Streptococcus suis is a pathogen associated with severe diseases in pigs and humans. Human infections have a zoonotic origin in pigs. To assess circulating strains, we characterized the serotypes, sequence types, and antimicrobial susceptibility of 78 S. suis isolates from diseased farmed pigs in Italy during 2017-2019. Almost 60% of infections were caused by serotypes 1/2 and 9. All but 1 of the serotype 2 and 1/2 isolates were confined to a single cluster, and serotype 9 isolates were distributed along the phylogenetic tree. Besides sequence type (ST) 1, the serotype 2 cluster included ST7, which caused severe human infections in China in 1998 and 2005. A large proportion of serotype 9 isolates, assigned to ST123, were resistant to penicillin. The emergence of this clone threatens the successful treatment of S. suis infection. The description of the pattern of livestock movements between herds provides essential information for both improving risk‐based surveillance and to understand the likely spread of infectious diseases. This study provides a description of the temporal pattern of pig movements recorded in Italy on a 4‐year period (2013–2016). Data, provided by the National Livestock registry, were described by social network analysis and the application of a walk‐trap algorithm for community detection. Our results show a highly populated community located in Northern Italy, which is the focal point of the Italian industrial pig production and as a general pattern an overall decline of medium and backyard farms and an increase in the number of large farms, in agreement with the trend observed by other EU pig‐producing countries. A seasonal pattern of all the parameters evaluated, including the number of active nodes in both the intensive and smaller production systems, emerged: that is characterized by a higher number of movements in spring and autumn, linked with the breeding and production cycle as pigs moved from the growing to the finishing phase and with periods of increased slaughtering at Christmas and Easter. The same pattern was found when restricting the analysis to imported pig batches. Outbreaks occurring during these periods would have a greater impact on the spread of infectious diseases; therefore, targeted surveillance may be appropriate. Finally, potential super‐spreader nodes have been identified and represent 0.47% of the total number of pig holdings (n = 477). Those nodes are present during the whole study period with a similar ranking in their potential of being super‐spreaders. Most of them were in Northern Italy, but super‐spreaders with high mean out‐degree centrality were also located in other Regions. Background and Aim: Balantidiasis, caused by Balantidium coli (syn. Neobalantidium coli or B. coli), represents a neglected parasitic infection of zoonotic significance affecting a variety of hosts, including domestic pigs that are the main reservoir. B. coli has a direct life cycle with a fecal-oral route transmission that occurs mainly by the ingestion of food and water contaminated with cysts. The ingestion of meat contaminated during inappropriate slaughtering processes may represent a new potential route of transmission. Only a few studies have investigated the prevalence of B. coli in domestic pigs in Italy, despite its high prevalence and zoonotic significance. This study aimed to improve the knowledge on B. coli prevalence in domestic swine reared both in intensive and "en plein air" breeding systems in the south of Italy. Materials and Methods: The infection rate of B. coli in pigs bred in 15 different pig farms and regularly slaughtered in South Italy, in the Calabria region, was investigated. From 2017 to 2019, 177 terminal parts of the rectums of pigs, of which 91 commercial hybrids and 86 autochthonous (Nero Calabrese), reared, respectively, in intensive and "en plein air" breeding system, were tested for parasite detection. After the slaughtering, the terminal part of the rectum was sampled and transported to the laboratories and immediately processed. For the detection of trophozoites, fresh smears of feces were examined at light microscopy. Moreover, flotation was performed using a sodium chloride solution, then smears of feces were dispersed on microscope slides and examined at light microscopy. The observed parasites were identified at the species level using morphological characteristics. Results: B. coli was detected in a total of 83 (i.e., 46.89%) pigs, with a significantly higher prevalence (p<0.0001) found in commercial hybrid (i.e., 59/91-64.84%) pigs rather than in autochthonous ones (i.e., 24/86-27.91%). It follows that the infection was more common in pigs reared in the intensive breeding system than in "en plein air" ones (p<0.0001). The infection prevalence was higher in males than in females and lower in elder animals than in younger ones (p=0.012 and p<0.0001, respectively). Conclusion: The breeding system is likely the main discriminant for the parasite spread as well as the hygienic condition of the farms. Good manufacturing and hygiene practices along the food production chain are crucial in preventing human balantidiasis transmission by meat consumption. The characterization and safeguarding of typical and local animal breeds were the focus of this study. These two aspects are crucial for their management, in particular for the Italian pig population. Indeed, Italy is among the top ten nations in terms of the pig population, with the local breeds famous worldwide for their great meat production. In this study we characterized three of these local pig breeds of Italy, i.e. Nero Siciliano (Black Sicily, BS), Casertana (C) and Nero Lucano (Black Lucano, BL), using cytogenetic and genomic techniques. These three local pig breeds of Italy were characterized by Aneuploidies, CAs (Chromosome Aberration) and SCEs (Sister Chromatid Exchange) and RAPD-PCR (Random Amplified Polymorphic DNA - Polymerase Chain Reaction) in order to clarify any genetic differences between the breeds to preserve biodiversity and to guarantee economic development of local pork industries. Comparing the data obtained from the analysis of Aneuploidies, CAs and SCEs, as well as the RAPD-PCR examination, it was concluded that the BS and C breeds are more similar and sufficiently different from the BL breed. The genetic diversity and the characteristic polymorphic profile make BL a breed of high value to be protected, particularly for its much appreciated meat. The research was carried out as a preliminary phase for the development program for local breeds and will provide useful information for the management, conservation and promotion of these three Italian pig breeds. Background Pigs are considered the main reservoir of genotypes 3 and 4 of hepatitis E virus (HEV), which is the major cause of acute hepatitis of viral origin in humans worldwide. An increasing number of autochthonous HEV infections have been observed in recent years in industrialized countries, most likely as a result of zoonotic transmission through the consumption of raw or undercooked meat products. Methods Two hundred and thirty-three blood and liver samples were collected at four different local slaughterhouses from domestic pigs bred in Abruzzo, a region of south-central Italy, where there is the highest human seroprevalence to HEV compared with the rest of Italy. An indirect enzyme-linked immunosorbent assay kit was used for detecting anti-HEV IgG in the sera, while the presence of HEV RNA was investigated by performing a real-time reverse transcriptase-polymerase chain reaction (RT-PCR). Results Between 87.3% and 100% of swine serum samples collected in different slaughterhouses of Abruzzo were positive for anti-HEV antibodies. Conversely, none of the liver samples collected from the same animals were positive for HEV by real-time RT-PCR. Conclusions The hypothesis of foodborne zoonotic transmission from local pigs as responsible for the hyperendemic status of Abruzzo cannot be corroborated. However, the high seroprevalence observed in pigs indicates that HEV is highly circulating in these territories. We propose to further investigate the role of wild fauna and trade in carrier pigs, and the maintenance of HEV virulence in the environment and meat supply chain to shed light on the possible sources of human infection and the degree of occupational risk. Objectives: To the best of our knowledge, we describe the first evidence in Europe of an MDR, blaNDM-4-positive Escherichia coli isolated from a food-producing animal, harboured by a novel IncFII plasmid of which we report the complete sequence.Methods: One blaNDM-4-positive E. coli isolated in 2019 from the caecal contents of a fattening pig in Italy was in-depth characterized by combined bioinformatic analysis of Oxford Nanopore long reads and Illumina short reads, for in silico typing, determination of the blaNDM-4 genetic context and full reconstruction of the blaNDM-4-carrying plasmid.Results: The isolate belonged to ST641 and to the genoserotype O108:H23 and tested positive for different virulence genes and plasmid replicons. The MDR phenotype of resistance to all β-lactams, carbapenems, sulfamethoxazole and trimethoprim was mediated by blaTEM-1B, blaNDM-4, sul1/sul3 and dfrA12, respectively. The blaNDM-4 gene was harboured by a novel 53 043 bp IncFII plasmid (pMOL412_FII) composed of four main genetic regions, including an MDR region (MRR-NDM-4) of 16 kb carrying blaNDM-4 and several antimicrobial resistance genes located in a class 1 integron. pMOL412_FII was closely related to another ∼90.3 kb plasmid (pM109_FII) harbouring blaNDM-4 in an E. coli isolated from a human patient in Myanmar.Conclusions: To the best of our knowledge, we have identified for the first time in Europe an NDM-producing Enterobacterales in livestock and resolved the complete sequence of the novel pMOL412_FII plasmid harbouring blaNDM-4 in an MRR. A global One Health approach, comparing genomic data from different sources and geographical areas, may help to trace back and control possible plasmid-borne carbapenemase gene transmission between animals and humans and along the food chain at international level. The scientific community considers the improper use of antimicrobials in farm animals among the causes of the insurgent bacterial resistance contributing to the wider pool of resistance at the animal/human interface with serious public health implications. The present study aims to describe the current perceptions regarding antimicrobial use (AMU) and resistance (AMR) and the prescribing behaviour of two different target groups: cattle and pig veterinary practitioners working in Italy. A semi-structured questionnaire was designed and administered using the Computer-Assisted Web Interviewing method. Among the 789 participants who completed the questionnaire, 53.5% and 12.2% were cattle and pig veterinarians, respectively. Differences between the two groups emerged in their opinions concerning different topics. For example, 69.4% and 85.8% of cattle veterinarians claimed to 'somewhat or strongly agree' with the statements 'prescribing broad-spectrum antibiotics in rearing farms increases AMR' and 'the preventive use of antibiotics fosters the development of AMR', against the observed 59.4% and 69.8% of pig veterinarians (p =.050 and p =.000, respectively). Moreover, 26% of pig veterinarians claimed to 'somewhat or strongly agree' with the statement 'alternative methods currently available could be an efficient alternative to antimicrobial treatment', against 16.1% of cattle veterinarians (p =.022). At the same time, no differences between the two groups emerged regarding the danger inadequate AMU in animals poses on both animal and human health. From collected data, the need to provide appropriate training strategies with the aim of adopting a behaviour more consistent with the guidelines for prudent AMU in cattle and pig production systems has emerged. 85.8% of cattle vs. 69.8% of pig veterinarians agreed with the statement 'the preventive use of antibiotics fosters the development of AMR'. 64.5% of cattle and 69.1% of pig veterinarians declared they suggest/prescribe alternative approaches to the use of antimicrobials. A total of 57 tissue samples of domestic pigs (Sus scrofa) were collected from the meat outlets of five north Indian states and examined for sarcocystosis by histological and molecular methods. The genomic DNA extracted from five representative positive isolates was subjected to PCR amplification of the partial 18S rRNA gene followed by cloning and sequencing. Sequence analysis of the newly generated Indian isolates recorded 96.9–100.0% identity with published sequences of Sarcocystis suihominis. Two new haplotypes that have not been previously described manifested 99.5–100.0% nucleotide homology within themselves. In the phylogenetic analysis, Indian isolates of S. suihominis grouped together with S. suihominis originating from Italy, and they collectively formed a sister clade with Sarcocystis miescheriana within a clade containing various Sarcocystis spp. of ruminants having felids as final hosts. At the same time, this clade separated from a sister clade containing Sarcocystis spp. of bovid or cervid ruminants using canids as known or surmised definitive host. The current study established the phylogenetic relationship of Indian isolates of S. suihominis with various Sarcocystis spp. as well as with other taxa of Sarcocystidae family based on 18S rRNA gene for the first time. Antimicrobial resistance (AMR) in bacteria is a frequent and widespread phenomenon. The European Food Safety Authority (EFSA) reports that multidrug resistant (MDR) Escherichia coli is considered an important hazard to public health. The lack of data on the correlation between the administration of antibiotics to pigs and the diffusion of MDR E. coli necessitates an in-depth study. The aims of our study were first of all to determine the presence of MDR and/or extended spectrum β-lactamase (ESβL) E. coli isolated from feces and carcasses of pigs; and second, to evaluate the correlation between antibiotic resistance and the antibiotic treatment administrated to the animals considered. The examined E. coli was isolated from 100 fecal swabs and 100 carcass sponges taken from farms and slaughterhouses located in Reggio Emilia province in Italy. The MDR isolates were tested following the protocol defined by EUCAST (2015). Subsequently, a real-time PCR and an endpoint-PCR were used for the genomic analysis. Data highlighted 76.5% of MDR E. coli with a marked presence of the ampicillin (AMP)-streptomycin (STRE)-tetracycline (TETRA) pattern. Moreover, 13 isolates were ESβL producers, and the blaCTXM gene was the most frequently observed in genomic analysis. Results confirm the complexity of the AMR phenomenon showing a partial correlation between the administration of antibiotics and the resistance observed. Pigs destined to the production of Protected Designation of Origin items are colonized by bacteria resistant to a wide range of antibiotic classes even if data are encouraging for colistin and third generation cephalosporin. Livestock-associated methicillin-resistant Staphylococcus aureus sequence type (ST) 398 (LA- MRSA ST398) is a genetic lineage for which pigs are regarded as the main reservoir. An increasing prevalence of LA-MRSA ST398 has been reported in areas with high livestock density throughout Europe. In this study, we have investigated the drivers contributing to the introduction and spread of LA-MRSA ST398 along the pig farming system in Southern Italy. Whole-genome sequencing (WGS) of LA-MRSA ST398 isolates collected in 2018 from pigs (n=53) and employees (n=14) from 10 farms in the Calabria region were comparatively analysed with previously published WGS data from Italian ST398 isolates (n=45), an international ST398 reference collection (n=89) and isolates from Danish pigs farms (n=283), which are the main suppliers of pigs imported to Italy. Single-nucleotide polymorphisms (SNP) were used to infer isolates relatedness and, together with data from animal trading, factors contributing to LA-MRSA ST398 dissemination were identified. The analyses support the existence of two concurrent pathways for the spread of LA-MRSA ST398 in Southern Italy: i) multiple introductions of LA-MRSA ST398 through the import of colonized pigs from other European countries including Denmark and France and; ii) the spread of distinct clones dependent on local trading of pigs between farms. Phylogenetically related Italian and Danish LA-MRSA ST398 isolates shared extensive similarities including carriage of antimicrobial resistance genes. Our findings highlight the potential risk of transboundary transmission of antimicrobial-resistant bacterial clones with a high zoonotic potential when importing pigs from countries with high LA-MRSA prevalence. Hepatitis E virus (HEV) is a singlestrand RNA virus that causes an acute viral hepatitis in humans. Among its eight recognized genotypes, HEV-3 and HEV-4 are zoonotic, infecting humans, pigs and wild boars. Recently, HEV-3 has been also detected in red deer, which represents another reservoir of HEV. Consumption of raw pork products (mainly liver sausages), undercooked wild boar meat, raw wild boar liver and deer meat has been responsible for foodborne HEV human worldwide. From November 2018 to March 2019, liver samples collected from 97 wild boars hunted in Emilia-Romagna region (Northern Italy) were tested for HEV RNA. The hunting area included two territories for an extension of 33 km2, named A (about 13 km2,natural park, deciduous wood) and B (about 20 km2, cultivated fields in proximity of a river) areas. Distance between the two areas ranged between 8 to 10 km. A total of 73 wild boars were hunted in area A, and 24 in area B. HEV RNA was detected by Real-time RT- PCR in 23/73 liver samples of wild boars living in area A only (31.5% - 95% CI: 22.0- 42.8%). The HEV sequences (n=13) clustered within genotype 3. The majority of positives belonged to animals < 12 months (12/25; 48%), followed by subadults (13-24 months) (7/16; 43.8%) and adults (4/32; 12.5%). This difference was found to be statistically significant (p=0.0024). In absence of pig farms, the restriction of HEV-positive animals to a well-defined territory of 13 km2 (Boschi di Carrega Regional Park) could hypothetically be related to the presence of red deer (Cervus elaphus), which lived in area A at the beginning of the hunting season. Aims: The diversity and the geographical distribution of swine papillomaviruses (PVs) are virtually unknown. The occurrence and the diversity of swine PV were therefore investigated in pig slurry collected in Italy, to contribute towards filling this gap in knowledge. Methods and Results: Twenty‐two slurry samples underwent analysis by nested PCR and DNA sequencing using published and newly designed specific primer pairs for Sus scrofa papillomavirus (SsPV) type 1 and 2 (SsPV1 and 2), along with degenerate PV‐specific primers targeting the major coat protein L1 and the helicase protein E1. Overall, three samples (13·6%) were positive for SsPV1 by specific primers, and nucleotide (nt) sequences showed 99–100% nt identity with SsPV1 variant a (EF395818), while SsPV2 was not found in any sample. Using generic primers, eight samples (36·4%) were tested positive for human papillomavirus (HPV), and were characterized as follows: β1‐HPV8, β1‐HPV14, β1‐HPV206, β2‐HPV113, β2‐HPV120 and γ1‐HPV173. Moreover, one unclassified γ‐type was detected. Conclusions: Both swine and human PVs were detected in pig slurry in this study. The unexpected presence of HPV in pig waste could be explained as the result of an improper use of the sewage collection pits and/or with improper procedures of the operators. Significance and Impact of the Study: This study reports the first detection of SsPV1 in Italy, along with the first detection of HPVs in pig slurry samples in Italy, and expands our knowledge about PV diversity and geographic distribution. Hepatitis E is an emerging disease in industrialized countries. The food-borne transmission of hepatitis E virus (HEV) is associated principally with products derived from the domestic pig, the wild boar, and deer; however, few quantitative data are available on HEV loads in animals used in food production. This study assessed HEV occurrence, viral load and genetic variability in wild boar hunted for domestic consumption in the district of Viterbo (Central Italy) where high anti-HEV IgG seroprevalence values are reported in humans. A total of 332 liver and 69 intestine samples were obtained from wild boar hunted between 2011 and 2014. The liver tissue in 54 of the animals (16.3%) was HEV-positive. Viral loads in quantifiable liver samples (n = 29) ranged between 3.2 × 102 and 3.8 × 105 genome copies (g.c.)/g with a mean value of 1.85 × 104 g.c./g. A statistically significant positive correlation was found between viral concentration in liver and intestinal tissue, though mean viral load in the intestines was lower (3.13 × 103 g.c./g). Twenty-six samples were characterized molecularly as genotype 3 (G3) and four subtypes (a, c, f and l) were detected. Finally, twelve samples with near identical sequences were identified as G3 but could not be assigned to any of the known subtypes, and could therefore represent a potentially new subtype. • HEV RNA prevalence in the livers of 332 wild boar hunted for domestic consumption in Central Italy was 16.3%. • The average viral load in quantifiable samples was 3.8 × 105 HEV g.c./g in liver and 3.13 × 103 g.c./g in intestine. • Four G3 subtypes (a, c, f and the putative subtype l) were detected in the HEV-positive animals. • A G3 sequence that could not be assigned to any known subtype was obtained from several samples. Methicillin-resistant Staphylococcus aureus (MRSA) is an important medical issue, since it causes serious and sometimes fatal infections in humans. Intensively reared swine may serve as reservoirs for MRSA that can infect swine workers, and also consumers (via contaminated meat). In this study, MRSA strains were isolated from 55 of the 85 (64.7%) intensive pig farms surveyed, and prevalence was greater on pig fattening farms than on breeding farms. In addition, we included in the study 63 foreign pigs imported for slaughter. Overall, the prevalence of MRSA in the 418 sampled swine was 59.1%; 12 genotypes were identified among the isolates; ST398 (96.4%) was most prevalent, followed by ST97 (2%), ST9 (0.8%) and ST1 (0.8%). MRSA isolates were also detected in 26 (17.3%) of the 150 operators included in the study; the genotypes detected were ST398 (85%), ST9 (7.6%), ST5 (3.8%) and ST1 (3.8%). All the strains were pvl negative and pia positive. Both swine and human strains displayed a multi-resistance pattern, and almost all were resistant to tetracycline. The results obtained in this study confirm the high prevalence of MRSA in swine reared and slaughtered in Italy, and underline the public health risk linked to the spread of antimicrobial-resistant Staphylococcus aureus among intensively reared pigs. • Our study shows genetic diversity of MRSA in pigs reared and slaughtered in Italy. • The foodborne risk linked to the consumption or handling the pig meat should be considered. • The professional risk for the farmers and the abattoir workers should be considered. • This is the first international report on the prevalence of MRSA in swine reared in Southern Italy. • Fifteen years of serological data on Leptospira in animals were presented. • An increase positivity to Leptospira in last years of investigation was recorded. • Rise or fall of some serovars were observed. • Some changes in hosts/serovars relations were detected. Leptospirosis is a re-emerging bacterial zoonosis. North-Central Italy is characterized by a geographic area that promote Leptospira circulation. Data on sero-epidemiological survey carried out from 2002 to 2016 in North-Central Italy were reported and discussed. Overall, 709 out of the 8488 (8.35%) tested sera were positive for Leptospira at the cut-off titer (1:100) and 218 (2.57%) at higher titer (≥1:400). The highest percentages of positivity was recorded for coypus (22.86%), swine (19.74%) and bovine (13.03%). Pomona and Australis resulted the serogroup more often detected, followed by Sejroe and Icterohaemorrhagiae; while, a low number of positive sera was detected for serogroups Ballum, Canicola and Tarassovi. Percentage of positive sera for each year slightly decreased from 2002 to 2008 and rose from 2009. High percentages of positive reactions were recorded in 2014 (17.23%), 2015 (19.61%) and 2016 (38.05%). In conclusion, the results of this investigation reported an increase of leptospirosis in North-Central Italy. Furthermore, several animals resulted infected as accidental hosts by unusual Leptospira serovars. Shiga toxin‐producing Escherichia coli (STEC) strains are food‐borne pathogens of public health concern. Despite ruminants are the most important reservoir, STEC human infections have also been attributed to pigs. We examined for the presence of STEC in 234 samples of swine caecal content collected during the year 2015 at Italian abattoirs in the framework of the harmonized monitoring of antimicrobial resistance (Decision 2013/652/EU). The presence of stx genes was detected in 122 (52.1%) samples, which were subsequently subjected to STEC isolation and characterization. The analysis of the 66 isolated STEC strains showed that the majority of the isolates (74.2%) possessed the stx2a gene subtype, in a few cases (16.7%) in combination with stx2b or stx2c. Only 25.8% of isolates possessed the stx2e subtype, typical of swine‐adapted STEC. None of the isolates possessed the intimin‐coding eae gene and the majority of them did not belong to serogroups commonly associated with human infections. The EU is supporting measures that stimulate enhanced value-added products in order to conserve local and threatened livestock breeds. Several Traditional Pork Products (TPP) and Innovative Traditional Pork Products (ITPP) with health innovations from four untapped pig breeds in Spain (Porc Negre Mallorquí), Croatia (Turopolje), Italy (Cinta Senese) and Slovenia (Krškopolje) were analysed. Consumers' "Non-hypothetical" willingness to pay (WTP) and hedonic evaluation were investigated. An integrated experimental approach using two Non-Hypothetical Discrete Choice Experiment (NH-DCE) was carried out before and after a hedonic evaluation test. Results showed that the health innovative products (ITPP) received similar and even lower WTP than the "control" products (TPP) from the untapped pig breeds. The TPP outperformed products enriched with healthy ingredients or with reduced undesirable compounds. The potential demand for traditional and "unaltered" product from the rustic pig breeds could contribute to their conservation. Bovine tuberculosis (bTB) caused by Mycobacterium bovis is an important re-emerging disease affecting livestock, wildlife and humans. Epidemiological studies are crucial to identifying the source of bTB infection, and its transmission dynamics and host preference, and thus to the implementation of effective strategies to contain it. In this study, we typed M. bovis isolates from livestock, and investigated their genetic diversity and distribution. A total of 204 M. bovis isolates were collected from cattle (n = 164) and Sicilian black pigs (n = 40) reared in a limited area of the province of Messina, northeastern Sicily, an area that had previously been identified as having the highest incidence of bTB in livestock on the island. All M. bovis isolates were typed by both spoligotyping and 12-loci MIRU-VNTR analysis. Results from both methods were then combined in order to improve the discriminatory power of M. bovis typing. We identified 73 combined genetic profiles. Thirty-five point six percent of the profiles were common to at least two animals, whereas 64.4% of profiles occurred in only one animal. A number of genetic profiles were predominant in either cattle or black pigs. We identified common genetic patterns in M. bovis isolates originating not only from neighboring districts, but also from non-neighboring districts. Our findings suggest that bTB is widespread in our setting, and is caused by a large number of genetically diverse M. bovis strains. The ecology and farming practices characteristic of the area may explain the substantial M. bovis heterogeneity observed, and could represent obstacles to bTB eradication. [ABSTRACT FROM AUTHOR]

Porcine circovirus type 2 (PCV2) is associated with multi-factorial syndromes, commonly known as porcine-circovirus–associated diseases, which cause severe economic losses in the swine industry worldwide. Four genotypes (PCV2a, PCV2b, PCV2c, and PCV2d) have been identified. Lately, the prevalence of PCV2d has been increasing in many countries, thereby prefiguring a global replacement of PCV2b. Wild boars are also susceptible to PCV2 infection, with virus prevalence similar to that of domestic pigs. This work was aimed at expanding the knowledge about the molecular epidemiology of PCV2 in Italy. For this purpose, we analysed 40 complete ORF-2 sequences from PCV2 strains isolated from domestic pigs and wild boars in Sardinia (Italy) over a period of 5 years (2009–2013). Phylogenetic and Bayesian analyses were performed on three data sets compiled from DNA sequences over a large geographical area. PCV2b was found to be dominant in Sardinia, whereas no PCV2a and PCV2c were found. This study indicates the presence of genotype PCV2d-2 infecting both domestic and wild pigs, thus confirming its circulation in Italy. Sardinian sequences clustered mostly with Italian isolates and with strains from China, Belgium, Croatia, Taiwan, Korea, and Portugal. Porcine reproductive and respiratory syndrome (PRRS) is one of the main diseases of pigs, leading to large economic losses in swine production worldwide. PRRSV high mutation rate and low cross-protection between strains make PRRS control challenging. Through a semi-longitudinal approach, we analysed the relationships among performance parameters, PRRSV-1 genetic diversity, coinfections and antimicrobial use (AMU) in pig nurseries. We collected data over the course of five years in five PRRS-positive nurseries belonging to an Italian multisite operation, for a total of 86 batches and over 200,000 weaners involved. The farm experienced a severe PRRS outbreak in the farrowing unit at the onset of the study, but despite adopting vaccination of all sows, batch-level losses in nurseries in the following years remained constantly high (mean±SE: 11.3 ± 0.5 %). Consistently with previous studies, our phylogenetic analysis of ORF 7 sequences highlighted the peculiarity of strains circulating in Italy. Greater genetic distances between the strain circulating in a weaners' batch and strains from the farrowing unit and the previous batch were associated with increased mortality (p < 0.0001). All the respiratory and enteric coinfections contributed to an increase in losses (all p < 0.026), with secondary infections by Streptococcus suis and enteric bacteria also inducing an increase in AMU (both p < 0.041). Summary: Hepatitis E virus (HEV) is the leading cause of human enterically transmitted viral hepatitis occurring around the world both as outbreaks and as sporadic cases. The accumulating literature indicates that domestic pigs and wild boars are the main reservoirs of genotype 3 and genotype 4 for human infections in industrialized countries. However, the recent identification of HEV from various animal species poses additional potential concerns for HEV zoonotic infection. In this study, the role of sheep as potential host of hepatitis E virus (HEV) was investigated. By screening 192 sheep from seven farms located in Abruzzo Region (Southern Italy), HEV‐specific antibodies were detected in the sera of 41 animals (21.3%) whilst the RNA of HEV, genotype 3, was detected in 20 faecal (10.4%) and three serum samples (1.6%). Upon sequence analyses of a partial ORF2 gene region of eight HEV positive samples, the sheep sequences all grouped together within HEV genotype 3 subtype c, being most closely related to HEV strains identified in goat and wild boar from Abruzzo. This is the first study that demonstrates, serologically and molecularly, the presence of HEV in sheep population in a European country. [ABSTRACT FROM AUTHOR]
