Fusarium disease and the consequent mycotoxin accumulation pose significant problem in maize cultivation, with fumonisins produced by Fusarium verticillioides posing a global health concern. To address this issue, a range of preventive measures (e.g. crop management techniques) can be implemented to minimize fungal infections. A promising strategy to counteract this issue involves the selection of genotypes with greater resistance to fungal pathogens. This approach has the potential to reduce the reliance on chemical inputs for controlling fungus growth or indirect infection vectors. Leveraging genetic approaches can help improve the economic sustainability of agriculture in the face of climate change challenges. In the present work, we assessed the importance of two husk leaf traits (coverage and number), their association with F. verticillioides infection, fumonisin content, and their potential influence on crop yield. The study was conducted in three locations in the North of Italy and 38 hybrids with varying resistance to F. Verticillioides were compared. The results obtained showed that husk coverage has a pivotal role not only in protecting maize ears from Fusarium infection but have also a significant impact on crop yield: a significant positive correlation was found between husk coverage and yield in all three locations (r = 0.33185; r = 0.51327 and r = 0.51207, respectively). Furthermore, in the field of Vicenza, a significant negative correlation was found between husk coverage and Fusarium severity (r = −0.41492). Husk coverage emerges as an important trait that merits inclusion in maize breeding programs, given its protective role against fungal infections and its favourable influence on both yield and grain quality. Brown and black spots, caused by Stemphylium and Alternaria species, are important fungal diseases affecting European pear (Pyrus communis) in orchards. Both fungal genera cause similar symptoms, which could favor misidentification, but Alternaria spp. are increasingly reported due to the changing climatic conditions. In this study, Alternaria spp. were isolated from symptomatic leaves and fruits of European pear, and their pathogenicity was evaluated on pear fruits from cultivar Abate Fétel, and molecular and chemical characterization were performed. Based on maximum likelihood phylogenetic analysis, 15 of 46 isolates were identified as A. arborescens species complex (AASC), 27 as A. alternata, and four as Alternaria sp. Both species were isolated from mature fruits and leaves. In pathogenicity assays on pear fruits, all isolates reproduced the symptoms observed in the field, by both wound inoculation and direct penetration. All but one isolate produced Alternaria toxins on European pears, including tenuazonic acid and alternariol (89.1% of the isolates), alternariol monomethyl ether (89.1%), altertoxin I (80.4%), altenuene (50.0%), and tentoxin (2.2%). These isolates also produced at least two mycotoxins, and 43.5% produced four mycotoxins, with an average total concentration of the Alternaria toxins exceeding 7.58 × 106 ng/kg. Our data underline the potential risks for human health related to the high mycotoxin content found on fruits affected by black spot. Mycotoxin represents a significant concern for the safety of food and feed products, and wheat represents one of the most susceptible crops. To manage this issue, fast, reliable, and low-cost test methods are needed for regulated mycotoxins. This study aimed to assess the potential use of the electronic nose for the early identification of wheat samples contaminated with deoxynivalenol (DON) above a fixed threshold. A total of 214 wheat samples were collected from commercial fields in northern Italy during the periods 2014–2015 and 2017–2018 and analyzed for DON contamination with a conventional method (GC-MS) and using a portable e-nose "AIR PEN 3" (Airsense Analytics GmbH, Schwerin, Germany), equipped with 10 metal oxide sensors for different categories of volatile substances. The Machine Learning approach "Classification and regression trees" (CART) was used to categorize samples according to four DON contamination thresholds (1750, 1250, 750, and 500 μg/kg). Overall, this process yielded an accuracy of >83% (correct prediction of DON levels in wheat samples). These findings suggest that the e-nose combined with CART can be an effective quick method to distinguish between compliant and DON-contaminated wheat lots. A two-year (2018/19 and 2019/20) field experiment was carried out to evaluate the efficacy of recently developed fungicide combinations (with different modes of action) towards fungal diseases on seven bread and eight durum wheat varieties. The trial was performed at the FIELDLAB experimental station of the University of Perugia (Italy). The diseases were assessed under natural pressure except for Fusarium head blight (FHB) for which artificial inoculation with a Fusarium culmorum deoxynivalenol (DON)-producing strain was performed at the full flowering stage (BBCH 65). Fungicides were sprayed at the fully extended flag leaf (BBCH 39) and full flowering (BBCH 65) stages. The incidence of different fungal diseases was visually evaluated and other parameters [grains production (t/ha), protein content (%), test weight (kg/hL), and DON accumulation in grain (μg/kg)] were also determined. In the two years, characterized by different climatic conditions, the fungicide treatments showed efficacy in controlling the observed diseases (Septoria tritici blotch and FHB) as well as in reducing DON contamination. No significant differences were found between treatments. The results highlight that, in the present scenario of commercially available durum and bread wheat varieties, the timely application of the most common fungicides plays a crucial role for FHB and DON management in the presence of climatic conditions that are favorable to the disease. Summary: Seed health is one of the quality characteristics that must be assessed during the seed certification process. In this study, the freezing blotter method was used to detect fungi present in a total of 102 and 75 maize seed lots produced in 2019 and 2020 respectively, by eight companies in different geographic areas. The analysis of the seed samples produced in 2019 showed significant differences among companies and countries in the percentage of infection with Aspergillus spp. and Fusarium spp. The results obtained with the samples produced in 2020 were not statistically different. PCR-based tests were carried out on 90 isolates to identify the fungal species and differentiate the mycotoxigenic isolates. The results showed that Fusarium verticillioides and Aspergillus flavus were the most common species infecting seed and that 46.5% of F. verticillioides and 57.1% of A. flavus isolates were toxigenic. The most effective seed treatments against Fusarium spp. and Aspergillus spp. contained azoxystrobin and prothioconazole. [ABSTRACT FROM AUTHOR]

Since 2003, non-compliant aflatoxin concentrations have been detected in maize produced in Italy. The most successful worldwide experiments in aflatoxin prevention resulted from distribution of atoxigenic strains of Aspergillus flavus to displace aflatoxin-producers during crop development. The displacement results in lower aflatoxin concentrations in harvested grain. The current study evaluated in field performances of two atoxigenic strains of A. flavus endemic to Italy in artificially inoculated maize ears and in naturally contaminated maize. Co-inoculation of atoxigenic strains with aflatoxin producers resulted in highly significant reductions in aflatoxin concentrations (>90%) in both years only with atoxigenic strain A2085. The average percent reduction in aflatoxin B1 concentration in naturally contaminated maize fields was 92.3%, without significant differences in fumonisins between treated and control maize. The vegetative compatibility group of A2085 was the most frequently recovered A. flavus in both treated and control plots (average 61.9% and 53.5% of the A. flavus, respectively). A2085 was therefore selected as an active ingredient for biocontrol products and deposited under provisions of the Budapest Treaty in the Belgian Co-Ordinated Collections of Micro-Organisms (BCCM/MUCL) collection (accession MUCL54911). Maize grain yields and profitability are enhanced in temperate growing areas as a result of an early sowing. However, cool conditions may constrain the uptake of nitrogen (N) and phosphorus (P) by the root system during the early and more susceptible growth stages. The application of starter fertilizers near the plant roots could ensure better N and P availability and improve early vigor of the crop. The aim of this study has been to quantify the overall benefits of a localized starter fertilization with N or P, or their combination, on maize (early development, grain yield, grain quality and sanitary traits). A 4-year field experiment was conducted in NW Italy during the 2014–17 growing seasons. The trial compared the sub-surface placement of N, P and NP in bands close to the maize seed furrows, as a starter fertilization strategy, with unfertilized controls on sandy silt loam (SSL), silt loam (SL) and silty clay loam (SCL) soils. These soils were placed side by side, and were characterized by low, high and medium N and plant-available P contents, respectively. The results showed that when N and P were applied together, the early vigor (plant height) and canopy development (NDVI values) of the maize were significantly enhanced, compared to both the controls and to the application of the single elements in all the soils. The positive growth responses to starter NP fertilization were combined with accelerated crop phenological development, so that anthesis occurred 2.4–3.1 days earlier. Thus, the grain yield was enhanced following the NP sub-surface placement (+7%, +15% and +11%, in SSL, SL and SCL, respectively), mainly due to an increase in the number of kernels per ear (+10%), while the grain moisture content was reduced, compared to the controls, in all the soils (−2.2%). The application of N or P alone did not consistently affect either the grain yield or the moisture content in any of the three soils. The starter fertilization strategy also affected both the qualitative and sanitary grain traits. NP and N both resulted in an increase in the grain hardness in the SL and SCL soils, but when P was applied alone, no significant effects were observed. The use of NP starter fertilization significantly reduced the ear injuries caused by European Corn Borer in all three soils (−44%), and reduced ear rot severity in the SSL and SL soils and the fumonisin content (−72%) in the SL and SCL soils, compared to the untreated controls. The synergistic effect of N and P applied as a sub-surface band at sowing had a consistent positive effect on the advancement of the flowering dates at different sites in different years, even in soils that have tested high to P or N. Thus, the application of NP starter fertilizer is a crop practice that should be considered in North Italy and for similar soils and environments. Aspergillus carbonarius colonizes grapes and its derived products and produces ochratoxin A (OTA). In previous studies we screened 107 strains of A. carbonarius isolated from grapes for production of OTA, and we selected three high OTA-producing strains for this study (AC05, AC06, and AC07). The effect of different values of three conditions [temperature 15, 25, 30, 35°C; water activity (aw) 0.98, 0.95, 0.90, 0.88; and pH 4.0, 7.0, 9.0, 10.0] on A. carbonarius growth and OTA production was examined. A. carbonarius AC07 produced higher levels of OTA than AC06 and AC05 at all the variables tested. At 30°C the strains of A. carbonarius produced more OTA than at the other temperatures. A water activity of aw 0.98 produced the greatest mycelial growth and OTA accumulation for all three A. carbonarius strains. A pH of 4.0 produced the highest levels of OTA in all the strains. No growth was seen at aw 0.88 or at pH 10.0 in any strain, except AC07, which grew at pH 10.0. The optimal conditions for growth and production of OTA by A. carbonarius strains were 30°C, aw 0.98 and pH 4.0. When all the three factors (aw, pH, T) were combined, aw 0.98, pH 4.00 and 30°C produced the highest levels of OTA in all the strains. Maximum amounts of OTA were found 7-9 days after inoculation in all the strains. Temperature, water activity, and pH had a great impact on OTA production by A. carbonarius and these factors should be taken into account when developing management practices for future research programmes. Field experiments were set up from 2000 to 2002 in northwest Italy to determine the effects of sowing date on the susceptibility of maize hybrids to ear rot and mycotoxin contamination in natural infection conditions. Three sowing dates (March, April and May) were compared using two hybrids with different maturity classes. The ears were rated for the incidence and severity of disease symptoms at harvest and the harvested kernels were analysed for mycotoxin fumonisin B1, zearalenone, deoxynivalenol, aflatoxin B1 and ochratoxin A. The last sowing date resulted in higher values of European corn borer incidence (+39%), kernel Fusarium infection and fungal ear rot severity (increased respectively by a factor of 4 and 3) than the plots sowed in March. The sowing date did not influence the type of mycotoxin found in the kernel, which only depended on the climatic conditions during the season and on their influence on the infection and the development of the fungal species. The natural occurrence of fumonisin B1 and deoxynivalenol was always significantly higher in late-sowed plots. A late sowing, after May 10, multiplies the risk of the occurrence of fumonisins and deoxynivalenol in the grains at harvest by a factor of 11.2 and 1.9, respectively. No significant difference was observed for the contamination of zearalenone or aflatoxin B1 for the different sowing dates. An earlier sowing time than April, resulted in a significant reduction in mycotoxin contamination only in the years in which the meteorological trend protracted the kernel dry down to the autumn months. The use of a late-maturity hybrid with late sowing, compared with a medium-maturity hybrid, could lead to a serious risk of mycotoxin contamination. Pear, peach, apricot, and mixed juices (125 samples) were purchased in Italian supermarkets and organic food shops and analysed for patulin content. An overall incidence of 34.4? was observed in the fruit juices, with a mean contamination level of 3.6 µg kg-1. No one sample exceeded 50 µg kg-1, the maximum permitted limit according to European legislation. However, 19 positive samples contained more than 10 µg kg-1 of patulin, which is the maximum level permitted for baby food. The incidence of patulin contamination was significantly higher in pear juices (64.1?) than in apricot, peach, or mixed juices. Statistical analysis (p = 0.002; Kruskal-Wallis test) showed a significantly higher level of patulin in pear (5.1 µg kg-1) and mixed juices (4.9 µg kg-1) than in the other juices. A slightly higher incidence of positive samples was found in conventional juices (35.7?) compared with the organic ones (29.6?). The magnitude between the mean contamination levels was similar in conventional (3.6 µg kg-1) and organic (3.3 µg kg-1) juices (p = 0.474; Mann-Whitney U-test). Samples were further divided based on the fruit content in fruit juices, fruit nectars, and fruit drinks. In fruit nectars the incidence of positive samples (37.8?) was slightly higher than in fruit drinks (27.2?), but the mean patulin contamination was higher in fruit drinks (5.2 µg kg-1) than in fruit nectars (3.4 µg kg-1). However, the magnitude between the two means was not statistically different (p = 0.734; Mann-Whitney U-test). Italian juices had a higher incidence of patulin (35.3?), with a lower mean content (3.5 µg kg-1), compared with the incidence (22.2?) and level (4.1 µg kg-1) foreign juices commercially available in Italy. The magnitude of the means between Italian and foreign juices was not statistically different, according to the Mann-Whitney U-test (p = 0.616). The estimated intake of patulin ranged from 0.1 to 1.5 ng kg-1 body weight for the whole population and from 0.3 to 5.1 ng kg-1 body weight for the consumers only. The highest patulin intake was estimated for consumers of pear juices, followed by consumers of mixed juices. The daily intake of patulin derived from the consumption of pear juice suggests that also pear juices, though a minor patulin source, could be monitored for their patulin content in order to control the mycotoxin contamination, especially in countries, such as Italy, where pear nectars are preferred as fruit drinks. Abstract: Aflatoxin B1, a common occurring type of aflatoxin in feed, is a potent cancer-causing agent. Lactating cows fed with aflatoxin-contaminated feed trasmit the toxin, as the metabolic form aflatoxin M1, into the milk with high risk for consumers. The aim of this study is to determine the contamination degree of raw milk and feed in Valle d’Aosta region. From the beginning of 2004 to the end of 2005 samples of raw cow’s milk and cattle feed has been analysed: in 2004, the presence of aflatoxin M1 in milk and B1 in feed was higher than the maximum allowable in 1.7% of raw milk samples and in 8.1% of feed samples. In 2005 the presence of these aflatoxins are below the limits of EU regulations. Abstract: A survey on the occurrence of patulin was conducted during 2005 on commercial pure apple juices (53 samples) and mixed apple juices (82 samples) marketed in Italy. The current study was undertaken to investigate the possible influence of the agro-food production process employed (conventional or organic), of the fruit percentage in the commercial product (higher or lower than 50%) and of the type of apple juice (clear or cloudy) on the occurrence and level of patulin contamination. Patulin could be quantified in 34.8% of the samples ranging from 1.58 to 55.41μgkg−1. With the exception of one sample, the level of patulin was lower than 50μgkg−1, the maximum permitted threshold in fruit juices according to the European legislation. Mean levels of patulin were significantly lower in mixed apple juices (4.54μgkg−1) than in pure apple juices (9.32μgkg−1). Levels of patulin contamination were comparable in clear and cloudy juices. A similar incidence of positive samples was found in conventional and organic apple based juices, and the magnitude between the mean contamination levels, although higher in organic (10.92μgkg−1) than in conventional juices (4.77μgkg−1), was not statistically significant (p =0.771; Mann–Whitney test). The magnitude between the means of patulin contamination in juices containing more than 50% fruit (11.26μgkg−1) and in juices with 50% or less fruit (3.35μgkg−1) was statistically significant (p =0.016; Mann–Whitney test). A study of apple juice products sold in Italy and South Africa was initially carried out on 20 samples bought in Cesena, Italy, and Tygerberg in Cape Town, South Africa. The samples were bought at random and analysed for patulin contamination. All 12 of the Italian samples had no detectable levels of patulin, except one, which was just slightly above the lowest regulatory limit of 10 ng ml-1. On the other hand, five of the eight South African samples were all contaminated with patulin levels above 10 ng ml-1, with one showing a concentration of 75 ng ml-1, well above the highest regulated limit of 50 ng ml-1. This latter result led to a more targeted investigation with 14 samples being purchased in the low-income areas of Tygerberg where the initial samples were sourced. These samples confirmed that there might be a problem of mycotoxin contamination in apple juices products sold to low-income consumers because half of the samples showed patulin contamination of which four had levels well above the acceptable limits. This is the first study in South Africa to look at apple juice products in low-income areas and it points to a need to intervene and introduce quality systems in the supply chain of the manufacture and packaging of apple juice products by independent small business. The occurrence of patulin was investigated in 100 conventional and 69 organic fruity foodstuffs samples commercially available in Italy by using an HPLC method with a limit of quantification of 0.5?µg?kg -1 . Patulin was detected in 26 (26%) conventional and 31 (45%) organic products with a significantly higher ( p <0.01) mean concentration in the organic products (4.78 vs. 1.15?µg?kg -1 ). Mean patulin concentrations in conventional apple juices, pear juices, other juices and fruits purees were 3.14, 0.22, 0.19, 0.11?µg?kg -1 , respectively, and 7.11, 11.46, 2.10, 0.18?µg?kg -1 in the relevant organic products. Four samples of juices (one conventional and two organic apple, and one organic pear) contained patulin at concentrations above the limit of 50?µg?kg -1 , four at concentrations between 10 and 25?µg kg -1 , and the remaining ones below 10?µg?kg -1 . Patulin was detected (<1?µg?kg -1 ) in only three of the 23 fruity baby food samples tested (homogenized fruits, 11 conventional and 12 organic). Based on the available data on Italian intakes of fruit juices, the estimated daily intakes of patulin, were 0.38 and 1.57?ng?kg -1 body weight (bw) from conventional and organic products, respectively. Estimated daily intakes of patulin for children were higher, 3.41?ng?kg -1 bw from conventional and 14.17?ng?kg -1 bw from organic products, but largely below the provisional maximum tolerable daily intake (PMTDI) of 400?ng?kg -1 bw. Patulin was also found in two samples of organic apple vinegar (<5?µg?kg -1 ) and in fresh apples with rotten spots (12 out of 24 samples) with maximum levels at 16?402 and 44?572?µg?kg -1 for conventional and organic apples, respectively. Ochratoxins are a group of mycotoxins that frequently occur as contaminants in agricultural commodities and foods, including dry-cured meats and cheeses. The fungus Aspergillus westerdijkiae is frequently isolated from aged foods and can produce ochratoxin A (OTA). However, individual strains of the fungus can have one of two OTA production phenotypes (chemotypes): OTA production and OTA nonproduction. Monitoring and early detection of OTA-producing fungi in food are the most effective strategies to manage OTA contamination. Therefore, we examined genome sequence data from five A. westerdijkiae strains isolated from the surface of cheese from southern Italy to identify genetic markers indicative of the twoOTA chemotypes. This analysis revealed a naturally occurring deletion of the OTA regulatory gene, otaR , in an OTA-nonproducing isolate.We used this information to design a polymerase chain reaction (PCR) method that could identify A. westerdijkiae and distinguish between the two OTA chemotypes. In this method, the PCR primers were complementary to conserved sequences flanking otaR and yielded different-sized amplicons from strains with the different chemotypes. The primers did not yield ota -region-specific amplicons from other OTA-producing species. Mycotoxins' exposure by inhalation and/or dermal contact can occur in different branches of industry especially where heavily dusty settings are present and the handling of dusty commodities is performed. This study aims to explore the possible contribution of the occupational exposure to aflatoxins by analysing urine samples for the presence of aflatoxins B 1 and M 1 and aflatoxin B 1 -N 7 -guanine adduct. The study was conducted in 2017 on two groups of volunteers, the workers group, composed by personnel employed in an Italian feed plant (n = 32), and a control group (n = 29), composed by the administrative employees of the same feed plant; a total of 120 urine samples were collected and analysed. A screening method and a quantitative method with high-resolution mass spectrometry determination were developed and fully validated. Limits of detections were 0.8 and 1.5 pg/mL urine for aflatoxin B 1 and M 1 , respectively. No quantitative determination was possible for the adduct aflatoxin B 1 -N 7 -guanine. Aflatoxin B 1 and its adduct were not detected in the analysed samples, and aflatoxin M 1 , instead, was found in 14 samples (12%) within the range 1.9-10.5 pg/mL urine . Only one sample showed a value above the limit of quantification (10.5 pg/mL urine ). The absence of a statistical difference between the mean values for workers and the control group which were compared suggests that in this specific setting, no professional exposure occurs. Furthermore, considering the very low level of aflatoxin M 1 in the collected urine samples, the contribution from the diet to the overall exposure is to be considered negligible.

The occurrence of ochratoxin A (OTA) in matured meat products can be attributed to mycotoxin coming from raw materials and/or molds that develop on the product surface during ripening. This work aimed to evaluate OTA production by Aspergillus westerdijkiae inoculated on the surface of sausages and its diffusion into the product throughout ripening, study the effect of relative humidity (RH) on the production of this mycotoxin, and investigate the presence of OTA in dry fermented sausages that naturally present intense contamination by A. westerdijkiae. Italian-type sausages were surface inoculated with A. westerdijkiae and two distinct experiments were performed. In the first, the sausages were matured in a chamber with decreasing RH (from 95 to 75%) for 35 days. In the other, the sausages were incubated under different RH (79, 85 or 95%) for 21 days. Samples were taken at the beginning of the experiments and every 7 days, subdividing into casing, outer border, and core for analyses. Sausage samples naturally spoiled by ochratoxigenic fungi were collected during sanitary inspection. Even in the presence of A. westerdijkiae mycelia, no OTA was detected for up to 7 days of sausage maturation. On the other hand, this study demonstrated that the growth of A. westerdijkiae on salami surface produces high amounts of OTA on the casing and allows its diffusion through the casing with contamination to the outer border of sausages. In the same way, it shows that under similar water activity values of substrate, RH influences the amount of OTA produced. Mycotoxins are fungal secondary metabolites identified in many agricultural products screened for toxigenic moulds. They have been reported to be carcinogenic, teratogenic, tremorogenic, haemorrhagic and dermatitic to a wide range of organisms. With the increasing stringent regulations for mycotoxins imposed by importing countries such as those of the European Union, many cereals that are not safe for human consumption are used in formulations intended for animal feed. Gamma-rays are reported in the scientific literature to destroy ochratoxin A and aflatoxin in food crops and feed. The present study provides preliminary data for establishing the effect of dose of gamma-irradiation, ranging from 0 to 15 kGy, on aflatoxins and ochratoxin A reduction in commercial animal feed. The mycotoxin levels were determined by means of immunoaffinity clean-up (IAC) and HPLC with fluorescence detection (HPLC-FLD). The maximum reductions found at 15 kGy were 23.9%, 18.2%, 11.0%, 21.1% and 13.6% for ochratoxin A, aflatoxin B₁, aflatoxin B₂, aflatoxin G₁ and aflatoxin G₂, respectively. Results showed that the gamma-rays even at 15 kGy were not effective in the complete destruction of ochratoxin A and aflatoxins in the tested feed.

In the present study, the occurrence of eighteen mycotoxins, nine trichothecenes (deoxynivalenol, 3-acetyl-deoxynivalenol, 15-acetyl-deoxynivalenol, nivalenol, neosolaniol, diacetoxyscirpenol, fusarenon-X, T-2 toxin and HT-2 toxin), three zearalenones (zearalenone, α-zearalenol and β-zearalenol), and six emergent mycotoxins, beauvericin and five enniatins (A, A1, B, B1 and B4), was monitored in different Italian organic cereals and cereal products by using a liquid chromatography coupled to triple quadrupole mass spectrometry method. A total of 93 organic cereal samples (wheat, barley, rye and oat) were collected from Italy. Limits of quantification ranged from 5 to 15 μg/kg. 80% of analyzed samples contained mycotoxins. The occurrence was 33%, 6.5%, 2%, 27%, 7%, 10% and 43% for deoxynivalenol, HT-2, T-2, nivalenol, zearalenone, beauvericin and enniatins, respectively. The major mycotoxin found was enniatin B4; it was detected in 40% of all analyzed samples and its levels ranged from 5.7 to 284.2 μg/kg.
