The metabolite fingerprinting of four Italian commercial bean seed cultivars, i.e., Phaseolus Cannellino (PCANN), Controne (PCON), Vellutina (PVEL), and Occhio Nero (PON), were investigated by Nuclear Magnetic Resonance (NMR) spectroscopy and multivariate data analysis. The hydroalcoholic and organic extract analysis disclosed more than 32 metabolites from various classes, i.e., carbohydrates, amino acids, organic acids, nucleosides, alkaloids, and fatty acids. PVEL, PCON, and PCANN varieties displayed similar chemical profiles, albeit with somewhat different quantitative results. The PON metabolite composition was slightly different from the others; it lacked GABA and pipecolic acid, featured a higher percentage of malic acid than the other samples, and showed quantitative variations of several metabolites. The lipophilic extracts from all four cultivars demonstrated the presence of omega-3 and omega-6 unsaturated fatty acids. After the determination of the total phenolic, flavonoids, and condensed tannins content, in vitro antioxidant activity was then assessed using the DPPH scavenging activity, the ABTS scavenging assay, and ferric-reducing antioxidant power (FRAP). Compared to non-dark seeds (PCON, PCANN), brown seeds (PVEL, PON) featured a higher antioxidant capacity. Lastly, only PON extract showed in vitro antifungal activity against the sclerotia growth of S. rolfsii, by inhibiting halo growth by 75%. [ABSTRACT FROM AUTHOR]

Many fungi belonging to Botryosphaeriaceae are well-known as causal agents of diseases in economically and ecologically important agricultural crops and forest trees. In Italy, the high diffusion of Botryosphaeriaceae infections observed over the last decade, has shown the importance of this group of fungi, which are becoming limiting factors for plant production in agricultural systems, nurseries and natural and urban landscapes. Global warming and stress factors such as occasional extreme climatic events can affect the susceptibility of host plants, as well as fungus behaviour, increasing the risk of future infections. Available reports of Botryosphaeriaceae in Italy have been examined, focusing on wood and fruit pathogens, resulting in a list of ten genera and 57 species. Diplodia is the most widespread genus in Italy with 76 records on 44 hosts, while at species level, Neofusicoccum parvum, Botryosphaeria dothidea and Diplodia seriata show the widest host ranges and many records. The ability of the pathogens to remain latent on asymptomatic plants, and uncontrolled trade of plant materials among countries, facilitate the dissemination and potential introduction of new Botryosphaeriaceae species. Preventive detection and adequate control strategies are always needed to limit the potential damage caused by Botryosphaeriaceae. Modeling phenological phases in a Mediterranean environment often implies tangible challenges to reconstructing regional trends over heterogenous areas using limited and scattered observations. The present investigation aimed to project phenological phases (i.e., sprouting, blooming, and pit hardening) for early and mid–late olive cultivars in the Mediterranean, comparing two phenological modeling approaches. Phenoflex is a rather integrated but data-demanding model, while a combined model of chill and anti-chill days and growing degree days (CAC_GDD) offers a more parsimonious and general approach in terms of data requirements for parameterization. We gathered phenological observations from nine experimental sites in Italy and temperature timeseries from the European Centre for Medium-Range Weather Forecasts, Reanalysis v5. The best performances of the CAC_GDD (RMSE: 4 days) and PhenoFlex models (RMSE: 5–9.5 days) were identified for the blooming and sprouting phases of mid–late cultivars, respectively. The CAC_GDD model was better suited to our experimental conditions for projecting pit hardening and blooming dates (correlation: 0.80 and 0.70, normalized RMSE: 0.6 and 0.8, normalized standard deviation: 0.9 and 1.0). The optimization of the principal parameters confirmed that the mid–late cultivars were more adaptable to thermal variability. Aim: Measuring the phenomenon of gambling and Internet addiction, with analysis of attitudes and psychophysical consequences among nurses working in different care settings. Methods: An observational, cross sectional, multicenter study was conducted from April to September 2020. Participants' socio-demographic information, the "Internet Addiction Test" (IAT) scale, and the "South Oaks Gambling Screen" (SOGS) were collected in order to assess the overuse of and whether an individual has a problematic relationship with gambling, respectively. Results: 502 nurses were enrolled in the study. Significant correlations were found (p <.001) between the IAT score and gender, number of years of work experience, job role, educational qualification; and between the SOGS and gender, number of years of work experience, job role and regions of Italy. Conclusions: The study highlighted an emerging social problem, and the results may be just the tip of the iceberg. Given the lack of knowledge of these phenomena and a high percentage of people who suffer from them but are afraid to admit it and get help, this study could also be useful in expanding knowledge and allow more professionals to get help and learn about possible treatments and cures for the resolution of these addictions. Forest management is a complex topic at the interface between sustainability and the resilience of socioeconomic and environmental systems. The influence of market forces, supranational, country and regional policies, as well as climate change, on forest goods and services, is expected to increase in the near future. Such a complex interplay between economic and environmental forces is common to most European countries. The aim of this study is to operationally delineate and discuss the transition of the environmental sustainability and economic viability of forestry in Italy. This country encompasses the typical Mediterranean ecosystems with broad forest coverage in mountainous and hilly areas, where expanding woodland areas have been observed in the last decades mainly as a consequence of the decline of agropastoral activities, especially in disadvantaged and marginal areas. The consequent increase in wood biomass was frequently conceived as an element of environmental criticality, exposing woods to high vulnerability to wildfires and a consequent reduction in their economic value, possibly exacerbated by local warming. These dynamics usually took place in contexts where only a part of the overall forest heritage was subjected to regular management, despite the efforts made through various policies such as the Constitutional Law 3/2001 and the recent Law on Forests and Forestry Supply Chains. The latter policy tool, enhancing the concept of "active forest management" aimed to establish a sustainable approach to forest resources, leading to a broader forest area for formal planning and controlled harvesting. These dynamics took place in parallel with the inherent expansion in forest certification schemes formally promoting long-term environmental sustainability and a wider spectrum of forest ecosystems. During the summer season, the Italian territory is vulnerable to extended wildfires, which can have dramatic impact on human activities and ecosystems. Such wildfire events are usually associated with the presence of drought conditions and are generally more severe in southern Italy, owing to the high temperatures and reduced precipitation that characterize this geographical region. In this work, we discuss the statistical analysis of the burned area (BA) in Italy and build simple data-driven models linking BA to different climatic drivers, comparing the relevance of direct surface soil moisture information to that provided by drought indices such as the Standardized Precipitation Index and the Standardized Precipitation Evapotranspiration Index (SPEI). We show that considering surface soil moisture alone is sufficient to produce reliable out-of-sample predictions in a large part of the country. Potatoes (Solanum tuberosum L.) are one of the most valuable agricultural crops, and the flesh of these tubers provides various classes of healthy compounds important for human nutrition. This work presents the results of a joint analysis of different chemical classes of compounds which provided insights on the metabolic characterization of pigmented and non-pigmented potato varieties collected from Italy. The identification of common or individual metabolic characteristics across the omic datasets (antioxidants, total polyphenolic content, polyphenols, and sugars) is conducted by Joint and Individual Variation Explained (JIVE), a data fusion multivariate approach. The common part of the multivariate model allowed the separation between non-pigmented and pigmented samples. Polyphenolic compounds were mainly responsible for the separation between purple-fleshed and red-skinned potatoes. An additional detailed analysis of the anthocyanin composition, including the acylated anthocyanins, allowed to pinpoint the diversities between the pigmented potato groups. Furthermore, the presence of an appreciable amount of hydroxycinnamic acids and anthocyanins in the purple-fleshed varieties, which are also characterized by a lower content of sugars, is found. Once a much more globally widespread crocodylian clade, Tomistominae is today represented by just one species, Tomistoma schlegelii (the false gharial), restricted to south-east Asia. Although tomistomine fossil occurrences are recognized from the early Eocene (∼55 Ma) onwards, their remains are often incomplete, making appropriate taxonomic classification within the group problematic. This is especially pertinent to several taxa from the Miocene of Europe, which were historically erected from fragmentary remains. Here we re-examine and describe four approximately contemporaneous taxa from Malta and Italy to determine their taxonomy and phylogenetic affinities: Melitosaurus champsoides, Tomistoma calaritanum, Tomistoma gaudense and Tomistoma lyceense. We place them into a phylogenetic analysis for the first time, comprising 70 taxa scored for 244 characters, several of which are revised or novel, and apply a number of character weighting strategies. Whereas 'Tomistoma lyceense' is deemed to be an indeterminate tomistomine, a unique combination of features confirms Melitosaurus champsoides, Tomistoma calaritanum and Tomistoma gaudense as three distinct species. These three taxa are recovered as derived tomistomines, with characters such as a posterior maxillary process between the lacrimal and nasal, large supratemporal fenestrae that are wider than long, and the posteromedial alignment of the last three premaxillary teeth, suggesting a close relationship with the approximately contemporaneous European taxa, Tomistoma lusitanica and Gavialosuchus eggenburgensis. It is unlikely that any of these species belong to Tomistoma, with the possibility that they can all be classified under Melitosaurus and Gavialosuchus. However, we retain them in open nomenclature pending reassessment of the remaining European Miocene tomistomines. Our taxonomic and phylogenetic revision helps to elucidate past tomistomine diversity in the Miocene of the Mediterranean region, prior to the group's extirpation, and is an important first step in resolving the complicated history of European tomistomine systematics. A geological map at 1:10,000 scale is presented that shows the distribution of the metamorphic rock units in the area of Montigiu (Mt.) Nieddu (NE Sardinia). These units belong to the High-Grade Metamorphic Complex in the Inner Zone of the Variscan chain of Sardinia. The rocks include different types of migmatite, amphibolite, and retrogressed eclogites. The geological map and cross section show the complex tectonic and metamorphic setting of the area, whereas the metamorphic assemblages and deformation framework represent a valuable tool for the reconstruction of the P-T metamorphic evolution for the crustal sectors involved in the Variscan orogeny. The metamorphic rock units reported in the map derived from the lower and upper continental plates involved in the Variscan collision and were tectonically juxtaposed during the early Carboniferous exhumation stage. Background: Following the development of the COVID-19 pandemic, a rigid public health strategy of reduced social contact and shelter-in-place has been adopted by the Italian Government to reduce the spread of the virus. In this paper, we aim at evaluating the impact that the COVID-19 pandemic, and the relative containment measures, have had on a real-life sample of patients suffering from substance use disorders (SUDs) and/or behavioral addictions. Methods: An anonymous questionnaire was filled out by 153 addicted patients, both outpatients and residential inpatients, recruited across Italy and highly representative of the current Italian population suffering from addictions. Psychopathological burden (anxiety and depressive symptomatology, somatization, irritability, and post-traumatic symptoms), quality of life, and craving changes in daily habits were assessed. Results: In our sample, we found moderate rates of depression (22.9%), anxiety (30.1%), irritability (31.6%), and post-traumatic stress (5.4%) symptoms. Psychopathological burden was globally higher among residential patients. Reported levels of craving were generally low. Discussion: This study is the first attempt to collect Italian data regarding the effects of the rigid quarantine period, during the COVID-19 pandemic, on patients suffering from a SUD and/or behavioral addictions. The presence of a moderate psychopathological burden correlated to poor quality of life and low craving scores represented the main outcomes. The pyroclastic rocks, belonging to the calcalkaline volcanic activity occurred in Sardinia between 32 and 11 Ma ago, were widely used as construction material in several Romanesque Churches of the easternmost Logudoro, as well as in the Sardinia territory. In this work was taken as a case study the Church of Nostra Signora di Otti (12th cent.). There are no historical or archaeological evidences of the ancient quarries. Based on the geochemical, petrographic and volcanological characters of several samples from an extensive field area (approximately 150 kmq), a geographical zoning of the Oligo-Miocenic volcanics has been recognised. Internally to the Oschiri sector, according to TiO vs Differentiation Index diagram, there are three different sub-zones with pyroclastics: less fractionated (D.I. ~70 ÷ 78); intermediate fractionated (D.I. ~76 ÷ 79); more fractionated (D.I. ~77 ÷ 82). To identify the origin of the ignimbrites of the Church, two statistical methods have been used: stepwise linear discriminant and canonical analysis. Moreover, to define the geochemical transformation processes induced by the alteration, by XRD analysis and XRF, SEM-EDS data, a comparative study of concentrations of major and trace elements measured between the surface portion and the innermost of the stone has been made. The present study assesses the level of sensitivity to desertification of forest types in Italy between 2000 and 2010 on a fine resolution scale using the Environmental Sensitive Area (ESA) scheme. The proposed methodology identifies and ranks the level of sensitivity of fourteen forest types and quantifies the changes in their level of sensitivity over time as a contribution to understanding of complex landscape-forest interactions in Mediterranean ecosystems. Only few forest types showed a relatively high sensitivity level, suggesting that forests may positively contribute to the mitigation of land degradation processes in the Mediterranean region. Forest types showing the highest sensitivity are native types mostly adapted to dry Mediterranean landscapes, introduced vegetation types and highly-fragmented and heterogeneous forest types. Results suggest that: (i) high-quality and biodiversity-rich forest types (e.g., beech, mountain pine forests) may act as vegetation buffer mitigating the increase of land sensitivity to desertification at the landscape scale; and (ii) the remaining forest types (especially highly fragmented, low-quality or low-biodiversity classes in areas with severe soil and climate conditions) may undergo increases in land sensitivity to desertification and should be protected through specific management measures as also implemented in the framework of the National Action Plans to Combat Desertification. In the last 30 years, the spread of emerging and invasive fungal pathogens has had a profound impact on plants, animals and humans, causing crop losses and damaging their health with economic and social impact. Awareness of the biodiversity of these pathogens is essential for their detection, identification and control. In this article, their diversity in Italy is reviewed taking into consideration the international context. Fungal endophytes, important ecological agents whose lifestyle can be switched by stochastic events from beneficial/neutral to pathogenic, are included in this review. [ABSTRACT FROM PUBLISHER]

Two phytotoxic metabolites, p-hydroxybenzaldehyde and indol-3-aldehyde, were isolated and identified by spectroscopic methods from solid cultures of Discula quercina, an endophytic fungal pathogen frequently associated with oak decline in Italy. In addition, the fungus produced an unusual acyclic keto acid as a major metabolite, which was identified by spectroscopic and chemical methods as 5-oxo-6 E,8 E-octadecadienoic acid. In leaf puncture assays on Quercus suber and Q. ilex leaves, indol-3-aldehyde proved to be more toxic than p-hydroxybenzaldehyde, while 5-oxo-6 E,8 E-octadecadienoic acid was inactive even at the highest concentration used (1 mg ml). Although indol-3-aldehyde is a known microbial metabolite, its phytotoxic activity has hitherto not been reported. In phenological studies, the plant developments are analysed considering their relationships with seasonal meteorological conditions; moreover, the influences of geographical features on biological responses have to be also considered. Different studies analysed the influence of latitude on phenological phases to investigate the possible different magnitude of biological response. In our experience, this type of geographic evaluation was conducted considering one of the more important plant species of Mediterranean shrub, the olive ( Olea europaea L.) in fifteen olive monitoring stations, four located in Tunisia and eleven in Italy, from the southern Zarzis area at 33° to the northern Perugia area at 43° of latitude. The olive flowering phenomenon was studied, utilising an aerobiological monitoring method through appropriate pollen traps located inside olive groves from 1999 to 2008. The olive monitored pollen grains were recognised and evaluated to obtain daily pollen concentrations to define the flowering dates in the different study areas. The biometeorological statistical analysis showed the 7°C threshold temperature and the single triangle method for growing degree days (GDD) yearly computing as the better ones in comparison to others. Moreover, the regression analysis between the dates of full flowering and the GDD amounts at the different monitoring latitudes permitted us to evidence the biological response of olive species in geographic regions with different climate patterns. The specific biological response at different latitude was investigated, the slope results, as flowering days per heat amounts, evidenced that olive species behaviours are very constant in consequence to similar meteorological conditions independently to latitude variations. Averagely, the relationships between plant's phenology, temperature trends and geographical features are very close, even if the yearly mesoscale meteorological variations force to consider, year by year, phenological advances or delays as local events. The metabolite profile of fresh Goji berries from two cultivars, namely Big Lifeberry (BL) and Sweet Lifeberry (SL), grown in the Lazio region (Central Italy) and harvested at two different periods, August and October, corresponding at the beginning and the end of the maturation, was characterized by means of nuclear magnetic resonance (NMR) and electrospray ionization Fourier transform ion cyclotron resonance (ESI FT-ICR MS) methodologies. Several classes of compounds such as sugars, amino acids, organic acids, fatty acids, polyphenols, and terpenes were identified and quantified in hydroalcoholic and organic Bligh-Dyer extracts. Sweet Lifeberry extracts were characterized by a higher content of sucrose with respect to the Big Lifeberry ones and high levels of amino acids (glycine, betaine, proline) were observed in SL berries harvested in October. Spectrophotometric analysis of chlorophylls and total carotenoids was also carried out, showing a decrease of carotenoids during the time. These results can be useful not only to valorize local products but also to suggest the best harvesting period to obtain a product with a chemical composition suitable for specific industrial use. Finally, preliminary studies regarding both the chemical characterization of Goji leaves generally considered a waste product, and the biological activity of Big Lifeberry berries extracts was also investigated. Goji leaves showed a chemical profile rich in healthy compounds (polyphenols, flavonoids, etc.) confirming their promising use in the supplements/nutraceutical/cosmetic field. Oaks are dominant and key tree species in Mediterranean forest ecosystems. However, in recent decades, oak forests have been heavily impacted by oak decline, a worldwide phenomenon exacerbated by climate change. The charcoal disease agent Biscogniauxia mediterranea is involved in the decline of Mediterranean oak formations in a variety of contexts. Here, we investigated the impact and role of B. mediterranea in the decline of oaks in Castelfidardo Forest, a relict wood of the late Holocene and a Site of Community Importance. We established five plots within which we recorded tree positions, any symptoms and signs of decline, association of B. mediterranea to declining trees, and deadwood and associated mycota. Of 471 oaks inspected, 7.0% showed brownish exudates on the stems, 46.9% showed epicormic shoots along the main trunk, and 24.4% showed black carbonaceous stromata on diseased branches and trunks. The decline was most severe for Quercus cerris, which comprised plots #4 and #5, at 50.0% (81/162 trees) and 29.0% (33/114), respectively; then for Quercus robur for plot #3, at 40.0% (38/95); and finally for Quercus pubescens for plots #1 and #2, at 13.7% (7/51) and 12.3% (6/49), respectively. Bark tissues were collected from trees with charcoal cankers and taken to the laboratory for microscopic examination and identification by mycological and molecular methods. This investigation revealed a close association between oaks with pronounced reduction of vitality and incidence of B. mediterranea. Deadwood was equally distributed among the five plots, and was heavily colonized by Basidiomycota. The high incidence of the charcoal canker pathogen B. mediterranea appeared to be related to environmental stresses. However, the absence of silvicultural management, high competition among physiologically mature trees, and the geographic isolation of this residual forest may have predisposed oaks to decline. The employment of multi-species starter cultures has growing importance in modern winemaking for improving the complexity and wine attributes. The assessment of compatibility for selected species/strains at the industrial-scale is crucial to assure the quality and the safety associated with fermentations. An aspect particularly relevant when the species belong to non-Saccharomyces, Saccharomyces spp. and malolactic bacteria, three categories with different biological characteristics and oenological significance. To the best of our knowledge, the present report is the first study regarding the utilization of a combined starter culture composed of three strains of non-Saccharomyces, Saccharomyces cerevisiae and Lactobacillus plantarum for production of wine at the industrial scale. More in-depth, this work investigated the oenological potential of the autochthonous characterized strains from the Apulian region (Southern Italy), Candida zemplinina (syn. Starmerella bacillaris) 35NC1, S. cerevisiae (NP103), and L. plantarum (LP44), in co-inoculation following a complete scale-up scheme. Microbial dynamics, fermentative profiles and production of volatile secondary compounds were assessed in lab-scale micro-vinification tests and then the performances of the mixed starter formulation were further evaluated by pilot-scale wine production. The above results were finally validated by performing an industrial-scale vinification on 100HL of Negroamaro cultivar grape must. The multi-starter formulation was able to rule the different stages of the fermentation processes effectively, and the different microbial combinations enhanced the organoleptic wine features to different extents. The findings indicated that the simultaneous inoculation of the three species affect the quality and quantity of several volatile compounds, confirming that the complexity of the wine can reflect the complexity of the starter cultures. Moreover, the results underlined that the same mixed culture could differently influence wine quality when tested at the lab-, pilot- and industrial-scale. Finally, we highlighted the significance of employment non-Saccharomyces and L. plantarum, together with S. cerevisiae, autochthonous strains in the design of custom-made starter culture formulation for typical regional wine production with pronounced unique quality.
