Background: Dermatophytes are a group of keratinophilic fungi of medical importance. Despite a relatively long history of molecular taxonomic studies, there is still a need for information on genetic polymorphism in wider variety of genomic loci., Objectives: Our goal was to study partial DNA topoisomerase 2 gene (TOP2) polymorphism in dermatophytes., Methods: We performed DNA sequencing of TOP2 in 26 dermatophyte species along with ribosomal internal transcribed spacer (ITS) sequencing., Results: The number of polymorphic sites in TOP2 data set was similar to that one in ITS data set. Nannizzia species formed paraphyletic group in TOP2 tree. Trichophyton simii was paraphyletic in concatenated TOP2-ITS tree, one of its two clades contained solely Iranian isolates., Conclusions: Our results revealed several unresolved problems in the taxonomy of dermatophytes, including probable polyphyly of the genus Nannizzia and the species T simii., (© 2020 Blackwell Verlag GmbH.)

Misidentifying with Microsporum gypseum has for a long time been accounted for less prevalence of the geophilic species, Microsporum fulvum in human dermatophytosis. We describe a new case of infection with the species in an Iranian young man. Direct examination of skin scrapings revealed a tinea corporis, and morphological study of the recovered isolate from the culture resulted in the identification of M. gypseum. However, PCR amplification of ITS1-5.8S rDNA-ITS2 region and subsequent ITS-RFLP and sequencing were indicative of M. fulvum as the true causative agent. To recognize M. fulvum in human infections and to validate the morphologically distinguished isolates of M. gypseum, the genetic-based identification is strongly recommended.

Microsporum ferrugineum, an uncommon causative agent of dermatophytosis, has restricted endemicity. Iranian strains suspected to be M. ferrugineum from two patients with tinea were analysed using the rDNA internal transcribed spacer (ITS) region and the partial β-tubulin (BT2) and translation elongation factor 1-α (TEF1) genes. Strains were compared to reference strains to differentiate M. ferrugineum from its relatives Microsporum canis and Microsporum audouinii. Inter-species differences for TEF1 and BT2 were found to be higher than for the ITS region, which is the current molecular standard for species identification in dermatophytes. Intra-species variation was zero for each of the markers. In silico analysis showed that the restriction enzymes BanI and BshNI were together sufficient to differentiate the three species based on TEF1, whereas a two-step digestion was needed with BT2 or the ITS region. The prevalence of M. ferrugineum in clinical samples in Iran appeared to be higher than suspected on the basis of routine phenotypic identification.
