Groundwater contamination in India exposes an estimated 70 million people to arsenic. Over 10 million people in the state of Bihar (India) are at risk of arsenic poisoning. As reported 27 districts of the state's thirty-eight districts are under a state of catastrophe. Symptoms noticed by those who have been exposed to the arsenic caused disease are-lack of appetite, neurobehavioral problems, hyperkeratosis, and melanosis on the skin. So, this study's primary objective is to find novel approaches to treat arsenic poisoning in rats using the Charles Foster model. Following permission from the Institutional Animal Ethics Committee, the animals were divided into three groups: one group served as a control, the second group received arsenic treatment, and the third group received Ganoderma lucidum extract after arsenic treatment. For the arsenic group, the rats were given 8 mg/kg body weight of sodium arsenite orally every day for 90 days, and then for 60 days, they were given 80 mg/Kg body weight of Ganoderma lucidum extract via gavage. Their biochemical values, including those of the liver and kidneys, were found to be increased. In addition, their levels of free radicals, including lipid peroxidation, were measured and found to be substantially higher. In addition, the levels of arsenic in the kidney and liver tissue were very high. However, biochemical and lipid peroxidation levels were significantly restored after administration of Ganoderma lucidum ethanolic extract. Arsenic levels in rat liver and kidney tissues were also found to be reduced. Ganoderma lucidum has a therapeutic impact against arsenicinduced toxicity, according to the present research. Arsenic contamination is a serious issue throughout the world and is substantial risk factor in most of the countries including India. Chronic arsenic exposure from drinking water to humans is causing major public health related issues. The present study aims to investigate the ameliorative effect of Citrullus lanatus (Watermelon) seeds against arsenic induced reproductive function and testicular toxicity in Charles Foster rats. In the present study, twenty-four male Charles Foster rats (120±5gm). The study group includes group - I (n=6) as control, and group - II (n=18) treated with sodium arsenite (8 mg per Kg body weight per day) for 90 days. The group - II was furthermore divided into three sub-groups, Group - II-a, Group - II-b and Group - II-c. The group - II-a were sacrificed to see the effect of arsenic toxicity effect after arsenic exposure. Group - II-b was left with normal food and water for next 60 days to observe auto recovery. The group - II-c rats were administered with Citrullus lanatus (Watermelon) hydroxyl ethanolic seeds extract at the dose of 400 mg per Kg body weight per day for 60 days for 90 days upon pre-treated sodium arsenite treated group (8 mg per kg body weight). After completion of the experiment, all the treated group of animals were sacrificed for biochemical, hormonal assay, histopathological, testicular and reproductive functions study. In the arsenic treated rats' group, there was significant (p<0.001) changes in serum levels of SGPT, SGOT, urea, uric acid and creatinine as well as in haematological parameters. There was decrease in the sperm counts and sperm motility accompanied by an increased incidence of sperm abnormalities and hormonal imbalance denotes infertility condition. In contrast, after the administration with C.lanatus (Watermelon) seeds hydroxy-ethanolic extract upon arsenic treated rats group, there was significant (p<0.001) improvement observed in the reproductive, hepatic and renal parameters. In the arsenic intoxicant rats' group, after administration with C. lanatus (Watermelon) seeds hydroxyl ethanolic extract, there was significant (p<001) reduction in the arsenic concentration in blood, liver, and kidney tissues as well as serum lipid peroxidation. The histopathological study also showed the C. lanatus (Watermelon) seeds hydroxy ethanolic extract significantly restored the cellular integrity of testicular cells leading to normal functioning of it against arsenic induced toxicity. An estimated 70 million population are exposed to arsenic poisoning in India in the 2020. The present study is aimed to develop the antidote for arsenic-induced toxicity in Charles Foster rats. A total of n=18 rats (12 weeks old) of an average weight of 160 ± 20 g were used for the study. The study group included three groups, n=6 control (Group I: Untreated) and n= 12 (group II) treated with sodium arsenite orally at the dose of 8mg/Kg b.w daily for 6 months. The n= 6 animals were dissected and the rest n=6 (Group III) was administered orally with Panax quinquefolius (Ginseng) root ethanolic extract at 300mg/Kg body weight per day for 8 weeks. All the animals were sacrificed after the completion of their respective doses and their blood samples were taken for haematological and biochemical evaluation, while the vital tissues such as liver and kidneys for the histopathological study. The study revealed significant fluctuation (p<0.0001/p<0.001/p<0.05) in the haematological parameters viz. leukocyte count, haemoglobin, red blood cell count, haematocrit percentage, MCV, MCH and MCHC and biochemical parameters such as SGPT, SGOT, ALP, bilirubin, urea, uric acid, creatinine and lipid peroxidation in arsenic-treated groups. There was a significant reduction (p<0.0001/p<0.001/p<0.05) in the levels of haematological and biochemical parameters after the administration of ginseng extract. Similarly, the histopathological study revealed a high magnitude of degeneration in the hepatocytes and nephrocytes after the treatment of arsenic, but after the administration of ginseng extract, there was significant restoration at the cellular level. Ethnopharmacological relevance Chronic arsenic poisoning due to contaminated subsoil water is a threat to society in West Bengal, India and in Bangladesh. The human being may also be affected by the exposed cattle from the affected area by consuming milk, egg, meat and others. In Ayurveda, several herbs like Haridra (turmeric), Shunthi (dried ginger root) and others are used for the management of arsenic poisoning. Aim of the study The study was conducted to find out the ameliorative effect of turmeric and ginger powder against experimentally induced arsenic toxicity in calves. Materials and methods Twenty four calves were divided into four groups (group I, II, III and IV) having six animals in each group. Animals of group I, II and III were orally administered with sodium arsenite at 1 mg/kg body weight for 90 days and in addition group II and group III animals were treated orally with turmeric and ginger powder respectively at 10 mg/kg body weight from 46th day onwards. Group IV animals were given food and water without drug and served as control. Arsenic content was estimated in faeces, hair, urine and plasma in every 15 days. Bio-chemical, haematological and anti-oxidant parameters were also assessed. Results Turmeric and ginger powder significantly (P<0.05) reduced the plasma and hair arsenic levels through increased excretion via faeces and urine. Haemoglobin level, TEC and TLC were decreased in groups I, II and III, however these were improved significantly (P<0.05) from 75th day onwards in turmeric and ginger treated groups. Increased activity of AST and ALT were significantly decreased (P<0.05) from 75th day onwards in group II and III. Blood urea nitrogen and plasma creatinine were also significantly decreased (P<0.05) in group II and III than group I from 60th day onwards. The SOD and catalase activity were significantly (P<0.05) reduced in groups I, II and III, but these were restored at the end of the experiment in turmeric and ginger treated groups. Conclusion The test drugs are found significantly effective not only to eliminate arsenic from the body but also give protection from possible damage caused by arsenic exposure, it may be concluded from the present study that turmeric and ginger can be helpful in the therapy of chronic arsenic toxicity in calves. The present study investigated the alleviative effect of aqueous extract of Trichosanthes dioica fruit (AQTD) against arsenic induced cardiotoxicity in Wistar albino rats. AQTD (50 and 100 mg/kg) was administered orally to rats for 20 consecutive days before oral administration of sodium arsenite (10 mg/kg) for 8 days. Then the body weights, heart weights, hematological profile, serum biochemical profile; myocardial antioxidative parameters viz. lipid peroxidation, reduced and oxidized glutathione, glutathione- S-transferase, glutathione peroxidase (GPx), glutathione reductase (GR), superoxide dismutase (SOD), catalase (CAT), and DNA fragmentation were evaluated. Pretreatment with AQTD markedly and significantly normalized body weights, heart weights, hematological profile, serum biochemical profile and significantly modulated all the myocardial antioxidative parameters and reduced DNA fragmentation in arsenic intoxicated rats. Therefore, T. dioica fruit possessed remarkable alleviative effects against arsenic induced myocardial toxicity in Wistar albino rats mediated by amelioration of arsenic induced myocardial oxidative stress by several mechanisms. Abstract: The freshwater wetland systems of India is a complex habitat that supports a broad range of diverse species including molluscs, which play an important role in supplementing third world countries. In the arsenic affected flood plains of West Bengal, a huge amount of arsenic laden groundwater is raised for the purpose of irrigation. Agricultural runoffs and flood water movement during monsoon may cause accumulation of arsenic in the adjacent freshwater aquifers, the common habitat of Lamellidens marginalis (Mollusca; Bivalvia; Eulamellibranchiata), a filter feeder, sensitive to altered environmental conditions. To examine the nature of toxicity induced by inorganic arsenic on both the haemocytes and tissues of the invertebrate heart, the animals were exposed to five different sublethal concentrations of sodium arsenite for a maximum time span of 30 days in vitro. Significant differences were recorded in the total haemocyte count, biochemical and histopathological parameters of the heart of L. marginalis under the arsenic induced stress. Our observations indicate the development of profound haematopoietic and cardiac stress under the sublethal inorganic arsenite exposure and it also implies the nature of risk imposed on the freshwater aquatic ecosystem under potential arsenic contamination. Abstract: Toxicity of arsenic was investigated in the gill of Lamellidens marginalis by exposing the animals to sublethal concentrations of sodium arsenite for a maximum period of 30 days in controlled laboratory conditions. Arsenite exposure inhibited the activities of acid phosphatase (ACP), alkaline phosphatase (ALP), glutamate oxaloacetate transaminase (GOT), glutamate pyruvate transaminase (GPT) and acetlycholinesterase (AChE) in a dose and time dependent manner. Depletion in cytotoxic molecule like nitric oxide (NO) and suppression of phenoloxidase (PO) activity suggests an immune compromise in the animal. Inhibition in the activities of glutathione-s-transferase (GST) and catalase (CAT) in the species indicate impairment of its vital detoxification process and elevated oxidative stress respectively. Histopathology of the gill indicates arsenite induced damage of the organ leading to its possible dysfunction. The toxic exposure ravaged the structure and impaired the functions of the gill of the animal which might restrict its proper gaseous exchange, filter feeding and elicitation of immune responses against pathogens. Chronic arsenic poisoning is one of the serious health hazards in West Bengal, India, and Bangladesh. It occurs due to contaminated subsoil water. The aim of this study is conducted to find out the ameliorative effect of turmeric and P. foetida powder on experimentally induced arsenic toxicity in sheep. Twelve sheep were divided into four groups; groups I, II and III were orally administered with sodium arsenite at 6.6 mg/kg body weight for 133 days; groups I and II animals were treated by turmeric and P. foetida powders respectively at 500 mg/kg dose for the last 49 days; the fourth group was control. Arsenic content was estimated in faeces, urine and wool in every 15 days. Biochemical, haematological, antioxidant parameters and DNA fragmentation were also assessed. Turmeric and P. foetida powder treatment significantly (P < 0.05) increased arsenic elimination through faeces, urine and wool. Haemoglobin content and TEC were decreased in groups I, II and III; however, these were improved significantly (P < 0.05) by turmeric and P. foetida powder treatment. Increased activity of AST, ALT, blood urea nitrogen and plasma creatinine were significantly (P < 0.05) decreased in groups I and II.
