Premise of research. Genome size has cascading effects at nuclear, cell, and whole-organism levels, with consequences in the biology of the species. Large genomes impose higher metabolic and energetic costs, which might be exceedingly limiting for species to thrive under stressing environments, such as island archipelagos. Methodology. We mined online databases to collate available data on chromosome numbers and genome sizes in the genus Artemisia , which colonized Macaronesia and Hawaii archipelagos. We applied phylogenetic-based modeling approaches to reconstruct the evolution of these two traits to estimate ancestral states as well as the main mechanisms of change. Pivotal results. On the basis of extant chromosome data, our analyses discard polyploidy as a likely mechanism for speciation in Macaronesia or Hawaii. However, contrasting genome size patterns are found. Hawaiian endemics account for the smallest genomes in Artemisia , while those of Macaronesian endemics fall well above the average size in the genus. Conclusions. The evidence gathered in Artemisia indicates that, rather than genome downsizing, the presence of small genomes most likely predated colonization of Hawaii, which could have been key for a long dispersal from an Asian ancestor. Such a pattern does not hold in Macaronesia, which is relatively closer to mainland compared with Hawaii, hence suggesting that geographical isolation might reduce the likelihood of a large-genomed taxon to successfully colonize remote island archipelagos. Background: The gut microbiome of animals is an important component that has strong influence on the health, fitness, and behavior of its host. Most research in the microbiome field has focused on human populations and commercially important species. However, researchers are now considering the link between endangered species conservation and the microbiome. In Hawaii, several threats (e.g., avian malaria and habitat loss) have caused widespread population declines of Hawaiian honeycreepers (subfamily: Carduelinae). These threats can have a significant effect on the avian gut microbiome and may even lead to disruption of microbial function. However, the gut microbiome of honeycreeper in the wild has yet to be explored. Methods: We collected 13 and 42 fecal samples, respectively, from two critically endangered honeycreeper species, the akikiki (Oreomystis bairdi) and the akekee (Loxops caeruleirostris). The 16S rRNA gene was sequenced and processed though a MOTHUR-based bioinformatics pipeline. Bacterial ASVs were identified using the DADA2 program and bacterial community analyses, including alpha and beta diversity measures, were conducted using R packages Phyloseq and vegan. Results: A total of 8,958 bacterial ASVs were identified from the fecal samples. Intraspecific differences in the gut microbiome among individual birds explained most of the variation present in the dataset, however differences between species did exist. Both species had distinct microbiomes with minimal overlap in beta diversity. 'Akikiki had a more diverse microbiome compared to 'akekee. Additionally, small but stastically significant differences in beta diversity also exist between sampling location and sexes in akikiki. Conclusion: Akikiki and akekee are currently the focus of captive breeding efforts and plans to translocate the two species to other islands are underway. Phylogenetic analyses of nucleotide sequences of the internal transcribed spacers and 5.8 regions of the nuclear ribosomal DNA and of the trnH-psbA spacer of the chloroplast genome confirm that the three taxa of the Jacquemontia ovalifolia (Choicy) Hallier f. complex (Convolvulaceae) form a monophyletic group. Levels of nucleotide divergence and morphological differentiation among these taxa support the view that each should be recognized as distinct species. These three species display unique intercontinental disjunction, with one species endemic to Hawaii ( Jacquemontia sandwicensis A. Gray.), another restricted to eastern Mexico and the Antilles [ Jacquemontia obcordata (Millspaugh) House], and the third confined to East and West Africa ( J. ovalifolia). The Caribbean and Hawaiian species are sister taxa and are another example of a biogeographical link between the Caribbean Basin and Polynesia. We provide a brief conservation review of the three taxa based on our collective field work and investigations; it is apparent that J. obcordata is highly threatened and declining in the Caribbean. The Hawaiian archipelago is arguably the world’s finest natural laboratory for the study of evolution and patterns of speciation. Arthropods comprise over 75% of the endemic biota of the Hawaiian Islands and a large proportion belongs to species radiations. We classify patterns of speciation within Hawaiian arthropod lineages into three categories: (i) single representatives of a lineage throughout the islands; (ii) species radiations with either (a) single endemic species on different volcanoes or islands, or (b) multiple species on each volcano or island; and (iii) single widespread species within a radiation of species that exhibits local endemism. A common pattern of phylogeography is that of repeated colonization of new island groups, such that lineages progress down the island chain, with the most ancestral groups (populations or species) on the oldest islands. While great dispersal ability and its subsequent loss are features of many of these taxa, there are a number of mechanisms that underlie diversification. These mechanisms may be genetic, including repeated founder events, hybridization, and sexual selection, or ecological, including shifts in habitat and/or host affiliation. Insular adaptive radiations in which repeated bouts of diversification lead to phenotypically similar sets of taxa serve to highlight predictability in the evolutionary process [1]. However, examples of such replicated events are rare. Cross-clade comparisons of adaptive radiations are much needed to determine whether similar ecological opportunities can lead to the same outcomes. Here, we report a heretofore uncovered adaptive radiation of Hawaiian stick spiders (Theridiidae, Ariamnes) in which different species exhibit a set of discrete ecomorphs associated with different microhabitats. The three primary ecomorphs (gold, dark, and matte white) generally co-occur in native forest habitats. Phylogenetic reconstruction mapped onto the well-known chronosequence of the Hawaiian Islands shows both that this lineage colonized the islands only once and relatively recently (2-3 mya, when Kauai and Oahu were the only high islands in the archipelago) and that the distinct ecomorphs evolved independently multiple times following colonization of new islands. This parallel evolution of ecomorphs matches that of "spiny-leg" long-jawed spiders (Tetragnathidae, Tetragnatha), also in Hawaii [2]. Both lineages are free living, and both have related lineages in the Hawaiian Islands that show quite different patterns of diversification with no evidence of deterministic evolution. We argue that repeated evolution of ecomorphs results from a rugged adaptive landscape, with the few peaks associated with camouflage for these free-living taxa against the markedly low diversity of predators on isolated islands. These features, coupled with a limited genetic toolbox and reduced dispersal between islands, appear to be common to situations of repeated evolution of ecomorphs., (Copyright © 2018 Elsevier Ltd.
