Livers of caribou and reindeer (Rangifer tarandus) from Canada (n = 146), Greenland (n = 30), Svalbard (n = 7), and Sweden (n = 60) were analyzed for concentrations of eight perfluoroalkyl carboxylic acids and four perfluoroalkane sulfonic acids. In Canadian caribou, PFNA (range < 0.01–7.4 ng/g wet weight, ww) and PFUnDA (<0.01–5.6 ng/g ww) dominated, whereas PFOS predominated in samples from South Greenland, Svalbard, and Sweden, although the highest concentrations were found in caribou from Southwest Greenland (up to 28 ng/g ww). We found the highest median concentrations of all PFAS except PFHxS in Akia-Maniitsoq caribou (Southwest Greenland, PFOS 7.2–19 ng/g ww, median 15 ng/g ww). The highest concentrations of ΣPFAS were also found in Akia-Maniitoq caribou (101 ng/g ww) followed by the nearby Kangerlussuaq caribou (45 ng/g ww), where the largest airport in Greenland is situated, along with a former military base. Decreasing trends in concentrations were seen for PFOS in the one Canadian and three Swedish populations. Furthermore, PFNA, PFDA, PFUnDA, PFDoDA, and PFTrDA showed decreasing trends in Canada's Porcupine caribou between 2005 and 2016. In Sweden, PFHxS increased in the reindeer from Norrbotten between 2003 and 2011. The reindeer from Västerbotten had higher concentrations of PFNA and lower concentrations of PFHxS in 2010 compared to 2002. Finally, we observed higher concentrations in 2010 compared to 2002 (albeit statistically insignificant) for PFHxS in Jämtland, while PFNA, PFDA, PFUnDA, PFDoDA, and PFTrDA showed no difference at all. There are few long-term datasets of volatile organic compounds (VOCs) in the High Arctic. Furthermore, knowledge about their source regions remains lacking. To address this matter, we report a multiseason dataset of highly time-resolved VOC measurements in the High Arctic from April to October 2018. We have utilized a combination of measurement and modeling techniques to characterize the mixing ratios, temporal patterns, and sources of VOCs at the Villum Research Station at Station Nord in northeastern Greenland. Atmospheric VOCs were measured using proton-transfer-reaction time-of-flight mass spectrometry. Ten ions were selected for source apportionment with the positive matrix factorization (PMF) receptor model. A four-factor solution to the PMF model was deemed optimal. The factors identified were biomass burning, marine cryosphere, background, and Arctic haze. The biomass burning factor described the variation of acetonitrile and benzene and peaked during August and September. The marine cryosphere factor was comprised of carboxylic acids (formic, acetic, and C3H6O2) as well as dimethyl sulfide (DMS). This factor displayed peak contributions during periods of snow and sea ice melt. A potential source contribution function (PSCF) showed that the source regions for this factor were the coasts around southeastern and northeastern Greenland. The background factor was temporally ubiquitous, with a slight decrease in the summer. This factor was not driven by any individual chemical species. The Arctic haze factor was dominated by benzene with contributions from oxygenated VOCs. This factor exhibited a maximum in the spring and minima during the summer and autumn. This temporal pattern and species profile are indicative of anthropogenic sources in the midlatitudes. This study provides seasonal characteristics and sources of VOCs and can help elucidate the processes affecting the atmospheric chemistry and biogeochemical feedback mechanisms in the High Arctic. Number concentrations of ice-nucleating particles (NINP) in the Arctic were derived from ground-based filter samples. Examined samples had been collected in Alert (Nunavut, northern Canadian archipelago on Ellesmere Island), Utqiaġvik, formerly known as Barrow (Alaska), Ny-Ålesund (Svalbard), and at the Villum Research Station (VRS; northern Greenland). For the former two stations, examined filters span a full yearly cycle. For VRS, 10 weekly samples, mostly from different months of one year, were included. Samples from Ny-Ålesund were collected during the months from March until September of one year. At all four stations, highest concentrations were found in the summer months from roughly June to September. For those stations with sufficient data coverage, an annual cycle can be seen. The spectra of NINP observed at the highest temperatures, i.e., those obtained for summer months, showed the presence of INPs that nucleate ice up to -5 ∘ C. Although the nature of these highly ice-active INPs could not be determined in this study, it often has been described in the literature that ice activity observed at such high temperatures originates from the presence of ice-active material of biogenic origin. Spectra observed at the lowest temperatures, i.e., those derived for winter months, were on the lower end of the respective values from the literature on Arctic INPs or INPs from midlatitude continental sites, to which a comparison is presented herein. An analysis concerning the origin of INPs that were ice active at high temperatures was carried out using back trajectories and satellite information. Background: Perfluoroalkyl acids (PFAAs) are persistent and bioaccumulating compounds, which are spread all over the globe. We aimed to compare the PFAA concentrations in serum from pregnant women in five birth cohorts from four countries (Denmark, China, Norway, and Greenland).Methods: Serum samples were obtained from the following five birth cohorts including a total of 4718 pregnant women: the Danish National Birth Cohort (DNBC, years 1996-2002, Denmark), the Aarhus Birth Cohort (ABC, years 2008-2013, Denmark), the Shanghai Birth Cohort (SBC, years 2013-2015, China), the Northern Norway Mother-Child Contaminant Cohort (MISA, years 2007-2009, Norway), and the Greenlandic Birth Cohort (ACCEPT, years 2010-2013, Greenland). The samples were analyzed using liquid chromatography triple-quadrupole mass spectrometry. To ensure comparability, all samples except for the MISA samples were measured in the same laboratory. We adjusted the log-transformed PFAA concentrations for age and parity using analysis of covariance.Results and Discussion: The geometric mean (GM) of the summed concentrations of the seven most abundant PFAAs (∑PFAA) was 35ng/mL in the DNBC, 25 ng/mL in the SBC, 18ng/mL in the ACCEPT, 12ng/mL in the MISA cohort, and 12ng/mL in the ABC. The DNBC concentration was highest presumably because these samples were taken in earlier years (i.e. 1996-2002) than the samples from the other cohorts (i.e. 2007-2015), and at a time when the production of PFAAs were at the highest. When excluding the DNBC samples, we found that the concentrations of all the perfluorinated sulfonic acids (PFSAs) and one of the four perfluorinated carboxylic acids (PFCAs) were highest in the Greenlandic women, whereas the other three PFCAs were highest in the Chinese women.Conclusion: The concentration and composition of serum PFAAs were similar for the Danish ABC women and the Norwegian MISA women but were otherwise different across the cohorts. The different exposure profiles might partly be related to differences in lifestyle and diet. As the concentrations and compositional patterns vary between the countries, we suggest that the health implications associated with high PFAA exposure might also differ between the countries. The Arctic Monitoring and Assessment Programme (AMAP) is a working group under the Arctic Council with the aim to monitor and assess temporal trends of contaminants in Arctic ecosystems. The Greenland AMAP Core programme was established to contribute to this effort. The Core programme includes three main components; routine monitoring, retrospective studies and new POP screening studies. The programme is based on an adaptive approach, which has led to changes throughout the years. An overview of the temporal trends during the last two to three decades is presently given together with selected examples of different characteristic trends of POPs. The results show how tissue banked samples and retrospective studies has helped in establishing time-series of compounds of emerging concern. Lastly, the statistical power of the Greenlandic time-series is discussed. The lesson learned is that trend monitoring improves with samples over time, and only pays off after decades of data are generated. [ABSTRACT FROM AUTHOR]

Atmospheric concentrations of organochlorine pesticides (OCPs), polybrominated diphenyl ethers (PBDEs) and neutral per- and polyfluoroalkyl substances (PFAS) have been measured at Villum Research Station, Station Nord (North Greenland) in the period 2008–2013. Atmospheric concentrations of OCPs at the same site have been previously reported for the years 2008–2010. The detection frequency and the average concentrations of OCPs have not significantly changed since the previous study. PBDE congeners (∑ 13 PBDEs) were measured for the first time in North Greenland at concentrations similar to those observed for other remote sites, confirming that these compounds are ubiquitous in the Northern Hemisphere. The ∑ 13 PBDEs concentration ranged from not detected (n.d.) to 6.26 pg m −3 . The BDE congeners found in more than 30% of the samples were BDE-17, BDE-28, BDE-47, BDE-71, BDE-99 and BDE-100. Also for neutral PFAS we present for the first time a multiyear series of measurements for North Greenland. The average sum of the seven measured neutral PFAS (∑ 7 PFAS) ranged from 1.82 to 32.1 pg m −3 . The most abundant compound was 8:2 FTOH (44% of ∑ 7 PFAS), followed by 6:2 FTOH and 10:2 FTOH. Perfluoroalkyl sulfonamides (FOSA) and perfluoroalkyl sulfonamidoethanols (FOSE) were also detected but at much lower concentrations than FTOHs. Temporal trends were investigated for all measured compounds but no significant trend in concentration was observed. Monthly average concentrations for the six years were calculated for each compound and the seasonal variation was investigated. Objectives: Perfluoroalkyl acids (PFAAs) have been detected in human blood, breast milk and umbilical cord blood across the globe. PFAAs do accumulate in the marine food chain in Arctic regions. In Greenland, increasing PFAA concentrations were observed during 1982-2006 in ringed seals and polar bears. However, until now, no data have been reported for PFAAs in Greenlandic Inuit. This study assesses the level and temporal trend of serum PFAAs in Greenlandic Inuit. Study design: Cross-section and temporal time trend survey. Methods: Serum PFAA levels were determined in 284 Inuit from different Greenlandic districts using liquid chromatography-tandem mass spectrometry with electrospray ionization. The temporal time trend of serum PFAAs in Nuuk Inuit during 1998-2005 and the correlation between serum PFAAs and legacy persistent organic pollutants (POPs) were explored. Results: Serum PFAA levels were higher in Nuuk Inuit than in non-Nuuk Inuit. Within the same district, higher PFAA levels were observed for males. An age-dependent, increasing trend of serum PFAA levels in the period from 1998-2005 was observed for Nuuk Inuit. For the pooled gender data, no significant association between PFAAs and legacy POPs was observed for Nuuk Inuit while for non-Nuuk Inuit this correlation was significant. No correlation between PFAAs and legacy POPs was found for male Inuit, whereas significant correlation was observed both for pooled female Inuit and for non-Nuuk Inuit females. Conclusions: We suggest that sources other than seafood intake might contribute to the observed higher PFAA levels in Nuuk Inuit compared to the pooled non-Nuuk Inuit. [ABSTRACT FROM AUTHOR]

Background: Breast cancer (BC) is the most common cancer for women in the western world. From very few cases an extraordinary increase in BC was observed in the Inuit population of Greenland and Canada although still lower than in western populations. Previous data suggest that exposure to persistent organic pollutants (POPs) might contribute to the risk of BC. Rat studies showed that perfluorinated compounds (PFCs) cause significantly increase in mammary fibroadenomas. This study aimed at evaluating the association between serum levels of POPs/PFCs in Greenlandic Inuit BC cases and their controls, and whether the combined POP related effect on nuclear hormone receptors affect BC risk.Methods: Thirty-one BC cases and 115 controls were sampled during 2000-2003 from various Greenlandic districts. The serum levels of POPs, PFCs, some metals and the combined serum POP related effect on estrogen- (ER), androgen- (AR) and Ah-receptor (AhR) transactivity were determined. Independent student t-test was used to compare the differences and the odds ratios were estimated by unconditional logistic regression models.Results: We observed for the very first time a significant association between serum PFC levels and the risk of BC. The BC cases also showed a significantly higher concentration of polychlorinated biphenyls at the highest quartile. Also for the combined serum POP induced agonistic AR transactivity significant association to BC risk was found, and cases elicited a higher frequency of samples with significant POP related hormone-like agonistic ER transactivity. The AhR toxic equivalent was lowest in cases.Conclusions: The level of serum POPs, particularly PFCs, might be risk factors in the development of BC in Inuit. Hormone disruption by the combined serum POP related xenoestrogenic and xenoandrogenic activities may contribute to the risk of developing breast cancer in Inuit. Abstract: Atmospheric concentrations of organochlorine pesticides (OCs), polybrominated diphenyl ethers (PBDEs) and polychloronaphthalenes (PCNs) were measured for the first time in Nuuk, Greenland in 2004 and 2005. The annual mean concentrations of the measured OCs were: α-HCH 20.2pgm−3, γ-HCH (lindane) 5.1pgm−3, endosulfan 4.8pgm−3 and dieldrin 1.9pgm−3. Concentrations of Σ-chlordanes, DDEs and heptachlor epoxide were generally similar and lower than those of α-HCH and γ-HCH. The concentrations of most chlorinated pesticides did not show any clear seasonal variation, with the exception of γ-HCH, which had maximum concentration in August in both years. The average annual mean for ΣPBDEs was 1.14±0.81pgm−3. The predominant congeners measured in Nuuk were BDE-47 and BDE-99 followed by BDE-100, -153 and -28, indicating the use of penta-BDE technical products as the main source. A clear seasonal variation of PBDE concentrations was observed with maximum concentrations occurring in the summer months. The ΣPCNs concentrations ranged between 0.062 and 0.258pgm−3 with an annual mean concentration of 0.161±0.004pgm−3. The PCNs profile was dominated by the tetra-PCNs (74% of the annual mean) and the penta-PCNs (18% of the annual mean). A seasonal trend for ΣPCNs was not observed. Atmospheric concentrations of the investigated compounds were correlated with temperature and anthropogenic CO in order to obtain information about their transport pattern. Positive correlations were found between CO and chlordanes, p,p′-DDE and trifluralin, while a negative correlation was found for γ-HCH. A significant correlation with temperature variations was found for dieldrin, heptachlor epoxide, α-HCH, γ-HCH, BDE-47, BDE-99 and tetra-PCNs, which indicates that re-emission of these compounds from previously contaminated surfaces as an important factor for the observed variations in concentrations. A well-defined subsample of 128 subadult (3-5 years) polar bears (Ursus maritimus) from 19 sampling years within the period 1984-2006 was investigated for perfluoroalkyl contaminants (PFCs). Linear regression analysis of logarithmic-transformed median concentrations showed significant annual increases for PFOS (4.7%), PFNA (6.1%), PFUnA (5.9%), PFDA (4.3%), PFTrA (8.5%), PFOA (2.3%), and PFDoA (5.2%). For four of the PECs, a LOESS smoother model provided significantly better descriptions, revealing steeper linear annual increases for PFOSA of 9.2% after 1990 and between 18.6 and 27.4% for PFOS, PFDA, and PFTrA after 2000. Concentrations of ΣPFCs, by 2006, exceeded the concentrations of all conventional OHCs (organohalogen compounds), of which several have been documented to correlate with a number of negative health effects, If the PFC concentrations in polar bears continue to increase with the steepest observed trends, then the lowest no-adverse-effect level (NOAEL) and lowest-adverse-effect level (LOAEL) detected for rats and monkeys will be exceeded in 2014-2024. In addition, the rapidly increasing concentrations of PFCs are likely to cause cumulative and combined effects on the polar bear, compounding the already detected threats from OHCs. The potential impact from exposure to high perfluorinated acid (PFA) levels (mean = 1280 ng g-1 wet weight; SD = 645; range = 114-3052; n = 57) was investigated on liver lesions as evidenced by the following parameters: Mononuclear cell infiltrations, lipid granulomas, steatosis, Ito cells, and bile duct hyperplasia/portal fibrosis in East Greenland polar bears (Ursus maritimus). Statistical analyses showed that prevalence of liver lesions was not significantly influenced by PFA concentrations. However, these findings are not unexpected as the PFA concentration was within a 4-fold range in all tissue samples. At present, it is not clear whether chronic exposure to PFA is associated with appearance of liver lesions in polar bears, however these hepatic lesions were similar to those produced by PFA under controlled lab conditions. The brominated flame retardants hexabromocyclododecane (HBCD) and tetrabromobisphenol A (TBBPA) are high-production-volume chemicals. In recent years, their presence has been reported in sediment and biota from the marine environment. In this study, an analytical method was developed for the simultaneous determination of HBCD, TBBPA, and the possible metabolite dimethyl-TBBPA. The method was applied in a preliminary screening of egg, liver, and adipose tissue of marine biota from Greenland and the Faroe Islands. α-HBCD was detected in 35 of 36 analysed samples from the Arctic, indicating a ubiquitous presence of α-HBCD in the environment. β- and γ-HBCD were found in 10 and 14 samples, respectively. TBBPA and dimethyl-TBBPA were not detected in any of the samples indicating limited or no transport of these compounds to remote areas. [ABSTRACT FROM AUTHOR]

Perfluorinated compounds (PFCs), such as perfluorooctane sulfonate (PFOS) and related compounds, have been identified as global pollutants and have shown their bioaccumulation into higher trophic levels in the food chain. PFCs have been found in remote areas far from sources, such as the Arctic. In this study spatial and temporal trends in the concentrations of selected PFCs were measured using archived liver samples of ringed seal (Phoca hispida) from East and West Greenland. The samples were collected in four different years at each location, between 1986 and 2003 in East Greenland and between 1982 and 2003 in West Greenland. PFOS was the major contributor to the burden of PFCs in samples, followed by perfluoroundecanoic acid (PFUnA). Perfluorononanoic acid (PFNA) and perfluorodecanoic acid (PFDA) were also detected in most samples. Perfluorohexane sulfonate (PFHxS) and perfluorooctane sulfonamide (PFOSA) were only found sporadically. Perfluorooctanoic acid was not found in detectable concentrations in any sample. Regression analysis of logarithmic transformed PFOS, PFDA, and PFUnA median concentrations indicated a significant temporal trend with increasing concentrations at both locations. A spatial trend in PFOS concentrations (ANOVA, p < 0.0001) was observed between the two sampling locations, with significantly higher concentrations in seals from East Greenland. Concentrations of hexabromocyclododecane (HBCD) were determined in a combination of archived and fresh blubber samples of juvenile ringed seals from East Greenland collected between 1986 and 2008. α-HBCD was the only diastereoisomer consistently above levels of quantification and showed a significant log-linear (exponential) increase from 2.0 to 8.7 ng/g lipid weight (median concentrations) with an annual rate of +6.1%. The concentrations were up to several orders of magnitude lower than those reported for marine mammals from industrialized areas. Previously presented time trends on polybrominated diphenyl ethers (PBDEs) and polychlorinated biphenyls (PCBs) have been extended with new data for 2006 and 2008. ΣPBDE in juvenile seals was the only parameter with a slight upward trend, however, dependent on the low 1986 concentration. Removing this data point resulted in a downward trend, which also was found for adult seals with a time trend starting in 1994. ΣPCB decreased significantly in juvenile seals, again due to the 1986 value, while no trend was found for the adult animals. This indicates stagnating PCB concentrations at a relatively high level, in some cases possibly exceeding tolerable daily intake rates for seal blubber as traditional Arctic food items. Persistent organic pollutants (POP) are environmental contaminants transported over long distances to the Arctic where they biomagnify in marine mammals subsistence hunted by Inuit and may therefore affect human health. Marine mammals in east Greenland are known to have the highest POP concentrations in the circumpolar Arctic area. Due to high intake of marine mammals, east Greenlandic Inuit likewise have the highest POP body burdens across the Arctic. This cross-sectional study aims to investigate the levels of POP and metals in Inuit with a high intake of top predatory species including killer whales and polar bears. Study participants include 37 men and 21 women from Kulusuk, Tasiilaq and Ittoqqortoormiit during year 2013–2015. Lipophilic POP (11 organochlorine-pesticides, 14 polychlorinated-biphenyls (PCB), 10 polybrominated diphenyl ethers), polyunsaturated fatty acids (PFUA) and cotinine were determined in plasma. Fifteen perfluoroalkylated substances (PFAS) were measured in serum and urine and the renal clearance was estimated. Finally the concentration of 10 metals were measured in whole blood. The median age was 38 years, Ittoqqortoormiit Inuit being the oldest. The smoking rate was around 70%, and Kulusuk participants had the lowest PFUA concentrations. Significant municipality differences were observed for lipophilic POP, serum PFAS, mercury, arsenic and selenium with highest concentrations in Ittoqqortoormiit Inuit. Males had higher blood concentrations of PFAS and lead. The estimated PFAS renal clearance and ratio of urine to serum were significantly higher for females, suggesting a sex difference in excretion via the kidney, maybe partly because men had higher serum PFAS concentrations. We observed that Inuit with intake of >200 g polar bear per week had significantly higher levels of PCB, PFAS, arsenic and selenium. In summary, the level of blood POP and heavy metals seems to relate to sex and the frequency intake of meat from marine mammals. Abstract: Organohalogen contaminants bioaccumulate to high concentrations in tissues of polar bears (Ursus maritimus). The exposure levels are in the order to be toxic to inner organs like liver and kidney. We therefore investigated the temporal development of lesions in liver (n=115) and kidney (n=122) samples from East Greenland polar bears taken over the 12year period from 1999 to 2010. Seven liver and seven kidney lesions were observed of which six were age-related. Controlling for this, the analyses showed that hepatic steatosis and renal cell infiltrations, glomerular sclerosis and tubular hyperplasia decreased over the investigated time period (all p<0.05). Similarly, hypertrophy of hepatic Ito cells, renal glomerular capillary wall thickening and interstitial fibrosis increased over the study period (all p<0.05). Regarding contaminant, concentrations of polybrominated diphenyl ethers in adipose tissue were negatively associated with hepatic mononuclear cell infiltrations (p=0.05) and a similar trend was found for Hg (p=0.09). Hexachlorobenzene was positively associated with portal cell infiltrations and hepatic lipid granulomas, while polychlorinated biphenyls were negatively associated with the prevalence of steatosis (both p<0.05) and a similar trend was found for hexachlorocyclohexanes (p=0.08). Mercury was positively correlated with the frequencies of hypertrophic Ito cells (p=0.002) and a similar trend was found for perfluorooctanesulfonic acid (p=0.07). In renal tissue, hexachlorocyclohexanes were positively associated with medullar hyaline casts (p=0.03) and a similar trend was found for cell infiltrations (p=0.08). There was a trend of trans-nonachlor being positively associated with glomerular sclerosis (p=0.06) while dichlorodiphenyltrichloroethanes were negatively associated with tubular hyperplasia (p=0.02). These results suggest that specific liver and renal lesions have decreased or increased over time and that long-range transported organohalogen contaminants and mercury may be among the co-factors responsible for these observations. Abstract: Polar bears (Ursus maritimus) feed mainly on ringed seal (Phoca hispida) and consume large quantities of blubber and consequently have one of the highest tissue concentrations of organohalogen contaminants (OHCs) worldwide. In East Greenland, studies of OHC time trends and organ system health effects, including reproductive, were conducted during 1990–2006. However, it has been difficult to determine the nature of the effects induced by OHC exposures on wild caught polar bears using body burden data and associated changes in reproductive organs and systems. We therefore conducted a risk quotient (RQ) evaluation to more quantitatively evaluate the effect risk on reproduction (embryotoxicity and teratogenicity) based on the critical body residue (CBR) concept and using a physiologically-based pharmacokinetic (PBPK) model. We applied modelling approaches to PCBs, p,p′-DDE, dieldrin, oxychlordane, HCHs, HCB, PBDEs and PFOS in East Greenland polar bears based on known OHC pharmacokinetics and dynamics in laboratory rats (Rattus rattus). The results showed that subcutaneous adipose tissue concentrations of dieldrin (range: 79–1271ngg−1 lw) and PCBs (range: 4128–53923ngg−1 lw) reported in bears in the year 1990 were in the range to elicit possible adverse health effects on reproduction in polar bears in East Greenland (all RQs⩾1). Similar results were found for PCBs (range: 1928–17376ngg−1 lw) and PFOS (range: 104–2840ngg−1 ww) in the year 2000 and for dieldrin (range: 43–640ngg−1 lw), PCBs (range: 3491–13243ngg−1 lw) and PFOS (range: 1332–6160ngg−1 ww) in the year 2006. The concentrations of oxychlordane, DDTs, HCB and HCHs in polar bears resulted in RQs<1 and thus appear less likely to be linked to reproductive effects. Furthermore, sumRQs above 1 suggested risk for OHC additive effects. Thus, previous suggestions of possible adverse health effects in polar bears correlated to OHC exposure are supported by the present study. This study also indicates that PBPK models may be a supportive tool in the evaluation of possible OHC-mediated health effects for Arctic wildlife. [Copyright &y& Elsevier]
