The use of a two-phase decanter (TwPD) for olive-oil extraction produces wastes and byproducts (a small volume of water from oil washing, olive leaves from the defoliator, and a high moisture pomace which can be destoned) that contain valuable bioactive compounds, such as phenolics and/or triterpenic acids. So far, there is no (water) or limited information (leaves and the destoned pomace fraction) on their content of bioactives, especially triterpenic acids. To contribute to the characterization of such streams from cultivars of international interest, in the present study, samples obtained from five mills from the region of Laconia (from one or two harvests) in Greece, where Koroneiki cv dominates, were screened for phenols and/or triterpenic acids. The leaves and pomace were dried at two temperatures (70 °C and/or 140 °C), and the pomace was also destoned before analysis. The liquid wastes contained low amounts of total (TPC) phenols (<140 mg gallic acid/L), hydroxytyrosol (<44 mg/L), and tyrosol (<33 mg/L). The olive leaves varied widely in TPC (12.8–57.4 mg gallic acid/g dry leaf) and oleuropein (0.4–56.8 mg/g dry leaf) but contained an appreciable amount of triterpenic acids, mainly oleanolic acid (~12.5–31 mg/g dry leaf, respectively). A higher drying temperature (140 vs. 70 °C) affected rather positively the TPC/oleuropein content, whereas triterpenic acids were unaffected. The destoned pomace TPC was 15.5–22.0 mg gallic acid/g dw, hydroxytyrosol 3.9–5.6 mg/g dw, and maslinic 5.5–19.3 mg/g dw. Drying at 140 °C preserved better its bioactive phenols, whereas triterpenic acids were not influenced. The present findings indicate that TwPD streams may have a prospect as a source of bioactives for added-value applications. Nowadays, olive leaf polyphenols have been at the center of scientific interest due to their beneficial effects on human health. The most abundant polyphenol in olive leaves is oleuropein. The biological properties of oleuropein are mainly due to the hydroxytyrosol moiety, a drastic catechol group, whose biological activity has been mentioned many times in the literature. Hence, in recent years, many nutritional supplements, food products, and cosmetics enriched in hydroxytyrosol have been developed and marketed, with unexpectedly positive results. However, the concentration levels of hydroxytyrosol in olive leaves are low, as it depends on several agricultural factors. In this study, a rapid and easy methodology for the production of hydroxytyrosol-enriched extracts from olive leaves was described. The proposed method is based on the direct acidic hydrolysis of olive leaves, where the extraction procedure and the hydrolysis of oleuropein are carried out in one step. Furthermore, we tested the in vitro bioactivity of this extract using cell-free and cell-based methods, evaluating its antioxidant and DNA-protective properties. Our results showed that the hydroxytyrosol-enriched extract produced after direct hydrolysis of olive leaves exerted significant in vitro antioxidant and geno-protective activity, and potentially these extracts could have various applications in the pharmaceutical, food, and cosmetic industries.

Legumes are considered to have beneficial health implications, which have been attributed to their phytochemical content. Polyphenols are considered the most important phytochemical compounds extensively studied for their antioxidant properties. The aim of the present study was to examine the effects of potent antioxidant legume plant extracts on xanthine oxidase (XO), catalase (CAT) and superoxide dismutase (SOD) activities. XO exerts a dual role, as it is the major contributor of free radicals during exercise while it generates uric acid, the most potent antioxidant molecule in plasma. CAT and SOD are two of the main enzymes of the antioxidant defence of tissues. We demonstrate that the majority of the extracts inhibited XO activity, but they had no effect on CAT inhibition and SOD induction when used at low concentrations. These results imply that the tested extracts may be considered as possible source of novel XO inhibitors. However, we have shown that allopurinol administration, a known XO inhibitor, before exercise reduces performance and induces oxidative stress in rats. Considering the fact that the extracts examined had an inhibitory effect on XO activity, possibly posing a restriction in their characterization as antioxidants, phytochemical antioxidant administration before exercise should probably be reconsidered.

The study reports the isolation and structural identification of two new flavonol triglycosides from the methanolic extract of Anthyllis hermanniae, exhibiting the same glycosylation pattern: quercetin 3-O-[α-L-rhamnopyranosyl-(1→2)-α-L-arabinopyranoside]-7-O-α-L-rhamnopyranoside (1) and kaempferol 3-O-[α-L-rhamnopyranosyl-(1→2)-α-L-arabinopyranoside]-7-O-α-L-rhamnopyranoside (2). A conformational study related to the central arabinoside moiety was carried out including the analysis of the contribution of NOE effects and acetylation to the elucidation of the 2-O-linked arabinoside configuration of the anomeric carbon. We also report the total synthesis of a model compound, quercetin 3-O-α-L-rhamnopyranosyl-(1→2)-α-L-arabinopyranoside (3), which verifies the structures of the isolated compounds.

A number of medicinal/culinary herbs have been reported to improve glucose metabolism and to yield hypoglycemic effects in patients with diabetes. Since stimulation of insulin sensitivity appears to be a potential mechanism, peroxisome proliferator-activated receptor (PPAR) γ is a likely target molecule for small lipophilic compounds derived from endogenous metabolism and nutrition. Functionally, PPAR γ integrates the control of energy, lipid, and glucose homeostasis. In addition, PPAR δ activity is involved in energy expenditure. Therefore the aim of this study was to investigate whether PPAR γ and PPAR δ as well as the stimulation of glucose uptake is activated by botanical products. CISTUS SALVIFOLIUS (Cistaceae) has been identified as a candidate botanical in a preliminary screening of extracts from medicinal plants of Greek flora. In a bioguided approach, crude extracts, fractions and in the end purified compounds have been evaluated for PPAR γ and PPAR δ specific activities using cell-based transactivation assays. Glucose uptake was measured by nonradioactive 2-[ N-(7-nitrobenz-2-oxa-1,3-diazol-4-yl)amino]-2-deoxyglucose (2-NBDG) uptake. Concerning PPAR γ several extracts induced reporter gene activity, and clear dose-response patterns (0.1-100 µg/mL) could be established in the case of the cyclohexane and dichloromethane extracts. Isolation of individual compounds from the cyclohexane extract revealed that at least 6 out of 7 compounds isolated were active with TRANS-cinnamic acid showing a clear dose-response pattern. In contrast, they were found to be inactive on PPAR δ. The same compounds, however, were also active in stimulating glucose uptake into 3T3-L1 adipocytes. In summary, the bioguided fractionation of CISTUS SALVIFOLIUS yields PPAR γ stimulating metabolites with differing chemical natures. The content of polyphenols in table olives is highly influenced by the olive variety and the debittering process applied on the fruits. Nine commercial types of Greek table olives were examined for their content in oleuropein and hydroxytyrosol. A very simple extraction procedure and a chromatographic methodology were applied for the simultaneous quantitation of oleuropein (OE) and hydroxytyrosol (HT) in drupes, using boiling water extraction followed by direct HPLC analysis. Hydroxytyrosol was found in all the types of olives that were studied. Kalamata olives and Green "tsakistes" of the variety Megaritiki contained the highest quantity of hydroxytyrosol (1.8-2.0 mg/fruit) followed by Greek-style "chondrolies" with quantity 1.0 mg/fruit. Oleuropein was found in small quantities in two cases, but in the case of Throuba Thassos which is processed by dry salt in a traditional Greek way, oleuropein was found in important quantities (1.2 mg/fruit) recorded over a 4-year period. This is the most important finding of this study showing that this particular table olive type is a nutritional rich source of oleuropein. Additionally, assuming a usual consumption of 20 olive fruits per day, an approximate quantity of 25 mg of oleuropein per day can be considered as safe for human use, since it can be found in the usual diet.

A sensitive and simple method was developed for the classification of wines according to variety, geographical origin, and vintage using NMR-based metabonomics. Polyphenol-rich extracts were prepared from 67 varietal wines from the principal wine-producing regions of Greece, using adsorption resin XAD-4. 1D (1)H NMR spectra obtained from the corresponding extracts were segmented, integrated, and normalized, and the data were subjected to principal component analysis. The chemometric classification of wines according to their phenolic profile allows discrimination between wines from different wineries of the same wine-producing zone and between different vintages for wines of the same variety.

An unprecedented natural acetophenone, polygonophenone (1), and a new resorcinol, polygonocinol (2), were isolated from the dichloromethane and methanol extracts of Polygonum maritimum and identified as 2-hydroxy-4-[(2-methoxyethoxy)methoxy] acetophenone and 2-methyl-5-nonadecylresorcinol, respectively. In addition, 11 known compounds were identified, namely, the sesquiterpenoid (+)-8-hydroxycalamene, four ferulic acid esters (tetracosyl, hexacosyl, octacosyl, and triacontyl ferulate), the arylpropane broussonin B, the flavonoids quercetin, quercitrin, and (+)-catechin, the hydroquinone glucoside isotachioside, and beta-sitosterol. The structures of the new compounds were elucidated on the basis of their NMR and MS data. It is noteworthy that polygonophenone (1) is the first naturally methoxyethoxymethyl (MEM)-substituted natural product, and its isolation gives support for the use of MEM protection in biomimetic synthetic schemes.

Phytochemical investigation of a methanol extract of Onobrychis ebenoides yielded five new 3-formyl-2-arylbenzofurans, namely, ebenfurans IV-VIII (1-5), together with the known compounds ebenfurans I, II (6), and III (7). Only 1 and 7 exhibited growth inhibitory activity against MCF-7 and Ishikawa cells, suggesting that the prenyl moiety at position C-5 is the key determinant of the cytotoxic activity of this group of compounds.

One new alkylresorcinol derivative, cichoriol B, and a mixture of three other ones, cichoriols A, C, and D, were isolated from the dichloromethane extract of Cichorium spinosum, a plant that is used traditionally in the Cretan diet. The methanol extract afforded one new sesquiterpene lactone, (4R)-3,4-dihydrolactucopicrin. The structures of the new compounds were determined by spectroscopic methods, mainly by the concerted application of 1D and 2D NMR techniques (HMQC, HMBC, NOESY).

The aerial parts of Cistus monspeliensis yielded 11 cis-clerodane type diterpenes. Five of these, 3, 4, 8,9, and 10, are new compounds. Their structures have been elucidated on the basis of their spectral data (FABMS, 1D and 2D NMR).

The aerial parts of Inula verbascifolia subsp. methanea yielded three new epoxygermacranolides, compounds 1-3, in addition to the previously known 9beta-hydroxyparthenolide. The structures of 1-3 have been elucidated on the basis of their spectral data (MS, CD, 1D and 2D NMR) and by quantum mechanical calculations. The in vitro cytotoxic activity of compounds 1-3 was evaluated against six human solid tumor cell lines.

The traditional uses of the roots of Paeonia taxa are described. The volatile constituents, obtained by steam distillation of the roots of Paeonia clusii Stern subsp. clusii, Paeonia mascula L. subsp. hellenica and Paeonia parnassica, were studied by GC/MS. Salicylaldehyde, paeonol, methyl salicylate and benzoic acid were found to be the major components among the twelve constituents identified. The evaluation of the in vitro antimicrobial activity of the volatile compounds against six bacteria and three fungi is also reported.
