Background: Due to ethical reasons, surgical castration of young male piglets in their first week of life without anesthesia will be banned in Germany from 2021. Breeding against boar taint is already implemented in sire breeds of breeding organizations but in recent years a low demand made this trait economically less important. The objective of this study was to estimate heritabilities and genetic relationships between boar taint compounds androstenone and skatole and maternal/paternal reproduction traits in 4′924 Landrace (LR) and 4′299 Large White (LW) animals from nucleus populations. Additionally, genome wide association analysis (GWAS) was performed per trait and breed to detect SNP marker with possible pleiotropic effects that are associated with boar taint and fertility. Results: Estimated heritabilities (h2) were 0.48 (±0.08) for LR (0.39 ± 0.07 for LW) for androstenone and 0.52 (±0.08) for LR (0.32 ± 0.07 for LW) for skatole. Heritabilities for reproduction did not differ between breeds except age at first insemination (LR: h2 = 0.27 (±0.05), LW: h2 = 0.34 (±0.05)). Estimates of genetic correlation (rg) between boar taint and fertility were different in LR and LW breeds. In LR an unfavorable rg of 0.31 (±0.15) was observed between androstenone and number of piglets born alive, whereas this rg in LW (− 0.15 (±0.16)) had an opposite sign. A similar breed-specific difference is observed between skatole and sperm count. Within LR, the rg of 0.08 (±0.13) indicates no relationship between the traits, whereas the rg of − 0.37 (±0.14) in LW points to an unfavorable relationship. In LR GWAS identified QTL regions on SSC5 (21.1–22.3 Mb) for androstenone and on SSC6 (5.5–7.5 Mb) and SSC14 (141.1–141.6 Mb) for skatole. For LW, one marker was found on SSC17 at 48.1 Mb for androstenone and one QTL on SSC14 between 140.5 Mb and 141.6 Mb for skatole. Conclusion: Knowledge about such genetic correlations could help to balance conventional breeding programs with boar taint in maternal breeds. QTL regions with unfavorable pleiotropic effects on boar taint and fertility could have deleterious consequences in genomic selection programs. Constraining the weighting of these QTL in the genomic selection formulae may be a useful strategy to avoid physiological imbalances. [ABSTRACT FROM AUTHOR]

Simple Summary: To suppress or prevent boar taint, many strategies are available, e.g., surgical or immunocastration or different housing and feeding conditions. In Germany, male piglets may no longer be castrated without anesthesia until the 7th day of life for animal welfare reasons. The aim of this study was to reduce skatole and indole, two of the boar taint-causing substances, by the feeding of charcoal. A fat coating applied on charcoal could delay the unspecific adsorption of charcoal until entry to the large intestine where skatole and indole should be adsorbed since the fat coating is digested during its passage through the small intestine. In total, 18 piglets were divided into three feeding groups and were fed for 19 days with the control feed or with the control feed plus 2% charcoal or plus 4% charcoal coated with a fat layer. The skatole and indole concentrations were analyzed in chyme retrieved from the colon and caecum as these substances need to be adsorbed by charcoal in the intestine to prevent resorption into the bloodstream and accumulation in fat. The reduction in skatole and indole contents underlines charcoal's adsorption capacity. The adsorption capacity was higher when the charcoal was coated. As charcoal reduced skatole and indole, feeding trials with adult boars are needed to observe the status of boar taint substances at the day of slaughter. To reduce the risk of boar taint, intact male piglets are immuno- or surgically castrated. One alternative is reducing skatole by adding skatole reducing or adsorbing substances to the boars' diet. Charcoal with a high capacity for adsorbing skatole and indole in vitro (tested before, data not shown) was fed to the boars to test the hypothesis that a fat coating prevents the unspecific adsorption of charcoal before entry into the large intestine while increasing skatole adsorption. Twelve male and six female weaning piglets with initial body weights of 7.74 ± 0.75 kg were fed for 18 (or 19) days with either 2% pure (untreated) charcoal or 4% coated (50% charcoal + 50% fat-coating) charcoal or no charcoal. After euthanasia, skatole and indole were quantified in caecum and colon chyme. Skatole and indole contents in caecum chyme were significantly lower (p < 0.05) in the group fed with coated charcoal (33 ± 4.2, 7 ± 2.8 µg/gDM, respectively) than in the group fed with pure charcoal (51 ± 7.3, 14 ± 3.0 µg/gDM) or with no charcoal (73 ± 12.6, 15 ± 1.7 µg/gDM). Similar effects were obvious for colon chyme. The results indicate that a fat coating of charcoal might prevent unspecific adsorption in the small intestine and might consequently lead to a higher adsorption capacity for skatole and indole in the large intestine, as skatole and indole concentrations in the chyme of caecum and colon were approximately 50% lower in the piglets who received coated charcoal. Simple Summary: Breeding against boar taint compounds androstenone and skatole can be an efficient alternative for surgical castration of young piglets during their first week of life to avoid boar taint. Physiological links of androstenone to steroid hormones in the synthesis pathway are documented and they have to be analyzed for their genetic effects on reproduction and fertility. While using boar taint and hormone data from Landrace and Large White pigs (commercial nucleus populations and herd book populations), the effects of breeding against androstenone on fertility were evaluated. Moreover, the genetic foundation of the chosen hormones' testosterone, 17β-estradiol, luteinizing hormone, follicle-stimulating hormone, and progesterone was analyzed and then checked for possible pleiotropic effects with boar taint compounds. The results showed consistent unfavorable side effects of breeding against androstenone on testosterone and 17β-estradiol in both breeds. The other hormones showed contrary results regarding unfavorable relationships between boar taint and endocrine fertility parameters. The genetic foundation showed a high potential of breeding against boar taint but the impact on fertility potential should be supervised. The surgical castration of young male piglets without anesthesia is no longer allowed in Germany from 2021. One alternative is breeding against boar taint, but shared synthesis pathways of androstenone (AND) and several endocrine fertility parameters (EFP) indicate a risk of decreasing fertility. The objective of this study was to investigate the genetic background between AND, skatole (SKA), and six EFP in purebred Landrace (LR) and Large White (LW) populations. The animals were clustered according to their genetic relatedness because of their different origins. Estimated heritabilities (h2) of AND and SKA ranged between 0.52 and 0.34 in LR and LW. For EFP, h2 differed between the breeds except for follicle-stimulating hormone (FSH) (h2: 0.28–0.37). Both of the breeds showed unfavorable relationships between AND and testosterone, 17-β estradiol, and FSH. The genetic relationships (rg) between SKA and EFP differed between the breeds. A genome-wide association analysis revealed 48 significant associations and confirmed a region for SKA on Sus Scrofa chromosome (SSC) 14. For EFP, the results differed between the clusters. In conclusion, rg partly confirmed physiologically expected antagonisms between AND and EFP. Particular attention should be spent on fertility traits that are based on EFP when breeding against boar taint to balance the genetic progress in both of the trait complexes. [ABSTRACT FROM AUTHOR]

From 2021 onwards, surgical castration of male piglets without pain relief will be banned in Germany. In Europe, stakeholders have committed themselves to end piglet castration from 2018 onwards. Alternatives to surgical castration are pork production with boars or immunocastrates. The competitiveness of these production systems is required to increase their market acceptance. The aim of this study was to test the profitability of pork production with boars and immunocastrates under different carcass pricing systems and penalty systems linked to boar taint. The calculations were based on the performance parameters of 36 animals (n = 12 immunocastrates, n = 12 boars, n = 12 barrows) from an experimental study. In order to analyze the economic effects of both alternatives under different regional German production systems, the performance data were set in relation to the data of agri benchmark. Both boars and immunocastrates performed economically worse than barrows in all the scenarios tested. If immunocastrates are sold according to the boar pricing system, the profitability of this technique is even lower, but still more profitable than boar fattening. Pork production with boars is the most unprofitable alternative in this study and will be further devalued if a penalty system linked to boar taint will be introduced. [ABSTRACT FROM AUTHOR]
