Background: The role of ferroptosis in tumorigenesis has been confirmed in previous studies. However, the comprehensive analysis of ferroptosis-related gene (FRG) to study the role of FRG in soft tissue sarcoma (STS) is lacking. Methods: RNA sequencing profile of TCGA-SARC cohort and GTEx were used to select differentially expressed FRGs (DEFRGs). Univariate, LASSO, and multivariate Cox analyses were selected to determine overall survival (OS)- and disease-free survival (PFS)-related FRGs. Two prognostic signatures were established and validated in two independent sets from Gene Expression Omnibus (GEO). Finally, the expression of key FRGs were validated with RT-qPCR. Results: In total, 198 FRGs (90.4%) were abnormally expressed in STS. Twelve DEFRGs were incorporated in the final signatures and showed favorable discrimination in both training and validation cohorts. Patients in the different risk groups not only showed different prognosis, but also showed different infiltration of immune cells. Two nomograms combining signature and clinical variables were established and the C-indexes were 0.852 and 0.752 for the OS and DFS nomograms, respectively. Finally, the expression of NOX5, HELLS, and RPL8 were validated with RT-qPCR. Conclusion: This comprehensive analysis of the FRG landscape in STS revealed novel FRGs related to carcinogenesis and prognosis. These findings have implications for prognosis and therapeutic responses, which revealed potential prognostic biomarkers and promote precision medicine. [ABSTRACT FROM AUTHOR]

In this paper, a real-life renewable energy-based system is considered and the reliability function of this system is evaluated with the help of Universal Generating Function technique. The renewable energy-based system is considered from the project in Germany where the wind turbine and hydro power plant are combined. In this project the wind turbines are the world's tallest wind turbines of about 584 feet and one reservoir have the capacity of 1.6 million gallons of water. The wind turbines are set at the top of the hills and the reservoirs are allowed to be filled with river water. This paper presents a renewable energy-based system where the wind turbine and hydro power plant are combined together to produce electricity in bulk. Universal generating function technique is used to obtain the reliability function of the proposed system. This paper intends to explore the impact of Information technology (IT) development on the legal concept of `signatures'. To what extent and in which way does it impact on the legal concept of `signatures'? This paper attempts to examine this issue from an international and comparative perspective. It was found that IT development has different levels of impact on the legal concept of `signatures' in different jurisdictions. In the Common Law system such as the UK and the US, it does not change the legal concept of `signatures'. However, it does put the legal concept on such an important position. Objective: Lipidomic changes were causally linked to metabolic diseases, but the scenario for colorectal cancer (CRC) is less clear. We investigated the CRC lipidome for putative tumor-specific alterations through analysis of 3 independent retrospective patient cohorts from 2 clinical centers, to derive a clinically useful signature., Design: Quantitative comprehensive lipidomic analysis was performed using direct infusion electrospray ionization coupled with tandem mass spectrometry (ESI-MS/MS) and high-resolution mass spectrometry (HR-MS) on matched nondiseased mucosa and tumor tissue in a discovery cohort (n = 106). Results were validated in 2 independent cohorts (n = 28, and n = 20), associated with genomic and clinical data, and lipidomic data from a genetic mouse tumor model (Apc 1638N )., Results: Significant differences were found between tumor and normal tissue for glycero-, glycerophospho-, and sphingolipids in the discovery cohort. Comparison to the validation collectives unveiled that glycerophospholipids showed high interpatient variation and were strongly affected by preanalytical conditions, whereas glycero- and sphingolipids appeared more robust. Signatures of sphingomyelin and triacylglycerol (TG) species significantly differentiated cancerous from nondiseased tissue in both validation studies. Moreover, lipogenic enzymes were significantly up-regulated in CRC, and FASN gene expression was prognostically detrimental. The TG profile was significantly associated with postoperative disease-free survival and lymphovascular invasion, and was essentially conserved in murine digestive cancer, but not associated with microsatellite status, KRAS or BRAF mutations, or T-cell infiltration., Conclusion: Analysis of the CRC lipidome revealed a robust TG-species signature with prognostic potential.
