Ground-level ozone (O3) has gained awareness as an agent of climate change. In this respect, key results are comprehended from a unique 8-year free-air O3-fumigation experiment, conducted on adult beech (Fagus sylvatica) at Kranzberg Forest (Germany). A novel canopy O3 exposure methodology was employed that allowed whole-tree assessment in situ under twice-ambient O3 levels. Elevated O3 significantly weakened the C sink strength of the tree–soil system as evidenced by lowered photosynthesis and 44% reduction in whole-stem growth, but increased soil respiration. Associated effects in leaves and roots at the gene, cell and organ level varied from year to year, with drought being a crucial determinant of O3 responsiveness. Regarding adult individuals of a late-successional tree species, empirical proof is provided first time in relation to recent modelling predictions that enhanced ground-level O3 can substantially mitigate the C sequestration of forests in view of climate change. Frequency and intensity of wildfire occurrences are dramatically increasing worldwide due to global climate change, having a devastating effect on the entire ecosystem including plants. Moreover, distribution of fire-smoke can influence the natural environment over very long distances, i.e. hundreds of kilometres. Dry plant matter contains 0.1-0.9% (w/w) sulphur, which is mainly released during combustion into the atmosphere as sulphur dioxide (SO 2 ) resulting in local concentrations of up to 3000 nL L -1 . SO 2 is a highly hazardous gas, which enters plants mostly via the stomata. Toxic sulphite is formed inside the leaves due to conversion of SO 2 . Plants as sessile organisms cannot escape from threats, why they evolved an impressive diversity of molecular defence mechanisms. In the present study, two recent wildfires in Germany were evaluated to analyse the effect of SO 2 released into the atmosphere on deciduous trees: the Meppen peat fire in 2018 and the forest fire close to Luebtheen in 2019. Collected leaf material from beech (Fagus sylvatica) and oak (Quercus robur) was examined with respect to detoxification of sulphur surplus due to the exposure to elevated SO 2 . An induced stress reaction in both species was indicated by a 1.5-fold increase in oxidized glutathione. In beech leaves, the enzymatic activities of the sulphite detoxification enzymes sulphite oxidase and apoplastic peroxidases were increased 5-fold and a trend of sulphate accumulation was observed. In contrast, oaks did not regulate these enzymes during smoke exposure, however, the constitutive activity is 10-fold and 3-fold higher than in beech. These results show for the first time sulphite detoxification strategies of trees in situ after natural smoke exposure. Beech and oak trees survived short-term SO 2 fumigation due to exclusion of toxic gases and different oxidative detoxification strategies. Beech seedlings originating from 11 German provenances with different climatic conditions were grown in pots and cultivated in a greenhouse. The composition of macro- and microelements in roots, axes and leaves was measured after half of the seedlings were subjected to a simulated summer drought. The recently described sensitivity of these provenances to drought was compared with drought-mediated changes in the elemental and ionic composition in organs of the seedlings; in addition, partitioning between roots and shoots was evaluated. A number of element concentrations were decreased in roots due to drought (K 94% of control, Mg 94%, Mn 75% and Zn 85%). However, chloride concentration increased in all organs (115-125%) and was the only element affected in leaves. Some changes in ionome can be related to sensitivity of provenances, but it is difficult to decide whether these changes are a result of, or a reason for, drought tolerance or sensitivity. Observed increases in chloride concentration in all plant parts of drought-treated beech seedlings can be explained by its function in charge balance, in particular since the level of phosphate was reduced. As a result of chloride accumulation, the sum of added charges of anions (and cations) in water extracts of leaf and root material was similar between drought and control plants. Since only the partitioning of Ca and Al (both only in axis) as well as Mn was affected and other elements (together with previously observed effects on C, N, S and P) remained unaffected by drought in all provenances, it can be concluded that direct effects by means of mass flow inhibition in xylem and phloem are unlikely. Secondary effects, for example on the pH of transport sap and the apoplastic space, cannot be excluded from the present study. These effects may affect partitioning between the apoplast and symplast and therefore may be significant for drought sensitivity.

We investigated the water balances of two beech stands (Fagus sylvatica L.) on opposite slopes (NE, SW) of a narrow valley near Tuttlingen in the southern Swabian Jura, a low mountain range in Southwest Germany. Our analysis combines results from continuous measurements of forest meteorological variables significant to the forest water balance, stand transpiration (ST) estimates from sap flow measurements, and model simulations of microclimate and water fluxes. Two different forest hydrological models (DNDC and BROOK90) were tested for their suitability to represent the particular sites. The investigation covers the years 2001-2007. Central aims were (1) to evaluate meteorological simulations of variables below the forest canopy, (2) to evaluate ST, (3) to quantify annual water fluxes for both beech stands using the evaluated hydrological models, and (4) to analyse the model simulations with regard to assumptions inherent in the respective model. Overall, both models were very well able to reproduce the observed dynamics of the soil water content in the uppermost 30 cm. However, the degree of fit depended on the year and season. The comparison of experimentally determined ST within the beech stand on the NE-slope during the growing season of 2007 with simulated transpiration did not yield a reliable statistical relationship. The simulation of water fluxes for the beech stand on the NE- and SW-slopes showed similar results for vegetation-related fluxes with both models, but different with respect to runoff and percolation flows. Overall, the higher evaporation demand on the warmer SW-slope did not lead to a significantly increased drought stress for the vegetation but was reflected mainly in decreased water loss from the system. This finding is discussed with regard to potential climate change and its impact on beech growth.

The effect of free-air ozone fumigation and crown position on antioxidants were determined in old-growth spruce (Picea abies) trees in the seasonal course of two consecutive years (2003 and 2004). Levels of total ascorbate and its redox state in the apoplastic washing fluid (AWF) were increased under double ambient ozone concentrations (2xO3), whilst ascorbate concentrations in needle extracts were unchanged. Concentrations of apoplastic and symplastic ascorbate were significantly higher in 2003 compared to 2004 indicating a combined effect of the drought conditions in 2003 with enhanced ozone exposure. Elevated ozone had only weak effects on total glutathione levels in needle extracts, phloem exudates and xylem saps. Total and oxidised glutathione concentrations were higher in 2004 compared to 2003 and seemed to be more affected by enhanced ozone influx in the more humid year 2004 compared to the combined effect of elevated ozone and drought in 2003 as observed for ascorbate.

In the present field study the role of ascorbate in scavenging the harmful atmospheric trace gases O3 and NO2 was examined. For this purpose ascorbate contents were determined in needles of adult Scots pine trees (Pinus sylvestris L.) during three consecutive years. Ascorbate contents were correlated with ambient tropospheric O3 and NO2 concentrations and with meteorological parameters. The results showed a strong correlation of atmospheric O3 but not of atmospheric NO2 concentrations with the apoplastic content of ascorbate during the seasonal course. Ascorbate contents in needle extracts did not correlate with ambient trace gas concentrations. In the apoplastic space, but not in needle extracts ascorbate contents correlate highly significantly with global radiation. From these results it is assumed that apoplastic ascorbate in Scots pine needles is adapted to the actual atmospheric O3 concentration to mediate immediate detoxification of O3, while the atmospheric O3 concentration itself is largely dependent on light intensity.

The flux of inorganic and organic nitrogen into the mistletoe Viscum album L. from the xylem sap of a deciduous (Populus x euamericana) and a coniferous host (Abies alba Mill.) was analyzed. For this purpose, a perfusion system was developed in which the xylem sap of the host was replaced by an artificial perfusion solution. With this system flux rates into the mistletoe were determined in feeding experiments either with the organic nitrogen source [1,2-13C2]glutamine at high and the inorganic nitrogen source 15NO3- at low concentration or vice versa. Glutamine influx was already saturated at the low concentration in the xylem sap and was--different from nitrate--not enhanced, when a 250-fold higher concentration was applied. Nitrate influx matched glutamine influx only at high inorganic/organic nitrogen ratios in the perfusion solution. This result indicates a preferential influx of glutamine over nitrate from the host xylem into the mistletoe at the concentrations found in the xylem sap of trees. Surprisingly, a high percentage of both N sources were accumulated in the mistletoe stem, indicating excessive N nutrition of the mistletoe leaves. Since 13C isotope signature was significantly reduced in the outflowing perfusion solution, either an upload of organic compounds from the phloem into the xylem, or an efflux of organic compounds from haustorium of mistletoe into the xylem has to be assumed. 15N isotope signatures enriched in the outflowing perfusion solution support the idea of a nitrate uptake system at the host xylem-haustorium interface, which favors the light N isotope of nitrate.
