Besides agricultural soils, temperate forest soils have been identified as significant sources of or sinks for important atmospheric trace gases (N2O, NO, CH4, and CO2). Although the number of studies for this ecosystem type increased more than tenfold during the last decade, studies covering an entire year and spanning more than 1-2 years remained scarce. This study reports the results of continuous measurements of soil-atmosphere C- and N-gas exchange with high temporal resolution carried out since 1994 at the Höglwald Forest spruce site, an experimental field station in Southern Germany. Annual soil N2O, NO and CO2 emissions and CH4 uptake (1994-2010) varied in a range of 0.2- 3.0 kgN2O-N ha-1 yr-1, 6.4-11.4 kg NO-N ha-1 yr-1, 7.0- 9.2 t CO2-C ha-1 yr-1, and 0.9-3.5 kgCH4-C ha-1 yr-1, respectively. The observed high fluxes of N-trace gases are most likely a consequence of high rates of atmospheric nitrogen deposition (>20 kgNha-1 yr-1) of NH3 and NOx to our site. For N2O, cumulative annual emissions were ≥0.8 kgN2O-N ha-1 yr-1 in years with freeze-thaw events (5 out 14 of years). This shows that long-term, multi-year measurements are needed to obtain reliable estimates of N2O fluxes for a given ecosystem. Cumulative values of soil respiratory CO2 fluxes tended to be highest in years with prolonged freezing periods, i.e. years with below average annual mean soil temperatures and high N2O emissions (e.g. the years 1996 and 2006). Furthermore, based on our unique database on trace gas fluxes we analyzed if soil temperature, soil moisture measurements can be used to approximate trace gas fluxes at daily, weekly, monthly, or annual scale. Our analysis shows that simple-to-measure environmental drivers such as soil temperature or soil moisture are suitable to approximate fluxes of NO and CO2 at weekly and monthly resolution reasonably well (accounting for up to 59% of the variance). However, for CH4 we so far failed to find meaningful correlations, and also for N2O the predictive power is rather low. This is most likely due to the complexity of involved processes and counteracting effects of soil moisture and temperature, specifically with regard to N2O production and consumption by denitrification and microbial community dynamics. At monthly scale, including information on gross primary production (CO2, NO), and N deposition (N2O), increased significantly the explanatory power of the obtained empirical regressions (CO2: r2 = 0.8; NO: r² = 0.67; N2O, all data: r² = 0.5; N2O, with exclusion of freeze-thaw periods: r² = 0.65). We investigated the magnitudes of temporal and spatial variabilities of gross ammonification and nitrification, in an N-saturated temperate forest ecosystem. Forest soil gross ammonification, gross nitrification and heterotrophic soil respiration were measured in the forest floor and uppermost mineral layer over a period of 3 years. Total annual gross fluxes for the organic layer and uppermost mineral horizon (0-4 cm) were in the range of 800-980 kg N ha−1 year−1 for gross ammonification and 480-590 kg N ha−1 year−1 for gross nitrification. Annual heterotrophic soil respiration was 8000-8900 kg C ha−1 year−1. Highest soil C and N turnover rates occurred in summer, and a consistent pattern was observed throughout the observation period, with highest values for plots located at a clear-cut area and lowest values for plots located at an unmanaged, approximately 100-year-old, spruce control site. Soil moisture, soil temperature and substrate availability accounted for most of the observed variability of C and N turnover rates. Because gross rates of inorganic N production were more than an order of magnitude larger than ecosystem N losses along hydrological and gaseous pathways, our study underlines the importance of internal microbial N turnover processes for ecosystem N cycling and retention. The article discusses a study which provides data on the stratification of nitric oxide and nitrous oxide production with soil dept under aerobic and anaerobic incubation conditions for different temperate forest sites in Germany and the Netherlands. It states that the exchange rates of environmental atmospheric trace gases are driven by a few microbiological processes in the soil. The forest sites in the two countries are exposed to high loads of atmospheric nitrogen deposition; The soil at all sites showed a typical check mark shape with most acidic conditions in the uppermost mineral layer.

Purpose: To describe findings on cerebral imaging in patients with COVID-19 and neurological symptoms at two German university hospitals., Materials and Methods: Patients with COVID-19 and neurological symptoms and cerebral imaging (CT or MRI) were included. A chart review regarding neurological symptoms, COVID-19 and imaging findings was conducted., Results: 12 patients (4 females, age 68 ± 12 years) could be included. Three patients had acute findings. Two patients had acute and subacute cerebral ischemia, one patient had additional intracranial hemorrhages and presumed central pontine myelinolysis. One patient had presumed COVID-19-associated pansinusitis., Conclusion: Findings on cerebral imaging in patients with COVID-19 are uncommon and nonspecific. Yield development of agricultural crops over time is not merely the result of genetic and agronomic factors, but also the outcome of a complex interaction between climatic and site-specific soil conditions. However, the influence of past climatic changes on yield trends remains unclear, particularly under consideration of different soil conditions. In this study, we determine the effects of single agrometeorological factors on the evolution of German winter wheat yields between 1958 and 2015 from 298 published nitrogen (N)-fertilization experiments. For this purpose, we separate climatic from genetic and agronomic yield effects using linear mixed effect models and estimate the climatic influence based on a coefficient of determination for these models. We found earlier occurrence of wheat growth stages, and shortened development phases except for the phase of stem elongation. Agrometeorological factors are defined as climate covariates related to the growth of winter wheat. Our results indicate a general and strong effect of agroclimatic changes on yield development, in particular due to increasing mean temperatures and heat stress events during the grain-filling period. Except for heat stress days with more than 31°C, yields at sites with higher yield potential were less prone to adverse weather effects than at sites with lower yield potential. Our data furthermore reveal that a potential yield levelling, as found for many West-European countries, predominantly occurred at sites with relatively low yield potential and about one decade earlier (mid-1980s) compared to averaged yield data for the whole of Germany. Interestingly, effects related to high precipitation events were less relevant than temperature-related effects and became relevant particularly during the vegetative growth phase. Overall, this study emphasizes the sensitivity of yield productivity to past climatic conditions, under consideration of regional differences, and underlines the necessity of finding adaptation strategies for food production under ongoing and expected climate change., (© 2020 The Authors. Importance: Anecdotal evidence suggests that deep brain stimulation (DBS) of the internal globus pallidus (GPi) is effective in ameliorating dystonia in X-linked dystonia parkinsonism (XDP), a disease that is usually refractive to medical therapy., Objective: To determine the efficacy of GPi-DBS in a cohort of patients with XDP in a prospective study and identify predictors of postoperative outcomes., Design, Setting, and Participants: This observational prospective cohort study enrolled patients in February 2013 and was completed in December 2014. The patients were followed up for up to 46 months. Patients from the Philippines were treated in a single center in Lübeck, Germany and followed up in the Philippines. Sixteen men with XDP (mean [SD] age, 40.9 [7.3] years; disease duration, 1-6 years) from the Philippines with predominant dystonia were selected., Exposures: All patients underwent bilateral GPi-DBS in Lübeck, Germany., Main Outcomes and Measures: Clinical assessment included the motor parts of the Burke-Fahn-Marsden scale (BFMDRS-M) and the Unified Parkinson's Disease Rating Scale (UPDRS-III). T1-based basal ganglia volumetry was performed and correlated with postoperative outcomes., Results: The study participants included 16 Filipino men (mean age, 40.9 years). Masked video ratings revealed significant improvements of dystonia severity 1 week (-55%; range, -94% to 59%; P < .01) and 6 months (-59%; range, -100% to 22%; P < .001) after surgery. The UDPRS-III score also improved, albeit to a lesser extent (-19%; range, -54% to 95%; and -27%; range, -70% to 124%; respectively). Unmasked long-term follow-up confirmed the continued efficacy of GPi-DBS up to 46 months after surgery. Important secondary end points improved, including activities of daily living, pain severity, weight, and quality of life. Caudate atrophy was a predictor of a less beneficial outcome (r = 0.817, P = .004)., Conclusions and Relevance: Internal globus pallidus DBS had a positive association in XDP with predominant dystonia (the primary end point) and contributed to an improved quality of life (the secondary end point). The response to DBS occurred within 1 week. Given the inverse correlation of postoperative benefit and caudate atrophy, GPi-DBS should be considered early during the disease course. Close international collaboration, training, and funding from multiple sources enabled the sustainable follow-up of patients with XDP in the Philippines.

Importance: Mutations in the GNAL gene have recently been shown to cause primary torsion dystonia. The GNAL-encoded protein (Gαolf) is important for dopamine D1 receptor function and odorant signal transduction. We sequenced all 12 exons of GNAL in 461 patients from Germany, Serbia, and Japan, including 318 patients with dystonia (190 with cervical dystonia), 51 with hyposmia and Parkinson disease, and 92 with tardive dyskinesia or acute dystonic reactions., Observations: We identified the following two novel heterozygous putative mutations in GNAL: p.Gly213Ser in a German patient and p.Ala353Thr in a Japanese patient. These variants were predicted to be pathogenic in silico, were absent in ethnically matched control individuals, and impaired Gαolf coupling to D1 receptors in a bioluminescence energy transfer (BRET) assay. Two additional variants appeared to be benign because they behaved like wild-type samples in the BRET assay (p.Ala311Thr) or were detected in ethnically matched controls (p.Thr92Ala). Both patients with likely pathogenic mutations had craniocervical dystonia with onset in the fifth decade of life. No pathogenic mutations were detected in the patients with hyposmia and Parkinson disease, tardive dyskinesias, or acute dystonic reactions., Conclusions and Relevance: Mutations in GNAL can cause craniocervical dystonia in different ethnicities. The BRET assay may be a useful tool to support the pathogenicity of identified variants in the GNAL gene.

Background: Restless legs syndrome (RLS) has a high familial aggregation. To date, several loci and genetic risk factors have been identified, but no causative gene mutation has been found., Methods: We evaluated a German family with autosomal dominantly inherited RLS in 7 definitely and 2 possibly affected members by genome-wide linkage analysis and exome sequencing., Results: We identified three novel missense and one splice site variant in the PCDHA3, WWC2, ATRN, and FAT2 genes that segregated with RLS in the family. All four exons of the PCDHA3 gene, the most plausible candidate, were sequenced in 64 unrelated RLS cases and 250 controls. This revealed three additional rare missense variants (frequency <1%) of unknown pathogenicity in 2 patients and 1 control., Conclusions: We present the first next-generation sequencing study on RLS and suggest PCDHA3 as a candidate gene for RLS., (Copyright © 2012 Movement Disorder Society.)

Objective: While several genes have been identified to cause Parkinson's disease (PD), monogenic forms explain only a small proportion of cases. We report clinical and genetic results in a large family with late-onset autosomal dominant PD., Methods: Thirty-eight family members of a five-generation Northern German PD family underwent a detailed neurologic examination, and transcranial sonography was performed in fifteen of them. Comprehensive mutation analysis of known PD-causing genes and a genome-wide linkage analysis were performed., Results: Late-onset definite PD was found in five subjects with a mean age at onset of 63 years. Another six individuals presented either with probable/possible PD or with subtle parkinsonian signs. Six members with a mean age of 79 years had an essential tremor phenotype. Mode of PD inheritance was compatible with autosomal dominant transmission. One of three examined patients with definite PD demonstrated an increased area of substantia nigra hyperechogenicity upon transcranial sonography. Comprehensive linkage and mutational analysis excluded mutations in known PD-causing genes. Genome-wide linkage analysis suggested a putative disease gene in an 11.3-Mb region on chromosome 7p15-21.1 with a multipoint LOD score of 2.0., Conclusions: The findings in this family further demonstrate genetic heterogeneity in familial autosomal dominant late-onset PD., (© 2011 John Wiley & Sons A/S.)

The multidrug resistance protein 1 (MDR1 or ABCB1) gene encodes a P-glycoprotein that protects the brain against neurotoxicants. Certain MDR1 genetic variants are known to compromise the function of this transporter and may thus be associated with Parkinson disease (PD). We therefore conducted a large case-control study investigating the potential relationship between MDR1 variants and PD. We determined the frequency of three MDR1 variants in 599 European PD patients and controls and further stratified the population by ethnicity, age at onset, and exposure to pesticides. We detected no relevant association in either the entire sample, or when separately investigating by ethnic origin or age at onset. However, the distribution of c.3435C/T differed significantly between PD patients exposed to pesticides compared to those non-exposed (odds ratio=4.74; confidence interval=[1.009; 22.306]); p=0.047), suggesting that common MDR1 variants might influence the risk to develop PD in conjunction with exposure to pesticides.
