NOx is an important primary air pollutant of major environmental concern which is predominantly produced by anthropogenic combustion activities. NOx needs to be accounted for in national emission inventories, according to international treaties. Constructing accurate inventories requires substantial time and effort, resulting in reporting delays of 1 to 5 years. In addition to this, difficulties can arise from temporal and country-specific legislative and protocol differences. To address these issues, satellite-based atmospheric composition measurements offer a unique opportunity for the independent and large-scale estimation of emissions in a consistent, transparent, and comprehensible manner. Here we test the multi-source plume method (MSPM) to assess the NOx emissions over Germany in the COVID-19 period from 2019–2021. For the years where reporting is available, the differences between satellite estimates and inventory totals were within 75–100 kt (NO2) NOx (<10 % of inventory values). The large reduction in the NOx emissions (∼15 %) concurrent with the COVID-19 lockdowns was observed in both the inventory and satellite-derived emissions. The recent projections for the inventory emissions of 2021 pointed to a recovery of the 2021 emissions towards pre-COVID-19 levels. In the satellite-derived emissions, however, such an increase was not observed. While emissions from the larger power plants did rebound to pre-COVID-19 levels, other sectors such as road transport did not, and the change in emissions is likely due to a reduction in the number of heavier transport trucks compared to the pre-COVID-19 numbers. This again illustrates the value of having a consistent satellite-based methodology for faster emission estimates to guide and check the conventional emission inventory reporting. The method described in this work also meets the demand for independent verification of the official emission inventories, which will enable inventory compilers to detect potentially problematic reporting issues, bolstering transparency and comparability, which are two key values for emission reporting. Insect diversity has been decreasing significantly during the past decades. A main driver causing this negative trend is agricultural intensification, which causes habitat destruction and a deterioration of habitat quality. We caught flying arthropods with 28 Malaise traps and two light traps across a heterogeneous Central European agro-environment in southern Germany over a period of four years. The arthropods captured with Malaise traps were assigned to barcode index numbers (BINs) based on metabarcoding. Lepidopterans caught with light traps and light towers were identified based on morphological characters. Within the four years study, a total of 11,984 insect operational taxonomic units (OTUs) related to unique BINs on BOLD (> 95% similarity) were recorded across this agro-environment. 7835 of them could be assigned to insect order and the vast majority of them represent different species. Different estimates of the total species richness ranged between 8188 and 11,512 OTUs (a proxy for species). This number corresponds to about 26–36% of the total German insect fauna. Light trapping also yielded a high species diversity and returned 502 Lepidoptera species (compared to 784 species captured with Malaise traps). The overlap between light and Malaise traps was less than 45%. The Malaise trap catches showed that insects are more diverse along forest edges than in the centre of meadows. Our findings underline the high value of heterogeneous agro-environments for biodiversity, and the high value when combining data collected with different sampling techniques. [ABSTRACT FROM AUTHOR]

Simple Summary: Tumors in the pancreas and colon are still too often an unmet clinical problem. Cells from these cancers and normal cells have different gene expression profiles. Such dysregulation can be exploited with novel drugs that modulate the acetylation of proteins. We present KH16, a novel compound that causes protein hyperacetylation and shifts the balance of protein expression towards cell death. Promisingly, KH16 kills tumor cells but not normal cells. Moreover, KH16 is more effective than clinically well-established and currently tested drugs with a similar mode of action. Future research can focus on KH16 and compounds with a similar chemotype for anti-cancer therapy. Epigenetic modifiers of the histone deacetylase (HDAC) family are often dysregulated in cancer cells. Experiments with small molecule HDAC inhibitors (HDACi) have proven that HDACs are a vulnerability of transformed cells. We evaluated a novel hydroxamic acid-based HDACi (KH16; termed yanostat) in human pancreatic ductal adenocarcinoma (PDAC) cells, short- and long-term cultured colorectal cancer (CRC) cells, and retinal pigment epithelial cells. We show that KH16 induces cell cycle arrest and apoptosis, both time and dose dependently in PDAC and CRC cells. This is associated with altered expression of BCL2 family members controlling intrinsic apoptosis. Recent data illustrate that PDAC cells frequently have an altered expression of the pro-apoptotic BH3-only protein NOXA and that HDACi induce an accumulation of NOXA. Using PDAC cells with a deletion of NOXA by CRISPR-Cas9, we found that a lack of NOXA delayed apoptosis induction by KH16. These results suggest that KH16 is a new chemotype of hydroxamic acid HDACi with superior activity against solid tumor-derived cells. Next to this temporal lag difficulties in composed inventories can arise from legislative and protocol differences between countries and over time in reporting of emissions. They give the possibility to check for compliance with emission reduction targets in a timely manner as well as to observe rapid emission reductions such as experienced during the COVID-19 lock-downs. Purpose: This study aims to identify pitfalls in the use of cooperation for the implementation of digital applications in rural cultural tourism marketing and derive practical implications for avoiding them. Design/methodology/approach: A total of 14 semi-structured in-depth interviews were conducted with the participants of the cooperation project around the Web app "The Legend of the Call of the Mountain" in the Zugspitz Region in Germany. Findings: The study has identified four key pitfalls: challenges in the long-term financing, destination-specific characteristics of rural areas, doubts about the use of evaluations and the limited horizon of many cultural institutions. Research limitations/implications: Because of the qualitative approach and the single case study, the results may lack generalisability. Therefore, future research should place the results to a larger scale, for example, with a mixed methods approach. Practical implications: Practical implications can be derived from the study for avoiding pitfalls and thus for a successful use of such cooperation projects. The implications draw attention to the different possibilities of long-term financing, the important role of a central tourism organisation, the special possibilities of evaluations and the important role of general awareness raising for the added values of such cooperation. Originality/value: By examining an exemplary cooperation project, the paper fills an existing research gap, as there has been little knowledge in empirical cultural tourism research about the special conditions for using cooperation to implement digital applications in rural cultural tourism marketing. 'Regent' is the fungal resistant grapevine cultivar with the highest acreage in Germany and an important resistance donor in international breeding programs. It carries the resistance loci Rpv3.1 as well as Ren3 and Ren9 against downy and powdery mildew, respectively. As the parents of 'Chambourcin', the resistant paternal ancestor of 'Regent', did not coincide with the breeder's information, the germplasm repository of JKI Geilweilerhof was screened to find the missing ancestors. SSR marker analysis revealed that 'Joannes Seyve 11369' and 'Plantet' are the true parents of 'Chambourcin' and not 'Seyve Villard 12-417' and 'Chancellor'. Furthermore, the origin of the resistance loci Ren3 and Ren9 could be traced back to the genotypes 'Seibel 4614' and 'Munson'. Since the breeder Hermann Jaeger mentioned 'Munson' as a direct descendant of Vitis aestivalis Michx. var. linsecomii (Buckley) L. H. Background/Aims: Reliable and especially widely accepted preventive measures are crucial to further reduce the incidence of colorectal cancer (CRC). Colon capsule endoscopy (CCE) might increase the screening numbers among patients unable or unwilling to undergo conventional colonoscopy. This registry trial aimed to document and determine the CCE indications, findings, complications, and adverse events in outpatient practices and clinics throughout Germany. Methods: Patients undergoing CCE between 2010 and 2015 were enrolled in this prospective multicenter registry trial at six German centers. Patient demographics, outcomes, and complications were evaluated. Results: A total of 161 patients were included. Of the CCE evaluations, 111 (68.9%) were considered successful. Pathological findings in the colon (n=92, 60.1%) and in the remaining gastrointestinal tract (n=38, 24.8%) were recorded. The main finding was the presence of polyps (n=52, 32.3%). Furthermore, five carcinomas (3.1%) were detected and histologically confirmed later. Adequate bowel cleanliness was more likely to be achieved in the outpatient setting (p<0.0001). Interestingly, 85 patients (55.6%) chose to undergo CCE based on personal motivation. Conclusions: CCE seems to be a reliable and safe endoscopic tool for screening for CRC and detecting other diseases. The number of insect species and insect abundances decreased severely during the past decades over major parts of Central Europe. Previous studies documented declines of species richness, abundances, shifts in species composition, and decreasing biomass of flying insects. In this study, we present a standardized approach to quantitatively and qualitatively assess insect diversity, biomass, and the abundance of taxa, in parallel. We applied two methods: Malaise traps, and automated and active light trapping. Sampling was conducted from April to October 2018 in southern Germany, at four sites representing conventional and organic farming. Bulk samples obtained from Malaise traps were further analyzed using DNA metabarcoding. Larger moths (Macroheterocera) collected with light trapping were further classified according to their degree of endangerment. Our methods provide valuable quantitative and qualitative data. Our results indicate more biomass and higher species richness, as well as twice the number of Red List lepidopterans in organic farmland than in conventional farmland. This combination of sampling methods with subsequent DNA metabarcoding and assignments of individuals according depending on ecological characteristics and the degree of endangerment allows to evaluate the status of landscapes and represents a suitable setup for large‐scale long‐term insect monitoring across Central Europe, and elsewhere. According to my files from many years ago, my first contact with Denis Lynn dates back to February 1974, when he sent me a letter with questions related to extrusomes in ciliates, the topic of my doctoral thesis. When in the early 1980s I began to think about editing a monograph with the title I Ciliates: Cells as Organisms i , the first colleague whom I approached with a request to compose a chapter on ciliate systematics was Denis. This study summarizes results of a DNA barcoding campaign on German Diptera, involving analysis of 45,040 specimens. The resultant DNA barcode library includes records for 2,453 named species comprising a total of 5,200 barcode index numbers (BINs), including 2,700 COI haplotype clusters without species‐level assignment, so called "dark taxa." Overall, 88 out of 117 families (75%) recorded from Germany were covered, representing more than 50% of the 9,544 known species of German Diptera. Until now, most of these families, especially the most diverse, have been taxonomically inaccessible. By contrast, within a few years this study provided an intermediate taxonomic system for half of the German Dipteran fauna, which will provide a useful foundation for subsequent detailed, integrative taxonomic studies. Using DNA extracts derived from bulk collections made by Malaise traps, we further demonstrate that species delineation using BINs and operational taxonomic units (OTUs) constitutes an effective method for biodiversity studies using DNA metabarcoding. As the reference libraries continue to grow, and gaps in the species catalogue are filled, BIN lists assembled by metabarcoding will provide greater taxonomic resolution. The present study has three main goals: (a) to provide a DNA barcode library for 5,200 BINs of Diptera; (b) to demonstrate, based on the example of bulk extractions from a Malaise trap experiment, that DNA barcode clusters, labelled with globally unique identifiers (such as OTUs and/or BINs), provide a pragmatic, accurate solution to the "taxonomic impediment"; and (c) to demonstrate that interim names based on BINs and OTUs obtained through metabarcoding provide an effective method for studies on species‐rich groups that are usually neglected in biodiversity research projects because of their unresolved taxonomy. Objective: To analyse stress in parents whose infants with very low birth weight have just concluded high-level care in a Neonatal Intensive Care Unit (NICU). More specifically, we aimed 1) to identify groups of parents in the NICU who are particularly at risk of experiencing stress, and 2) to explore the effects of clinical staffś communication on parental stress.Methods: Our multi-center-study evaluated views from 1277 parents about care for 923 infants in 66 German NICUs. Answers were linked with separately evaluated medical outcomes of the infants. Separate generalised mixed models estimated the influence of personal, medical and communication-related characteristics on specific parental stress.Results: Parents of a younger age and those of infants with severe prognoses were more likely to experience stress. While empathetic communication as one aspect of staffś communication was shown as appropriate in reducing parental stress, an initial introduction and the quantity of information were only slightly associated with lower levels of stress.Conclusion: Results provide evidence for the need to involve parents empathetically from the beginning of their child's stay in the NICU.Practice Implications: Staff in the NICU should communicate empathetically and help to reduce stress in parents particularly at risk. Residential wood burning is becoming an increasingly important cause of air quality problems since it has become a popular source of alternative energy to fossil fuel. In order to characterize the contribution of residential wood burning to local particle pollution, a field campaign was organized at the village of Seiffen (Saxony, Germany). During this campaign, an Aerosol Mass Spectrometer (AMS) was deployed in parallel to a PM1 high volume filter sampler. The AMS mass spectra were analyzed using Positive Matrix Factorization (PMF) to obtain detailed information about the organic aerosol (OA). Biomass-burning organic aerosol (BBOA), Hydrocarbon-like organic aerosol (HOA), and Oxygenated Organic Aerosol (OOA) were identified and represented 20 %, 17% and 62% of total OA, respectively. Additionally, Polycyclic Aromatic Hydrocarbons (PAH) were measured by the AMS with an average concentration of 10 ngm-3 and short term events of extremely high PAH concentration (up to 500 ngm-3) compared to the mean PAH value were observed during the whole measurement period. A comparison with the results from PM1 filter samples showed that the BBOA factor and the AMS PAH are good indicators of the total concentration of the different monosaccharide anhydrides and PAH measured on the filter samples. Based on its low correlation with CO and the low car traffic, the HOA factor was considered to be related to residential heating using liquid fuel. An influence of the time of the week (week vs. weekend) on the diurnal profiles of the different OA components was observed. The weekdays were characterized by two maxima; a first one early in the morning and a stronger one in the evening. During the weekend days, the different OA components principally reached only one maximum in the afternoon. Finally, the PAH emitted directly from residential wood combustion was estimated to represent 1.5% of the total mass of the BBOA factor and around 62%of the total PAH concentration measured at Seiffen. This result highlights the important contribution of residential wood combustion to air quality and PAH emissions at the sampling place, which might have a significant impact on human health. Moreover, it also emphasizes the need for a better time resolution of the chemical characterization of toxic particulate compounds in order to provide more information on variations of the different sources through the days as well as to better estimate the real human exposure. Introduction: Minimized perfusion circuits (MPCs), although aiming at minimizing the adverse effects of cardiopulmonary bypass, have not yet gained popularity. This can be attributed to concerns regarding their safety, as well as lack of sufficient evidence of their benefit.Methods: Described is a randomized, multicentre study comparing the MPC - ROCsafeRX to standard cardiopulmonary bypass in patients undergoing elective coronary artery bypass grafting and/ or aortic valve replacement.Results: Five hundred patients were included in the study (252 randomized to the ROCsafeRX group and 248 to standard cardiopulmonary bypass). Both groups were well matched for demographic characteristics and type of surgery. No operative mortality and no device-related complications were encountered.Transfusion requirement (333 ± 603 vs. 587 ± 1010 ml; p=0.001), incidence of atrial fibrillation (16.3% vs. 24.2%; p=0.03) and the incidence of major adverse events (9.1% vs. 16.5%; p=0.02) were all in favour of the MPC group.Conclusion: These results confirm both the safety and efficacy of the ROCsafeRX MPC for a large variety of cardiac patients. Background: Weight change was analyzed in a cohort of obese patients whose eating habits were changed individually mainly on the basis of food energy density (ED) to evaluate the feasibility of this concept for a larger controlled trial. Method: Five hundred and thirteen outpatients were treated between January 2003 and December 2006. Dietary counseling was based on a pretreatment food diary. In January 2008, a follow-up (FU) was made. For pre- and post-change eating habits, 5184 dietary protocols of 189 patients were analyzed. Results: During 10.5 months of treatment, patients lost weight from an initial BMI of 38.8 ± 8.5 by −0.195 kg/m per month; 36% had weight loss >5%, 44% lost 0-4.9% and 20% had weight gain. At follow-up, 413 patients (80.5%) were reached of whom 80 were still in treatment while 333 were considered as self-treatment (ST) group. The ST group had further weight loss by −0.053 kg/m per month over 16.8 months (40% weight loss, 46% maintenance and 14% weight gain), and 164 patients with type-2-diabetes had greater weight loss compared to those without diabetes during ST (Δ-BMI-0.166 vs. −0.028 points/month; p < 0.0001). Energy intake (EI) was reduced by lower ED, beverages and number of meals. Average carbohydrate, fat and protein intake was reduced by 28, 42, and 7%, respectively. Conclusion: In a unselected cohort of substantially obese patients, the individual change of eating habits based primarily on food ED in conjunction with beverage intake and meal frequency weight loss continued beyond the supported treatment phase indicating a good patient adherence. Simple Summary: Roughly two-thirds of the insect species described from Germany belong to the orders Diptera (flies) or Hymenoptera (wasps, bees, ants and sawflies). However, both orders contain several species-rich families that have received little taxonomic attention until now. This study takes the first step in assessing these "dark taxa" families and provides species estimates for four challenging groups of Diptera (Cecidomyiidae, Chironomidae, Phoridae and Sciaridae). The estimates given in this paper are based on the sequencing results of over 48,000 fly specimens that have been collected in southern Germany via Malaise traps that were operated for one season each. We evaluated the fraction of species in our samples belonging to well-known fly families in order to estimate the species richness of the challenging "dark taxa" (DT families hereafter). Our results suggest a surprisingly high proportion of undetected biodiversity in a supposedly well-investigated country: at least 1800–2200 species await discovery and description in Germany in these four families. Determining the size of the German insect fauna requires better knowledge of several megadiverse families of Diptera and Hymenoptera that are taxonomically challenging. This study takes the first step in assessing these "dark taxa" families and provides species estimates for four challenging groups of Diptera (Cecidomyiidae, Chironomidae, Phoridae, and Sciaridae). These estimates are based on more than 48,000 DNA barcodes (COI) from Diptera collected by Malaise traps that were deployed in southern Germany. We assessed the fraction of German species belonging to 11 fly families with well-studied taxonomy in these samples. The resultant ratios were then used to estimate the species richness of the four "dark taxa" families (DT families hereafter). In Germany, the financing of cultural activities is heavily dependent on state funding. About 90% of cultural funding comes front the public sector. However, because of the precarious state of public finances, funding for cultural activities has been steadily reduced in the last years. Against this background, German cultural institutions are essentially compelled to develop appropriate measures in order to improve their competitive e situation in the context of scarce public resources, and to seek additional sources of finance in order to stabilise their situation over the longer term. One of the more suitable means of increasing revenue on an ongoing basis, is to improve and extend the marketing and visitor-related activities themselves. But how should the marketing or visitor orientation be formulated so that it contributes towards the organisation's social mission as well as improves the financial position of the cultural institution? With respect to the particular financing situation of German cultural institutions, this paper will try to give an answer to this question. [ABSTRACT FROM AUTHOR]

Because of the generally precarious state of public finance in Germany, at federal, state and community levels, expenditure for cultural purposes is being reduced constantly. Therefore, cultural institutions such as heritage sites are virtually compelled to find additional sources of funding to improve their financial situation and ensure their long-term survival. One of the more suitable means of increasing the revenue situation on an ongoing basis is to improve and expand the cultural tourism offerings. Despite this significance of cultural tourism, an empirical study established that German heritage sites have by no means exploited the real potential of cultural tourism in a comprehensive manner. Thus, the objective of this paper is, on the basis of a well-founded delineation of the concept and demand-side situation, to demonstrate the potential success factors for developing cultural tourism and to do so from a marketing perspective. The article features Professor Christoph Cremer and cites his contributions to the field of biophysics. Cremer studied Physics in Munich, Germany and obtained his first doctor of philosophy (Ph.D.) in biophysics and genetics at the University of Freiburg in 1976. His contributions to biophysics reportedly prompted several research activities all over the world. He has collaborated with colleagues from around the world, making him one of the major figures of far-field microscopy.

Treatment of relapse after allogeneic hematopoietic stem cell transplantation (alloHSCT) remains a great challenge. Aiming to evaluate the combination of venetoclax and hypomethylating agents (HMAClax) for the treatment of relapse of myeloid malignancies after alloHSCT, we retrospectively collected data from 32 patients treated at 11 German centers. Venetoclax was applied with azacitidine (n = 13) or decitabine (n = 19); 11 patients received DLI in addition. HMAClax was the first salvage therapy in 8 patients. The median number of cycles per patient was 2 (1-19). All but 1 patient had grade 3/4 neutropenia. Hospital admission for grade 3/4 infections was necessary in 23 patients (72%); 5 of these were fatal. In 30 evaluable patients, overall response rate (ORR) was 47% (14/30, 3 CR MRD neg , 5 CR, 2 CRi, 1 MLFS, 3 PR). ORR was 86% in first salvage patients versus 35% in later salvage patients (p = 0.03). In 6 patients with molecular relapse (MR), ORR was 67% versus 42% in patients with hematological relapse (HR) (n = 24, p = n.s.). After a median follow-up of 8.4 months, 25 patients (78%) had died and 7 were alive. Estimated median overall survival was 3.7 months. Median survival of patients with HMAClax for first versus later salvage therapy was 5.7 and 3.4 months (p = n.s.) and for patients with MR (not reached) compared to HR (3.4 months, p = 0.024). This retrospective case series shows that venetoclax is utilized in various different combinations, schedules, and doses. Toxicity is substantial and patients who receive venetoclax/HMA combinations for MR or as first salvage therapy derive the greatest benefit.

DNA metabarcoding was utilized for a large-scale, multiyear assessment of biodiversity in Malaise trap collections from the Bavarian Forest National Park (Germany, Bavaria). Principal component analysis of read count-based biodiversities revealed clustering in concordance with whether collection sites were located inside or outside of the National Park. Jaccard distance matrices of the presences of barcode index numbers (BINs) at collection sites in the two survey years (2016 and 2018) were significantly correlated. Overall similar patterns in the presence of total arthropod BINs, as well as BINs belonging to four major arthropod orders across the study area, were observed in both survey years, and are also comparable with results of a previous study based on DNA barcoding of Sanger-sequenced specimens. A custom reference sequence library was assembled from publicly available data to screen for pest or invasive arthropods among the specimens or from the preservative ethanol. A single 98.6% match to the invasive bark beetle Ips duplicatus was detected in an ethanol sample. This species has not previously been detected in the National Park., (© 2020 The Authors. Background: The State of Bavaria is involved in a research program that will lead to the construction of a DNA barcode library for all animal species within its territorial boundaries. The present study provides a comprehensive DNA barcode library for the Geometridae, one of the most diverse of insect families., Methodology/principal Findings: This study reports DNA barcodes for 400 Bavarian geometrid species, 98 per cent of the known fauna, and approximately one per cent of all Bavarian animal species. Although 98.5% of these species possess diagnostic barcode sequences in Bavaria, records from neighbouring countries suggest that species-level resolution may be compromised in up to 3.5% of cases. All taxa which apparently share barcodes are discussed in detail. One case of modest divergence (1.4%) revealed a species overlooked by the current taxonomic system: Eupithecia goossensiata Mabille, 1869 stat.n. is raised from synonymy with Eupithecia absinthiata (Clerck, 1759) to species rank. Deep intraspecific sequence divergences (>2%) were detected in 20 traditionally recognized species., Conclusions/significance: The study emphasizes the effectiveness of DNA barcoding as a tool for monitoring biodiversity. Open access is provided to a data set that includes records for 1,395 geometrid specimens (331 species) from Bavaria, with 69 additional species from neighbouring regions. Taxa with deep intraspecific sequence divergences are undergoing more detailed analysis to ascertain if they represent cases of cryptic diversity.
