Monitoring of tickborne diseases is critical for prevention and management. We analyzed 418 ticks removed from 359 patients during 2014-2021 in Marseille, France, for identification and bacteria detection. Using morphology, molecular methods, or matrix-assisted laser desorption/ionization time-of-flight mass spectrometry, we identified 197 (47%) Ixodes, 136 (33%) Dermacentor, 67 (16%) Rhipicephalus, 8 (2%) Hyalomma, 6 (1%) Amblyomma, 2 (0.5%) Argas, and 2 (0.5%) Haemaphysalis tick species. We also detected bacterial DNA in 241 (58%) ticks. The most frequent bacterial pathogens were Rickettsia raoultii (17%) and R. slovaca (13%) in Dermacentor ticks, Borrelia spp. (9%) in Ixodes ticks, and R. massiliae (16%) in Rhipicephalus ticks. Among patients who were bitten, 107 had symptoms, and tickborne diseases were diagnosed in 26, including scalp eschar and neck lymphadenopathy after tick bite and Lyme borrelioses. Rapid tick and bacteria identification using a combination of methods can substantially contribute to clinical diagnosis, treatment, and surveillance of tickborne diseases. [ABSTRACT FROM AUTHOR]

Introduction Lactobacilli are Gram-positive rods, commensals of the normal human flora. Generally, these lactic acid-producing bacteria are considered contaminants, however over the last years their clinical relevance is reevaluated. Lactobacillus delbrueckii is very rarely isolated and only a few cases of L. delbrueckii urinary tract infections (UTIs) have been reported, mainly in females. Case report We report the case of a L. delbrueckii UTI in an 82-year-old male suffering from benign prostate hyperplasia with repeated episodes of acute urinary retention over the last month before presenting to our hospital. The catheter urine culture grew >105 CFUs/mL of pure L. delbrueckii on Columbia CNA blood agar and on Trypticase soy agar. Identification was achieved by matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF MS), using VITEK MS (bioMérieux, France). The patient was successfully treated with cefixime for ten days. A follow-up urine culture performed 7 days after antibiotic discontinuation was sterile. Conclusions To our knowledge the present is the second case of L. delbrueckii urinary tract infection in a male patient. Rare fungal pathogens are emerging as agents of invasive fungal infections. We analyzed 13 cases of fungal infections caused by Kazachstania (Arxiozyma) spp. in Strasbourg University Hospital, Strasbourg, France. Among the cases, 4 patients had proven fungal disease (3 cases of invasive fungal disease and 1 mucocutaneous infection) and 9 were colonized by Kazachstania (Arxiozyma) spp. Candida albicans was also isolated from 11 of the 13 patients. None of the patients with proven invasive fungal disease met host criteria, but most had underlying diseases. All strains were identified as K. telluris by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry, and 3 were confirmed as K. bovina by internal transcribed spacer sequencing. For all tested strains, the MICs for fluconazole were >2 μg/mL. Emergence of this rare fungal infection might be explained by the increasing number of patients with immunocompromised conditions and gastroesophageal diseases. [ABSTRACT FROM AUTHOR]

We aimed to assess the prevalence, risk factors, and visual outcome of Moraxella keratitis. We retrospectively reviewed the medical charts of patients diagnosed with Moraxella spp. keratitis at the Quinze-Vingts National Ophthalmology Hospital, Paris, France, between January 2016 and December 2018. Definitive microbiological identification was performed on archival strains using matrix-assisted laser desorption ionization time of flight coupled to mass spectrometry. One hundred one culture-proven cases of Moraxella keratitis were identified. The most common isolates were Moraxella lacunata (50%) and Moraxella nonliquefasciens (38%). Systemic predisposing factors, principally diabetes mellitus (13%) were identified in 28% of patients, and 87% of patients had ocular surface conditions, including blepharitis (25%), prior ocular surgery (21%), glaucoma (17%), exposure keratopathy (16%), and trauma (16%). Severely affected inpatients were treated empirically with fortified antibiotics including vancomycin, piperacillin, and gentamicin. The presence of hypopyon and being over the age of 60 years were associated with a poorer final visual acuity (p < 0.05). Adjuvant treatment, mostly amniotic membrane transplantation, was required for 31 eyes. The prognostic factors significantly associated with the need for adjuvant treatment were a larger infiltrate and hypoesthesia. The clinical features including ulcer healing, treatment duration, and infiltrate size were not different between Moraxella species. Keratitis caused by Moraxella spp. are rare in France but may threaten sight. The early identification of patients with a poor ocular surface, particularly those with neurotrophic keratopathy and anesthetic cornea, is crucial to prevent delayed healing of ulcers and the need for adjuvant treatment. The ASTA MicroIDSys system (ASTA, Suwon, Korea) is a newly developed Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) system for identification of microorganisms. We compared the performance of the ASTA MicroIDSys system with that of the VITEK MS system (bioMérieux, Marcy l'Etoile, France) for identifying clinical microorganisms. A total 2055 isolates including 1910 bacteria and 145 yeasts were tested. Among them, the VITEK MS correctly identified 1999 (97.3%) isolates to species level and 26 (1.3%) to the genus level. The ASTA MicroIDSys correctly identified 1988 (96.7%) isolates to species level and 28 (1.4%) to the genus level. The VITEK MS and ASTA MicroIDSys misidentified one isolate and four (0.2%) isolates, respectively, and provided no identification for 29 (1.4%) and 35 (1.7%) isolates, respectively. The performance of the ASTA MicroIDSys was comparable to that of the VITEK MS for identification of clinically relevant bacterial and yeast isolates. [ABSTRACT FROM AUTHOR]

Objective: Resistant Gram-negative bacteria isolated from health-related infections are a worldwide problem. Increasing frequency of infections particularly caused by Enterobacteriaceae producing expanded spectrum beta lactamase, leads to the use of more carbapenem group antibiotics which, in turn, leads to bacterial resistance. In this study, we aimed to evaluate carbapenem resistance in Klebsiella pneumoniae (K. pneumoniae) isolates, the mechanisms causing this resistance and the clonal relationship between these isolates. Methods: Ninety-one K. pneumoniae strains isolated from clinical samples obtained in our laboratory were included to the study. The identification of the bacteria was performed with Matriks assisted laser desorption ionization time of flight mass spectrometry (bioMérieux, Marcy-I’Étoile, France) and antimicrobial susceptibility with VITEK-2 (bioMérieux), and the carbapenem resistance was confirmed by ertapenem E-test (bioMérieux). Reverse transcription polymerase chain reaction method was used for the investigation of genes causing carbapenemase production (blaOXA-48, blaNDM-1, blaKPC, blaIMP, blaVIM-1). The clonal relationship between isolates was investigated by pulsed-field jel elektroforez. Results: In carbapenem resistant isolates, blaOXA-48 positivity was found to be 55%, blaNDM-1positivity 37.4%, blaKPC and blaVIM-1 positivity 1.1%. A total of 10 isolates was identified with different resistance genes. In 73 of the isolates included in the study, the clonal relationship was examined, and 16 different groups were identified. Twenty isolates were not clonally associated with any other isolates. The most common resistance mechanism causing the carbapenem resistance was blaOXA-48 gene that is known to be endemic in Turkey. Conclusion: As a result, the carbapenem resistance that we found as 3.13% in our study is similar to the rates obtained in other studies performed in our country which indicates that this resistance is not at a high level yet in our country. However, the ability of carbapenem resistance genes to spread between strains can be a major problem in the near future. Molecular methods are gold standard in carbapenemase detection, but because of having high cost they can not be used in laboratories routinely. Aim: To discuss the significance and applied value in the rapid identification and drug susceptibility test for blood stream infection (BSI) using matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF MS) combined with flow cytometry (FCM).Methods: The bacteria were separated from the positive blood culture bottle using the separation gel-adsorption method system, and then applying MALDI-TOF MS combined with FCM to identify pathogen and drug susceptibility test quickly.Results: The efficiency of the separation gel-adsorption method for gram-negative bacterium, gram-positive bacteria, and fungi is 71%, 74%, and 88%, respectively. The results of identifying pathogens using MALDI-TOFMS are in agreement with results obtained using VITEK®2 (bioMérieux, Marcy l'Etoile, France); both methods can identify 90% of bacteria to species. For fungi, MALDI-TOF MS can identify 75% fungi to species, which is superior to VITEK2, which identifies 60% fungi to species. The results of drug susceptibility test using FCM are almost identical to VITEK2; additionally, the addition of fluorescein diacetate can identify the heterogenic drug-resistant strains.Conclusions: We can quickly identify pathogen and drug-susceptibility test based on MALDI-TOF MS combined with FCM, which is consistent with traditional methods and can shorten the report time from 36-72 hour to 3 hours. As part of the Chemistry-Aerosol Mediterranean Experiment (ChArMEx), a series of aerosol and gas-phase measurements were deployed aboard the SAFIRE ATR42 research aircraft in summer 2014. The present study focuses on the four flights performed in late June early July over two forested regions in the south of France. We combine in situ observations and model simulations to aid in the understanding of secondary organic aerosol (SOA) formation over these forested areas in the Mediterranean and to highlight the role of different gas-phase precursors. The non-refractory particulate species measured by a compact aerosol time-of-flight mass spectrometer (cToF-AMS) were dominated by organics (60 to 72 %) followed by a combined contribution of 25% by ammonia and sulfate aerosols. The contribution from nitrate and black carbon (BC) particles was less than 5% of the total PM1 mass concentration. Measurements of nonrefractory species from off-line transmission electron microscopy (TEM) showed that particles have different mixing states and that large fractions (35 %) of the measured particles were organic aerosol containing C, O, and S but without inclusions of crystalline sulfate particles. The organic aerosol measured using the cToF-AMS contained only evidence of oxidized organic aerosol (OOA), without a contribution of fresh primary organic aerosol. Positive matrix factorization (PMF) on the combined organic--inorganic matrices separated the oxidized organic aerosol into a more-oxidized organic aerosol (MOOA), and a less-oxidized organic aerosol (LOOA). The MOOA component is associated with inorganic species and had higher contributions of m/z 44 than the LOOA factor. The LOOA factor is not associated with inorganic species and correlates well with biogenic volatile organic species measured with a proton-transfer-reaction mass spectrometer, such as isoprene and its oxidation products (methyl vinyl ketone, MVK; methacroleine, MACR; and isoprene hydroxyhydroperoxides, ISOPOOH). Despite a significantly high mixing ratio of isoprene (0.4 to 1.2 ppbV) and its oxidation products (0.2 and 0.8 ppbV), the contribution of specific signatures for isoprene epoxydiols SOA (IEPOXSOA) within the aerosol organic mass spectrum (m/z 53 and m/z 82) were very weak, suggesting that the presence of isoprene-derived SOA was either too low to be detected by the cToF-AMS, or that SOA was not formed through IEPOX. This was corroborated through simulations performed with the Polyphemus model showing that although 60 to 80% of SOA originated from biogenic precursors, only about 15 to 32% was related to isoprene (non-IEPOX) SOA; the remainder was 10% sesquiterpene SOA and 35 to 40% monoterpene SOA. The model results show that despite the zone of sampling being far from industrial or urban sources, a total contribution of 20 to 34% of the SOA was attributed to purely anthropogenic precursors (aromatics and intermediate or semi-volatile compounds). The measurements obtained during this study allow us to evaluate how biogenic emissions contribute to increasing SOA concentrations over Mediterranean forested areas. Directly comparing these measurements with the Polyphemus model provides insight into the SOA formation pathways that are prevailing in these forested areas as well as processes that need to be implemented in future simulations. Abstract: The large time-of-flight (TOF) array is one of the main detectors devoted to charged hadron identification in the mid-rapidity region of the ALICE experiment at the LHC. It allows separation among pions, kaons and protons up to a few GeV/c, covering the full azimuthal angle and . The TOF exploits the innovative MRPC technology capable of an intrinsic time resolution better than 50 ps with an efficiency close to 100% and a large operational plateau; the full array consists of 1593 MRPCs covering a cylindrical surface of 141m2. The TOF detector has been efficiently taking data since the first pp collisions recorded in ALICE in December 2009. Abstract: At the end of the 2011 run, the CERN CNGS neutrino beam has been briefly operated in lower intensity mode with and with a proton beam structure made of four LHC-like extractions, each with a narrow width of , separated by 524 ns. This very tightly bunched beam allowed a very accurate time-of-flight measurement of neutrinos from CERN to LNGS on an event-by-event basis. The ICARUS T600 detector (CNGS2) has collected 7 beam-associated events, consistent with the CNGS collected neutrino flux of and in agreement with the well-known characteristics of neutrino events in the LAr-TPC. The time of flight difference between the speed of light and the arriving neutrino LAr-TPC events has been analysed. The result is compatible with the simultaneous arrival of all events with speed equal to that of light. This is in a striking difference with the reported result of OPERA (OPERA Collaboration, 2011) claiming that high energy neutrinos from CERN arrive at LNGS earlier than expected from luminal speed. An Aerosol Time-Of-Flight Mass Spectrometer (ATOFMS) was deployed to investigate the size-resolved chemical composition of single particles at an urban background site in Paris, France, as part of the MEGAPOLI winter campaign in January/February 2010. ATOFMS particle counts were scaled to match coincident Twin Differential Mobility Particle Sizer (TDMPS) data in order to generate hourly size-resolved mass concentrations for the single particle classes observed. The total scaled ATOFMS particle mass concentration in the size range 150-1067 nm was found to agree very well with the sum of concurrent High-Resolution Time-of-Flight Aerosol Mass Spectrometer (HR-ToF-AMS) and Multi-Angle Absorption Photometer (MAAP) mass concentration measurements of organic carbon (OC), inorganic ions and black carbon (BC) (R² = 0.91). Clustering analysis of the ATOFMS single particle mass spectra allowed the separation of elemental carbon (EC) particles into four classes: (I) EC attributed to biomass burning (ECbiomass), (ii) EC attributed to traffic (ECtraffic), (iii) EC internally mixed with OC and ammonium sulfate (ECOCSOx), and (iv) EC internally mixed with OC and ammonium nitrate (ECOCNOx). Average hourly mass concentrations for ECcontaining particles detected by the ATOFMS were found to agree reasonably well with semi-continuous quantitative thermal/optical EC and optical BC measurements (r² = 0.61 and 0.65-0.68 respectively, n = 552). The EC particle mass assigned to fossil fuel and biomass burning sources also agreed reasonably well with BC mass fractions assigned to the same sources using seven-wavelength aethalometer data (r² = 0.60 and 0.48, respectively, n = 568). Agreement between the ATOFMS and other instrumentation improved noticeably when a period influenced by significantly aged, internally mixed EC particles was removed from the intercomparison. 88% and 12% of EC particle mass was apportioned to fossil fuel and biomass burning respectively using the ATOFMS data compared with 85% and 15% respectively for BC estimated from the aethalometer model. On average, the mass size distribution for EC particles is bimodal; the smaller mode is attributed to locally emitted, mostly externally mixed EC particles, while the larger mode is dominated by aged, internally mixed ECOCNOx particles associated with continental transport events. Periods of continental influence were identified using the Lagrangian Particle Dispersion Model (LPDM) "FLEXPART". A consistent minimum between the two EC mass size modes was observed at approximately 400 nm for the measurement period. EC particles below this size are attributed to local emissions using chemical mixing state information and contribute 79%of the scaled ATOFMS EC particle mass, while particles above this size are attributed to continental transport events and contribute 21% of the EC particle mass. These results clearly demonstrate the potential benefit of monitoring size-resolved mass concentrations for the separation of local and continental EC emissions. Detailed investigations of the chemical and microphysical properties of atmospheric aerosol particles were performed at the puy-de-Dôme (pdD) research station (1465 m) in autumn (September and October 2008), winter (February and March 2009), and summer (June 2010) using a compact Time-of-Flight Aerosol Mass Spectrometer (cToFAMS). Over the three campaigns, the average mass concentrations of the non-refractory submicron particles ranged from 10 µgm-3 up to 27 µgm-3. Highest nitrate and ammonium mass concentrations were measured during the winter and during periods when marine modified airmasses were arriving at the site, whereas highest concentrations of organic particles were measured during the summer and during periods when continental airmasses arrived at the site. The measurements reported in this paper show that atmospheric particle composition is strongly influenced by both the season and the origin of the airmass. The total organic mass spectra were analysed using positive matrix factorisation to separate individual organic components contributing to the overall organic particle mass concentrations. These organic components include a low volatility oxygenated organic aerosol particle (LV-OOA) and a semi-volatile organic aerosol particle (SV-OOA). Correlations of the LV-OOA components with fragments of m/z 60 and m/z 73 (mass spectral markers of wood burning) during the winter campaign suggest that wintertime LV-OOA are related to aged biomass burning emissions, whereas organic aerosol particles measured during the summer are likely linked to biogenic sources. Equivalent potential temperature calculations, gas-phase, and LIDAR measurements define whether the research site is in the planetary boundary layer (PBL) or in the free troposphere (FT)/residual layer (RL). We observe that SV-OOA and nitrate particles are associated with air masses arriving from the PBL where as particle composition measured from RL/FT airmasses contain high mass fractions of sulphate and LV-OOA. Abstract: The aim of this work was to reliably identify odour-active compounds in dry-cured ham using powerful analysis methods for the volatile fraction. For this purpose, dynamic headspace gas chromatography combined with eight-way olfactometry using a panel of eight sniffers was used. One- and two-dimensional gas chromatography coupled with mass spectrometry and (or) olfactometry were also used. More than 600 compounds from the volatile fraction of dry-cured ham were identified and their biochemical origins are discussed. They covered a wide diversity of structures and chemical functions. Only 29 of them proved odour-active. Comparison of the results of GC–O analysis with those obtained by orthonasal sniffing of the dry-cured ham helped to gain a better understanding of how these substances contributed to the overall aroma of the product. Thus, “Fruity–Floral”, “Green–Vegetable” or “Plastic–Chemical” odours intensively perceived by GC–O have been poorly perceived by orthonasal sniffing. By contrast, “Animal–Meat products” or “Butter–Lactic–Cheesy” odours have been much better perceived by orthonasal sniffing. These results indicate that to understand the interactions between odour-active compounds, experimental doping with carefully selected odour-active compounds will be necessary. Abstract: Head-space solid-phase microextraction (HS-SPME)-based procedure, coupled to comprehensive two-dimensional gas chromatography–time-of-flight mass spectrometry (GC×GC–TOF-MS), was employed for fast characterisation of honey volatiles. In total, 374 samples were collected over two production seasons in Corsica (n =219) and other European countries (n =155) with the emphasis to confirm the authenticity of the honeys labelled as “Corsica” (protected denomination of origin region). For the chemometric analysis, artificial neural networks with multilayer perceptrons (ANN-MLP) were tested. The best prediction (94.5%) and classification (96.5%) abilities of the ANN-MLP model were obtained when the data from two honey harvests were aggregated in order to improve the model performance compared to separate year harvests. New Candida species such as Candida auris have emerged recently as important invasive fungal diseases. We report a case of C. bovina bloodstream infection in a 94-year-old patient in France. Lactones are important flavor compounds in lots of foodstuffs. They also play an important role in brandy, but have not been studied at large. In this study, solid-phase extraction (SPE) and stir bar sorptive extraction (SBSE) combined with comprehensive two-dimensional gas chromatography and time-of-flight mass spectrometry (GC × GC-TOFMS) were applied to identify and quantify lactones in brandies between China and France. Totally 17 lactones were identified, four of which were detected only in SBSE. Among them, γ-valerolactone, γ-heptalactone, δ-octalactone, γ-undecanolactone and δ-dodecalactone were detected in brandy for the first time. The results of partial least squares-discriminant analysis (PLS-DA) revealed that lactones distinguished regional characteristics among different brandies. The omission test showed that four lactones (OAV > 1) had direct impact on the aroma of brandy, and other seven lactones at sub-threshold (0.1 < OAV < 1) provided peach and apricot aroma characteristics through synergistic effects. • Inexpensive and handheld sensor for carcinogenic formaldehyde detection. • Detects 5 ppb of formaldehyde within 2 min without interference. • Proven performance in indoor air and with wood product emissions. • Applicable for interconnected formaldehyde monitoring in "smart buildings". Formaldehyde is a carcinogenic indoor air pollutant emitted from wood-based furniture, building materials, paints and textiles. Yet, no low-cost sensor exists for on-site monitoring to fulfill stringent current and upcoming (e.g., 8 parts-per-billion by volume, ppb, in France by 2023) exposure guidelines. Here, we present an inexpensive and handheld formaldehyde detector with proven performance in real indoor air. Selectivity is achieved by a compact packed bed column of nanoporous polymer sorbent that separates formaldehyde from interferants present in ambient air. Downstream, a highly sensitive nanoparticle-based chemoresistive Pd-doped SnO 2 sensor detects formaldehyde in the relevant concentration range down to 5 ppb within 2 min. As a proof-of-concept, we measured formaldehyde in indoor air and from different wood product emissions, in excellent agreement (R2 > 0.98) with high-resolution proton-transfer-reaction time-of-flight mass spectrometry. This detector is simple-in-use and readily applicable for on-site formaldehyde exposure monitoring at home or work. Chlorinated paraffins (CPs) are high-volume chemicals used in numerous industrial applications. Their quantitative analysis is extremely challenging and this work presents the optimization of an analytical method based on gas chromatography hyphenated with electron capture negative ionization time-of-flight high-resolution mass spectrometry (GC-ECNI-TOF HRMS) for the simultaneous determination of short-chain and medium-chain CPs (SCCPs and MCCPs, respectively) in fish tissues (i.e. dorsal muscle). The resolution of the TOF-MS analyzer reduced or eliminated isobaric interferences and the CP response was optimized through Design of Experiment. A simple clean-up procedure based on adsorption chromatography further removed some potentially interfering organochlorines. Good selectivity, linearity and accuracy were achieved; method detection limits or limits of reporting were compatible with expected levels in wild fish (0.03-0.35 ng g -1 wet weight, ww, depending on the congener). This method was proven suitable for the analysis of CPs in tissues of common barbel Barbus, a fish species frequently used for water quality monitoring purposes in Europe. SCCPs and MCCPs were found to be widespread within the Rhône river basin (France). At all locations, MCCP concentrations (1.3-72.7 ng g -1 ww) were higher than those of SCCPs (0.3-10.6 ng g -1 ww) and levels were systematically lower than the proposed Predicted No Effect Concentrations (PNECs). The article discusses the highlights of a workshop on trends in cold neutron time-of-flight (CNTOF) spectroscopy held at the Institut Laue-Langevin (ILL) in Grenoble, France on November 26 and 27, 2009. Poster presentations included instrumental and sample environment developments, data acquisition and treatment and planned CNTOF instruments. It cites presentations of instrument projects from Korea, Australia, Germany and France.
