Background: Leptospirosis in cattle is recognized as a major cause of reproductive loses due to abortions, early embryonic death, and infertility. The type of Leptospira serovars that are currently circulating in Colombian herds is largely unknown due to the lack of national reports. Objective: The prevalence of antibodies specific to 13 Leptospira spp serovars was investigated in unvaccinated cows from 26 herds in San Pedro de los Milagros (Province of Antioquia) dairy region. Methods: Microscopic agglutination test was used on serum samples, following WHO recommendations. Results: A total of 112/900 (12.4%) of the animals had antibody titers of 1:100 or greater to at least one Leptospira serovar. Of those positive, 34/900 (3,8%) animals seroconverted to two or more serovars. In order of decreasing prevalence, the serovars detected were: hardjo prajitno, pomona, grippotyphosa, tarassovi, copenhageni, canicola, cellodeni and bratislava. There were 28 animals seropositive to serovar hardjo. Conclusion: Our results indicate that leptospirosis should be included as a differential diagnosis for various reproductive problems in this region. [ABSTRACT FROM AUTHOR]

A captive woolly monkey (Lagothrix lagothricha) displayed severe lethargy, vomiting and diarrhea. Laboratory tests revealed anemia, leukopenia, hypoproteinemia, severe azotemia and positive Leptospira IgM ELISA. The monkey was humanely euthanized and the necropsy revealed a multifocal tubulointerstitial glomerulonephritis; in addition to splenic lymphoid depletion, and interstitial pneumonia, all of which are compatible with leptospirosis. Rodent control and biosecurity measures should be done at all zoological collections in order to prevent transmission to facility personnel and to threatened mammals maintained in captivity. Abstract: Leptospirosis is caused by Leptospira, gram negative spirochaetes whose microbiologic identification is difficult due to their low rate of growth and metabolic activity. In Colombia leptospirosis diagnosis is achieved by serological techniques without unified criteria for what positive titers are. In this study we compared polymerase chain reaction (PCR) with microbiological culture and dark field microscopy for the diagnosis of leptospirosis. Microbiological and molecular techniques were performed on 83 samples of urine taken from bovines in the savannahs surrounding Bogotá in Colombia, with presumptive diagnosis of leptospirosis. 117 samples of urine taken from healthy bovines were used as negative controls. 83 samples were MAT positive with titers≥1:50; 81 with titers≥1:100; and 66 with titers≥1:200. 36% of the total samples (73/200) were Leptospira positives by microbiological culture, 32% (63/200) by dark field microscopy and 37% (74/200) by PCR. Amplicons obtained by PCR were 482 base pair long which are Leptospira specific. An amplicon of 262 base pairs typical of pathogenic Leptospira was observed in 71 out of the 74 PCR positive samples. The remaining 3 samples showed a 240 base pair amplicon which is typical of saprophytic Leptospira. PCR as a Leptospira diagnosis technique was 100% sensitive and 99% specific in comparison to microbiological culture. Kappa value of 0.99 indicated an excellent concordance between these techniques. Sensitivity and specificity reported for MAT when compared to microbiological culture was 0.95 and 0.89 with a≥1:50 cut off. PCR was a reliable method for the rapid and precise diagnosis of leptospirosis when compared to traditional techniques in our study. Objective. To determe leptospirosis seroprevalence and dominant serovars in canine population of three municipalities of Tolima department of Colombia. Materials and Methods. A cross-sectional study was carried, out (subjects being selected by sampling in 62 districts). Samples from 900 dogs during June, July and August 2007 were collected and processed using Microscopic Agglutination Test (MAT). Five serovars: hardjo, pomona, grippotyphosa, canicola, icterohaemorrhagiae and bratislava were tested. The MAT was considered positive when 50% or more leptospiras were agglutinated with one or more serovars in a serum dilution of 1:100. Results. At least one serovar showed evidence of infection in 21.4 % of the canines. The most dominant serovars were grippotyphosa: (82.4%) and pomona-(8.2%). It was evident the decrease in the frequency of reactivity of canicola and icterohaemorrhagiae serovars (0.5 and 2.2% respectively). Co-agglutinations were presented in 4.3% canine sera samples. The sera-reactivity showed statistically significant differences when was compared for sex and age (p=0.038). Conclusions. These findings suggested that dogs are potential reservoirs of Leptospira in the three municipalities evaluated. It is necessary to conduct studies that permit the isolation and characterization of the serovars prevalent in endemic areas to guide preventive measures to avoid the risk of disease transmission to human. Naturally acquired acute leptospirosis in monkeys is uncommon. This study reports an outbreak of severe leptospirosis among 52 capuchin (Cebus) monkeys that had been rescued from homes and housed in a wildlife rehabilitation center in Colombia in 2007. Case confirmation consisted of Leptospira isolation followed by a polymerase chain reaction targeting the LipL32 gene. The attack and mortality rates were 71% and 27%, respectively. Sixteen cases were confirmed. Necropsy revealed diffuse jaundice and pulmonary hemorrhage. Multi-locus sequence typing identified the agent to be Leptospira interrogans sequence type 17, indicating rats as the source of infection. An environmental survey confirmed rodent infestation as the cause of the outbreak. The extent of Leptospira transmission between humans and monkeys is unknown. Improper husbandry of non-human primates could create new reservoirs and transmission routes for Leptospira threatening conservation efforts and public health. [ABSTRACT FROM AUTHOR]
