Evaluating changes in the accumulation of grain minerals, including nitrogen (N), copper (Cu), iron (Fe), potassium (K), magnesium (Mg), manganese (Mn), phosphorus (P), and zinc (Zn), across different genotypes can provide valuable information for the development of nutrient-enriched maize varieties. Meanwhile, N rates can affect maize yield and quality, but their effects on element accumulation remain to be elucidated. Here, field experiments were conducted at two locations in China over 2 years (2010 and 2011). Under a normal N application rate (240 kg N ha−1), 24 maize cultivars that had been bred and released between 1930 and 2010 were evaluated for the elemental concentrations in the grains. Cultivars Yedan 13 and Zhengdan 958, representing old- and new-era cultivars respectively, were selected to investigate grain element accumulation in response to different levels of N (0, 60, 120, 180, and 240 kg N ha−1). The results showed that element concentrations were significantly affected by year, genotype (G), N rates, and N × G interaction. Grain yield tended to increase with the year of cultivar released, while the concentrations of N, Cu, Mn, and Zn in the grain significantly declined in the new-era. The element concentrations of grains were mainly influenced by N rate or N × G interactions. As N levels increased, N, Cu, Fe, Mg, and Mn concentrations rose, while K, P, and Zn concentrations decreased. Compared with old-era cultivars, new-era cultivars showed an increase in grain yield of 25.39%; however, they demonstrated decreases in N, Cu, Fe, K, Mg, P, and Zn concentrations. In the new-era varieties, the reduction in Cu, Fe, K, and P concentrations were significantly exacerbated by high N rates, but this was not the case in the old-era varieties. The concentration of grain Cu, K, Mg, P, and Zn were higher under N-limited condition (N0), but grain yield was also lower. However, the optimal N rate (120–180 kg N ha−1) could increase N, Fe, Mg, and Mn concentrations without affecting grain yield in new-era varieties. It is concluded that maize breeding processes have improved grain yield, but reduced grain nutrient element concentrations. Background and aims: Modern maize breeding has increased maize yields worldwide. The changes in above-ground traits accompanying yield improvement are well-known, but less information is available as to the effect of modern plant breeding on changes in maize root traits. Methods: Root growth, nitrogen uptake, dry matter accumulation and yield formation of six maize hybrids released from 1973 to 2000 in China were compared. Experiments were conducted under low and high nitrogen supply in a black soil in Northeast China in 2010 and 2011. Results: While nitrogen accumulation, dry matter production and yield formation have been increased, modern maize breeding in China since 1990 has reduced root length density in the topsoil without much effect on root growth in the deeper soil. The efficiency of roots in acquiring N has increased so as to match the requirement of N accumulation for plant growth and yield formation. The responses of root growth, nitrogen and dry matter accumulation, and grain yield to low-N stress were similar in the more modern hybrids as in the older ones. Conclusions: Modern maize breeding has constitutively changed root and shoot growth and plant productivity without producing any specific enhancement in root responsiveness to soil N availability. The impact of global changes on food security is of serious concern. Breeding novel crop cultivars adaptable to climate change is one potential solution, but this approach requires an understanding of complex adaptive traits for climate-change conditions. In this study, plant growth, nitrogen (N) uptake, and yield in relation to climatic resource use efficiency of nine representative maize cultivars released between 1973 and 2000 in China were investigated in a 2-year field experiment under three N applications. The Hybrid- Maize model was used to simulate maize yield potential in the period from 1973 to 2011. During the past four decades, the total thermal time (growing degree days) increased whereas the total precipitation and sunshine hours decreased. This climate change led to a reduction of maize potential yield by an average of 12.9% across different hybrids. However, the potential yield of individual hybrids increased by 118.5 kg ha−1 yr−1 with increasing year of release. From 1973 to 2000, the use efficiency of sunshine hours, thermal time, and precipitation resources increased by 37%, 40%, and 41%, respectively. The late developed hybrids showed less reduction in yield potential in current climate conditions than old cultivars, indicating some adaptation to new conditions. Since the mid-1990s, however, the yield impact of climate change exhibited little change, and even a slight worsening for new cultivars. Modern breeding increased ear fertility and grain-filling rate, and delayed leaf senescence without modification in net photosynthetic rate. The trade-off associated with delayed leaf senescence was decreased grain N concentration rather than increased plant N uptake, therefore N agronomic efficiency increased simultaneously. It is concluded that modern maize hybrids tolerate the climatic changes mainly by constitutively optimizing plant productivity. In recent years, agricultural growth in China has accelerated remarkably, but most of this growth has been driven by increased yield per unit area rather than by expansion of the cultivated area. Looking towards 2030, to meet the demand for grain and to feed a growing population on the available arable land, it is suggested that annual crop production should be increased to around 580 Mt and that yield should increase by at least 2% annually. Crop production will become more difficult with climate change, resource scarcity (e.g. land, water, energy, and nutrients) and environmental degradation (e.g. declining soil quality, increased greenhouse gas emissions, and surface water eutrophication). To pursue the fastest and most practical route to improved yield, the near-term strategy is application and extension of existing agricultural technologies. This would lead to substantial improvement in crop and soil management practices, which are currently suboptimal. Two pivotal components are required if we are to follow new trajectories. First, the disciplines of soil management and agronomy need to be given increased emphasis in research and teaching, as part of a grand food security challenge. Second, continued genetic improvement in crop varieties will be vital. However, our view is that the biggest gains from improved technology will come most immediately from combinations of improved crops and improved agronomical practices. The objectives of this paper are to summarize the historical trend of crop production in China and to examine the main constraints to the further increase of crop productivity. The paper provides a perspective on the challenge faced by science and technology in agriculture which must be met both in terms of increased crop productivity but also in increased resource use efficiency and the protection of environmental quality. Modern maize cultivars have high nitrogen responsiveness of grain yield. However, the underlying physiological mechanism is unclear. In the present study, from 17 cultivars released from 1950 s to 2010 s in China, three maize cultivars released in 1950 s (BMY), 1970 s (ZD2), and 1990 s (ZD958) were selected according to their growth responses to nitrogen supply. Nitrogen responsiveness of shoot biomass was highest in the new cultivar ZD958, and the lowest in the old cultivar BMY, with the medium cultivar ZD2 in between. Shoot biomass was closely related to leaf elongation rate. Leaf elongation was further analyzed using kinematic analysis method. Cell division rate and cell production rate were greater and more responsive to nitrogen supply in ZD958 than in the other cultivars. In agreement, there were more cells in the elongation zone of ZD958 and they were more responsive to increasing supply, resulting a longer leaf elongation zone. The interaction effect between cultivar and nitrogen supply on leaf elongation was in consistence with the change in gibberellin (GA 1 and GA 3) concentration in leaf growth zone. Accordingly, the genes encoding gibberellin biosynthesis or signal transduction was more up-regulated in ZD958, which was positively correlated to the up-regulation of genes related to cell cycle and DNA replication. On the contrary, the genes related to the inactivation of gibberellin was more down-regulated in ZD958. Exogenous application of bioactive gibberellin or its biosynthesis inhibitor eliminated the interaction effect between cultivar and nitrogen supply on leaf elongation rate. In conclusion, modern maize breeding may have increased nitrogen responsiveness in maize by enhancing the gibberellins-dependent leaf elongation response to high nitrogen supply. • Nitrogen responsiveness is partly explained by leaf elongation rate. • Nitrogen responsiveness of leaf elongation is driven by gain in cell division. • Nitrogen-dependent gibberellin biosynthesis and signal transduction promote cell division in leaf growth zone. In order to understand the regulation of source-sink relationship on leaf senescence in maize, we investigated the effect of pollination prevention on leaf senescence, post-silking dry matter and nitrogen accumulation and remobilization in different vegetative organs in one landrace and ten representative maize hybrids released between 1973 and 2000 in China. When pollination was prevented by covering silk with paper bags at silking stage, leaf senescence of Nongda 60, Yedan 13, Shendan 7 and Zhengdan 958 was delayed as shown by the remaining green leaf area per plant at maturity. However, pollination-prevention did not have obvious effect on the leaf senescence of Baimaya, Danyu 6, Yedan 2 and Xianyu 335. By contrast, pollination-prevention accelerated the leaf senescence of maize hybrids Zhongdan 2, Danyu13 and Yedan 4. It was found that leaf senescence of the late-released (since 1985) Chinese hybrids tended to be delayed by pollination-prevention, except XY335. From silking to physiological maturity, nitrogen content increased in the stem (plus the sheath, cob, husk, and tassel) and root of the non-pollinated plants. However, there was still a net reduction in leaf nitrogen content. We also found that pollination-prevention reduced leaf nitrogen remobilization efficiency, with genotypic difference and variation between the two years of testing. The results suggested that the response of leaf senescence to pollination-prevention is at least partially due to the change of leaf nitrogen remobilization efficiency. Genotypic difference in nitrogen responsiveness of maize grain yield has gained considerable attention, however, the underlying physiological mechanism is less understood. Leaf area and plant type are important factors determining biomass production, and both of them are regulated by nitrogen supply. The current research aims to assess whether high nitrogen responsiveness is related to leaf growth and plant type. A 2-year field experiment was conducted with three maize cultivars (BMY, ZD2 and ZD958) differing in nitrogen responsiveness under a wide range of nitrogen supply (0, 60, 120, 180 and 240 kg N ha-1). The links between grain yield, leaf area dynamics, leaf angle and orientation value, and radiation use efficiency were investigated. Nitrogen responsiveness, defined as yield increment per unit of increased nitrogen supply, was 8.3–29.1% higher in ZD958 compared with the other cultivars. The maximum grain yield of ZD958 was 21.4–74.6% higher than that of the other cultivars. With increasing nitrogen supply, the increments in maximum leaf area and linear growth rate of leaf area were greater in ZD958, which was positively correlated with grain yield. The interaction effect between cultivar and nitrogen supply was significant on leaf length, but not on leaf width. Across the combinations between cultivar and nitrogen supply, the variation in grain yield was explained by leaf length rather than leaf width. High nitrogen supply increased leaf angle and reduced leaf orientation value, so as to make the leaf more horizontally distributed. However, this change was less significant in ZD958, indicating that the plant type was more compact under high nitrogen supply in maize cultivars with high nitrogen responsiveness. Radiation use efficiency, defined as the slope of the linear relationship between above-ground biomass produced and cumulated intercepted photosynthetic active radiation (PAR) was 5.2–13.2% higher in ZD958 than that of BMY. In conclusion, with increasing nitrogen supply, the cultivars with fast leaf growth and compact plant type have higher conversion efficiency of intercepted PAR into the above-ground biomass and get higher grain yield. • Nitrogen increased leaf area to a larger extent in the maize cultivar with high N responsiveness. • A compact plant type contributed to high N responsiveness under high N supply. • Grain yield increased via gains in radiation use efficiency under high N supply. Selection of high-yielding and nitrogen (N)-efficient cultivars under realistic agronomic conditions can benefit farmers in their production practices. In addition, phenotyping the root characteristics of high-yielding and N-efficient cultivars under N stress conditions is important to guide maize breeding for abiotic stress tolerance. The objective of this study was to (1) evaluate the potential increase in yield and reduction in required fertilizer input in maize cultivars, and (2) examine the root characteristics of high-yielding and N-efficient cultivars. Under the high-N (240 kg N ha−1) and the low-N (no N supplied) conditions, grain yield and nodal root traits of 10–67 dominant maize cultivars grown in North and Northeast China were investigated in field trials at six environments. The low-N conditions reduced grain yield by 24%, crown root number (CRN, shoot-borne roots formed at consecutive underground shoot nodes) by 8%, brace root number (BRN, shoot-borne roots formed at consecutive aboveground shoot nodes) by 44%, and brace root whorl number (BRWN, whorls of roots that arise from aboveground shoot nodes) by 50%. The grain yield showed a significant positive correlation with CRN and crown root length (CRL) under both N treatments. The cultivars were classified into four types based on the average grain yield under high-N and low-N treatments: EE, efficient under both high-N and low-N treatments; HNE, N efficient only under high-N treatment; LNE, N efficient only under low-N treatment; and NN, inefficient under both high-N and low-N treatments. The cultivars with EE and HNE types had 5%–10% greater grain yield and reduced N fertilizer input requirements by 11%–50%. Under the low-N conditions, the cultivars classified as EE and HNE types showed 4%–20% higher CRN and 14%–22% longer CRL compared with NN types. Cultivation of the EE and HNE maize cultivars in Northeast and North China may reduce N input requirements without compromising yield. Compared with NN type cultivars, the EE and HNE cultivars had greater CRN and CRL, which may be contributed to improving N acquisition and grain yield formation in response to N stress. Abstract: Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease with unknown pathophysiological mechanisms. Profilin 1 gene (PFN1) has been identified as a causative gene, which accounts for 1% to 2% of familial ALS. In this study, we investigated the mutation spectrum of PFN1 in Chinese patients with ALS. A total of 550 ALS patients (including 540 sporadic ALS [SALS] and 10 familial ALS) from the Department of Neurology, West China Hospital of Sichuan University, were recruited for the study. From the same region, 545 healthy control individuals (HC) were recruited as a control group. The encoding regions of the PFN1 gene were screened by direct sequencing. Novel candidate mutations or variations were confirmed by polymerase chain reaction-restriction fragment length polymorphism. A novel nonsynonymous p.R136W mutation was identified in an early-onset SALS female patient. A novel synonymous mutation p.L88L detected in a late-onset SALS female patient was considered nonpathogenic, as it was also detected in a control subject. No mutations were found in 10 familial ALS patients. Moreover, we found a significant difference in the genotype distribution of reported rs13204 (p.L112L) between SALS patients and HC (p = 0.0030). The frequency of minor allele ‘T’ of rs13204 in the SALS group was significantly lower than that in HC (p = 0.0040, OR = 0.7270, 95% CI = 0.5848–0.9039). Our results suggest that PFN1 mutation is an uncommon cause of ALS in the Han Chinese population. The SNP rs13204 of the PFN1 gene may have an important function in ALS development. To discuss combinations of traditional screening and noninvasive prenatal screening (NIPS) and to compare which traditional screening is the most suitable first-line screening approach to NIPS, pregnant women were recruited in this retrospective observational study. Pregnant women underwent one of four traditional screening tests. The 9 contingent models were combined by high risk cut-offs of 1:50, 1:100, 1:270 and intermediate risk cut-offs of 1:1000, 1:1500, 1:2000. We analyzed cost and performance of various screening models with contingent screening of different risk cut-offs. Compared with other screening tests, combined first-trimester screening (CFTS) had the lowest proportion of high risk (≥1:270) with the highest detection rate (DR) (78.79%) and the lowest proportion of intermediate risk (1:271~1:1000). When intermediate risk was 1:51 ~1:1500, CFTS as first-line screening had the lowest cost with DR of 93.94%. Other screening tests as the first-line screening with intermediate risk of 1:51~1:1000 had the lowest cost, there DR were 90.91%, 84.62%, 91.67%, respectively. Our study demonstrated if only one traditional screening was allowed to screen pregnant women, CFTS was recommended as the first choice. According to local health and economic conditions, adopting appropriate traditional screening with suitable cut-offs as first-line screening will contributed to a cost-effective screening model.

Mutations in the sequestosome 1 gene (SQSTM1) have recently been identified in patients with amyotrophic lateral sclerosis, accounting for 1.11%-4.92% of familial ALS and 2.42%-4.37% of sporadic amyotrophic lateral sclerosis (SALS). The mutation spectrum of SQSTM1 in Chinese patients with SALS remains unknown. Three hundred and six patients with SALS from the Department of Neurology, West China Hospital of Sichuan University were recruited for this study. From the same region, 350 healthy individuals were recruited as a control group. The encoding regions of SQSTM1 were screened by direct sequencing. Three novel nonsynonymous mutations- p. I99L, p. D337E, and p. L341V-were identified in 3 patients with SALS, none of which were found in healthy controls. The male patient carrying mutation p. I99L presented limb symptom at age of 34 and died in 34 months. Two late-onset patients carrying D337E and p. L341V mutations had bulbar and limb onset, respectively. Moreover, a c.1166-14_1166-11delTACT mutation in the intron 7 was found in a living male patient with limb onset at age of 62. None of the patients carrying SQSTM1 mutation showed clinical evidence of concomitant Paget disease of bone or mutation of the valosin-containing protein gene. Root and rhizosphere research has been conducted for many decades, but the underlying strategy of root/rhizosphere processes and management in intensive cropping systems remain largely to be determined. Improved grain production to meet the food demand of an increasing population has been highly dependent on chemical fertilizer input based on the traditionally assumed notion of 'high input, high output', which results in overuse of fertilizers but ignores the biological potential of roots or rhizosphere for efficient mobilization and acquisition of soil nutrients. Root exploration in soil nutrient resources and root-induced rhizosphere processes plays an important role in controlling nutrient transformation, efficient nutrient acquisition and use, and thus crop productivity. The efficiency of root/rhizosphere in terms of improved nutrient mobilization, acquisition, and use can be fully exploited by: (1) manipulating root growth (i.e. root development and size, root system architecture, and distribution); (2) regulating rhizosphere processes (i.e. rhizosphere acidification, organic anion and acid phosphatase exudation, localized application of nutrients, rhizosphere interactions, and use of efficient crop genotypes); and (3) optimizing root zone management to synchronize root growth and soil nutrient supply with demand of nutrients in cropping systems. Experiments have shown that root/rhizosphere management is an effective approach to increase both nutrient use efficiency and crop productivity for sustainable crop production. The objectives of this paper are to summarize the principles of root/rhizosphere management and provide an overview of some successful case studies on how to exploit the biological potential of root system and rhizosphere processes to improve crop productivity and nutrient use efficiency.

Root growth has a fundamental role in nitrogen (N) use efficiency. Nevertheless, little is known about how modern breeding progress has affected root growth and its responses to N supply. The root and shoot growth of a core set of 11 representative Chinese maize (Zea mays L.) hybrids released between 1973 and 2009 were investigated under high N (4 mmol L(-1), HN) and low N (0.04 mmol L(-1), LN) levels in a solution culture system. Compared with LN, HN treatment decreased root dry weight (RDW), the root: shoot ratio (R/S), and the relative growth rate for root dry weight (RGR(root)), but increased the total root length (TRL) and the total lateral root length (LRL). The total axial root length (ARL) per plant was reduced under HN, mostly in hybrids released before the 1990s. The number of seminal roots (SRN) was largely unaffected by different N levels. More recently released hybrids showed higher relative growth rates in the shoot under both HN and LN. However, the roots only showed increased RGR under HN treatment. Correspondingly, there was a positive linear relationship with the year of hybrid release for TRL, LRL and ARL under HN treatment. Together, these results suggest that while shoot growth of maize has improved, its root growth has only improved under high N conditions over the last 36 years of selective breeding in China. Improving root growth under LN conditions may be necessary to increase the N use efficiency of maize.
