Background: The herbaceous peony (Paeonia lactiflora Pall.) is extensively cultivated in China due to its root being used as a traditional Chinese medicine known as 'Radix Paeoniae Alba'. In recent years, it has been discovered that its seeds incorporate abundant unsaturated fatty acids, thereby presenting a potential new oilseed plant. Surprisingly, little is known about the full-length transcriptome sequencing of Paeonia lactiflora, limiting research into its gene function and molecular mechanisms. Results: A total of 484,931 Reads of Inserts (ROI) sequences and 1,455,771 full-Length non-chimeric reads (FLNC) sequences were obtained for CDS prediction, TF analysis, SSR analysis and lncRNA identification. In addition, gene function annotation and gene structure analysis were performed. A total of 4905 transcripts were related to lipid metabolism biosynthesis pathway, belonging to 28 enzymes. We use these data to identify 10 oleosin (OLE) and 5 diacylglycerol acyltransferase (DGAT) gene members after de-redundancy. The analysis of physicochemical properties and secondary structure showed them similarity in gene family respectively. The phylogenetic analysis showed that the distribution of OLE and DGAT family members was roughly the same as that of Arabidopsis. Quantitative real-time polymerase chain reaction (qRT–PCR) analyses revealed expression changes in different seed development stages, and showed a trend of increasing and then decreasing. Conclusion: In summary, these results provide new insights into the molecular mechanism of triacylglycerol (TAG) biosynthesis and storage during the seedling stage in Paeonia lactiflora. It provides theoretical references for selecting and breeding oil varieties and understanding the functions of oil storage as well as lipid synthesis related genes in Paeonia lactiflora. The main objective of this study was to evaluate the quality and select the best varieties from 85 olive varieties in the Liangshan region, China, from the perspective of oil content, fatty acid, and triacylglycerol compositions. Although 85 varieties of olive oil showed no difference in the type of fatty acid composition and the distribution of triglycerides, they varied greatly in the oil content, the relative proportion of fatty acids, and triglycerides. Principal component analysis (PCA) and cluster and heatmap analysis clearly divided the 85 samples into three groups. Group A included 36 varieties and was characterized by high oleic acid, mono-unsaturated fatty acids (MUFA), eicosenoic acid, OOO (glyceryl trioleate), and OOL (1-oleic acid-2-oleic acid-3-linoleic acid glycerides) contents, but rare palmitoleic acid, SFA (saturated fatty acid), and POP (1-palmitic acid-2-oleic acid-3-palmitic acid glycerides) levels. Group B contained 9 varieties of olive, with the main characteristics of high oleic acid, linolenic acid, MUFA, MUFA/PUFA, SFA, and OLnO (1-oleic acid-2-linolenic acid-3-oleic acid glycerides) content, but low in linoleic acid, PUFA (polyunsaturated fatty acid), PLL levels and ratio of omega-6 to omega-3 polyunsaturated fatty acids. Group C comprised 40 varieties, identified by high linoleic acid and PUFA levels but low ratios of oleic acid, MUFA, OOO, MUFA/PUFA, and the ratio of omega-6 to omega-3 polyunsaturated fatty acids. Combined with the oil content (FW) (%) of 85 varieties, we suggested four distinguished varieties in Group A, namely Lechín, Coratina, Koroneiki, and Arbosana; three distinguished varieties in Group B, namely Picual, Ezhi, Cornicabra; two distinguished varieties in Group C namely Frantoio and Arbequina as the objects of large-scale cultivation by Chinese cultivators. More importantly, the obtained data also demonstrated that some locally bred specific varieties, such as Zhongze-3 and Yuntai, had interesting unsaturated fatty acids and had cultivation and popularization value in China. Amaranthus tricolor and Amaranthus dubius are the most prevalent vegetable amaranths in China. The fatty acid composition, triacylglycerol (TAG) profile, vitamin E and sterol content, and thermal properties of seed oils extracted from these two vegetable amaranths were investigated in this work. The results reveal that the seed contains ∼8% oil. The main fatty acids in the oils are linoleic (51.5–53.5%), oleic (14.8–18.4%), palmitic (15.7–17.1%) and stearic (3.6–4.1%). PLL (∼22%), LLL (∼20%), LLO (∼17%) and POL (∼11%) are the dominant TAGs in the oils. The amaranth seed oils exhibit a distinct melting and crystallization profile that consists of 5 endothermic peaks and 2 exothermic peaks. The oils are rich in vitamin E (705–829 mg/kg) with δ-tocopherol as the major component. The two oils have similar total sterol contents (2488.7 and 2762.1 mg/100 g) but different individual sterol percentages. The higher contents of oil, linoleic acid, vitamin E, and sterols make the amaranth seed oil desirable in terms of nutrition. • Properties of seed oil from vegetable amaranth were comparatively investigated. • The oil of vegetable amaranth contained more linoleic acid. • The vitamin E and sterol profile of vegetable amaranth oil is different from those of grain amaranth. • The thermal properties of amaranth seed oil were first reported. • Amaranth oil is a good source for developing high-quality vegetable oil. Background/aims: Hepatitis E virus (HEV)-triggered acute-on-chronic liver failure (ACLF) has unacceptably high short-term mortality. However, it is unclear whether the existing predictive scoring models are applicable to evaluate the prognosis of HEV-triggered ACLF., Methods: We screened datasets of patients with HEV-triggered ACLF from a regional tertiary hospital for infectious diseases in Shanghai, China, between January 2011 and January 2021. Clinical and laboratory parameters were recorded and compared to determine a variety of short-term mortality risk factors, which were used to develop and validate a new prognostic scoring model., Results: Out of 4952 HEV-infected patients, 817 patients with underlying chronic liver disease were enrolled in this study. Among these, 371 patients with HEV-triggered ACLF were identified and allocated to the training set (n = 254) and test set (n = 117). The analysis revealed that hepatic encephalopathy (HE), ascites, triacylglycerol and apolipoprotein A (apoA) were associated with 90-day mortality (P < 0.05). Based on these significant indicators, we designed and calculated a new prognostic score = 0.632 × (ascites: no, 1 point; mild to moderate, 2 points; severe, 3 points) + 0.865 × (HE: no, 1 point; grade 1-2, 2 points; grade 3-4, 3 points) - 0.413 × triacylglycerol (mmol/L) - 2.171 × apoA (g/L). Compared to four well-known prognostic models (MELD score, CTP score, CLIF-C OFs and CLIF-C ACLFs), the new scoring model is more accurate, with the highest auROCs of 0.878 and 0.896, respectively, to predict 28- and 90-day transplantation-free survival from HEV-triggered ACLF. When our model was compared to COSSH ACLF IIs, there was no significant difference. The test data also demonstrated good concordance., Conclusions: This study is one of the first to address the correlation between hepatitis E and serum lipids and provides a new simple and efficient prognostic scoring model for HEV-triggered ACLF., (© 2023. Background: Bioactive lipids play an important role in insulin secretion and sensitivity, contributing to the pathophysiology of type 2 diabetes (T2D). This study aimed to identify novel lipid species associated with incident T2D in a nested case-control study within a long-term prospective Chinese community-based cohort with a median follow-up of ~ 16 years., Methods: Plasma samples from 196 incident T2D cases and 196 age- and sex-matched non-T2D controls recruited from the Hong Kong Cardiovascular Risk Factor Prevalence Study (CRISPS) were first analyzed using untargeted lipidomics. Potential predictive lipid species selected by the Boruta analysis were then verified by targeted lipidomics. The associations between these lipid species and incident T2D were assessed. Effects of novel lipid species on insulin secretion in mouse islets were investigated., Results: Boruta analysis identified 16 potential lipid species. After adjustment for body mass index (BMI), triacylglycerol/high-density lipoprotein (TG/HDL) ratio and the presence of prediabetes, triacylglycerol (TG) 12:0&#95;18:2&#95;22:6, TG 16:0&#95;11:1&#95;18:2, TG 49:0, TG 51:1 and diacylglycerol (DG) 18:2&#95;22:6 were independently associated with increased T2D risk, whereas lyso-phosphatidylcholine (LPC) O-16:0, LPC P-16:0, LPC O-18:0 and LPC 18:1 were independently associated with decreased T2D risk. Addition of the identified lipid species to the clinical prediction model, comprised of BMI, TG/HDL ratio and the presence of prediabetes, achieved a 3.8% improvement in the area under the receiver operating characteristics curve (AUROC) (p = 0.0026). Further functional study revealed that, LPC O-16:0 and LPC O-18:0 significantly potentiated glucose induced insulin secretion (GSIS) in a dose-dependent manner, whereas neither DG 18:2&#95;22:6 nor TG 12:0&#95;18:2&#95;22:6 had any effect on GSIS., Conclusions: Addition of the lipid species substantially improved the prediction of T2D beyond the model based on clinical risk factors. Preterm infants with physiological immaturity require higher lipid provision than term infants. This study compared the lipid composition, including total, sn-2 fatty acid, and triacylglycerol (TAG) compositions in 14 preterm formulas and 25 term formulas in the Chinese market, in 2020-2021. Preterm formula had higher medium-chain fatty acid (MCFA) and comparable C22:6n-3 (DHA) contents than term formula. Notably, significantly lower C16:0 and C18:0 (p < 0.001) were distributed on the sn-2 position in preterm formula. Two hundred and thirteen kinds of TAG molecular species were identified using UPLC-Q-TOF-MS. In preterm formula, significantly higher Ca-Ca-Cy and Ca-Ca-Ca (p < 0.001) existed. These clear distinctions showed the challenge of the progress in preterm formula, such as DHA status, MCFA pattern, and TAG esterified with palmitic acid on the sn-2 position., (Copyright © 2022 Elsevier Ltd. • Characterization of five PSOs presented in TAGs, fatty acids, two micronutrients. • PSOs could enhance the contents of unsaturated fatty acidsand micronutrients. • Principal components analysis allows distinction of cultivation areas of PSOs. Peony (Paeonia lactiflora pall) is a unique plant in China, which is planted on a large scale in different areas. The study characterized the triacylglycerol (TAG), fatty acid (FA), phytosterol, and tocopherol in peony seed oil (PSO) from five cultivation areas in China. The FAs in PSO were mainly C 18 fatty acyl chains, especially α-linolenic acid (C 18:3n-3). The triacylglycerol of PSO was identified by UPLC-Q-ToF/MS, and the predominant TAGs were LnLnLn, LLnLn, LLLn, LnLO/LLL, and OOLn/OLL. As for the micronutrients, γ-tocopherol and β-sitosterol were main components of tocopherol and phytosterol, respectively. The obtained data were further analyzed by principal components analysis (PCA), which allowed differentiation of five PSOs by their cultivation areas. For the great potential in unsaturated fatty acid and micronutrients, PSO could be a good dietary resource of plant oil. [ABSTRACT FROM AUTHOR]

The nutritional composition and chemical properties of the Chinese highland barley bran oil were characterized in this study. The barley bran oil extracted with solvent possessed relatively high acid value and peroxide value, indicating that the oil should be further refined before using. The fatty acid composition of the oil showed that the content of unsaturated fatty acids was 80.12 g/100 g, in which the content of polyunsaturated fatty acids was as high as 60.41 g/100 g. The overall triacylglycerol profile showed that the oil contained 27 TAGs including 21 regioisomers. Major TAGs included LLL (21.08 g/100 g), PLL (19.27 g/100 g), LLO (12.24 g/100 g), and LLLn (12.17 g/100 g). The total unsaponifiable matter of the oil reached up to 10.74 g/100 g oil. The total phytosterol content reached 7.90 g/100 g oil, in which β-sitosterol was the most predominant, with the content of 5.69 g/100 g oil. Other important sterols included campesterol (1.32 g/100 g oil), lanosterol (0.70 g/100 g oil) and stigmasterol (0.19 g/100 g oil).

Screening suitable strains with high temperature adaptability is of great importance for reducing the cost of temperature control in microalgae cultivation, especially in summer. To obtain high temperature-tolerant diatoms, water samples were collected in summer from 7 different regions of China across the Northeast, North and East. A total of 731 water samples was collected and from them 131 diatom strains were isolated and identified based on the 18S rRNA sequences. Forty-nine strains out of the 131 diatoms could survive at 30°C, and 6 strains with relatively high biomass and lipid content at high temperature were selected and were found to be able to grow at 35°C. Cyclotella sp. HB162 had the highest dry biomass of 0.46 g/l and relatively high triacylglycerol (TAG) content of 237.4 mg/g dry biomass. The highest TAG content of 246.4 mg/g dry biomass was obtained in Fistulifera sp. HB236, while Nitzschia palea HB170 had high dry biomass (0.33 g/l) but relatively low TAG content (105.9 mg/g dry biomass). N. palea HB170 and Fistulifera sp. HB236 presented relatively stable growth rates and lipid yields under fluctuating temperatures ranging from 28 to 35°C, while Cyclotella HB162 maintained high lipid yield at temperatures below 25°C. The percentage of saturated fatty acids and monounsaturated fatty acids in all the 6 strains was 84-91% in total lipids and 90-94% in TAGs, which makes them the ideal feedstock for biodiesel.

Triacylglycerol (TAG) composition of breast milk plays an important role in improving digestion, absorption, and metabolism when consumed by infants. This study characterized the TAG profile of human colostrum, transitional, and mature milk samples from 103 women. Significant differences in the TAGs composition of breast milk fat from three lactation stages were observed. The TAGs with high molecular weight and unsaturated fatty acid (such as 1,3-olein-2-palmitin (OPO) and 1(3)-olein-2-palmitin-3(1)-linolein (OPL)) were enriched in colostrum, while the TAGs containing medium-chain fatty acids were more abundant in transitional and mature milk than that in colostrum. Of note, OPL was the most common TAG in breast milk of Chinese women while the most common TAG in breast milk of Western women was OPO. This data will promote the development of infant formulas in terms of the TAG composition more suitable for infants.

Aims/hypothesis: Glucokinase regulatory protein gene (GCKR) variant rs780092 is a novel genetic variant associated with serum triacylglycerol (TG) identified in a genome-wide association study in East Asians. We aimed to investigate associations of rs780092 with incident type 2 diabetes and dyslipidaemia, and the longitudinal changes in glucose and lipid levels., Methods: A community-based prospective cohort study was conducted at baseline in 2008, including 5,613 non-diabetic participants (37% male, mean age 57.6 years) with 5 years of follow-up. Blood glucose and lipid was measured at baseline and follow-up., Results: Each rs780092 T-allele was associated with a 17% lower risk of incident type 2 diabetes (HR 0.83 [95% CI 0.73, 0.95]) and 36% higher risk of incident hypertriacylglycerolaemia (OR 1.36 [95% CI 1.08, 1.72]), after adjustment for baseline fasting glucose and TG and other confounders. The T-allele was associated with a 5 year increasing level of log10 TG (β ± SE, 0.01 ± 0.004, p = 0.005). Mediation analysis showed that both baseline TG and the 5 year increase in log10 TG were significant mediators in the associations of rs780092 with risk of diabetes. The risk of incident type 2 diabetes associated with 1 SD increase in total and LDL-cholesterol was 35% and 22% lower in TT carriers compared with CC carriers, respectively (both p for interaction ≤ 0.04)., Conclusions/interpretation: The GCKR rs780092 variant showed opposite-directional associations with type 2 diabetes and hypertriacylglycerolaemia in a Chinese population. Both baseline level and 5 year change in serum TG were mediators of the association between the genetic variant and type 2 diabetes.

Lemnaceae (duckweeds) are widely distributed aquatic flowering plants. Their high growth rate, starch content and suitability for bioremediation make them potential feedstock for biofuels. However, few natural duckweed resources have been investigated in China, and there is no information about total fatty acid (TFA) and triacylglycerol (TAG) composition of duckweeds from China. Here, the genetic diversity of a natural duckweed population collected from Lake Chao, China, was investigated using multilocus sequence typing (MLST). The 54 strains were categorised into four species in four genera, representing 12 distinct sequence types. Strains representing Lemna aequinoctialis and Spirodela polyrhiza were predominant. Interestingly, a surprisingly high degree of genetic diversification within L. aequinoctialis was observed. The four duckweed species revealed a uniform fatty acid composition, with three fatty acids, palmitic acid, linoleic acid and linolenic acid, accounting for more than 80% of the TFA. The TFA in biomass varied among species, ranging from 1.05% (of dry weight, DW) for L. punctata and S. polyrhiza to 1.62% for Wolffia globosa. The four duckweed species contained similar TAG contents, 0.02% mg · DW(-1). The fatty acid profiles of TAG were different from those of TFA, and also varied among the four species. The survey investigated the genetic diversity of duckweeds from Lake Chao, and provides an initial insight into TFA and TAG of four duckweed species, indicating that intraspecific and interspecific variations exist in the content and composition of both TFA and TAG in comparison with other studies., (© 2015 German Botanical Society and The Royal Botanical Society of the Netherlands.)
