Alternative polyadenylation (APA) plays a crucial role in cancer biology. Here, we used data from the 3′aQTL-atlas, GTEx, and the China Nanjing Lung Cancer GWAS database to explore the association between apaQTL/eQTL-SNPs and the risk of lung adenocarcinoma (LUAD). The variant T allele of rs277646 in NIT2 is associated with an increased risk of LUAD (OR = 1.12, P = 0.015), lower PDUI values, and higher NIT2 expression. The 3′RACE experiment showed multiple poly (A) sites in NIT2, with the rs277646-T allele causing preferential use of the proximal poly (A) site, resulting in a shorter 3′UTR transcript. This leads to the loss of the hsa-miR-650 binding site, thereby affecting LUAD malignant phenotypes by regulating the expression level of NIT2. Our findings may provide new insights into understanding and exploring APA events in LUAD carcinogenesis. Since the release of the 13th Five-Year Plan, the construction plan of the smart city and Internet of Things (IoT) has once again pushed the sensors into the limelight. In the current situation, whether in China or around the world, the construction of smart communities has become an irreversible trend. In this environment, the sensor as a smart city "bridge" will certainly usher in an industrial explosion. "Smart city" is a new concept and model for urban development. Academics have also explored the concept and theoretical model of smart cities from different perspectives in different forms. With the gradual establishment of the national smart city system, the Internet of Things and other technologies are being widely used to serve other aspects of human social life, such as urban smart communities, intelligent transportation, and intelligent home security systems. The application of technology is gradually penetrating into the life of our human society. In the context of building a smart city, scholars have also brought an unprecedented broader idea. By seeking a reasonable balance between human and urban nature, the concept of "park city," which aims to create a good ecological habitat where people and nature can coexist and develop in harmony, has emerged as China's urbanization process accelerates and the conflict between people and land becomes more and more prominent. As an important factor to promote the harmonious development of humans and nature, the planning and construction of urban parks is an important means to meet the needs of people for a better life and to improve the ecological environment of cities; therefore, the planning and design of urban parks under the concept of "park city" is getting more and more attention. Qingdao, as a typical coastal city, has a mosaic of green areas and urban construction land, so urban ecological parks with the carrier of nonconstruction land exist in large numbers in the city and become a key link in the practice of the "park city" concept. However, while urban ecological parks provide ecological and social services to citizens, their special land use has also led to a series of problems. At present, there is not enough basis for the planning and management of such special parks in Qingdao, and it is not possible to guide the planning and construction of such parks with universal standards, so how to effectively protect and reasonably utilize them to promote the construction of "park city" is an urgent problem to be solved. Based on the above background, this study takes Qingdao urban ecological park as the research object and summarizes the typical problems in the process of planning, construction, and use of this kind of park through relevant data research and extensive studies. Based on this, we propose three aspects, namely, the location and layout of parks at a macro level, the construction of the park system at meso-level, and the design optimization of parks at a micro level. The specific research content includes the following aspects: the first part defines the background, purpose, and significance of the topic and the research object and summarizes and reviews the existing research results at home and abroad, and then proposes the overall idea and framework of the research. The second part analyzes the problems of Qingdao urban ecological parks with the concept of "park city." Based on the connotation and characteristics of the "park city" concept, the impact of the concept on urban parks is analyzed, and the significance of the "park city" concept on the planning and construction of urban ecological parks is clarified based on the special characteristics of urban ecological parks. Finally, we provide the basis for the proposed strategy by combining case studies at different levels. Species of Mycena sect. Calodontes are representative of the Mycena genus as a whole and are easily recognised by the pinkish, reddish, purplish to brownish pileus and larger basidiomata. Furthermore, the colour of the pileus in the species of sect. Calodontes often has a transition or changes in different stages and the combination of the colour of the pileus with cystidia and basidiospores can be used to recognise taxa within this section. To date, 19 species of Mycena sect. Calodontes have been reported worldwide. Including our recent description of M. yuezhuoi, five species of sect. Calodontes have been recorded in China. During examination of specimens collected in coniferous forests or mixed broadleaf-conifer forests in temperate regions of China, additional taxa assigned to sect. Calodontes were identified. Four new species are recognised, based mostly on characters of the pileus and cystidia. Phylogenetic analysis of sequence data from multiple DNA regions (ITS + rpb1 + tef1) supported the morphological evidence. Here, we propose M. polycystidiata, M. rufobrunnea, M. shengshanensis and M. subulata as new species in Mycena sect. Calodontes. Only three Gerronema (Porotheleaceae) species have been previously recorded in China. Here, we report collections of a fourth species in China: G. nemorale Har. Takah., which is widely distributed in Chinese temperate to subtropical zones. We also formally describe three new species, collected from Anhui, Fujian, and Zhejiang provinces: G. baishanzuense sp. nov., G. microcarpum sp. nov., and G. zhujian sp. nov. Furthermore, we include the results of a phylogenetic analysis of Porotheleaceae based on a multi-locus (ITS + nLSU) dataset. The results, which indicate that Gerronema is polyphyletic, support the taxonomic recognition of the three new species. Morphological descriptions, photographs, line drawings, and comparisons with closely related taxa are presented for the new and newly recorded species. Only a few Crepidotus species have been previously reported from subalpine areas of China. Members of Crepidotus possessing an orange-yellow pileus are rare in China as well. Here, we describe Crepidotus yuanchui, a new species characterized by an orange-yellow pileus, ovoid basidiospores, and abundant cylindric to narrowly utriform cheilocystidia. This species is widely distributed above 2000 m asl in Yunnan Province. In addition, C. caspari, which is a distinct species based on its combined characteristics of smooth basidiospores and clamp connections, is newly recorded and detailed from subalpine China. The results of phylogenetic analyses of ITS + nLSU sequences based on maximum likelihood and Bayesian inference methods support the recognition of these two species. An updated description of the genus Atheniella, combining macro- and micromorphological characters that elaborate on the original generic characterisation, is presented. Atheniella is characterised by a brightly coloured pileus, all tissues inamyloid and pileipellis covered with simple to branched excrescences. Previously, nine Atheniella species were known globally, of which three species were accepted in China. Three newly-recognised species classified in the genus are here formally described from Yunnan Province: Atheniella flavida sp. nov., A. rutila sp. nov. and A. taoyao sp. nov. The new species are characterised by a yellow, orange, pink or red pileus, fusiform cheilocystidia and pleurocystidia, non-smooth pileipellis, stipitipellis smooth or with cylindrical ornamentation, caulocystidia fusiform or subglobose, if present and all tissues inamyloid. Morphological descriptions, photographs, line drawings and comparisons with closely-related taxa are presented for the new species. A phylogenetic analysis of sequence data for the rDNA internal transcribed spacer region and nuclear large ribosomal subunit (ITS + nLSU) supported that Atheniella is resolved as monophyletic and also supported the taxonomic recognition of the new species. The vibration and friction triggered by the long-term operation of stacker inevitably lead to the damage problem of stacker’s track. In order to ensure the stable operation of the stacker, a novel scheme is proposed to address an accurate damage localization problem of the stacker’s track in industrial environment based on ensemble empirical mode decomposition-empirical mode decomposition (EEMD-EMD) and density-based spatial clustering of applications with noise (DBSCAN). First, the original electric current signal with unbalanced distribution is processed by fixed-interval smoothing and cubic Hermite interpolation algorithms. Second, the EEMD-EMD method, which solves the mode-mixing problem, is adopted to decompose the preprocessed signal into well-defined intrinsic mode functions (IMFs). Third, based on Hilbert–Huang transform (HHT) of the IMFs, the preprocessed signal and the difference of average instantaneous amplitudes in two adjacent time points are considered as the 2-D feature vector input of the DBSCAN algorithm. As such, damage locations of the stacker’s track are detected by means of the outliers that are obtained by the DBSCAN algorithm. The efficiency of the proposed localization scheme and its superiority over the existing cooperative damage localization methods, namely, the box-plot method, the pulse coupling neural network (PCNN) and wavelet transform (WT) theory method, and the box-plot and WT method, are verified through experiments using the data provided by State Grid Measuring Center of China. Genetic variation has an important role in the development of non-small-cell lung cancer (NSCLC). However, genetic factors for lung cancer have not been fully identified, especially in Chinese populations, which limits the use of existing polygenic risk scores (PRS) to identify subpopulations at high risk of lung cancer for prevention. We therefore aimed to identify novel loci associated with NSCLC risk, and generate a PRS and evaluate its utility and effectiveness in the prediction of lung cancer risk in Chinese populations. To systematically identify genetic variants for NSCLC risk, we newly genotyped 19 546 samples from Chinese NSCLC cases and controls from the Nanjing Medical University Global Screening Array Project and did a meta-analysis of genome-wide association studies (GWASs) of 27 120 individuals with NSCLC and 27 355 without NSCLC (13 327 cases and 13 328 controls of Chinese descent as well as 13 793 cases and 14 027 controls of European descent). We then built a PRS for Chinese populations from all reported single-nucleotide polymorphisms that have been reported to be associated with lung cancer risk at genome-wide significance level. We evaluated the utility and effectiveness of the generated PRS in predicting subpopulations at high-risk of lung cancer in an independent prospective cohort of 95 408 individuals from the China Kadoorie Biobank (CKB) with more than 10 years' follow-up. We identified 19 susceptibility loci to be significantly associated with NSCLC risk at p≤5·0 × 10−8, including six novel loci. When applied to the CKB cohort, the PRS of the risk loci successfully predicted lung cancer incident cases in a dose-response manner in participants at a high genetic risk (top 10%) than those at a low genetic risk (bottom 10%; adjusted hazard ratio 1·96, 95% CI 1·53–2·51; p trend =2·02 × 10−9). Specially, we observed consistently separated curves of lung cancer events in individuals at low, intermediate, and high genetic risk, respectively, and PRS was an independent effective risk stratification indicator beyond age and smoking pack-years. We have shown for the first time that GWAS-derived PRS can be effectively used in discriminating subpopulations at high risk of lung cancer, who might benefit from a practically feasible PRS-based lung cancer screening programme for precision prevention in Chinese populations. Phylogenetic reconstruction revealed that Mycena stirps Amparoina, which is traditionally classified in sect. Sacchariferae, should be treated at section level. Section Amparoina is characterised by the presence or absence of cherocytes, the presence of acanthocysts and spinulose caulocystidia. Eight species referred to Mycena sect. Amparoina sect. nov. are recognised in China. Of these taxa, four new species classified in the new section are formally described: M. bicystidiata sp. nov., M. griseotincta sp. nov., M. hygrophoroides sp. nov. and M. miscanthi sp. nov. The new species are characterised by the absence of both cherocytes and a basal disc, along with the presence of acanthocysts on the pileus, spinulose cheilocystidia and caulocystidia. Background Epidermal growth factor receptor-tyrosine kinase inhibitors ( EGFR-TKIs) effectively treat advanced non-small cell lung cancer with EGFR-mutation. However, most patients develop acquired resistance without effective therapy subsequent to EGFR-TKI failure. We evaluated the efficacy of subsequent treatment strategies for EGFR-TKI resistance. Methods We retrospectively analyzed 240 patients with advanced lung adenocarcinoma with EGFR-TKI failure and following subsequent treatment. According to the first subsequent strategies after EGFR- TKI failure, patients were divided into groups of EGFR-TKI continuation (21 cases), EGFR-TKI continuation with chemotherapy (23 cases), chemotherapy alone (143 cases), and best supportive care ( BSC) (53 cases). Results Except for 53 cases of BSC, the disease control rates ( DCR) of the remaining 187 patients in the EGFR-TKI continuation, EGFR-TKI continuation with chemotherapy, and chemotherapy alone groups were 66.7%, 73.9%, and 44.8%, respectively. The median post-progression progression-free survival ( PFS) for the three groups was 3.0, 3.3, and 2.0 months, respectively. The DCR for the EGFR-TKI continuation with chemotherapy group was significantly higher than the chemotherapy alone group ( P = 0.006). The post-progression PFS of the EGFR-TKI continuation with chemotherapy group was significantly longer than the chemotherapy alone group ( P = 0.037). The median overall survival in the EGFR-TKI continuation, EGFR-TKI continuation with chemotherapy, chemotherapy alone, and BSC groups were 6.9, 11.6, 8.8, and 0.9 months, respectively. Compared to the BSC group, all groups achieved a survival benefit ( P < 0.001). Background To evaluate the efficacy and safety of Icotinib - an orally administered, highly potent selective inhibitor of epidermal growth factor receptor ( EGFR) and its active mutations, in the treatment of patients with advanced non-small cell lung cancer ( NSCLC). Methods A total of 101 patients with stage IIIb/ IV NSCLC were treated with 125 mg Icotinib three times a day until disease progression or intolerable toxicity. Response rate was evaluated using response evaluation criteria in solid tumors and progression-free survival ( PFS) was collected. Results The overall response rate ( ORR) and disease control rate ( DCR) were 37.6% (38/101) and 79.2% (80/101), respectively. The median PFS was 6.5 months. Multivariate analysis showed that female gender ( P = 0.048, 95% confidence interval [ CI] 1.010-6.016) and occurrence of rash ( P = 0.002, 95% CI 1.667-9.809) were the independent predictive factors for ORR, while a performance status ( PS) score of 0-1 ( P = 0.001, 95% CI 0.024-0.402) and rash ( P = 0.042, 95% CI 1.089-76.557) were the independent predictive factors for DCR. In addition, PS scores of 0-1 ( P < 0.001, 95% CI 0.135-0.509), and non-smoking ( P = 0.017, 95% CI 0.342-0.900) were found to be independent influencing factors for PFS. Moreover, patients with EGFR mutations had better PFS than patients with wild type EGFR, while patients with EGFR exon 19 deletion had better survival than those with EGFR exon 21 mutation. The most common adverse effects of Icotinib were rash (35.6%) and diarrhea (17.8%), which was tolerable. • The addition of THAM increased the hydrophilicity and electronegativity of the composite membrane surface. • TPAPD/PES composite membrane has excellent flux and phosphate rejection rate. • High performance of phosphate removal was observed in the real actual wastewater. In this study, polyaniline-p-phenylenediamine (PAPD) was employed as an amino monomer to fabricate a polyaniline-p-phenylenediamine composite membrane (PAPD/PES) with a nanoparticle-like surface morphology. Additionally, we used interfacial polymerization of Tris(hydroxymethyl)aminomethane (THAM) to hydroxylate the PAPD/PES composite membrane, resulting in the formation of a TPAPD/PES composite membrane. THAM is an economical and environmentally friendly monomer, rarely used for surface modification of composite membranes through interfacial polymerization. Following the introduction of THAM, a significant abundance of hydroxyl groups appeared on the membrane's surface, enhancing its hydrophilicity and electronegativity. This, in turn, improved both flux and rejection capabilities. Simultaneously, the introduction of THAM led to the formation of a hydration layer growing on the membrane's surface, further enhancing its antifouling performance. The optimal polymerization concentration (0.5 mol/L) of THAM was used to synthesize the TPAPD/PES composite membrane, resulting in a flux of 84.18 L m−2 h−1 bar−1, which was 1.46 times higher than that of the PAPD/PES composite membrane before polymerization. Additionally, the phosphate rejection rate was determined to be 92.14 % (with an initial phosphate concentration of 10 mg P/L). Long-time running and operating pressure tests results confirmed the remarkable stability of the TPAPD/PES composite membrane. Furthermore, the phosphate separation property of the TPAPD/PES composite membrane was evaluated using real wastewater samples (with an initial phosphate concentration of 1.03 mg P/L) as the research object. After separation, the permeate displayed a phosphate concentration of 0.28 mg P/L, meeting the first-class A standard for urban sewage treatment in China (0.5 mg P/L). Carbon emissions and water consumption are both important factors affecting sustainable development. Therefore, it is necessary to put them in the same research framework and investigate the synergy. In this study, the dynamic evolution characteristics of the synergistic effect of reducing carbon and saving water (RCSW) were analyzed. Then, taking the Yangtze River Delta Urban Agglomerations (YRDUA) as the research object, the influencing factors and specific paths of the synergistic effect were clarified. The results showed that the low-carbon emission efficiency (LCEE) had a stable synergy with the intensive utilization efficiency of water resources (IUEWR) in the YRDUA. Government financial expenditure, actual use of foreign capital, and population density were the most significant driving forces for the synergistic effect of RCSW, with q values of 0.561, 0.363, and 0.240, respectively. Notoginseng leaf triterpenes (PNGL), derived from the dried stems and leaves of P. notoginseng, is a phytoestrogen that exerts many neuroprotective effects in vivo and in vitro of ischemic stroke. However, its impact on neurological restoration specifically in relation to angiogenesis following ischemic stroke remains unexplored. The aim of this study was to assess the effects of PNGL on angiogenesis subsequent to ischemic stroke. Male Sprague-Dawley rats were utilized in this study and were subjected to middle cerebral artery occlusion/reperfusion (MCAO/R). Post-ischemia, PNGL were administered through intraperitoneal (i.p.) injection. The high-performance liquid chromatography (HPLC) fingerprinting, triphenyltetrazolium chloride (TTC) staining, immunofluorescent staining, network pharmacology and western blot analyses were assessed to determine the therapeutical effect and molecular mechanisms of PNGL on cerebral ischemia/reperfusion injury. Our findings demonstrate that PNGL effectively reduced infarct volume, enhanced cerebral blood flow, and induced angiogenesis in rats subjected to MCAO/R. Notably, PNGL also facilitated neuronal proliferation and migration in HUMECs in vitro. The proangiogenic effects of PNGL were found to be linked to the activation of the nuclear factor erythroid 2-related factor 2 (Nrf2) pathway and the AMPK/SIRT1-mediated PGC-1/ERα axis, as well as the activation of neurological function. Our study provides evidence that PNGL hold promise as an active ingredient of inducing proangiogenic effects, potentially through the activation of the Nrf2 pathway and the AMPK/SIRT1-mediated PGC-1/ERα axis. We present the largest whole-genome sequencing (WGS) study of non-small cell lung cancer (NSCLC) to date among 6,004 individuals of Chinese ancestry, coupled with 23,049 individuals genotyped by SNP array. We construct a high-quality haplotype reference panel for imputation and identify 20 common and low-frequency loci (minor allele frequency [MAF] ≥ 0.5%), including five loci that have never been reported before. For rare loss-of-function (LoF) variants (MAF < 0.5%), we identify BRCA2 and 18 other cancer predisposition genes that affect 5.29% of individuals with NSCLC, and 98.91% (181 of 183) of LoF variants have not been linked previously to NSCLC risk. Promoter variants of BRCA2 also have a substantial effect on NSCLC risk, and their prevalence is comparable with BRCA2 LoF variants. The associations are validated in an independent case-control study including 4,410 individuals and a prospective cohort study including 23,826 individuals. Recent studies have found multiple single nucleotide variants (SNVs) associated with DNA damage. However, previous association analysis may ignore the potential interaction effects between SNVs. Therefore, we used an improved random forest (RF) analysis to identify the SNVs related to personal DNA damage in exon-focused genome-wide association study (GWAS). A total of 301 subjects from three independent centers (Zhuhai, Wuhan, and Tianjin) were retained for analysis. An improved RF procedure was used to systematically screen key SNVs associated with DNA damage. Furthermore, we used genetic risk score (GRS) and mediation analysis to investigate the integrative effect and potential mechanism of these genetic variants on DNA damage. Besides, gene set enrichment analysis was conducted to identify the pathways enriched by key SNVs using the Data-driven Expression Prioritized Integration for Complex Traits (DEPICT). Finally, a set of 24 SNVs with the lowest mean square errors (MSE) were identified by improved RF analysis. Both weighted and unweighted GRSs were associated with increased DNA damage levels (P weight  < 0.001 and P unweight  < 0.001). Gene set enrichment analysis indicated that these loci were significantly enriched in several biological features associated with DNA damage. These findings suggested the role of SNVs in modifying DNA damage levels. Chinese lung cancer patients have distinct epidemiologic and genomic features, highlighting the presence of specific etiologic mechanisms other than smoking. Here, we present a comprehensive genomic landscape of 149 non-small cell lung cancer (NSCLC) cases and identify 15 potential driver genes. We reveal that Chinese patients are specially characterized by not only highly clustered EGFR mutations but a mutational signature (MS3, 33.7%), that is associated with inflammatory tumor-infiltrating B lymphocytes (P = 0.001). The EGFR mutation rate is significantly increased with the proportion of the MS3 signature (P = 9.37 × 10 -5 ). TCGA data confirm that the infiltrating B lymphocyte abundance is significantly higher in the EGFR-mutated patients (P = 0.007). Additionally, MS3-high patients carry a higher contribution of distant chromosomal rearrangements >1 Mb (P = 1.35 × 10 -7 ), some of which result in fusions involving genes with important functions (i.e., ALK and RET). Thus, inflammatory infiltration may contribute to the accumulation of EGFR mutations, especially in never-smokers.

FOSL1 (FOS like antigen 1) is one kind of proto-oncogene, and may play a vital role in carcinogenesis of multiple cancers. However, studies about the relationship between SNPs in FOSL1 and gastric cancer are still lacking. Thus, we investigated the association of seven SNPs in FOSL1 with gastric cancer using case-control design in a two-stage strategy (Screening stage: 1,140 gastric cancer cases and 1,547 controls; Replication stage: 1,006 cases and 2,273 controls). We found that rs1892901 was significantly associated with increased risk of gastric cancer in additive model (adjusted OR = 1.25, 95%CI: 1.06-1.47, P = 0.008) in first stage. Following replication results revealed that the relationship between rs1892901 and gastric cancer risk was consistent with our primary results. In silico analysis showed that rs1892901 might alter multiple regulatory motifs, disturb protein binding, and affect the expression of FOSL1 and other important gastric cancer-related genes such as EGR1, CHD, EP300, FOS, JUN and FOSL2. Our findings indicated that functional SNP rs1892901 in FOSL1 might affect the expression of FOSL1, and ultimately increase the risk of gastric cancer. Background: Alternative splicing is an important biological step during mRNA processing. Misregulation of alternative splicing can produce aberrant protein isoforms, thus contributing to cancer. We hypothesized that variants in 5 critical splicing factor-associated genes might play an important role in carcinogenesis of lung cancer., Materials and Methods: A case-control study including 1,341 non-small cell lung cancer (NSCLC) cases and 1,982 cancer-free controls were conducted to evaluate the associations of 16 tagging/functional polymorphisms in 5 splicing factor-associated genes with lung cancer risk., Results: We found altogether 8 SNPs were associated with lung cancer risk with adjustment of age, gender, and smoking status after multiple corrections (FDR). Among these, six SNPs were related with SRSF7(rs10197412, OR (95% CI)=1.23 (1.06-1.43), P for FDR=0.018; rs12621103, OR(95% CI)=1.25 (1.08-1.46), P for FDR=0.016; rs13024811, OR (95% CI)=1.25 (1.07-1.46), P for FDR=0.016; rs2037875, OR (95% CI)=1.23 (1.06-1.42), P for FDR=0.018; rs3134628, OR (95% CI)=1.25(1.07-1.45), P for FDR=0.016 and rs6715866, OR (95% CI)=1.23 (1.07-1.43), P for FDR=0.016); one SNP was near PTBP2 (rs12566237: OR (95% CI)=1.16 (1.05-1.28), P for FDR=0.016) and one SNP in HNRNPQ (rs16876385: OR (95% CI)=1.17 (1.04-1.32), P for FDR=0.022)., Conclusions: Our findings indicated that genetic variants in these splicing-associated genes might modify individual susceptibility to lung cancer in Chinese population.
