The Xinjiang Uygur Autonomous Region (Xinjiang) borders eight countries and has a complex geographic environment. There are almost 45.696 million herded sheep in Xinjiang, which occupies 13.80% of China's sheep farming industry. However, there is a scarcity of reports investigating the role of sheep or ticks in Xinjiang in transmitting tick-borne diseases (TBDs). A total of 894 ticks (298 tick pools) were collected from sheep in southern Xinjiang. Out of the 298 tick pools investigated in this study, Rhipicephalus turanicus (Rh. turanicus) and Hyalomma anatolicum (H. anatolicum) were identified through morphological and molecular sequencing. In the southern part of Xinjiang, 142 (47.65%), 86 (28.86%), and 60 (20.13%) tick pools were positive for Rickettsia spp., Theileria spp., and Anaplasma spp., respectively. Interestingly, the infection rate of Rickettsia spp. (73%, 35.10%, and 28.56–41.64%) was higher in Rh. turanicus pools than in H. anatolicum pools (4%, 4.44%, and 0.10–8.79%) in this study. Fifty-one tick pools were found to harbor two pathogens, while nineteen tick pools were detected to have the three pathogens. Our findings indicate the presence of Rickettsia spp., Theileria spp., and Anaplasma spp. potentially transmitted by H. anatolicum and Rh. turanicus in sheep in southern Xinjiang, China. Artemisiae argyi Folium is a traditional herbal medicine used for moxibustion heat therapy in China. The volatile oils in A.argyi leaves are closely related to its medicinal value. Records suggest that the levels of these terpenoids components within the leaves vary as a function of harvest time, with June being the optimal time for A. argyi harvesting, owing to the high levels of active ingredients during this month. However, the molecular mechanisms governing terpenoid biosynthesis and the time-dependent changes in this activity remain unclear. In this study, GC–MS analysis revealed that volatile oil levels varied across four different harvest months (April, May, June, and July) in A. argyi leaves, and the primarily terpenoids components (including both monoterpenes and sesquiterpenes) reached peak levels in early June. Through single-molecule real-time (SMRT) sequencing, corrected by Illumina RNA-sequencing (RNA-Seq), 44 full-length transcripts potentially involved in terpenoid biosynthesis were identified in this study. Differentially expressed genes (DEGs) exhibiting time-dependent expression patterns were divided into 12 coexpression clusters. Integrated chemical and transcriptomic analyses revealed distinct time-specific transcriptomic patterns associated with terpenoid biosynthesis. Subsequent hierarchical clustering and correlation analyses ultimately identified six transcripts that were closely linked to the production of these two types of terpenoid within A. argyi leaves, revealing that the structural diversity of terpenoid is related to the generation of the diverse terpene skeletons by prenyltransferase (TPS) family of enzymes. These findings can guide further studies of the molecular mechanisms underlying the quality of A. argyi leaves, aiding in the selection of optimal timing for harvests of A. argyi. Q fever, spotted fever rickettsioses and equine piroplasmosis, are some of the most serious equine tick-borne diseases caused by Coxiella burnetii , Rickettsia spp., Babesia caballi and/or Theileria equi. This study surveyed and molecularly characterized these pathogens infecting horses in ten ranches from XUAR, China using molecular technology. Among 200 horse blood samples, 163 (81.5%) were infected with at least one of the pathogens. Rickettsia spp. was the most prevalent pathogen (n = 114, 57.0%), followed by C. burnetii (n = 79, 39.5%), T. equi (n = 79, 39.5%) and B. caballi (n = 49, 24.5%). Co-infections were observed in 61.3% of positive samples in this study. Statistically significant differences were observed between the sampling regions for C. burnetii , B. caballi and T. equi , and also in different age group for C. burnetii and T. equi. The genotype analysis indicated that C. burnetii htpB , Rickettsia spp. ompA, B. caballi rap-1 , B. caballi 18S rRNA, T. equi EMA-1 and T. equi 18S rRNA gene sequences from horses in XUAR were variable. To the best of our knowledge, this study is the first report of C. burnetii and Rickettsia spp. infection and co-infected with piroplasma in horses in China. Overall, this study revealed the high infection rate of the pathogens in horses in XUAR, China. The current findings are expected to provide a basis for better tick-borne disease control in the region. The Qinghai-Tibetan Plateau Area (QTPA) is a plateau with the highest average altitude, located in Northwestern China. There is a risk for interspecies disease transmission, such as spotted fever rickettsioses. However, information on the molecular characteristics of the spotted fever group (SFG) Rickettsia spp. in the area is limited. This study performed screenings, and detected the DNA of human pathogen, SFG Rickettsia spp., with 11.3% (25/222) infection rates in yaks (Bosgrunniens). BLASTn analysis revealed that the Rickettsia sequences obtained shared 94.3–100% identity with isolates of Rickettsia spp. from ticks in China. One Rickettsia sequence (MN536161) had 100% nucleotide identity to two R. raoultii isolates from Chinese Homo sapiens, and one isolate from Qinghai Dermacentorsilvarum. Meanwhile, another Rickettsia sequence (MN536157) shared 99.1–99.5% identity to one isolate from Dermacentor spp. in China. Furthermore, the phylogenetic analysis of SFG Rickettsia spp. ompA gene revealed that these two sequences obtained from yaks in the present study grouped with the R. slovaca and R. raoultii clades with isolates identified from Dermacentor spp. and Homo sapiens. Tick-borne diseases cause significant losses to livestock production in tropical and subtropical regions. However, information about the tick-borne infections in cattle in Xinjiang Uygur Autonomous Region (XUAR), northwestern China, is scarce. In this study, nested polymerase chain reaction (PCR) assays and gene sequencing were used to detect and analyze epidemiological features of Babesia bovis, B. bigemina, Coxiella burnetii and Anaplasma bovis infections in XUAR. Out of 195 samples tested, 24 (12.3%), 67 (34.4%), 40 (20.5%) and 10 (5.1%) were positive for B. bovis, B. bigemina, C. burnetii and A. bovis , respectively. Sequencing analysis indicated that B. bovis SBP-4, B. bigemina Rap1a, C. burnetii htpB and A. bovis 16S rRNA genes from XUAR showed 99%–100% identity with documented isolates from other countries. Phylogenetic analyses revealed that B. bovis SBP-4, B. bigemina Rap1a, C. burnetii htpB and A. bovis 16S rRNA gene sequences clustered in the same clade with isolates from other countries. To the best of our knowledge, this is the first report of C. burnetii infection of cattle in XUAR. Furthermore, this study provides important data for understanding the distribution of tick-borne pathogens, and is expected to improve the approach for prevention and control of tick-borne diseases in China. • A. ovis , T. luwenshuni , T. uilenbergi and B. motasi sp. L/N/T were detected infecting sheep from Qinghai, China. • B. motasi sp. L/N/T, T. luwenshuni and T. uilenbergi were analyzed in sheep in Qinghai for the first time. • Mixed infection with the four tick-borne pathogens was firstly identified in sheep in Qinghai, China. Information regarding the prevalence and genetic characterization of Anaplasma spp., Babesia spp. and Theileria spp. infecting sheep in Qinghai province, northwestern China, is limited. In this study, 277 sheep blood samples were collected from 7 counties in Qinghai and tested by PCR to detect and identify the above mentioned tick-borne pathogens. Among the 277 sheep, 176 (63.5%) were infected with at least one pathogen. A. ovis was the most prevalent pathogen (n = 112, 40.4%), followed by T. luwenshuni (n = 108, 39.0%), T. uilenbergi (n = 65, 23.5%) and B. motasi sp. L/N/T (n = 11, 4.0%). Interestingly and importantly, 83 (30.0%) infections with two or more pathogens were identified. The analysis of the nucleotide sequences of targeted genes showed that A. ovis ms p4, B. motasi sp. L/N/T rap-1b , T. luwenshuni 18S rRNA and T. uilenbergi 18S rRNA sequences from Qinghai were not conserved. These sequences shared 97.7–99.7%, 87.0–99.8%, 98.2–100% and 91.3–100% identities with previously published isolates, respectively. The phylogenetic analysis revealed that all A. ovis ms p4 sequences belonged to the same clade. On the other hand, B. motasi sp. L/N/T rap-1b isolates showed a closer relationship with B. motasi sp. BQ1/Lintan. One T. luwenshuni and one T. uilenbergi sequence from this study formed branches divergent from the remaining T. luwenshuni and T. uilenbergi isolates. Overall, these results provide a better understanding of the prevalence and molecular characterization of TBPs in sheep in Qinghai and suggest that these pathogens require more attention from veterinarians. • Dermacentor marginatus glutathione s-transferase mu 1 (DmGstm1) was cloned and in silico analysis was performed. • Gene expression level of DmGstm1 was measured. • The recombinant GST was efficiently expressed through E. coli. • It's the first tick vaccine trial against Dermacantor marginatus using recombinant GST in northwestern China. Dermacentor marginatus is one of the main tick species in northwestern China, and is a vector of various tick-borne pathogens. Tick control method largely depends on chemical agents, but the disadvantages of using such approach would cause environmental damage and the risk of developing tick resistance to acaricides. Vaccination of tick protective antigen is an eco-friendly approach which is an alternative and promising method to mitigate tick infestation in livestock. In the study, a mu-class glutathione S-transferase (GST) sequence of D. marginatus was cloned and the recombinant protein (rDmGST) was expressed. Transcriptional level of the GST was measured together with native GST activity of the tick. Finally, A vaccine trial on rabbits against D. marginatus was proceeded to evaluate the anti-tick effect of rDmGST. Results reveled that the CDs of the D. margiantus glutathione S-transferase mu 1 gene has 669 base pair nucleotide sequence encoding a 223 amino acid. The deduced GST protein sequence had over 95 % similarity with that of D. variabilis. The rDmGST was efficiently expressed soluble and purified by His trap affinity chromatography. Enzyme activity of native GST and transcriptional profiles of the GST showed up-regulation in different stages and organs of D. marginaus during blood feeding. Polyclonal antibody reacted with rDmGST in Western blotting. Tick challenge on rDmGST inoculated rabbits showed reductions in adult female engorgement rate, total egg mass and egg hatching rate with an overall vaccine efficacy of 43.69 %. Hyalomma tick species are considered the competent vector tick species that carry and transmit Crimean-Congo hemorrhagic fever virus (CCHFV) to humans and animals. Hyalomma asiaticum is one of the major tick species widespread in the Xinjiang Uygur Autonomous Region (XUAR) of China. To determine the potential prevalence of H. asiaticum in XUAR, species distribution modeling was performed using MaxEnt algorithm assembled with bioclimatic variables and curated tick presence records. The results indicate that potential habitats of H. asiaticum mainly cover the northern and western XUAR. The suitable habitats included the west rim of the Taklimakan Desert, Turpan Basin, and Junggar Basin. The models show a mean area under the curve of 0.865 ± 0.068 for H. asiaticum based on 10-fold cross-validation. Despite limited tick presence records used in the study, this work describes the potential distribution and the association of important bioclimatic variables that are crucial for the survival of H. asiaticum in many arid areas in XUAR. Ticks carry and transmit a wide range of pathogens (bacteria, viruses, and protozoa) that are of importance to humans and animals globally. However, information about the tick-borne pathogens harbored by ticks in the Xinjiang Uygur Autonomous Region (XUAR), northwestern China, is scarce. This study investigated the occurrence of tick species of domestic animals and tick-borne pathogens by using morphological molecular identification and sequence analysis in Turpan, Qitai, Altay, Hejing, Nileke, and Zhaosu counties (XUAR). A total of 5822 adult ticks (females and males) from 12 tick species were identified from 5 animal species (cattle, goats, sheep, camels, and horses) in 6 counties in the XUAR. Collected tick species included Dermacentor marginatus (24.7 %), Dermacentor nuttalli (20.8 %), Hyalomma anatolicum (13.7 %), Dermacentor niveus (13.1 %), Haemaphysalis punctata (10.7 %), Dermacentor silvarum (7.1 %), Dermacentor pavlovskyi (3.9 %), Hyalomma asiaticum (2.2 %), Rhipicephalus pumilio (1.9 %), Rhipicephalus sanguineus sensu lato (0.7 %), Rhipicephalus turanicus (0.6 %), and Hyalomma asiaticum kozlovi (0.6 %). Furthermore, 750 partially engorged adult ticks (females and males), including H. anatolicum (250), D. nuttalli (250), and D. marginatus (250), were individually separated according to species and sampling site, used for DNA extraction, and then screened for tick-borne pathogens. The most common pathogen was Rickettsia raoultii (36.80 %), followed by Brucella sp. (26.2 %), Anaplasma ovis (22.4 %), Babesia caballi (14.8 %), Theileria equi (8.7 %), and Theileria ovis (8.5 %). The sequencing of 6 genes showed a 96-100 % nucleotide identity between the sequences in this study and those deposited in GenBank.
