The uncertainty and unknowability of emerging infectious diseases have caused many major public health and security incidents in recent years. As a new tick‐borne disease, Dabieshan tick virus (DBTV) necessitate systematic epidemiological and spatial distribution analysis. In this study, tick samples from Liaoning Province were collected and used to evaluate distribution of DBTV in ticks. Outbreak points of DBTV and the records of the vector Haemaphysalis longicornis in China were collected and used to establish a prediction model using niche model combined with environmental factors. We found that H. longicornis and DBTV were widely distributed in Liaoning Province. The risk analysis results showed that the DBTV in the eastern provinces of China has a high risk, and the risk is greatly influenced by elevation, land cover, and meteorological factors. The risk geographical area predicted by the model is significantly larger than the detected positive areas, indicating that the etiological survey is seriously insufficient. This study provided molecular and important epidemiological evidence for etiological ecology of DBTV. Metschnikowia bicuspidata is a globally distributed pathogenic yeast with a wide range of aquatic hosts. A new strain, M. bicuspidata LNES0119, isolated from the Chinese mitten crab Eriocheir sinensis , has caused a serious reduction in production and marked economic loss for the aquaculture industry in China. Therefore, the whole-genome sequence of M. bicuspidata LNES0119 was sequenced using Illumina and Oxford Nanopore technology; whole-genome annotation and comparative genomic analyses of this pathogen were performed as well. A high-quality genome of M. bicuspidata LNES0119 was 16.13 Mb in size, with six scaffolds and six contigs, and encoded 5,567 putative predicted genes. Of these, 1,467 genes shared substantial homology with genes in the pathogen–host interactions database. Comparative genomic analyses of three M. bicuspidata strains and one non-pathogenic yeast, M. aff. pulcherrima , showed 331 unique genes in M. bicuspidata LNES0119, 30 of which were putatively related to pathogenicity. Overall, we identified several meaningful characteristics related to pathogenicity and virulence that may play essential roles in the infection and pathogenicity of M. bicuspidata LNES0119. Our study will aid in identifying potential targets for further exploration of the molecular basis of the pathogenicity of M. bicuspidata as well as the therapeutic intervention of M. bicuspidata infection. Tachaea chinensis Thielemann, 1910 (Isopoda: Corallanidae) is a branchial ectoparasite that attaches onto shrimps and prawns. However, the distribution of T. chinensis in China, especially its epidemiology, remains unclear. We determined the prevalence of T. chinensis on the ridgetail white prawn (Exopalaemon carinicauda Holthuis, 1950) in Jiangsu Province. Fifty ponds in 10 shrimp farms were assessed. Isopod species were identified by morphological features and mitochondrial 16S rRNA gene analysis. A literature review was performed to determine the geographical distribution of T. chinensis in China. Published data revealed that T. chinensis was geographically distributed throughout five provinces in China, including Liaoning, Tianjin, Henan, Hubei, and Guangxi. A total of 998 T. chinensis were collected from 50 ridgetail white prawn ponds in Yancheng City and Rudong County. Tachaea chinensis prevalence ranged from 0.98% to 4.42% in Yancheng City and 0.62% to 0.92% in Rudong County. This is the first study to investigate the geographical distribution of T. chinensis in China and determine the prevalence of T. chinensis on the ridgetail white prawn in Jiangsu Province. Background: Schistosomiasis japonica remains a significant public health problem in China and Southeast Asian countries. The most typical and serious outcome of the chronic oriental schistosomiasis is the progressive granuloma and fibrosis in the host liver, which has been a major medical challenge. However, the molecular mechanism underling the hepatic pathogenesis is still not clear. Methodology and Principal Findings: Using microarrays, we quantified the temporal gene expression profiles in the liver of Schistosoma japonicum-infected BALB/c mice at 15, 30, and 45 day post infection (dpi) with that from uninfected mice as controls. Gene expression alternation associated with liver damage was observed in the initial phase of infection (dpi 15), which became more magnificent with the onset of egg-laying. Up-regulated genes were dominantly associated with inflammatory infiltration, whereas down-regulated genes primarily led to the hepatic functional disorders. Simultaneously, microRNA profiles from the same samples were decoded by Solexa sequencing. More than 130 miRNAs were differentially expressed in murine liver during S. japonicum infection. MiRNAs significantly dysregulated in the mid-phase of infection (dpi 30), such as mmu-miR-146b and mmu-miR-155, may relate to the regulation of hepatic inflammatory responses, whereas miRNAs exhibiting a peak expression in the late phase of infection (dpi 45), such as mmu-miR-223, mmu-miR-146a/b, mmu-miR-155, mmu-miR-34c, mmu-miR-199, and mmu-miR-134, may represent a molecular signature of the development of schistosomal hepatopathy. Further, a dynamic miRNA-gene co-expression network in the progression of infection was constructed. Conclusions and Significance: This study presents a global view of dynamic expression of both mRNA and miRNA transcripts in murine liver during S. japonicum infection, and highlights that miRNAs may play a variety of regulatory roles in balancing the immune responses during the development of hepatic pathology. IES was the first knowledge-based cost estimating system with distinctive Chinese features and has been developed over the past four years, increasing the speed of estimating twelvefold in Shandong Province. GIE, the system for the whole of China is being tested in Jiangsu Province and the authors anticipate that other provinces will adopt the method, thus raising estimating standards in China as a whole. L'IES, developpe au cours des quatre dernieres annees, constitue le premier systeme intelligent d'estimation des couts dote de caracteristiques propres a l'environnement chinois. Son utilisation, dans la province de Shandong, a permis d'accelerer de douze fois la rapidite d'estimation des couts de projets de construction. Les premiers essais du systeme GIE, destine a l'ensemble de la Chine, sont en cours dans la province de Jiangsu. Tachaea chinensis is a parasitic isopod that negatively affects the production of several commercially important shrimp species in China. To date, there have been no reports on the antioxidant and immune responses of host shrimps to isopod parasite infection or their underlying molecular mechanisms. In this study, we examined the specific activities of the immune and antioxidant enzymes of the shrimp Macrobrachium nipponense during the course of a 15-day isopod infection and evaluated expression of related genes. Acid phosphatase (ACP) and alkaline phosphatase (AKP) activities and malondialdehyde (MDA) levels showed significant peaks over 15 days of exposure in both the hepatopancreas and muscle (P < 0.05), whereas catalase (CAT) activity increased continuously during infection (P < 0.05), and lysozyme (LZM) activity increased only in the hepatopancreas (P < 0.05). After 6 days of exposure, expressions of glutathione S -transferase (GST), ACP, and AKP were significantly higher than at 12 days. Compared with the control group, at 12 days, S -(hydroxymethyl) glutathione dehydrogenase activity and glutathione metabolism pathways were significantly inhibited (P < 0.05). Furthermore, the NOD-like receptor signaling pathway and antigen processing and presentation pathways were also significantly inhibited at 12 days compared with that at 6 days (P < 0.05), indicating that T. chinensis parasitism could perturb the antioxidant and immune systems of shrimp hosts during the latter stages of infection. Additionally, the molting and mortality rates of M. nipponense increased the duration of parasitism. These findings indicate that M. nipponense can activate antioxidant and immune defense systems during the early period during isopod parasitism, whereas the parasite can negatively affect these host defense systems during the latter period. Abstract: Acquisition of Toxoplasma gondii infections is mainly through ingestion of parasite-contaminated food. T. gondii oocyst distribution in the living environment of human and livestock is directly linked to the prevalence of the parasite infection in humans and domestic animals. In this study, we investigated the sero-prevalence of T. gondii infection in free-range as well as caged chicken in northeast China. The purpose of this study was to investigate the potential prevalence of T. gondii oocysts in the environments. Sera of 308 free-range chickens and 210 caged chickens collected in three areas in northeast China were tested for anti-T. gondii antibodies with ELISA assays. The infection rates of free-range and caged chickens were 34.7% and 2.8% respectively, indicating that the parasite is widely distributed in the environment and poses threatens to the health of people living in those areas. The Chinese mitten crab, Eriocheir sinensis , is an economically important crustacean species in China. A recent epidemic of "milky disease" in Panjin, China resulted in a large loss of cultured E. sinensis. Clinical signs of the disease include weak response, swollen pereopod and claw joints, opaque or whitish muscles, and milky hemolymph, culminating in tissue or organ failure and death. Histological analysis of the crabs showed a marked disorder of fibers, discrete necrotic lesions, and high yeast infiltration in the muscles, heart, gills, and other organs. The yeast cells (LNES0119) isolated from the diseased crab tissues were subjected to polymerase chain reaction analysis. The resulting product, the D1/D2 domain of 26S rDNA of the LNES0119 yeast isolate, was used to identify the pathogen for milky disease, which was found to have 98% sequence identity with the sequence of Metschnikowia bicuspidata (GenBank accession nos. U44822 and U94944), a pathogenic yeast found in aquatic invertebrates. A laboratory challenge experiment showed that the pathogen was infectious and caused the same pathological alterations in crab hosts. The present findings demonstrated that the pathogen isolated from the Chinese mitten crab with milky disease was the yeast M. bicuspidata. In this study, we describe in detail the life cycle of Tachaea chinensis (Isopoda: Corallanidae), a branchial ectoparasitic isopod that infests the freshwater shrimp Palaemonetes sinensis in China. We obtained 14 ovigerous T. chinensis females (8.22-11.92 mm in length) and observed the development of embryos through 5 sequential ontogenetic stages within the brood pouches (marsupium) of these females. The number of eggs or mancae (post-larval juveniles) held in the female marsupium ranged from 31 to 86, with a mean ± SD of 61.25 ± 16.16 eggs. Female T. chinensis were semelparous, i.e. individuals died following the release of mancae from the marsupium. Released mancae were non-planktonic and immediately infective to host shrimps. However, only a few mancae successfully established contact with a host, and it is thus assumed that the remainder were predated by shrimp. Attached T. chinensis fed on the host hemolymph, and subsequent to host death, these isopods typically searched for a new host. We also found that T. chinensis exhibits a host preference: most mancae attached to P. sinensis rather than to Neocaridina sp. or Macrobrachium nipponense. This study provides valuable empirical data that will support future research on the prevention and control of parasitic isopod infections.

The "milky disease" of the Chinese mitten crab, Eriocheir sinensis , is a highly lethal fungal disease caused by Metschnikowia bicuspidata infection. To elucidate the immune responses of the hemolymph of E. sinensis to M. bicuspidata infection, a comparative analysis of the hemolymph of E. sinensis infected with M. bicuspidata and that treated with phosphate buffered saline was performed using label-free quantitative proteomics. A total of 429 proteins were identified. Using a 1.5-fold change in expression as a physiologically significant benchmark, 62 differentially expressed proteins were identified, of which 38 were significantly upregulated and 24 were significantly downregulated. The upregulated proteins mainly included cytoskeleton-related proteins (myosin regulatory light chain 2, myosin light chain alkali, tubulin α-2 chain, and tubulin β-1 chain), serine protease and serine protease inhibitor (clip domain-containing serine protease, leukocyte elastase inhibitor, serine protein inhibitor 42Dd), catalase, transferrin, and heat shock protein 70. Upregulation of these proteins indicated that phenoloxidase system, phagocytosis and the ROS systems were induced by M. bicuspidata . The downregulated proteins were mainly organ and tissue regeneration proteins (PDGF/VEGF-related factor protein, integrin-linked protein kinase homing pat-4 gene) and hemagglutination-associated proteins (hemolymph clottable protein, hemocyte protein-glutamine gamma-glutamyltransferase). Downregulation of these proteins indicated that M. bicuspidata inhibited hemocyte regeneration and hemolymph agglutination. Fifteen differentially expressed proteins related to immunity were verified using a parallel reaction monitoring method. The expression trend of these proteins was similar to that of the proteome. To the best of our knowledge, this is the first report on the proteome of E. sinensis in response to M. bicuspidata infection. Chinese grass shrimp ( Palaemonetes sinensis ) is an economically important crustacean in Chinese aquaculture. Recently, we found that shrimp in Panjin city were infected with microsporidia, a group of fungi. The hepatopancreas of several infected shrimp showed white turbidity and pathological changes that negatively affected the health and appearance of the shrimp. Histopathology and transmission electron microscopy were used to examine the development of the parasite within its parasitophorous vacuole. Our results indicated that microsporidia developed asynchronously within the same parasitophorous vacuole. The spores were predominantly small, and rod or oval-shaped. The sizes of fresh spores were approximately 3.1 × 2.4 μm and fixed spores were 1.9 × 1.1 μm. The polar filament was isofilar with 5-6 coils and the thickness was 103.2 nm. Merogonial divisions occurred by binary fission and sporogonial division occurred by plasmotomy. The small subunit ribosomal DNA sequence (1295 bp) from the parasite was highly similar to the previously reported parasite Enterocytospora artemiae (99% nucleotide identity, JX915760). Using maximum likelihood to analyze the phylogenetic relationships, we found that this microsporidian should be grouped within Clade IV, an Enterocytospora -like clade, of the Microsporidia phylum. Based on this parasite's life cycle characteristics, morphology, and small subunit ribosomal DNA sequence, the parasite described here is likely E. artemiae , which has previously only been described in Europe and North America. As a unique feature among otherwise hermaphroditic trematodes, Schistosoma species are gonochoric parasites whose sex is genetically determined (ZZ for males and ZW for females). However, schistosome larvae are morphologically identical, and sex can only be discriminated by molecular methods. Here, we integrated published Schistosoma. japonicum transcriptome and genome data to identify W chromosome-specific genes as sex biomarkers. Three W chromosome-specific genes of S. japonicum were identified as sex biomarkers from a panel of 12 genes expressed only in females. An efficient duplex real-time PCR (qPCR) method for sexing cercariae was developed which could identify the sex of cercariae within 2 h without DNA extraction. Moreover, this method can be used to identify not only single-sex but also mixed-sex schistosome-infected snails. We observed a nearly equal proportion of single-male, single-female, and mixed-sex schistosome infections in artificially infected snails. Sex-known schistosome-infected snail models can be efficiently constructed with the aid of duplex qPCR. A field study revealed that single-sex schistosome infections were predominant among naturally infected snails. Finally, a schistosomiasis mouse model based on sex-known cercariae infection was shown to be more reliable than a model based on sex-unknown cercariae infection. Tachaea chinensis, a parasitic isopod, negatively affects the production of several commercially important shrimp species in China. The mechanism of parasite-host interaction cannot be accurately described by transcriptomic and proteomic approaches individually. Here, comparative metabolite profiling was used to achieve a broad coverage of primary metabolite changes in Chinese grass shrimp Palaemonetes sinensis following T. chinensis parasitization. In total, 66 metabolites were significantly differentially accumulated between the control and infected groups; of these, 19 were upregulated and 47 were downregulated after T. chinensis infection. Moreover, the Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis revealed that 10 pathways were significantly enriched. The protein digestion and absorption pathways were highly enriched, followed by the mineral absorption, aminoacyl-tRNA biosynthesis, biosynthesis of amino acids, and metabolic metabolism pathways. Parasitization by T. chinensis enhanced the glycolytic pathway and tricarboxylic acid (TCA) cycle in P. sinensis, thereby releasing more energy for swimming, foraging, and evading predation. Glucogenic amino acids such as alanine, histidine, glutamine, and proline were consumed to generate glutamate and enhance the TCA cycle. Nucleotide-related metabolic pathways were downregulated, possibly because T. chinensis can secrete molecules to degrade nucleotides and inhibit hemostasis and inflammatory responses. These results suggest that the isopod parasite can increase the host's metabolic burden by enhancing the host's TCA cycle and secreting molecules to degrade host proteins, thereby enabling the parasite to feed on the host and inhibit an inflammatory response. The results will be a valuable contribution to understanding the metabolic responses of crustaceans to isopod parasitism.

Giardia duodenalis is an important zoonotic intestinal parasite responsible for diarrhea in humans and other animals worldwide. The present study was conducted to assess the prevalence of bovine giardiosis and to perform molecular characterization of Giardia duodenalis in the northeast of China. A total of 655 fecal specimens were collected from dairy cattle in 15 farms located in three different provinces. G. duodenalis assemblages and subtypes were determined by sequence analysis of the triosephosphate isomerase (TPI) gene. As a whole, the G. duodenalis infection rate in dairy cattle was 7.9% (52/655), as determined by Lugol's iodine staining. Two assemblages were identified, namely, the potentially zoonotic assemblage A (n = 1), the livestock-specific assemblage E (n = 50), and a mixed infection case of assemblages A and E. Seven distinct subtypes of E assemblages were identified and E-XI, E-I and E-III are the major subtypes. Only subtype A-I was identified in assemblage A. Findings relevant to assemblage A are of public health importance. The results indicated the livestock-specific assemblage E is the major genotype and zoonotic assemblage A or B occurs very seldomly which is significantly different with previous report in the same area. Background: Toxoplasmosis is one of the most common parasitic zoonoses. The seroprevalence of Toxoplasma gondii infection in humans varies widely worldwide. Detection of Toxoplasma-specific antibodies has been a gold standard method for both epidemiological investigation and clinical diagnosis. Genetic investigation indicated that there is a wide distribution of different genome types or variants of the parasite prevalent in different areas. Thus the reliability of using antigens from parasites of a single genome type for diagnosis and epidemiology purposes needs to be extensively evaluated., Methods: In this study, the prevalence of T. gondii infection among 880 clinically healthy individuals in China was systematically tested using crude soluble native antigens and purified recombinant antigens of type I and II T. gondii. The T. gondii-specific IgG and IgM in the sera was further confirmed using commercial Toxoplasmosis Diagnosis Kits and Western blot assays., Results: The sero-prevalence of T. gondii-specific IgG detected with crude native Type I and type II antigens was 12.2% and 11.3% respectively. Whereas the overall prevalence was more than 20% when combined with the results obtained with recombinant tachyzoite and bradyzoite antigens. There was an obvious variation in immune-recognition of parasite antigens among the individuals studied., Conclusions: The general prevalence of anti-T. gondii IgG in the study population was likely much higher than previously reported. The data also suggested that there is more genetic diversity among the T. gondii isolates in China. Further, combination of recombinant antigens with clear immuno-recognition will be able to generate more sensitive diagnostic results than those obtained with crude antigens of T. gondii tachyzoites.

Background: Upper gastrointestinal (UGI) symptoms are common in hemodialysis (HD) patients, while gastric metaplasia (GM) and Helicobacter pylori infections are key causes for UGI symptoms. This study is targeted to compare GM and H. pylori infections in patients with different durations of HD., Methods: A total of 406 subjects from Ningbo Urology and Nephrology Hospital were included. The mean age of subjects was 44.7 ± 13.5 years; 62.9% were male; and subjects were divided into four groups according to different HD durations. Upper endoscopy and lesion were performed in these patients and methylene blue staining was used in detecting H. pylori and GM., Results: Erosive gastritis was the most common symptom in uremic subjects. GM was found in 139 patients. The longer the dialysis duration, the higher the incidence rate of GM (p < 0.05). H. pylori infection accounted for 24.1% in HD patients. The occurrence of H. pylori infection decreased as dialysis periods progressed within the first 4-year follow-up after the start of HD., Conclusions: Almost all patients with HD experienced gastrointestinal discomfort in the current patient cohort. The most common mucosal lesion observed in our study pool was chronic erosive gastritis. The overall incidence of GM was normal at 35.0%, since quite a part of patients are the elderly group in this study. We need not worry about this too much, unless the HD patients have registered for renal transplantation or are suffering from severe gastrointestinal discomfort.

In this study, the prevalence of Enterocytozoon bieneusi in China was investigated. Twelve genotypes of E. bieneusi were identified, of which 10 were novel genotypes. Further, 41.6% of the genotypes were found in both humans and animals. This is the first report of E. bieneusi in China.

Background: Toxoplasmosis is an important zoonotic parasitic disease worldwide. In immune competent individuals, Toxoplasma gondii preferentially infects tissues of central nervous systems, which might be an adding factor of certain psychiatric disorders. Congenital transmission of T. gondii during pregnancy has been regarded as a risk factor for the health of newborn infants. While in immune-compromised individuals, the parasite can cause life-threatening infections. This study aims to investigate the prevalence of T. gondii infection among clinically healthy individuals and patients with psychiatric disorders in China and to identify the potential risk factors related to the vulnerability of infection in the population., Methods: Serum samples from 2634 healthy individuals and 547 patients with certain psychiatric disorders in Changchun and Daqing in the northeast, and in Shanghai in the south of China were examined respectively for the levels of anti-T. gondii IgG by indirect ELISA and a direct agglutination assay. Prevalence of T. gondii infection in the Chinese population in respect of gender, age, residence and health status was systematically analyzed., Results: The overall anti-T. gondii IgG prevalence in the study population was 12.3%. In the clinically healthy population 12.5% was sero-positive and in the group with psychiatric disorders 11.3% of these patients were positive with anti-T. gondii IgG. A significant difference (P = 0.004) was found between male and female in the healthy population, the seroprevalence was 10.5% in men versus 14.3% in women. Furthermore, the difference of T. gondii infection rate between male and female in the 20-19 year's group was more obvious, with 6.4% in male population and 14.6% in female population., Conclusion: A significant higher prevalence of T. gondii infection was observed in female in the clinically healthy population. No correlation was found between T. gondii infection and psychiatric disorders in this study. Results suggest that women are more exposed to T. gondii infection than men in China. The data argue for deeper investigations for the potential risk factors that threat the female populations.
