Livestock grazing and the collection of bamboo shoots are the main threats to giant panda (Ailuropoda melanoleuca) habitat in the Liangshan Mountains in China. It is important to clarify the effect of these disturbances to the giant panda to formulate targeted management policies. Based on species distribution models and daily activity models, we investigated the effects of livestock grazing and bamboo shoot collection on giant pandas from May 2021 to July 2022. Our results indicated the giant panda's suitable habitat in the reserve covered 51.83 km² (15.02% of the reserve area). Grazing and bamboo shoot collection led to losses of 19.08 km² and 7.68 km² of suitable habitat, respectively. Together, the 2 activities resulted in a loss of 28.35 km² of suitable habitat, which was more than half of the area of panda habitat. The areas of suitable habitat for giant pandas significantly overlapped with the areas affected by both disturbances. Giant pandas did not show significant differences in daily activity rhythms under a single disturbance, but the daily activity rhythms of giant pandas differed when we compared the area combining the 2 disturbances with the undisturbed area. Our study reveals that the anthropogenic disturbances in the reserve have varying effects on the suitable habitat range and daily activity rhythm of giant pandas and evidence of a synergistic effect. Therefore, when formulating relevant conservation policies, it is important to fully evaluate the extent and characteristics of anthropogenic disturbances in shaping the population distribution and habitat preferences of the giant panda and other wildlife to enhance the efficacy of conservation management practices. Analyses of animal social networks have traditionally been conducted on species that exhibit social behaviors such as group living, whereas relatively less work has been done on species that are thought of as solitary, are cryptic, and that communicate through scent-marking cues. We employed noninvasive fecal genetic sampling, conducted from March 2015 to February 2016, to identify individuals of one such species, the giant panda (Ailuropoda melanoleuca), across a study population in southwestern China. We then used spatiotemporal proximity thresholds to infer scent mark–based association networks and conduct social network analyses of the population. Results show social clustering in which cluster members preferentially associated with each other. Genetic relatedness was a positive predictor of associations outside the mating season but a negative predictor of associations in the mating season (Mar–Jun), potentially indicating a behavioral change that would reduce risk of inbreeding and kin competition that we term "kin-space recognition." Giant panda Ailuropoda melanoleuca exhibits are popular attractions for zoos and wildlife parks. However, it remains to be investigated whether such exhibits enhance visitor knowledge about pandas and broader conservation issues. We conducted questionnaire surveys at giant panda exhibits at three city zoos and five wildlife parks in China. Although visitors were generally interested in the giant panda, this was not reflected in their post-exhibit knowledge of giant panda biology. Socio-demographically, men were more knowledgeable of giant panda biology than women. Knowledge correlated positively with respondent level of education. Younger respondents (< 45 years) knew most about giant pandas and expressed an interest in learning more about them using social media. The most informed respondents had visited other giant panda exhibits previously. Respondents were generally satisfied with the giant panda exhibits (mean score 4.44/5). Wildlife parks delivered a better educational outcome than city zoos. We recommend approaches to improve the visitor experience further and to leverage public interest in broader conservation engagement and action in China. [ABSTRACT FROM AUTHOR]

Simple Summary: The giant panda is an endemic species in China and the flagship species of global wildlife conservation. Habitat studies of the giant panda corridor can reveal their survival status, habitat environment, and the threats they face, which is crucial for giant panda population recovery and habitat conservation. The results of this study show that due to the opening of the National Road 5 (G5) Niba Mountain tunnel and the completion of the Niba Mountain giant panda corridor plan, the recovery of vegetation within the Niba Mountain giant panda corridor has led to the emergence of giant panda activity in the area, which may have spread to the central part of the reserve through the corridor. The habitat selection characteristics of the giant pandas in the corridor were clarified by investigating the microhabitats of the giant panda corridor in Niba Mountain. These findings can provide a reference for scientists to formulate practical habitat conservation and management measures for giant pandas in the study area. Habitat reduction and increased fragmentation are urgent issues for the survival and recovery of the giant panda (Ailuropoda melanoleuca). However, changes in the distribution and microhabitat selection of giant panda habitats in different seasons in the same region have rarely been assessed. To further understand giant panda habitat requirements, this study analyzed the giant panda habitat selection characteristics and differences using the sample data of the giant panda occurrence sites collected during 2020–2022. The results showed that the giant panda in both seasons selected medium altitudes (2000–2400 m), southeastern slopes, slopes less than 15°, taller tree layers (8–15 m) with a larger diameter at breast height (17–25 cm) and medium density (25–55%), shorter shrub layers (<4 m) with sparse density (<30%), and taller bamboo (>2 m) with high density (>35%). The giant panda microhabitat survey in the Niba Mountain corridor clarified the characteristics of suitable habitat selection for the giant panda in the corridor. We used four anthelmintics—pyrantel pamoate (PYR), mebendazole (MBZ), albendazole (ABZ), and ivermectin (IVM)—on 22 enrolled giant pandas. The fecal egg count reduction (FECR) proportions were calculated using both the Markov chain Monte Carlo (MCMC) Bayesian mathematical model and the arithmetic mean based on fecal egg count data. Anthelmintic resistance (AR) was assessed based on the criteria recommended by the World Association for the Advancement of Veterinary Parasitology (WAAVP). We found that the nematode was suspected to be resistant to PYR. The number of eggs per gram in the feces of giant panda enrolled in the present study was increased near the end of the experiment. Baylisascaris schroederi is one of the main health risks threatening both wild and captive giant pandas. The administration of anthelmintics is a common method to effectively control B. schroederi infection, but there is a notable risk of anthelmintic resistance (AR) after long-term, constant use of anthelmintics. Four anthelmintics—pyrantel pamoate (PYR), mebendazole (MBZ), albendazole (ABZ), and ivermectin (IVM)—were each administered separately at intervals of 2 months to 22 enrolled giant pandas. The fecal egg count reduction (FECR) proportions were calculated by both the Markov chain Monte Carlo (MCMC) Bayesian mathematical model and the arithmetic mean. AR was assessed based on the criteria recommended by the World Association for the Advancement of Veterinary Parasitology (WAAVP). The estimated prevalence of B. schroederi infection was 34.1%. After treatment with PYR, MBZ, ABZ, and IVM, it was determined that MBZ, ABZ, and IVM were efficacious against B. schroederi, while nematodes were suspected to be resistant to PYR according to the fecal egg count reduction (FECR) proportions. The use of camera traps is prevalent in the ecological study of giant pandas (Ailuropoda melanoleuca). The reliability of camera-trap surveying results greatly depends on sampling designs that significantly influence the detection probability of the target species. Few studies have tested the efficacy of sampling designs on camera-trap surveys for monitoring giant pandas in a heterogeneous landscape. In this study, we conducted camera trapping of giant pandas based on two different sampling schemes in Changqing National Nature Reserve of China, and evaluated their outcomes based on three aspects: occupancy analysis, photographic rate, and activity pattern. The results demonstrated that both climate heterogeneity and distance to the nearest road had a strong positive influence on site occupancy, and slope and forest cover had a significant negative impact on site occupancy. Significant differences in the direction or magnitude of variables' influences indicated that there were apparently spatial-temporal dynamics of giant panda distribution between two sampling schemes. The low detection probabilities indicated that both sampling schemes were not robust to accurately monitor giant pandas in the whole study area. We recommended that more suitable sampling designs with local covariates be developed for camera-trap surveys monitoring giant pandas to account for temporal variability and small-scale variation in heterogeneous landscapes. In November 1928, Theodore Jr. and Kermit Roosevelt led an expedition to China with the expressed purpose of being the first Westerners to kill the giant panda (Ailuropoda melanoleuca). The expedition lasted 8 months and resulted in the brothers shooting a giant panda in the mountains of Sichuan Province. Given the concurrent attention in the popular press describing this celebrated expedition, the giant panda was poised to be trophy hunted much like other large mammals around the world. Today, however, the killing of giant pandas, even for the generation of conservation revenue, is unthinkable for reasons related to the species itself and the context, in time and space, in which the species was popularized in the West. We found that the giant panda's status as a conservation symbol, exceptional charisma and gentle disposition, rarity, value as a nonconsumptive ecotourism attraction, and endemism are integral to the explanation of why the species is not trophy hunted. We compared these intrinsic and extrinsic characteristics with 20 of the most common trophy‐hunted mammals to determine whether the principles applying to giant pandas are generalizable to other species. Although certain characteristics of the 20 trophy‐hunted mammals aligned with the giant panda, many did not. Charisma, economic value, and endemism, in particular, were comparatively unique to the giant panda. Our analysis suggests that, at present, exceptional characteristics may be necessary for certain mammals to be excepted from trophy hunting. However, because discourse relating to the role of trophy hunting in supporting conservation outcomes is dynamic in both science and society, we suspect these valuations will also change in future. Livestock grazing occurs in many protected areas for wildlife and has become a threat to wildlife worldwide. Livestock grazing within protected areas causes negative effects to rare wildlife (e.g., giant panda [Ailuropoda melanoleuca]) and their habitat. We used the 2,000‐km2 Wolong National Nature Reserve, Sichuan Province, southwestern China, to document the effects of livestock on the giant panda and its habitat. We monitored arrow bamboo (Bashania fangiana), wildlife sign (i.e., feces and tracks), and characteristics of plant communities in intact habitat (IH; limited livestock grazing) and disturbed habitat (DH; with grazing disturbance) to assess the effects of livestock grazing and the responses of giant pandas and sympatric species across spatial and temporal scales. Bamboo coverage and the height and basal diameter of bamboo in IH were greater than those in DH, whereas the number of herbaceous species and herbaceous coverage in IH were lower than those in DH. Wildlife signs in IH were greater than those in DH; specifically, giant panda and red panda (Ailurus fulgens) signs were greater, whereas signs of sambar (Rusa unicolor) and tufted deer (Elaphodus cephalophus) in IH were similar to those in DH. Livestock grazing reduced bamboo, which may threaten the long‐term survival of the giant panda. Giant pandas (Ailuropoda melanoleuca) use chemical cues to determine identity, gender, and sexual receptivity of conspecifics. We hypothesized that volatile chemical profiles of free-ranging giant pandas are detectable in air. Therefore, we aimed to identify volatile compounds produced by free-ranging giant pandas residing in the Wolong Nature Reserve (Sichuan, China) through field air analysis/solid phase microextraction/gas chromatography-mass spectrometry (FAA/SPME/GCMS). From 28 June to 12 July 2017, 3 SPME fibers were secured to trees that appeared to have previous marking activity. Trail cameras confirmed that a single giant panda performed scent-marking behavior at one sampling location. The abundance of 7 compounds were elevated in samples collected from the tree visited by the giant panda compared with controls. Three ot these compounds (Ethane, 1,1 -dinitro-; Octane, 4-ethyl-; 2(lH)-Pyridinethione, 3-ethoxy-6-methyl-) were unique to the giant panda visit. Wild populations of giant pandas (Ailuropoda melanoleuca) have steadily increased in the past 2 decades, but the species' distribution remains highly fragmented. Since 2009, an introduction program has worked to rescue the giant panda population of Liziping National Nature Reserve in southwestern Sichuan Province, China. Using Global Positioning System and activity collar data collected between May 2011 and March 2016, we investigated the post-release behavior of the first 5 pandas introduced to Liziping, 4 of which were bred in captivity. Using a change-point analysis, we tested several models of post-release adjustment to the habitat. We found that it took 3-4 months for captive-bred individuals to exhibit movement patterns characteristic of their long-term behavior. Furthermore, we found that, for these individuals, post-adjustment behavior varied by season, with activity levels peaking between May and July, a period of high resource availability. This also corresponded with a decrease in large movement events, where individuals were less likely to travel long distances quickly during these months. Unlike wild giant pandas in more northerly reserves, the 5 pandas released in Liziping (both captive-bred and translocated) did not exhibit any seasonal migration between elevations. Finally, we found that our study individuals had 2 daily periods of activity, which was comparable to those reported in the literature for wild individuals. Enterocytozoon bieneusi is an emerging and opportunistic enteric pathogen triggering diarrhea and enteric disease in humans and animals. Despite extensive research on this pathogen, the prevalence and genotypes of E. bieneusi infection in precious wild animals of giant and red pandas have not been reported. In the present study, 82 faecal specimens were collected from 46 giant pandas ( Ailuropoda melanoleuca ) and 36 red pandas ( Ailurus fulgens ) in the northwest of China. By PCR and sequencing of the internal transcribed spacer (ITS) region of the ribosomal RNA (rRNA) gene of E. bieneusi , an overall infection rate of 10.98% (9/82) was observed in pandas, with 8.70% (4/46) for giant pandas, and 13.89% (5/36) for red pandas. Two ITS genotypes were identified: the novel genotype I-like ( n = 4) and genotype EbpC ( n = 5). Multilocus sequence typing (MLST) employing three microsatellites (MS1, MS3 and MS7) and one minisatellite (MS4) showed that nine, six, six and nine positive products were amplified and sequenced successfully at four respective loci. A phylogenetic analysis based on a neighbor-joining tree of the ITS gene sequences of E. bieneusi indicated that the genotype EbpC fell into 1d of group 1 of zoonotic potential, and the novel genotype I-like was clustered into group 2. The present study firstly indicated the presence of E. bieneusi in giant and red pandas, and these results suggested that integrated strategies should be implemented to effectively protect pandas and humans from infecting E. bieneusi in China. Two subfossil partial skeletons of male giant pandas (Ailuropoda melanoleuca) were recovered, along with remains of 16 other mammalian species, from a natural sinkhole on Jiangdong Mountain (south-western Yunnan, China). The panda and other mammalian bones from the sinkhole's upper chamber yielded tightly clustered accelerator mass spectrometry corrected radiocarbon ages of 8470–8250 yr BP, and the panda remains from the lower chamber were found to be 5025 ± 35 yr BP. The bones represent a natural accumulation of mostly large mammals (>30 kg), which had fallen accidentally into the sinkhole. The Jiangdong Mountain mammal assemblage comprises mostly obligate forest-dwellers such as the giant panda, Asian elephant and Sumatran rhinoceros, but the presence of the horse and gaur suggests a more open woodland habitat. Giant pandas were distributed widely in southern China during the Pleistocene, but are now restricted to dense stands of bamboo within temperate forests surrounding the Sichuan Basin. The evidence from Jiangdong Mountain indicates that suitable habitat for pandas extended west of the Salween River through the mid-Holocene. Rapid deforestation of the area in historical times is suggested by an accumulation of cave fill below the opening of the sinkhole containing pieces of an early to middle Ming Dynasty bowl. Understanding present patterns of genetic diversity is critical in order to design effective conservation and management strategies for endangered species. Tangjiahe Nature Reserve (NR) is one of the most important national reserves for giant pandas Ailuropoda melanoleuca in China. Previous studies have shown that giant pandas in Tangjiahe NR may be threatened by population decline and fragmentation. Here we used 10 microsatellite DNA markers to assess the genetic variability in the Tangjiahe population. The results indicate a low level of genetic differentiation between the Hongshihe and Motianling subpopulations in the reserve. Assignment tests using the Bayesian clustering method in STRUCTURE identified one genetic cluster from 42 individuals of the two subpopulations. All individuals from the same subpopulation were assigned to one cluster. This indicates high gene flow between subpopulations. F statistic analyses revealed a low FIS-value of 0.024 in the total population and implies a randomly mating population in Tangjiahe NR. Additionally, our data show a high level of genetic diversity for the Tangjiahe population. Mean allele number (A), Allelic richness (AR) and mean expected heterozygosity (HE) for the Tangjiahe population was 5.9, 5.173 and 0.703, respectively. The effect of landscape on populations is of great importance, especially given the number of species inhabiting patchy landscapes. Developments in geographical information systems are facilitating a greater application of spatial analyses to threatened species, such as the Endangered giant panda Ailuropoda melanoleuca, for which habitat patchiness and quality are key limiting factors. Of all factors that influence the suitability of habitat for the giant panda, topography is not subject to change. Here, we report a spatial and statistical analysis of the high quality topographic habitat preferred by the giant panda across its stronghold in the Qinling Mountains, China. High quality topographic habitat, as indicated by a combination of elevation, slope and aspect, covers 92,788 ha, accounting for 15% of the current range of the species. The distribution of the giant panda closely follows patterns of topography and areas with patches of high quality topographic habitat are strongly associated with areas supporting greater numbers of giant pandas. However, comparisons between our model and the existing reserve system reveals a number of inadequacies. Some of the reserves contain little high quality topographic habitat and many patches of high quality topographic habitat are unprotected. Given the importance of topography and the decisive role it plays in shaping habitat, landscapes containing high quality topographic features must be a critical consideration in the design of reserves for the giant panda. Abstract: Serum samples from 92 giant pandas in three captive facilities were tested for antibodies against five viruses of carnivores. Antibody titers against canine distemper virus (CDV) in two facilities in which giant pandas were vaccinated were variable. The canine adenovirus (CAV-1) and canine parvovirus (CPV) titers in vaccinated group were both positive, but titers were not high and varied among individual except one vaccinated panda had extremely high CAV-1 titer, indicating infection with the field virus following vaccination. Our results suggest that the vaccines used for these giant pandas do not elicit consistent antibody titers. Antibody titers against CDV, CPV and CAV-1 in unvaccinated giant pandas were highly variable, especially CPV titer. Almost half of sera were CPV antibody positive, and CPV titers were high enough to suggest infection with the virus. Canine coronavirus (CCV) and canine parainfluenza virus (CPIV) titers were not detected in all serum samples. Aim To examine the effects of forest fragmentation on the distribution of the entire wild giant panda ( Ailuropoda melanoleuca) population, and to propose a modelling approach for monitoring the spatial distribution and habitat of pandas at the landscape scale using Moderate Resolution Imaging Spectro-radiometer (MODIS) enhanced vegetation index (EVI) time-series data. Location Five mountain ranges in south-western China (Qinling, Minshan, Qionglai, Xiangling and Liangshan). Methods Giant panda pseudo-absence data were generated from data on panda occurrences obtained from the third national giant panda survey. To quantify the fragmentation of forests, 26 fragmentation metrics were derived from 16-day composite MODIS 250-m EVI multi-temporal data and eight of these metrics were selected following factor analysis. The differences between panda presence and panda absence were examined by applying significance testing. A forward stepwise logistic regression was then applied to explore the relationship between panda distribution and forest fragmentation. Results Forest patch size, edge density and patch aggregation were found to have significant roles in determining the distribution of pandas. Patches of dense forest occupied by giant pandas were significantly larger, closer together and more contiguous than patches where giant pandas were not recorded. Forest fragmentation is least in the Qinling Mountains, while the Xiangling and Liangshan regions have most fragmentation. Using the selected landscape metrics, the logistic regression model predicted the distribution of giant pandas with an overall accuracy of 72.5% (κ = 0.45). However, when a knowledge-based control for elevation and slope was applied to the regression, the overall accuracy of the model improved to 77.6% (κ = 0.55). Main conclusions Giant pandas appear sensitive to patch size and isolation effects associated with fragmentation of dense forest, implying that the design of effective conservation areas for wild giant pandas must include large and dense forest patches that are adjacent to other similar patches. The approach developed here is applicable for analysing the spatial distribution of the giant panda from multi-temporal MODIS 250-m EVI data and landscape metrics at the landscape scale. Because habitat loss and fragmentation threaten giant pandas (Ailuropoda melanoleuca), habitat protection and restoration are important conservation measures for this endangered species. However, distribution and value of potential habitat to giant pandas on a regional scale are not fully known. Therefore, we identified and ranked giant panda habitat in Foping Nature Reserve, Guanyinshan Nature Reserve, and adjacent areas in the Qinling Mountains of China. We used Mahalanobis distance and 11 digital habitat layers to develop a multivariate habitat signature associated with 247 surveyed giant panda locations, which we then applied to the study region. We identified approximately 128 km² of giant panda habitat in Foping Nature Reserve (43.6% of the reserve) and 49 km² in Guanyinshan Nature Reserve (33.6% of the reserve). We defined core habitat areas by incorporating a minimum patch-size criterion (5.5 km²) based on home-range size. Percentage of core habitat area was higher in Foping Nature Reserve (41.8% of the reserve) than Guanyinshan Nature Reserve (26.3% of the reserve). Within the larger analysis region, Foping Nature Reserve contained 32.7% of all core habitat areas we identified, indicating regional importance of the reserve. We observed a negative relationship between distribution of core areas and presence of roads and small villages. Protection of giant panda habitat at lower elevations and improvement of habitat linkages among core habitat areas are important in a regional approach to giant panda conservation. We studied the denning ecology of giant pandas (Ailuropoda melanoleuca) in the Foping Nature Reserve, China. We identified 17 used and 21 unused cavities of appropriate size to accommodate denning and measured several variables potentially affecting the suitability of these cavities for panda denning. Principal component analysis, combined with traditional univariate tests, indicated that maternal females preferred deeper cavities with a high interior-to-entrance ratio for height and width, suggesting a preference for narrow entrances and roomy chambers. Microhabitat features, including slope and distance to water, were also useful in predicting den use by maternal females. We believe that the availability of suitable dens may limit population size, especially in areas where tree dens have been eliminated by logging of old growth forests. Conservation managers in giant panda reserves can use our data to determine the number of suitable panda dens that are in an area, estimate whether there are enough for the adult female population to use as birthing dens, and--if needed--construct suitable artificial dens to increase the rate of reproduction. The article presents a research on the temporal changes in the connectivity of Giant Panda Habitat across boundaries of the Wolong Nature Reserve in China. A study is undertaken to analyze the changes in the habitat of giant panda (Ailuropoda melanoleuca) that are inside the Wolong Nature Reserve in Sichuan, China and within its buffer area and boundaries. The study utilized a time series of classified satellite imagery and field observations. It is found out that the habitat connectivity between the inside and outside the reserve area diminished between 1965 and 2001. However, images between 1997 and 2001 present an increase in habitat connectivity due to changes in socioeconomic activities.

The natal den is important to reproduction success of the giant panda Ailuropoda melanoleuca. In this study, we tested the differences between natal dens of the giant panda and reference caves by three statistical methods, and derived the main factors influencing den selection by the giant pandas by principal component analysis (PCA). The results of difference tests indicated that nine variables, including visibility at a height of 1 m, rain proofing, aspect, position, gradient, canopy cover, coverage of bamboo, distance to water and human disturbance were significantly different. The result of PCA indicated that canopy cover was the most important factor in natal den selection by giant pandas. Population viability of the giant panda ( Ailuropoda melanoleuca) is threatened by small population sizes in scattered isolated habitat areas. Designing a conservation plan for protecting and connecting the fragmented habitat will improve the chances for survival of this endangered species. For such a plan, this study assessed the overall habitat suitability for the species in the Qionglai mountain range (Sichuan, China) using Landsat TM imagery acquired in 2001, geographical data, field surveys, and information acquired in previous researches. Results show that the habitat is separated by roads and rivers, as well as by human settlements and cropland areas, into four main habitat blocks. Overlapping these four habitat blocks with the current nature reserve network reveals that only 36% of the total habitat is protected within nature reserves. Thus, the current nature reserve network is failing to preserve essential habitat for dispersal and genetic exchange. In this study, five key areas and four linkage areas were identified and suggested as nature reserves and/or corridors. These areas, together with the six currently established nature reserves in the mountain range, will form a conservation unit for facilitating the exchange of giant panda individuals among previously isolated habitat blocks. Policies recently implemented by the Chinese government, including the Natural Forest Conservation Program (NFCP) and the Grain-to-Green Program (GTGP), could aid in the formation of such a conservation unit. Little is known about habitat selection of the giant panda (Ailuropoda melanoleuca), especially about the relationship between giant panda presence and bamboo and tree structures. We presented data on giant panda habitat use and selection in Foping Nature Reserve (NR), China. We used 1,066 radiotracking locations for 6 collared individuals to analyze giant panda habitat selection, and we used 110 plots to reveal the structure of giant panda habitat and its relationship with giant panda presence. We found that (1) giant pandas in Foping NR selected mostly 3 habitats: conifer forest, deciduous broadleaf forest, and Fargesia bamboo groves. (2) In winter, giant pandas selected deciduous broadleaf forest within elevations of 1,600 to 1,800 m with a south-facing slope of 10 to 20 degrees. In summer, giant pandas selected conifer forest within elevations of 2,400 to 2,600 m and a slope of 20 to 30 degrees. (3) Giant pandas selected the Bashaina fargesii bamboo area with short and dense culms in winter, while they selected the Fargesia qinlingensis bamboo area with a high coverage of tall and thick culms in summer. We concluded that giant pandas in Foping NR do select their preferred habitats. Studies assessing the potential negative consequences of radiocollars on endangered mammals are few. No such studies have been conducted on giant panda (Ailuropoda melanoleuca), an endangered species potentially sensitive to invasive monitoring technologies and handling. Lack of knowledge about the effects of radiocollars and concern for the well-being of radiocollared pandas led to a moratorium on radiocollaring the species in the wild in 1995. To assess the potential effect of radiocollars on the behavior of giant pandas, 4 captive giant pandas (2 male, 2 female) were immobilized and fitted with radiocollars during February and March 2000, just prior to the mating season. We evaluated potential behavioral and endocrine indices of stress daily for 2 weeks before and after immobilization and attachment of radiocollars. We found no significant change in stereotypic behaviors, activity levels, or cortisol levels for any of the 4 individuals. Both adult radiocollared pandas mated successfully. Our results suggest that any potential short-term negative effects from radiocollaring giant pandas are negligible, and with proper planning and use, radiotelemetry can be safely utilized in the study of free-ranging giant pandas. The giant panda Ailuropoda melanoleuca, endemic to China, is perhaps the most powerful and wellknown global icon of biodiversity conservation, having been the symbol of WWF since it was formed in 1961. It is categorized as Endangered on the IUCN Red List. Although there have been conservation successes for the giant panda in the last 2 decades, with 59 reserves established for the species, many conservation challenges remain and the species' status on the IUCN Red List has not changed since 1990, when it was first categorized as Endangered. Habitat fragmentation, synchronous bamboo flowering and die-off, and mismanagement of reserves are continuing challenges. [ABSTRACT FROM AUTHOR]

Scientific research provides essential information for conservation of threatened species. Data deficiency due to insufficient research impedes the design of conservation plans, and research bias may mistakenly direct limited resources to low biodiversity regions or less threatened species. Here, we conducted a systematic review of published papers, grants, and graduate student training on carnivorans in China to identify species bias and research gaps. Furthermore, we collected intrinsic and extrinsic features of carnivorans, and identified features that impact research intensity using generalized linear models. We found that the amount of research on carnivorans increased markedly after 2000, but species bias existed. Bears and big cats received the greatest research attention, while most small- and medium-sized carnivorans received little attention, thus showing the 80-20 phenomenon. Species with a higher level of endemism and protection under Chinese law received more consideration. As an animal conservation icon in China, the giant panda ( Ailuropoda melanoleuca ) attracted more than 50% of overall carnivoran research resources. However, the giant panda also showed spillover effects, i.e., post-doctoral graduates who studied the giant panda shifted their research focus to other species after graduation, which may help improve research on other species. Thus, to improve and strengthen Carnivora research and conservation, we suggest investing greater effort in species of less concern, training of more graduate students, and reinforcing academic exchange. If such actions are not taken, many carnivoran species will continue being data deficient and threatened.

Background: Parasitic infections are among the important causes of death of giant pandas (Ailuropoda melanoleuca) that hamper their survival in the wild. There are about 35 species of parasites which have been identified in giant pandas, but no information is currently available regarding the infection of Babesia in giant pandas. Babesia spp. are common intraerythrocytic parasite in wildlife, transmitted by ixodid ticks, which cause babesiosis. Clinical signs of babesiosis include fever, hemolysis, anemia, jaundice and death., Methods: A species of Babesia was detected in the blood of a giant panda based on morphology and PCR amplification of the 18S rRNA gene. The phylogenetic relationship of Babesia sp. infecting giant panda was assessed by gene sequence alignment and phylogenetic analysis., Results: Our analysis revealed that the Babesia isolate detected was most similar to an unidentified species of Babesia identified in black bears (Ursus thibetanus japonicus) from Japan (Babesia sp. Iwate, AB586027.1) with a 99.56% sequence similarity, followed by Babesia sp. EBB (AB566229.1, 99.50%) and Babesia sp. Akita (AB566229.1, 99.07%)., Conclusions: To our knowledge, this is the first report of Babesia detected in the giant panda. The results indicate that this Babesia sp. may be a novel species, currently named Babesia sp. strain EBP01.

Interspecific killing is a primary reason for the low survival rates of some animal species. The giant panda ( Ailuropoda melanoleuca ) is an altricial eutherian mammal and thus, in comparison to other infants, panda cubs are highly vulnerable, which may significantly influence the selection of breeding sites by females. Here, we used infrared camera traps to monitor giant panda dens for 5.5 years in Foping National Nature Reserve (FNNR) to determine how interspecific factors affect den selection by wild female pandas. Results indicated that Asian black bears ( Ursus thibetanus ), yellow-throated martens ( Martes flavigula ), leopard cats ( Prionailurus bengalensis ), and masked palm civets ( Paguma larvata ) visited the dens frequently, and the presence of these species negatively influenced den selection by female pandas. Interestingly, the presence of rodents and terrestrial birds appeared to indicate den safety, and female giant pandas were not averse and even preferred dens with a high abundance index of rodents and terrestrial birds. The den suitability index (DSI) was a reliable tool for evaluating whether dens were suitable for female giant pandas to give birth to and rear cubs, with preference for dens with high DSI values. This study increases our understanding of the den selection criteria of female giant pandas and the main threats to the survival of their cubs, thus providing important guidance for the conservation and management of this species.

Historically, the giant panda was widely distributed from northern China to southwestern Asia [ 1 ]. As a result of range contraction and fragmentation, extant individuals are currently restricted to fragmented mountain ranges on the eastern margin of the Qinghai-Tibet plateau, where they are distributed among three major population clusters [ 2 ]. However, little is known about the genetic consequences of this dramatic range contraction. For example, were regions where giant pandas previously existed occupied by ancestors of present-day populations, or were these regions occupied by genetically distinct populations that are now extinct? If so, is there any contribution of these extinct populations to the genomes of giant pandas living today? To investigate these questions, we sequenced the nuclear genome of an ∼5,000-year-old giant panda from Jiangdongshan, Tengchong County in Yunnan Province, China. We find that this individual represents a genetically distinct population that diverged prior to the diversification of modern giant panda populations. We find evidence of differential admixture with this ancient population among modern individuals originating from different populations as well as within the same population. We also find evidence for directional gene flow, which transferred alleles from the ancient population into the modern giant panda lineages. A variable proportion of the genomes of extant individuals is therefore likely derived from the ancient population represented by our sequenced individual. Although extant giant panda populations retain reasonable genetic diversity, our results suggest that this represents only part of the genetic diversity this species harbored prior to its recent range contractions. • First nuclear paleogenome of the giant panda • Represents a population that forms the sister group to all extant populations • Differential admixture between extinct and extant panda populations • Part of the extinct, ancient population survives in the modern gene pool Sheng et al. present the first nuclear paleogenome of the giant panda, one of the most iconic mammalian species. The dramatic growth of the captive giant panda (Ailuropoda melanoleuca) population exemplifies how the application of scientific findings to animal care and reproductive management can improve conservation breeding outcomes. Detailed behavioral studies of giant panda estrus, pregnancy and cub rearing have demonstrated the importance of husbandry management that supports natural reproductive behavior to enhance breeding success. Natural breeding has been valuably augmented by the development of assisted reproductive techniques founded through detailed studies of the reproductive physiology of the giant panda and outlining fundamental information about reproductive seasonality, male fertility and characterization of the estrous cycle. The resultant holistic understanding of giant panda reproduction has improved reproductive success in the captive population to such an extent that it is now self-sustaining and provides surplus animals for reintroduction. Despite these significant advances, there are knowledge gaps and remaining challenges to be addressed. Pregnancy detection remains the single biggest challenge when determining if natural mating or assisted breeding have been successful. Because pregnancy can only be determined in the few weeks prior to parturition, there are gaps in understanding and detecting delayed implantation and early embryonic loss. Additionally, dynamic management practices and standard of care for reproductive assistance needs to be developed. Only large breeding centers in China have the ability to promote normal reproductive behaviors and allow mate choice for the giant panda. These challenges need to be addressed in the near future in order to maintain a self-sustaining, genetically diverse and behaviorally competent captive population. This chapter documents the development of successful giant panda managed breeding programs by focusing on three key areas, (1) the development of science-driven reproductive techniques to improve fecundity in a species where the mating system was poorly understood, (2) how targeted research and adaptive management of social settings surrounding estrus and breeding improved reproductive success, and (3) insights and solutions to challenges faced across the program's history with future directions for research.

Aim Temporal transferability is an important issue when habitat models are used beyond the time frame corresponding to model development, but has not received enough attention, particularly in the context of habitat monitoring. While the combination of remote sensing technology and habitat modelling provides a useful tool for habitat monitoring, the effect of incorporating remotely sensed data on model transferability is unclear. Therefore, our objectives were to assess how different satellite-derived variables affect temporal transferability of habitat models and their usefulness for habitat monitoring. Location Wolong Nature Reserve, Sichuan Province, China. Methods We modelled giant panda habitat with the maximum entropy algorithm using panda presence data collected in two time periods and four different sets of predictor variables representing land surface phenology. Each predictor variable set contained either a time series of smoothed wide dynamic range vegetation index (WDRVI) or 11 phenology metrics, both derived from single-year or multi-year (i.e. 3-year) remotely sensed imagery acquired by the Moderate Resolution Imaging Spectroradiometer (MODIS). We evaluated the ability of models obtained with these four variable sets to predict giant panda habitat within and across time periods by using threshold-independent and threshold-dependent evaluation methods and five indices of temporal transferability. Results Our results showed that models developed with the four variable sets were all useful for characterizing and monitoring giant panda habitat. However, the models developed using multi-year data exhibited significantly higher temporal transferability than those developed using single-year data. In addition, models developed with phenology metrics, especially when using multi-year data, exhibited significantly higher temporal transferability than those developed with the time series. Main conclusions The integration of land surface phenology, captured by high temporal resolution remotely sensed imagery, with habitat modelling constitutes a suitable tool for characterizing wildlife habitat and monitoring its temporal dynamics. Using multi-year phenology metrics reduces model complexity, multicollinearity among predictor variables and variability caused by interannual climatic fluctuations, thereby increasing the temporal transferability of models. This study provides useful guidance for habitat monitoring through the integration of remote sensing technology and habitat modelling, which may be useful for the conservation of the giant panda and many other species. We confirmed infection with influenza A(H1N1)pdm09 in giant pandas in China during 2009 by using virus isolation and serologic analysis methods. This finding extends the host range of influenza viruses and indicates a need for increased surveillance for and control of influenza viruses among giant pandas.
