Araucaria araucana is an ancient conifer, native to the mountain ranges in Chile and Argentina. These trees host a large number of organisms, mainly insects, strongly or even exclusively associated with them. The recent emergence of a novel canker disease on A. araucana has emphasised the importance of fungi associated with these iconic trees and has resulted in the discovery of various new species. In this study, we considered the identity of an unknown calicioid fungus consistently found on resin on the branches of A. araucana. Preliminary phylogenetic analyses placed isolates in the recently described sub-class Cryptocaliciomycetidae, closest to Cryptocalicium blascoi. However, the morphology of the ascomata and its occurrence in a unique niche suggested that the closest relative could be Resinogalea humboldtensis (Bruceomycetaceae, incertae sedis), a fungus with similar sporing structures found on resin of Araucaria humboldtensis in New Caledonia. There are no living cultures or sequence data available for either R. humboldtensis or its supposed closest relative, Bruceomyces castoris, precluding sequence-based comparisons. Morphological comparisons of the sporing structures on A. araucana confirmed that the ascomatal morphology of our unknown calicioid fungus and R. humboldtensis are almost identical and resemble each other more so than B. castoris or Cr. blascoi. A phylogenetic analysis based on the small subunit (SSU), internal transcribed spacer (ITS) and large subunit (LSU) rDNA regions resolved our strains into two clades with Cr. blascoi as its closest relative. Further analyses applying the Genealogical Concordance Phylogenetic Species Recognition (GCPSR) based on ITS, mini chromosome maintenance protein complex (MCM7), RNA polymerase II second largest subunit (RPB2) and translation elongation factor 1-alpha (TEF) gene regions, confirmed that strains represent two new species. Based on our morphological observations and phylogenetic analyses, we introduce two new Resinogalea species, R. araucana and R. tapulicola, and reclassify the genus in the subclass Cryptocaliciomycetidae. The taxonomy of Armillaria in southern South America has received little attention since the work of Singer and others. In this study we examine the morphological traits and cultural features for taxa representing the lineages revealed based on molecular phylogenyi and we link them to previously described taxa based on morphology. Lineages I-IV were identified as Armillaria novae-zelandiae, A. montagnei, A. umbrinobrunnea comb. nov. and A. sparrei respectively. They could be differentiated morphologically based on dimension, features of the epicutis, annulus, stipe, hymenophoral trama and flavor and characteristics in culture. Furthermore there was no evidence of host preference for the species recognized. This is the first study integrating the phylogeny and morphology of Armillaria species from Patagonia, and it provides a foundation for future research on these fungi in South America. Abstract The nematode Deladenus siricidicola is the primary biological control agent of Sirex noctilio, a globally invading woodwasp pest of Pinus species. Preliminary studies on the diversity of populations of D. siricidicola revealed very low diversity in the Southern Hemisphere where they have been introduced for the purpose of biological control. The potential to augment biocontrol efficacy by increasing genetic diversity in biocontrol programs motivated this study, which investigated the patterns of genetic diversity in D. siricidicola across eight countries, including the presumed native range (Spain), areas of accidental introduction (Canada and the USA) and countries D. siricidicola has been intentionally released (Argentina, Brazil, Chile, New Zealand and South Africa). Nematodes were screened using mitochondrial COI sequence data and twelve microsatellite markers. Analyses of these data identified three distinct lineages from North America (Lineage A), the Southern Hemisphere (Lineage B) and Spain (Lineage C). Strains from Chile were an exception as they appear to represent an admixture of lineages A and B. This suggests a common origin of populations throughout the Southern Hemisphere, with a second introduction from North America into Chile. The introduction into North America is distinct from that in the Southern Hemisphere and probably originated from Europe. It is evident that substantial genetic diversity exists in D. siricidicola globally, which could be exploited to augment the reduced diversity in some populations used in biocontrol programs. Abstract: Phytophthora pinifolia is the causal agent of the recently discovered needle disease of Pinus radiata in Chile, referred to as “Daño Foliar del Pino” (DFP). The genetic structure of the pathogen population is unknown, which hinders our understanding of its appearance and spread in Chile since 2004. In this study, a population of 88 cultures of P. pinifolia isolated from P. radiata at several localities in Chile was evaluated for genotypic diversity using amplified fragment length polymorphisms (AFLPs). Results of the AFLP analyses showed that the P. pinifolia population in Chile consists of two near identical genotypes but with no genetic differentiation based on geography, year of isolation or the part of the tree from which the isolates were obtained. Mating experiments did not lead to the production of gametangia suggesting that the organism is sterile. The fact that a single clonal genotype dominates the population of P. pinifolia in Chile supports the hypothesis that P. pinifolia was recently introduced into this country and that its impact is due to a new and susceptible host encounter. Ophiostoma ips is a common fungal associate of various conifer-infesting bark beetles in their native ranges and has been introduced into non-native pine plantations in the Southern Hemisphere. In this study, we used 10 microsatellite markers to investigate the population biology of O. ips in native (Cuba, France, Morocco and USA) and non-native (Australia, Chile and South Africa) areas to characterize host specificity, reproductive behaviour, and the potential origin as well as patterns of spread of the fungus and its insect vectors. The markers resolved a total of 41 alleles and 75 haplotypes. Higher genetic diversity was found in the native populations than in the introduced populations. Based on the origin of the insect vectors, the populations of O. ips in Australia would be expected to reflect a North American origin, and those in Chile and South Africa to reflect a European origin. However, most alleles observed in the native European population were also found in the native North American population; only the allele frequencies among the populations varied. This admixture made it impossible to confirm the origin of the introduced Southern Hemisphere (SH) populations of O. ips. There was also no evidence for specificity of the fungus to particular bark beetle vectors or hosts. Although O. ips is thought to be mainly self-fertilizing, evidence for recombination was found in the four native populations surveyed. The higher genetic diversity in the North American than in the European population suggests that North America could be the possible source region of O. ips.
