Nothofagus nitida (Phil.) Krasser (Nothofagaceae) regenerates under the canopy in microsites protected from high light. Nonetheless, it is common to find older saplings in clear areas and adults as emergent trees of the Chilean evergreen forest. We hypothesized that this shade to sun transition in N. nitida is supported by an increase in photochemical and non-photochemical energy dissipation capacities of both photosystems in parallel with the increase in plant size and light availability. To dissect the relative contribution of light environment and plant developmental stage to these physiological responses, the photosynthetic performance of both photosystems was studied from the morpho-anatomical to the biochemical level in current-year leaves of N. nitida plants of different heights (ranging from 0.1 to 7 m) growing under contrasting light environments (integrated quantum flux (IQF) 5–40 mol m−2 day−1). Tree height (TH) and light environment (IQF) independently increased the saturated electron transport rates of both photosystems, as well as leaf and palisade thickness, but non-photochemical energy flux, photoinhibition susceptibility, state transition capacity, and the contents of D1 and PsbS proteins were not affected by IQF and TH. Spongy mesophyll thickness and palisade cell diameter decreased with IQF and TH. Amax, light compensation and saturation points, Rubisco and nitrogen content (area basis) only increased with light environment (IQF), whereas dark respiration (Rd) decreased slightly and relative chlorophyll content was higher in taller trees. Overall, the independent effects of more illuminated environment and tree height mainly increased the photochemical instead of the non-photochemical energy flux. Regardless of the photochemical increase with TH, carbon assimilation only significantly improved with higher IQF. Drought stress conditions (DC) reduce plant growth and nutrition, restraining the sustainable reestablishment of Nothofagus dombeyi in temperate south Chilean forest ecosystems. Ectomycorrhizal symbioses have been documented to enhance plant nitrogen (N) and phosphorus (P) uptake under drought, but the regulation of involved assimilative enzymes remains unclear. We studied 1-year-old N. dombeyi (Mirb.) Oerst. plants in association with the ectomycorrhizal fungi Pisolithus tinctorius (Pers.) Coker & Couch. and Descolea antartica Sing. In greenhouse experiments, shoot and root dry weights, mycorrhizal colonization, foliar N and P concentrations, and root enzyme activities [glutamate synthase (glutamine oxoglutarate aminotransferase (GOGAT), EC 1.4.1.13-14), glutamine synthetase (GS, EC 6.3.1.2), glutamate dehydrogenase (GDH, EC 1.4.1.2-4), nitrate reductase (NR, EC 1.6.6.1), and acid phosphomonoesterase (PME, EC 3.1.3.1-2)] were determined as a function of soil-water content. Inoculation of N. dombeyi with P. tinctorius and D. antartica significantly stimulated plant growth and increased plant foliar N and P concentrations, especially under DC. Ectomycorrhizal inoculation increased the activity of all studied enzymes relative to non-mycorrhizal plants under drought. We speculate that GDH is a key enzyme involved in the enhancement of ectomycorrhizal carbon (C) availability by fuelling the tricarboxylic acid (TCA) cycle under conditions of drought-induced carbon deficit. All studied assimilative enzymes of the ectomycorrhizal associations, involved in C, N, and P transfers, are closely interlinked and interdependent. The up-regulation of assimilative enzyme activities by ectomycorrhizal fungal root colonizers acts as a functional mechanism to increase seedling endurance to drought. Nothofagus dombeyi (Mirb.) Blume and Nothofagus nitida (Phil.) Krasser are closely related evergreen trees native to south central Chile. Nothofagus dombeyi is a pioneer in habitats subject to high daytime irradiances and nighttime freezing temperatures and has a wider altitudinal and latitudinal distribution than N. nitida, which is restricted to more oceanic climates. We postulated that N. dombeyi has a greater cold-acclimation capacity, expressed as a greater capacity to maintain a functional photosynthetic apparatus at low temperatures, than N. nitida. Because cold-acclimation may be related to the accumulation of cryoprotective substances, we investigated relationships between ice nucleation temperature (IN), freezing temperature (FT), and the temperature causing injury to 50% of the leaf tissues (LT50) on the one hand, and concentrations of total soluble carbohydrates (TSC), starch and proline on the other hand. Observations were made throughout a seasonal cycle in adults and seedlings in the field and in seedlings in the laboratory under cold-acclimation inductive and non-inductive conditions. In adults, LT50 values were lower in N. dombeyi than in N. nitida, suggesting that N. dombeyi is the more frost tolerant species. Adults of both species tolerated freezing in autumn and winter but not in spring and summer. In the fall and winter, seedlings of N. dombeyi had a much lower LT50 than those of N. nitida. Nothofagus nitida seedlings, in autumn and winter, exhibited freezing avoidance mechanisms. Although elevated TSC and proline concentrations may contribute to freezing tolerance in adults of both species, an increase in proline concentration is unlikely to be the dominant frost tolerance response in adults because proline concentrations were higher in N. nitida than in N. dombeyi. In seedlings, however, there were large differences in proline accumulation between species that may account for the difference between them in freezing tolerance. Starch concentration in both species decreased during winter. Chlorophyll fluorescence indicated that maximal photochemical efficiency (Fv/Fm) remained at optimal values (∼0.8) throughout the year. The effective photochemical efficiency of PSII (ΦPSII) and relative electron transport rates (ETRr) decreased in winter in both species. In seedlings, fluorescence parameters were more affected in winter in N. nitida than in N. dombeyi. We concluded that adults and seedlings of N. dombeyi are hardier than adults and seedlings of N. nitida, which is consistent with its wider latitudinal and altitudinal distribution. Gastric cancer is the third main cause of cancerous tumors in humans in Chile. It is well-accepted that a diet rich in antioxidant plants could help in fighting cancer. Blueberry is a fruit crop with a high content of antioxidants. Methyl jasmonate (MeJA) is a phytohormone involved in plant defenses under stress conditions. The exogenous application of MeJA can improve the antioxidant properties in plants. We studied in vitro and in vivo anticancer action on human gastric cancer (cell line AGS) and the antioxidant properties of extracts from blueberry plants untreated and treated with MeJA. The results demonstrated that leaf extracts displayed a higher inhibition of cancer cell viability as well as greater antioxidant properties compared to fruit extracts. Besides, MeJA applications to plants improved the antioxidant properties of leaf extracts (mainly anthocyanins), increasing their inhibition levels on cell viability and migration. It is noteworthy that leaf extract from MeJA-treated plants significantly decreased cancer cell migration and expression of gastric cancer-related proteins, mainly related to the mitogen-activating protein kinase (MAPK) pathway. Interestingly, in all cases the anticancer and antioxidant properties of leaf extracts were strongly related. Despite highlighted outcomes, in vivo results did not indicate significant differences in Helicobacter pylori colonization nor inflammation levels in Mongolian gerbils unfed and fed with blueberry leaf extract. Our findings demonstrated that MeJA increased antioxidant compounds, mainly anthocyanins, and decreased the viability and migration capacity of AGS cells. In addition, leaf extracts from MeJA-treated plants were also able to decrease the expression of gastric cancer-related proteins. Our outcomes also revealed that the anthocyanin-rich fraction of blueberry leaf extracts showed higher in vitro antiproliferative and anti-invasive effects than the crude leaf extracts. However, it is still uncertain whether the leaf extracts rich in anthocyanins of blueberry plants are capable of exerting a chemopreventive or chemoprotective effect against gastric cancer on an in vivo model. Nothofagus dombeyi (Mirb.) Blume and Nothofagus nitida (Phil.) Krasser, two evergreens in the South Chilean forest, regenerate in open habitats and under the canopy, respectively. Both overtop the forest canopy when they are in the adult stage, suggesting that their photoprotective mechanisms differ in ontogenetic dynamics. We postulated that N. nitida, a shade-tolerant species increases its capacity to tolerate photoinhibitory conditions (low temperature and high irradiance) by thermal energy dissipation of excess energy during its transition from the seedling to the adult stage, whereas N. dombeyi, a shade-intolerant species, maintains a high capacity for photoprotection by thermal energy dissipation from the seedling to the adult stage. To test this hypothesis, the main photoprotective mechanisms in plants – the fast- and slow-relaxing components of thermal energy dissipation (NPQ, non-photochemical quenching) NPQF and NPQS, respectively, and state transitions – were studied in seedlings and adults of both species grown in their natural habitats and in a common garden. In adults, NPQF and NPQS did not differ between species and seasons. The greatest differences in these parameters were observed in seedlings. The xanthophyll cycle was more active in N. dombeyi seedlings than in N. nitida seedlings at low temperature and high irradiance, consistent with a higher NPQF in N. dombeyi. Under all study conditions, N. nitida seedlings had higher NPQS than N. dombeyi seedlings. The state transition capability was higher in N. nitida seedlings than in N. dombeyi seedlings. Therefore, although (shade-intolerant) N. dombeyi was able to thermally dissipate the excess absorbed energy, under natural conditions its photochemical energy quenching was efficient in both developmental stages, decreasing its need for thermal dissipation. In contrast, the seedlings of N. nitida were more sensitive to photoinhibition than the adult trees, suggesting a change from shade-grown to sun-exposed phenotype from the seedling to the adult stage. These results help to explain the differences in the regeneration patterns of N. nitida and N. dombeyi and the presence of N. nitida adult stage in the upper canopy. [ABSTRACT FROM AUTHOR]

In acid soils, manganese (Mn) concentration increases, becoming toxic to plants. Mn toxicity differentially affects physiological processes in highbush blueberry (Vaccinium corymbosum L.) cultivars. However, the mechanisms involved in Mn toxicity of the new and traditionally established cultivars are unknown. To understand Mn toxicity mechanisms, four traditionally established (Legacy, Brigitta, Duke, and Star) cultivars and two recently introduced to Chile (Camellia and Cargo) were grown under hydroponic conditions subjected to control Mn (2 μM) and Mn toxicity (1000 μM). Physiological, biochemical, and molecular parameters were evaluated at 0, 7, 14, and 21 days. We found that the relative growth rate was reduced in almost all blueberry cultivars under Mn toxicity, except Camellia, with Star being the most affected. The photosynthetic parameters were reduced only in Star by Mn treatment. Leaf Mn concentrations increased in all cultivars, exhibiting the lowest levels in Camellia and Cargo. Brigitta and Duke exhibited higher β-carotene levels, while Cargo exhibited a reduction under toxic Mn. In Legacy, lutein levels increased under Mn toxicity. Traditionally established cultivars exhibited higher antioxidant activity than the new cultivars under Mn toxicity. The Legacy and Duke cultivars increased VcMTP4 expression with Mn exposure time. A multivariate analysis separated Legacy and Duke from Camellia; Star and Cargo; and Brigitta. Our study demonstrated that Mn toxicity differentially affects physiological, biochemical, and molecular features in the new and traditionally established cultivars, with Legacy, Duke, Camellia, and Cargo as the Mn-resistant cultivars differing in their Mn-resistance mechanisms and Star as the Mn-sensitive cultivar. • The new and traditionally established highbush blueberry cultivars differ in their Mn-resistance mechanisms. • The new cultivars present the Mn-resistant exclusion mechanism; meanwhile, Legacy and Duke the Mn-tolerance mechanism. • Star cultivar reduces photosynthetic performance and plant growth by Mn toxicity. • Legacy and Duke showed an increase of VcMTP4 expression concomitant with their shoot Mn levels under toxic Mn. It is possible to incorporate a lot of natural antioxidants into the human organism by consuming berries which can prevent diseases generated by the action of free radicals. Antioxidants neutralize free radicals and thus protect the organism from the oxidative damage of lipids, proteins, and nucleic acids. Berries stand out as one of the richest sources of antioxidant phytonutrients among various fruits and vegetables. The objective of this research was to determine antioxidant capacity (AC), total anthocyanins (TA), and total phenols (TP) of wild and cultivated berries in different localities of La Araucanía and Los Ríos Regions in Chile. These parameters were analyzed by using the free radical 2,2-diphenyl-1-picrylhydrazyl hydrate (DPPH) method, pH-differential, and Folin-Ciocalteu method. Percentages of DPPH discoloration of different berries studied were between 67.8% and 95.3% for red sarsaparilla and rosehip, respectively. Maqui berries showed a significantly higher TA content (2240.2 and 1445.3 mg L-1 cyanidin 3-glucoside) than other berries, and a mean for all berries of 335.5 mg L-1. Higher phenol content levels were obtained in two cultivars of saskatoon (773.9 and 1001.9 mg L-1 gallic acid) and wild rosehip (1457.0 and 1140.4 mg L-1 gallic acid). We conclude that there are significant differences in antioxidant capacity of wild and cultivated Chilean berries in this study which show a strong correlation between AC and TP content. In April and December 1989, 35 fish from Lake Huillinco (42 degrees 48'S, 74 degrees 02'W) and 36 fish from Lake Natri (42 degrees 48'S, 73 degrees 50'W), in the Great Island of Chiloé (Chile) were examined. Coprological samples from 159 persons, 17 dogs, 19 pigs and 4 cats from around both lakes were examined for Diphyllobothrium spp. infection. In the Lake Huillinco the following helminths of fishes were determined: Contracaecum sp. and Hysterothylacium sp. in Salmo trutta, Cauque mauleanum and Eleginops maclovinus; Dichelyne (Cucullanellus) dichelyneformis in S. trutta and E. maclovinus and Scolex pleuronectis in S. trutta. One specimen of Mugil cephalus did not show helminth parasites. Prevalence of infection were greater for Contracaecum sp. in S. trutta (75.0%) and C. mauleanum (76.0%); and Hysterothylacium sp. in E. maclovinus (75.0%). Mean intensity was higher for D. (C.) dichelyneformis in E. maclovinus. Contracaecum sp. in S. trutta, Oncorhynchus mykiss, Oncorhynchus kisutch and Galaxias maculatus; Acanthocephalus sp. in S. trutta and G. maculatus, S. pleuronectis in O. mykiss and Cystidicoloides sp. in G. maculatus were determined at Lake Natri. Prevalence and intensity of infection were higher for Contracaecum sp. in S. trutta and O. kisutch. Infection by Diphyllobothrium sp. was determined in one domestic cat. Prevalence of infection by intestinal protozoan and helminths in human population only showed significative differences for Ascaris lumbricoides and Trichuris trichiura that were higher in the Lake Huillinco. Importance of natural infection by helminth parasites for fish in cultured condition and possible mechanisms of infections in relation to the diet of fishes are discussed.
