Wildfire is the dominant disturbance in boreal forests and fire activity is increasing in these regions. Soil fungal communities are important for plant growth and nutrient cycling postfire but there is little understanding of how fires impact fungal communities across landscapes, fire severity gradients, and stand types in boreal forests. Understanding relationships between fungal community composition, particularly mycorrhizas, and understory plant composition is therefore important in predicting how future fire regimes may affect vegetation. We used an extreme wildfire event in boreal forests of Canada's Northwest Territories to test drivers of fungal communities and assess relationships with plant communities. We sampled soils from 39 plots 1 year after fire and 8 unburned plots. High‐throughput sequencing (MiSeq, ITS) revealed 2,034 fungal operational taxonomic units. We found soil pH and fire severity (proportion soil organic layer combusted), and interactions between these drivers were important for fungal community structure (composition, richness, diversity, functional groups). Where fire severity was low, samples with low pH had higher total fungal, mycorrhizal, and saprotroph richness compared to where severity was high. Increased fire severity caused declines in richness of total fungi, mycorrhizas, and saprotrophs, and declines in diversity of total fungi and mycorrhizas. The importance of stand age (a surrogate for fire return interval) for fungal composition suggests we could detect long‐term successional patterns even after fire. Mycorrhizal and plant community composition, richness, and diversity were weakly but significantly correlated. These weak relationships and the distribution of fungi across plots suggest that the underlying driver of fungal community structure is pH, which is modified by fire severity. This study shows the importance of edaphic factors in determining fungal community structure at large scales, but suggests these patterns are mediated by interactions between fire and forest stand composition. Background. Biological invasions are major drivers of environmental change that can significantly alter ecosystem function and diversity. In plants, soil microbes play an important role in plant establishment and growth; however, relatively little is known about the role they might play in biological invasions. A first step to assess whether root microbes may be playing a role in the invasion process is to find out if invasive plants host different microbes than neighbouring native plant species. Methods. In this study we investigated differences in root associated microbes of native sugar maple (Acer saccharum Marsh.) and exotic Norway maple (A. platanoides L.) collected from a forested reserve in eastern Canada. We used microscopy to examine root fungi and high-throughput sequencing to characterize the bacterial, fungal and arbuscular mycorrhizal communities of both maple species over one growing season. Results. We found differences in root associated bacterial and fungal communities between host species. Norway maple had a higher bacterial and fungal OTU (operational taxonomic units) richness compared to sugar maple, and the indicator species analysis revealed that nine fungal OTUs and three bacterial OTUs had a significant preference for sugar maple. The dominant bacterial phyla found on the roots of both maple species were Actinobacteria and Proteobacteria. The most common fungal orders associated with the Norway maple roots (in descending order) were Helotiales, Agaricales, Pleosporales, Hypocreales, Trechisporales while the Agaricales, Pleosporales, Helotiales, Capnodiales and Hypocreales were the dominant orders present in the sugar maple roots. Dark septate fungi colonization levels were higher in the sugar maple, but no differences in arbuscular mycorrhizal fungal communities and colonization rates were detected between maple species. Discussion. Our findings show that two congeneric plant species grown in close proximity can harbor distinct root microbial communities. These findings provide further support for the importance of plant species in structuring root associated microbe communities. The high colonization levels observed in Norway maple demonstrates its compatibility with arbuscular mycorrhizal fungi in the introduced range. Plant-associated microbial communities can affect host fitness and function in many ways; therefore, the observed differences suggest a possibility that biotic interactions can influence the dynamics between native and invasive species. Field studies were conducted over 4 years at two locations in Manitoba, Canada to evaluate effects of preceding crop, tillage and phosphorus (P) fertilization on Cd and Zn concentration in oilseed flax (linseed— Linum usitatissimum L.). Canola ( Brassica napus L.) and spring wheat ( Triticum aestivum L.) were grown under conventional and reduced tillage with three rates of monoammonium phosphate. Flax was seeded the year following the spring wheat or canola, with or without P fertilizer, and Cd and Zn concentration and accumulation were determined in the flax tissue at 5 weeks and in the mature seed. Flax following canola had lower Zn concentration and accumulation and higher Cd concentration and Cd:Zn ratio in the tissue and seed relative to flax following wheat. Phosphorus fertilization tended to increase Cd concentration and Cd:Zn ratio and decrease Zn concentration in the tissue and seed. Effects of tillage and interactions among tillage, preceding crop, and P fertilization were inconsistent. Changes in flax Cd and Zn concentration may be due to changes in mycorrhizal colonization or by the high concentration of Cd in the decomposing canola residue. Crop sequence and P management can be used to improve flaxseed food quality, by increasing Zn and decreasing Cd concentration. [ABSTRACT FROM AUTHOR]

Genera in the tribe Pyroleae (subfamily Monotropoideae, family Ericaceae) occur as understory plants in northern temperate zones where some form major components of ecosystems. Most have been poorly studied in terms of their association with symbiotic fungi. In this study, colonization patterns of mycorrhizal roots of five members of the Pyroleae (Pyrola asarifolia Michx., Pyrola chlorantha Sw., Orthilia secunda (L.) House, Chimaphila umbellata (L.) W. Bart., Moneses uniflora (L.) Gray) were explored. Root samples were processed for light, fluorescence, and laser scanning confocal, scanning electron, and transmission electron microscopy, as well as for immunocytochemistry. Roots of all species had enlarged epidermal cells containing hyphal complexes, Hartig nets confined to the epidermis, and mantles. Epidermal cells were penetrated by hyphae originating from the Hartig net at more than one site either along the inner tangential wall or radial walls. The outer tangential wall of epidermal cells of all species, except M. uniflora, was thicker than radial and inner tangential walls and consisted of two layers, the outer containing nonesterified pectins that were labeled with JIM 5 antibodies. Radial walls and inner tangential walls did not label, but cortical cell walls did. Intracellular hyphal complexes developed initially around centrally positioned, enlarged epidermal cell nuclei and, through branching, occupied most of the cell volume. Senescence and degradation of the complexes followed. The fungal species in these symbiotic associations may be important functionally in nutrient exchange, as well as in contributing to broader linkages with other hosts in these plant communities. Arbuscular mycorrhizae (AM) are seldom reported from high latitudes. We found that Asteraceae (Arnica, Erigeron, and Taraxacum) at a site on Axel Heiberg Island (approximately 80°N) have abundant AM and fine endophytes (FE). We used standard microscopic methods for examination and quantification, plus high-resolution confocal fluorescence imaging. AM in Arctic Asteraceae were compared with those in congeners from Saskatoon and with those in some other Arctic species. Arctic AM had 6 µm wide aseptate hyphae producing abundant arbuscules, vesicles, and inter- and intra-cellular hyphae. AM colonization exceeded 80% for Arctic Asteraceae, similar to 66%–90% for prairie Taraxacum and Erigeron, the first of this type of comparison. AM/FE abundance in Arctic Ranunculus was 68%. Within Taraxacum roots, hyphal coils predominated near the epidermis and arbuscules near the vascular cylinder. Arctic AM colonization did not vary with soil depth, although permafrost was approximately 15 cm below the surface. FE were abundant in our High Arctic samples, where they may have functional roles comparable with those of AM. Thus, low abundance of AM reported previously at the community level for high-latitude sites may reflect a combination of biotic and abiotic factors. A combination of light microscopy (including differential interference contrast) and laser scanning confocal microscopy was used to document the colonization patterns of epidermal cells and details of intracellular hyphal complexes of five native ericaceous hosts: Vaccinium oxycoccos L. (bog cranberry), Ledum groenlandicum Oeder. (Labrador tea), Vaccinium myrtilloides L. (velvet-leaf blueberry), Kalmia angustifolia L. (sheep laurel), and Gaultheria procumbens L. (wintergreen). Colonization patterns, hyphal complex morphology, and the structure of thick-walled epidermal cells varied considerably among hosts. Multiple hyphal connections were observed between adjacent epidermal cells, indicating that one fungal entry point may result in the colonization of more than one epidermal cell. Further field observations combined with fungal isolations from field-collected plants, identification, and reinoculation studies of other species in the large Ericaceae family are required to determine the full range of structural details in ericoid mycorrhizas. Summary • This study compares DNA and culture-based detection of fungi from 15 ericoid mycorrhizal roots of salal (Gaultheria shallon ), from Vancouver Island, BC Canada. • From the 15 roots, we PCR amplified fungal DNAs and analyzed 156 clones that included the internal transcribed spacer two (ITS2). From 150 different subsections of the same roots, we cultured fungi and analyzed their ITS2 DNAs by RFLP patterns or sequencing. We mapped the original position of each root section and recorded fungi detected in each. • Phylogenetically, most cloned DNAs clustered among Sebacina spp. (Sebacinaceae, Basidiomycota). Capronia sp. and Hymenoscyphus erica (Ascomycota) predominated among the cultured fungi and formed intracellular hyphal coils in resynthesis experiments with salal. • We illustrate patterns of fungal diversity at the scale of individual roots and compare cloned and cultured fungi from each root. Indicating a systematic culturing detection bias, Sebacina DNAs predominated in 10 of the 15 roots yet Sebacina spp. never grew from cultures from the same roots or from among the > 200 ericoid mycorrhizal fungi previously cultured from different roots from the same site.
