Wastewater treatment plant effluent introduces a mixture of pollutants into marine environments; however, the impacts of chronic sublethal exposures on populations are often unclear. Presence of estrogenic agents in sediments and uptake of these compounds by demersal flatfishes has been reported at the Orange County Sanitation District (OCSD) wastewater outfall. Furthermore, estrogenic activity has been identified in male flatfish in the area, potentially contributing to observed population declines in the OCSD region. Rising ocean temperatures may further contribute to flatfish declines as relationships between temperature and abundance have been reported in the Southern California Bight. To investigate declines, sex ratios, condition factor, organ health indices, hormones, and vitellogenin were quantified in flatfish collected at OCSD outfall and reference sites. Additionally, historical temperature data was examined for trends with population abundances. Rather than being linked to estrogenic activity, results indicated that population declines were more correlated to increases in ocean temperature., (Copyright © 2018 Elsevier Ltd. Sentinel fish hornyhead turbot (Pleuronichthysverticalis) captured near wastewater outfalls are used for monitoring exposure to industrial and agricultural chemicals of ~ 20 million people living in coastal Southern California. Although analyses of hormones in blood and organ morphology and histology are useful for assessing contaminant exposure, there is a need for quantitative and sensitive molecular measurements, since contaminants of emerging concern are known to produce subtle effects. We developed a second generation multi-species microarray with expanded content and sensitivity to investigate endocrine disruption in turbot captured near wastewater outfalls in San Diego, Orange County and Los Angeles California. Analysis of expression of genes involved in hormone [e.g., estrogen, androgen, thyroid] responses and xenobiotic metabolism in turbot livers was correlated with a series of phenotypic end points. Molecular analyses of turbot livers uncovered altered expression of vitellogenin and zona pellucida protein, indicating exposure to one or more estrogenic chemicals, as well as, alterations in cytochrome P450 (CYP) 1A, CYP3A and glutathione S-transferase-α indicating induction of the detoxification response. Molecular responses indicative of exposure to endocrine disruptors were observed in field-caught hornyhead turbot captured in Southern California demonstrating the utility of molecular methods for monitoring environmental chemicals in wastewater outfalls. Moreover, this approach can be adapted to monitor other sites for contaminants of emerging concern in other fish species for which there are few available gene sequences.

Constructed wetlands (CW) offer a mechanism to meet increasingly stringent regulatory standards for wastewater treatment while minimizing energy inputs. Additionally, harvested wetland phytomass subjected to anaerobic digestion can serve as a source of biogas methane. To investigate CW wastewater polishing activities and potential energy yield we constructed a pair of secondary wastewater-fed channelized CW modules designed to retain easily harvestable floating aquatic vegetation and maximize exposure of water to roots and sediment. Modules that were regularly harvested averaged a nitrate removal rate of 1.1 g N m(-2) d(-1); harvesting, sedimentation and gasification were responsible for 30.5%, 8.0% and 61.5% of the N losses, respectively. Selective harvesting of a module to maintain dominance of filamentous algae had no effect on nitrate removal rate but lowered productivity by one-half. The average monthly productivity for unselectively harvested modules was 9.3 ± 1.7 g dry wt. m(-2) d(-1) (±SE). Cessation of harvesting in one module resulted in a significant increase in nitrate removal rate and decrease in phosphate removal rate. Compared to the influent, the effluent of the harvested module had significantly lower levels of estrogenic activity, as determined by a quantitative PCR-based juvenile trout bioassay, and significantly lower densities of E. coli. In mixed vertical-flow reactors anaerobic co-digestion of equal dry weight proportions of harvested aquatic vegetation, wine yeast lees and dairy manure was greatly improved when the manure was replaced with the crude glycerol by-product of biodiesel production. Remaining solids were vermicomposted for use as a soil amendment. Our results indicate that incorporation of constructed wetlands into an integrated treatment system can simultaneously enhance the economic and energetic feasibility of wastewater and organic waste treatment processes.

The Santa Ana River (SAR) is the primary source of groundwater recharge for the Orange County Groundwater Basin in coastal southern California. Approximately 85% base flow in the SAR originates from wastewater treatment plants operated by three dischargers. An on-line, flow-through bioassay using Japanese medaka (Oryzias latipes) as a means of judging potential public health impacts was employed to evaluate the water quality of the surface water and shallow groundwater originating from the SAR. Three chronic (3-4.5 mo) exposures using orange-red (outbred, OR) and see-through (color mutant, ST-II) Japanese medaka as bioindicators were conducted to evaluate endocrinologic, reproductive, and morphologic endpoints. No statistically significant differences in gross morphological endpoints, mortality, gender ratios, and vitellogenin induction were observed in fish from SAR groundwater treatment compared to the group tested in solute reconstituted reverse osmosis-treated or granular activated carbon (GAC)-treated control waters. Significant differences were observed in egg reproduction and the time to hatch in SAR groundwater; however, total hatchability was not significantly lower. To evaluate the estrogenic activity of the surfacewater source of the groundwater, SAR surface water was evaluated for vitellogenin and gonadal histopathology in juvenile medaka with no effects observed. These results demonstrate that OR Japanese medaka may be a sensitive strain as an on-line monitor to predict potential impacts of water quality, but further studies are needed to elicit causative agents within the water mixture.
