Scarlet Royal, a mid-season ripening table grape, is one of the popular red grape varieties in California. However, its berries develop an undesirable astringent taste under certain conditions. Among the various factors contributing to the degradation of berry attributes, the levels and compositions of polyphenols play a fundamental role in defining berry quality and sensory characteristics. To comprehend the underlying mechanism of astringency development, Scarlet Royal berries with non-astringent attributes at the V7 vineyard were compared to astringent ones at the V9 vineyard. Biochemical analysis revealed that the divergence in berry astringency stemmed from alterations in its polyphenol composition, particularly tannins, during the late ripening stage at the V9 vineyard. Furthermore, transcriptomic profiling of berries positively associated nineteen flavonoid/proanthocyanidins (PAs) structural genes with the accumulation of PAs in V9 berries. The identification of these genes holds significance for table grape genetic improvement programs. At a practical level, the correlation between the taste panel and tannin content revealed a threshold level of tannins causing an astringent taste at approximately 400 mg/L. Additionally, berry astringency at the V9 vineyard was linked to a lower number of clusters and yield during the two study seasons, 2016 and 2017. Furthermore, petiole nutrient analysis at bloom showed differences in nutrient levels between the two vineyards, including higher levels of nitrogen and potassium in V9 vines compared to V7. It's worth noting that V9 berries at harvest displayed a lower level of total soluble solids and higher titratable acidity compared to V7 berries. In conclusion, our results indicate that the accumulation of tannins in berries during the ripening process results in a reduction in their red color intensity but significantly increases the astringency taste, thereby degrading the berry quality attributes. Mineral dust aerosols often observed over California in winter and spring, associated with long-range transport from Asia and the Sahara, have been linked to enhanced precipitation based on observations. Local anthropogenic pollution, on the other hand, was shown in previous observational and modeling studies to reduce precipitation. Here we incorporate recent developments in ice nucleation parameterizations to link aerosols with ice crystal formation in a spectralbin cloud microphysical model coupled with the Weather Research and Forecasting (WRF) model in order to examine the relative and combined impacts of dust and local pollution particles on cloud properties and precipitation type and intensity. Simulations are carried out for two cloud cases (from the CalWater 2011 field campaign) with contrasting meteorology and cloud dynamics that occurred on 16 February (FEB16) and 2 March (MAR02). In both cases, observations show the presence of dust and biological particles in a relative pristine environment. The simulated cloud microphysical properties and precipitation show reasonable agreement with aircraft and surface measurements. Model sensitivity experiments indicate that in the pristine environment, the dust and biological aerosol layers increase the accumulated precipitation by 10-20% from the Central Valley to the Sierra Nevada for both FEB16 and MAR02 due to a ~40% increase in snow formation, validating the observational hypothesis. Model results show that local pollution increases precipitation over the windward slope of the mountains by a few percent due to increased snow formation when dust is present, but reduces precipitation by 5-8%if dust is removed on FEB16. The effects of local pollution on cloud microphysics and precipitation strongly depend on meteorology, including cloud dynamics and the strength of the Sierra Barrier Jet. This study further underscores the importance of the interactions between local pollution, dust, and environmental conditions for assessing aerosol effects on cold-season precipitation in California. Objective: To analyze the transcriptomic profile of endometrial gene alterations during the window of implantation in infertile obese patients.Design: Multicenter, prospective, case-control study.Setting: Three academic medical centers for reproductive medicine.Patient(s): Infertile patients, stratified into body mass index (BMI) categories according to the World Health Organization guidelines, were included in the study.Intervention(s): Endometrial samples were obtained from women undergoing standardized estrogen and P replacement cycles after 5 days of vaginal P supplementation.Main Outcome Measure(s): To identify endometrial gene expression alterations that occur during the window of implantation in infertile obese patients as compared with infertile normal-weight controls using a microarray analysis.Result(s): XCL1, XCL2, HMHA1, S100A1, KLRC1, COTL1, COL16A1, KRT7, and MFAP5 are significantly dysregulated during the window of implantation in the receptive endometrium of obese patients. COL16A1, COTL1, HMHA1, KRCL1, XCL1, and XCL2 were down-regulated and KRT7, MFAP5, and S100A1 were up-regulated in the endometrium of obese patients. These genes are mainly involved in chemokine, cytokine, and immune system activity and in the structural extracellular matrix and protein-binding molecular functions.Conclusion(s): Obesity is associated with significant endometrial transcriptomic differences as compared with non-obese subjects. Abstract: In-situ aircraft measurements of aerosol chemical and cloud microphysical properties were conducted during the CalWater campaign in February and March 2011 over the Sierra Nevada Mountains and the coastal waters of central California. The main objective was to elucidate the impacts of aerosol properties on clouds and precipitation forming processes. In order to accomplish this, we compared contrasting cases of clouds that ingested aerosols from different sources. The results showed that clouds containing pristine oceanic air had low cloud drop concentrations and started to develop rain 500m above their base. This occurred both over the ocean and over the Sierra Nevada, mainly in the early morning when the radiatively cooled stable continental boundary layer was decoupled from the cloud base. Supercooled rain dominated the precipitation that formed in growing convective clouds in the pristine air, up to the −21°C isotherm level. A contrasting situation was documented in the afternoon over the foothills of the Sierra Nevada, when the clouds ingested high pollution aerosol concentrations produced in the Central Valley. This led to slow growth of the cloud drop effective radius with height and suppressed and even prevented the initiation of warm rain while contributing to the development of ice hydrometeors in the form of graupel. Our results show that cloud condensation and ice nuclei were the limiting factors that controlled warm rain and ice processes, respectively, while the unpolluted clouds in the same air mass produced precipitation quite efficiently.
