Abstract: A new entomopathogenic nematode, Steinernema brazilense n. sp., was isolated from a single soil sample collected from a natural forest in Mato Grosso do Sul state, Brazil. S. brazilense n. sp. is characterized morphologically by features of infective juveniles (IJ), males and females. For the IJ, body length averaging 1157 (1023–1284)μm, distance from anterior end to excretory pore 95 (87–102)μm, from anterior end to end of esophagus 148 (139–153)μm, tail length 85 (80–104)μm, D% and E% values 63 (58–70) and 106 (95–118.0), respectively. Lateral field pattern variable; the formula for the arrangement of ridges from head to tail is: 2, 4, 6, 8, 6, 2. For the male, the diagnostic characters include spicule averaging 83 (75–89)μm; D% about 65; the ratio SW% about 192. The length of spicule head is greater than width. Lateral field with one narrow ridge. First generation females are characterized by the presence of a ventral postanal swelling. S. brazilense n. sp. is morphologically close to Steinernema diaprepesi. It can be differentiated from S. diaprepesi by its longer IJ body length (1157 vs 1002μm), longer distance from anterior end to excretory pore (110 vs 75μm), a longer tail length (103 vs 83μm); males of the new species with longer spicule (83 vs 79μm). The new species can be distinguished further from other members of Steinernema glaseri group by characteristics of rDNA of ITS and D2D3 regions. [Copyright &y& Elsevier]

In a survey of entomopathogenic nematodes in Brazil, a nematode isolate of the genus Heterorhabditis was found. The nematode was collected from soil by the insect-baiting technique and maintained in the laboratory on last instar Galleria mellonella (L.) larvae. Morphological and molecular studies of the isolate showed that the nematode is a new species. Light and scanning electron microscopy, DNA characterisation and phylogeny were used for this description. Heterorhabditis amazonensis n. sp. is morphologically similar to H. baujardi, H. floridensis, H. mexicana and H. indica, and can be distinguished from these species mainly by male and female characters. Fifty percent of Heterorhabditis amazonensis n. sp. males have two pairs of bursal papillae in the terminal group; 25% with two papillae on one side and one papilla on the other side and 25% with one pair of papillae. Twenty percent of the population has a curved gubernaculum. The percentage of the gubernaculum to spicule length (GS%) is lower than that of H. mexicana (50 vs 56), and the length of the spicule relative to anal body diam. (SW%) is lower than that of H. mexicana (152 vs 167) and H. baujardi (152 vs 182). The female can be differentiated from related species by its unique vulva pattern. In a phylogenetic tree, the new species, H. floridensis, H. baujardi and H. mexicana form a monophyletic group, a sister group to H. indica. The new species has evolved three autapomorphic nucleotide character states, differing from its sister taxa H. mexicana at 22, and H. baujardi at 15, aligned positions. [ABSTRACT FROM AUTHOR]

In Brazil, countless insect species attack and damage sugarcane (Saccharum officinarum L.), which is an extremely important crop since it is planted on more than 10 million hectares. Among these insects, the sugarcane billbug, Sphenophorus levis (Coleoptera: Curculionidae), is of great importance as the larvae open tunnels in the rhizome of the plant, causing high damage and losses. This insect is attracted mainly to vinasse, which is the liquid fraction generated from the alcohol production and discarded onto the sugarcane fields for fertigation. Toward a novel control method for S. levis, the native entomopathogenic nematode Steinernema rarum (Pam 25) was compared with S. carpocapsae (IL 1) and Heterorhabditis bacteriophora (HBEN01) in respect to their ability to search for larvae of two insect hosts (Galleria mellonella and S. levis) within the cane rhizome. The selected nematode S. rarum was also assessed for rate effects, its survival in vinasse and field efficacy to control sugarcane pests S. levis, Hyponeuma taltula (Lepidoptera, Erebidae) and Leucothyreus alvarengai (Coleoptera: Melolonthidae). Steinernema rarum exhibited superior virulence to G. mellonella and S. levis larvae inserted into the cane rhizomes (75–78% mortality) compared to S. carpocapsae (30–53%) and H. bacteriophora (18–28%). Vinasse affected S. rarum when infective juveniles were suspended in the liquid compost for more than 6 h but did not affect the nematode when kept on the straw and soil treated with the compost. Steinernema rarum tested at 1–3 × 108 infective juveniles/ha on the sugarcane field caused 74.1, 56.3 and 50.6% control of S. levis, L. alvarengai and H. taltula, respectively. Heterorhabditis is a nematode found in the soil that is used as an important biological control agent against various organisms. However, few studies have been performed of its use against snails and the present study is the first to investigate the effect of experimental exposure of Bradybaena similaris to Heterorhabditis indica LPP1. Two groups of 16 snails were formed: the control group (not exposed) and the treatment, which was exposed for three weeks to infective juveniles (J3) of H. indica LPP1. The entire experiment was conducted in duplicate, using a total of 64 snails. After this period, the snails were dissected to collect the hemolymph to evaluate the possible physiological alterations, namely total proteins, uric acid and hemolymph urea, as well as the activities of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) as a result of the infection. The terrariums were analyzed on alternate days throughout the experiment to count the dead snails. Intense proteolysis was observed in the infected snails. An increase in the level of uric acid and reduction of the hemolymph urea content indicated that the infection by H. indica results in the inversion of the excretion pattern of the host snail. Variations in the aminotransferase activities were also observed, with the infected group presenting significantly higher values (p<0.05) than the control group for both ALT and AST. The exposure to H. indica LPP1 caused 55% mortality, with the highest rate observed in the first week after exposure (30%).
