The proliferation of Raphidiopsis raciborskii blooms has sparked concerns regarding potential human exposure to heightened saxitoxins (STXs) levels. Thus, comprehending how environmental elements drive the proliferation of this STXs-producing species can aid in predicting human exposure risks. This study aimed to explore the link between cyanobacteria R. raciborskii, STXs cyanotoxins, and environmental factors in 37 public supply reservoirs in the tropical region and assess potential health hazards these toxins pose in the reservoir waters. A Structural Equation Model was used to assess the impact of environmental factors (water volume and physical and chemical variables) on R. raciborskii biomass and STXs levels. Furthermore, the potential risk of STXs exposure from consuming untreated reservoir water was evaluated. Lastly, the cumulative distribution function (CDF) of STXs across the reservoirs was computed. Our findings revealed a correlation between R. raciborskii biomass and STXs concentrations. Total phosphorus emerged as a critical environmental factor positively influencing species biomass and indirectly affecting STXs levels. pH significantly influenced STXs concentrations, indicating different factors influencing R. raciborskii biomass and STXs. Significantly, for the first time, the risk of STXs exposure was gauged using the risk quotient (HQ) for untreated water consumption from public supply reservoirs in Brazil's semi-arid region. Although the exposure risks were generally low to moderate, the CDF underscored the risk of chronic exposure due to low toxin concentrations in over 90% of samples. These outcomes emphasize the potential expansion of R. raciborskii in tropical settings due to increased phosphorus, amplifying waterborne STXs levels and associated intoxication risks. Based on a literature survey, we evaluated the periods of cyanobacterial dominance in Brazil. We hypothesized that variability of environmental forces along the country will promote or facilitate temporal and spatial mosaic in cyanobacterial dominance. The most striking outcomes are related to the dominance of Cylindrospermopsis, Dolichospermum, and Microcystis. Although they share important adaptive strategies (e.g., aerotopes, large size and toxins production), our findings suggest that they have different environmental preferences. Dolichospermum and Microcystis dominated mainly in warm-rainy periods whereas Cylindrospermopsis was more common during dry periods and in mixed systems, or formed perennial dominance. Maximum phosphorus concentrations were observed in reservoirs dominated by Cylindrospermopsis. Although the main genera reached high biomass levels individually, different abilities to form dominance and co-dominance were observed. The number of co-dominance of Chroococales and Nostocales was almost the same as the individual occurrence of the main genera from these groups. This dataset reveals patterns of dominance of these cyanobacteria and also indicates that physiological features will cause differences in the mechanisms of interactions between species. The understanding of these processes and their relationship to environmental conditions will promote better understanding of cyanobacterial dominance and increase our ability to predict and manage these events. Abstract: Longitudinal heterogeneity in reservoirs is especially related to increase in sedimentation and water transparency along the river/dam axis. Consequently, primary production tends to reach higher values in intermediate regions where there is a balance between the availability of the main resources (light and nutrients) suitable for phytoplankton growth. Many factors such as reservoir morphometry, retention time, thermal stratification and geographical location can affect the boundaries between these regions. The tropical Funil Reservoir (Brazil), despite a low retention time, has experienced severe eutrophication in recent decades, with persistent cyanobacteria blooms. During the course of 1 year, samples were collected at four stations along the reservoir (fluvial, intermediate and lentic compartments) to evaluate if spatial heterogeneity could affect the occurrence and distribution of these blooms along the reservoir. Although the reservoir has a short annual retention time (mean 41.5 days), the typical zonation pattern was observed for the main abiotic variables and phytoplankton abundance. However, higher biomass occurred in the lentic compartment rather than in the intermediate zone. Despite the peculiar heterogeneity in total biomass, the phytoplankton composition and seasonal variability were very similar along the entire reservoir, with a few marked differences only in the fluvial zone. Phytoplankton total biomass in Funil Reservoir was high, even in periods of lower seasonal retention time (around 15 days), and was especially related to high input of nutrients. Moreover, retention time directly affects the spatial heterogeneity of phytoplankton biomass, since strong variability was only observed during the cold–dry season, corresponding to periods of longer retention time (around 80 days). While high availability of nutrients promoted high cyanobacterial biomass in the entire system, the few periods of heterogeneous spatiality seemed to be related to changes in retention time. Blooms of Cylindrospermopsis raciborskii and Anabaena spiroides were studied in relation to their toxins composition, geographical locations, and other characteristics of the waters in the southern region of Brazil. All forms of Cylindrospermopsis were paralytic shellfish toxin producers, with a similar profile of the toxins. Anabaena blooms were studied in relation to the production of anatoxin-a(S). In all samples containing Anabaena spiroides, a positive result was found when the AChE inhibition technique was used. Methods applied for both studies are very convenient for monitoring this large region and give a reasonable view of the present situation of water reservoirs in southern Brazil. Toxic cyanobacteria blooms are a frequent problem in subtropical reservoirs and freshwater systems. The purpose of this study was to investigate the occurrence of potentially toxic cyanobacteria and the environmental conditions associated with the presence of cyanotoxins in a Brazilian subtropical reservoir. Five collections were carried out at seven sampling locations in the reservoir, during the rainy and dry seasons, between the years 2016 and 2017. There was permanent occurrence of Raphidiopsis raciborskii (Woloszynska) Aguilera, Berrendero Gómez, Kastovsky, Echenique & Salerno (Phycologia 57(2):130-146, 2018), ranging between dominant and abundant, with an average biomass of 38.8 ± 29.9 mg L -1 . Also abundant were Dolichospermum solitarium, D. planctonicum, Planktothrix isothrix, and Aphanizomenon gracile. Saxitoxin (STX) was detected in all the collected samples (0.11 ± 0.05 µg L -1 ). Microcystin (MC) was also detected, but at lower concentrations (0.01 ± 0.0 µg L -1 ). Low availability of NO 3 - and phosphorus limitation had significant effects on the R. raciborskii biomass and the levels of STX and MC. It was observed that R. raciborskii was sensitive to thermal stratification, at the same time that STX levels were higher. This suggested that STX was produced under conditions that restricted the growth of R. raciborskii. • Toxic cyanobacterium Raphidiopsis raciborskii was found blooming in a coastal lake whose water is used for human consumption. • The toxic biomass is transported downstream reaching coastal and estuarine waters. • R. raciborskii strains can survive, grow and increase the production of saxitoxins at salinities up to 6. • Saxitoxins of limnic origin can reach coastal ecosystems and accumulate in marine shellfish, causing poisoning. Raphidiopsis raciborskii (formerly Cylindrospermopsis raciborskii) is a freshwater cyanobacterium potentially producing saxitoxins (STX) and cylindrospermopsin. Its ecophysiological versatility enables it to form blooms in the most diverse types of environments, from tropical to temperate, and from relatively pristine to polluted. In Peri Lake, located in the subtropical south of Brazil, growing populations of STX-producing R. raciborskii have been detected since 1994, posing risks to the use of its waters that supply a population of about 100,000 inhabitants. Despite the existence of a monitoring system for the presence and toxicity of cyanobacteria in Peri Lake water, no assessment has been made in the coastal region, downstream of outflowing lake water, thereby potentially making available a toxic biomass to natural and cultivated shellfish populations in the salt water ecosystem. To address this problem, the present study evaluated environmental variables and STX concentration by profiling the outflowing waters between Peri Lake and the adjacent coastal zone. Laboratory experiments were carried out with three strains of R. raciborskii in order to confirm the effect of salinity on STX production and verify if Perna Perna mussels fed with R. raciborskii cultures would absorb and accumulate STX. Results showed that environmental concentrations of STX reach high levels (up to 6.31 µg L−1 STX eq.), especially in the warmer months, reaching the coastal zone. In laboratory tests, it was found that the strains tolerate salinities between 4 and 6 and that salinity influences the production of STX. In addition, mussels fed with R. raciborskii effectively absorb and accumulate STX, even in typically marine salinities (22 to 30), suggesting that R. raciborskii biomass remains available and toxic despite salinity shock. These results draw attention to the ecological and health risk associated with R. raciborskii blooms, both in the lake environment and in the adjacent marine environment, calling attention to the need to improve the monitoring and management systems for water and shellfish toxicity in the region of interest, as well as other places where toxic cyanobacteria of limnic origin can reach the coastal zone. Geoengineering techniques have been used to control phosphorus and cyanobacteria in lakes promising greater and quicker chemical and ecological recovery. Techniques that use coagulants and clays to remove particulates and dissolved phosphorus from the water column have received great. In this study, bench-scale "flock & sink" assays were carried out to evaluate the efficiency of the coagulants aluminium sulphate (SUL), polyaluminium chloride (PAC) and chitosan (CHI), alone and combined with natural bentonite clays (BEN) and lanthanum-modified bentonite (LMB), to remove of phosphorus from a eutrophic reservoir in a semi-arid region of Brazil. In addition, the study seeks to assess the effects on the cyanobacteria density and the intra- and extracellular concentrations of cyanotoxins after the application of these geoengineering materials. The SUL and PAC coagulants effectively reduced the total phosphorus (TP), reactive soluble phosphorus (SRP), turbidity, chlorophyll-a, cyanobacteria density and intracellular microcystin, whereas CHI showed a low removal efficiency. Lanthanum-modified bentonite proved to be more effective than BEN; however, the application of the coagulants only was sufficient to successfully remove phosphorus and cyanobacteria from the water column. In addition, the efficiency of the "flock & sink" technique in cell removal varied among the cyanobacteria species. Small colonial species such as Aphanocapsa delicatissima, Merismopedia glauca and Merismopedia tenuissima were removed regardless of the treatment used, including those with CHI and BEN. As for the filamentous cyanobacteria, Cylindrospermopsis raciborskii, Geitlerinema amphibium, Planktothrix agardhii and Pseudanabaena catenata, removal was achieved only using PAC, SUL and LMB alone or when combined. The intracellular concentrations of saxitoxin and cylindrospermopsin and the extracellular fraction of these cyanotoxins and of microcystin were not influenced by the application of coagulants and clays. This indicates that cell lysis did not occur with the addition of the geoengineering materials. Camorim is a small, eutrophic reservoir in Rio de Janeiro, Brazil, with a phytoplankton community dominated most of the year by the filamentous diatom Aulacoseira spp. and the toxic cyanobacterium Cylindrospermopsis raciborskii. As filamentous species can be a poor food for grazers, we hypothesize that phytoplankton from this reservoir would constrain cladoceran fitness due to nutritional limitation and/or toxicity when animals fed mixtures of cultured green algae and natural seston. Clones of different cladoceran species were exposed either to seston from Camorim reservoir sampled in different seasons or to a C. raciborskii strain (CYLCAM-2) isolated from the reservoir. In short-term assays, cladocerans were exposed to either 100% seston or mixtures of 50% seston added to green algae (200 μg C L -1 ), and their survivorship and somatic growth were measured for 4 days. In life table assays, neonates were exposed to the same seston treatments over 14 days and age at first reproduction, survivorship, fecundity, total offspring, and the intrinsic rate of natural increase (r) were assessed. In general, seston negatively affected cladoceran survivorship and fitness (r), but this response was seasonally and species specific. Stronger effects of CYLCAM-2 than those caused by seston on survivorship, somatic growth, and r were found for all cladoceran species, especially when the proportion of CYLCAM-2 was higher than 50% in relation to green algae in a fixed total food concentration. Our results suggest that both nutritional (C/P and morphology) and toxicity factors can act to impair cladoceran fitness and help explain the absence of cladocerans in Camorim reservoir.

This study investigated the dynamics of cyanobacteria in two deep, eutrophic reservoirs in a semi-arid region of Brazil during periods of stratification and destratification. Four collections were carried out at each reservoir at two depths at three-month intervals. The following abiotic variables were analyzed: water temperature, dissolved oxygen, pH, turbidity, water transparency, total phosphorus, total dissolved phosphorus, orthophosphate and total nitrogen. Phytoplankton density was quantified for the determination of the biomass of cyanobacteria. The data were analyzed using CCA. Higher mean phytoplankton biomass values (29.8 mm(3).L(-1)) occurred in the period of thermal stratification. A greater similarity in the phytoplankton communities also occurred in this period and was related to the development of cyanobacteria, mainly Cylindrospermopsis raciborskii (>3.9 mm(3).L(-1)). During the period of thermal destratification, this species co-dominated the environment with Planktothrix agardhii, Geitlerinema amphibium, Microcystis aeruginosa and Merismopedia tenuissima, as well as with diatoms and phytoflagellates. Environmental instability and competition among algae hindered the establishment of blooms more during the mixture period than during the stratification period. Thermal changes in the water column caused by climatologic events altered other physiochemical conditions of the water, leading to changes in the composition and biomass of the cyanobacterial community in tropical reservoirs.
