Background: Lucilia cuprina (Wiedemann, 1830) (Diptera: Calliphoridae) is the main causative agent of flystrike of sheep in Australia and New Zealand. Female flies lay eggs in an open wound or natural orifice, and the developing larvae eat the host's tissues, a condition called myiasis. To improve our understanding of host-seeking behavior, we quantified gene expression in male and female antennae based on their behavior., Methods: A spatial olfactometer was used to evaluate the olfactory response of L. cuprina mated males and gravid females to fresh or rotting beef. Antennal RNA-Seq analysis was used to identify sensory receptors differentially expressed between groups., Results: Lucilia cuprina females were more attracted to rotten compared to fresh beef (> fivefold increase). However, males and some females did not respond to either type of beef. RNA-Seq analysis was performed on antennae dissected from attracted females, non-attracted females and males. Transcripts encoding sensory receptors from 11 gene families were identified above a threshold (≥ 5 transcript per million) including 49 ATP-binding cassette transporters (ABCs), two ammonium transporters (AMTs), 37 odorant receptors (ORs), 16 ionotropic receptors (IRs), 5 gustatory receptors (GRs), 22 odorant-binding proteins (OBPs), 9 CD36-sensory neuron membrane proteins (CD36/SNMPs), 4 chemosensory proteins (CSPs), 4 myeloid lipid-recognition (ML) and Niemann-Pick C2 disease proteins (ML/NPC2), 2 pickpocket receptors (PPKs) and 3 transient receptor potential channels (TRPs). Differential expression analyses identified sex-biased sensory receptors., Conclusions: We identified sensory receptors that were differentially expressed between the antennae of both sexes and hence may be associated with host detection by female flies. The most promising for future investigations were as follows: an odorant receptor (LcupOR46) which is female-biased in L. cuprina and Cochliomyia hominivorax Coquerel, 1858; an ABC transporter (ABC G23.1) that was the sole sensory receptor upregulated in the antennae of females attracted to rotting beef compared to non-attracted females; a female-biased ammonia transporter (AMT_Rh50), which was previously associated with ammonium detection in Drosophila melanogaster Meigen, 1830. Background: Larvae of the Australian sheep blowfly, Lucilia cuprina, parasitise sheep by feeding on skin excretions, dermal tissue and blood, causing severe damage known as flystrike or myiasis. Recent advances in -omic technologies and bioinformatic data analyses have led to a greater understanding of blowfly biology and should allow the identification of protein families involved in host-parasite interactions and disease. Current literature suggests that proteins of the SCP (Sperm-Coating Protein)/TAPS (Tpx-1/Ag5/PR-1/Sc7) (SCP/TAPS) superfamily play key roles in immune modulation, cross-talk between parasite and host as well as developmental and reproductive processes in parasites., Methods: Here, we employed a bioinformatics workflow to curate the SCP/TAPS protein gene family in L. cuprina. Protein sequence, the presence and number of conserved CAP-domains and phylogeny were used to group identified SCP/TAPS proteins; these were compared to those found in Drosophila melanogaster to make functional predictions. In addition, transcription levels of SCP/TAPS protein-encoding genes were explored in different developmental stages., Results: A total of 27 genes were identified as belonging to the SCP/TAPS gene family: encoding 26 single-domain proteins each with a single CAP domain and a solitary double-domain protein containing two conserved cysteine-rich secretory protein/antigen 5/pathogenesis related-1 (CAP) domains. Surprisingly, 16 SCP/TAPS predicted proteins formed an extended tandem array spanning a 53 kb region of one genomic region, which was confirmed by MinION long-read sequencing. RNA-seq data indicated that these 16 genes are highly transcribed in all developmental stages (excluding the embryo)., Conclusions: Future work should assess the potential of selected SCP/TAPS proteins as novel targets for the control of L. cuprina and related parasitic flies of major socioeconomic importance.

The last 50 years of research into infections in Australia and New Zealand caused by larvae of the sheep blowfly, Lucilia cuprina, have significantly advanced our understanding of this blowfly and its primary host, the sheep. However, apart from some highly effective drugs it could be argued that no new control methodologies have resulted. This review addresses the major areas of sheep blowfly research over this period describing the significant outcomes and analyses, and what is still required to produce new commercial control technologies. The use of drugs against this fly species has been very successful but resistance has developed to almost all current compounds. Integrated pest management is becoming basic to control, especially in the absence of mulesing, and has clearly benefited from computer-aided technologies. Biological control has more challenges but natural and perhaps transformed biopesticides offer possibilities for the future. Experimental vaccines have been developed but require further analysis of antigens and formulations to boost protection. Genetic technologies may provide potential for long-term control through more rapid indirect selection of sheep less prone to flystrike. Finally in the future, genetic analysis of the fly may allow suppression and perhaps eradication of blowfly populations or identification of new and more viable targets for drug and vaccine intervention. Clearly all these areas of research offer potential new controls but commercial development is perhaps inhibited by the success of current chemical insecticides and certainly requires a significant additional injection of resources., (Crown Copyright © 2014. Objective: To determine the effect of applying plastic clips to the breech and tail of lambs on the perineal and tail bare areas, breech wrinkle, dag (faecal) accumulation, urine stain, body weight and survival of lambs, compared with unclipped, unmulesed lambs and mulesed lambs., Method: The study was conducted on five commercial wool-growing farms in southern Australia. On each study site lambs (2-12-weeks-old) were divided into three groups: (1) unclipped, unmulesed control (tail dock only); (2) treated with clips; and (3) treated with the mules operation. Evaluations of effects, including visual scoring, bare area measurements and body weight, were performed before treatment and on or approximately days 30, 60, 90 and 180 after treatment. On each occasion, lambs were recorded as either present or absent to allow estimates of survival., Results: The clip treatment increased the size of the perineal and tail bare areas compared with the unclipped, unmulesed control lambs (P < 0.05), although the increases were less than in mulesed lambs (P < 0.05). The clips reduced breech wrinkle, dag and urine stain to levels partway between those recorded in the unclipped, unmulesed controls and the mulesed lambs (P < 0.05). Clipped lambs weighed more than mulesed lambs after treatment (days 30-90 P < 0.001; day 180 P < 0.01) and had higher cumulative percentage survival to 90 days after treatment (P = 0.03)., Conclusions: The clips successfully modified the breech region of lambs, although the changes were less than with mulesing. The clips did not reduce the body weight of lambs, compared with the unclipped, unmulesed control lambs, and improved 90-day cumulative percentage survival compared with mulesed lambs., (© 2012 The Authors. The blowfly, Lucilia cuprina Wiedemann (Diptera: Calliphoridae), is the primary myiasis (strike) fly of sheep in Australia. Most strike occurs in the anal-perineum area (crutch), but strike to the neck, shoulders, back and withers (body) is also important. Regression analysis was used to determine the extent to which the weekly incidence of flystrike can be explained by variations in fly abundance and/or recent changes in weather, pasture conditions or flock management. Strike and flock management data were collected by questionnaire surveys of 30-60 sheep properties in each of three major sheep-producing areas in southeastern Australia, namely, Gunning (southern New South Wales), Inverell (northern New South Wales) and Flinders Island (Bass Strait). After using simulation modelling to remove effects due to shearing, crutching and/or insecticide treatment, pasture growth index was found to be the most important explanatory variable affecting the incidence of all forms of myiasis. Others were average weekly air temperature, the amount and frequency of rainfall, relative humidity, dung quality index and a factor denoting seasonal effects. Together, these variables accounted for 48.4% of the variation in body strike, 56.8% of that in crutch strike and 51.9% of that in other forms of strike. Prediction was improved by the inclusion of additional lagged variables describing previous strike, fly abundance and fly activity. With these additions, the variation explained increased to 60.4% for body strike, 68.0% for crutch strike and 58.3% for other strikes.

Breeding for disease resistance in Merino sheep in Australia has attracted considerable research and development attention. Increased labour costs, the reduced efficacy of common anthelmintics and insecticides, consumer demand for products which are free of chemicals and the poor prognosis of alternative long-term control strategies are all forcing sheep breeders to contemplate the best animal health options available, including selective breeding for resistance. The three major diseases which affect sheep production include gastrointestinal nematode parasites, flystrike (cutaneous myiasis) and footrot. Genetic improvement has been shown to be feasible in all these diseases, possibly with little adverse effect on genetic progress in other production traits. Programmes for resistance to internal parasites to be included in practical breeding programmes are now in progress. This paper deals with the incentives for focusing on the three major diseases in Merino sheep, the potential sources of genetic variation, and the means to exploit these sources of variation. The authors also highlight gains and benefits achieved in experimental selection flocks, and the difficulties and options available for commercial breeders.

To consider the evolutionary origin of the ectoparasitic habit in the blowfly genus Lucilia (Diptera:Calliphoridae), phylogenetic analyses of mitochondrial DNA sequence data were performed for 10 species, including all the common Lucilia agents of myiasis, collected from Africa, Australasia, North America and Europe. Complementary genetic distance and parsimony analyses are used to consider inter and intraspecific relationships within the genus with reference to previous morphological work. The results support the hypothesis of independent multiple evolution of the ectoparasitic habit in Lucilia sericata, Lucilia cuprina and the Lucilia caesar/Lucilia illustris group and suggest that it has coevolved in relatively recent history along with the domestication and husbandry of sheep. The geographic differences in pathogenic importance of various species of Lucilia also suggest that there is a strong climatic influence determining which species has dominated. Lucilia cuprina has become the predominant pathogenic species in sub-tropical and warm temperate habitats (e.g., Australia and South Africa), L. sericata in cool temperate habitats (e.g., Europe and New Zealand) and L. caesar and L. illustris become more common in sheep myiasis in more northerly Palaearctic regions.

A field survey was undertaken between 1972-76 to reappraise the nature of the blowfly problem in New South Wales. For 2 years, 1972-1974, some 80,000 sheep were kept under observation and 12,481 strikes were reported, most due to Lucilia cuprina. Breech strike was still the basic problem but tail strike associated with scouring had become an important component where pastures have been improved. Breech strike was controlled at minimal cost by managerial practices such as docking tails the correct length--second joint-space palpable ventrally (midway down the vulval orifice in ewes) for radically mulesed lambs and the third joint-space (tip of vulva in ewes) for all other lambs-mulesing at lamb marking, mid-season crutching, determining the cause of scouring and applying the appropriate preventative or remedial measures. Thus the use of insecticides could be reserved for the control of body strike in young sheep in the odd wet years and poll strike in horned rams. Major outbreaks of body strike occurred in 1973/74. Body strike worried graziers most because of its unpredictability, sudden onset and scale. and only failing insecticides were available for control.
