A pot experiment was conducted in an open greenhouse to explore the use of citrate-coated cobalt ferrite nanoparticles (CoFe 2 O 4 NPs) as a source for Fe fortification of three wheat lines ( Triticum aestivum L.). Two of the three wheat lines tested differ in their efficiency concerning Zn storage in their grains (efficient and inefficient), and one had inefficient P-absorption. The NPs were supplied by foliar or soil application of Fe at 330 mg L -1 , and 46 or 68 mg kg -1 soil, respectively. A positive control (Fe-EDTA salt, a conventional iron fertilizer) and a negative control (no fertilization) were also included to compare the efficiency of NP fertilization. Soil fertilization with NPs improved the grain yield and Fe concentration in the grains compared with the foliar application of NPs and conventional Fe fertilizer. Application of soil NPs at 68 mg kg -1 increased the grain yield by 52% and 21% compared with the control and soil Fe-EDTA treatments, respectively. Likewise, grain Fe concentration increased by 96% and 72% compared with the control and soil Fe-EDTA treatments, respectively. The phytic acid concentration in grains and the phytic acid:Fe ratio decreased by 6% and 62%, respectively, due to the soil application of NPs (68 mg Fe per kg). The Fe grain concentration of lines inefficient for Zn storage and P-uptake in plants from soil fertilized with NPs (68 mg Fe per kg) was 1.37 and 0.26 fold above the target biofortification concentration (60 mg Fe per kg). Cobalt concentration in grains ranged from 9 to 16 mg kg -1 . These concentrations were below the maximum allowable limit of Co in grains (50 mg kg -1 ) recommended by FAO and the WHO. Our results showed that Fe supplied as NPs may improve the nutritional quality of wheat grains, and the economic yield. However, there remains a long way to go to achieve effective and economic use of nanotechnology for the nutritional development of wheat.

A comparative study of the effects of exposure to high Cd 2+ (50 µM) and excess Zn 2+ (600 µM) on photosynthetic performance of hydroponically-grown durum wheat seedlings was performed. At day 8, Cd and Zn were added to the nutrient solution. After 7-days exposure, the chosen concentrations of both metals resulted in similar relative growth rate (RGR) inhibitions of about 50% and comparable retardations of the CO₂ assimilation rates (about 30%) in the second developed leaf of wheat seedlings. Analysis of chlorophyll a fluorescence indicated that both metals disturbed photosynthetic electron transport processes which led to a 4- to 5-fold suppression of the efficiency of energy transformation in Photosystem II. Non-specific toxic effects of Cd and Zn, which prevailed, were an inactivation of part of Photosystem II reaction centres and their transformation into excitation quenching forms as well as disturbed electron transport in the oxygen-evolving complex. The specificity of the Cd and Zn modes of action was mainly expressed in the intensity of the toxicity effects: despite the similar inhibitions of the CO₂ assimilation rates, the wheat photochemistry showed much more sensitivity to Cd than to Zn exposure., Competing Interests: The authors declare no conflict of interest.

Plant growth promoting endophytic bacteria (PGPB) isolated from Brassica napus were inoculated in two cultivars of Helianthus tuberosus (VR and D19) growing on sand supplemented with 0.1 mM Cd or 1 mM Zn. Plant growth, concentrations of metals and thiobarbituric acid (TBA) reactive compounds were determined. Colonization of roots of H. tuberosus D19 by Pseudomonas sp. 262 was evaluated using confocal laser scanning microscopy. Pseudomonas sp. 228, Serratia sp. 246 and Pseudomonas sp. 262 significantly enhanced growth of H. tuberosus D19 exposed to Cd or Zn. Pseudomonas sp. 228 significantly increased Cd concentrations in roots. Serratia sp. 246, and Pseudomonas sp. 256 and 228 resulted in significantly decreased contents of TBA reactive compounds in roots of Zn exposed D19 plants. Growth improvement and decrease of metal-induced stress were more pronounced in D19 than in VR. Pseudomonas sp. 262 - green fluorescent protein (GFP) colonized the root epidermis/exodermis and also inside root hairs, indicating that an endophytic interaction was established. H. tuberosus D19 inoculated with Pseudomonas sp. 228, Serratia sp. 246 and Pseudomonas sp. 262 holds promise for sustainable biomass production in combination with phytoremediation on Cd and Zn contaminated soils., Competing Interests: The authors declare no conflict of interest. Plants on contaminated mining soils often show a reduced growth due to nutrient depletion as well as trace elements (TEs) toxicity. Since those conditions threat plant's survival, plant growth-promoting rhizobacteria (PGPRs), such as rhizobia, might be of crucial importance for plant colonization on TE-contaminated soils. Native rhizobia from mining soils are promising candidates for bioaugmented phytoremediation of those soils as they are adapted to the specific conditions. In this work, rhizobia from Zn- and Cd-contaminated mining soils were in vitro screened for their PGP features [organic acids, indole-3-acetic acid (IAA), and siderophore (SID) production; 1-aminocyclopropane-1-carboxylate (ACC) deaminase activity; and Ca 3 (PO 4 ) 2 solubilization] and Zn and Cd tolerance. In addition, some type and reference rhizobia strains were included in the study as well. The in vitro screening indicated that rhizobia and other native genera have great potential for phytoremediation purposes, by exerting, besides biological N 2 fixation, other plant growth-promoting traits. Leucaena leucocephala-Mesorhizobium sp. (UFLA 01-765) showed multielement tolerance and an efficient symbiosis on contaminated soil, decreasing the activities of antioxidative enzymes in shoots. This symbiosis is a promising combination for phytostabilization.

The interaction between plant growth-promoting bacteria (PGPB) and plants can enhance biomass production and metal tolerance of the host plants. This work aimed at isolating and characterizing the cultivable bacterial community associated with Brassica napus growing on a Zn-contaminated site, for selecting cultivable PGPB that might enhance biomass production and metal tolerance of energy crops. The effects of some of these bacterial strains on root growth of B. napus exposed to increasing Zn and Cd concentrations were assessed. A total of 426 morphologically different bacterial strains were isolated from the soil, the rhizosphere, and the roots and stems of B. napus. The diversity of the isolated bacterial populations was similar in rhizosphere and roots, but lower in soil and stem compartments. Burkoholderia, Alcaligenes, Agrococcus, Polaromonas, Stenotrophomonas, Serratia, Microbacterium, and Caulobacter were found as root endophytes exclusively. The inoculation of seeds with Pseudomonas sp. strains 228 and 256, and Serratia sp. strain 246 facilitated the root development of B. napus at 1,000 µM Zn. Arthrobacter sp. strain 222, Serratia sp. strain 246, and Pseudomonas sp. 228 and 262 increased the root length at 300 µM Cd.

The impact of metal pollution on plant communities has been studied extensively in the past, but little is known about the effects of metal pollution on fungal communities that occur in metal-polluted soils. Metal-tolerant ecotypes of the ectomycorrhizal fungus Suillus luteus are frequently found in pioneer pine forests in the Campine region in Belgium on metal-polluted soils. We hypothesized that metal pollution would play an important role in shaping below-ground fungal communities that occur in these soils and that Suillus luteus would be a dominant player. To test these hypotheses, the fungal communities in a young pine plantation in soil polluted with zinc, and cadmium were studied using 454 amplicon pyrosequencing. Results show that zinc, cadmium and soil organic matter content were strongly correlated with the fungal community composition, but no effects on fungal diversity were observed. As hypothesized, S. luteus was found to be a dominant member of the studied fungal communities. However, other dominant fungal species, such as Sistotrema sp., Wilcoxina mikolae and Cadophora finlandica were found as well. On Zn-polluted soils, populations of the ectomycorrhizal basidiomycete Suillus bovinus exhibit an elevated Zn tolerance when compared to populations on non-polluted sites. To elucidate the mechanism of Zn tolerance, the time-course of Zn uptake was studied in isolates with contrasting Zn tolerance. Unidirectional fluxes and subcellular compartmentation of Zn were investigated through radiotracer flux analyses. Fluorescence imaging was used to support the subcellular Zn compartmentation. After 2 h of exposure to 200 μM Zn, significantly more Zn was accumulated in Zn-sensitive isolates compared to tolerant isolates, despite similar short-term uptake kinetics and similar extracellular Zn sequestration in cell walls. In Zn-sensitive isolates twice as much Zn accumulated in the cytoplasm and 12 times more Zn in the vacuole. (65)Zn efflux analyses revealed a considerably faster Zn export in the Zn-tolerant isolate. The adaptive Zn tolerance in S. bovinus is therefore achieved by a preferential removal of Zn out of the cytoplasm, back into the apoplast, instead of the usual transfer of Zn into the vacuole. Zn exclusion in the fungal symbiont eventually contributes to a lower Zn influx in host plants.

Short rotation coppice (SRC) of willow and poplar might be a promising phytoremediation option since it uses fast growing, high biomass producing tree species with often a sufficient metal uptake. This study evaluates growth, metal uptake and extraction potentials of eight willow clones (Belders, Belgisch Rood, Christina, Inger, Jorr, Loden, Tora and Zwarte Driebast) on a metal-contaminated agricultural soil, with total cadmium (Cd) and zinc (Zn) concentrations of 6.5 +/- 0.8 and 377 +/- 69 mg kg(-1) soil, respectively. Although, during the first cycle, on average generally low productivity levels (3.7 ton DM (dry matter) ha(-1) y(-1)) were obtained on this sandy soil, certain clones exhibited quite acceptable productivity levels (e.g. Zwarte Driebast 12.5 ton DM ha(-1) y(-1)). Even at low biomass productivity levels, SRC of willow showed promising removal potentials of 72 g Cd and 2.0 kg Zn ha(-1) y(-1), which is much higher than e.g. energy maize or rapeseed grown on the same soil Cd and Zn removal can be increased by 40% if leaves are harvested as well. Nevertheless, nowadays the wood price remains the most critical factor in order to implement SRC as an acceptable, economically feasible alternative crop on metal-contaminated agricultural soils.

Adaptive Zn and Cd tolerance have evolved in populations of the ectomycorrhizal fungus Suillus luteus. When exposed to high concentrations of both metals in vitro, a one-sided antagonism was apparent in the Zn- and Cd-tolerant isolates. Addition of high Zn concentrations restored growth of Cd-stressed isolates, but not vice versa. The antagonistic effect was not detected in a S. luteus isolate from non-contaminated land and in Paxillus involutus. The fungi were inoculated on pine seedlings and subsequently exposed to ecologically relevant Zn and Cd concentrations in single and mixed treatments. The applied doses severely reduced nutrient acquisition of non-mycorrhizal pines and pines inoculated with metal-sensitive S. luteus. Highest translocation of Zn and Cd to shoots occurred in the same plants. Seedlings inoculated with fungi collected from the polluted site reduced metal transfer to their host and maintained nutrient acquisition under high metal exposure. The isolate showing highest tolerance in vitro also offered best protection in symbiosis. The antagonistic effect of high Zn on Cd toxicity was confirmed in the plant experiment. Scots pine (Pinus sylvestris L.) seedlings inoculated or not (NM) by a Zn-sensitive or a Zn-tolerant isolate of the ectomycorrhizal fungus Suillus bovinus (L. Fr.) Roussel were exposed to 0.1 or 150 muM Zn(2+) for 9 months. We hypothesized that inoculation with a Zn-tolerant S. bovinus isolate should result in added Zn resistance of the host plant. Plant and fungal growth as well as nutrient profiles and photosynthetic pigments in pine needles were quantified. In NM plants and in plants colonized by the Zn-sensitive isolate, plant growth, N, P, Mg and Fe assimilation were strongly inhibited under Zn stress and concurred with significantly reduced chlorophyll concentrations. In contrast, plants colonized by the Zn-tolerant isolate grew much better and remained physiologically healthier when exposed to elevated Zn. These results provide further evidence for the important role metal-adapted mycorrhizal fungi play as an effective biological barrier against metal toxicity in trees.

Zn pollution has triggered evolution for adaptive Zn tolerance in populations of Suilloid ectomycorrhizal fungi. The objectives of this study were to determine differential physiological responses that are linked to the Zn tolerance trait and to obtain more insight in the general mechanism responsible for the differential growth in Zn-enriched medium. Therefore, we identified intrinsic growth rates and element profiles in Zn-sensitive and Zn-tolerant genotypes. Isolates from Zn-polluted and unpolluted sites were exposed in vitro to increasing Zn(2+) stress. The Zn concentration which inhibits growth by 50% (EC(50)) was determined, and element (Zn, Fe, Mn, Cu, Mg, Ca and P) profiles in the mycelia were analysed. The intraspecific variation in growth rate and nutrient content of the in vitro grown mycelia is great and was not reduced in Zn-tolerant populations. The Zn resistance was not correlated to the intrinsic mycelial growth rate of the isolates or to the concentrations of the elements analysed, except for Zn. At low external Zn, Zn-resistant genotypes had lower Zn concentrations than sensitive isolates. At high external Zn, the differential Zn accumulation pattern between resistant and sensitive isolates became very prominent. Zn-exclusion mechanisms are most likely involved in the naturally selected adaptive Zn resistance. Other mechanisms of Zn detoxification such as sequestration of Zn on cell wall compounds or intracellular chelation and/or compartmentation are probably active but cannot explain the differential Zn sensitivity of the isolates.

The atmospheric pollution history of a former Belgian Zn-smelter complex is preserved in organic sediments of a nearby peat bog pool. The stratigraphy of trace metals, Pb-isotope ratios and mineralogy indicate extreme heavy metal pollution in recent sediments. In the pollutant trend, extremes coincide with maxima in 20th century metal production, minima during major war conflicts and the final shutdown of the smelter. Peak pollution concentrations measure up to 4.7 wt.% Zn, 1.1 wt.% Pb and 0.1 wt.% Cd, which correspond to calculated atmospheric deposition rates of 9.0, 1.6 and 0.16 g m(-2) yr(-1), respectively. 206Pb/207Pb isotope ratios show higher values in the polluted interval (1.135-1.162) relative to local geogenic values deeper down-core (1.194-1.215). Within the polluted interval, three significantly different 208Pb/206Pb plateaus are recognized and suggested to indicate changes in the origins of processed ores. Microprobe analyses on sediment thin sections show extensive in situ FeS2 and ZnS precipitation, which suggests that anoxic processes are responsible for the immobilization of the atmospheric metal inputs. The occurrence of oxidized smelter dusts in an independent surface soil sample indicates a rapid diagenetic transformation of metal oxides into sulfides. Morphology and chemical characteristics allow the distinction between smelter related and diagenetic mineral deposits, and give evidence for dust from open-air ore repositories, as well as smelter operation without dust filters.

Phytoextraction could be a potential management option for diffusely Cd-Zn-Pb-polluted agricultural land in Northeast Belgium. The use of high yielding crops with a sufficiently high metal accumulation is preferred as these are expected to both gradually decontaminate the soil while generating an income through biomass valorization. To find out which high biomass crop possessed the highest and most constant (in time) phytoextraction potential on these soils, different plant species and different mutants or clones of each species, were evaluated during consecutive years. Biomass production and metal accumulation of pre-selected tobacco somaclonal variants (Nicotiana tabacum L.) and pre-selected sunflower mutants (Helianthus annuus L.) were investigated for two productivity years, while the phytoextraction potential of experimental poplar (Populus) and willow (Salix) in short rotation coppice (SRC) was assessed at the end of the second cutting cycle (after two times four growing seasons). The tobacco clones and the sunflower mutants showed efficient extraction of, respectively, Cd and Zn, while the highest simultaneous extractions of Cd and Zn were gained with some SRC clones. Variation in biomass production and metal accumulation were high for all crops over the years. The highest biomass production was observed for the experimental poplar clone of the crossing type Populus deltoides (P. maximowiczii x P. trichocarpa) with 9.9 ton DW per ha per year. The remediation period to reach legal threshold values for the pseudo-total content of Cd in this specific soil was estimated to be at least 60 years. Combining estimated phytoextraction potential and economic and environmental aspects, the SRC option is proposed as the most suitable crop for implementing metal phytoextraction in the investigated area. Short rotation coppice (SRC) of willow and poplar is proposed for economic valorization and concurrently as remediation strategy for metal contaminated land in northeast-Belgium. However, metal phytoextraction appears insufficient to effectuate rapid reduction of soil metal contents. To increase both biomass production and metal accumulation of SRC, two strategies are proposed: (i) in situ selection of the best performing clones and (ii) bioaugmentation of these clones with beneficial plant-associated bacteria. Based on field data, two experimental willow clones, aSalix viminalisand aSalix alba x albaclone, were selected. Compared to the best performing commercial clones, considerable increases in stem metal extraction were achieved (up to 74% for Cd and 91% for Zn). From the selected clones, plant-associated bacteria were isolated and identified. All strains were subsequently screened for their plant growth-promoting and metal uptake enhancing traits. Five strains were selected for a greenhouse inoculation experiment with the selected clones planted in Cd-Zn-Pb contaminated soil. Extraction potential tended to increase after inoculationof S. viminalisplants with aRahnella sp.strain due to a significantly increased twig biomass. Background and Scope Plant responses to the toxic effects of soil contaminants, such as excess metals or organic substances, have been studied mainly at physiological, biochemical and molecular levels, but the influence on root system architecture has received little attention. Nevertheless, the precise position, morphology and extent of roots can influence contaminant uptake. Here, data are discussed that aim to increase the molecular and ecological understanding of the influence of contaminants on root system architecture. Furthermore, the potential of plant-associated bacteria to influence root growth by their growth-promoting and stress-relieving capacities is explored. Methods Root growth parameters of Arabidopsis thaliana seedlings grown in vertical agar plates are quantified. Mutants are used in a reverse genetics approach to identify molecular components underlying quantitative changes in root architecture after exposure to excess cadmium, copper or zinc. Plant-associated bacteria are isolated from contaminated environments, genotypically and phenotypically characterized, and used to test plant root growth improvement in the presence of contaminants. Key Results The molecular determinants of primary root growth inhibition and effects on lateral root density by cadmium were identified. A vertical split-root system revealed local effects of cadmium and copper on root development. However, systemic effects of zinc exposure on root growth reduced both the avoidance of contaminated areas and colonization of non-contaminated areas. The potential for growth promotion and contaminant degradation of plant-associated bacteria was demonstrated by improved root growth of inoculated plants exposed to 2,4-di-nitro-toluene (DNT) or cadmium. Conclusions Knowledge concerning the specific influence of different contaminants on root system architecture and the molecular mechanisms by which this is achieved can be combined with the exploitation of plant-associated bacteria to influence root development and increase plant stress tolerance, which should lead to more optimal root systems for application in phytoremediation or safer biomass production. Seed endophyte inoculation can enhance the plant tolerance to pollutants, which allows plant cultivation on trace element (TE) polluted soils. Methylobacterium sp. CP3 and Kineococcus endophyticus CP19 were tested in vitro for their tolerance to Zn and Cd and their plant growth promotion traits. The in vivo effects of bioaugmentation with individual strains or both strains were tested using two poplar cultivars, Populus deltoides x (P. trichocarpa x P. maximowiczii) 'Dender' and 'Marke', grown in TE polluted soil for six weeks. Methylobacterium sp. was found to grow on media enriched with 0.4 and 0.8 mM Cd, and both endophytes tolerated 0.6 and 1 mM Zn, due to the presence of genes involved in Zn and Cd tolerance and transport. Methylobacterium sp. showed an extracellular ion sequestration mechanism. Production of indole-3-acetic acid by Methylobacterium sp. and K. endophyticus, as well as phosphorus solubilization by Methylobacterium sp. were observed. Bioaugmentation with both endophytes increased the shoot length of Populus 'Marke' and enhanced the Mg uptake in both cultivars. Inoculation with Methylobacterium sp. reduced the bioaccumulation of Zn in 'Marke', conferring it an excluder strategy. Woody cuttings from two Salix genotypes (genotype I - clone LUC-31, Salix alba and genotype II- clone STOTT, Salix viminalis) were grown hydroponically for 14 days at increasing concentrations of Zn: control, 50, 100 or 150 μ M Zn. Genotype tolerance to excess zinc (Zn) was evaluated using a root elongation test. The changes in growth, Zn, iron (Fe), copper (Cu) and manganese (Mn) concentrations as well as photosynthetic performance were used as additional evaluation criteria. Photosynthetic pigments concentrations in Zn-exposed cuttings of genotype II decreased as compared with genotype I, which corresponded well with the higher leaf Zn accumulation, decreased Fe concentrations as well as lowered photosynthetic rate. Pb-210 dating of two metal-polluted organic sedimentcores obtained near a former pyrometallurgical zinc smelter in Lommel, Belgiumhave been used to reconstruct atmospheric lead deposition rates during the20th century. Independent knowledge concerning historical pollutionevents and 137Cs fall-out profiles has allowed a criticalevaluation of the CRS, CIC and CF-CS models for the 210Pb ageinterpretation. Resulting ages for the three models suggest that, in this case,the CIC model gives the most accurate interpretation of historical pollutionevents and atmospheric lead fall-out. The 210Pbwater-sediment flux was estimated at 141–1158Bq·m−2·yr−1 for one site and62–106 Bq·m−2·yr−1 at theother site, during the last century. The large difference illustrates thatsediment focusing was important on a small spatial scale (10 m).The direction of focusing correlates with the predominant wind direction.Maximum atmospheric lead deposition rates were found to be 1.63 ± 0.59g·m−2·yr−1 around 1968 AD,which is 2 orders of magnitude larger than the Belgian average in 1980 AD, and5 orders larger than Holocene atmospheric lead deposition. Abstract: Treatment and/or disposal of metal contaminated biomass are still an unsolved problem. Knowledge of the metal distribution is of prime importance concerning the application of pyrolysis product streams. Sorghum bicolor (L.) Moench was cultivated in a semi-hydroponic system to assess its potential use in phytoremediation and biomass production. Plants were grown in a greenhouse using perlite as substrate, half-strength Hoagland's solution as control (CTR) and the same solution supplemented with a mixture of Ni and Zn (CTM; 10 g m−3 each). Shoot and root biomass were determined and analyzed for their metals content. Flash and slow pyrolysis were performed on S. bicolor shoots at 450 °C. Biomass and pyrolysis products were analyzed focusing on metal distribution. Mass and energy balances were determined. S. bicolor delivered good shoot biomass with relatively moderate concentrations of Ni and Zn. Metal concentrations in the pyrolysis oils were below detection limits and almost all metals accumulated in the char. In fact, 99% of Ni and 98% of Zn were recovered in the char when a slow pyrolysis process was applied, while in flash pyrolysis conditions both metals were found back in the char and in the heating transfer medium. Furthermore, the percentages of char and oil were higher in slow pyrolysis compared to flash pyrolysis. Energy recovery in the char from slow pyrolysis was higher than flash pyrolysis. Flash and slow pyrolysis can likely offer a valuable processing method for metal contaminated biomass, thus limiting the waste disposal problem associated with phytoremediation of metals.
