Bio-adhesives used clinically, commonly have the ability to fill surgical voids and support wound healing, but which are devoid of antibacterial activity, and thus, could not meet the particular needs of the infected wound site. Herein, a series of natural polyphenolic antibacterial bio-adhesives were prepared via simple mixing and heating of polyphenols and acid anhydrides without any solvent or catalyst. Upon the acid anhydride ring opening and acylation reactions, various natural polyphenolic bio-adhesives could adhere to various substrates ( i.e. , tissue, wood, glass, rubber, paper, plastic, and metal) based on multi-interactions. Moreover, these bio-adhesives showed excellent antibacterial and anti-infection activity, rapid hemostatic performance and appropriate biodegradability, which could be widely used in promoting bacterial infection wound healing and hot burn infection wound repair. This work could provide a new strategy for strong adhesives using naturally occurring molecules, and provide a method for the preparation of novel multifunctional wound dressings for infected wound healing.

Background: During wound healing, the overproduction of reactive oxygen species (ROS) can break the cellular oxidant/antioxidant balance, which prolongs healing. The wound dressings targeting the mitigation of ROS will be of great advantages for the wound healing. puerarin (PUE) and ferulic acid (FA) are natural compounds derived from herbs that exhibit multiple pharmacological activities, such as antioxidant and anti-inflammatory effects. Polydopamine (PDA) is made from natural dopamine and shows excellent antioxidant function. Therefore, the combination of natural antioxidants into hydrogel dressing is a promising therapy for wound healing., Results: Hydrogel wound dressings have been developed by incorporating PUE or FA via PDA nanoparticles (NPs) into polyethylene glycol diacrylate (PEG-DA) hydrogel. This hydrogel can load natural antioxidant drugs and retain the drug in the gel network for a long period due to the presence of PDA NPs. Under oxidative stress, this hydrogel can improve the activity of superoxide dismutase and glutathione peroxidase and reduce the levels of ROS and malondialdehyde, thus preventing oxidative damage to cells, and then promoting wound healing, tissue regeneration, and collagen accumulation., Conclusion: Overall, this triple antioxidant hydrogel accelerates wound healing by alleviating oxidative injury. Our study thus provides a new way about co-delivery of multiple antioxidant natural molecules from herbs via antioxidant nanoparticles for wound healing and skin regeneration., (© 2021. Shape amphiphiles refer to a class of molecular conjugates composed by distinct building blocks with incommensurate shapes and chemical interactions. Whilst extensive efforts have been devoted to the solid state self-assembly and nanolithography applications of shape amphiphiles, only very limited examples of their usage in the biomedical field have been explored. In this work, we reported a series of gallic acid functionalized polyhedral oligomeric silsesquioxane (POSS)-based shape amphiphiles with various macromolecular architectures for efficient and accelerated wound healing. These antioxidant giant amphiphiles demonstrated good biocompatibility and strong free radical scavenging activity to prevent cell oxidative damage and then promote wound recovery. We describe these POSS-based antioxidant shape amphiphiles in an initial proof-of-concept study and propose that this family of functional giant amphiphiles will be useful in regenerative biomedicine.

Although smart bio‐glues have been well documented, the development of internal bio‐glues for non‐invasive or minimally invasive surgery is still met with profound challenges such as safety risk and the lack of deep tissue penetration stimuli for internal usage. Herein, a series of smart internal bio‐glues are developed via the integration of o‐nitrobenzene modified biopolymers with up‐conversion nanoparticles (UCNPs). Upon irradiation by near‐infrared (NIR) light, the prepared smart bio‐glues can undergo a gelation process, which may further induce strong adhesion between tissues under both dry and wet conditions based on multi‐interactions. Moreover, those NIR light‐responsive bio‐glues with deeper tissue penetration ability demonstrate good biocompatibility, excellent hemostatic performance, and the potent ability to accelerate wound healing for both external and internal wounds. Wound healing dressing is increasingly needed in clinical owing to the large quantity of skin damage annually. Excessive reactive oxygen species (ROS) produced through internal or external environmental influences can lead to lipid peroxidation, protein denaturation, and even DNA damage, and ultimately have harmful effects on cells. Aiming to sufficiently contact with the wound microenvironment and scavenge ROS, superabsorbent poly (acrylic acid) and antioxidant poly (ester amide) (PAA/PEA) hybrid hydrogel has been developed to enhance wound healing. The physical and chemical properties of hybrid hydrogels were studied by Fourier-transform infrared (FTIR) absorption spectrum, compression, swelling, degradation, etc. Besides, the antioxidant properties of hybrid hydrogels can be investigated through the free radical scavenging experiment, and corresponding antioxidant indicators have been tested at the cellular level. Hybrid hydrogel scaffolds supported the proliferation of human umbilical vein endothelial cells and fibroblasts, as well as accelerated angiogenesis and skin regeneration in wounds. The healing properties of wounds in vivo were further assessed on mouse skin wounds. Results showed that PAA/PEA hybrid hydrogel scaffolds significantly accelerated the wound healing process through enhancing granulation formation and re-epithelialization. In summary, these superabsorbent and antioxidative hybrid hydrogels could be served as an excellent wound dressing for full-thickness wound healing. [ABSTRACT FROM AUTHOR]

Abstract: The objective of this work was to prepare chitosan/silk fibroin (CS/SF) blending membranes crosslinked with alginate dialdehyde (ADA) as wound dressings and to evaluate the physical properties and biocompatibility of the membranes. The morphology of membrane was observed by scanning electron microscopy (SEM) which showed that the well consistency of these two compositions. Further, the stability, water absorption and water vapor permeability of the ADA fixed CS/SF membranes could meet the needs of wound dressing. Furthermore, the biocompatibility of ADA fixed membranes was investigated by MTT assays and SEM in vitro, and the membranes were found to promote the cell attachment and proliferation. These results suggest that ADA fixed CS/SF blending membranes with a suitable ratio could be a promising candidate for wound healing applications. [Copyright &y& Elsevier]

Although bio‐adhesives have achieved many progresses, creating robust bio‐glues that could withstand the complex physiological microenvironment remains a significant challenge such as limited bioadhesion strength and stability, as well as slow adhesion speed caused by pH, enzymes, salt, and bacteria. Herein, a series of strong bio‐glues are successfully fabricated using a polyphenol–acrylamide–ruthenium crosslinking (PARC) reaction under visible light irradiation, involving acrylamides radical polymerization, polyphenols (extracts) coupling reaction, and ruthenium‐mediated photochemistry. The resulting bio‐glues underwent rapid sol–gel transition and exhibited adhesive properties within a short of time (≈5 s), which could quickly bond to various bio‐tissues. Moreover, these bioadhesives demonstrate high adhesion strength and stability with mucosal tissue in the oral environment, effectively promoting rat and pig oral mucosal wound healing. It is anticipated that this class of visible light‐responsive polyphenolic bio‐glues could serve as a robust tool for addressing oral mucosal defects in the clinic, offering a promising alternative to traditional treatments for oral mucosal disorders. • Reduction of polydopamine nanoparticles were used as a simple strategy to improve the antioxidative activity. • Reduction of polydopamine nanoparticles incorporated polysaccharide hydrogels have been fabricated. • The hydrogels possess enhanced antioxidant property and contact antibacterial activity. • These hydrogels can accelerate the wound healing process obviously. Hydrogels with antioxidative and antibacterial properties have emerged as potential dressings for accelerated wound healing. Herein, a series of reduced polydopamine nanoparticles (rPDA NPs) incorporated oxidized dextran/chitosan hybrid hydrogels have been designed for wound healing due to their excellent antioxidative property and antibacterial activity. The physicochemical properties as well as the antioxidative activities of the hydrogels were carefully characterized. The results demonstrated rPDA NPs have better antioxidative activity than the untreated PDA NPs. And the rPDA NPs incorporated oxidized dextran/chitosan hybrid hydrogels had excellent antioxidative properties to protect cells against external oxidative stress. Besides, the hydrogels also showed antibacterial ability to protect the wound against infections. In vitro and in vivo investigations concluded that rPDA NPs incorporated oxidized dextran/chitosan hybrid hydrogels could be served as an effective dressing for accelerated wound healing. • Enhance antioxidant activity of hydrogel is a promising way for wound healing. • The well-designed hybrid hydrogel is natural and simple. • Arginine derivatives enhance the dopamine antioxidation for hyaluronic acid hydrogel. Scaffolds with antioxidant capability have attracted considerable interests for wound healing. For enhanced wound healing, new antioxidant materials are still highly needed. In this study, new antioxidant materials, arginine derivatives (AD), were introduced into dopamine-functionalized hyaluronic acid (HA-DA) to develop novel hydrogels (HA-DA/AD) with enhanced antioxidant activity. The HA-DA/AD hydrogels showed greater DPPH and hydroxyl radical scavenging rates than the HA-DA hydrogel. Moreover, the HA-DA/AD hydrogels exhibited significantly better protection for cells against external oxidative stress (decreased ROS and MDA levels and increased SOD and GPx enzyme activity) and better wound healing (enhanced VEGF and CD 31 expression and enhanced tissue remodeling).
