RPM1 is a CC-NBS-LRR protein that was first shown to be required for resistance to Pseudomonas syringae pv. maculicola in Arabidopsis thaliana. Our previous study showed that TaRPM1 gene in wheat was upregulated about six times following infection by Puccinia striiformis f. sp. tritici (Pst) under high temperature, compared with normal temperature. To study the function of TaRPM1 in wheat high-temperature seedling-plant (HTSP) resistance to Pst , the full length of TaRPM1 was cloned, with three copies each located on chromosomes 1A, 1B, and 1D. Transient expression of the TaRPM1-GFP fusion protein in Nicotiana benthamiana indicated that TaRPM1 localizes in the cytoplasm and nucleus. Profiling TaRPM1 expression indicated that TaRPM1 transcription was rapidly upregulated upon Pst inoculation under high temperature. In addition, TaRPM1 was induced by exogenous salicylic acid hormone application. Silencing TaRPM1 in wheat cultivar Xiaoyan 6 (XY 6) resulted in reduced HTSP resistance to Pst in terms of reduced number of necrotic cells and increased uredinial length, whereas no obvious phenotypic changes were observed in TaRPM1 -silenced leaves under normal temperature. Related defense genes TaPR1 and TaPR2 were downregulated in TaRPM1 -silenced plants under high temperature. We conclude that TaRPM1 is involved in HTSP resistance to Pst in XY 6. Stripe rust, caused by Puccinia striiformis f. sp. tritici (Pst), is a destructive disease of wheat worldwide. The disease is preferably controlled by growing resistant cultivars. Wheat cultivar Xiaoyan 6 (XY 6) has been resistant to stripe rust since its release. In the previous studies, XY 6 was found to have higher-temperature seedling-plant (HTSP) resistance. However, the molecular mechanisms of HTSP resistance were not clear. To identify differentially expressed genes (DEGs) involved in HTSP resistance, we sequenced 30 cDNA libraries constructed from XY 6 seedlings exposed to several temperature treatments. Compared to the constant normal (15°C) and higher (20°C) temperature treatments, 1395 DEGs were identified in seedlings exposed to 20°C for 24 h (to activate HTSP resistance) and then kept at 15°C. These DEGs were located on all 21 chromosomes, with 29.2%on A, 41.1%on B and 29.7%on D genomes, by mapping to the Chinese Spring wheat genome. The 1395 DEGs were enriched in ribosome, plant-pathogen interaction and glycerolipid metabolism pathways, and some of them were identified as hub proteins (phosphatase 2C10), resistance protein homologs, WRKY transcription factors and protein kinases. The majority of these genes were up-regulated in HTSP resistance. Based on the differential expression, we found that phosphatase 2C10 and LRR receptor-like serine/threonine protein kinases are particularly interesting as theymay be important for HTSP resistance through interactingwith different resistance proteins, leading to a hypersensitive response. WRKY transcription factors (TFs) play crucial roles in plant resistance responses to pathogens. Wheat stripe rust, caused by the fungal pathogen Puccinia striiformis f. sp. tritici (Pst), is a destructive disease of wheat (Triticum aestivum) worldwide. In this study, the two WRKY genes TaWRKY49 and TaWRKY62 were originally identified in association with high-temperature seedling-plant resistance to Pst (HTSP) resistance in wheat cultivar Xiaoyan 6 by RNA-seq. Interestingly, the expression levels of TaWRKY49 and TaWRKY62 were down- and up-regulated, respectively, during HTSP resistance in response to Pst. Silencing of TaWRKY49 enhanced whereas silencing TaWRKY62 reduced HTSP resistance. The enhanced resistance observed on leaves following the silencing of TaWRKY49 was coupled with increased expression of salicylic acid (SA)- and jasmonic acid (JA)-responsive genes TaPR1.1 and TaAOS, as well as reactive oxygen species (ROS)-associated genes TaCAT and TaPOD; whereas the ethylene (ET)-responsive gene TaPIE1 was suppressed. The decreased resistance observed on leaves following TaWRKY62 silencing was associated with increased expression of TaPR1.1 and TaPOD, and suppression of TaAOS and TaPIE1. Furthermore, SA, ET, MeJA (methyl jasmonate), hydrogen peroxide (H2O2) and abscisic acid (ABA) treatments increased TaWRKY62 expression. On the other hand, MeJA did not affect the expression of TaWRKY49, and H2O2 reduced TaWRKY49 expression. In conclusion, TaWRKY49 negatively regulates while TaWRKY62 positively regulates wheat HTSP resistance to Pst by differential regulation of SA-, JA-, ET and ROS-mediated signaling. Stripe rust, caused by Puccinia striiformis f. sp. tritici (Pst), is a devastating disease of wheat (Triticum aestivum) worldwide. Wheat high-temperature seedling plant (HTSP) resistance to Pst is non-race-specific and durable. WRKY transcription factors have been proven to play important roles in plant defence responses to attacks by several pathogens. However, there is no direct evidence as to whether WRKY transcription factors play a role in HTSP resistance to Pst. We isolated a WRKY gene, named TaWRKY70, from wheat cultivar Xiaoyan 6. The expression level of TaWRKY70 was increased significantly when exposed to high temperatures (HTs) during the initial symptom expression stage of Pst infection. The expression of this gene increased in plants treated with ethylene (ET), salicylic acid (SA) and cold (4°) stresses, but decreased in plants treated with methyl jasmonate (MeJA) and heat (40°) stresses. Silencing of TaWRKY70 led to greater susceptibility to Pst (in terms of the increase in length of uredinial pustules and the decrease in the number of necrotic cells) compared with non-silenced plants when exposed to HT during the initial symptom expression stage of Pst infection, coinciding with expression changes of the ET- and SA-responsive genes TaPIE1 and TaPR1.1. In contrast, the expression level of the jasmonic acid (JA)-responsive gene TaAOS was not affected by TaWRKY70. Both wheat powdery mildew severities and nitrogen input levels can lead to changes in spectral reflectance, but they have been rarely studied simultaneously for their effect on spectral reflectance. To determine the effects and influences of different nitrogen input levels on monitoring wheat powdery mildew and estimating yield by near-ground hyperspectral remote sensing, Canopy hyperspectral reflectance data acquired at Feekes growth stage (GS) 10.5.3, 10.5.4, and 11.1 were used to monitor wheat powdery mildew and estimate grain yield under different nitrogen input levels during the 2016–2017, 2017–2018, 2018–2019 and 2019–2020 seasons. The relationships of powdery mildew and grain yield with vegetation indices (VIs) derived from spectral reflectance data across the visible (VIS) and near-infrared (NIR) regions of the spectrum were studied. The relationships of canopy spectral reflectance or first derivative spectral reflectance with powdery mildew did not differ under different nitrogen input levels. However, the dynamics of VIs differed in their sensitivities to nitrogen input levels, disease severity, grain yield, The area of the red edge peak (Σdr680–760 nm) was a better overall predictor for both disease severity and grain yield through linear regression models. The slope parameter estimates did not differ between the two nitrogen input levels at each GSs. Hyperspectral indices can be used to monitor wheat powdery mildew and estimate grain yield under different nitrogen input levels, but such models are dependent on GS and year, further research is needed to consider how to incorporate the growth stage and year-to-year variation into future applications.
