In aquatic environments the concurrent exposure of molluscs to microplastics (MPs) and estrogens is common, as these pollutants are frequently released by wastewater treatment plants into estuaries. Therefore, this study aimed to evaluate the independent and co-exposure impacts of polyethylene microplastics (PE-MPs) and estrogenic endocrine-disrupting chemicals (EEDCs) at environmentally relevant concentrations on polar metabolites and morphological parameters of the Sydney rock oyster. A seven-day acute exposure revealed no discernible differences in morphology; however, significant variations in polar metabolites were observed across oyster tissues. The altered metabolites were mostly amino acids, carbohydrates and intermediates of the Kreb's cycle. The perturbation of metabolites were tissue and sex-specific. All treatments generally showed an increase of metabolites relative to controls - a possible stimulatory and/or a potential hormetic response. Micro- and nano-plastics (MNPs) are emerging contaminants found in air, water, and food. Ageing and weathering processes convert aquatic plastics into MNPs which, due to their small size, can be assimilated by organisms. The accumulation of MNPs in aquatic life (e.g., fish, oysters, and crabs) will, in turn, pose risks to the health of ecosystems and human. This study focuses on the uptake, biodistribution, and size-dependent toxicity of polystyrene nano-plastics (PS-NPs) in a commercially important food web, the Australian Bass (Macquaria novemaculeata). Fish were fed artemia containing PS-NPs of various sizes (ranging from 50 nm to 1 μm) for durations of 5 and 7 days. The findings revealed that smaller NPs (50 nm) accumulated in the brain and muscle tissues at higher concentrations, whereas larger NPs (1 μm) were primarily found in the gills and intestines. In addition, an inverse correlation was observed between the size of NPs and the rate of trophic transfer, with smaller PS-NPs resulting in a higher transfer rate from artemia to fish. Polystyrene NPs caused both activation of the enzyme superoxide dismutase and damage to the DNA of fish tissues. These effects were size dependent. Metabolomic analysis revealed that indirect exposure to different-sized PS-NPs resulted in altered metabolic profiles within fish intestines, potentially impacting lipid and energy metabolism. Microplastics (MPs) exposed to the natural environment provide an ideal surface for biofilm formation, which potentially acts as a reactive phase facilitating the sorption of hazardous contaminants. Until now, changes in the contaminant sorption capacity of MPs due to biofilm formation have not been quantified. This is the first study that compared the capacity of naturally aged, biofilm-covered microplastic fibers (BMFs) to adsorb perfluorooctane sulfonate (PFOS) and lead (Pb) at environmentally relevant concentrations. Changes in the surface properties and morphology of aged microplastic fibers (MF) were studied by surface area analysis, infrared spectroscopy, and scanning electron microscopy. Results revealed that aged MFs exhibited higher surface areas because of biomass accumulation compared to virgin samples and followed the order polypropylene>polyethylene>nylon>polyester. The concentrations of adsorbed Pb and PFOS were 4-25% and 20-85% higher in aged MFs and varied among the polymer types. The increased contaminant adsorption was linked with the altered surface area and the hydrophobic/hydrophilic characteristics of the samples. Overall, the present study demonstrates that biofilms play a decisive role in contaminant-plastic interactions and significantly enhance the vector potential of MFs for toxic environmental contaminants. We anticipate that knowledge generated from this study will help refine the planetary risk assessment of MPs.

The long-term aging of plastic leads to weathering and biofouling that can influence the behavior and fate of plastic in the marine environment. This is the first study to fingerprint the contaminant profiles and bacterial communities present in plastic-associated inorganic and organic matter (PIOM) isolated from 10 year-aged plastic. Plastic sleeves were sampled from an oyster aquaculture farm and the PIOM was isolated from the intertidal, subtidal, and sediment-buried segments to investigate the levels of metal(loid)s, polyaromatic hydrocarbons (PAHs), per-fluoroalkyl substances (PFAS) and explore the microbial community composition. Results indicated that the PIOM present on long-term aged high-density polyethylene plastic harbored high concentrations of metal(loid)s, PAHs, and PFAS. Metagenomic analysis revealed that the bacterial composition in the PIOM differed by habitat type, which consisted of potentially pathogenic taxa including Vibrio , Shewanella , and Psychrobacter . This study provides new insights into PIOM as a potential sink for hazardous environmental contaminants and its role in enhancing the vector potential of plastic. Therefore, we recommend the inclusion of PIOM analysis in current biomonitoring regimes and that plastics be used with caution in aquaculture settings to safeguard valuable food resources, particularly in areas of point-source contamination.

Besides the ecotoxicological consequences of microplastics and associated chemicals, the association of microbes on plastics has greater environmental implications as microplastics may select for unique microbiome participating in environmentally significant functions. Despite this, the functional potential of the microbiome associated with different types of plastics is understudied. Here, we investigate the interaction between plastic and marine biofilm-forming microorganisms through a whole-genome sequencing approach on four types of microplastics incubated in the marine environment. Taxonomic analysis suggested that the microplastic surfaces exhibit unique microbial profiles and niche partitioning among the substrates. In particular, the abundance of Vibrio alginolyticus and Vibrio campbellii suggested that microplastic pollution may pose a potential risk to the marine food chain and negatively impact aquaculture industries. Microbial genera involved in xenobiotic compound degradation, carbon cycling, and genes associated with the type IV secretion system, conjugal transfer protein TraG, plant-pathogen interaction, CusA/CzcA family heavy metal efflux transfer proteins, and TolC family proteins were significantly enriched on all the substrates, indicating the variety of processes operated by the plastic-microbiome. The present study gives a detailed characterization of the rapidly altering microbial composition and gene pools on plastics and adds new knowledge surrounding the environmental ramifications of marine plastic pollution.

Increasing our understanding of the bioavailable fractions of polycyclic aromatic compounds (PACs) in an aquatic environment is important for the assessment of the environmental and human health risks posed by PACs. More importantly, the behaviour of polar polycyclic aromatic hydrocarbons (polar PAHs), which are metabolites of legacy PAHs, are yet to be understood. We, therefore, carried out a study involving Sydney rock oysters (Saccostrea glomerata) sourced from two locations, that had been exposed to PAH contamination, within an Australian south-east estuary. Biomonitoring of these oysters, following relocation from the estuary to a relatively isolated waterway, was done at 24 and 72 h after deployment and subsequently at 7, 14, 28, 52 and 86 days. Control samples from Camden Haven River were sampled for PAC analyses just before deployment, after 28 days and at the end of the study (day 86). Lipid-normalised concentrations in oyster tissues across the 86-day sampling duration, elimination rate constants (k 2 ), biological half-lives (t 1/2 ) and time required to reach 95% of steady-state (t 95 ) were reported for parent PAHs and the less-monitored polar PAHs including nitrated/oxygenated/heterocyclic PAHs (NPAHs, oxyPAHs and HPAHs) for the three differently sourced oyster types. Most of the depurating PAHs and NPAHs, as well as 9-FLO (oxyPAH), had k 2 values significantly different from zero (p < 0.05). All other oxyPAHs and HPAHs showed no clear depuration, with their concentrations remaining similar. Improving risk assessment and remediation rests on better understanding of contaminant bioavailability. Despite their strong toxicological attributes, little is known about the partitioning behaviour and bioavailability of polar polycyclic aromatic hydrocarbons (PAHs) in aquatic environments. The present study provides an insight into the bioavailable fractions of polar PAHs and their parent analogues in the tissues of the Sydney rock oyster, Saccostrea glomerata, a model aquatic bio-indicator organism. The concentration and distribution patterns of parent and polar PAHs including oxygenated PAHs (oxyPAHs), nitrated PAHs (NPAHs) and heterocyclic PAHs (HPAHs) were determined in water, sediment and oysters from an ecologically and economically important estuary of New South Wales, Australia. Total concentrations of PAHs, oxyPAHs, NPAHs and HPAHs were higher in sediments compared to oyster tissue and water. For most polar PAHs, total concentrations for water, sediment and oyster samples were <1 μg/g (μg/l for water) while parent PAH concentrations were several orders of magnitude higher. Computed biota-sediment accumulation factors (BSAFs) on lipid-normalized oyster concentrations revealed that while ∑oxyPAHs and ∑HPAHs exhibited low accumulation from sediment to oyster tissues (BSAF <1), ∑PAHs and ∑NPAH were found to be accumulated at high levels (BSAF >1). BSAF individual computation showed that bioaccumulation of nine investigated HPAHs in oyster tissues were relatively low and only 2-EAQ (oxyPAH) and 1N-NAP (NPAH) showed high levels of accumulation in oyster tissues, similar to parent PAHs. To the best of our knowledge, this is the first known study on the bioavailability of polar and non-polar PAHs in an Australian aquatic environment. Great ecological and human health risks may arise from the presence of polycyclic aromatic hydrocarbons (PAHs) in aquatic environments and particularly in sediments, where they often partition. In spite of the apparent risk, knowledge about PAHs and their polar derivatives in sediments is limited. We, therefore, carried out an assessment of the concentrations of parent PAHs and their derivatives (polar PAHs) in sediments of Lake Macquarie: the largest saltwater lake in the southern hemisphere. A total of 31 sediment samples along the pollution prone western shoreline of the estuary were analysed. Multiple source apportionment methods were used to investigate PAH sources contributing to parent and polar PAH concentrations in the estuarine sediments. Concentration levels were highest for high molecular weight (HMW) PAHs compared to low molecular weight (LMW) PAHs. The highest PAH concentrations were recorded for oxygenated PAHs (oxy-PAHs) compared to parent and other polar PAHs. Polycyclic aromatic hydrocarbon diagnostic ratios and compositional analysis showed that PAHs in Lake Macquarie were predominantly pyrogenic exhibiting strong positive correlation (R 2  = 0.972) with total PAH concentrations. Principal Component Analysis (PCA) identified three groupings of PAHs with oxy-PAHs and NPAHs dominating (40.2%). Carbazole, a heterocyclic PAH, was also a prominent contributor to sediment PAH concentrations. Microplastics are an emerging contaminant in aquatic environments. Information on the occurrence and characteristics of microplastics in Australia is limited and their interactions with chemical contaminants have not been addressed. Therefore, the aim of this study was to generate baseline information on the physical and chemical characteristics of microplastics on Australian shorelines to facilitate further detailed risk assessment. Field collected microplastics were categorised by colour, shape and polymer type. Plastic particles were primarily clear, blue, white and green and consisted mainly of fragments (57.80%) and pellets (30.68%). Polymer characterisation revealed that shoreline microplastics were polyethylene (53.17%), polypropylene (35.17%), polystyrene (6.61%) and polyethylene terephthalate (1.85%). Analysis of metal(loid)s found that concentrations of Mn, Cr, Cu, As, Zn and Pb were significantly higher on microplastics associated with industrial locations compared with other land uses, indicating that aged microplastics have the potential to adsorb toxic metals and that metals levels may be location-dependent., Competing Interests: Declaration of competing interest The authors declare that they have no known competing financial interests or personal relationships that could have appeared to influence the work reported in this paper., (Crown Copyright © 2020. Plastic litter has become one of the most serious threats to the marine environment. Over 690 marine species have been impacted by plastic debris with small plastic particles being observed in the digestive tract of organisms from different trophic levels. The physical and chemical properties of microplastics facilitate the sorption of contaminants to the particle surface, serving as a vector of contaminants to organisms following ingestion. Bioaccumulation factors for higher trophic organisms and impacts on wider marine food webs remain unknown. The main objectives of this review were to discuss the factors influencing microplastic ingestion; describe the biological impacts of associated chemical contaminants; highlight evidence for the trophic transfer of microplastics and contaminants within marine food webs and outline the future research priorities to address potential human health concerns. Controlled laboratory studies looking at the effects of microplastics and contaminants on model organisms employ nominal concentrations and consequently have little relevance to the real environment. Few studies have attempted to track the fate of microplastics and mixed contaminants through a complex marine food web using environmentally relevant concentrations to identify the real level of risk. To our knowledge, there has been no attempt to understand the transfer of microplastics and associated contaminants from seafood to humans and the implications for human health. Variable and occasionally high concentrations of cadmium in wild oysters at a remote location with the potential to develop aquaculture enterprises motivated research into the distribution and sources of metals in oysters, seawater, sediment, suspended solids and phytoplankton. Saccostrea mytiloides and Saccostrea mordax contained cadmium concentrations exceeding the food standard maximum level (ML) at three of four sites. At one site with high zinc levels in sediment, oyster cadmium levels were below the ML. Metal levels in seawater were not correlated with cadmium levels in oysters but high cadmium/zinc ratios were measured in Trichodesmium erythraeum blooms. We suggest that oysters accumulate cadmium mainly from annual phytoplankton blooms except at sites where zinc availability is sufficiently high to prevent uptake though a mechanism of antagonistic exclusion. Knowledge of the source and uptake mechanisms of cadmium in oysters should lead to new management strategies to reduce cadmium levels in farmed oysters., (Copyright © 2017 Elsevier Ltd. Previous work suggests that larvae from Sydney rock oysters that have been selectively bred for fast growth and disease resistance are more resilient to the impacts of ocean acidification than nonselected, wild-type oysters. In this study, we used proteomics to investigate the molecular differences between oyster populations in adult Sydney rock oysters and to identify whether these form the basis for observations seen in larvae. Adult oysters from a selective breeding line (B2) and nonselected wild types (WT) were exposed for 4 weeks to elevated pCO2 (856 μatm) before their proteomes were compared to those of oysters held under ambient conditions (375 μatm pCO2 ). Exposure to elevated pCO2 resulted in substantial changes in the proteomes of oysters from both the selectively bred and wild-type populations. When biological functions were assigned, these differential proteins fell into five broad, potentially interrelated categories of subcellular functions, in both oyster populations. These functional categories were energy production, cellular stress responses, the cytoskeleton, protein synthesis and cell signalling. In the wild-type population, proteins were predominantly upregulated. However, unexpectedly, these cellular systems were downregulated in the selectively bred oyster population, indicating cellular dysfunction. We argue that this reflects a trade-off, whereby an adaptive capacity for enhanced mitochondrial energy production in the selectively bred population may help to protect larvae from the effects of elevated CO2 , whilst being deleterious to adult oysters., (© 2015 John Wiley & Sons Ltd.)

The Sydney rock oyster, Saccostrea glomerata, has been demonstrated as a useful biomonitor of estrogenic compounds following laboratory exposures, yet its utility in the assessment of estrogenic exposure and effects under field conditions requires investigation. To achieve this aim, S. glomerata were deployed in Newcastle, Australia in the effluent receiving marine waters of Burwood Beach WWTP (Burwood Beach "near", <50 m from outfall and Burwood Beach "far", 100-150 m from outfall) and reference locations (Redhead, Fingal Island 1 and Fingal Island 2) at depths of 4, 8 and 12 m for six weeks. Effluent receiving waters of Burwood Beach WWTP were found to be a suitable impact location, demonstrated via measurement of estrogenic compounds and activity throughout the deployment. Estrogenic compounds were detected (average of combined solids and liquid fractions) at average concentrations of: 1.42 ng/L for estrone, 0.69 ng/L for 17β estradiol, 3.83 ng/L for estriol (E3), 0.56 ng/L for 17α-ethynylestradiol, 64.2 ng/L for bisphenol A, 7.51 ng/L for 4-nonylphenol and 5.93 ng/L for 4-tert-octylphenol. Total estrogenic activity was estimated at 4.48 ng/L EEQ via the Yeast Estrogen Screen (YES(®)) assay (average of combined solid and liquid fractions). Female vitellogenin gene expression was highest at Burwood Beach locations, yet no significant differences were detected among locations for either sex. Vitellogenin protein was significantly higher (p<0.05) in S. glomerata at Burwood Beach Near compared to reference locations for the 4 and 12 m depths. Increased proportions of females were found at Burwood Beach Near, at 4m depth (p<0.05). Both Burwood Beach locations had higher proportions of mature female gonadal development stages compared to reference locations (p<0.05). Oocyte area was highest at both Burwood Beach locations, but no significant differences were detected among locations. Although mounting evidence suggests exposure to estrogenic contaminants increases vitellogenin production in molluscs, demonstration of dose-response relationships and knowledge of the temporal nature of the vitellogenin response with continual exposure is currently lacking for biomarker utility. To address this knowledge gap, adult Sydney rock oysters, Saccostrea glomerata, were exposed to a range of environmentally relevant concentrations of 17α-ethynylestradiol (EE2) (0, 6.25, 12.5, 25 or 50 ng/l) in seawater under laboratory conditions. Vitellogenin induction and gonadal development was assessed following 4, 21 and 49 days exposure to EE2. Vitellogenin was found to increase in a dose dependent manner with EE2 exposure for females (4 and 49 days) and males (4 and 21 days). Histological examination of gonads revealed a number of individuals exhibited intersex (ovotestis) in 50 ng/l EE2 (after 21 days) and in 6.25 and 12.5 ng/l EE2 (after 49 days). Furthermore, a significant shift towards females was observed following 49 days exposure at 50 ng/l EE2 suggesting estrogenic exposure is capable of facilitating a progression for protandric males from male-intersex-female gametal status. Increases in female vitellogenin (4 days) were predictive of later increases in female developmental stages at 21 days and increases in oocyte area following 49 days. Male vitellogenin (4 days) was predictive of decreased male percentages and lower male developmental stages at 49 days. Vitellogenin in S. glomerata is a predictive biomarker of estrogenic exposure and effect if sampled soon after exposure and at the commencement of a gonadal development cycle.

Adult Saccostrea glomerata were exposed to environmentally relevant concentrations of 4-nonylphenol (1microg/L and 100microg/L) and 17alpha-ethynylestradiol (5ng/L and 50ng/L) in seawater over 8 weeks. Exposures were performed to assess effects on vitellogenin induction and gonadal development during reproductive conditioning. Chronic direct estrogenicity within gonadal tissue was assessed via an estrogen receptor-mediated, chemical-activated luciferase reporter gene-expression assay (ER-CALUX). Estradiol equivalents (EEQ) were greatest in the 100microg/L 4-nonylphenol exposure (28.7+/-2.3ng/g tissue EEQ) while 17alpha-ethynylestradiol at concentrations of 50ng/L were 2.2+/-1.5ng/g tissue EEQ. Results suggest 4-nonylphenol may be accumulated in tissue and is partly resistant to biotransformation; maintaining its potential for chronic estrogenic action, while 17alpha-ethynylestradiol, although exhibiting greater estrogenic potency on biological endpoints possibly exerts its estrogenic action before being rapidly metabolised and/or excreted. A novel methodology was developed to assess vitellogenin using high-performance liquid chromatography (HPLC). Exposure to both 17alpha-ethynylestradiol (50ng/L) and 4-nonylphenol (100microg/L) produced increases in vitellogenin for females, whereas males exhibited increases in vitellogenin when exposed to 50ng/L 17alpha-ethynylestradiol only. Females exhibited greater vitellogenin responses than males at 50ng/L 17alpha-ethynylestradiol only. Histological examination of gonads revealed a number of individuals exhibiting intersex (ovotestis) in 50ng/L 17alpha-ethynylestradiol exposures. Male individuals in 1microg/L and 100microg/L 4-nonylphenol exposures and 5ng/L 17alpha-ethynylestradiol were at earlier stages of spermatogenic development than corresponding controls.

The Akoya pearl oyster (Pinctada imbricata) was experimentally exposed to (a) constant levels of lead (Pb) at 180 microg L(-1) for nine weeks, or (b) two short term (pulse) exposures of Pb at 180 microg L(-1) (three weeks each) with an intervening depuration period (three weeks), to assess its utility as an (i) accumulative monitor of Pb contamination and an (ii) archival monitor for discriminating constant versus pulsed Pb exposure events. P. imbricata showed similar reductions in growth (based on shell morphology and wet weight) and Pb accumulation patterns for whole tissue and shell in response to both Pb exposure regimes. Thus the whole oyster was deemed an inappropriate accumulative monitor for assessing short-term temporal variation of Pb exposure and effect. However, using secondary ion mass spectrometry, Pb was shown to accumulate in the successively deposited nacreous layers of the shell of P. imbricata, documenting the exposure history of constant versus pulsed Pb events. Patterns of Pb deposition not only reflected the frequency of Pb exposure events but also their relative durations. Thus, the shell of P. imbricata may be employed as a suitable biological archive of Pb exposure.

The use of pearl oysters has recently been proposed as an environmental remediation tool in coastal ecosystems. This study quantified the nitrogen, phosphorus and heavy metal content of the tissue and shell of pearl oysters harvested from a small pearl oyster farm at Port Stephens, Australia. Each tonne of pearl oyster material harvested resulted in approximately 703 g metals, 7452 g nitrogen, and 545 g phosphorus being removed from the waters of Port Stephens. Increasing current farm production of 9.8 tyr(-1) to 499 tyr(-1) would balance current nitrogen loads entering Port Stephens from a small Sewage Treatment Plant (STP) located on its southern shores. Furthermore, manipulation of harvest dates to coincide with oyster condition would likely remove substantially greater quantities of nutrients. This study demonstrates that pearl aquaculture may be used to assist in the removal of pollutants from coastal waters while producing a commercially profitable commodity.
