Freeze-induced stress causing aggregation of proteins has typically been primarily attributed to the ice-water interface. However, we hypothesize that the underlying observed and perceived detrimental effect of ice is, to some extent, attributed to air bubbles expelled from ice crystal lattices or to nanobubbles existing prior to freezing. The reduction of dissolved air was achieved via a deaeration process by placing samples in a reduced pressure chamber, while the reduction of nanobubbles was achieved by filtering samples via a syringe filter. The results showed that the reduction of both dissolved air molecules and stable colloidal nanobubbles in a bovine IgG solution prior to freezing led to a significant decrease in aggregation after thawing compared to untreated samples (∼6,000 vs. ∼ 40,000 particles/mL at a freezing rate of 100 K/s, respectively). The deaeration-filtration treatment works additively with cryoprotectants such as trehalose, further reducing the freeze-induced aggregation of IgG. The results also demonstrated that air-water interfacial aggregation of IgG in bulk liquid samples is a time-dependent process. The number of IgG subvisible particles increased with time and temperature, suggesting that random collisions of denatured molecules promoted the formation of aggregates with spherical morphology. In contrast, the IgG subvisible count after freeze-thawing had already reached its nominal value, suggesting a time-independent process where denatured protein molecules were compressed between ice crystals into filament-like aggregates. In summary, the findings shift the current paradigm from ice crystals being the main destabilizing factor during freezing to air bubbles, although the two are intertwined. The effect of concentration and origin of dissolved organic matter (DOM) on acenaphthene (Ace) photodegradation in liquid water and ice was investigated, and the components in DOM which were involved in Ace photodegradation were identified. The DOM samples included Suwannee River fulvic acid (SRFA), Elliott soil humic acid (ESHA), and an effluent organic matter (EfOM) sample. Due to the production of hydroxyl radical ( • OH) and triplet excited-state DOM ( 3 DOM*) which react with Ace, DOM had promotion effects on Ace photodegradation. However, the promotion effects of DOM were prevailed over by their suppressing effect of DOM including screening light effect, intermediates reducing effect and RS quenching effect, and thus, the photodegradation rates of Ace decreased in the presence of the three DOM with concentrations of 0.5-7.5 mg C/L in liquid water and ice. ESHA had higher light absorption and thus had higher screening light effect on Ace photodegradation in liquid water than SRFA and EfOM. At each DOM concentration, ESHA exhibited higher promotion effect on Ace photodegradation than SRFA and EfOM, in liquid water and ice. The binding of Ace with DOM was indicated by decreases in fluorescence intensity of Ace when coexisted with DOM. However, the binding of Ace to DOM played an unimportant role in suppressing Ace photodegradation. The photodegradation behavior of fluorophores in Ace with DOM present in ice was not similar to that in liquid water. C-O, C═O, carboxyl groups O-H and aliphatic C-H functional groups in DOM were involved in the interaction of DOM with Ace. This study assessed the influence of deep learning reconstruction (DLR) on the quality of diffusion-weighted images (DWI) and apparent diffusion coefficient (ADC) using an ice-water phantom. An ice-water phantom with known diffusion properties (true ADC = 1.1 × 10 -3 mm 2 /s at 0 °C) was imaged at various b-values (0, 1000, 2000, and 4000 s/mm 2 ) using a 3 T magnetic resonance imaging scanner with slice thicknesses of 1.5 and 3.0 mm. All DWIs were reconstructed with or without DLR. ADC maps were generated using combinations of b-values 0 and 1000, 0 and 2000, and 0 and 4000 s/mm 2 . Based on the quantitative imaging biomarker alliance profile, the signal-to-noise ratio (SNRs) in DWIs was calculated, and the accuracy, precision, and within-subject parameter variance (wCV) of the ADCs were evaluated. DLR improved the SNR in DWIs with b-values ranging from 0 to 2000s/mm 2 ; however, its effectiveness was diminished at 4000 s/mm 2 . There was no noticeable difference in the ADCs of images generated with or without implementing DLR. For a slice thickness of 1.5 mm and combined b-values of 0 and 4000 s/mm 2 , the ADC values were 0.97 × 10 -3 and 0.98 × 10 -3 mm 2 /s with and without DLR, respectively, both being lower than the true ADC value. Furthermore, DLR enhanced the precision and wCV of the ADC measurements. Magnetic field individually applied on the maximum ice crystal formation zone (MMF) and the whole freezing process (WMF) was compared to improve the quality of multiple freezing-thawing treated dough. All treatments showed that the breadmaking performances of magnetic field-assisted freezing were better than the conventional freezing. Especially, the WMF-treated breads exhibited higher resilience and lower firmness than MMF-treated breads. WMF treatment made dough remained a continuous and compact gluten-starch matrix while the starches and glutens got separated in MMF-treated dough. It could keep the gluten macropolymer from freezing-induced depolymerization with the decreased free sulfhydryl by 7.09% and more ordered secondary structure. WMF had positive effects on the homogeneous water distribution and high water-binding ability in frozen dough where the freezable water decreased from 32.47% to 30.77%. Ice slurry ingestion during prolonged exercises may improve performance in hot environments; however, the ideal amount and timing of ingestion are still uncertain. We determined whether ad libitum ice slurry ingestion influences physiological and perceptual variables and half-marathon performance while comparing the effects of the amount and moment of ingestion between ice slurry and water at 37 °C. Ten trained participants (28 ± 2 years; mean and SD) were required to run two half marathons while consuming either ice slurry (-1 °C; Ad-1) or water (37 °C; 37 CE) ad libitum. They then performed two other half marathons where, during one, they were required to ingest an amount of water equivalent to the amount consumed during the Ad-1 trial (Pro37), and in the other, to ingest ice slurry in the amount consumed during the 37 CE trial (Pro-1). During the half marathons, dry-bulb temperature and relative humidity were controlled at 33.1 ± 0.3 °C and 60 ± 3%, respectively. Ad-1 ingestion (349.6 ± 58.5 g) was 45% less than 37 CE ingestion (635.5 ± 135.8 g). Physical performance, heart rate, perceived exertion, body temperatures, and thermal perception were not influenced by the temperature or amount of beverage ingestion. However, a secondary analysis suggested that lower beverage ingestion was associated with improved performance (Ad-1 + Pro37 vs. 37 CE + Pro-1: -4.0 min, Cohen's d = 0.39), with a significant relationship between lower beverage ingestion and faster running time (b = 0.02, t = 4.01, p < 0.001). In conclusion, ice slurry ingestion does not affect performance or physiological or perceptual variables during a half marathon in a hot environment. Freeze-thawing (F/T) is an effective method of sludge dewatering preconditioning and has been studied in many studies. However, previous studies have taken landfill sludge from different regions, filled for different length of time or at different depth, resulting in large differences in initial water content and different treatment effects. Therefore, the effect of initial water content on the dewatering characteristics of F/T preconditioned landfill sludge has been investigated. The sludge with different initial water contents was firstly preconditioned by one F/T cycle. Then the F/T sludge was vacuum filtered and compared with the dewatering performance of FeCl 3 preconditioned sludge with the same water content. Finally, the mechanism of the initial water content on the effect of F/T preconditioning was analyzed by the change of sludge internal composition. The results show that the higher the initial water content of the sludge, the greater the improvement of its dewatering performance after F/T preconditioning. The specific resistance and water content after filtration of sludge after F/T conditioning decreased greatly with the increase of the initial water content, reaching their respective minimum values of 13.3 × 10 12  m/kg and 58.3% at 85% and 87.5%. These values are lower than the optimal values observed for the sludge conditioned by FeCl 3 . With the rise in initial water content, the driving force at the ice-water interface gains strength. Small particles aggregate into larger flocs, forming stable drainage channels that enhance the dewatering performance of sludge. Antifreeze proteins (AFPs) are biodegradable inhibitors that effectively prevent the formation of natural gas hydrates that block pipelines. In this study, molecular dynamics simulations were employed to establish a kinetic model of the hyperactive insect antifreeze protein ( Tenebrio molitor , Tm AFP) and its mutants to inhibit the growth of sI natural methane hydrate. Simulations revealed that the hydrophobic and hydrophilic groups of threonine (Thr) residues at hydrate-binding sites played a synergistic role in binding hydrates. The hydrophobic groups anchored Tm AFP to the hydrate surface through residues Thr 39 -Thr 65 by migrating pendant hydrophobic methyl groups to the hydrate semicages. The hydrophilic groups stabilized Tm AFP by hydrogen bonding with water molecules and integrating them into a quasi-hydrate structure, which more effectively inhibited hydrate growth. The results suggest that the hydrate growth inhibition is attributed to both the shape complementarity and the flexibility of binding residues. The synergy between hydrophobic and hydrophilic groups provides guidance for the design of more effective hydrate inhibitors.

Biological ice nucleation plays a key role in the survival of cold-adapted organisms. Several species of bacteria, fungi, and insects produce ice nucleators (INs) that enable ice formation at temperatures above -10 °C. Bacteria and fungi produce particularly potent INs that can promote water crystallization above -5 °C. Bacterial INs consist of extended protein units that aggregate to achieve superior functionality. Despite decades of research, the nature and identity of fungal INs remain elusive. Here, we combine ice nucleation measurements, physicochemical characterization, numerical modeling, and nucleation theory to shed light on the size and nature of the INs from the fungus Fusarium acuminatum . We find ice-binding and ice-shaping activity of Fusarium IN, suggesting a potential connection between ice growth promotion and inhibition. We demonstrate that fungal INs are composed of small 5.3 kDa protein subunits that assemble into ice-nucleating complexes that can contain more than 100 subunits. Fusarium INs retain high ice-nucleation activity even when only the ~12 kDa fraction of size-excluded proteins are initially present, suggesting robust pathways for their functional aggregation in cell-free aqueous environments. We conclude that the use of small proteins to build large assemblies is a common strategy among organisms to create potent biological INs.

Water-to-ice transformation results in a 10% increase in volume, which can have a significant impact on biopharmaceuticals during freeze-thaw cycles due to the mechanical stresses imparted by the growing ice crystals. Whether these stresses would contribute to the destabilization of biopharmaceuticals depends on both the magnitude of the stress and sensitivity of a particular system to pressure and sheer stresses. To address the gap of the "magnitude" question, a phospholipid, 1,2-dipalmitoyl- sn -glycero-3-phosphocholine (DPPC), is evaluated as a probe to detect and quantify the freeze-induced pressure. DPPC can form several phases under elevated pressure, and therefore, the detection of a high-pressure DPPC phase during freezing would be indicative of a freeze-induced pressure increase. In this study, the phase behavior of DPPC/water suspensions, which also contain the ice nucleation agent silver iodide, is monitored by synchrotron small/wide-angle X-ray scattering during the freeze-thaw transition. Cooling the suspensions leads to heterogeneous ice nucleation at approximately -7 °C, followed by a phase transition of DPPC between -11 and -40 °C. In this temperature range, the initial gel phase of DPPC, Lβ', gradually converts to a second phase, tentatively identified as a high-pressure Gel III phase. The Lβ'-to-Gel III phase transition continues during an isothermal hold at -40 °C; a second (homogeneous) ice nucleation event of water confined in the interlamellar space is detected by differential scanning calorimetry (DSC) at the same temperature. The extent of the phase transition depends on the DPPC concentration, with a lower DPPC concentration (and therefore a higher ice fraction), resulting in a higher degree of Lβ'-to-Gel III conversion. By comparing the data from this study with the literature data on the pressure/temperature Lβ'/Gel III phase boundary and the lamellar lattice constant of the Lβ' phase, the freeze-induced pressure is estimated to be approximately 0.2-2.6 kbar. The study introduces DPPC as a probe to detect a pressure increase during freezing, therefore addressing the gap between a theoretical possibility of protein destabilization by freeze-induced pressure and the current lack of methods to detect freeze-induced pressure. In addition, the observation of a freeze-induced phase transition in a phospholipid can improve the mechanistic understanding of factors that could disrupt the structure of lipid-based biopharmaceuticals, such as liposomes and mRNA vaccines, during freezing and thawing.

Using a randomized crossover protocol, 10 young men completed four 180 min exposures (38 °C, 60% relative humidity), alternating between 30 min of walking and 30 min of sitting where fluid or ice slurry were served. Participants underwent four trials: (i) 5 mL·kg body mass -1 of 20 °C water (CON); (ii) 5 mL·kg body mass -1 of 20 °C water + fan at 4 m·s -1 (FAN); (iii) fan + 3 mL·kg body mass -1 of ice slurry + 2 mL·kg body mass -1 of 4 °C water (FAN + ISCW); and (iv) FAN + ISCW + lower-leg immersion in 20 °C water (FAN + ISCW + LLI). Sweat and body mass losses were higher with FAN than CON, FAN + ISCW, and FAN + ISCW + LLI. Mean and peak changes in Δrectal temperature, heart rate, and perceived heat and thirst from baseline were not statistically and practically different between FAN and CON. FAN + ISCW + LLI decreased sweat loss compared with FAN + ISCW and mean and peak changes in Δrectal temperature, heart rate, and perceived thirst compared with CON. FAN + ISCW + LLI also attenuated the changes in Δrectal temperature (peak) and thirst (mean and peak) compared with FAN. In conclusion, FAN slightly exacerbates fluid loss but does not attenuate the changes in Δrectal temperature, heart rate, and perceived heat and thirst during intermittent physical activity in hot-humid conditions. However, coupling ISCW or ISCW + LLI with the use of a fan attenuates the increase in these outcomes compared with no intervention and to a greater extent for rectal temperature and thirst when fan is coupled with ISCW + LLI than when it is used in isolation., Competing Interests: The authors declare that there is no conflict of interest.

Background: Ice machines in healthcare facilities have been suspected and even linked to outbreaks and pseudo-outbreaks. Guidelines exist for maintenance of these devices but there is no clear independent infection control standard, and little is known about their microbial contamination., Aim: To evaluate the microbial contamination, amplification, and presence of opportunistic pathogens in ice-water machines in a healthcare facility., Methods: Concentrations of general microbial indicators (heterotrophic plate counts (HPC), total and intact cells), faecal indicators (enterococci) and opportunistic pathogens (Pseudomonas aeruginosa, non-tuberculous mycobacteria (NTM), Candida spp.) were measured in 36 ice-water machines on patient wards of a 772-bed hospital. Profile sampling was performed on five ice-water machines and adjacent faucets to identify sites of microbial proliferation., Findings: Candida spp. were found in half of ice-water samples while enterococci and P. aeruginosa were present in six and 11 drain inlets respectively. NTM were measured in all ice-water samples and 35 out of 36 biofilms. Pre-filters and ice machines are sites for additional amplification: NTM densities were on average 1.3 log 10 higher in water of ice machine flushed 5 min compared to flushed adjacent tap water., Conclusion: Ice machine design needs to be adapted to reduce microbial proliferation. The absence of correlation between HPC densities (current microbial indicators) and NTM concentrations suggests a need for cleaning efficiency indicators better correlated with opportunistic pathogens. Precise data on the non-variant equilibrium of the four phases (vapor-aqueous solution-ice-gas hydrate) in P - T coordinates are highly desired for developing accurate thermodynamic models and can be used as reference points (similar to the triple point of water). Using the two-component hydrate-forming system CO 2 -H 2 O, we have proposed and validated a new express procedure for determining the temperature and pressure of the lower quadruple point Q 1 . The essence of the method is the direct measurement of these parameters after the successive formation of the gas hydrate and ice phases in the initial two-phase gas-water solution system under intense agitation of the fluids. After relaxation, the system occurs in the same equilibrium state ( T = 271.60 K, P = 1.044 MPa), regardless of the initial parameters and the order of crystallization of the CO 2 hydrate and ice phases. Considering the combined standard uncertainties (±0.023 K, ±0.021 MPa), the determined P and T values agree with the results of other authors obtained by a more sophisticated indirect method. Validating the developed approach for systems with other hydrate-forming gases is of great interest.

Time-resolved structural investigations of crystallization of water in lipid/protein/salt mesophases at cryogenic temperatures are significant for comprehension of ice nanocrystal nucleation kinetics in lipid membranous systems and can lead to a better understanding of how to experimentally retard the ice formation that obstructs the protein crystal structure determination. Here, we present a time-resolved synchrotron microfocus X-ray diffraction (TR-XRD) study based on ∼40,000 frames that revealed the dynamics of water-to-ice crystallization in a lipid/protein/salt mesophase subjected to cryostream cooling at 100 K. The monoolein/hemoglobin/salt/water system was chosen as a model composition related to protein-loaded lipid cubic phases (LCP) broadly used for the crystallization of proteins. Under confinement in the nanoscale geometry, metastable short-living cubic ice (I c ) rapidly crystallized well before the formation of hexagonal ice (I h ). Cholic acid is a trihydroxy bile acid with a nice peculiarity: the average distance between the oxygen atoms (O7 and O12) of the hydroxy groups located at C7 and C12 carbon atoms is 4.5 Å, a value which perfectly matches with the O/O tetrahedral edge distance in Ih ice. In the solid phase, they are involved in the formation of hydrogen bonds with other cholic acid units and solvents. This fact was satisfactorily used for designing a cholic dimer which encapsulates one single water molecule between two cholic residues, its oxygen atom (Ow) being exactly located at the centroid of a distorted tetrahedron formed by the four steroid hydroxy groups. The water molecule participates in four hydrogen bonds, with the water simultaneously being an acceptor from the 2 O12 (hydrogen lengths are 2.177 Å and 2.114 Å) and a donor towards the 2 O7 (hydrogen bond lengths are 1.866 Å and 1.920 Å). These facts suggest that this system can be a nice model for the theoretical study of the formation of ice-like structures. These are frequently proposed to describe the water structure found in a plethora of systems (water interfaces, metal complexes, solubilized hydrophobic species, proteins, and confined carbon nanotubes). The above tetrahedral structure is proposed as a reference model for those systems, and the results obtained from the application of the atoms in molecules theory are presented here. Furthermore, the structure of the whole system allows a division into two interesting subsystems in which water is the acceptor of one hydrogen bond and the donor of another. The analysis of the calculated electron density is performed through its gradient vector and the Laplacian. The calculation of the complexation energy used correction of the basis set superposition error (BSSE) with the counterpoise method. As expected, four critical points located in the H…O bond paths were identified. All calculated parameters obey the proposed criteria for hydrogen bonds. The total energy for the interaction in the tetrahedral structure is 54.29 kJ/mol, while the summation obtained of the two independent subsystems and the one between the alkyl rings without water is only 2.5 kJ/mol higher. This concordance, together with the calculated values for the electron density, the Laplacian of the electron density, and the lengths of the oxygen atom and the hydrogen atom (involved in the formation of each hydrogen bond) to the hydrogen bond critical point, suggests that each pair of hydrogen bonds can be considered independent of each other., Competing Interests: The authors declare no conflict of interest.

Heterogeneous ice nucleation occurs vis-à-vis nucleating agents already present in solution yet can occur within a rather broad range of temperatures (0 to ca. -38 °C). Controlling this temperature and the subsequent growth of resulting ice crystals is crucial for the survival of biological organisms (certain insects, fish, and plants that endure subzero temperatures), as well as in the context of medical cryopreservation and food science. In these environments, uncontrolled crystal shape and size can rupture the cell membrane causing irreversible and catastrophic damage. Antifreeze (AF) proteins and synthetic AF analogs address this issue to restrict crystal growth and to shape ice crystals. Yet, if the nucleation temperature is not controlled and occurs in a lower temperature range, nascent ice crystals will have grown to a significantly larger size before the AF agents can be active on their surface to halt or slow the Ostwald ripening process during recrystallization. At a higher nucleation temperature, diffusion of AF macromolecules is enhanced, and dynamic crystal shaping can start earlier, producing smaller crystals overall. While antifreeze proteins, the inspiration for these synthetic analogs, are always applied in a salt buffer aqueous environment (most typically phosphate-buffered saline (PBS) buffer), the heterogeneous nucleation events are stochastic and occur within a wide temperature range. Silver iodide (AgI), however, is a highly effective ice nucleation promoter as its crystal lattice structure is a 98% lattice match to the basal plane of hexagonal ice (Ih) crystals acting as a template for water molecule orientation and decreasing the interfacial free energy. Here, we expose the advantage of purposely seeding such nascent ice crystals with AgI at a defined and higher temperature (-7 °C) in ultrapure water (UPW) such that nucleation can only come from AgI (and also in AgI/PBS), resulting in the most potent synthetic IRI observed to date (at concentrations as low as 0.001 mg·mL -1 ).

Antifreeze glycoproteins (AFGPs) are a special kind of antifreeze proteins with strong flexibility. Whether their antifreeze activity is achieved by reversibly or irreversibly binding to ice is widely debated, and the molecular mechanism of irreversible binding remains unclear. In this work, the antifreeze mechanism of the smallest AFGP isoform, AFGP8, is investigated at the atomic level. The results indicate that AFGP8 can bind to ice both reversibly through its hydrophobic methyl groups (peptide binding) and irreversibly through its hydrophilic disaccharide moieties (saccharide binding). Although peptide binding occurs faster than saccharide binding, free-energy calculations indicate that the latter is energetically more favorable. In saccharide binding, at least one disaccharide moiety is frozen in the grown ice, resulting in irreversible binding, while the other moieties significantly perturb the water hydrogen-bonding network, thus inhibiting ice growth more effectively. The present study reveals the coexistence of reversible and irreversible bindings of AFGP8, both contributing to the inhibition of ice growth and further provides molecular mechanism of irreversible binding.

A potential function is presented for describing a system of flexible H 2 O molecules based on the single-center multipole expansion (SCME) of the electrostatic interaction. The model, referred to as SCME/f, includes the variation of the molecular quadrupole moment as well as the dipole moment with changes in bond length and angle so as to reproduce results of high-level electronic structure calculations. The multipole expansion also includes fixed octupole and hexadecapole moments, as well as anisotropic dipole-dipole, dipole-quadrupole, and quadrupole-quadrupole polarizability tensors. The model contains five adjustable parameters related to the repulsive interaction and damping functions in the electrostatic and dispersion interactions. Their values are adjusted to reproduce the lowest energy isomers of small clusters, (H 2 O) n with n = 2-6, as well as measured properties of the ice Ih crystal. Subsequent calculations of the energy difference between the various isomer configurations of the clusters show that SCME/f gives good agreement with results of electronic structure calculations and represents a significant improvement over the previously presented rigid SCME potential function. Analysis of the vibrational frequencies of the clusters and structural properties of ice Ih crystal show the importance of accurately describing the variation of the quadrupole moment with molecular structures.

Coarse-grained water models are ∼100 times more efficient than all-atom models, enabling simulations of supercooled water and crystallization. The machine-learned monatomic model ML-BOP reproduces the experimental equation of state (EOS) and ice-liquid thermodynamics at 0.1 MPa on par with the all-atom TIP4P/2005 and TIP4P/Ice models. These all-atom models were parametrized using high-pressure experimental data and are either accurate for water's EOS (TIP4P/2005) or ice-liquid equilibrium (TIP4P/Ice). ML-BOP was parametrized from temperature-dependent ice and liquid experimental densities and melting data at 0.1 MPa; its only pressure training is from compression of TIP4P/2005 ice at 0 K. Here we investigate whether ML-BOP replicates the experimental EOS and ice-water thermodynamics along all pressures of ice I. We find that ML-BOP reproduces the temperature, enthalpy, entropy, and volume of melting of hexagonal ice up to 400 MPa and the EOS of water along the melting line with an accuracy that rivals that of both TIP4P/2005 and TIP4P/Ice. We interpret that the accuracy of ML-BOP originates from its ability to capture the shift between compact and open local structures to changes in pressure and temperature. ML-BOP reproduces the sharpening of the tetrahedral peak of the pair distribution function of water upon supercooling, and its pressure dependence. We characterize the region of metastability of liquid ML-BOP with respect to crystallization and cavitation. The accessibility of ice crystallization to simulations of ML-BOP, together with its accurate representation of the thermodynamics of water, makes it promising for investigating the interplay between anomalies, glass transition, and crystallization under conditions challenging to access through experiments.

To understand the crystallization of aqueous solutions in the atmosphere, biological specimens, or pharmaceutical formulations, the rate at which ice nucleates from pure liquid water must be quantified. There is still an orders-of-magnitude spread in the homogeneous nucleation rate of water measured using different instruments, with the most important source of uncertainty being that of the measured temperature. Microfluidic platforms can generate hundreds to thousands of monodisperse water-in-oil droplets, unachievable by most other techniques. However, most microfluidic devices previously used to quantify homogeneous ice nucleation rates have reported high temperature uncertainties, between ±0.3 and ±0.7 K. We use the recently developed Microfluidic Ice Nuclei Counter Zurich (MINCZ) to observe the freezing of spherical water droplets with two diameters (75 and 100 μm) at two cooling rates (1 and 0.1 K min -1 ). By varying both droplet volume and cooling rate, we were able to probe a temperature range of 236.5-239.3 K with an accuracy of ±0.2 K, providing reliable data where previously determined nucleation rates suffered from large uncertainties and inconsistencies, especially at temperatures above 238 K. From these data and from Monte Carlo simulations, we demonstrate the importance of obtaining a sufficiently large dataset so that underlying nucleation rates are not overestimated at higher temperatures. Finally, we obtain new parameters for a previous parameterisation by fitting to our newly measured nucleation rates, enabling its use in applications where ice formation needs to be predicted.

The extent of freeze dehydration of mesophyll cells in response to extracellular ice varies from supercooling to severe freezing cytorrhysis. The structural factors involved are poorly understood. In a comparison of mesophyll cells of 11 species, the factors "cell wall", "cellular" and "tissue" traits were investigated. The extent of freeze dehydration was quantified as reduction in the sectional area during controlled freezing in the presence of ice. The cell wall thickness, cell size, cell area and the relative area of intercellular spaces were determined. The modulus of elasticity was determined by psychrometry. To grasp the relationships between factors and with freeze dehydration, we applied a principal component analysis. The first two components explain 84% of the variance in the dataset. The first principal component correlated negatively with the extent of freeze dehydration and relative area of intercellular spaces, and positively with the squared cell wall thickness to cell size ratio, elasticity and cell wall thickness. The cell size parameters determined the second principal component. Supercooling appeared preferable in cells with a high squared cell wall thickness to cell size ratio and a low relative area of intercellular spaces. Such factors are hypothesised to affect the magnitude of negative turgor pressure being built up below the turgor loss point. Negative turgor pressure slows dehydration by reducing the water potential gradient to the extracellular ice. With high levels of freeze dehydration, sufficient intercellular spaces for extracellular ice accommodation are needed. Sound affects the medium it propagates through and studies on biological systems have shown various properties arising from this phenomenon. As a compressible media and a "collective mirror", water is influenced by all internal and external influences, changing its molecular structure accordingly. The water molecular structure and its changes can be observed as a whole by measuring its electromagnetic (EMG) spectrum. Using near-infrared spectroscopy and aquaphotomics, this pilot study aimed to better describe and understand the sound-water interaction. Results on purified and mineral waters reported similar effects from the applied 432 Hz and 440 Hz frequency sound, where significant reduction in spectral variations and increased stability in water were shown after the sound perturbation. In general, the sound rearranged the initial water molecular conformations, changing the samples' properties by increasing strongly bound, ice-like water and decreasing small water clusters and solvation shells. Even though there was only 8 Hz difference in applied sound frequencies, the change of absorbance at water absorbance bands was specific for each frequency and also water-type-dependent. This also means that sound could be effectively used as a perturbation tool together with spectroscopy to identify the type of bio, or aqueous, samples being tested, as well as to identify and even change water functionality.

Antifreeze glycoproteins (AFGPs) inhibit ice recrystallization by a mechanism remaining largely elusive. The dynamics of AFGPs' hydration water and its involvement in the antifreeze activity, for instance, have not been identified conclusively. We herein, by simulation and theory, examined the picosecond site-specific water dynamics in the first hydration layer of a solvated AFGP8. Using a hydrogen bond switch event-based treatment, we strictly excluded the non-first layer water contribution. The observed water dynamics is much more retarded and inhomogeneous compared to the result of other commonly adopted treatments with non-first layer water contributions included. A molecular jump model analysis, with the cross-correlation between hydrogen bond switch molecular frames included, further indicates that excluding the non-first layer water contribution enhances the slow component in water dynamics, which couples strongly with the local environment. Further comparison with the structured ubiquitin protein revealed that, although the overall relaxation time distributions are similar between two proteins, a significant portion (>30%) of water hydrogen bond switching processes on the AFGP8 surface are considerably slower since they are trapped between the disaccharides and other protein regions. AFGP8 therefore resembles much the situation of an enzyme binding cleft or a DNA groove, where considerable slowdown of hydration water dynamics is observed due to the confinement. When bound to the ice surface, these slow, disordered water molecules may become a factor hindering the ice growth.

New high-resolution imaging methods for biological samples such as atom probe tomography (APT), facilitated by the invention of laser-pulsed atom probes and cryo-transfer procedures, have recently emerged. However, ensuring the vitreous state of the fabricated aqueous needle-shaped APT samples remains a challenge despite it being crucial for characterizing biomolecules such as proteins and cellular architectures in their near-native state. Our work investigated three potential approaches: (1) open microcapillary (OMC) method, (2) high-pressure freezing method (HPF), and (3) graphene encapsulation method. Diffraction patterns of the needle specimens acquired by cryo-TEM have demonstrated the vitreous state of the ice needles, although limited to the tip regions, has been achieved with the three proposed approaches. With the capability to prepare vitreous ice needles from hydrated samples of up to ∼200 μm thickness (HPF), combined use of the three approaches opens new avenues for future near-atomic imaging of biological cells in their near-native state.

Long-term cryopreservation of human umbilical vein endothelial cells (HUVECs) is important and beneficial for a variety of biomedical research and applications. In this study, we investigated HUVEC's cryobiological characteristics and parameters that are indispensable for predicting and determining an optimal cooling rate to prevent lethal intracellular ice formation (IIF) and severe cell dehydration during the cryopreservation processes. The parameters include cell membrane hydraulic conductivity (i.e., cell membrane water permeability), L p , cell membrane water permeability activation energy, E lp , and osmotically inactive volume of a cell V b . Cryomicroscopy was used to study the IIF phenomena and cell volume excursion at various cooling rates, 1, 10, and 20°C/min, respectively, based on which the cryobiological parameters were determined using biophysical and mathematical models. Results from this research work laid an important cryobiological foundation for the optimization of HUVEC's cryopreservation conditions.

Hypothesis: Understanding the crystallization of atmospheric water can require levitation techniques to avoid the influence of container walls. Recently, an acoustic levitation device called the TinyLev was designed, which can levitate multiple droplets at room temperature. Proximal crystallization may affect droplet phase change and morphological characteristics., Methodology: In this study, acoustically levitated pure water droplets were frozen individually and in pairs or triplets using a TinyLev device. Nucleation, bulk crystal growth, and melting were observed using digital and infrared cameras concurrently., Findings: Initially, the acoustic field forced the droplets into an oblate spheroid shape, though the counteracting force of the cooling stream caused them to circularize. Droplet geometry was thus the net result of streaming forces and surface tension at the acoustic boundary layer/air-liquid interface. Nucleation was determined to be neither homogeneous nor heterogeneous but secondary, and thus dependent on the cooling rate and not on the degree of supercooling. It was likely initiated by aerosolized ice particles from the air or from droplets that had already nucleated and broken up. The latter secondary ice production process resulted in multi-drop systems with statistically identical nucleation times. Notably, this meant that the presence of interfacial rupture at an adjacent droplet could influence the crystallization behaviour of another. After the formation of an initial ice shell around the individual droplets, dendritic protrusions grew from the droplet surface, likely seeded by the same ice particles that caused nucleation, but at a quasi-liquid layer. When freezing was complete, it was determined that the frozen core had undergone a volumetric expansion of 30.75%, compared to 9% for pure, sessile water expansion. This significantly greater expansion may have resulted from entrained air bubbles at the inner solid-liquid interface and oscillations at the moving phase boundary caused by changes in local acoustic forces. The local environment within a hydrogel influences the properties of water, including the propensity for ice crystallization. Water-swollen amphiphilic copolymers produce tunable nanoscale environments, which are defined by hydrophobic associations, for the water molecules. Here, the antifreeze properties for equilibrium-swollen amphiphilic copolymers with a common hydrophilic component, hydroxyethyl acrylate (HEA), but associated through crystalline (octadecyl acrylate, ODA) or rubbery (ethylhexyl acrylate, EHA) hydrophobic segments, are examined. Differences in the efficacy of the associations can be clearly enunciated from compositional solubility limits for the copolymers in water (<2.6 mol % ODA vs ≤14 mol % EHA), and these differences can be attributed to the strength of the association. The equilibrium-swollen HEA-ODA copolymers are viscoelastic solids, while the swollen HEA-EHA copolymers are viscoelastic liquids. Cooling these swollen copolymers to nearly 200 K induces some crystallization of the water, where the fraction of water frozen depends on the details of the nanostructure. Decreasing the mean free path of water by increasing the ODA composition from 10 to 25 mol % leads to fractionally more unfrozen water (66-87%). The swollen HEA-EHA copolymers only marginally inhibit ice (<13%) except with 45 mol % EHA, where nearly 60% of the water remains amorphous on cooling to 200 K. In general, the addition of the EHA leads to less effective ice inhibition than analogous covalently crosslinked HEA hydrogels (19.9 ± 1.8%). These results illustrate that fluidity of confining surfaces can provide pathways for critical nuclei to form and crystal growth to proceed.

Aqueous supercooling provides a method by which to preserve biological matter at subfreezing temperatures without the deleterious effects of ice formation. The extended longevity of the preserved biologic is a direct result of a reduction in the rate of metabolism with decreasing temperature. However, because the nucleation of ice from a supercooled solution is a stochastic process, supercooled preservation carries the risk of random ice nucleation. Theoretical supercooled biopreservation research to date has largely treated these biological and thermophysical phenomena separately. Here, we apply a statistical model of stochastic ice nucleation to demonstrate how the possible reduction in metabolic rate is inherently related to supercooling stability (i.e., the likelihood of ice nucleation). We develop a quantitative approach by which to weigh supercooling stability versus potential metabolic reduction, and further show how the stability-metabolism relationship varies with system size for two assumed modes of nucleation. The ice crystallization and melting in systems where the equilibrium state is difficult to reach is one of the growing areas in pharmaceutical freeze-drying research. The quality of the final freeze-dried product depends on the parameters of the cooling step, which affect the ice nucleation and growth. In this paper, we present a DSC study of ice crystallization and melting in a sucrose-water system. Using two different types of thermal cycles, we examine the influence of cooling and heating rates on the thermal behavior of sucrose-water solutions with water contents between 50 and 100 wt%. The DSC results show that low cooling rates provide crystallization at higher temperatures and lead to lower amount of non-freezing water. Consequently, the glass transition and ice melting properties observed upon heating depend on the cooling conditions in the preceding step. Based on the experimental results, we investigate the reasons for the existence of the two steps on DSC heating curves in sucrose-water systems: the glass transition step and the onset of ice melting. We show that diffusion of water can be the limiting factor for ice growth and melting in the sucrose-water system when the amorphous phase is in a liquid state. In particular, when the diffusion coefficient drops below 10 -14 m 2 /sec, the ice crystals growth or melting becomes strongly suppressed even above the glass transition temperature. Experiments and computer simulations have established that liquid water's surfaces can deviate in important ways from familiar bulk behavior. Even in the simplest case of an air-water interface, distinctive layering, orientational biases, and hydrogen bond arrangements have been reported, but an overarching picture of their origins and relationships has been incomplete. Here we show that a broad set of such observations can be understood through an analogy with the basal face of crystalline ice. Using simulations, we demonstrate a number of structural similarities between water and ice surfaces, suggesting the presence of domains at the air-water interface with ice-like features that persist over 2-3 molecular diameters. Most prominent is a shared characteristic layering of molecular density and orientation perpendicular to the interface. Lateral correlations of hydrogen bond network geometry point to structural similarities in the parallel direction as well. Our results bolster and significantly extend previous conceptions of ice-like structure at the liquid's boundary and suggest that the much-discussed quasi-liquid layer on ice evolves subtly above the melting point into a quasi-ice layer at the surface of liquid water.

The adsorption of acetamide on low density amorphous (LDA) ice is investigated by grand canonical Monte Carlo computer simulations at the temperatures 50, 100, and 200 K, characteristic of certain domains of the interstellar medium (ISM). We found that the relative importance of the acetamide-acetamide H-bonds with respect to the acetamide-water ones increases with decreasing temperature. Thus, with decreasing temperature, the existence of the stable monolayer, characterizing the adsorption at 200 K, is gradually replaced by the occurrence of marked multilayer adsorption, preceding even the saturation of the first layer at 50 K. While isolated acetamide molecules prefer to lay parallel to the ice surface to maximize their H-bonding with the surface water molecules, this orientational preference undergoes a marked change upon saturation of the first layer due to increasing competition of the adsorbed molecules for H-bonds with water and to the possibility of their H-bond formation with each other. As a result, molecules stay preferentially perpendicular to the ice surface in the saturated monolayer. The chemical potential value corresponding to the point of condensation is found to decrease linearly with increasing temperature. We provide, in analogy with the Clausius-Clapeyron equation, a thermodynamic explanation of this behavior and estimate the molar entropy of condensed phase acetamide to be 34.0 J/mol K. For the surface concentration of the saturated monolayer, we obtain the value 9.1 ± 0.8 µmol/m 2 , while the heat of adsorption at infinitely low surface coverage is estimated to be -67.8 ± 3.0 kJ/mol. Our results indicate that the interstellar formation of peptide chains through acetamide molecules, occurring at the surface of LDA ice, might well be a plausible process in the cold (i.e., below 50 K) domains of the ISM; however, it is a rather unlikely scenario in its higher temperature (i.e., 100-200 K) domains.

Pseudomonas syringae is a widely spread plant pathogen known to have ice-nucleating proteins that serve as crystallization sites promoting ice growth at near-zero temperatures. Three temperatures that characterize water freezing and ice melting are (i) the freezing point of water, (ii) the temperature of coexistence of ice and water, and (iii) the melting point of ice. Here we show the influence of different concentrations of P. syringae on these three parameters. P. syringae appears to affect both the freezing point of water and the temperature of the coexistence of ice and water. Bacterial ice nucleators (INs) are among the most effective ice nucleators known and are relevant for freezing processes in agriculture, the atmosphere, and the biosphere. Their ability to facilitate ice formation is due to specialized ice-nucleating proteins (INPs) anchored to the outer bacterial cell membrane, enabling the crystallization of water at temperatures up to -2 °C. In this Perspective, we highlight the importance of functional aggregation of INPs for the exceptionally high ice nucleation activity of bacterial ice nucleators. We emphasize that the bacterial cell membrane, as well as environmental conditions, is crucial for a precise functional INP aggregation. Interdisciplinary approaches combining high-throughput droplet freezing assays with advanced physicochemical tools and protein biochemistry are needed to link changes in protein structure or protein-water interactions with changes on the functional level.

To investigate the dielectric relaxations of ice in low-concentration protein-water mixtures, broadband dielectric spectroscopy measurements were performed on partially crystallized bovine serum albumin (BSA)-water mixtures with BSA concentrations of 1-10 wt% at temperatures in the range of 123-298 K. The temperature dependence of the relaxation time of ice observed in all these mixtures changes twice at T C1 (∼240 K) and T C2 (200-160 K) ( T C1 , changes. Below 200 K, the relaxation of ice separates as 3-4 relaxations with different T C2 values. The presence of the multiple ice relaxations is the same as that observed for the gelatin-water mixtures (T. Yasuda, K. Sasaki, R. Kita, N. Shinyashiki and S. Yagihara, i.e. , at which the apparent activation energy, E a , changes. Below 200 K, the relaxation of ice separates as 3-4 relaxations with different T are different. The relaxation interpreted to be due to uncrystallized water in 20 wt% and 40 wt% BSA-water mixtures reported (N. Shinyashiki, W. Yamamoto, A. Yokoyama, T. Yoshinari, S. Yagihara, R. Kita, K. L. Ngai and S. Capaccioli, C2 and E , 14448) was re-examined and concluded to be due to one of the multiple relaxations of ice.a values. Hypothesis: Loss of anti-icing properties of slippery liquid-infused porous surfaces (SLIPS) in conditions of repetitive shear stresses is the intrinsic process related to peculiarities of SLIPS structure., Experiments: The study of the evolution of the ice adhesion strength to superhydrophobic surfaces (SHS) and SLIPS during repetitive icing/de-icing cycles measured by a centrifugal method was supplemented with the estimation of change in capillary pressure inside the pores, and SEM analysis of the effect of multiple ice detachments on surface morphology., Findings: Obtained data indicated that although for freshly prepared SLIPS, the ice shear adhesion strength at -25 °C was several times lower than for SHS, repetitive icing-deicing cycles resulted in dramatic SLIPS degradation. In contrast, SHS showed weak degradation at least during 50 cycles. Additional to the depletion of an impregnating oil layer, other mechanisms of SLIPS degradation were hypothesized and tested. It was shown that lower capillary pressure required to displace air by water from the surface texture for SLIPSs compared to SHSs resulted in deeper water/ice penetration inside the grooves. Spin probe electron paramagnetic resonance spectroscopy is applied to characterize the dynamics of concentric hydration and mesophase solvent domains that surround proteins within the ice boundary in frozen aqueous solutions. The solvent dynamics are tuned by variation of temperature (190-265K) and by the degree of ice boundary confinement, which is modulated by the volume of added cryosolvent (0-~50Å separation distance from protein surface). Goals are to: (1) characterize the protein-coupled solvent dynamics on correlation time scales of ~10 -10 <τ<10 -7 s, and spatial scales from protein surface to periphery of the surrounding solution, from the perspective of a free, small-molecule (~7Å diameter) probe, and (2) reveal properties of the solvent-protein coupling that can be correlated with protein functions, that are measureable under the same conditions. Rotational mobility of the nitroxide spin probe, TEMPOL, resolves and tracks two solvent components, the protein-associated domain (PAD; akin to hydration layer) and surrounding mesodomain, through their distinct temperature- and confinement-dependent values of τ and normalized weight. Detailed protocols are described for simulation of two-component nitroxide EPR spectra, which are categorized by line shape regime and guided by a library of template spectra and simulation parameters derived from two model soluble globular proteins. The order-disorder transition in the PAD, which is a universal feature of protein-coupled solvent dynamics, provides a well-defined, tunable property for elucidating mechanism in solvent-protein-function dynamical coupling. In this paper, a mathematical model based on spherical differential unit cell is proposed as a model for studying seasonal freeze-thaw soil space infinitesimal differential unit cell. From this model, the basic equations of permafrost moisture and heat flow motion are directly derived, then the linked equations form the permafrost water-heat coupled transport model. On this basis, the one-dimensional seasonal permafrost water-heat transport equation is derived. The model reduces the original spatial three-variable coordinate system (parallel hexahedron) into a coupled equation with a single spherical radius (R) as the independent variable, so the iterations of the numerical simulation algorithm is greatly reduced and the complexity is decreased. Finally, the model is used to simulate the seasonal freeze-thaw soil in the ShiHeZi region of Xinjiang, China. The principle of the simulation is to collect the soil temperature and humidity values of the region in layers and fixed-points using a homemade freeze-thaw soil sensor, after that we solve it by numerical calculation using MATLAB. The analysis results show that the maximum relative error of the model we proposed is 4.36, the minimum error is 0.98, and the average error is 2.515. The numerical simulation results are basically consistent with the measured data, then the proposed model is consistent with the matching states of permafrost moisture content and soil temperature in the region at different times. Precise control over the size and shape of ice crystals is a key factor to consider in designing antifreezing and cryoprotecting molecules for cryopreservation of cells. Here, we report that a poly(ethylene glycol)-poly(l-alanine) (PEG-PA) block copolymer exhibits excellent cryoprotecting properties for stem cells and antifreezing properties for water. As the molecular weight of PA increased from 500, 760, and 1750 Da (P1, P2, and P3) at the same PEG molecular weight of 5000 Da, the β-sheet content decreased and α-helix content increased. Comparing P2 (PEG-PA; 5000-760) and P4 (PEG-PA: 1000-750), β-sheets increased as the PEG block length decreased. The critical micelle concentration of the PEG-PA block copolymers was in a range of 0.5-3.0 mg/mL and was proportional to the hydrophobicity of the PEG-PA block copolymers. The P1, P2, and P3 self-assembled into spherical micelles, whereas P4 formed micelles with cylindrical morphology. The difference in the block copolymer structure affected ice recrystallization inhibition (IRI) activity and cryopreservation of cells. IRI activity was assayed via mean largest grain size (MLGS), and interactions between polymers and ice crystal surfaces were studied by dynamic ice-shaping studies. The MLGS decreased to 58 → 53 → 45 → 35 → 23% of that of PBS, as the polymer (PEG-PA 5000-500) concentration increased from 0.0 (PBS; control) → 1.0 → 5.0 → 10 → 30 → 50 mg/mL. The MLGS of PEG 5k solutions (negative control) decreased to 74 → 71 → 64 → 44 → 37% of that of PBS in the same concentration range. P3 and P4 with a longer hydrophobic PA block developed elongated ice crystals at above 30 mg/mL. The dynamic ice-shaping study exhibited that ice crystals became needle-shaped, as the hydrophobicity of the polymer increased as in P2-P4. The cell recovery in the P1 system after cryopreservation at -196 °C for 7 days was 87% of that of the dimethyl sulfoxide (DMSO) 10% system (positive control). The cell recovery was 48% for the P2 system and drastically decreased to less than 30% of that of the DMSO 10% system in the P3, P4, PEG 5k, PEG 1k, PVA 80H, and PVA 100H systems. Current studies suggest that IRI activity, round ice crystal shaping, and membrane stabilization activity of P1 cooperatively provide excellent cell recovery among the candidate systems. Recovered stem cells exhibited excellent proliferation and multilineage differentiation into osteocytes, chondrocytes, and adipocytes. To conclude, the PEG-PA (5000-500) block copolymer is suggested to be a promising antifreezing cryoprotectant for stem cells.

A springtail (Collembola) identified as Granisotoma rainieri was collected from snow in Hokkaido, Japan, in late winter when nighttime temperatures were below zero. Extracts of these arthropods showed antifreeze activity by shaping ice crystals and stopping their growth. The glycine-rich proteins responsible for this freezing point depression were isolated by ice-affinity purification and had principal masses of ~ 6.9 and 9.6 kDa. We identified a transcript for a 9.6-kDa component and produced it as a His-tagged recombinant protein for structural analysis. Its crystal structure was solved to a resolution of 1.21 Å and revealed a polyproline type II helical bundle, similar to the six-helix Hypogastrura harveyi AFP, but with nine helices organized into two layers held together by an extensive network of hydrogen bonds. One of the layers is flat, regular, and hydrophobic and likely serves as the ice-binding side. Although this surface makes close protein-protein contacts with its symmetry mate in the crystal, it has bound chains of waters present that resemble those on the basal and primary prism planes of ice. Molecular dynamic simulations indicate most of these crystal waters would preferentially occupy these sites if exposed to bulk solvent in the absence of the symmetry mate. These prepositioned waters lend further support to the ice-binding mechanism in which AFPs organize ice-like waters on one surface to adsorb to ice. The application and functionalization of cellulose has been attracting increased attention in academic and industrial studies because of its wide range of sources, short renewable cycle, and low environmental impact. In order to enhance the application field of cellulose and decrease the environmental pollution for organic solvent associated with its preparation, cellulose foam with a vertically hierarchically porous structure similar to wood was designed and fabricated successfully from a cellulose aqueous solution using an ice templated in this study. The cellulose foam prepared using a 3 wt % concentration possessed a uniform vertical hierarchically porous structure, which could provide a pathway for the flow of water or air based on the capillary effect. The highest water wicking rate and flux were 7.8184 mm·s -1 and 29.49 mL·min -1 ·g -1 , respectively, for the porous foam prepared using a 3 wt % concentration. The mechanical testing experiment showed that the porous structure did not reduce the amount of stress that the sample could endure before being damaged. The compression strength increased with increasing cellulose concentration in solution. Therefore, the hierarchical structure formed in the prepared cellulose foam effectively improved the water flux behavior and provided a structural basis for future applications of cellulose scaffolds.

Antifreeze proteins (AFPs) inhibit ice growth by adsorbing onto specific ice planes. Microbial AFPs show diverse antifreeze activity and ice plane specificity, while sharing a common molecular scaffold. To probe the molecular mechanisms responsible for AFP activity, we here characterized the antifreeze activity and crystal structure of TisAFP7 from the snow mold fungus Typhula ishikariensis. TisAFP7 exhibited intermediate activity, with the ability to bind the basal plane, compared with a hyperactive isoform TisAFP8 and a moderately active isoform TisAFP6. Analysis of the TisAFP7 crystal structure revealed a bound-water network arranged in a zigzag pattern on the surface of the protein's ice-binding site (IBS). While the three AFP isoforms shared the water network pattern, the network on TisAFP7 IBS was not extensive, which was likely related to its intermediate activity. Analysis of the TisAFP7 crystal structure also revealed the presence of additional water molecules that form a ring-like network surrounding the hydrophobic side chain of a crucial IBS phenylalanine, which might be responsible for the increased adsorption of AFP molecule onto the basal plane. Based on these observations, we propose that the extended water network and hydrophobic hydration at IBS together determine the TisAFP activity.

Ice-nucleation active (INA) bacteria can promote the growth of ice more effectively than any other known material. Using specialized ice-nucleating proteins (INPs), they obtain nutrients from plants by inducing frost damage and, when airborne in the atmosphere, they drive ice nucleation within clouds, which may affect global precipitation patterns. Despite their evident environmental importance, the molecular mechanisms behind INP-induced freezing have remained largely elusive. We investigate the structural basis for the interactions between water and the ice-nucleating protein InaZ from the INA bacterium Pseudomonas syringae. Using vibrational sum-frequency generation (SFG) and two-dimensional infrared spectroscopy, we demonstrate that the ice-active repeats of InaZ adopt a β-helical structure in solution and at water surfaces. In this configuration, interaction between INPs and water molecules imposes structural ordering on the adjacent water network. The observed order of water increases as the interface is cooled to temperatures close to the melting point of water. Experimental SFG data combined with molecular-dynamics simulations and spectral calculations show that InaZ reorients at lower temperatures. This reorientation can enhance water interactions, and thereby the effectiveness of ice nucleation.

Taste disorders are common adverse effects of cancer chemotherapy that can reduce quality of life and impair nutritional status. However, the molecular mechanisms underlying chemotherapy-induced taste disorders remain largely unknown. Furthermore, there are no effective preventive measures for chemotherapy-induced taste disorders. We investigated the effects of a combination of three anticancer drugs (TPF: docetaxel, cisplatin and 5-fluorouracil) on the structure and function of mouse taste tissues and examined whether the drinking of ice-cold water after TPF administration would attenuate these effects. TPF administration significantly increased the number of cells expressing apoptotic and proliferative markers. Furthermore, TPF administration significantly reduced the number of cells expressing taste cell markers and the magnitudes of the responses of taste nerves to tastants. The above results suggest that anticancer drug-induced taste dysfunction may be due to a reduction in the number of taste cells expressing taste-related molecules. The suppressive effects of TPF on taste cell marker expression and taste perception were reduced by the drinking of ice-cold water. We speculate that oral cryotherapy with an ice cube might be useful for prophylaxis against anticancer drug-induced taste disorders in humans.

Background: Under atmospheric pressure, the identifiable phases of ice crystals are hexagonal (stable) and cubic (metastable)., Objective: This study aimed to test the hypothesis that water crystallizes into the cubic phase at the beginning and then changes to the hexagonal phase., Materials and Methods: Aqueous solutions of 40% (w/w) and 50% (w/w) glucose, and 40% (w/w) ammonium hydrogen sulfate, as well as emulsified water, were investigated., Results: The cubic-to-hexagonal ice phase transition was detected in 40% (w/w) glucose solution within a 1 s integration interval, whereas the cubic ice formed in 50% (w/w) glucose solution did not transition to the hexagonal phase. The cubic phase was also confirmed in the 40% (w/w) ammonium hydrogen sulfate solution, but not in emulsified water., Conclusion: The cubic-to-hexagonal ice phase transition was detected in three aqueous solutions tested upon freezing. It was not possible to clearly capture the transition process in emulsified water under the study condition.

Nucleobases are of significant importance to all known organisms, may be an important building block of life, and could be important biosignatures of current or past life. Given their potential significance to the field of astrobiology, it is important to understand the survival of these molecules when subjected to ionizing radiation as is present in a range of extraterrestrial environments. In this work, we present data on the kinetics of the radiolytic destruction of pure thymine and water + thymine ice mixtures at temperatures from 13 to 150 K. Rate constants were measured using in situ infrared spectroscopy, and radiolytic half-lives for thymine were computed for different planetary and interstellar environments. Our results demonstrate that the survival of thymine decreases as the dilution of thymine in water increases. Additionally, we find that thymine survival increases with ice temperature and that this decrease may be related to structure of the ice matrix.

The accumulation of ice on surfaces brings dangerous and costly problems to our daily life. Thus, it would be desirable to design anti-icing coatings for various surfaces. We report a durable anti-icing coating based on mussel-inspired chemistry, which is enabled via fabricating a liquid water layer, achieved by modifying solid substrates with the highly water absorbing property of sodium alginate. Dopamine, the main component of the mussel adhesive protein, is introduced to anchor the sodium alginate in order to render the coating applicable to all types of solid surfaces. Simultaneously, it serves as the cross-linking agent for sodium alginate; thus, the cross-linking degree of the coatings could be easily varied. The non-freezable and freezable water in the coatings with different cross-link degrees all remain liquid-like at subzero conditions and synergistically fulfill the aim of decreasing the temperature of ice nucleation. These anti-icing coatings display excellent stability even under harsh conditions. Furthermore, these coatings can be applied to almost all types of solid surfaces and have great promise in practical applications.

The ice-water interface is commonly encountered in our life, and comes into play in a wide number of natural phenomena. Here, attention will be focused on its effects on protein stability, with specific reference to the case of pharmaceutical proteins. This field represents a fascinating, and not yet fully understood, subject of investigation. Some background information on the ice-water phase diagram, as well as to the mechanisms of nucleation and crystal growth, will be provided. We will eventually discuss the effect of ice on protein activity, reviewing the mechanisms of ice-induced denaturation that have been proposed so far and discussing the strategies that may help prevent, or minimize, undesired loss of therapeutic activity., (Copyright © 2020 American Pharmacists Association®.
