Helichrysum decorum DC, Helichrysum lepidissimum S. Moore, and Helichrysum umbraculigerum are three species traditionally used in the South African medicine. The present work deals with the investigation of the spontaneous emission and the essential oils obtained from these plants cultivated in open field under uniform conditions. Fractions of the volatile organic compounds of the three species were rich in monoterpene hydrocarbons, representing more than 70% of the total composition. Pinene isomers were the most representative compounds: β-pinene in H. decorum (53.0%), and α-pinene in H. lepidissimum (67.9%) and H. umbraculigerum (54.8%). These latter two species evidenced an important amount of sesquiterpene hydrocarbons (SH) especially represented by γ-curcumene ( H. lepidissimum ) and α- and β-selinene ( H. umbraculigerum ). On the contrary, in the EOs, sesquiterpenes compounds prevailed, representing more than 64% of the identified fraction to reach more than 82 and 87% in H. umbraculigerum and H. lepidissimum, respectively. Although the chemical classes and their relative abundances were comparable among the three species, the individual compounds of EOs showed large differences. In fact, caryophyllene oxide (26.7%) and γ-curcumene (17.4%) were the main constituents in H. decorum , and H. lepidissimum respectively, while neo -intermedeol (11.2%) and viridiflorol (10.6%) characterized H. umbraculigerum .

Spontaneous emissions of S. dentata Aiton and S. scabra Thunb., as well as the essential oil (EO) composition of the cited species, together with S. aurea L., were investigated. The chemical profile of the first two species is reported here for the first time. Moreover, in vitro tests were performed to evaluate the antifungal activity of these EOs on Trichophyton mentagrophytes , Microsporum canis , Aspergillus flavus , Aspergillus niger , and Fusarium solani . Secondly, the EO antibacterial activity against Escherichia coli,   Staphylococcus aureus , and Staphylococcus pseudointermedius was examined, and their antiviral efficacy against the H1N1 influenza virus was assessed. Leaf volatile organic compounds (VOCs), as well as the EOs obtained from the arial part of Salvia   scabra , were characterized by a high percentage of sesquiterpene hydrocarbons (97.8% and 76.6%, respectively), mostly represented by an equal amount of germacrene D (32.8% and 32.7%, respectively). Both leaf and flower spontaneous emissions of S. dentata , as well as the EO composition, showed a prevalence of monoterpenes divided into a more or less equal amount of hydrocarbon and oxygenated compounds. Interestingly, its EO had a non-negligible percentage of oxygenated sesquiterpenes (29.5%). S. aurea EO, on the contrary, was rich in sesquiterpenes, both hydrocarbons and oxygenated compounds (41.5% and 33.5%, respectively). S. dentata EO showed good efficacy (Minimal Inhibitory Concentration (MIC): 0.5%) against M. canis . The tested EOs were not active against E. coli and S. aureus , whereas a low inhibition of S. dentata EO was observed on S. pseudointermedius (MIC = 10%). Once again, S. dentata EO showed a very good H1N1 inhibition; contrariwise, S. aurea EO was completely inactive against this virus. The low quantity of S. scabra EO made it impossible to test its biological activity. S. dentata EO exhibited interesting new perspectives for medicinal and industrial uses.

In the present work the composition of biogenic volatile organic compounds (BVOCs) and the essential oil (EO) of Helichrysum araxinum Takht. ex Kirp. aerial parts, together with the antimicrobial activity, were investigated. The results showed the prevalence of sesquiterpene hydrocarbons in both spontaneous emissions as well as in the EO. The main compounds of BVOCs were γ-curcumene (10.7%), γ-muurolene (9.2%), and β-selinene (8.5%). This latter constituent also showed a similar amount in the EO and represented the most abundant compounds together with α-selinene (8.0%). It is Interesting to note the same percentage of monoterpene hydrocarbons (MHs) in both the aroma profile and the EO (18.0%) with the same most abundant compounds: β-pinene (6.3% in BVOCs vs. 5.1% in EO, respectively) and limonene (4.5% in VOCs vs. 4.9% in EO, respectively). With regard to the antimycotic activity, the EO showed to be inactive against the tested strains, while a moderate antibacterial activity was shown against Staphylococcus isolates.

Helichrysum genus was used in folk South African medicine to treat various human disorders. As a part of our on-going research addressing the exploitation of South African plants belonging to this genus, five species were investigated for their volatile and antimicrobial activities. The volatile organic compounds (VOCs) and the essential oils (EOs) were analysed by gas chromatography mass spectrometry (GC-MS). Microdilution was the method used for assessing both antimycotic and antibacterial activities, which was also tested by Kirby-Bauer agar disc diffusion. Total monoterpenes (TMs) dominated the VOCs of four species ( H. trilineatum (70.6%), H. edwardsii (79.3%), H. cooperi (84.5%), and H. pandurifolium (57.0%)). H. cooperi and H. edwardsii EOs showed the predominance of TMs (68.2% and 84.5%, respectively), while H. pandurifolium and H. trilineatum EOs were characterized by the prevalence of TSs (86.5% and 43.6%, respectively). H. odoratissimum EO evidenced a similar amount of both TMs (49.5%) and TSs (46.4%). Microsporum canis was more sensitive to these EOs. The lowest minimum inhibitory concentration (MIC) was observed with H. pandurifolium and H. edwardsii EOs (0.25%). H. pandurifolium and H. trilineatum had a good effect on Staphylococcus aureus (MIC 5%). These findings open new perspectives for the exploitation of these natural compounds for application in cosmetics and pharmaceutics.

The volatile profiles (VOC) and the essential oil (EO) composition from the aerial parts of Salvia broussonetii were analysed. Sesquiterpene hydrocarbons dominate the VOCs from leaves (95.7%) and flowers (67.6%), followed by monoterpene hydrocarbons (2.6 and 29.7%, respectively). The main common compounds are germacrene D, β-bourbonene, α-pinene, α-copaene and α-gurjunene, even if with divergent relative abundances. In the leaf EOs the sesquiterpenes prevail, even if not overwhelmingly (about 50.0%), followed by monoterpenes (23.0-35.0%) and by minor fractions of diterpene hydrocarbons and non-terpene derivates. The most abundant common compounds across the two sampling periods are α-pinene, β-pinene, isobornyl acetate, α-gurjenene, germacrene D and bifloratriene. A morphological characterisation of the trichomes responsible for the productivity in terpenes was also performed. Four different morphotypes were observed on the above ground organs of S. brussonetii: peltates and capitates of type II and III resulted the only producers of volatile substances.

Rhus coriaria, also known as Sumac, has been traditionally used in many countries as spice, condiment, dying agent, and medicinal herb. The chemical composition of essential oils (EOs) and the volatile emissions from different organs of this species collected in Sicily (Italy) were analyzed by gas chromatography-flame ionization detection and gas chromatography/mass spectrometry. Monoterpene and sesquiterpene hydrocarbons were the most abundant class in the volatile emissions with β-caryophyllene and α-pinene were the main constituents in the majority of the examined samples. The EO composition was characterized by high amount of monoterpene and sesquiterpene hydrocarbons together with diterpenes. The main compounds in the EO obtained from the leaves and both stages of fruit maturation were cembrene and β-caryophyllene, while α-pinene and tridecanoic acid were the key compounds in the flower EO. The chemical composition of spontaneous volatile emission from Rubus ulmifolius flowers and fruits during different stages of development was evaluated by HS-SPME-GC/MS. In total, 155 chemical compounds were identified accounting 84.6 - 99.4% of whole aroma profile of flowers samples and 92.4 - 96.6% for fruit samples. The main constituents were α-copaene, β-caryophyllene, germacrene D, (E,E)-α-farnesene, 1,7-octadien-3-one,2-methyl-6-methylene, tridecane, (E)-2-hexenol acetate, (E)-3-hexenol acetate and cyperene. We analyzed the spontaneous volatile emission of different aerial parts of the caper (Capparis spinosa L.) by HS-SPME-GC/MS. We identified 178 different compounds of which, in different proportions based on the sample type, the main ones were (E)-β-ocimene, methyl benzoate, linalool, β-caryophyllene, α-guaiene, germacrene D, bicyclogermacrene, germacrene B, (E)-nerolidol, isopropyl tetradecanoate, and hexahydrofarnesyl acetone. The multivariate statistical analyses seem to point out that the parameter leading the emission patterns is the function of the analyzed sample; the flower samples showed differences in the emission profile between their fertile and sterile portions and between the other parts of the plant. The green parts emission profiles group together in a cluster and are different from those of seeds and fruits. Flowering plants employ a wide variety of signals, including scent, to attract pollinators. The aim of this work was to examine whether flower volatiles in four closely related Crocus species are linked to species divergence and to the current knowledge on their pollination syndromes. Fragrances of freshly opened flowers in Crocus etruscus, C. ilvensis, C. neglectus, and C. vernus, all belonging to ser. Verni, were analyzed using GC/MS. Results coincide with present knowledge about systematic relationships among taxa. The four species fall into two main fragrance types, based on similarities of their volatile compounds. In C. etruscus, C. ilvensis, and C. neglectus, oxygenated monoterpenes (lilac aldehyde B and A) are most abundant, while C. vernus has a fragrance rich in monoterpene hydrocarbons (α-pinene and limonene). Our results point towards outcrossing mating strategies for C. etruscus, C. ilvensis, and C. neglectus, whose volatile compounds are known as pollinator attractants. This is in line with their flower architecture, showing a style of variable height, often overtopping stamens. On the other hand, a self-pollination strategy was repeatedly suggested in the literature for C. vernus, marked by flowers with the style deeply inserted in the stamens and also by a completely different flower bouquet.

Aromatic plant species present in the natural Park of Tuscany Archipelago are used as flavoring agents and spices, as dietary supplements and in cosmetics and aromatherapy. The plants are usually collected from wild stands, inducing a depletion of the natural habitat. Therefore, micropropagation of these aromatic plants can play a role in the protection of the natural ecosystem, can guarantee a massive sustainable production and can provide standardized plant materials for diverse economical purposes. The aim of this study is to compare the volatile organic compounds produced by the wild plants with those from in vitro plantlets using headspace solid phase micro-extraction (HS-SPME) followed by capillary gas-chromatography coupled to mass spectrometry (GC-MS). Typical plants of this natural area selected for this work were Calamintha nepeta L., Crithmum maritimum L., Lavandula angustifolia L., Myrtus communis L., Rosmarinus officinalis L., Salvia officinalis L. and Satureja hortensis L. Different explants were used: microcuttings with vegetative apical parts, axillary buds and internodes. Sterilization percentage, multiplication rate and shoot length, as well as root formation were measured. The volatile aromatic profiles produced from in vitro plantlets were compared with those of the wild plants, in particular for C. maritimum, R. officinalis, S. officinalis and S. hortensis. This study indicated that the micropropagation technique can represent a valid alternative to produce massive and sterile plant material characterised by the same aromatic flavour as in the wild grown plants.

Samples of fresh pollen grains, collected from capitula in full bloom from two genotypes of sunflower (Helianthus annuus L.) and characterized by a different color, i.e., white-cream (WC) and orange (O), were analyzed by the HS-SPME (headspacesolid phase microextraction)/GC/MS technique. This study defined for the first time the fingerprint of the sunflower pollen, separated from the disc flowers, to define its contribution to the inflorescence aroma. In the GC/MS fingerprints of the WC and O genotypes, 61 and 62 volatile compounds were identified, respectively. Monoterpene hydrocarbons (34% in O vs. 28% in WC) and sesquiterpene hydrocarbons (37% in O vs. 31% in WC) were ubiquitous in all samples analyzed and represented the main chemical classes. α-Pinene (21% in O vs. 20% in WC) and sabinene (11% in O vs. 6% in WC) were the dominant volatiles, but also a full range of aliphatic hydrocarbons and their oxygenated derivatives gave a decisive contribution to the aroma composition (10% in O vs. 12% in WC). In addition, dendrolasin (3% in O vs. 4% in WC) and some minor constituents such as (E)-hex-2-en-1-ol (0.4% in O vs. 0.1% in WC) were pointed out not only for their contribution to the pollen scent, but also for their well-known role in the plant ecological relationships. Having evaluated two pollen morphs with different carotenoid-based colors, the study sought to highlight also the presence of some volatile precursors or derivatives of these pigments in the aroma. In the circular economy framework, hydrosols, by-products of the essential oil industry, are gaining attention for their potential in waste reduction and resource reuse. This study analyzed hydrosols from six edible flowers, investigating their chemical composition (VOC-Hyd) and antibacterial properties alongside volatile organic compounds of fresh flowers (VOC-Fs) and essential oils (EOs). Antirrhinum majus exhibited ketones as major VOC-Fs (62.6%) and VOC-Hyd (41.4%), while apocarotenoids dominated its EOs (68.0%). Begonia cucullata showed alkanes (33.7%) and aldehydes (25.7%) as primary VOC-Fs, while alkanes were prevalent in both extracts (65.6% and 91.7% in VOC-Hyd and in EOs, respectively). Calandula officinalis had monoterpenoids in VOC-Fs and VOC-Hyd (89.3% and 49.7%, respectively), while its EOs were rich in sesquiterpenoids (59.7%). Dahlia hortensis displayed monoterpenoid richness in both VOC-Fs and extracts. Monocots species' VOC-Fs (Polianthes tuberosa, Tulbaghia cominsii) were esters-rich, replaced by monoterpenoids in VOC-Hyd. P. tuberosa EO maintained ester richness, while T. cominsii EOs contained a significant percentage of sulfur compounds (38.1%). Antibacterial assays indicated comparable minimum inhibitory concentration profiles across VOC-Hyd: B. calcullata and P. tuberosa against Staphylococcus aureus and Salmonella enterica ser. typhimurium, T. cominsii against Escherichia coli and S. enterica, A. majus and C. officinalis against S. aureus, and D. hortensis against S. enterica. (1) Background: The leaves of some plants are reported for their culinary uses, while in edible flowers, they are one of the discarded products in the supply chain. We investigated the volatile profile (VP) and the essential oil (EO) compositions of leaves from 12 Lamiaceae species, of which nine belong to the Mentheae tribe and three to the Ocimeae tribe. (2) Methods: Phytochemical analyses were performed using a GC-MS instrument. (3) Results: More than 53% of the Ocimeae tribe VP was represented by sesquiterpene hydrocarbons (SH), followed by phenylpropanoids, except for O. × citriodorum, where oxygenated monoterpenes (OM) were the second main class. OM prevailed in six species of the Mentheae tribe except for Agastache 'Arcado Pink', Salvia discolor, and S. microphylla, where SH dominated. The EO composition of Ocimeae tribe showed a similar behavior to that of VP concerning the predominant classes. O. basilicum 'Blue Spice' (Ob-BS) was an exception, since it showed oxygenated sesquiterpenes (OS: 29.6%) as a second principal class. Sesquiterpene compounds were also present in a high amount in two species of the Salviinae subtribe (S. microphylla and S. discolor) and two of the Nepetinae subtribe (Nepeta × faasenii and A. 'Arcado Pink'). The remaining species of the Mentheae tribe were characterized by OM. As it mimics olfactory perception, headspace analysis is frequently used for examination of products like chocolate, in which aroma is a key feature. Chemical analysis by itself, however, only provides half the picture, as final consumer's perception cannot be compared to that of a Gas Chromatography-Mass Spectrometry (GC-MS) port, but rather to a panel test assessment. The aim of the present study was the evaluation of combined chemical (by means of headspace solid-phase microextraction and GC-MS) and panel test data (by means of a sensory evaluation operated by 6 untrained panelists) obtained for 24 dark chocolate bars to assess whether these can discriminate between bars from different brands belonging to different commercial segments (hard discount, HD; supermarket, SM; organic bars, BIO). In all samples, with the only exception of one supermarket bar (in which esters exhibited the highest relative abundance), pyrazines were detected as the most abundant chemical class (HD: 56.3–74.2%; BIO: 52.0–76.4%; SM: 31.2–88.9%). Non-terpene alcohols, aldehydes, and esters followed as quantitatively relevant groups of compounds. The obtained data was then subjected to hierarchical cluster (HCA) and principal component (PCA) analysis. The statistical distribution of samples obtained for the chemical data did not match that obtained with panelists' sensorial data. Moreover, although an overall ability of grouping samples of the same commercial origin was evidenced for hard discount and supermarket bars, no sharp grouping was possible. [ABSTRACT FROM AUTHOR]

Volatile organic compounds (VOC) and essential oils (EO) extracted from different organs of Sambucus nigra (leaves, flower buds, flowers, unripe and ripe fruits), were evaluated in this work. VOC analyses highlighted that oxygenated monoterpenes were the major class for both flowers and ripe fruits with percentages of 71.1% and 35.1%, respectively. The ripe fruit showed a considerable amount of linalyl acetate (26.3%) while cis-linalool oxide (pyranoide) (38.6%) was the main constituent of the flowers. This latter compound was also one of the major constituents of the leaf extracts (10.3% of the total identified fractions). The leaves evidenced a high percentage of esters (31.6%) with (Z)-3-hexenol acetate (15.1%) as the major constituent. Alcohols were exclusively represented by 1-hexanol (42.5%) and (E)-3-hexen-1-ol (31.1%) in the unripe fruits, while β-caryophyllene (42.0%) was the characteristic component of the buds. All the studied EOs showed non-terpene derivatives as the main class of volatiles even though its percentage varied in an organ-dependent manner. Alkanes were predominant and especially represented by heneicosane (the highest percentage of 32.9% in the fresh flowers) and nonadecane (the highest percentage of 17.2% in the buds,). Benzaldehyde was the main aldehyde in the leaves (17.8%) while acids, chiefly n-hexadecanoic acid, were also found in quite high amounts in the ripe fruits (14.3%). [ABSTRACT FROM AUTHOR]

The differences in volatile profile of Echinacea purpurea plants not-inoculated (EpC) and inoculated with their endophytes from roots (EpR) and stem/leaves (EpS/L) were analysed and compared by GC-FID/GC–MS in an in vitro model. Non-terpenes and sesquiterpene hydrocarbons were the most abundant classes with an opposite behaviour of EpS/L showing a decreased emission of sesquiterpenes and an increase of non-terpene derivatives. The main compounds obtained from EpS/L were (Z)-8-dodecen-1-ol and 1-pentadecene, while germacrene D and β-caryophyllene were the key compounds in EpC and EpR. For the first time, this work indicates that bacterial endophytes modify the aroma profiles of infected and non-infected in vitro plants of the important medicinal plant E. purpurea. Therefore, our model of infection could permit to select endophytic strains to use as biotechnological tool in the production of medicinal plants enriched in volatile bioactive compounds. Lavandula angustifolia Mill. has a great economic importance in perfumery, cosmetics, food manufacturing, aromatherapy, and pharmaceutical industry. This species finds its phytosociological optimum in the sub-Mediterranean region. Latitudinal and altitudinal gradients are expected to affect species diversification in peripheral alpine populations. In this study, phenotypic traits including morphometric parameters, volatile organic compounds (VOCs) and essential oils (EOs) were analyzed in lavender peripheral populations selected in order to explore different ecological conditions. Plants were cultivated under uniform conditions to observe variations due to the genetic adaptation to native environments and to exclude the short-term response to environmental factors. Results showed qualitatively and quantitatively intra-specific variations in secondary metabolites, mainly along the latitudinal gradient, while minor effect was attributable to the altitude. This latter affected more the morphometric parameters. As the latitude augmented, VOCs showed lower content of monoterpene hydrocarbon (mh) and higher content of oxygenated monoterpenes (om); whereas EOs showed higher content of mh and non-terpene derivatives (nt) and lower content of sesquiterpene hydrocarbons (sh). Lavender aroma and EO composition varied in every population, for a total of 88 and 104 compounds identified, respectively. Eleven and 13 compounds were responsible for 95% of the dissimilarity, with linalool, linalyl acetate and 1,8-cineole as major contributors. As the latitude augmented, linalool decreased and 1,8-cineole increased while linalyl acetate content was unaffected. These results are discussed with regards to the potential adoption of the lavender peripheral alpine populations for the improvement of quality and productivity of lavender cultivations, especially in mountainous areas. The present study aimed to elucidate the salinity influence on the bioactive metabolites of Lawsonia inermis L. (henna) plants. Young henna plants were cultivated under salinity stress with two NaCl concentrations (75 mM and 150 mM) in controlled environmental conditions and the leaves were investigated to check their adaptative responses. The modulation of photosynthetic performance to salinity stress was demonstrated by gas exchange and chlorophyll fluorescence parameters. The partial stomatal closure triggered an enhanced water-use efficiency, and a proline accumulation was observed, leading to an osmotic adjustment. The increased capacity to dissipate the excess excitation energy at photosystem II as heat was associated with changes in chlorophylls, anthocyanins, and carotenoids. The higher antioxidant activity at 150 mM salt level suggested its scavenger role on reactive oxygen species (ROS) dissipation and photoprotection. The reduced CO2 uptake and the higher metabolic costs necessary to sustain the henna tolerance mechanism against high NaCl concentration negatively affected lawsone production. Leaf volatile organic compounds (VOCs) showed changes in the amount and composition of VOCs with increasing salinity level. Overall, this study revealed efficient physiological and biochemical adaptations of henna leaves to salt stress despite an altered production of important economic metabolites such as lawsone. The profile of volatile organic compounds (VOC), the composition of the essential oils (EOs) and the micromorphology of the leaf trichomes were investigated in two rosemary hybrids, Salvia×mendizabalii (Sagredo ex Rosúa) Roma-Marzio & Galasso (S × m) and Salvia x lavandulacea (de Noé) Roma-Marzio & Galasso (S × l). The phytochemical analysis highlighted the dominance of monoterpene hydrocarbons in both aroma profiles (65.8 % in S × m and 77.9 % in S × l , respectively), although they displayed different main compounds except for α-pinene (28.2 % in S × m and 24.5 % in S × l). In S × m the EOs extracted from both fresh and dried leaves exhibited camphor (29.7 % and 25.9 %, respectively), α-pinene (21.0 % and 18.2 %, respectively) and 1,8-cineole (11.1 % and 13.6 %, respectively) as major compounds. Noteworthy is the occurrence of β-pinene (2.6 %) in the EO dried samples, whereas sabinene and limonene were exclusive of the fresh samples. In S × l the EOs from the fresh and dried leaves shared the same main compounds: camphor (24.0 % and 27.5 %, respectively), myrcene (14.9 % and 14.8 %, respectively) and α-pinene (13.1 % and 12.2 %, respectively). The micro-morphological observations on leaves proved the occurrence of non-glandular dendritic hairs with smooth cuticle in both hybrids. The glandular trichomes include three main morphotypes: peltate, short capitate and medium-long capitate. The peltate and the short capitates are common to both hybrids, however in the peltate the number of the head secreting cells is different, eight in S × m and sixteen in S × l. Volatilomes emitted from edible flowers of two species of Agastache (A. aurantiaca (A.Gray) Lint & Epling, and A. mexicana (Kunth) Lint & Epling) and from two hybrids (Agastache 'Arcado Pink' and Agastache 'Blue Boa') were investigated using a solid-phase microextraction technique as well as the extraction of its essential oils. Oxygenated monoterpenes were almost always the predominant class (>85%) of volatile organic compounds (VOCs) in each sample of A. aurantiaca, A. 'Blue Boa' and A. mexicana, with the exception of A. 'Arcado Pink' (38.6%). Pulegone was the main compound in A. aurantiaca (76.7%) and A. 'Blue Boa' (82.4%), while geranyl acetate (37.5%) followed by geraniol (16%) and geranial (17%) were the principal ones in A. mexicana. The essential oil composition showed the same behavior as the VOCs both for the main class as well as the major constituent (pulegone) with the same exception for A. mexicana. Total soluble sugars, secondary metabolites (polyphenols, flavonoids and anthocyanins) and antioxidant activity were also investigated to emphasize the nutraceutical properties of these edible flowers. The essential oils extracted from the peels of two Tuscany Citrus of the Massa province have been characterised. Moreover, the flavedo of these species has been used in the production of two Citrus olive oils (COOs) obtained with an innovative method in which the citrus peels are cryomacerated and then pressed with the olives. The presence of functional compounds, such as carotenoids, naringenin and minor phenolics, classifies these COOs as nutraceuticals with the potential to develop enriched foods able to promote a healthy diet. Moreover, the increased presence of tyrosol and hydroxytyrosol, compared to the unflavoured oil, further highlights the nutritional value to the two COOs, being these phenolic compounds recognized as good possible therapeutic candidates for the inhibition of neurodegenerative diseases as the Parkinson's disease. The volatiles of different pomegranate plant parts were extracted using headspace solid phase micro-extraction (HS-SPME) and analyzed for the first time by gas chromatography-mass spectrometry (GC–MS) and flame ionization detector (GC-FID). All the data were submitted to multivariate statistical analysis evidencing many differences amongst the selected plant parts. Sesquiterpene hydrocarbons were the most abundant class in the aroma of leaves and in all the flower organs where the main constituents were β-caryophyllene, ( E , E )-α-farnesene and trans -α-bergamotene. Stem aroma showed similar content of sesquiterpene hydrocarbons and non-terpenes with α-humulene as main compound. • Nemo's Garden® underwater biospheres are a green, alternative agriculture system. • Underwater basil did not show micromorphological changes of the leaf indumentum. • Relevant differences were detected in the essential oil and head space compositions. • More photosynthetic pigments and polyphenols were produced in the underwater plants. • Basil seems well adapted: studies on other species are needed to evaluate a scale-up. The need for alternative cultivation methods is urgent for regions of the world where cultivable land is scarce: underwater areas are unexploited and vast. Nemo's Garden® Project aims at creating a green and alternative agriculture system: its biospheres are underwater greenhouses, developed for areas where plants growth is difficult in terrestrial conditions, due to climate change. Basil was chosen as model plant to study its phytochemical, physiological, and micromorphological parameters, in comparison with the same plants grown in terrestrial conditions in the Sanremo Research Centre for Vegetable and Ornamental Crops (CREA-Centro di Ricerca Orticoltura e Florovivaismo) greenhouse. While the micromorphological analyses showed no detectable differences between control and biospheres samples, the phytochemical investigations evidenced a switch of the essential oil chemotype from methyl eugenol/linalool to methyl eugenol, respectively. The headspaces were also different: sesquiterpenes dominated the biospheres samples, whereas oxygenated monoterpenes accounted for half the control sample emission. Differences also emerged in the physiological investigation: total chlorophyll, total carotenoids and total polyphenols were present in higher amounts in the biospheres samples, with a 31.52% and 13.3% increase in the antioxidant activity and polyphenols content, respectively. Basil was well adapted in the biosphere environment, whose influence should be studied in different species to assess the viability of an industrial scale-up of the Nemo's Garden®.
