Severe acute respiratory syndrome (SARS) emerged in China late in 2002 and spread globally; causing 8,422 reported infections with 916 (11%) fatalities. The causative agent was identified as a coronavirus (SARS-CoV), and related viruses found in palm civets (Paguma larvata), raccoon dogs (Nyctereutes procyonoides), and insectivorous bats in Asia clustered phylogenetically with SARS-CoV. In this study 705 African bat sera from four different locations in Southern and Central Africa were tested. Antibody reactive with SARS-CoV antigen was detected by serological assays in 6.7% (47/705) of bat sera comprising seven out of 26 bat species implying that African bats may harbour agents related to SARS-CoV. Highest prevalences could be found in the fruit bat Rousettus aegyptiacus (16.4%) and the insectivorous bat Mops condylurus (12.2%). Moreover, in order to define and characterize target cells of SARS- CoV, the susceptibility of 23 different permanent and primary eukaryotic cell cultures to SARS-CoV was studied. In addition to monkey kidney Vero E6 cells, SARS-CoV infection could also be demonstrated in two pig cell lines (POEK, PS) and one human hepatoma cell line (Huh-7) using an indirect immunofluorescence assay (IFA) and a quantitative real-time reverse transcriptase PCR. The identification of liver and kidney cell lines as putative target cells for SARS-CoV is coherent with pathological findings in SARS-CoV patients. In all susceptible cell lines mRNA of the angiotensin-converting enzyme 2 (ACE2), the functional receptor for SARS-CoV infection, could be detected by RT-PCR. The presented results show that there is a correlation between the abundance of ACE2 mRNA and SARS-CoV susceptibility. Group I coronaviruses like the novel HCoV-NL63 are commonly associated with respiratory tract infections. Due to the lack of knowledge on HCoV-NL63 proteins this thesis focuses on the structural membrane (M) and nucleocapsid (N) proteins and in particular on the hitherto uncharacterized open reading frame 3 (ORF3), a homologous protein of SARS-CoV ORF3a. This SARS-CoV unique ORF3a was shown to be a triple-membrane spanning structural glycoprotein (O-glycosylated) able to form potassium ion channels that modulate virus release. The in silico analysis of the deduced amino acid (aa) sequence of HCoV-NL63 ORF3 comprising 225 aa (26 kDa) also revealed a triple-membrane spanning protein. By means of of an N-terminal FLAG tagged ORF3 and an ORF3 C-terminus specific antisera an extracellular N-terminus and a cytosolic C-terminus could be identified. By in vitro translation and subsequent endoglycosidase H digestion both ORF3 and M protein were revealed to be N-glycosylated. By means of produced antisera against ORF3, M and N, a protein expression in infected cells could be confirmed by Western blot analysis and IFA. Additionally, ORF3 could be localized at the plasma membrane, the endoplasmatic reticulum Golgi intermediate compartment (ERGIC) and the Golgi complex in transiently transfected and infected cells by confocal laser scanning microscopy. Using green fluorescent protein (GFP) tagged structural proteins envelope (E), M and N it could be shown that FLAG- ORF3 co-localizes extensively with GFP-E and GFP-M within the ERGIC where virus assembly and budding takes place. The resemblance of HCoV-NL63 ORF3 with SARS-CoV ORF3a is intriguing and encourages a more detailed analysis., Das schwere akute respiratorische Syndrom (SARS), welches 2002 in China ausbrach, führte zu weltweit gemeldeten 8422 Infektionen mit 916 (11%) Todesfällen. Der Auslöser konnte als ein Coronavirus (SARS-CoV) identifiziert werden. Verwandte Coronaviren, welche in Schleichkatzen (Paguma larvata), Maderhunden (Nyctereutes procyonoides) sowie in asiatischen Fledermäusen (Rhinolophus spec.) gefunden wurden, wiesen eine nahe phylogenetische Beziehung auf. Um zu untersuchen, ob SARS-ähnliche Viren auch außerhalb Asiens in Fledermäusen vorkommen, wurden in der vorliegenden Studie 705 Serumproben von afrikanischen Fledermäusen analysiert. Insgesamt wiesen 6,7% (47/705) dieser Serumproben Antikörper auf, die mit einem SARS-CoV Antigen reagierten. Die höchsten Prävalenzen konnten mit 16,4% in den frugivoren Flughunden der Spezies Rousettus aegyptiacus und in der insectivoren Fledermausspezies Mops condylurus (12,2%) festgestellt werden. Die Ergebnisse deuten darauf hin, dass akute oder persistierende Infektionen mit Coronaviren der Gruppe IV (SARS-CoV) in afrikanischen Fledermäusen wahrscheinlich sind. Darüber hinaus wurden in weiteren Studien mögliche Zielzellen für das SARS-CoV charakterisiert, indem die Suszeptibilität von 23 verschiedenen eukaryotischen Zelllinien und primären Zellen für das SARS-CoV analysiert wurde. Neben der Affennierenzelllinie Vero E6 wurden SARS-CoV Infektionen ebenfalls in zwei Schweinezelllinien (POEK, PS) und einer humanen Leberzelllinie (Huh-7) mittels indirekter Immunfluoreszenztests (IFT) und einer quantitativen real-time reverse Transkriptase PCR nachgewiesen. In allen suszeptiblen Zelllinien wurde mRNA des Angiotensin-converting Enzyms 2 (ACE2), welches als funktioneller Rezeptor für das Virus bekannt ist, detektiert. Die Resultate zeigten zudem, dass es eine Korrelation zwischen dem Vorkommen von ACE2 mRNA und einer SARS- CoV Suszeptibilität gibt. Gruppe I Coronaviren wie das kürzlich entdeckte HCoV-NL63 werden allgemein mit Infektionen der Atemwege in Verbindung gebracht. Bisher ist sehr wenig über die HCoV-NL63 Strukturproteine bekannt, so dass in dieser Arbeit das Membran- (M) und das Nucleocapsid (N) Protein sowie insbesondere das bislang uncharakterisierte Offene Leserahmen 3 (ORF3) Protein untersucht wurden. Bereits bekannt ist, dass ein homologes Protein des SARS-CoV, genannt ORF3a, ein dreifach membrandurchspannendes Glykoprotein ist. Durch die Bildung von Homotetrameren ist dieses in der Lage, Kaliumkänale zu bilden, die die Virusfreisetzung modulieren. Eine in dieser Arbeit präsentierte in silico Analyse der abgeleiteten Aminosäuresequenz (225 Aminosäuren; 26 kDa) des HCoV-NL63 ORF3 zeigte ebenfalls ein dreifach membrandurchspannendes Protein. Mit Hilfe einer N-terminalen FLAG Markierung und eines ORF3 C-terminalen spezifischen Antiserums konnte ein extrazellulärer N-Terminus und ein cytosolischer C-Terminus identifiziert werden. Mittels in vitro Translation und nachfolgendem Endoglykosidase H Verdau wurde für das ORF3 und das M Protein eine N-Glykosylierung nachgewiesen. Durch Antiseren gegen das ORF3, M und N Protein konnte die Expression der viralen Proteine mittels IFT und Western Blot Analysen bestätigt werden. Zudem wurde das ORF3 Protein mit Hilfe der konfokalen Laserscanning Mikroskopie an der Plasmamembran, im ERGIC und im Golgi Apparat in transient transfizierten und in infizierten Zellen lokalisiert. Durch Ko-Transfektionsstudien von GFP markierten Strukturproteinen Envelope (E), M und N mit FLAG markiertem ORF3 in humanen Zelllinien konnte eine Ko-Lokalisation von GFP-E und GFP-M innerhalb des ERGIC Kompartiments gezeigt werden. Das ORF3 Protein von HCoV-NL63 weist demnach eine sehr große Ähnlichkeit zum ORF3a Protein des SARS-CoV auf, so dass weitere Analysen hinsichtlich der Interaktion mit anderen viralen und zellulären Proteinen sowie der generellen Funktion folgen sollten.
