The temporal relationship between changes in cervical dilatation, uterine electromyographic (EMG) activity, and maternal plasma concentrations of estradiol 17beta (E(2)), progesterone (P(4)), and 13,14-dihydro-15-keto-prostaglandin-F(2alpha) (PGFM), was investigated in six parturient cows. Calving was induced with a single injection of a synthetic analogue of prostaglandin F(2alpha) (PG) on Day 274 of gestation. Cervical dilatation was measured continuously by measuring the transit time between two implanted ultrasound crystals while at the same time uterine EMG activity was measured through two silver electrodes sutured on the myometrial surface until the expulsive stage of calving had been reached. In blood samples collected at 4-h intervals, starting at the moment of PG injection, the mean plasma E(2) concentration gradually increased and was significantly elevated at 28 h after PG injection. At 4 h after PG treatment, the mean P(4) concentration had dropped significantly and continued to decrease until a value of around 1 ng/ml was reached, where it stayed until the onset of expulsion. Mean plasma PGFM concentrations increased steadily after PG injection, reaching significantly elevated concentrations at 20 h after treatment. In the five cows that delivered calves in anterior positions, uterine EMG activity, expressed as root mean square (RMS in microV), started to increase at a mean interval (+/- SD) of 13.1 +/- 3.7 h following PG treatment. The increase in EMG activity was significantly correlated with changes in plasma PGFM concentrations. In these cows, dilatation of the caudal cervix started after a mean (+/- SD) interval of 28.5 +/- 1.5 h following PG treatment and dilatation progressed at a mean (+/- SD) rate of 2.25 +/- 0.24 cm/h. In one cow with a calf in the posterior position, uterine EMG activity and dilatation started at 15.8 h and 31.8 h, respectively, after induction of calving. We conclude that a predictable sequence of physiological changes occurs around induction of calving, which allows specific timing of future studies on cellular and biochemical changes within the cervix during parturition.

The mRNA expression patterns of activin beta(A) and follistatin in the uterus and embryo, the mRNA expression of the activin receptor II in the embryo, and the localization in the uterus of the immunoreactive activin beta(A) and the receptor II proteins in the uterus were examined at gestation days 7-12 after ovulation in pig. Activin was located predominantly at the mesometrial side of the uterus during all stages of pregnancy studied. Follistatin mRNA was absent in the uterus during these stages, suggesting that activin of uterine origin is not inhibited by intra-uterine follistatin. The receptor was localized throughout the glandular and luminal epithelium of the uterus. In the embryo, activin was expressed predominantly in the epiblast before unfolding, but after unfolding of the epiblast activin expression shifted to the trophoblast. The expression pattern of follistatin mRNA was contrarily to that of activin, i.e., before unfolding predominantly in the trophoblast (days 8-9), and shifted to the epiblast at day 10. During streak stages, follistatin was detected in the node and primitive streak. Activin receptor II mRNA was first detected at day 8 in the embryoblast. At day 11, it was expressed in trophoblast cells near the epiblast, and in the first ingressing mesoderm cells. During the streak stages, it was expressed predominantly in the trophoblast. The presence of activin and its receptor in uterine epithelium and early embryonic tissues indicate that both embryonic and uterine activin are involved in intra-uterine processes, such as attachment and early embryonic development. To determine the effect of dose and day of oxytocin treatment on intrauterine pressure, 6 normal mares were treated with 10 or 25 IU oxytocin 2 days before ovulation, on the day of ovulation and 2 days after ovulation. Intrauterine pressure (IUP) was measured using micro-tip-catheters (one placed intrauterine, a second and third serving as reference sensors in the vagina and external to the mare) and transmitted by telemetry for 30 min to establish a baseline before saline was administered, iv, and for an additional 30 min after saline administration. Oxytocin was then given, iv, and IUP was recorded for 60 min. No change in IUP was observed after saline injection. The administration of both 10 (n=16) and 25 (n=10) IU oxytocin induced a response (P<0.01). The intensity of response depended on the day of administration (P<0.01) and the dose of oxytocin (P<0.001). The variation of response was significantly greater after 10 IU oxytocin (CV 15.78%) compared with 25 IU oxytocin (CV 6.42%). The uterine response was greatest on Day 2 prior to ovulation and lowest on Day 2 after ovulation. The response was negatively correlated to increasing plasma progesterone (10 IU oxytocin: r = -0.435, 25 IU oxytocin: r = -0.265). There was no correlation between the uterine response and plasma estradiol-17beta concentration (P<0.01). In conclusion the results of this study show that oxytocin administration to mares before ovulation provides a greater response than after ovulation. A decline in the intensity of response after ovulation can be compensated for with a higher dose of oxytocin. Furthermore, the use of the multiple catheter technique is an effective method for assessing changes in uterine pressure.

Hydrometra is a pathological condition of the uterus which is characterized by accumulation of aseptic fluid in the presence of a persistent corpus luteum. It forms a major cause of subfertility in goats. Active immunization against prostaglandin F2 alpha (PGF2 alpha), the luteolytic hormone in this species, was used to explore the possibility for artificial induction of this pseudopregnant condition. During the breeding season, 11 goats (group I) were immunized with 5 mg PGF2 alpha-ovalbumin conjugate in Freund's complete adjuvant, 4 goats were control-immunized (group CI) and 5 goats remained untreated (group C). Jugular blood samples were taken twice a week (monday and thursday) for measurements of plasma progesterone and binding of 3HPGF2 alpha. In conjunction with blood sampling, transcutaneous ultrasonographic examination of the uterus took place to detect the presence of fluid in the uterus. Before and immediately after immunization, the mean (+/-SD) duration of luteal phases (progesterone concentrations > or = 1 ng ml-1) was 16.7 +/- 1.6 (n = 39), 17.8 +/- 1.3 (n = 23) and 16.9 +/- 1.1 (n = 18) days in animals of group I, CI and C respectively. Ten goats of group I developed an antibody titre. Persistence of luteal function (mean duration +/-SD: 150.3 +/- 23.5 days) occurred in 6 of these animals and in 1 goat of group CI. Accumulation of fluid in the uterus in group I was first observed between day 31 and 38 of the prolonged luteal phase. Discharge of uterine fluid occurred as soon as the plasma progesterone concentration reached a level lower than 0.5 ng ml-1. It is concluded that immunization against PGF2 alpha is an effective method to induce pseudopregnancy in goats, providing a model for studies on luteal maintenance and uterine function in the absence of a conceptus.

A 5-MHz sector transducer (experiment 1) and a 7.5 MHz linear-array transducer (experiment 2) were used for early pregnancy diagnosis in two Hungarian dairy herds under field conditions. In the first experiment, ultrasound scanning was conducted three times at 3 or 4-day intervals between 24 and 33 days after artificial insemination (AI), while in the second experiment, it was performed two times at 7-day intervals between 27 and 38 days post-insemination. Significantly more incorrect non-pregnancy diagnoses were made between 24 and 33 days (first experiment) and between 27 and 38 days (second experiment) after AI in cows in which the uterus was located far cranial to the pelvic inlet than in animals in which the uterus was located within or close to the pelvic inlet at the first ultrasonographic examination.

The use of a transabdominal or a transrectal scanning technique to visualize the uterus in goats is discussed. The results of sonographic imaging of normal (non-pregnant, pregnant, postpartum) and abnormal (endometritis, hydrometra, fetal death) uterine structures are reviewed. In the goat, special attention must be paid to distinguishing between the sonographic image of hydrometra and early pregnancy. A case of hydrometra that developed in the presence of three dead fetuses is reported. It is concluded that, under field conditions, transabdominal scanning of goats, performed between days 40 and 70 after mating, yields information about pregnancy, fetal viability, and single or multiple pregnancy, while at the same time making possible a reliable differential diagnosis of pregnancy and hydrometra.

15-Hydroxy-prostaglandin dehydrogenase activity (PGDH) was measured in endometrium, allanto-chorion, allanto-amnion and the placenta from six cows during late gestation and six parturient cows during caesarean section. Under saturated substrate conditions PGDH activity was lower than 138 pmol g wet tissue-1 min-1 in all uterine tissues in both groups. Human placental reference samples showed a PGDH activity of 16,800 +/- 1200 pmol per g wet weight min-1 (n = 4). Under subsaturational conditions enzyme activity was demonstrated in endometrium and both fetal membranes. These results indicate that PGF2 alpha catabolism in bovine uterine tissue on day 262 and during parturition, contributes little to changing plasma PGFM concentrations.

In this study of the pregnant uterus of the ewe and the pygmy goat, the simultaneous use of real-time ultrasound scanning and electromyography is introduced. In unanaesthetized animals it was possible to visualize the uterine wall, the placentomes, parts of the fetal skeleton, the fetal kidneys, different parts of the fetal stomach and several structures of the fetal heart and vascular system. From these features the intra-abdominal position of the fetus could be determined. It was observed that Episodes of Myometrial Electrical Activity (EMEA's), as registered by chronically implanted uterine surface electrodes, coincided with passive displacements of the fetus in the direction of the maternal pelvis. These passive displacements could clearly be discerned from active fetal body movements. In both species the number of active fetal movements was significantly reduced during and/or shortly after an EMEA. In one of the pygmy goats, in which three different types of movement were discerned, this reduction of fetal movements could be studied in more detail. It is concluded that real-time ultrasound scanning of the fetus in small ruminants might contribute to studies of the complex maternal-fetal interactions that exist during late gestation.

Uterine position and presentation of fetuses in 17 minipigs during pregnancy were compared with their order and presentation at birth. Fetuses from 1 or both uterine horns were marked between day 80 and day 105 of gestation by 1 or more transuterine injections of either a 2.5% solution of dianil blue (3 animals) or a radiopaque substance (14 animals). Roentgenograms of the newborn pigs, which were ear-notched immediately after birth, enabled making individual identification. Pigs seemed to be delivered randomly from the 2 uterine horns. They either followed a neighbor from the same horn (n = 42) or followed a pig from the contralateral horn (n = 52). Only in 3 dams did 1 uterine horn become empty before delivery from the other horn started. In 4 dams, 1 pig passed its neighbor from the same horn, thereby being born earlier than expected. Moreover, passing of pigs could have occurred and gone undetected in those dams from which the pigs of only 1 horn were marked. This passing might have taken place within the uterine body. From 18 of 95 pigs (19%) , the presentation at birth differed from the one observed at laparotomy.

Bipolar electrodes were implanted into the myometrium of three cycling ewes. Natural mating took place 35, 47 and 91 days after surgery, and all three animals became pregnant. Two of them were slaughtered 18 and 40 days after mating, the third delivered twin lambs 144 days after mating. The uterine electrical activity was recorded during pregnancy and parturition. From the 3rd until the 27th day after mating the uterus appeared to be quiescent. From the 5th wk of pregnancy until induction of parturition on day 141, electrical activity was observed to occur in episodes of 5--10 min, with a frequency of 1--3 per 2 h. The episodes are separated by periods of inactivity. The amplitudes of the electrical discharges increased gradually during pregnancy, although the plasma progesterone levels increased also. It is concluded that (1) the implanted electrodes did not interfere with normal reproductive processes such as conception, maternal recognition of pregnancy, implantation and fetal development: (2) a distinct pattern of electrical activity can be detected as early as the 5th wk of pregnancy.

To quantify the pattern of myometrial activity during gestation and parturition, one bipolar electrode was implanted on each uterine horn of four bilaterally pregnant pygmy goats between days 102 and 120 of gestation. After a recovery period, electromyographic recordings were made for at least six hours per day and continuously when parturition was judged to be imminent. During late gestation myometrial activity occurred as discrete episodes of myometrial electrical activity (EMEA) with a mean duration ranging from 6.2 +/- 2.07 to 8.3 +/- 1.60 minutes. The mean interval between two successive EMEAs ranged from 45.8 +/- 19.95 to 74.7 +/- 42.27 minutes. In three of the four goats these characteristics were not significantly different from the two uterine horns. Parturition was preceded by a prolonged period (eight to 12 hours) of myometrial quiescence. It was only from 19 to 15 hours before expulsion of the first kid that total duration of EMG activity increased. This finally resulted in the regular occurrence of bursts which occupied 25 to 30 per cent of the recording time. The results demonstrate that, as in the sheep, cow and pig, the myometrium is active during late pregnancy. It is postulated that luteolysis coincides with the prolonged period of myometrial quiescence which precedes the onset of the parturient pattern of uterine activity.

During late pregnancy (greater than 50 days) the pattern of uterine electrical activity is characterized by episodes of activity lasting 3-10 min and recurring at a low frequency (maximum 2.5/h). During the last 7 days before delivery there is a progressive qualitative change in activity which is correlated with the decrease in plasma progesterone concentration. Together with significant quantitative changes in uterine activity which occur during the last 24 h before parturition and after the sharp fall in progesterone concentration this suggests that progesterone plays a crucial role in the process of parturition in the bitch. Experiments with a calcium-channel antagonist have demonstrated the important role of Ca2+ ions in uterine contractility in dogs, but further investigations will be necessary to illustrate the clinical significance of hypocalcaemia in relation to primary uterine inertia. By marking all fetuses in utero before term by means of a radio-opaque substance, it was demonstrated that, when one or more pups were left in each uterine horn after birth of a pup, 78.2% of the pups were expelled from the contralateral horn. Assessment of the blood gas and acid-base status in spontaneously born puppies indicated that the initial state of acidosis is more severe than in several other domestic species. A mild to severe combined respiratory-metabolic acidosis occurs in almost every newborn puppy.

Uterine electromyography was performed by means of chronically implanted surface electrodes in 3 Pony mares during spontaneous oestrous cycles and following luteolysis induced by a prostaglandin analogue (fluprostenol). Three distinct patterns were recognized during the oestrous cycle. (1) During oestrus well defined phases of activity with closely grouped high-amplitude spikes were separated by long periods (10-45 min) of complete inactivity. (2) During dioestrus more diffuse phases of activity with low-amplitude spikes were separated by variable periods of relative inactivity. (3) During luteolysis, short and frequently occurring phases of activity were propagated between the two electrodes on one uterine horn; a similar pattern also occurred between 1 and 3 h after injection of fluprostenol. Peripheral plasma progesterone, but not total inconjugated oestrogen, concentrations were closely related to characteristics of the myographic activity during the cycle. Insemination during oestrus and injection of fluprostenol during dioestrus caused a marked and prolonged increase in myometrial electrical activity. Almost any non-specific environmental stimulus, including entry by palpation of the genital tract per rectum and vaginoscopic examination, but these were of brief duration and the normal resting pattern of activity was quickly re-established after completion of the manipulations.
