The mitochondria are the main source of cellular energy production and have an important role in development, fertility, and thermal limitations. Adaptive mitochondrial DNA mutations have the potential to be of great importance in determining aspects of the life history of an organism. Phylogenetic analyses of the globally invasive marine ascidian Didemnum vexillum using the mitochondrial cytochrome c oxidase 1 (COX1) coding region, revealed two distinct clades. Representatives of one clade (denoted by 'B') are geographically restricted to D. vexillum's native region (north-west Pacific Ocean, including Japan), whereas members of the other clade (denoted by 'A') have been introduced and become invasive in temperate coastal areas around the world. Persistence of clade B's restricted distribution may reflect it being inherently less invasive than clade A. To investigate this we sought to determine if the two clades differ significantly in other mitochondrial genes of functional significance, specifically, alterations in amino acids encoded in mitochondrial enzyme subunits. Differences in functional mitochondrial genes could indicate an increased ability for clade A colonies to tolerate a wider range of environmental temperature. Full mitochondrial genomic sequences from D. vexillum clades A and B were obtained and they predict significant sequence differences in genes encoding for enzymes involved in oxidative phosphorylation. Diversity levels were relatively high and showed divergence across almost all genes, with p-distance values between the two clades indicating recent divergence. Both clades showed an excess of rare variants, which is consistent with balancing selection or a recent population expansion. Results presented here will inform future research focusing on examining the functional properties of the corresponding mitochondrial respiration enzymes, of A and B clade enzymes. By comparing closely related taxa that have differing distributions it is possible to identify genes and phenotypes suited to particular environments. The examination of mitochondrial genotypes, and associated enzyme functioning, across populations may aid in our understanding of thermal tolerance and environmental adaptation., (Copyright © 2014 Elsevier B.V. Considerable progress in our understanding of the population genetic changes associated with biological invasions has been made over the past decade. Using selectively neutral loci, it has been established that reductions in genetic diversity, reflecting founder effects, have occurred during the establishment of some invasive populations. However, some colonial organisms may actually gain an ecological advantage from reduced genetic diversity because of the associated reduction in inter-colony conflict. Here we report population genetic analyses, along with colony fusion experiments, for a highly invasive colonial ascidian, Didemnum vexillum. Analyses based on mitochondrial cytochrome oxidase I (COI) partial coding sequences revealed two distinct D. vexillum clades. One COI clade appears to be restricted to the probable native region (i.e., north-west Pacific Ocean), while the other clade is present in widely dispersed temperate coastal waters around the world. This clade structure was supported by 18S ribosomal DNA (rDNA) sequence data, which revealed a one base-pair difference between the two clades. Recently established populations of D. vexillum in New Zealand displayed greatly reduced COI genetic diversity when compared with D. vexillum in Japan. In association with this reduction in genetic diversity was a significantly higher inter-colony fusion rate between randomly paired New Zealand D. vexillum colonies (80%, standard deviation ±18%) when compared with colonies found in Japan (27%, standard deviation ±15%). The results of this study add to growing evidence that for colonial organisms reductions in population level genetic diversity may alter colony interaction dynamics and enhance the invasive potential of newly colonizing species.

Botrylloides simodensis, belonging to the family Botryllidae, is a common species on the rocky shore around Shimoda. Since this species was first reported in 1981, it has been noticed that some B. simodensis colonies have distinctly different ground colors and color patterns from colonies including type specimens (holotype and all of paratypes) of B. simodensis. However, as there is no remarkable difference among them in morphology other than the colonial colors and color patterns, they have been considered to belong to the same species. In the present work, colonies of B. simodensis sensu lato were divided into three groups, two major and one minor, based on their color-morphs and periods of sexual reproduction. In one of the major groups, B. simodensis sensu strict, the period of sexual reproduction was limited from July to September, as shown in the original description. The other major group engaged in sexual reproduction for much longer than the former group, and they frequently made clusters of vascular vessels and underwent vascular budding. Therefore, the group with the long period of sexual reproduction was described as a new species of the genus Botrylloides here. In addition, phylogenetic analysis based on mtDNA sequences of these groups indicated that each group had a peculiar sequence, and that they were genetically separated.

We investigated whether Vasa was a germline-specific marker in the colonial ascidian Botryllus primigenus, and whether it was inducible epigenetically in the adult life span. We cloned a Botryllus Vasa homologue (BpVas). The deduced open reading frame encoded 687 amino acid residues. It was expressed specifically by germline cells such as the loose cell mass, oogonia and juvenile oocytes in the ovary, and the primordial testis (compact cell mass), spermatogonia and juvenile spermatocytes in the testis. The loose cell mass, the most primitive germline cells, showed an ultrastructure of undifferentiated cells known as hemoblasts. The hemoblasts did not contain electron-dense materials or a mitochondrial assembly in the cytoplasm. These organelles appeared later in the oogonia and oocytes. When the loose cell mass and developing germ cells were eliminated by extirpating all zooids and buds from the colonies, BpVas transcripts disappeared completely from the vascularized colonies. After 14 days, when the colonies regenerated by vascular budding, BpVas-positive cells reappeared in some cases, and in 30 day colonies, BpVas-positive germ cells were observed in all the regenerated colonies. These results show that in B. primigenus, germ cells are inducible de novo from the Vasa-negative cells even at postembryonic stages.

The allometric scaling of metabolic rate of organisms, the three-quarters power rule, has led to a questioning of the basis for the relation. We attacked this problem experimentally for the first time by employing the modular organism, the ascidian that forms a single layered flat colony, as a model system. The metabolic rate and colony size followed the three-quarters power relation, which held even after the colony size was experimentally manipulated. Our results established that the three-quarters power relation is a real continuous function, not an imaginary statistical regression. The fact that all the hypotheses failed to explain why the two-dimensional organism adhered to the three-quarters power relation led us to propose a new hypothesis, in which the allometric relation derives from the self-organized criticality based on local interaction between modulus-comprising organisms.

Ascidians inhabit both shallow water and the deep sea. The phylogenetic position of deep-sea ascidians has not been sufficiently investigated because of their unusual habitats. The family Octacnemidae is one such enigmatic deep-sea ascidian. In this report, we determined the sequences of the 18SrDNA and a mitochondrial protein gene of Megalodicopia hians belonging to the family Octacnemidae, and we analyzed its phylogenetic relationship with other ascidians. A phylogenetic relationship of this family with the families Cionidae and/or Corellidae has been suspected based on a small number of morphological characteristics. However, our results suggested that M. hians has a close relationship to the family Corellidae and might originate from them. This is the first report of the molecular phylogenetic analysis of a deep-sea ascidian.

The morphology and life history of a strange and unidentified botryllid ascidian were investigated. This ascidian was first collected from the stony shore of Ebisu Island in Shimoda, a city on Izu peninsula in central Japan. Unlike other botryllid ascidians, whose colonies are flat and smooth, this ascidian's colonies are rugged. In each colony, zooids are arranged into several oval systems, each of which has a thick part containing zooids and very thin parts that do not. The arrangement of ovary and testis in this species is the same as in other species of the genus Botryllus; the ovary is situated anterior to the testis. The embryo of this ascidian develops in the peribranchial cavity of its mother zooid without any brooding organs, as is the case with Botryllus scalaris and Botryllus puniceus. Meanwhile, the results of cut colony assay experiments did not show the existence of colony specificity in this ascidian. Even when two syngeneic colonies were brought into contact at their growing edges, none fused together. On the other hand, when two colonies were brought into contact with each other at their cut surfaces, they always fused into a single colony, regardless of their origin. Therefore, this species may be the only species that lacks colony specificity among the botryllids studied so far.

The metabolic rate and its scaling relationship to colony size were studied in the colonial ascidian Botrylloides simodensis. The colonial metabolic rate, measured by the oxygen consumption rate (V(O2) in millilitres of O(2) per hour) and the colony mass (wet weight M(w) in grams) showed the allometric relationship (V(O2) = 0.0412 M(w)(0.799). The power coefficient was statistically not different from 0.75, the value for unitary organisms. The size of the zooids and the tunic volume fraction in a colony were kept constant irrespective of the colonial size. These results, together with the two-dimensional colonial shape, excluded shape factors and colonial composition as possible causes of allometry. Botryllid ascidians show a takeover state in which all the zooids of the parent generation in a colony degenerate and zooids of a new generation develop in unison. The media for connection between zooids such as a common drainage system and connecting vessels to the common vascular system experienced reconstruction. The metabolic rate during the takeover state was halved and was directly proportional to the colonial mass. The scaling thus changed from being allometric to isometric. The alteration in the scaling that was associated with the loss of the connection between the zooids strongly support the hypothesis that the allometry was derived from mutual interaction among the zooids. The applicability of this hypothesis to unitary organisms is discussed.

The morphology and life history of a new species of the genus Botryllus belonging to the family Botryllidae are described in detail. This ascidian was collected from the stony shore in the cove near Shimoda Marine Research Center, University of Tsukuba (Shimoda, Shizuoka Prefecture, Japan). The ascidian colony was easily distinguished from colonies of other botryllids because it was very thin and bright pink in color. The arrangement of ovary and testis in this ascidian was the same as that in other species of the genus Botryllus. This ascidian was prolific, with 1-5 embryos on each side of a zooid, and the embryos of this ascidian developed in the peribranchial cavity without any brooding organs as in Botryllus scalaris. We observed the processes and features of the allorecognition reaction in colony specificity and found that allorejection occurred after fusion of the vascular system between two incompatible colonies. This manner of allorejection is also shown in B. scalaris and Botryllus delicatus; however, the reaction speed of allorejection is faster than that of B. delicatus and similar to that of B. scalaris. These results indicate that this ascidian might be closely related to B. scalaris.

Ultrastructures of circulating hemocytes were studied in 9 botryllid ascidians. The hemocytes are classified into five types: hemoblasts, phagocytes, granulocytes, morula cells, and pigment cells. These five types are always found in the 9 species. They should represent the major hemocyte types of the circulating cells in the blood. Hemoblasts are small hemocytes having a high nucleus/cytoplasm ratio. There are few granular or vacuolar inclusions in the cytoplasm. Phagocytes have phagocytic activity and their shape is variable depending on the amount of engulfed materials. In granulocytes, shape and size of granules are different among the species. Morula cells are characterized by several vacuoles filled with electron dense materials. In pigment cells, the bulk of the cytoplasm is occupied by one or a few vacuoles containing pigment granules. We also described some other hemocyte types found in particular species. Furthermore, we encountered free oocytes circulating in the blood in two species, Botryllus primigenus and Botrylloides lentus.

Xenogeneic rejection was observed among colonies of three botryllids, Botryllus scalaris, Botryllus primigenus, and Botrylloides simodensis. Allogeneic recognition occurs in each of these species, but the manner of allogeneic rejection differs among them. We studied xenogeneic rejection reactions among these species under the following conditions: colony contact at natural growing edges, colony contact at artificially cut surfaces, and injection of xenogeneic blood plasma into a vascular vessel. In the first two cases, xenogeneic rejection occurred only in Botryllus primigenus and Botrylloides simodensis. The features of that xenogeneic rejection were similar to those of allogeneic rejection in each of these two botryllids. Injection of xenogeneic blood plasma induced responses similar to those of allogeneic rejection in all three botryllids. It is interesting to note that colonies of Botryllus scalaris never showed any response against injected blood plasma from allogeneic incompatible colonies, unlike the responses seen in colonies of the other two botryllids under the same conditions. On the basis of these results, the relationship between allogeneic and xenogeneic rejection in botryllids is discussed.

Megalodicopia hians Oka is a solitary ascidian belonging to the family Octacnemidae inhabiting the bathyal /abyssal zone as well as other octacnemid ascidians. The phylogenetic relationship of octacnemids is open to argument because of its extraordinary morphological features due to habitat adaptation, e.g., a pharynx lacking ciliated stigmata. Aggregations of M. hians were discovered by the manned submersible Shinkai 2000 in the bathyal seafloor of Toyama Bay, Japan Sea, in 2000; this was the first in situ observation of M. hians in the Japanese coastal waters. In 2001, a total of 36 M. hians specimens were collected from the bay (592 to approximately 978 m deep). In situ observation indicated that M. hians usually opens its large oral apertures to engulf the drifting food particles in the water current. Microscopical observation of the gut contents also showed that M. hians is a non selective macrophagous feeding on small crustaceans, diatoms, detritus, and so on. Along with the position of the intestinal loop and gonad, the morphological characteristics of the tunic (integument of ascidians) suggest that M. hians is closely related to Cionidae and/or Corellidae. Some symbiotic/parasitic organisms were occasionally found in the tunic, including rod-shaped bacteria, fungi-like multicellular structure, and spawns of unknown animals.

Morphology and life history of a new species of the genus Botryllus belonging to the family Botryllidae were described in detail. This ascidian was collected from the stony shore in the vicinity of Shimoda (Shizuoka prefecture, Japan). The arrangement of ovary and testis in this ascidian was the same as that in other species of the genus Botryllus, while the embryo developed in a brood pouch formed from the invagination of peribranchial epithelium, as in the other genus Botrylloides. The processes and features of the allorecognition reaction of this ascidian were observed. The reaction showed the same processes as that in the species of the genus Botrylloides. Therefore, this ascidian has both features of the two genera of the family Botryllidae, which strongly suggests the necessity of reconsidering on the classificatory criteria of botryllid ascidians.

Xenogeneic rejection reactions were histologically examined among four compound ascidians of the genus Botrylloides; B. simodensis, B. lentus, B. fuscus and B. violaceus, to compare with the allogeneic rejections of these species. When the incompatible conspecifics were brought into contact, hemolytic rejections occurred at the point where the tunic of the two colonies was partially fused. Xenogeneic contact at their growing edges induced hemolytic rejection in some combinations (B. simodensis-B. lentus, B. lentus-B. fuscus, and B. fuscus-B. violaceus), while conspicuous reaction was not found in the other combinations. Since the hemolytic rejection requires the partial fusion of tunic, the occurrence of hemolytic rejection suggests that the tunic cuticle of the colonies does not discriminate the facing colony from conspecifics. On the other hand, whereas cut surface contact between incompatible conspecifics induced intense rejection in B. simodensis, it resulted in fusion (formation of vascular connection) even in the combination in which the growing edge contact resulted in rejection. In xenogeneic combination, the cut surface contact of colonies always resulted in an intense rejection reaction except for B. fuscus-B. violaceus in which hemolytic reactions did not occur. The absence of hemolytic rejection suggests that the effector system for rejection reaction is not activated in this combination. Activity of phenoloxidase, a key enzyme of the rejection reaction, indicates lower reactivity in B. lentus, B. fuscus and B. violaceus than that in B. simodensis.
